Prooxidant and anti-inflammatory potential of Garcinia xanthochymus fruit and its phytochemical characterisation by UHPLC-Q-Orbitrap HRMS

Pro-oxidants play a crucial role in cancer by causing oxidative stress that leads to apoptosis. The present study demonstrates the prooxidant and anti-inflammatory potential of ethyl acetate and methanolic extracts of Garcinia xanthochymus fruit. Oxidation of Trolox and NADH activity indicated the pro-oxidant capacity of the extracts. Significant decrease in cell viability in B16F10 and MDA-MB-231 cancer cell lines and significant increase in caspase 3 activity after treatment with extracts indicated pro-oxidant induced apoptosis. Pre-treatment with the extracts significantly inhibited ROS, reduced NO production, inhibited LPS-induced COX-2 and suppressed IL-6 and TNF-α expression. HRMS analysis showed the presence of compounds like biflavonoids, xanthones, phloroglucinols, benzophenones, etc. The fruit is rich in total phenolic and flavonoid contents, and have DPPH radical scavenging, ferric reducing antioxidant and metal chelating potential. This study report for the first time about the anticancer and anti-inflammatory properties of G. xanthochymus whole fruit.

Appraisal of chemical composition and biological activities of thyme essential oil: an insight to in-silico molecular docking studies

The current study aims to ascertain the chemical composition and biological activities of thyme leaves essential oil (TEO), with an emphasis on its in silico molecular docking studies targeting the antibacterial properties of its constituents. TEO was extracted from dried leaves using a standardized procedure, with a yield of 0.75%. The chemical constituents of TEO were identified using Gas Chromatography-Mass Spectrometry (GC-MS). The primary compounds detected included carvacrol, o-acetyl thymol, α-pinene, and spathulenol. Subsequently, the antioxidant activity of the isolated oil was measured, and the results showed that the total phenolic contents (TPC) was 4.69 ± 0.17 GAE mg/100 g, the total flavonoid contents (TFC) was 1.45 ± 0.04 CE mg/100 g, the ferric reducing antioxidant potential (FRAP) ranged from 0.07-0.81, and the DPPH radical scavenging activity was 42.88-86.94%. TEO exhibited notable antibacterial activity against certain strains of Staphylococcus aureus and Escherichia coli. The molecular docking studies were then conducted to further investigate the interaction of TEO’s potent metabolites with key bacterial enzymes, identifying spathulenol and caryophyllene as the best-interacting ligands. Hemolytic activity tests of TEO indicated negligible cytotoxic effects. These findings support that TEO holds great potential for the development of functional food and nutra-pharmaceuticals.

Physicochemical quality retention of ‘Sucrier’ bananas (Musa acuminata AA group cv. ‘Kluai Khai’) by jicama starch-based postharvest treatment

The objective of this research was to develop a new type of edible coating using jicama starch (JS) in order to control the postharvest deterioration of ‘Sucrier’ bananas (Musa acuminata AA group cv. ‘Kluai Khai’) during the ripening process in ambient condition (25 ± 1 °C, 85-90 % RH). The preliminary experiment was carried out to investigate the impact of JS concentration (0, 2.5, 5.0, and 7.5 % w/v) on the postharvest quality of bananas. The application of JS demonstrated a delay in banana ripening, including color development, weight loss, and fruit firmness. Within the JS-treated banana groups, 5.0 % treated bananas displayed higher firmness compared to the other groups. Therefore, a validation experiment was conducted to verify the subsequent physicochemical alterations in bananas after subjecting bananas to 5.0 % JS coating in comparison to non-treated bananas during storage at ambient condition. The JS treatment maintained the color and visual appearance by minimizing the formation of malondialdehyde content and electrical conductivity during storage. Furthermore, it resulted in a delay in the increases in total soluble solid, titratable acidity, BrimA, and total carotenoid concentrations in the pulp. Nevertheless, the ascorbic acid and phenolic contents and 2,2-diphenyl-2-picrylhydrazyl scavenging activity of the control samples were likely to be higher than those of the treated bananas, whereas the ferric reducing antioxidant potential of both control and treated bananas showed a comparable trend throughout storage. The outcomes demonstrate that the utilization of JS is an effective approach for improving the overall storage capacity of ‘Sucrier’ bananas.

Evaluation of the proximate composition and antioxidant capacity of some seaweeds from the Konkan coast of India

Minerals, antioxidants, and other nutrients that promote health can be found in abundance in seaweeds. In this study, we evaluated the proximate composition and antioxidant capacity of some seaweeds from Konkan coast of India. There were notable differences in the proximate compositions between the species. The highest protein content was found in Pyropia vietnamensis (~49 mg. g−1 dw), whereas the highest carbohydrate (~83 mg. g−1 dw), lipids (56 mg. g−1 dw) and vitamins were found in Stoechospermum marginatum. Total phenolic content (TPC) and antioxidant activity were estimated from a Methanolic extracts. The antioxidant capacity of the seaweeds was evaluated using 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, ABTS (2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid), ferric reducing antioxidant power (FRAP) assay and hydrogen peroxide free radical scavenging activity assay. Folin-Ciocalteu method was used to quantify total phenolic content (TPC), and Ulva fasciata had the highest TPC (24.30 ± 3 mg GAE. g−1 dry weight). In U. fasciata, the antioxidant activity was ~62 %, with an IC50 value of 0.057 ± 0.01 mM. Acanthophora spicifera was found to have a strong reducing power, with an IC50 value 0.012 ± 0.002 mM. H2O2 scavenging activity (~79 %) with an IC50 value of 0.006 ± 0.001 mM and 67 % ABTS radical scavenging activity (IC50 = 0.005) was highest in S. marginatum. Analysis showed that TPC exhibited strong positive correlation with both the ferric reducing antioxidant potential and DPPH antioxidant activity. These results showed that these seaweeds have significant antioxidant capacity and rich nutritional properties, which suggest they could be useful sources of functional ingredients for dietary supplements and other food products.

Morphological, biochemical and molecular characterization of short-day tropical Indian garlic (Allium sativum L.)

Garlic, an asexually propagated bulbous crop, displays a wide diversity based on its morphological traits and biochemical compositions. This study investigated the genetic variability of Indian garlic through morphological, biochemical, and molecular markers. Twenty-nine genotypes along with three Allium species as outgroup were included in the present study. Observations were recorded on 14 quantitative traits, 17 qualitative traits, and 9 biochemical traits in fresh garlic. Significant variability was observed among genotypes for different characters. All the morphological and biochemical traits showed higher phenotypic coefficient of variation (PCV) than genotypic coefficient of variation (GCV) revealing the role of environment in trait expression. High to moderate heritability and genetic advance as percent mean were recorded for different traits except dry matter and Total Soluble Solids (TSS). Correlation analysis revealed the highest positive correlation between total yield, marketable yield, Ferric Reducing Antioxidant Potential (FRAP) and 2,2-diphenyl-1-picrylhyrazyl (DPPH). Cluster analysis differentiated all the genotypes into three major clusters based on morphological and biochemical traits. 214 Simple Sequence Repeats (SSRs) were screened and nine markers exhibited polymorphism. Cluster analysis using molecular markers revealed 4 distinct clusters. The observations from this study will help in the identification of diverse garlic germplasm for its efficient management and duplicate identification of germplasm resources.

Dietary total antioxidant capacity in relation to disease severity and risk of mortality in cirrhosis; results from a cohort study

Liver Cirrhosis, defined as the final stage of chronic liver disease, may become more prevalent in the lower level of body defense against oxidation and inflammation. Therefore, we assessed the association of dietary total antioxidant capacity (DTAC) with the severity and mortality of cirrhosis in a cohort study. 120 newly diagnosed cirrhosis patients from Tehran, Iran, participated in this study. The patients’ habitual diet was assessed using a 168-item validated food frequency questionnaire. Both ferric-reducing antioxidant potential (FRAP) and oxygen radical scavenging capacity (ORAC) methods were computed to achieve DTAC scores. The association between DTAC with disease severity and mortality was estimated by multivariate linear regression and cox proportional hazards regression models. Dietary total antioxidant capacity-ORAC had a significant inverse association with disease severity in both crude and adjusted models (P for trend: <0.001 and 0.016 respectively). The risk of mortality in the first and second tertiles of ORAC was 5.56 (95 % CI: 2.25–13.75; P = 0.002) and 3.20 (95 % CI: 1.25–8.19; P = 0.015) higher than those in the third category, respectively. In conclusion, a higher antioxidant capacity of diet is associated with less disease severity and mortality risk in cirrhosis.

CBD oil by-product (Hemp flakes): Evaluation for nutritional composition, heavy metals and functionality as a food ingredient

BackgroundThe recent interest among consumers in industrial hemp due to health and wellness benefits has led to several products from industrial hemp, including cannabidiol (CBD) oil. CBD oil extraction from hemp buds and flowers generates by-product biomass (hemp flakes), often posing disposal challenges and with little or no applications. We hypothesized that hemp flakes possess residual compounds with nutritional and health value that could be used to improve utilization. MethodsLocally sourced hemp flakes were compared to three commercial hemp protein products. The nutritional composition (proximate analysis), heavy metals (Al, Cu, As, Pb, Co, Cd), and functional composition (phenolic and antioxidant properties–total phenolic compounds (TPC), total flavonoid compounds (TFC), ferric reducing antioxidant potential (FRAP), 1,1–diphenyl-1-picrylhydrazyl (DPPH), Trolox equivalent antioxidant capacity (TEAC)), (CBD, cannabiodiolic acid–CBDA, cannabichromene–CBC, cannabigerol–CBG, and cannabinol–CBN) contents were determined and compared. FindingsHemp flakes had a similar nutritional composition to commercial hemp protein products, with heavy metal levels within FDA allowed limits. The by-product had significantly higher CBDA levels than commercial products. Overall, hemp flakes had comparable nutrient composition and antioxidant capabilities. Based on the protein composition of hemp flakes (31.62 %) versus the highest commercial product (43 %), hemp flakes are an acceptable functional food ingredient.

Evaluation of Chemical Composition among the Multi Colored Germplasm of Abrus precatorius L.

The medicinal plant Abrus precatorius L. was traditionally used in the Siddha and Ayurvedic systems of medicine in India. The Indian center of origin holds a vast variability in its seed color. The objective of this study was to assess the total monomeric anthocyanin, flavonol, as well as the antioxidative potential, protein content and ash content among the accessions. A total of 99 accessions conserved in the Indian National Genebank were used in this study. The methods used for the estimation of total monomeric anthocyanin, flavonol, as well as the antioxidative potential, protein content and ash content were the pH differential method, Oomah method, Ferric Reducing Antioxidant Potential, Dumas method and gravimetric method, respectively. The completely black colored accession was recorded with highest total monomeric anthocyanin (51.95 mg/100 g of cyanidin 3-glucoside equivalent) and flavonol content (66.41 mg/g of quercetin equivalent). Red + black colored accessions have recorded the maximum value with respect to antioxidants (14.18 mg/g of gallic acid equivalent). The highest amount of protein content was found in a completely white colored accession (20.67%) and the maximum ash content was recorded in red + black colored accession (4.01%). The promising accessions identified can be used by pharmaceutical companies in drug development and in curing degenerative diseases.

Association of dietary total antioxidant capacity with general and abdominal obesity in type 2 diabetes mellitus patients.

The dual existence of Type 2 Diabetes Mellitus (T2DM) and obesity within a single individual may describe a combined adverse health effects, including impaired quality of life and increased risk for cardiovascular diseases (CVDs). Oxidative stress is a contributing factor to the pathogenesis of obesity. Meanwhile, dietary antioxidants may improve the antioxidant defense system and thereby decrease oxidative injury. Dietary total antioxidant capacity (TAC) is usually used to investigate the potential health effects of dietary antioxidant intake on several oxidative stress induced chronic diseases. This study aimed to examine the association of dietary TAC with obesity-related features in T2DM patients. The present study included 254 type 2 diabetes outpatients with a mean (SD) age of 54.52 (7.21) years and mean (SD) diabetes duration of 8.2 (6.4) years. Data on dietary intake was assessed using a validated food frequency questionnaire. Dietary TAC was estimated by ferric reducing antioxidant potential (FRAP) method. Anthropometric, clinical and lifestyle characteristics were all collected. In linear regression analyses, dietary antioxidant capacity was inversely associated with body mass index (β = -0,231; 95% CI, -0,419 to -0,042), waist circumference (β = -0,427; 95% CI, -0,849 to -0,006) and fat mass percentage (β = -0,328; 95% CI, -0,545 to -0,112) independently of the assessed confounding variables. Interestingly, dietary TAC showed positive and significant associations with vitamin A, vitamin C, β-carotene, magnesium, folic acid and iron intakes, after adjusting for age and daily energy intake. Higher intake of dietary TAC was in association with lower indices of general and central obesity in T2DM patients. Therefore, dietary recommendations for counteracting obesity in patients with T2DM should take into account a high dietary TAC.

Studies on the Nutritional, Minerals, Mineral Ratio, Anti-nutritional, Molar Ratio and Antioxidant Compositions of Four Selected Leafy Wild Vegetables in Ado-Ekiti, Ekiti State, Nigeria

The study investigated the nutritional, mineral Anti-nutritional factors, molar ratios and antioxidants compositions in the leaves of four selected wild edible vegetables (Piper guinensis, Piper umbelatum, Celosia argentia and Jatropha tajorensis) collected in Ado Ekiti, Ekiti State, Nigeria. The proximate, minerals and anti-nutrient compositions of the samples were determined using standard procedures. Also antioxidant activity of the samples were detected using standard assays such as 1, 1, diphenyl-2-2 picrylhydrazyl (DDPH) and Ferric Reducing Antioxidant Potential (FRAP).The mineral ratio, molar ratio and Mineral Safety Index were calculated from mineral results. The results of the proximate compositions for the four wild vegetables for moisture content, ash, crude fat, crude fiber, crude protein and carbohydrate ranged from 11.16 - 12.15%, 2.76-4.03%, 4.07 -7.31%, 13.33-16.73%, 7.36 – 10.09%, and 52.09 -58.22% respectively,. The results of the mineral composition revealed high content of K, moderate content of Ca, P, and low contents of Mg, Fe, Zn, Cu and very low concentrations of Pb and Ni. Cd was absent in all the samples investigated. Zn/Cu and Fe/Cu ratios indicated low copper load. Antinutrients factors in the vegetable were low with the exception of phytate which can be reduced to non-toxic level through boiling or cooking. The calculated mole ratio for [Phy][Zn], [Ca]/[Phy], [Phy]/[Fe] and [Ca][Phy]/[Zn] conformed with the standard values. The results of the antioxidant properties showed that the wild vegetables are good natural antioxidant sources. The assessment of the vegetables showed that they are rich sources of crude protein, crude fiber and carbohydrate with high energy calories. The mineral ratios and mineral safety index suggested that the minerals could not pose any mineral load to human body. Hence, the vegetables could be explored as supplement diet for man. The vegetables could provide the needed nutrition health benefit and also help in addressing the problem of food security in Nigeria.

Berberine attenuates brain aging via stabilizing redox homeostasis and inflammation in an accelerated senescence model of Wistar rats.

Aging is a multifaceted and progressive physiological change of the organism categorized by the accumulation of deteriorating processes, which ultimately compromise the biological functions. The objective of this study was to investigate the anti-aging potential of berberine (BBR) in D-galactose (D-Gal) induced aging in rat models. In this study, male Wistar rats were divided into four groups: The control group was given only vehicle, the BBR group was treated with berberine orally, the D-Gal group was treated with D-galactose subcutaneously and the BBR + D-Gal group was treated with D-galactose and berberine simultaneously. D-galactose exposure elevated the pro-oxidants such as malondialdehyde (MDA) level, protein carbonyl and advanced oxidation protein products (AOPP) in the brain. It decreased the anti-oxidants such as reduced glutathione (GSH) and ferric reducing antioxidant potential (FRAP) in the brain. D-galactose treatment also reduced the mitochondrial complexes (I, II, III and IV) activities and elevated the inflammatory markers such as interleukine-6 (IL-6), tumor necrosis factor- α (TNF-α) and C-reactive protein (CRP). The mRNA expressions of IL-6 and TNF-α in the brain were upregulated following D-galactose exposure. Berberine co-treatment in D-galactose induced aging rat model prevented the alteration of pro-oxidant and anti-oxidant in the brain. Berberine treatment restored the mitochondrial complex activities in the brain and also normalized the inflammatory markers. Based on these findings we conclude that berberine treatment has the potential to mitigate brain aging in rats via stabilizing the redox equilibrium and neuroinflammation.

Evaluation of in vitro antioxidant activities, total phenolic and elemental contents of common herbs and spices (Moringa oleifera leaves, Allium sativum (Garlic) and Momordica charantia (ejinrin) leaves) in South-West Nigeria

Abstract Natural remedies are being widely utilized in some disease’s conditions including cancer, diabetes, neurodegenerative diseases, atherosclerosis hypertension and other cardiovascular diseases. The therapeutic intervention of medicinal plants and spices cannot be overemphasized in the management and control of diseases but the exact modes of action of these herbs and plants have not been fully elucidated. This research work was designed to study the expression of selected elements, zinc (Zn), iron (Fe), copper (Cu), selenium (Se), lead (Pb) and cadmium (Cd); the phytochemical and the in vitro antioxidant properties of ethanolic extracts of Moringa oleifera leaves, Allium sativum (Garlic) and Momordica charantia (ejinrin) leaves for their potential involvement in the prevention and management of cardiovascular diseases and cancer through different chemical methods. In vitro antioxidant properties were assayed by investigating 2,2-diphenyl-2-picryl-hydrazyl (DPPH) free radicals scavenging potentials and Ferric Reducing Antioxidant Potential (FRAP); total phenolic content was determined by using Folin-Ciocalteu assay and the elemental contents of these extracts was investigated through the use of Inductively Coupled Plasma Optical Emission Spectroscopy (ICP-OES). All tests were run in triplicates and analysed. Our results revealed that the ethanolic extracts of M. oleifera leaves, A. sativum (Garlic) and M. charantia (ejinrin) leaves possess significant antioxidant activities and express important and beneficial elements (in marginal and trace amounts) and phytochemicals. These results suggest that the medicinal attributes of these plants may be linked to the radical scavenging abilities, beneficial elements and phytochemicals expression of their ethanolic extracts which may likely be a good direction in the area of drug discovery and development.

A water-based ultrasound-assisted extraction system to obtain natural blue colorant from Genipap (Genipa americana L.)

This study aimed to optimize the extraction of a blue colorant using ultrasound-assisted technology employing water as the solvent from genipap, an Amazon fruit. Process conditions were optimized by multiresponse analysis through the antioxidant activity by the ferric reducing antioxidant potential (FRAP) assay, the total phenolic content, and the b* and C* color parameters. The optimized conditions of 70 °C, 325 W, solid: solvent ratio of 1:40 (m/v), and time of 10 min were validated externally at a level of 90 % confidence. The composition of aqueous blue extract obtained under the greatest conditions was evaluated by ultra-performance liquid chromatography (UPLC). Besides, in vitro bioactivities such as digestive enzymatic inhibitions assays, and antiproliferative activity through human cells assays were evaluated. Genipin, geniposide, and other ten iridoids were identified in the aqueous blue extract. The natural colorant obtained presented the capacity of inhibiting enzymes involved in lipid and glucose digestion, suggesting a potential control of obesity and glucose levels in blood without showing cytotoxicity. Therefore, this study presented, for the first time, the optimal operational conditions to obtain a natural blue colorant from genipap in a green approach with interesting biological properties for the food and pharmaceutical industries.

Effects of Green and Gold Kiwifruit Varieties on Antioxidant Neuroprotective Potential in Pigs as a Model for Human Adults.

Kiwifruit (KF) has shown neuroprotective potential in cell-based and rodent models by augmenting the capacity of endogenous antioxidant systems. This study aimed to determine whether KF consumption modulates the antioxidant capacity of plasma and brain tissue in growing pigs. Eighteen male pigs were divided equally into three groups: (1) bread, (2) bread + Actinidia deliciosa cv. 'Hayward' (green-fleshed), and (3) bread + A. chinensis cv. 'Hort16A' (yellow-fleshed). Following consumption of the diets for eight days, plasma and brain tissue (brain stem, corpus striatum, hippocampus, and prefrontal cortex) were collected and measured for biomarkers of antioxidant capacity, enzyme activity, and protein expression assessments. Green KF significantly increased ferric-reducing antioxidant potential (FRAP) in plasma and all brain regions compared with the bread-only diet. Gold KF increased plasma ascorbate concentration and trended towards reducing acetylcholinesterase activity in the brain compared with the bread-only diet. Pearson correlation analysis revealed a significant positive correlation between FRAP in the brain stem, prefrontal cortex, and hippocampus with the total polyphenol concentration of dietary interventions. These findings provide exploratory evidence for the benefits of KF constituents in augmenting the brain's antioxidant capacity that may support neurological homeostasis during oxidative stress.

Exploring the antioxidant, antidiabetic, and antibacterial potential of postbiotic compounds derived from Lactiplantibacillus plantarum O7S1.

Probiotic bacteria are distinguished by their ability to produce various functional postbiotic metabolites. Therefore, this study aimed to explore the in vitro antioxidant, antidiabetic, and antibacterial properties of two postbiotics generated by Lactiplantibacillus plantarum O7S1 (Lpb. plantarum O7S1) during the fermentation process: cell-free supernatant (CFS) and exopolysaccharides (EPS). The antioxidant potential of these postbiotics was assessed using various radical scavenging assays and ferric-reducing antioxidant potential. The antidiabetic activity was evaluated through α-amylase inhibitory assays, while antibacterial activity was determined using agar well diffusion assays. The results of the present study revealed that CFS exhibited significant antioxidant and antidiabetic efficacy in contrast to EPS (P < 0.001). Specifically, CFS displayed remarkable scavenging ability against DPPH, hydroxyl, and superoxide radicals, with inhibition rates of 88.78, 78.91, and 34.85%, respectively, while EPS showed comparatively lower inhibition rates. Additionally, CFS demonstrated higher reducing activity (0.30 optical density units at 700 nm) and potent α-amylase inhibitory activity (95.87%) compared to EPS (67.17%) (P < 0.001). The agar well diffusion assay reported that CFS showed significant antimicrobial activity against both Gram-positive and Gram-negative pathogens, while no activity was observed with EPS. Furthermore, carbohydrate fermentation analysis indicated the strain's ability to metabolize various carbohydrates and their derivatives, potentially enhancing digestive health. These findings suggest that both CFS and EPS exhibit promising hypoglycemic, antioxidant, and antibacterial properties, making them potential candidates for incorporation into functional foods and pharmaceuticals aimed at preventing oxidative damage, diabetes, and pathogenic bacterial infections.

Antioxidant and Antidiabetic Properties of Pectin Extracted from Pomegranate (Punica granatum) Peel

Diabetes mellitus is a chronic disease resulting from defects in insulin secretion, insulin action or both. The prevalence of diabetes has been increasing gradually over the last few decades. It has emerged as a global health concern that burdens public health and socioeconomic development. Oxidative stress plays a crucial role in developing diabetic complications affecting the eyes, kidneys, nerves, and heart. There are various established adverse effects of current conventional antidiabetic drugs. Due to this fact, plants have been chosen for developing new drugs as the bioactive compounds in plants have higher antidiabetic and antioxidant properties with low toxicity. Pomegranate peel is an agro-industrial waste that is rich in pectin. Pectin is a natural polysaccharide that has gained importance due to its pharmacological properties. In this study, pectin from pomegranate peel was extracted using ultrasound-assisted extraction to characterize the pectin and to evaluate its antioxidant and antidiabetic activity. Pomegranate peel pectin (PPP) was extracted using citric acid (solid-to-liquid ratio of 1:30 g/mL) at pH 2 under optimized conditions at 75°C for 40 min. A white sponge-like solid was obtained upon freeze-drying with a yield of 18.17%. The moisture content of pectin was 18.26% and it was found to be a low-methoxylated pectin with a degree of esterification (DE) of 31.07%. Fourier Transform Infrared Spectroscopy (FTIR) results showed that pectin from pomegranate peel was quite similar to commercial pectin. The antioxidant activity was evaluated using scavenging assays, 1,1- diphenyl-2-picrylhydrazyl (DPPH), Ferric Reducing Antioxidant Potential (FRAP) and 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) at 1 mg/mL (78.26% with IC50 value of 326.8 μg/mL, 79.12% with IC50 value of 83.63 μg /mL and 442.7 mg Fe2+/g, respectively). The antidiabetic activity was assessed using an α-amylase inhibition assay at 1 mg/mL (43.82%). These results suggest that PPP has the potential to be used as an antidiabetic and antioxidant agents.

Evaluation of phenolic and antioxidant profiles of pink Guava peel (Psidium guajava L. cv Arka kiran) during fruit ripening and its in silico Anti SARS-CoV-2 property

Guava (Psidium guajava L.) is a highly nutritious and economically important fruit. Although fruit peel is generally regarded as a waste, researchers believe that the peel of the guava is rich in bioactive constituents, even higher than the fruit's flesh. The present study aimed to estimate phenolic content (total phenolic and total flavonoid) and assess antioxidant properties of guava fruit peel (pink variety, cv Arka kiran) by 2,2-di (4-tert-octylphenyl)-1-picryl-hydrazyl (DPPH), 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and Ferric Reducing Antioxidant Potential (FRAP) assays at five different ripening stages (stage 1 to 5). The TPC and TFC assays were performed by Folin-Ciocalteu and aluminium chloride (AlCl3) methods, respectively. The molecular docking experiment between the major phenolic of guava peel, Catechin and the spike protein of SARS-CoV-2 was performed by the Dockthor online server. Results showed that the peel had high phenolic (highest TPC and TFC, 7307.3 mg gallic acid equivalent/g dry weight [DW] and 433.9 mg quercetin equivalent/g DW, respectively) and antioxidant values (highest DPPH, ABTS and FRAP values 4784.8, 206.6 and 2451 mg ascorbic acid equivalent/g DW, respectively) throughout all stages, although there was a gradual decline in the activity at the later stages. Furthermore, it was found that catechin had a strong binding affinity (-7.591 kcal mol-1) with the spike protein, in silico when compared with the control drug ceftazidime (-7.250 kcal mol-1). The overall outcome of our experiemnts revealed that guava peel could be explored for future pharmacological applications through in vivo studies, and the ‘green mixed with the yellow’ stage of ripening is optimum for such studies.

DES-based microwave- and ultrasound-assisted extraction of natural products from Rumex hastatus as per response surface methodology

Rumex hastatus D. Don is a medicinal wild herb that abundantly grows in many parts of the world. The current work explores the extraction of its polyphenolic and antioxidant compounds using glycerol-choline chloride deep eutectic solvent and two modern techniques, ultrasound-assisted extraction (UAE) and microwave-assisted extraction (MAE). The optimization was done as per the response surface methodology. Six responses were studied including total phenolic content (TPC), total flavonoid content (TFC), total condensed tannin content (TCT), ferrous iron-chelating (FIC) activity, DPPH radical scavenging activity (RSA), and ferric reducing antioxidant potential (FRAP). The optimal factors for MAE were 10 s extraction time, 229.40 W power, 40 mL/g SSR (solvent-to-solid ratio) and 60 % DES concentration with the predicted TPC, TFC, TCT, FIC, RSA and FRAP as 64.46 mg GAE/g PM, 155.78 mg RE/g PM, 129.73 mg CE/g PM, 51.46 mg EDTAE/g PM, 235.84 mg AAE/g PM and 129.14 mg AAE/g PM, respectively. For UAE, the predicted optimal factors were 24.30 min sonication time, 47 °C extraction temperature, 40 mL/g SSR and 60 % DES concentration with the predicted TPC, TFC, TCT, FIA, RSA, and FRAP as 56.54 mg GAE/g PM, 148.24 mg RE/g PM, 155.60 mg CE/g PM, 57.83 mg EDTAE/g PM, 240.56 mg AAE/g PM and 111.71 mg AAE/g PM, respectively. Both the models were strongly supported by the validation study with minimal % errors; 2.53–6.38 % for MAE and 1.14–5.30 % for UAE. MAE was more efficient than UAE in terms of time input and recovery of total polyphenols. Therefore, the method based on DES-MAE can safely be recommended for use on an industrial level for obtaining bioactives from R. hastatus.

Stabilization of the Antioxidant Properties in Spray-Dried Microcapsules of Fish and Chia Oil Blends.

Even with healthy foods, there is still a need to protect the functionality during processing. The stabilization and enrichment of fish oil (FO) extracted from fish fillets using solvent extraction might make this healthy oil more available. FO was stabilized by mixing it with chia seed oil (CSO) at 50:50 at room temperature. The antioxidant properties of the blends were evaluated using the total phenolic content (TPC), free radical scavenging activity (DPPH), ferric reducing antioxidant potential (FRAP), and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) activities with FO and CSO as controls. The blends of FO and CSO increased the oxidative stability, while FO was the most susceptible to degradation. The stability and bioactivity of antioxidants against environmental factors were improved by using encapsulation. Response surface methodology (RSM) was used to optimize spray-drying operating conditions for spray-dried microcapsules (SDMs). The independent variables were the inlet air temperature (IAT), which varied from 125 to 185 °C; wall material (WM) concentration, which varied from 5 to 25%; pump speed (PS), which varied from 3 to 7 mL/min; and needle speed (NS), which varied from 3 to 11 s. The results indicated that the maximum antioxidant activity of SDM was obtained at 140 °C IAT, 10% WM, 4 mL/min PS, and 5 s NS, while the minimum value was obtained at 170 °C IAT, 20% WM, 6 mL/min PS, and 9 s NS. The IAT had a significant effect on the antioxidant activities, and the stability of SDMs was increased. These SDMs can be used in the formulation of food matrices due to their therapeutic and nutritional properties.

Sorghum- and Chickpea-Based Ready-to-Eat Extrudate Flakes: Quality Attributes and In Vitro Digestibility

The study is aimed at the development of whole sorghum-based ready-to-eat extruded snacks using chickpea at different barrel temperature (BT) (130, 145, and 170°C) and feed moisture (FM) (14, 16, and 18%) conditions, and quality changes on the functionality aspects of prepared snacks were evaluated. Data revealed that with increment in barrel temperature and feed moisture, the antioxidant properties, viz., ferric reducing antioxidant potential (FRAP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) inhibition, total phenolic content (TPC), and hardness of the flakes amplified; however, elevated barrel temperature and decreased feed moisture enhanced the expansion ratio, water solubility index, and overall acceptability of lakes. The results of the ( L ∗ , a ∗ , and b ∗ ) color values indicated that the rates of Maillard’s reaction were higher when the BT and FM processes were intensified. Enhanced extrusion conditions (BT and FM) lead to higher levels of resistant starch and improved in vitro protein digestibility. The estimated glycemic index of the flakes ranges from 52 to 54, thus showing low estimated glycemic index (E-GI) of flakes. Slowly digestible starch and Exp-GI reveal a positive correlation among themselves. Results from the present study showed that highly acceptable flakes with commercialization properties can be successfully formulated using extrusion processing.
