Berberine attenuates brain aging via stabilizing redox homeostasis and inflammation in an accelerated senescence model of Wistar rats.

Aging is a multifaceted and progressive physiological change of the organism categorized by the accumulation of deteriorating processes, which ultimately compromise the biological functions. The objective of this study was to investigate the anti-aging potential of berberine (BBR) in D-galactose (D-Gal) induced aging in rat models. In this study, male Wistar rats were divided into four groups: The control group was given only vehicle, the BBR group was treated with berberine orally, the D-Gal group was treated with D-galactose subcutaneously and the BBR + D-Gal group was treated with D-galactose and berberine simultaneously. D-galactose exposure elevated the pro-oxidants such as malondialdehyde (MDA) level, protein carbonyl and advanced oxidation protein products (AOPP) in the brain. It decreased the anti-oxidants such as reduced glutathione (GSH) and ferric reducing antioxidant potential (FRAP) in the brain. D-galactose treatment also reduced the mitochondrial complexes (I, II, III and IV) activities and elevated the inflammatory markers such as interleukine-6 (IL-6), tumor necrosis factor- α (TNF-α) and C-reactive protein (CRP). The mRNA expressions of IL-6 and TNF-α in the brain were upregulated following D-galactose exposure. Berberine co-treatment in D-galactose induced aging rat model prevented the alteration of pro-oxidant and anti-oxidant in the brain. Berberine treatment restored the mitochondrial complex activities in the brain and also normalized the inflammatory markers. Based on these findings we conclude that berberine treatment has the potential to mitigate brain aging in rats via stabilizing the redox equilibrium and neuroinflammation.

Evaluation of in vitro antioxidant activities, total phenolic and elemental contents of common herbs and spices (Moringa oleifera leaves, Allium sativum (Garlic) and Momordica charantia (ejinrin) leaves) in South-West Nigeria

Abstract Natural remedies are being widely utilized in some disease’s conditions including cancer, diabetes, neurodegenerative diseases, atherosclerosis hypertension and other cardiovascular diseases. The therapeutic intervention of medicinal plants and spices cannot be overemphasized in the management and control of diseases but the exact modes of action of these herbs and plants have not been fully elucidated. This research work was designed to study the expression of selected elements, zinc (Zn), iron (Fe), copper (Cu), selenium (Se), lead (Pb) and cadmium (Cd); the phytochemical and the in vitro antioxidant properties of ethanolic extracts of Moringa oleifera leaves, Allium sativum (Garlic) and Momordica charantia (ejinrin) leaves for their potential involvement in the prevention and management of cardiovascular diseases and cancer through different chemical methods. In vitro antioxidant properties were assayed by investigating 2,2-diphenyl-2-picryl-hydrazyl (DPPH) free radicals scavenging potentials and Ferric Reducing Antioxidant Potential (FRAP); total phenolic content was determined by using Folin-Ciocalteu assay and the elemental contents of these extracts was investigated through the use of Inductively Coupled Plasma Optical Emission Spectroscopy (ICP-OES). All tests were run in triplicates and analysed. Our results revealed that the ethanolic extracts of M. oleifera leaves, A. sativum (Garlic) and M. charantia (ejinrin) leaves possess significant antioxidant activities and express important and beneficial elements (in marginal and trace amounts) and phytochemicals. These results suggest that the medicinal attributes of these plants may be linked to the radical scavenging abilities, beneficial elements and phytochemicals expression of their ethanolic extracts which may likely be a good direction in the area of drug discovery and development.

A water-based ultrasound-assisted extraction system to obtain natural blue colorant from Genipap (Genipa americana L.)

This study aimed to optimize the extraction of a blue colorant using ultrasound-assisted technology employing water as the solvent from genipap, an Amazon fruit. Process conditions were optimized by multiresponse analysis through the antioxidant activity by the ferric reducing antioxidant potential (FRAP) assay, the total phenolic content, and the b* and C* color parameters. The optimized conditions of 70 °C, 325 W, solid: solvent ratio of 1:40 (m/v), and time of 10 min were validated externally at a level of 90 % confidence. The composition of aqueous blue extract obtained under the greatest conditions was evaluated by ultra-performance liquid chromatography (UPLC). Besides, in vitro bioactivities such as digestive enzymatic inhibitions assays, and antiproliferative activity through human cells assays were evaluated. Genipin, geniposide, and other ten iridoids were identified in the aqueous blue extract. The natural colorant obtained presented the capacity of inhibiting enzymes involved in lipid and glucose digestion, suggesting a potential control of obesity and glucose levels in blood without showing cytotoxicity. Therefore, this study presented, for the first time, the optimal operational conditions to obtain a natural blue colorant from genipap in a green approach with interesting biological properties for the food and pharmaceutical industries.

Effects of Green and Gold Kiwifruit Varieties on Antioxidant Neuroprotective Potential in Pigs as a Model for Human Adults.

Kiwifruit (KF) has shown neuroprotective potential in cell-based and rodent models by augmenting the capacity of endogenous antioxidant systems. This study aimed to determine whether KF consumption modulates the antioxidant capacity of plasma and brain tissue in growing pigs. Eighteen male pigs were divided equally into three groups: (1) bread, (2) bread + Actinidia deliciosa cv. 'Hayward' (green-fleshed), and (3) bread + A. chinensis cv. 'Hort16A' (yellow-fleshed). Following consumption of the diets for eight days, plasma and brain tissue (brain stem, corpus striatum, hippocampus, and prefrontal cortex) were collected and measured for biomarkers of antioxidant capacity, enzyme activity, and protein expression assessments. Green KF significantly increased ferric-reducing antioxidant potential (FRAP) in plasma and all brain regions compared with the bread-only diet. Gold KF increased plasma ascorbate concentration and trended towards reducing acetylcholinesterase activity in the brain compared with the bread-only diet. Pearson correlation analysis revealed a significant positive correlation between FRAP in the brain stem, prefrontal cortex, and hippocampus with the total polyphenol concentration of dietary interventions. These findings provide exploratory evidence for the benefits of KF constituents in augmenting the brain's antioxidant capacity that may support neurological homeostasis during oxidative stress.

Antioxidant and Antidiabetic Properties of Pectin Extracted from Pomegranate (Punica granatum) Peel

Diabetes mellitus is a chronic disease resulting from defects in insulin secretion, insulin action or both. The prevalence of diabetes has been increasing gradually over the last few decades. It has emerged as a global health concern that burdens public health and socioeconomic development. Oxidative stress plays a crucial role in developing diabetic complications affecting the eyes, kidneys, nerves, and heart. There are various established adverse effects of current conventional antidiabetic drugs. Due to this fact, plants have been chosen for developing new drugs as the bioactive compounds in plants have higher antidiabetic and antioxidant properties with low toxicity. Pomegranate peel is an agro-industrial waste that is rich in pectin. Pectin is a natural polysaccharide that has gained importance due to its pharmacological properties. In this study, pectin from pomegranate peel was extracted using ultrasound-assisted extraction to characterize the pectin and to evaluate its antioxidant and antidiabetic activity. Pomegranate peel pectin (PPP) was extracted using citric acid (solid-to-liquid ratio of 1:30 g/mL) at pH 2 under optimized conditions at 75°C for 40 min. A white sponge-like solid was obtained upon freeze-drying with a yield of 18.17%. The moisture content of pectin was 18.26% and it was found to be a low-methoxylated pectin with a degree of esterification (DE) of 31.07%. Fourier Transform Infrared Spectroscopy (FTIR) results showed that pectin from pomegranate peel was quite similar to commercial pectin. The antioxidant activity was evaluated using scavenging assays, 1,1- diphenyl-2-picrylhydrazyl (DPPH), Ferric Reducing Antioxidant Potential (FRAP) and 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) at 1 mg/mL (78.26% with IC50 value of 326.8 μg/mL, 79.12% with IC50 value of 83.63 μg /mL and 442.7 mg Fe2+/g, respectively). The antidiabetic activity was assessed using an α-amylase inhibition assay at 1 mg/mL (43.82%). These results suggest that PPP has the potential to be used as an antidiabetic and antioxidant agents.

Evaluation of phenolic and antioxidant profiles of pink Guava peel (Psidium guajava L. cv Arka kiran) during fruit ripening and its in silico Anti SARS-CoV-2 property

Guava (Psidium guajava L.) is a highly nutritious and economically important fruit. Although fruit peel is generally regarded as a waste, researchers believe that the peel of the guava is rich in bioactive constituents, even higher than the fruit's flesh. The present study aimed to estimate phenolic content (total phenolic and total flavonoid) and assess antioxidant properties of guava fruit peel (pink variety, cv Arka kiran) by 2,2-di (4-tert-octylphenyl)-1-picryl-hydrazyl (DPPH), 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and Ferric Reducing Antioxidant Potential (FRAP) assays at five different ripening stages (stage 1 to 5). The TPC and TFC assays were performed by Folin-Ciocalteu and aluminium chloride (AlCl3) methods, respectively. The molecular docking experiment between the major phenolic of guava peel, Catechin and the spike protein of SARS-CoV-2 was performed by the Dockthor online server. Results showed that the peel had high phenolic (highest TPC and TFC, 7307.3 mg gallic acid equivalent/g dry weight [DW] and 433.9 mg quercetin equivalent/g DW, respectively) and antioxidant values (highest DPPH, ABTS and FRAP values 4784.8, 206.6 and 2451 mg ascorbic acid equivalent/g DW, respectively) throughout all stages, although there was a gradual decline in the activity at the later stages. Furthermore, it was found that catechin had a strong binding affinity (-7.591 kcal mol-1) with the spike protein, in silico when compared with the control drug ceftazidime (-7.250 kcal mol-1). The overall outcome of our experiemnts revealed that guava peel could be explored for future pharmacological applications through in vivo studies, and the ‘green mixed with the yellow’ stage of ripening is optimum for such studies.

DES-based microwave- and ultrasound-assisted extraction of natural products from Rumex hastatus as per response surface methodology

Rumex hastatus D. Don is a medicinal wild herb that abundantly grows in many parts of the world. The current work explores the extraction of its polyphenolic and antioxidant compounds using glycerol-choline chloride deep eutectic solvent and two modern techniques, ultrasound-assisted extraction (UAE) and microwave-assisted extraction (MAE). The optimization was done as per the response surface methodology. Six responses were studied including total phenolic content (TPC), total flavonoid content (TFC), total condensed tannin content (TCT), ferrous iron-chelating (FIC) activity, DPPH radical scavenging activity (RSA), and ferric reducing antioxidant potential (FRAP). The optimal factors for MAE were 10 s extraction time, 229.40 W power, 40 mL/g SSR (solvent-to-solid ratio) and 60 % DES concentration with the predicted TPC, TFC, TCT, FIC, RSA and FRAP as 64.46 mg GAE/g PM, 155.78 mg RE/g PM, 129.73 mg CE/g PM, 51.46 mg EDTAE/g PM, 235.84 mg AAE/g PM and 129.14 mg AAE/g PM, respectively. For UAE, the predicted optimal factors were 24.30 min sonication time, 47 °C extraction temperature, 40 mL/g SSR and 60 % DES concentration with the predicted TPC, TFC, TCT, FIA, RSA, and FRAP as 56.54 mg GAE/g PM, 148.24 mg RE/g PM, 155.60 mg CE/g PM, 57.83 mg EDTAE/g PM, 240.56 mg AAE/g PM and 111.71 mg AAE/g PM, respectively. Both the models were strongly supported by the validation study with minimal % errors; 2.53–6.38 % for MAE and 1.14–5.30 % for UAE. MAE was more efficient than UAE in terms of time input and recovery of total polyphenols. Therefore, the method based on DES-MAE can safely be recommended for use on an industrial level for obtaining bioactives from R. hastatus.

Stabilization of the Antioxidant Properties in Spray-Dried Microcapsules of Fish and Chia Oil Blends.

Even with healthy foods, there is still a need to protect the functionality during processing. The stabilization and enrichment of fish oil (FO) extracted from fish fillets using solvent extraction might make this healthy oil more available. FO was stabilized by mixing it with chia seed oil (CSO) at 50:50 at room temperature. The antioxidant properties of the blends were evaluated using the total phenolic content (TPC), free radical scavenging activity (DPPH), ferric reducing antioxidant potential (FRAP), and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) activities with FO and CSO as controls. The blends of FO and CSO increased the oxidative stability, while FO was the most susceptible to degradation. The stability and bioactivity of antioxidants against environmental factors were improved by using encapsulation. Response surface methodology (RSM) was used to optimize spray-drying operating conditions for spray-dried microcapsules (SDMs). The independent variables were the inlet air temperature (IAT), which varied from 125 to 185 °C; wall material (WM) concentration, which varied from 5 to 25%; pump speed (PS), which varied from 3 to 7 mL/min; and needle speed (NS), which varied from 3 to 11 s. The results indicated that the maximum antioxidant activity of SDM was obtained at 140 °C IAT, 10% WM, 4 mL/min PS, and 5 s NS, while the minimum value was obtained at 170 °C IAT, 20% WM, 6 mL/min PS, and 9 s NS. The IAT had a significant effect on the antioxidant activities, and the stability of SDMs was increased. These SDMs can be used in the formulation of food matrices due to their therapeutic and nutritional properties.

Sorghum- and Chickpea-Based Ready-to-Eat Extrudate Flakes: Quality Attributes and In Vitro Digestibility

The study is aimed at the development of whole sorghum-based ready-to-eat extruded snacks using chickpea at different barrel temperature (BT) (130, 145, and 170°C) and feed moisture (FM) (14, 16, and 18%) conditions, and quality changes on the functionality aspects of prepared snacks were evaluated. Data revealed that with increment in barrel temperature and feed moisture, the antioxidant properties, viz., ferric reducing antioxidant potential (FRAP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) inhibition, total phenolic content (TPC), and hardness of the flakes amplified; however, elevated barrel temperature and decreased feed moisture enhanced the expansion ratio, water solubility index, and overall acceptability of lakes. The results of the ( L ∗ , a ∗ , and b ∗ ) color values indicated that the rates of Maillard’s reaction were higher when the BT and FM processes were intensified. Enhanced extrusion conditions (BT and FM) lead to higher levels of resistant starch and improved in vitro protein digestibility. The estimated glycemic index of the flakes ranges from 52 to 54, thus showing low estimated glycemic index (E-GI) of flakes. Slowly digestible starch and Exp-GI reveal a positive correlation among themselves. Results from the present study showed that highly acceptable flakes with commercialization properties can be successfully formulated using extrusion processing.

Evaluating the in vitro antidiabetic, antibacterial and antioxidant properties of copper(II) Schiff base complexes: An experimental and computational studies

In recent years, metal-based compounds have been explored as potential remedies for majorhealth issues such as diabetes, bacterial infections, and oxidative stress, which impact the general well-being of humans globally. Diabetes, bacterial infections, and oxidative stress are major health concerns affecting millions worldwide. Researchers have been exploring the use of metal-based compounds as potential treatments for these conditions. In our quest towards identifying biologically viable metal-based compounds, three novel Cu(II) complexes were synthesized from halogen-substituted ON donor Schiff base ligands. These ligands were obtained by condensing 2-hydroxybenzldehyde with three different halogen-substituted anilines. The ligands and their complexes were characterized using various spectroscopic techniques such as 1H and 13C nuclear magnetic resonance, Fourier Transform-Infrared, Ultraviolet–visible spectra, elemental analysis, mass spectrometry, and single-crystal X-ray spectroscopy to elucidate their structures. The compounds were screenedfor antidiabetic (α-glucose and α-amylase enzymes inhibition assay), antioxidant (2,2-diphenyl-1-picrylhydrazyl(DPPH), ferric reducing antioxidant potential (FRAP) assay, and nitric oxide(NO) radical scavenging), and antimicrobial activities. The pharmacological activity revealed that the efficacy of the ligands was enhanced upon coordination, and CuL3 is more potent among the complexes, having higher inhibition activity on α-glucosidase (IC50 = 100.1 μM) than the control, whose IC50 is 112.4 μM on the same enzyme. For radical scavenging activity, the results showed that the complexes have higher scavenging activity on DPPH radicals than their ligands and the control. For the FRAP assay, the complexes show higher activity than their ligands. Similarly, NO radical scavenging activity results indicate the ligands have higher activity than their complexes, while the complexes outperformed the control at all concentrations tested. The In vitro antibacterial results on some selected Gram-positive and Gram-negative bacteria revealed the complexes have higher antibacterial activity than their corresponding ligands, with CuL3 taking the lead, showing a MIC value of 4 g/mL on some of the bacteria tested. Also, density functional theory calculations at different levels of DFT and basis sets were used to investigate the optimized geometry, electronic structure (energy of HOMO and LUMO, Mulliken atomic charge, dipole moment, hardness, and softness, electrophilicity, and spectral properties) of the synthesized compounds. Molecular docking studies were carried out to investigate the binding affinity of the receptor proteins to other proteins and to establish the mechanism of action at the active sites.

Infrared Spectroscopy for the Analysis of Bioactive Analytes in Wheat: A Proof-of-Concept Study

This study compared the performance of near-infrared spectroscopy (NIRS) and mid-infrared spectroscopy (MIRS) for the prediction of moisture, protein, total phenolic content (TPC), ferric reducing antioxidant potential (FRAP) and total monomeric anthocyanin (TMA) content in 65 samples of Australian wheat flour. Models were constructed on 50 of the wheat samples, with the 15 remaining samples used as a dependent test set. NIRS showed excellent results for the prediction of protein content (R2test = 0.991; RMSEP = 0.22% w/v) and acceptable to good results for TPC (R2test = 0.83; RMSEP = 3.9 mg GAE/100 g), FRAP (R2test = 0.92; RMSEP = 5.4 mg TE/100 g) and moisture content (R2test = 0.76, RMSEP = 0.62% w/v). Similarly, MIRS showed the best results for protein prediction (R2test = 0.93, RMSEP = 0.62% w/v) and acceptable results for moisture content (R2test = 0.83, RMSEP = 0.65% w/v), FRAP (R2test = 0.83, RMSEP = 7.0 mg TE/100 g) and TPC (R2test = 0.73, RMSEP = 5.6 mg GAE/100 g). However, the TMA content could not be predicted. Finally, moving window analysis was conducted to determine the optimum wavelength ranges for predicting selected analytes. On average, this improved RMSECV values by an average of 18–20% compared to the corresponding full wavelength models, when using the same component selection method. The results confirm that infrared spectroscopy may be useful for the real-time quantitation and/or screening of key quality parameters in wheat, such as protein, TPC and antioxidant capacity.

Evaluation of Total Isoflavones in Chickpea (Cicer arietinum L.) Sprouts Germinated under Precursors (p-Coumaric Acid and L-Phenylalanine) Supplementation.

Cicer arietinum L. (Bengal gram, chickpea) is one of the major pulse crops and an important part of traditional diets in Asia, Africa, and South America. The present study was conducted to determine the changes in total isoflavones during sprouting (0, 3, and 7 days) along with the effect of two precursor supplementations, p-coumaric acid (p-CA) and L-phenylalanine (Phe), in C. arietinum. It was observed that increasing sprouting time up to the seventh day resulted in ≈1282 mg 100 g-1 isoflavones, which is approximately eight times higher than chickpea seeds. The supplementation of Phe did not affect the total length of sprouts, whereas the supplementation of p-CA resulted in stunted sprouts. On the third day of supplementation with p-CA (250 mg L-1), the increase in the total phenolic content (TPC) (80%), daidzein (152%), and genistin (158%) contents were observed, and further extending the supplementation reduced the growth of sprouts. On the seventh day of supplementation with Phe (500 mg L-1), the increase in TPC by 43% and genistin content by 74% was observed compared with non-treated sprouts; however, the total isoflavones content was found to be 1212 mg 100 g-1. The increased TPC was positively correlated with the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity (r = 0.787) and ferric-reducing antioxidant potential (FRAP) (r = 0.676) activity. This study suggests that chickpea sprouts enriched in TPC and antioxidants can be produced by the appropriate quantity of precursor supplementation on a particular day. The results indicated major changes in the phytochemical content, especially daidzein and genistin. It was also concluded that the consumption of 100 g of seventh-day sprouts provided eight times higher amounts of isoflavones in comparison to chickpea seeds.

Effect of fruit maturity on the composition of high-value components and biological activities of olives from newly developed cultivars

The present study evaluates the variations in the composition of targeted high-value components (phenolics, fatty acids, organic acids and natural sugars) and biological activities of olive fruits, collected at different maturity stages, from two locally developed cultivars BARI Zaitoon-1 (BZ1) and BARI Zaitoon-2 (BZ2). Ultrasound-assisted extraction (UAE) was employed to extract phenolic compounds. The oil yield was significantly increased during the maturation, while a decrease in bioactives extract yield was noted. The maximum oil yield (33.8 %) was recovered from BZ1 olives at the fully ripen stage, while the maximum extract yield (34.5 %) was given by BZ2 at the unripen stage. According to HPLC analysis, total phenolic acids, including sinapic, caffeic, gallic, chlorogenic and benzoic acids in BZ1 and BZ2 olives, were higher at semi-ripen and unripen stages, respectively. Myricetin was detected as the major flavonoid in both cultivars, but its concentration gradually decreased during fruit maturation. A good correlation of total phenolic contents (TPC) with 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and total flavonoid contents (TFC) with ferric reducing antioxidant potential (FRAP) was observed. Similarly, an increase in the contents of total sugars and organic acids was noticed corresponding to fruit maturity. Furthermore, the tested olives fruit extracts inhibited the growth of Gram-positive bacterial strains (B. subtilis and S. aureus) more effectively than Gram-negative strains (K. oxytoca, S. typhi, and E. coli). The GC–MS analysis of extracted olive oil revealed that oleic acid was the major fatty acid found at three maturity stages for both cultivars. The current research revealed considerable variations in the selected components profile and biological activities of the tested olive cultivars as a function of fruit maturity. Based on these findings, the uses of olives can be recommended for specific applications to maximize the functional food and nutraceutical benefits of this valuable fruit.

Phenolic Profile and Antioxidant Potential of Aqueous Extracts of Selected Traditional Anti-cough Plants

The recent surge in attention towards plant-based antioxidants can be attributed to their perceived advantageous impact on human health. The present study assessed the phenolic profile, antioxidant potential, and free radical scavenging ability of hot aqueous leaf extracts obtained from Vernonia amygdalina, Piper guineense, and Gongronema latifolium. High-Performance Liquid Chromatography (HPLC) was used to examine phenolic profile. Antioxidant capacity was conducted using total flavonoid content (TFC), total phenol content (TPC), total antioxidant capacity (TAC), polyphenol oxidase (PPO) activity, while ferric reducing antioxidant potential (FRAP), nitric oxide (NO), hydroxyl (OH), and 2, 2-diphenyl-2-picrylhydrazyl (DPPH) were analysed for their radicals scavenging activities. G. latifolium displayed notable phenolic compounds, such as lunamarin (76.32mg/ml), ribalinidine (64.08 mg/ml), gallocatechin (48.58mg/ml), aglycone (41.69 mg/ml), flavonone (33.45mg/ml), and flavan-3-ol (30.49mg/ml). The plant extracts contained resveratol, kaempferol, and ribalinidine in a consistent manner across all samples. The findings suggest that the plants exhibits noteworthy antioxidant potential, with G. latifolium displaying the highest antioxidant capacity (96.18 ± 0.11µg/mL) and total phenolic content (28.50 ± 0.04 µg/mL). V. amgydalina showed highest polyphenol oxidase activity, at 7.14 ± 0.10 µg/mL G. latifolium as well showed high •NO scavenging radicals and FRAP activity, while exhibiting the least efficacy in •OH scavenging radicals. The results indicate that all three samples demonstrated a noteworthy increase in DPPH scavenging activity, which was statistically significant at p<0.05 compared to standard. Results reveals that plant extracts showed significant phenolic compounds, antioxidant and free radicals property. These observations provides a rationale for the traditional use of these extracts in folk medicine for management and treatment of cough.

Comparative analysis of physicochemical characteristics, bioactive components, and volatile profile of sour cherry (Prunus cerasus)

This study provides a detailed report on the physicochemical, bioactive components, and volatile profiles of diverse sour cherry ( Prunus cerasus) cultivars to identify the cultivar(s) containing high health-promoting components. Physiological characteristics (fruit weight, size dimensions, moisture, color attributes, total soluble solids, pH, titratable acidity, maturity index, nutritional bioactive components (total phenolic, total anthocyanin content, and total flavonoids), antioxidant activity, and volatile profile of 10 sour-cherry cultivars, consisting of dark red Morello type and clear fruit flesh Amarelle type, were studied. The total phenolic content was in the range of 123.24–289.91 mg gallic acid equivalent/100 g FW (fresh weight), total flavonoids (1340.23–2831.91 mg quercetin equivalent/100 g FW), and total anthocyanins (225.43–485.66 mg cyanidin-3-glucoside equivalent (CGE)/100 g FW) in different sour-cherry cultivars, showing significant diversity in such health-promoting compounds. In vitro antioxidant activity assessed by ferric reducing antioxidant potential was observed in the range of 658.18–1483.37 mg Trolox equivalent (TE)/100 g FW and by 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) at 384.36 and 931.85 mg TE/100 g FW. A total of 10 phenolic components, including five hydroxycinnamic acids, three flavonoids (flavanols), and one anthocyanin, have been identified and quantified by high-performance liquid chromatography. Hydroxycinnamic acids represented 40%–60% of total phenolic components, while flavonoids and anthocyanins amounted to 20% each in total phenolic composition. The volatile profile of sour-cherry cultivars revealed that aldehydes, alcohols, ketones, esters, monoterpenes, acids, sugars, and hydrocarbons were the predominant volatiles present in sour cherry.

Curcumin displays a potent caloric restriction mimetic effect in an accelerated senescent model of rat.

Curcumin, a strong natural compound with numerous health benefits, is extracted from the Curcuma longa. According to recent research findings, it also acts as a calorie restriction mimetic. We examined established aging biomarkers in erythrocytes and plasma and tested a persistent oral dietary dose of curcumin in young and D-galactose-induced accelerated rat aging models. For four weeks, D-gal (300mg/kg b.w. subcutaneously) and curcumin (200mg/kg b.w. oral) were administered simultaneously to test the protective effects of curcumin against D-galactose-induced accelerated aging and oxidative stress. In the accelerated senescent rat model, we discovered a significant rise in protein carbonyl, malonaldehyde (MDA), and advanced oxidation protein products. Increased levels of catalase, superoxide dismutase, ferric-reducing antioxidant potential, and reduced glutathione (GSH) were observed. Our findings reveal that curcumin has characteristics resembling a calorie restriction mimic and can successfully maintain redox equilibrium throughout the aging process in rat erythrocytes and plasma.

In Vitro Potent Anticancer, Antifungal, and Antioxidant Efficacy of Walnut (Juglans regia L.) Genotypes

Walnuts are one of the healthiest foods in the world because they are one of the best sources of beneficial nutrients, minerals, and antioxidants. They also contain key sources of energy. Despite several traditional uses, the leaves of Juglans regia L. have received little attention regarding phytochemical and pharmacological potential. Thus, the current study intended to find the walnut genotypes with the greatest antioxidant, anticancer, and antifungal activity. The total polyphenolic, flavonoid, and flavanol contents of leaves from 14 walnut genotypes were determined. Genotypes that accumulate flavonoid/flavonol contents (99.8–111.93 mg/g quercetin equivalent (QE) and 101.67–111.83 mg/g QE) showed significantly higher ferric reducing antioxidant potential (FRAP) activity (128.2–148.1 μM Fe2+/g dry weight (DW)] than other genotypes. Maximum divergence in the quercetin content (0.8–1.23 mg/g) of walnut genotypes was obtained by cluster analysis. The active component, quercetin, was measured using RP-HPLC. Moreover, the extracts were investigated for antifungal and anticancer assays. We report the significant antifungal potential of walnut leaf genotypes against Candida glabrata, Candida albicans, and Candida tropicalis, with 57.7–93.6%, 26.8–51.5%, and 26.8–51.5% inhibition, respectively. The most significant antiproliferative effect was shown by Opex Culchry, which exhibited 9.4% cell viability at a concentration of 25 µL (0.75 mg) against lung (A549) cell lines. Chenovo exhibited 2.9, 6.2, and 2.2% cell viability, Opex Culchry exhibited 2, 1.5, and 2.4% cell viability, and Sulieman showed 7.6, 0.9, and 7% cell viability against the colon (HCT116) cell lines. The results showed that walnut leaves possess enormous potential as antioxidants, and as anticancer and antifungal agents.

Evaluation of Antioxidant Potential of Asystasia Chelonoides var.chelonoides Nees.(Acanthaceae)

The ethanolic leaf extract of Asystasia chelonoides var. chelonoides Nees. was evaluated using various in vitro and ex vivo assays. Evaluations were made for DPPH radical scavenging activity, NO radical scavenging activity, Super oxide radical scavenging activity ,Ferric Reducing Antioxidant Potential (FRAP) assay, Total antioxidant activity , Reducing Power activity and Lipid Peroxidation inhibition. Over all findings revealed that A.chelonoides leaf exhibited four extracts namely (AC HEX, AC CHL, AC ETH and AC CRD) and were selected for the study, AC ETH and AC CRD showed potent antioxidant activity by inhibiting DPPH radical in a concentration dependent manner . The nitric oxide radical scavenging activity of all the samples increased as their concentration increased from 20 µg/mL to 320 µg/ mL. AC CRD showed its maximum superoxide radical scavenging of 78.56 % at 320 µg/mL and the EC50 was found to be 93.26 µg/mL. The extracts exhibited a dose dependent reducing power activity within the applied concentrations (20 µg/mL to 100 µg/mL) and AC CRD at 100 µg/mL concentration showed highest activity, which was almost comparable to standard ascorbic acid. In FRAP assay the crude extract of A. chelonoides (AC CRD ) showed 138.14 µg trolox equivalent/g of dry extract respectively. Total antioxidant activity of AC CRD, AC HEX, AC CHL and AC ETH were found to be 98.27, 14.86, 20.16 and 72.56 µg of ascorbic acid equivalent/mg of dry extract respectively. A.chelonoides crude extract and ethanolic fraction showed potent inhibition of FeCl2AA stimulated rat liver lipid peroxidation in concentration dependent manner.

The Antioxidant Activity and Induction of Apoptotic Cell Death by Musa paradisiaca and Trigona sp. Honey Jelly in ORL115 and ORL188 Cells.

Head and neck cancer patients usually need nutritional support due to difficulties in swallowing and chewing. Therefore, this study aimed to formulate Musa paradisiaca and Trigona sp. honey jelly (MTJ) as a convenient functional food. The antioxidant properties were analysed using 2,2'-diphenyl-1 picrylhydrazyl (DPPH), ferric reducing antioxidant potential (FRAP) and 2,2'-azinodi 3-ethylbenthiazolinesulfonate (ABTS) assays. Cytotoxicity was assayed using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) test and the induction of apoptosis was observed via caspase-3/7 activity assay. The identification of phenolic compounds was done via ultra-high-performance-liquid chromatography coupled to mass spectrometer (UHPLC-MS/MS). The antioxidant analysis exhibited: the half inhibitory concentration (IC50) of DPPH inhibition, 54.10 (SD = 4.51) μg/mL; the FRAP value, 30.07 (SD = 0.93) mM TEQ/100 g; and the ABTS value, 131.79 (SD = 8.73) mg TEQ/100 g. Cinnamic acid was the most abundant phenolic compound, followed by maleic acid and salicylic acid. The IC50 for ORL115 and ORL188 were 35.51 mg/mL and 43.54 mg/mL, respectively. The cells became rounded and dissymmetrical which reduced in number and size. The apoptotic cell death in ORL115 and ORL188 was deduced as caspase-3/7 activities that significantly increased (P < 0.05). The study evidenced that the antioxidant activity of MTJ could influence the induction of apoptosis in ORL115 and ORL188 in future investigations and verifications.

Assessment of phytochemical screening and antioxidant potential of Heteropogon contortus (L.) whole plant

Heteropogon contortus (L.) belongs to the family Poaceae, which is known to have bioactivities like anti-inflammatory, antioxidant, membrane and mast cell stabilization, antimicrobial and many more. The aim of the present study is to explore occurrence of phytochemicals and antioxidant activity by using the whole plant (including leaf, stem, and root) crude extract in three different solvents (methanol, hydroethanol, and aqueous). Using standard methods, presence of various bioactive compounds was determined. Quantitative evaluation of total phenol, flavonoid and tannin content were done by using the spectrophotometric techniques. For antioxidant property analysis, in-vitro techniques like ferric reducing antioxidant potential (FRAP) assay, diphenyl1-2-picrylhydrazyl (DPPH) and metal chelation assay were used. In plant extract preparation, the green colour extract of methanol with solid consistency observed to have highest % yield of 4.46 %, followed by hydroethanolic (3.86 %), and aqueous extract (2.95 %). Various phytocompounds (e.g., alkaloids, steroids, phenols, flavonoids, glycosides, carbohydrates, and resins) were observed in higher concentration in methanol, hydroethanol, and aqueous extract. Out of all extracts, the methanol extract observed to have highest flavonoid (30.07 ± 0.09 mg Quercetin equivalent (QE)/g), phenol (34.12 ± 0.028 mg of the Gallic acid equivalent (GAE)/g), and tannin (26.61 ± 0.008 mg of the Tannic acid equivalent (TAE)/g) content. In DPPH and metal chelation assay, the effective inhibitor was the methanol extract (42.94 ± 0.0061 µg/ml, 100.57 ± 0.085 µg/ml) in order of potency inhibitory concentration (IC50) while the FRAP value was observed maximum in the hydroethanol extract (258.41 ± 0.0085 mg/g). Thus, overall investigation of the plant extracts shows that the occurrence of variety of phytochemicals having antioxidant properties makes the plant a promising candidate for usage in the traditional medicine.
