Effect of ethanol extract of Ganoderma lucidum from Tasikmalaya against the growth of Salmonella sp bacteria and some Pathogenic Fungi using the Kirby-Bauer method in vitro

<p><em>Ganoderma lucidum (lingzhi) can be used as traditional medicine. This mushroom has many properties, one of which is as an antimicrobial. Antimicrobial functions as antibacterial, antiviral, and antifungal. Lingzhi contains organic alkaloid compounds, flavonoids, and triterpenoids. One of the bacteria that often cause disease in Indonesia is Salmonella sp and several dermatophyte diseases caused by infection with several pathogenic fungi such as Malassezia furfur, Trichophyton rubrum, and Trichophyton mentagrophytes . This study aimed to determine the inhibitory power of Lingzhi from Tasikmalaya ethanol extract against Pathogenic Microorganism. The method used is tested against the anti-microbial used by using the diffusion method using disc paper or Kirby Baure. The analysis showed that the 70% ethanol extract of Lingzhi mushrooms had antibacterial activity against salmonella sp as evidenced by the presence of an average inhibition zone at a concentration of 50% (16,57mm), 60% (18,1mm); 70% (18,55mm), 80% (19.17mm), 90% (21,2mm), and 100% (21,8mm). </em><em>Inhibitory ability against several pathogenic fungi was able to inhibit at a concentration of 100% with a diameter of Trichophyton rubrum</em><em>, </em><em>Trichophyton mentagrophytes</em><em> </em><em>and </em><em>Malassezia furfur</em><em> </em><em>with an average diameter of inhibition of 12.9 mm, 9.4 mm, and 6 mm</em><em>.</em></p>

Assessment of Cosmetic Properties and Safety of Use of Model Washing Gels with Reishi, Maitake and Lion's Mane Extracts.

Natural cosmetics are becoming more and more popular every day. For this reason, this work investigates the properties of mushroom extracts, which are not as widely used in the cosmetics industry as plant ingredients. Water extracts of Grifolafrondosa (Maitake), Hericiumerinaceus (Lion’s Mane) and Ganoderma lucidum (Reishi) were tested for their antioxidant properties, bioactive substances content, skin cell toxicity, ability to limit TEWL, effect on skin hydration and pH, and skin irritation. Our research showed that Maitake extract contained the highest amount of flavonoids and phenols, and also showed the most effective scavenging of DPPH and ABTS radicals as well as Chelation of Fe2+ and FRAP radicals, which were 39.84% and 82.12% in a concentration of 1000 µg/mL, respectively. All tested extracts did not increase the amount of ROS in fibroblasts and keratinocytes. The addition of mushroom extracts to washing gels reduced the irritating effect on skin, and reduced the intracellular production of free radicals, compared with the cosmetic base. Moreover, it was shown that the analyzedcosmetics had a positive effect on the pH and hydration of the skin, and reduced TEWL.

PRODUCTION OF GANODERMA LUCIDUM EXTRACT LOADED GELATIN-SODIUM ALGINATE MICROSPHERES, INVESTIGATION OF RELEASE KINETICS AT DIFFERENT pH VALUES AND EVALUATION OF KINETIC MODELS

In this study, pH sensitive microsphere polymeric drug carriers were produced by using biodegradable natural gelatin and sodium alginate polymers. Microspheres were loaded with prepared Ganoderma lucidum extract that is a medicinal mushroom and has the potential to be used in several diseases’ treatment. Extract release kinetics of the microspheres were examined by spectrophotometric method by using an UV spectrometer. Buffer solutions with different pH values were used as release medium for examination of drug release kinetics of the produced microspheres. The Ganoderma lucidum release of microspheres was presented in terms of percent cumulative release (CR%) defined as the percentage ratio of the instantaneous amount of Ganoderma lucidum released at a certain time of incubation to the initial amount of Ganoderma lucidum loadings. As a result, it was seen that the release of the extract accelerated as pH of the release medium increased and the fastest extract release was observed in the pH 7. The release kinetic models of the microspheres were examined. The release kinetics of microspheres fitted Higuchi model for pH 1.3, pH 5.0 and pH 6.0 and first-order model for pH 3.0 and pH 7.0.

Production Ganoderma lucidum extract nanoparticles by expansion of supercritical fluid solution and evaluation of the antioxidant ability

Due to the growing human tendency to treat with natural substances, fungi such as Ganoderma lucidum can be a good source to meet this need. Effectiveness, ease of use and a rich source of active ingredients such as ganoderic acids have caused G. lucidum to be considered in the pharmaceutical and food industries. In this project, G. lucidum was applied to extraction using supercritical carbon dioxide. Then expansion of supercritical fluid solution (ESS) was used as, novel, repeatable and green method to yield nanoparticles from G.lucidum extract. The response surface method was used to improve the Extraction efficiency, antioxidant activity, and improving the nanoparticles production status. Optimal conditions were observed at the extraction step by setting pressure at 27.5 MPa, dynamic time of 46 min, and modifier volume of 162 μL. The optimum point for the production of nanoparticles was obtained as follows: pressure drop at 25 MPa, 20 min for collection time, and 40° C for temperature. Under these conditions, the size and count were 86.13 nm, and 98, respectively. Nanoparticles were analyzed by FESM and, the DPPH was used for antioxidant activity evaluation. The LC–MS identified various ganoderic acids from G.lucidum that are famous to be highly oxygenated triterpenoids.

Protective Effects of Amauroderma rugosum on Doxorubicin-Induced Cardiotoxicity through Suppressing Oxidative Stress, Mitochondrial Dysfunction, Apoptosis, and Activating Akt/mTOR and Nrf2/HO-1 Signaling Pathways.

Clinical outcomes for doxorubicin (Dox) are limited by its cardiotoxicity but a combination of Dox and agents with cardioprotective activities is an effective strategy to improve its therapeutic outcome. Natural products provide abundant resources to search for novel cardioprotective agents. Ganoderma lucidum (GL) is the most well-known edible mushroom within the Ganodermataceae family. It is commonly used in traditional Chinese medicine or as a healthcare product. Amauroderma rugosum (AR) is another genus of mushroom from the Ganodermataceae family, but its pharmacological activity and medicinal value have rarely been reported. In the present study, the cardioprotective effects of the AR water extract against Dox-induced cardiotoxicity were studied in vitro and in vivo. Results showed that both the AR and GL extracts could potentiate the anticancer effect of Dox. The AR extract significantly decreased the oxidative stress, mitochondrial dysfunction, and apoptosis seen in Dox-treated H9c2 rat cardiomyocytes. However, knockdown of Nrf2 by siRNA abolished the protective effects of AR in these cells. In addition, Dox upregulated the expression of proapoptotic proteins and downregulated the Akt/mTOR and Nrf2/HO-1 signaling pathways, and these effects could be reversed by the AR extract. Consistently, the AR extract significantly prolonged survival time, reversed weight loss, and reduced cardiac dysfunction in Dox-treated mice. In addition, oxidative stress and apoptosis were suppressed, while Nrf2 and HO-1 expressions were elevated in the heart tissues of Dox-treated mice after treatment with the AR extract. However, the GL extract had less cardioprotective effect against Dox in both the cell and animal models. In conclusion, the AR water extract demonstrated a remarkable cardioprotective effect against Dox-induced cardiotoxicity. One of the possible mechanisms for this effect was the upregulation of the mTOR/Akt and Nrf2/HO-1-dependent pathways, which may reduce oxidative stress, mitochondrial dysfunction, and cardiomyocyte apoptosis. These findings suggested that AR may be beneficial for the heart, especially in patients receiving Dox-based chemotherapy.

GC/MS Analysis and In vitro Effect of Ganoderma lucidum Solvent Extracts on Microorganisms Isolated from the Armpit, Scalp and Urinary Tract

Aims: The study was to assess the antibacterial effect of solvent extracts of Ganoderma lucidum on isolates from the human scalp, armpit, and urinary tract.
 Study Design: Descriptive comparative.
 Place and Duration of Study: the Bells University of Technology, between December 2019 and October 2020.
 Methodology: Test organisms isolated from the scalp, armpit, and urine were identified using conventional methods and Analytical Profile Index kits (API). Ganoderma lucidum was rinsed with 2.5% potassium hydroxide and sterile distilled water, dried at 50℃ for 72 h, and pulverized. The mushroom powder (10%) was extracted in methanol, acetone, and petroleum ether at 65oC for 48 h. The filtrate was evaporated and two-fold serial dilutions were prepared in Dimethyl sulfoxide (DMSO). Antibacterial activity was done by the agar well diffusion method. Inoculum adjusted to 0.5 McFarland standard was inoculated onto Mueller Hinton agar. Wells of 6mm were filled with 100ml of the extracts. Antibiotic sensitivity was by disc diffusion method. Plates were incubated at 37oC for 24 h. Gas chromatography-mass spectrophotometer (GC-MC) analysis of methanol extract was carried out.
 Results: The isolates were identified as follows; scalp (Corynebacterium kutscheri and Enterobacter intermedius), armpit (Acinetobacter baumannii), and urine (Pseudomonas aeruginosa). The solvents exhibited varying degrees of inhibition on test organisms. The methanol extract was most inhibitory on all organisms. Petroleum ether showed the least inhibition. P. aeruginosa was most resistant to the extracts. Fluoroquinolones and aminoglycosides inhibited all the organisms and recorded a higher zone of inhibition, unlike the beta-lactams. GC/MS of methanol extract revealed the presence of 48 compounds amongst them were sugar, alcohols, nitrates, alkaloids, amines, fatty acids, methyl esters, and steroids. Also found was Phenol, 2-methoxy butylated hydroxyl anisole (BHA) an antioxidant.
 Conclusion: Ganoderma lucidum contained bioactive compounds that are antimicrobial and showed utility for use in the medical-pharmaceutical industry.

Synergistic in vitro anticancer actions of decorated selenium nanoparticles with fucoidan/Reishi extract against colorectal adenocarcinoma cells

Abstract Cancer-fighting needs more effective and targeted drugs, desirably with least side-effects and from natural bases. The assessment and synergistic actions of multiple anticancer drug candidates were investigated. Fucoidan (Fu) was extracted from Sargassum cinereum, which was attained from the Saudi Red-Sea coast. The extract of Ganoderma lucidum or Reishi mushroom (Re) was achieved, intermixed with Fu solution, and they were used for reducing/decorating/capping selenium nanoparticles (SeNPs). The infrared analysis of produced/synthesized agents emphasized their biochemical structures and interactions. The decorated Fu/Re/SeNPs had negative (−30.6 mV) zeta potential and the SeNPs mean particle size was 6.5 nm. The transmission imaging of SeNPs indicated their spherical profiles and well-dispersion within Fu/Re composite. The anticancer potentiality of Fu/Re and Fu/Re/SeNPs against human colorectal adenocarcinoma cells was investigated using MTT, 4′,6-diamidino-2-phenylindole staining and comet assay. Both composites exhibited potent bioactivities toward adenocarcinoma cells; the reduced cells survival was detected with IC50 of 57.82 and 16.43 µg·mL−1 for Fu/Re and Fu/Re/SeNPs, respectively. The constrained apoptosis was notably observed from treated cells with the composites; substantial molecular damages were also verified via the comet assay, especially in Fu/Re/SeNPs treated cells. The innovative innocuous composite from Fu/Re/SeNPs is highly recommended to deactivate/destruct colorectal adenocarcinoma cells.

Natural triterpenoids of Ganoderma lucidum as new, green, and effective corrosion inhibitor for steel in acidic medium: characterization, experimental and theoretical investigations

Ganoderma lucidum, a wood-decaying fungus, has been at the center of pharmaceutical research for its disease curative and preventive abilities due to its abundant antioxidant-rich phytoconstituents. Herein, we have explored its outstanding inhibition properties in the protection of steel from environmental degradation. Its triterpenoid constituents were extracted, characterized by liquid chromatography/multi-stage mass spectrometry, and applied in both experiments (electrochemical, gravimetric, 3 D optical profiling, SEM, and FTIR tests) and computation (quantum chemical calculations and molecular dynamics simulation) to reveal its metallic corrosion suppression efficacy. 36 constituent triterpenoids were effectively characterized from the extract. In comparison with corresponding electrochemical tests conducted with the crude extract of Ganoderma lucidum, an inhibition efficiency of 83.1% at 1.0 g/L was recorded for the triterpenoid extract against 93.9% for 1.0 g/L crude extract, and both extracts displayed mixed-typed corrosion inhibition properties, exhibiting improved performances with increasing concentrations and exposure time while their effects were gradually decreased with temperature increments. The potential presence of the triterpenoids was confirmed on the carbon steel surface by the surface analytical techniques, and the experimental data were in full compliance with the Langmuir adsorption isotherm model. Computational methods estimated the adsorption performances of some selected triterpenoids on Fe (110) surface. An exclusive study of the corrosion inhibition influence of triterpenoids for metal protection in an acid environment is hereby presented for the first time, and the reported results will benefit future explorations of triterpenoids in inhibitors design and synthesis.

The use of antibiotics is one of the most important ways to deal with the spread and treatment of pathogenic microbial infections. The search for new antibiotic sources continues to be carried out to anticipate the emergence of microbial resistance. One of the natural resources that has the potential as an antimicrobial source is a member of the macrofungi of the Genus Ganoderma. The purpose of this study was to evaluate the antimicrobial performance of the ethanol extracts of Ganoderma lucidum, G. applanatum dan Ganorderma sp. against fungi (Candida albicans dan Cryptococcus neoformans), gram positive bacteria (Bacillus cereus dan Staphylococcus aureus) and gram negative bacteria (Escherichia coli dan Shigella sp.). Macrofungi samples were taken from the forest area of Nature Tourism Park (TWA) Gunung Tunak, TWA Kerandangan, TWA Suranadi, TWA Nuraksa Sesaot, TWA Lemor and Pusuk forest. The stages of the method performed are sample collection, sample preparation, extraction, and testing of antimicrobial activity using the well difusion method. The ethanol extract concentrations for testing were 20%, 40%, 60% and 80%. The results showed that the three Ganoderma species had anti-fungal and antibacterial activity and that different levels of concentration had an effect on inhibition. The size of the inhibition zone is directly proportional to the higher the extract concentration. The antimicrobial activity of the ethanol extract of G. lucidum was higher when compared to G. applanatum and Ganoderma sp. both against fungi (Candida albicans and Cryptococcus neoformans) as well as against gram-positive and gram-negative test bacteria. In addition, G. applanatum showed very weak inhibition against both groups of tested bacteria. Key words: candidiasis; cryptococcosis; comorbid infections; macerations; pathogens

Ganoderma Lucidum Extract Regulates Gut Morphology and Microbial Community in Lipopolysaccharide-Challenged Broilers

This study was conducted to investigate the effect of Ganoderma lucidum extract (GLE) on the gut morphology and cecal microbial community of broilers challenged with lipopolysaccharide (LPS). 144 one-day-old unsexed broiler chicks were randomly distributed into four treatments: non-challenged broilers fed a basal diet; LPS-challenged broilers fed a basal diet; LPS challenged broilers fed a basal diet supplemented with 1 mL/L of GLE in the drinking water; and LPS challenged broilers fed a basal diet supplemented with 1.33 mL/L of GLE in the drinking water. Results showed that supplementationwith1.33 mL/L of GLE alleviated intestinal inflammatory gene expression in LPS-challenged broilers (p≤0.05). Supplementation of GLE (1 and 1.33 mL/L) increased the villus height in the jejunum and ileum of LPS-challenged broilers (p≤0.001). Weighted principal coordinate analysis, heat map of species abundance, and microbial function pathway revealed distinct separation between the groups treated with LPS only and LPS in combination with GLE supplementation (1 and 1.33 mL/L). The abundance of the genus Faecalibacterium was increased in the cecal digesta of LPS-challenged broilers receiving GLE(1 and 1.33 mL/L) compared with the LPS challenge-only group (p≤0.001). The growth performance parameter of broilers was positively associated with the abundance of the genus Faecalibacterium in the cecal digesta. In conclusion, GLE supplementation could modulate gut morphology and cecal microbiota composition of broilers under inflammatory challenge.

Ganoderma lucidum polysaccharide peptide alleviates hyperuricemia by regulating adenosine deaminase and urate transporters.

Hyperuricemia (HUA) affects human health and is involved in the pathogenesis of common chronic diseases. Previous studies showed that Ganoderma lucidum extract lowered HUA in animals. However, the active ingredient and pharmacological mechanism of Ganoderma lucidum extract in the improvement of HUA are unknown. The purpose of this study was to determine the anti-HUA efficacy and related mechanism of Ganoderma lucidum polysaccharide peptide (GLPP) using a potassium oxonate (PO)-induced mouse model and an adenosine-induced cell model. The experimental results showed that blood uric acid (UA) was decreased up to 40.6% by GLPP in HUA mice in a dose-dependent manner. Additionally, GLPP significantly reduced UA production by inhibiting the hepatic and blood adenosine deaminase (ADA) activity and increased UA excretion by decreasing the expression of glucose transporter 9 (GLUT9) and increasing the expression of organic anion transporter 1 (OAT1) in kidney. The adenosine-induced cell model showed that the inhibitory effect of GLPP on ADA activity may be the main reason for the alleviation of HUA by GLPP. Furthermore, PO-induced renal histopathological damage was also alleviated by GLPP in a dose-dependent manner. The experimental results in this study indicated that GLPP exerted anti-HUA effects via regulating the UA production and excretion, suggesting that GLPP could be developed into a therapeutic agent for HUA.

Inhibitory effects of Thai herbal extracts on the cytochrome P450 3A-mediated the metabolism of gefitinib, lapatinib and sorafenib

Herbal products are widely used in cancer patients via co−administration with chemotherapy. Previous studies have demonstrated that pharmacokinetic interactions between herbs and anticancer drugs exist due to inhibition of drug−metabolizing enzymes, particularly cytochrome P450s (CYPs). The aim of this study was to determine the inhibitory effects of Andrographis paniculata, Curcuma zedoaria, Ganoderma lucidum, Murdannia loriformis and Ventilago denticulata extracts on the metabolism of gefitinib, lapatinib and sorafenib. The activities of CYP3A in human liver microsome on the metabolism of gefitinib, lapatinib and sorafenib in the absence and presence of Thai herbal extracts were assayed using high-performance liquid chromatography analysis. Curcuma zedoaria extract potently inhibited CYP3A−mediated lapatinib and sorafenib metabolism with IC50 values of 4.18 ± 3.20 and 7.59 ± 1.23 μg/mL, respectively, while the metabolism of gefitinib was strongly inhibited by Murdannia loriformis and Ventilago denticulata extracts with IC50 values of 7.53 ± 2.87 and 7.06 ± 1.23 μg/mL, respectively. Andrographis paniculata and Ganoderma lucidum extracts had less effect on the metabolism of the tested anticancers (IC50 values >10 μg/mL). In addition, kinetic analysis of the ability of Curcuma zedoaria extract to inhibit CYP3A-mediated metabolism of anticancer drugs was best described by the noncompetitive and competitive inhibition models with Ki values of 20.08 and 11.55 μg/mL for the metabolism of gefitinib and sorafenib, respectively. The present study demonstrated that there were potential pharmacokinetic interactions between tyrosine kinase inhibitors and herbal extracts.

Immunomodulatory Effects of Extract of Lingzhi or Reishi Medicinal Mushroom Ganoderma lucidum (Agaricomycetes) Basidiocarps Cultivated on Alternative Substrate.

Ganoderma lucidum is a medicinal mushroom exhibiting numerous health benefits primarily based on strong immunostimulatory effects. The study aimed to investigate if there were differences in effects of extracts of commercially (GC) and alternatively (wheat straw) (GA) cultivated G. lucidum basidiocarps on properties of peritoneal macrophages (PM) and monocyte-derived dendritic cells (MoDCs). Differences in immunomodulatory effects of GC/GA extracts were studied. The viability of treated PMs, their adhesive and phagocytic capability, and their capacity to produce reactive oxygen species (ROS) and NO were tested. Immature MoDCs generated from human monocytes were treated with poly I:C (10.0 μg/ml) and loxoribine (34.0 μg/ml), a selective TLR3 and TLR7 agonists, respectively, and with/without GC/GA extract (100.0 μg/ml). The effect of each combination on phenotypic properties, cytokines production by MoDCs, and their proliferation and Th polarizing capacity was studied. GA extract stimulated the metabolic and phagocytic activity of PMs, their adhesion capability, and ability to produce ROS and NO more strongly compared to GC. Both tested extracts significantly increased allostimulatory and Th1 polarization capacity of simultaneous TLR3 and TLR7-activated MoDCs, but GA extract was more effective. The extract of alternatively cultivated G. lucidum basidiocarps increased production of ROS and NO by TLR4 stimulated PMs and upregulated production of certain cytokines as well as allostimulatory and Th1 polarization capacity of MoDCs. GA extract could be a potent immunostimulatory agent for activation of MoDCs with the simultaneous engagement of TLRs, which seems to be a promising strategy for the preparation of DC-based anti-tumor vaccines.

Aqueous Extracts of Medicinal Mushrooms Ganoderma lucidum and Pleurotus eryngii (Agaricomycetes) Can Affect Seed Germination and Seedling Survival.

Rapid and complete seed germination results in the rapid initial growth of plants. In this study, aqueous extracts (boiling, maceration + filtration, and maceration + autoclave) of Ganoderma lucidum and Pleurotus eryngii were used to improve seed germination of three crops (maize, clover, and alfalfa). The results revealed that boiled water extractions of G. lucidum positively affected seed germination of clover and alfalfa. Moreover, P. eryngii boiling extract could increase the vigor index II in alfalfa. The results achieved are comparable with those gained by phytohormones and in some cases were even better. From these findings, it can be concluded that the existing metabolites in aqueous extracts of G. lucidum and P. eryngii can be used to enhance seed germination effectively and even increase the leaf area of crop plants.

Mycosynthesis of Zinc Oxide Nanoparticles Coated with Silver using Ganoderma lucidum (Curtis) P. Karst and Its Evaluation of In Vitro Antidiabetic and Anticancer Potential

Nanotechnology is an evolving interdisciplinary field of research interspersing material science and nanobiotechnology. Nanoparticles are studied extensively for their specific catalytic, magnetic, electronic, optical, antimicrobial, theranostic, diagnosis, wound healing, and anti‐inflammatory properties. ZnO nanoparticles (NPs) have many applications owing to their unique characteristics, which include low cost, nontoxicity, abundance in nature, and the ability to prepare compounds with varying morphologies having different properties. The main aim of the study is to biosynthesis of ZnO nanoparticles coated with silver from the aqueous extract of Ganoderma lucidum (Curtis) P. Karst and to evaluate its antidiabetic potential by performing alpha‐glucosidase inhibition and alpha‐amylase inhibition assays and to evaluate the anticancer potential by cytotoxicity (MTT) assay against human breast cancer MDA‐MB 231 cell lines. The biosynthesis of ZnO nanoparticles coated with Ag was characterized by UV–vis spectroscopy, Fourier transform infrared spectroscopy, energy dispersive X‐ray analysis, scanning electron microscope, and transmission electron microscopy. An increasing concentration in the biosynthesized ZnO nanoparticles coated with Ag produces strong antidiabetic activity through enzyme inhibition effect and anticancer activity through the reduction of cell viability. The present study recommended that the “Biological” method of biological nanoparticle production is a promising approach that allows synthesis in aqueous conditions, with low energy requirements and low costs. In the future, the mycosynthesized nanoparticles might be used in the medical arena to treat and prevent diseases.

Self-nanoemulsifying composition containing curcumin, quercetin, Ganoderma lucidum extract powder and probiotics for effective treatment of type 2 diabetes mellitus in streptozotocin induced rats

Liquid self-nanoemulsifying drug delivery system (L-SNEDDS) of curcumin and quercetin were prepared by dissolving them in isotropic mixture of Labrafil M1944CS®, Capmul MCM®, Tween-80® and Transcutol P®. The prepared L-SNEDDS were solidified using Ganoderma lucidum extract, probiotics and Aerosil-200® using spray drying. These were further converted into pellets using extrusion-spheronization. The mean droplet size and zeta potential of L-SNEDDS were found to be 63.46 ± 2.12 nm and − 14.8 ± 3.11 mV while for solid SNEDDS pellets, these were 72.46 ± 2.16 nm and −38.7 ± 1.34 mV, respectively. The dissolution rate for curcumin and quercetin each was enhanced by 4.5 folds while permeability was enhanced by 5.28 folds (curcumin) and 3.35 folds (quercetin) when loaded into SNEDDS pellets. The Cmax for curcumin and quercetin containing SNEDDS pellets was found 532.34 ± 5.64 ng/mL and 4280 ± 65.67 ng/mL, respectively. This was 17.55 and 3.48 folds higher as compared to their naïve forms. About 50.23– and 5.57-folds increase in bioavailability was observed for curcumin and quercetin respectively, upon loading into SNEDDS pellets. SNEDDS pellets were found stable at accelerated storage conditions. The developed formulation was able to normalize the levels of blood glucose, lipids, antioxidant biomarkers, and tissue architecture of pancreas and liver in streptozotocin induced diabetic rats as compared to their naïve forms.

Advancement of Microwave-Assisted Biosynthesis for Preparing Au Nanoparticles Using Ganoderma lucidum Extract and Evaluation of Their Catalytic Reduction of 4-Nitrophenol.

This study describes the biosynthesis of gold nanoparticles (AuNPs) using the extract of Ganoderma lucidum in the buffer zone of Bach Ma National Park, Vietnam, as a reducing and protecting agent using microwave-assisted synthesis. The as-synthesized AuNPs were characterized using transmission electron microscopy, scanning electron microscopy, X-ray diffraction, energy-dispersive X-ray spectroscopy, and Fourier transform infrared spectroscopy. Compared to the conventional method, the proposed microwave-assisted method produced AuNPs having a small size of 22.07 ± 8.11 nm in a short synthesis time period. In excess NaBH4, the as-prepared AuNPs demonstrated good catalytic activity for reducing 4-nitrophenol to 4-aminophenol. Furthermore, AuNPs demonstrated improved reusability after four cycles. The pseudo-first-order apparent rate constant was estimated to be 0.086 min–1 at 303 K. Both the catalytic mechanism and reaction path of reduction were proposed. Moreover, activation energy and thermodynamic parameters, including activation enthalpy and entropy, were examined.

Ganoderic Acid A Attenuates IL-1β-Induced Inflammation in Human Nucleus Pulposus Cells Through Inhibiting the NF-κB Pathway.

Intervertebral disc (IVD) degeneration is a major cause of low back pain associated with several pathological changes in the IVD, including dysfunction of nucleus pulposus (NP) cells. Ganoderic Acid A (GAA), one of triterpenoid extracts of Ganoderma lucidum (G. lucidum), has been reported to possess anti-inflammatory effect. In the current study, we aimed to evaluate the effect of Ganoderic Acid A (GAA) on the interleukin-1β (IL-1β)-induced inflammation in human NP cells. Our results showed that the IL-1β-stimulated production of inflammatory mediators including nitric oxide (NO), prostaglandin E2 (PGE2), inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 were suppressed by GAA. In addition, treatment of NP cells with GAA significantly inhibited the production of inflammatory cytokines tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in IL-1β-stimulated human NP cells. GAA improved the reduced expression levels of extracellular matrix (ECM) proteins, collagen II and aggrecan in IL-1β-stimulated human NP cells. GAA also alleviated IL-1β-induced the levels of matrix metalloproteinase (MMP)-3 and MMP-13. Furthermore, GAA inhibited the IL-1β-induced upregulation of the phosphorylation of p65 and downregulation of IκBα. Taken together, these findings indicated that GAA alleviated IL-1β-induced inflammation and ECM degradation in NP cells through regulating NF-κB pathway.
