Optimizing the performance of silica nanoparticles functionalized with a near-infrared fluorescent dye for bioimaging applications

Modified fluorescent nanoparticles continue to emerge as promising candidates for drug delivery, bioimaging, and labeling tools for various biomedical applications. The ability of nanomaterials to fluorescently label cells allow for the enhanced detection and understanding of diseases. Silica nanoparticles have a variety of unique properties that can be harnessed for many different applications, causing their increased popularity. In combination with an organic dye, fluorescent nanoparticles demonstrate a vast range of advantageous properties including long photostability, surface modification, and signal amplification, thus allowing ease of manipulation to best suit bioimaging purposes. In this study, the Stöber method with tetraethyl orthosilicate (TEOS) and a fluorescent dye sulfo-Cy5-amine was used to synthesize fluorescent silica nanoparticles. The fluorescence spectra, zeta potential, quantum yield, cytotoxicity, and photostability were evaluated. The increased intracellular uptake and photostability of the dye-silica nanoparticles show their potential for bioimaging.

Targeted Biodegradable Near-Infrared Fluorescent Nanoparticles for Colorectal Cancer Imaging.

Colorectal cancer (CRC) is the third leading cause of cancer death in the U.S., and early detection and diagnosis are essential for effective treatment. Current methods are inadequate for rapid detection of early disease, revealing flat lesions, and delineating tumor margins with accuracy and molecular specificity. Fluorescence endoscopy can generate wide field-of-view images enabling detection of CRC lesions and margins; increased signal intensity and improved signal-to-noise ratios can increase both speed and sensitivity of cancer detection. For this purpose, we developed targeted near-infrared (NIR) fluorescent silica nanoparticles (FSNs). We tuned their size to 50-200 nm and conjugated their surface with an antibody to carcinoembryonic antigen (CEA) to prepare CEA-FSNs. The physicochemical properties and biodegradable profiles of CEA-FSN were characterized, and molecular targeting was verified in culture using HT29 (CEA positive) and HCT116 (CEA negative) cells. CEA-FSNs bound to the HT29 cells to a greater extent than to the HCT116 cells, and smaller CEA-FSNs were internalized into HT29 cells more efficiently than larger CEA-FSNs. After intravenous administration of CEA-FSNs, a significantly greater signal was observed from the CEA-positive HT29 than the CEA-negative HCT116 tumors in xenografted mice. In F344-PIRC rats, polyps in the intestine were detected by white-light endoscopy, and NIR fluorescent signals were found in the excised intestinal tissue after topical application of CEA-FSNs. Immunofluorescence imaging of excised tissue sections demonstrated that the particle signals coregistered with signals for both CRC and CEA. These results indicate that CEA-FSNs have potential as a molecular imaging marker for early diagnosis of CRC.

Combining Bioorthogonal Chemistry with Fluorescent Silica Nanoparticles for the Ultrasensitive Detection of the HIV-1 p24 Antigen.

Early detection and viral concentration monitoring of human immunodeficiency virus in resource-poor settings are important to control disease spread and reduce mortality. Nucleic acid amplification tests are expensive for low-resource settings. Lateral flow antibody tests are not sensitive if testing is performed within 7-10 days, and these tests are not quantitative. We describe a signal enhancement technique based on fluorescent silica nanoparticles and bioorthogonal chemistries for the femtomolar detection of the HIV-1 p24 antigen. We developed a magnetic bead-based assay, wherein we used fluorescent-dye-encapsulated silica nanoparticles as reporters. The number of reporters was increased by using bioorthogonal chemistry to provide signal enhancement. The limit and range of detection of the sandwich immunoassay using alternating multiple layers for p24 in human serum were found to be 46 fg/mL (1.84 fM) and 46 fg/mL to 10 ng/mL, respectively. This simple assay was 217-fold higher in sensitivity compared to that of commercial enzyme-linked immunoassays (limit of detection of 10 pg/mL).

A non-residue surface modification strategy for active-targeting fluorescent silica nanoparticles to cellular organelles.

Silica nanoparticles (SiNPs) with a chemically modified surface typically have a complicated chemical composition, which can significantly differ from their intended design. In this study, we systematically studied the effects of two surface modification methods on active-targeting of intracellular organelles of SiNPs: (1) the widely used step-by-step approach, which involves modifying SiNPs in two steps, i.e., the outer surface of SiNPs was firstly modified with amino groups and then these amino groups were linkedwith targeting groups, and (2) a newly developed one-step approach in which the ligand-silane complex is initially synthesized, followed by chemically immobilizing the complex on the surface of SiNPs. In the one-step approach, the molar ratio of reactants was precisely tuned so that there are no reactive groups left on the outer surface of SiNPs. Two essential organelles, mitochondria and the nucleus, were selected to compare the targeting performances of SiNPs synthesized via these two approaches. By characterizing physicochemical properties, including structural properties, the number of amino groups, surface charge, polydispersity, and cell colocalization, we demonstrated that SiNPs synthesized via the one-step approach with no residual linkage groups on their surface showed significantly improved mitochondria- and nucleus-targeting performances. This precise control of surface properties allows for optimized biological behavior and active-targeting efficiency of SiNPs. We anticipate that such simple and efficient synthetic strategies will enable the synthesis of effective SiNPs for active-targeting organelles in various biological applications.

Usefulness of a new immunochromatographic assay using fluorescent silica nanoparticles for serodiagnosis of Thai patients with amebiasis.

A fluorescence immunochromatography (FIC) kit was developed recently using fluorescent silica nanoparticles coated with a recombinant C-terminal fragment of the surface lectin intermediate subunit (C-Igl) of Entamoeba histolytica to establish rapid serodiagnosis of amebiasis. We further evaluated the system using serum samples from 52 Thai patients with amebiasis. Of the patients, 50 (96%) tested positive using FIC. The samples were also tested using enzyme-linked immunosorbent assay (ELISA) with C-Igl as the antigen. Two samples were negative on ELISA but positive on FIC. The correlation coefficient between the fluorescence intensity using FIC and the optical density value using ELISA was 0.5390, indicating a moderate correlation between the two tests. Serum samples from 20 patients with malaria and 22 patients with Clostridioides difficile infection were also tested using FIC. The false-positive rates were 4/20 (20%) and 1/22 (4%) in patients with malaria and C. difficile infection, respectively. Combining the data from the present study with our previous study, the sensitivity and specificity of FIC were determined to be 98.5% and 95.2%, respectively. The results of the 50 samples were studied using a fluorescence scope and a fluorescence intensity reader, and the findings were compared. Disagreements were found in only two samples showing near-borderline fluorescence intensity, indicating that the use of scope was adequate for judging the results. These results demonstrate that FIC is a simple and rapid test for the serodiagnosis of amebiasis.

An ultra-sensitive SARS-CoV-2 antigen optical biosensor based on angiotensin converting enzyme 2 (ACE-2) functionalized magnetic-fluorescent silica nanoparticles.

This work reports on the design and synthesis of an angiotensin-converting enzyme 2 (ACE-2) functionalized magnetic fluorescent silica nanoparticles (Fe-FSNP) as a biosensing platform to detect severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) antigen. Iron oxide (Fe3O4) nanoparticles were synthesized via ultrasonic-assisted coprecipitation and then coated with fluorescent silica nanoparticles (FSNP) through thesol-gelmethod forming the Fe-FSNP samples. Silica obtained from local geothermal powerplant was used in this work and Rhodamine B was chosen as the incorporated fluorescent dye, hence this reports for the first time ACE-2 was immobilized on the natural silica surface. The Fe-FSNP nanoparticle consists of a 18-25 nm magnetic core and a silica shell with a thickness of 30 nm as confirmed from the transmission electron microscopy image. Successful surface functionalization of the Fe-FSNP with ACE-2 as bioreceptor was conducted through hydrosylilation reaction and confirmed through the Fourier transform infrared spectroscopy. The detection of SARS-Cov-2 antigen by Fe-FSNP/ACE2 was measured through the change in its maximum fluorescence intensity at 588 nm where fluorescence- quenching had occurred. The biosensing platform showed a rapid response at 30 min with a linear range of 10-6to 10-2μg ml-1. The magnetic-fluorescent properties of the nanoparticle enables an ultra-sensitive detection of SARS-Cov-2 antigen with the limit of detection as low as 2 fg ml-1.

Development of amine-functionalized fluorescent silica nanoparticles from coal fly ash as a sustainable source for nanofertilizer

Scaling up the synthesis of fluorescent silica nanoparticles to meet the current demand in diverse applications involves technological limitations. The present study relates to the hydrothermal synthesis of water-soluble, crystalline, blue-emitting amine-functionalized silica nanoparticles from coal fly ash sustainably and economically. This study used tertiary amine (trimethylamine) to prepare amine-functionalized fluorescent silica nanoparticles, enhancing fluorescence quantum yield and nitrogen content for nanofertilizer application. The TEM and FESEM studies show that the silica nanoparticles have a spherical morphology with an average diameter of 4.0 nm. The x-ray photoelectron and Fourier transform infrared spectroscopy studies reveal the presence of the amine group at the surface of silica nanoparticles. The silica nanoparticles exhibit blue fluorescence with an emission maximum of 454 nm at 370 nm excitation and show excitation-dependent emission properties in the aqueous medium. With the perfect spectral overlap between silica nanoparticle emission (donor) and chlorophyll absorption (acceptor), fluorescent silica nanoparticles enhance plant photosynthesis rate by resonance energy transfer. This process accelerates the photosynthesis rate to improve the individual plant’s quality and growth. These findings suggested that the fly ash-derived functionalized silica nanoparticles could be employed as nanofertilizers and novel delivery agents.

Near-Infrared Fluorescent Silica Nanoparticles Based on Gold-Silver Alloy Nanoclusters for Clinical Diagnosis.

In clinical diagnosis, fluorescent particles are applied to detect analytes in biofluids, such as blood and saliva. However, current fluorescence detection methods have not been optimized to account for the overlapping autofluorescence peaks of biological substances. Gold and silver nanoclusters are known to the novel fluorescent materials and their emission wavelengths depend on cluster size. In this study, we developed fluorescent silica nanoparticles using gold-silver alloy nanoclusters and chitosan (CS) (NH2-SiO2@Au@CS@AuAg) by the layer-by-layer method. Under UV-light irradiation at 365 nm, the emission wavelength of NH2-SiO2@Au@CS@AuAg reached 750 nm in the near-IR region. Scanning electron microscopy images revealed that the shape of NH2-SiO2@Au@CS@AuAg was uniform and spherical. The fluorescence spectrum of horse blood obtained in the presence of NH2-SiO2@Au@CS@AuAg contained a specific fluorescence peak attributed to NH2-SiO2@Au@CS@AuAg, which was distinguishable from the autofluorescence peaks. These results showed that NH2-SiO2@Au@CS@AuAg has advantageous fluorescence properties for clinical diagnostic applications.

Overcoming Barriers Associated with Oral Delivery of Differently Sized Fluorescent Core-Shell Silica Nanoparticles.

Oral delivery, while a highly desirable form of nanoparticle-drug administration, is limited by challenges associated with overcoming several biological barriers. Here, the authors study how fluorescent and poly(ethylene glycol)-coated (PEGylated) core-shell silica nanoparticles sized 5 to 50nm interact with major barriers including intestinal mucus, intestinal epithelium, and stomach acid. From imaging fluorescence correlation spectroscopy studies using quasi-total internal reflection fluorescence microscopy, diffusion of nanoparticles through highly scattering mucus is progressively hindered above a critical hydrodynamic size around 20nm. By studying Caco-2 cell monolayers mimicking the intestinal epithelia, it is observed that ultrasmall nanoparticles below 10nm diameter (Cornell prime dots, [C' dots]) show permeabilities correlated with high absorption in humans from primarily enhanced passive passage through tight junctions. Particles above 20nm diameter exclusively show active transport through cells. After establishing C' dot stability in artificial gastric juice, in vivo oral gavage experiments in mice demonstrate successful passage through the body followed by renal clearance without protein corona formation. Results suggest C' dots as viable candidates for oral administration to patients with a proven pathway towards clinical translation and may generate renewed interest in examining silica as a food additive and its effects on nutrition and health.

Combined toxicity of fluorescent silica nanoparticles with cadmium in Ceriodaphnia dubia: Interactive effects of natural organic matter and green algae feeding

The environmental risks of silica nanoparticles (SiNP) reported in the literature are contradictory and bring into question its safety for use in consumer applications. Organisms are never exposed to NPs alone in the real environment, while studies of the combined toxicity of SiNP are limited. To address this, we compared the acute toxicity of fluorescent core-shell SiNPs alone and in mixtures with Cd2+ to Ceriodaphnia dubia in the absence and presence of NOM. We identified biodistribution and feeding behaviour in addition to the traditional endpoints. NOM increased the colloidal stability of SiNPs in reconstituted water. In immobility tests, no significant effects were observed from Cd2+ exposure with NOM and varying concentrations of SiNPs. A similar pattern of curve dose-response was observed for varying concentrations of SiNPs and increasing Cd2+ concentration and constant NOM. Fluorescence microscopy verified a dose-dependent bioaccumulation of SiNPs in C. dubia. Co-exposure to 10 mg L−1 SiNP with NOM and Cd2+ resulted in a stimulated stress feeding response at the lower Cd2+ concentrations which declined at the higher dose due to a functional impairment of the digestive tract. Alterations in feeding behaviour and the increasing bioaccumulation of SiNP indicate a potential ecological risk for Ceriodaphnia dubia from the mixture exposure.

Preparation of polyacrylonitrile-based solution blow spinning fluorescent nanofibers from perylene-doped silica nanoparticles

ABSTRACT Fluorescent agents have been used to improve the anticounterfeiting of marketable products. It has been vital to enhance the engineering processing of fluorescent agents and materials matrices to prepare a mechanically reliable nanocomposite material with anticounterfeiting properties. Herein, new fluorescent and mechanically reliable inorganic-organic hybrid nanofibrous were developed by solution blowing spinning of polyacrylonitrile (PAN) encapsulating perylene-doped silica nanoparticles (PSiN). Two different types of fluorescent nanofibers were prepared using PSiN as an active doping agent and polyacrylonitrile as a hosting material. Fluorescent nanofibers were provided by solution blowing spinning technology utilizing perylene pigments, including perylene potassium (PMIK) and mono-imide sodium (PMINa) salts, and perylene diimide (PDI). In order to create a transparent nanofibrous film, fluorescent perylene-doped silica nanoparticles must be efficiently dispersed in a polyacrylonitrile solution without aggregation. The prepared fluorescent silica nanoparticles (SiN) displayed diameters ranging between 45 and 65 nm. The provided solution blow spun fluorescent PAN nanofibers integrated with PMINa, PMIK or PDI, showed smooth surface with diameters ranging between 350 and 550 nm. The prepared fluorescent PSiN and PSiN@PAN nanofibers were examined by several techniques, including transmission electron microscopy (TEM), scanning electron microscopy (SEM), and infrared spectroscopy (FT-IR). The solution blow spun nanofibers demonstrated an absorbance band at 458 nm and a fluorescence peak at 647 nm. The prepared nanofibers displayed a reversible emission upon repeated illumination with UV light. The emission properties of the prepared PSiN@PAN nanofibers were studied. The cytotoxic properties of the nanofibrous films were also explored. The current strategy introduces fluorescent nanostructured fibers that pave the way for the preparation of optical textiles such as optical devices, anticounterfeiting applications, and flexible displays.

Fluorescent silica nanoparticles as nano-chemosensors for the sequential detection of Pb2+ ions and bacterial-spore biomarker dipicolinic acid (DPA) in aqueous solution

Herein, we report fluorescein-labelled silica nanoparticles (FSNP) which serve as fluorescent nano-chemosensors for sequential detection of Pb2+ (which is a toxic heavy metal) and dipicolinic acid (DPA) (which is a distinctive indicator biomarker of bacterial spores) with high sensitivity and selectivity. The fluorescence of FSNP is quenched because of the complex formation between Pb2+ ions and surface amide groups, however, the fluorescence is recovered in contact with DPA, resulting from the association of DPA with surface bound Pb2+ ions. FSNP-Pb2+ complexes show high sensitivity towards DPA with a low detection limit of 850 nM which is approximately seventy times lower than the infectious dosage of bacterial spores (60 μM). Lateral flow test platform was further developed to show the applicability and practicability of our system.

Flowing through Gastrointestinal Barriers with Model Nanoparticles: From Complex Fluids to Model Human Intestinal Epithelium Permeation.

Most nanomaterial-based medicines are intravenously applied since oral administration comprises challenging-related biological obstacles, such as interactions with distinct digestive fluids and their transport through the intestinal barrier. Moreover, there is a lack of nanoparticle-based studies that faithfully consider the above-cited obstacles and boost oral-administered nanomedicines' rational design. In this study, the physicochemical stability of fluorescent model silica nanoparticles (f-SiO2NPs) passing through all simulated gastrointestinal fluids (salivary, gastric, and intestinal) and their absorption and transport across a model human intestinal epithelium barrier are investigated. An aggregation/disaggregation f-SiO2NPs process is identified, although these particles remain chemically and physically stable after exposure to digestive fluids. Further, fine imaging of f-SiO2NPs through the absorption and transport across the human intestinal epithelium indicates that nanoparticle transport is time-dependent. The above-presented protocol shows tremendous potential for deciphering fundamental gastrointestinal nanoparticles' evolution and can contribute to rational oral administration-based nanomedicine design.

Intravital imaging of osteocyte integrin dynamics with locally injectable fluorescent nanoparticles

Osteocytes are the resident mechanosensory cells in bone. They are responsible for skeletal homeostasis and adaptation to mechanical cues. Integrin proteins play a prominent role in osteocyte mechanotransduction, but the details are not well stratified. Intravital imaging with multiphoton microscopy presents an opportunity to study molecular level mechanobiological events in vivo and presents an opportunity to study integrin dynamics in osteocytes. However, fluorescent imaging limitations with respect to excessive optical scattering and low signal to noise ratio caused by mineralized bone matrix make such investigations non-trivial. Here, we demonstrate that ultra-small and bright fluorescent core-shell silica nanoparticles (<7 nm diameter), known as Cornell Prime Dots (C'Dots), are well-suited for the in vivo bone microenvironment and can improve intravital imaging capabilities. We report validation studies for C'Dots as a novel, locally injectable in vivo osteocyte imaging tool for both non-specific cellular uptake and for targeting integrins. The pharmacokinetics of C'Dots reveal distinct sex differences in nanoparticle intracellular dynamics and clearance in osteocytes, which represents a novel topic of study in bone biology. Integrin-targeted C'Dots were used to study osteocyte integrin dynamics. To the best of our knowledge, we report here the first evidence of osteocyte integrin endocytosis and recycling in vivo. Our results provide novel insights in osteocyte biology and will open up new lines of investigation that were previously unavailable in vivo.

Synthesis of core-shell fluorescent silica nanoparticles with opposite surface charges for transport studies of nanofluids in porous media

Silica nanoparticles (SiNPs) have been extensively studied as additives in injected fluids for enhanced oil recovery. Typically, surfactants are used to increase colloidal stability, but little is known about the transport of SiNPs in porous media because of the scarcity of reliable quantification techniques. In this study, we investigated the use of core-shell silica-based fluorescent SiNPs as tracers for transport studies of nanofluids containing surfactants. Two synthetic strategies were used to obtain distinct structures of fluorescent SiNPs with similar cores but different surface groups, namely silanol and aminopropyl, comparable to those reported in nanofluid applications. The synthesised NPs had comparable sizes and morphologies but displayed opposite surface charges when dispersed in aqueous solutions. The main colloidal properties and time stability of the fluorescence response were determined in the presence of an anionic surfactant and compared with regard to their potential use as tracers for transport studies. Both fluorescent SiNPs were successfully quantified in the effluent after flowing through the glass bead column. The SiNP-containing aminopropyl surface groups showed the greatest stability and lowest adsorption compared with those grafted with silanol groups. We attribute this behaviour to the effect of stronger attractive interactions between the NPs and surfactants. The potential of these fluorescent NPs as sensitive tracers for transport and adsorption studies in nanofluids containing surfactants was demonstrated.

Highly bright full-color emission from dye-doped silica nanoparticles with prevention of dye self-quenching

Although fluorescent dye-doped silica nanoparticles (SiOx NPs) have been widely studied for a broad range of optical probes and devices, it is still challenging to fabricate highly bright dye-doped SiOx NPs because the luminescence quantum yields of dye-doped SiOx NPs are often lower than those of the dye alone. Herein, we report highly bright full-color emissions from dye-doped SiOx NPs that prevent dye self-quenching. Green and red fluorescence-emitting dye-doped silica nanoparticles (G-SiOx and R-SiOx NPs) were synthesized from 3-(aminopropyl) triethoxysilane (APTES) and fluorescence dyes (fluorescein, Rhodamine B). The quantum yields (QY) of G-SiOx and R-SiOx NPs were 80.9% and 25.5%, respectively; doping these dyes into the SiOx NPs did not decrease the QY values of the dyes. The combination of bule luminescent SiOx NPs (QY = 32.3%) with G-and R-SiOx NPs displayed an intense pure white (x = 0.33, y = 0.33) fluorescence emission.

Preparation of fluorescent ink using perylene-encapsulated silica nanoparticles toward authentication of documents

Novel inorganic–organic hybrid fluorescent ink was produced for anticounterfeiting applications. Pigment/resin ink formulations were prepared from the thermally and photostable fluorescent perylene-doped silica nanoparticles (PSN). To create a colorless printed film from pigment/resin ink, the fluorescent perylene must be physically disseminated without clumping. Using screen-printing technique, the pigment/resin combination was successfully applied to sheets of cellulose paper. CIE Lab tests showed that pigment concentrations as low as 1 % result in a red emission when a uniform fluorescent layer was produced on the pale yellow cellulose paper surface. These printed sheets displayed an absorption band at 436 nm and an emission band at 625 nm. The fluorescent PSN exhibited diameters of 6–33 nm. The printed paper sheet was analyzed using various spectroscopic techniques, including scanning electron microscopy (SEM), infrared spectroscopy (FT-IR), energy-dispersive X-ray (EDX), and wavelength dispersive X-ray fluorescence (WDXRF). Upon repeated exposure to UV light, the prints exhibited a reversible fluorescence. Investigations into the ink rheological characteristics, stability, and printability were conducted.

Bioactivity, Uptake, and Distribution of Prothioconazole Loaded on Fluorescent Double-Hollow Shelled Mesoporous Silica in Soybean Plants.

Prothioconazole (PTC) has been widely utilized for plant fungal disease control, but its metabolite prothioconazole-desthio (PTC-d) exhibits reproductive toxicity. In the present study, carbon quantum dot (CQD)-modified fluorescent double-hollow shelled mesoporous silica nanoparticles (FL-MSNs) loaded with PTC, referred to as PTC@FL-MSNs, were constructed with an average size of 369 nm and a loading capacity of 28.1 wt %, which could increase the antifungal efficiency of PTC. In addition, upright fluorescence microscope and UPLC-MS/MS studies showed that PTC@FL-MSNs could be effectively transported via root uptake and foliar spray in soybean plants. Compared to a 30% PTC dispersible oil suspension agent, the PTC@FL-MSN treatment group showed higher concentrations (leaves: 0.50 > 0.48 mg/kg), longer half-lives for degradation (leaves: 3.62 > 3.21 d; roots: 3.39 > 2.82 d), and fewer metabolites. These findings suggest that sustained pesticide release and toxicity reduction are potential applications for PTC nanofungicide delivery technology.

Engineered Ultrasmall Nanoparticle Drug-Immune Conjugates with "Hit and Run" Tumor Delivery to Eradicate Gastric Cancer.

Despite advances by recently approved antibody-drug conjugates in treating advanced gastric cancer patients, substantial limitations remain. Here, several key obstacles are overcome by developing a first-in-class ultrasmall (sub-8-nanometer (nm)) anti-human epidermal growth factor receptor 2 (HER2)-targeting drug-immune conjugate nanoparticle therapy. This multivalent fluorescent core-shell silica nanoparticle bears multiple anti-HER2 single-chain variable fragments (scFv), topoisomerase inhibitors, and deferoxamine moieties. Most surprisingly, drawing upon its favorable physicochemical, pharmacokinetic, clearance, and target-specific dual-modality imaging properties in a "hit and run" approach, this conjugate eradicated HER2-expressing gastric tumors without any evidence of tumor regrowth, while exhibiting a wide therapeutic index. Therapeutic response mechanisms are accompanied by the activation of functional markers, as well as pathway-specific inhibition. Results highlight the potential clinical utility of this molecularly engineered particle drug-immune conjugate and underscore the versatility of the base platform as a carrier for conjugating an array of other immune products and payloads.

Preliminary Study of Modified Fluorescent Silica Nanoparticles for the Detection of IgY Antibody

Biosensors must display high sensitivity and selectivity with fast response and be cost-effective. Silica nanoparticles are profoundly promising as versatile signal platforms opening up new possibilities for bioassays and dynamic units in biosensing systems. Fluorescent silica nanoparticles (FSNP) may be a key nanomaterial for labeling antibodies or antigens for biosensor application. Integration FSNP with functional molecules utilizing different surface modification techniques can bring out great improvements such as selective recognition, sensitive detection, and signal amplification because FSNP can bind with a large number of dye molecules within a silica matrix. The FSNP conjugated with anti-IgY can be applied for the detection of IgY antibody. FSNP was fabricated using the sol-gel method with hydrosilylation techniques for silica surface modification. N-H of amide groups from FSNP-anti-IgY ware observed in the FTIR spectra at 1648 cm-1 indicating that the conjugation was successful. The fluorescence intensity of FSNP-anti-IgY after detection of IgY was applied for immunodetection and measured using fluorescence spectroscopy. The intensity of FSNP-anti-IgY was 156.82, 368.31, and 648.00 a.u for the 0.05, 0.1, 0.5 µg/ml of the sample concentrations, respectively. The results showed that the modified FSNP-anti-IgY may be used for the fluorescence detection of antibodies.
