Optimization of extraction of antioxidants from aromatic herbs and their synergistic effects in a lipid model system

Response surface methodology was applied to improve the polyphenol extraction process of rosemary, oregano, sage, and thyme. Aqueous ethanol (EtOH 50%) rendered the highest polyphenol extraction yield for all tested samples. Based on their total phenolic contents, rosemary, oregano, and thyme were selected for evaluation of their scavenging activities towards DPPH radical and ABTS radical cation and application in an oil model system. All extracts decreased the production of primary oxidation compounds during Schaall oven test storage. The induction period, as evaluated by the Rancimat test, was also reduced. There was an agreement between both oil model system assays, and rosemary extract showed the highest antioxidant capacity, followed by thyme and oregano. A centroid simplex design was used to evaluate the synergistic effect among the samples. Rosemary was able to play a synergistic effect when combined with thyme and oregano, or when used in binary mixtures.

Acetone effects on Buddleja scordioides polyphenol extraction process and assessment of their cellular antioxidant capacity and anti-inflammatory activity

The use of water, acetone, and hydroacetonic solutions as an extraction solvent for polyphenol compounds from Buddleja scordioides has been investigated. The phenolic profiles determined by liquid chromatography—electrospray ionization—mass spectrometry/mass spectrometry (LC–ESI–MS/MS) were compared with those obtained with the traditional aqueous infusion used for the treatment of gastrointestinal disorders. It was evaluated the effects of polyphenol extract at physiologically relevant conditions against H2O2-induced oxidative stress via attenuation of reactive oxygen species (ROS) levels and lipopolysaccharide (LPS)-induced inflammation in human HT-29 cells. The use of hydroacetonic solutions allows obtaining phytochemical extracts enriched with constituents that have antioxidant and anti-inflammatory activity in intestinal cells. This suggests that polyphenols of low and middle polarity from Buddleja scordioides may be better extracted with hydroacetonic solutions, and showing high nutraceutical potential to reduce oxidative stress associated with the onset and progress of inflammatory diseases.

Extraction Process of Polyphenols from Soybean (Glycine max L.) Sprouts: Optimization and Evaluation of Antioxidant Activity

This research aimed to optimize the total polyphenol content (TPC) extracted from soybean sprout powder under different experimental parameters, including ethanol concentration (60–100% v/v), extraction temperature (40–80 °C), extraction time (15–150 min), material:solvent ratio (1:4–1:10 g/mL), the number extraction cycles (1, 2 and 3 times), the age of sprout (0–7 days), and the used part of the sprout (cotyledon, hypocotyl, or radicle). The obtained results were used in response surface methodology, in combination with a central composite design, to model the total polyphenol content (TPC) with respect to three variables, including ethanol concentration, extraction temperature, and material:solvent ratio. The experimental conditions for optimal recovery of TPC consisted of ethanol concentration of 88% (v/v), extraction temperature of 59 °C, material:solvent ratio of 1:6.5 g/mL, extraction time of 60 min, and 2 cycles of maceration. In addition, for maximal TPC, the sprout should undergo the germination of 5 days and the radicle fraction should be used. Based on the suggested optimum conditions, the obtained and verified TPC was 19.801 mg genistein (GE)/g dry weight (d.w.). The obtained dried extract also exhibited low antioxidant activity.

Optimization and Kinetics of the Solid-liquid Extraction Process of Polyphenols from Black Mulberry Fruit

This paper investigates the influence of operating conditions and extraction techniques (maceration and ultrasonic) on the yield and extraction of dry extract, total phenolic, flavonoids and anthocyanins from black mulberry fruit (Morus nigra L.). The optimal extraction conditions and kinetic parameters of the extraction process are determined.

Extraction of polyphenols from olive pomace: Mathematical modeling and technological feasibility in a high temperature and high pressure stirred reactor

A procedure to evaluate the technological feasibility at pilot scale of the extraction process of polyphenols from olive pomace is presented in this work. The proposed approach takes into consideration the extended kinetic route coupled with mathematical simulation. Detailed physically-based dynamic mathematical models, taking into account mass and energy balance equations, are adopted to describe both the lab-scale and the pilot-scale reactors. Chemical physical parameters appearing in the models are estimated from the experimental data at lab-scale or are partially taken from literature. Different heating systems are designed for the pilot scale reactor and their performance is tested by simulation. Characteristic times are evaluated also during start-ups and different control loops are analyzed in order to set-up the best process and operating variables. Average yields in polyphenols are finally evaluated for both the batch and the continuous operated pilot reactor, by considering feed variability and fluctuations of process parameters.

Pressurized liquid extraction of polyphenols from Goldenberry: Influence on antioxidant activity and chemical composition

This work investigates the extraction process of polyphenols (phenolic acids and flavonoids) from Goldenberry using pressurized ethanol. The influence of pressure (100 and 200bar), flow rate (1–3mLmin−1) and time (0–60min) on the antioxidant capacity (DPPH) and chemical composition were evaluated. Results indicated that the highest antioxidant potentials were obtained at 10min and ranged from 34.01% to 76.33%. Gallic acid and ellagic acid were extracted at 100bar and 200bar, respectively, regardless of the flow. On the other hand, caffeic acid, rutin and mangiferin were extracted in all conditions, regardless of the flow and pressure applied. In general, high antioxidant capacities were modulated by specific experimental conditions and the value may not be related only to content, but probably due to the presence of some compounds with strong power of radical scavenging (DPPH).

Optimization of the extraction process of polyphenols from Thymus serpyllum L. herb using maceration, heat- and ultrasound-assisted techniques

Abstract Despite the fact that Thymus serpyllum is well-known medicinal plant and its chemical profile and biological activity have been investigated, there is no detailed study regarding the influence of different techniques and conditions on the extraction of polyphenolic compounds from Serpylli herba. The aim of this study was optimization of the extraction parameters that improves the efficiency of polyphenols extraction from T. serpyllum: particle size, solid-to-solvent ratio, solvent type and extraction time, by using maceration, heat- and ultrasound-assisted extraction (HAE and UAE). The extraction efficiency was expressed via total polyphenol content (TPC) and total flavonoid content (TFC). The statistical analysis (one-way ANOVA and full factorial design) has revealed that the optimal conditions for achieving the best polyphenols yield were particle size of 0.3 mm, 1:30 solid-to-solvent ratio and 50% ethanol, as environmentally friendly extraction medium, while extraction time has not shown statistically significant influence on polyphenols concentration, in all procedures. Under these conditions, the measured TPC was 26.6 mg GAE/L in maceration, 29.8 mg GAE/L in HAE and 32.7 mg GAE/L in UAE, which was in agreement with the predicted values, while TFC was 14.3 mg CE/L, 12.4 mg CE/L and 16.7 mg CE/L for maceration, HAE and UAE, respectively. According to total polyphenols yield, the efficiency of the extraction methods for all variables was ranked by significance in the following order: UAE > HAE > maceration, whereas total flavonoids yield was the highest in UAE, although there was no statistically significant difference between maceration and HAE. According to our results, UAE could be selected as the most successful and suitable technique for extraction of bioactive polyphenolic compounds from Serpylli herba. Using LC/MS and HPLC analysis, 9 polyphenolic compounds were identified and quantified: 6,8-Di-C-glucosylapigenin, chlorogenic acid, 6-hydroxyluteolin 7-O-glucoside, caffeic acid, luteolin 7-O-glucuronide, apigenin glucuronide, salvianolic acid K isomer, rosmarinic acid and salvianolic acid I. This study was an initial step in production of polyphenols-rich wild thyme extracts aimed to be used for formulation of foodstuffs and medicines.

Optimization of the extraction process of polyphenols from cashew apple agro-industrial residues

The goal of this study was to determine the chemical composition of cashew apples agro-industrial residue and optimize the process of polyphenols extraction in this residue. The extraction process conditions were defined using a 24-1 fractional factorial experimental design using acetone and methanol as solvents. The independent variables were: time (30 to 90 min), temperature (30 to 50 °C), solvent concentrations (50% to 90%), agitation speed (100 to 300 rpm); the dependent variables were: total phenolic content and DPPH scavenging capacity. The optimized process was carried out by applying the Central Composite Rotational Design (CCRD) considering the results obtained with the 24-1 fractional factorial experimental design. The residue presented bioactive compounds in its composition, with emphasis on the content of total phenolic compounds (1975.64 mg/ 100 g). The extraction process was not affected by methanol; however, acetone affected the amounts of extracted phytochemicals. Extracts with high levels of polyphenols and strong DPPH scavenging capacity (> 80%) were obtained using 55% acetone, 30 minutes, 30 °C, and 150 rpm. The results showed that cashew apple residue is a potential natural source of bioactive compounds with strong antioxidant capacity. These compounds could be used partially or totally to replace synthetic antioxidants.

Green extraction process of tannins obtained from Moroccan Acacia mollissima barks by microwave: Modeling and optimization of the process using the response surface methodology RSM

The effect of extraction conditions on polyphenols contents and condensed tannins by microwave-assisted extraction (MAE) was studied for the first time to our knowledge. Moroccan barks of Acacia mollissima was used to extract phenolic compounds. The variables studied are the following: power extraction, time extraction and solvent nature. Five powers extraction were tested: 150W, 250W, 300W, 450W and 600W. A significant effect of power extraction on the extractable nature was proved by ANOVA and Student test. The yields were also affected by time extraction. Different solvent (water, ethanol, methanol and ethyl acetate) were tested to evaluate the best extraction solvent according to the extractable nature. Highest polyphenols contents were obtained with methanol. The proportion of this solvent, time extraction and power extraction were optimized using the response surface methodology (RSM). A face-centered composite design (FCCD) was applied to evaluate the effects of these variables on the polyphenols and condensed tannins contents. For each experiment, the extraction yield, the total polyphenolic contents and the condensed tannins contents were quantified using colorimetric essays. The extracts were characterized by their reactivity to formaldehyde and reverse phase high pressure liquid chromatography (RP-HPLC). The highest polyphenols content was obtained at 156W using 80% of methanol during 5min. For condensed tannins, the highest content of cyanidin was obtained at 182W using 20% of methanol during 3.66min. RSM applied in MAE, permitted to develop green extraction process of polyphenols and tannins extracted, using lower microwave power and methanol proportion with a shortest time extraction and in the same time improve the quantity of extractables obtained from renewable natural resource.

Garlic (Allium sativum L.) husk waste as a potential source of phenolic compounds: Influence of extracting solvents on its antimicrobial and antioxidant properties

This paper reports the extraction process of polyphenols from the industrially disposed garlic husk (GH) using different studied solvents (water, methanol, ethanol and 50% aqueous solutions of methanol and ethanol). The highest extraction yield was achieved with water (26.5%) and high bioactive potential was shown by the samples extracted with 50/50 methanol/water (25mg GAEs/g dry GH, 0.617mg QE/g dry GH, IC50=0.26mg/mL for DPPH assay, RP0.5AU=2.8mg/mL for reducing power, and IC50=0.45mg/mL for hydroxyl radical assay). All the extracts exhibited antimicrobial activity against Gram-positive bacteria, when applied at different concentrations (1–10mg/mL). Only 50/50 methanol/water and absolute methanol extracts inhibited the growth of Pseudomonas aeruginosa and Klebsiella pneumonia. These interesting biological properties could be attributed to the major identified phenolic compounds, such as caffeic, p-coumaric, ferulic and di-ferulic acids. With this procedure, the GH waste polyphenols could be used as a cheap source of natural compounds, with potential applications in the food and health sectors.

Extraction process optimization of polyphenols from Indian Citrus sinensis – as novel antiglycative agents in the management of diabetes mellitus

BackgroundDiabetes mellitus is a chronic metabolic disorder characterized by increased blood glucose level. It has become an epidemic disease in the 21st century where, India leads the world with largest number of diabetic subjects. Non-enzymatic glycosylation (glycation) is severe form of diabetes, occurs between reducing sugar and proteins which results in the formation of advanced glycation end products (AGEs) that leads to the other complicated secondary disorders. In this context, Mangifera indica (Mango), Syzygium cumini (Jambul), Vitis vinifera (Grapes), Citrus sinensis (Orange), Artocarpus heterophyllus (Jackfruit), Manilkara zapota (Sapodilla) seeds were evaluated for their antiglyation activity. Attempts were made to isolate the polyphenols in the seeds that have recorded the maximum activity.MethodsDifferent extraction methods (shake flask, centrifugation and pressurized hot water) using various extractants (organic solvents, hot water and pressurized hot water) were adopted to investigate the in vitro antiglycation activity. Central composite (CCD) design based Response Surface Methodology (RSM) was espoused to optimize the extraction process of polyphenols from the fruit seeds that have recorded poor antiglycation activity. The PTLC analysis was performed to isolate the polyphenols (Flavonoids and phenolic acids) and LC-PDA-MS analysis was done for structure prediction.ResultsPressurized hot water extraction of Artocarpus heterophyllus (87.52%) and Citrus sinensis seeds (74.79%) was found to possess high and low antiglycation activity, respectively. The RSM mediated optimization process adopted for the Citrus sinensis seeds have revealed that 1:15 solvent ratio (hexane to heptane), 6 minutes and 1:20 solid to liquid ratio as the optimal conditions for the extraction of polyphenols with a maximum antiglycation activity (89.79%). The LC-PDA-MS analysis of preparative thin layer chromatography (PTLC) eluates of Artocarpus heterophyllus seed has showed the presence of compounds like quercetin (301.2), 4-hydroxy phenyl acetic acid (149.0), rhamnosyl-di-hexosyl quercetin sulphate (857.6), quercetin-3-O-xyloside (428.2), rutin (613.4), diosmetin (298.1) and luteolin (283.0).ConclusionThe Artocarpus heterophyllus was observed to possess a significant antiglycation activity and the activity of Citrus sinensis was improved after the optimization process, which proved that both the seeds may be used as a traditional medicine in the management of chronic diabetes mellitus.

Optimization of Phenolic Antioxidant Extraction from Wuweizi (Schisandra chinensis) Pulp Using Random-Centroid Optimazation Methodology

The extraction optimization and composition analysis of polyphenols in the fresh pulp of Wuweizi (Schisandra chinensis) have been investigated in this study. The extraction process of polyphenols from Wuweizi pulp was optimized using Random-Centroid Optimization (RCO) methodology. Six factors including liquid and solid ratio, ethanol concentration, pH, temperature, heating time and extraction times, and three extraction targets of polyphenol content, antioxidant activity and extract yield were considered in the RCO program. Three sets of optimum proposed factor values were obtained corresponding to three extraction targets respectively. The set of optimum proposed factor values for polyphenol extraction given was chosen in further experiments as following: liquid and solid ratio (v/w) 8, ethanol 67.3% (v/v), initial pH 1.75, temperature 55 °C for 4 h and extraction repeated for 4 times. The Wuweizi polyphenol extract (WPE) was obtained with a yield of 16.37 mg/g and composition of polyphenols 1.847 mg/g, anthocyanins 0.179 mg/g, sugar 9.573 mg/g and protein 0.327 mg/g. The WPE demonstrated high scavenging activities against DPPH radicals.

Kinetics on the Extraction of Polyphenol from Infructescence of <i>Platycarya strobilacea sieb. et zucc</i>

In this paper, 60% ethanol was used as solvent to extract polyphenol from infructescence ofPlatycarya Strobilacea Sieb.et Zucc(IPSSZ), and the mass transfer kinetics of polyphenol at different temperature was investigated. According to Fick’s second law of diffusion, the kinetic equation forIPSSZ.extraction process was established with plate model, and the parameters, such asK(rate constant),Ea(activation energy) were gained. The results can provide the valuable theory basis for the technical design and further research of total polyphenol extraction process.

Temperature and solvent effects on polyphenol extraction process from chestnut tree wood

Polyphenols are a group of chemical substances found in plants, characterized by the presence of more than one phenol unit or building block per molecule. Polyphenols are generally divided into hydrolysable tannins (gallic acid esters of glucose and other sugars) and phenylpropanoids, such as lignins, flavonoids, and condensed tannins. They are widely found in natural products, nowadays they are extensively used in food and beverage industry and in pharmaceutical and nutraceutical industry for their positive effects on human health. In conventional processes polyphenols are extracted from vegetable material using water as solvent in a temperature range from 40 to 90 °C. The aim of this work is to characterize chestnut tree wood in terms of total extractable polyphenols and assess the temperature and solvent type effects on the polyphenol extraction process. To this end, experimental data on equilibrium distribution of polyphenols between solid and liquid phases for different temperatures (ranging from 60 to 80 °C) and different solvent solutions (water and ethanol–water solutions) are reported. The obtained results have been correlated by means of Freundlich isotherm. Experimental data are also reported about the batch extraction kinetics of polyphenols from solid phase and analysed by a mathematical model to estimate the polyphenol diffusion coefficient inside the chestnut wood particles and the mass transfer coefficient in the liquid phase.

Optimum extraction process of polyphenols from Bridelia grandis stem bark using experimental design

Euphorbiaceae barks are known to contain an appreciable amount of polyphenolic compounds responsible for several biological activities. Preliminary extraction from Bridelia grandis stem bark afforded high content of polyphenols, determined by spectrophotometric methods such as Folin-Ciocalteu (for total phenols, TP) and n-butanol-HCl (for condensed tannins, CT). A preliminary Plackett-Burman screening design was used to identify the key factors that influence the TP and CT extraction. Between all the variables known to influence the extraction from vegetable matrixes, six were selected; maceration was chosen as traditional extraction methodology. To investigate the effect of solvents and extraction method, methanol, acetone 70% (v/v in water), centrifugation and ultrasound were chosen. A full factorial design 2(3) was applied to optimize the extraction procedure. The responses were obtained analyzing the extracts for their TP and CT contents determined by the above-mentioned spectrophotometric methods. The results confirm that, within the explored domain, the optimum solvent is methanol and the optimum method is one-cycle centrifugation. Finally, it was also compared with the effect of maceration on the considered responses. It has never given results better than centrifugation, whereas in the case of CT it represents an advantage to employ a three-cycle centrifugation instead of one.

Enhancing polyphenol extraction from unripe apples by carbohydrate-hydrolyzing enzymes

The effects of process variables such as enzyme types, enzyme ratio, reaction temperature, pH, time, and ethanol concentration on the extraction of unripe apple polyphenol were investigated. The results indicated that Viscozyme L had the strongest effect on polyphenols extraction and was selected to study the polyphenol composition. The ratio of enzyme (Viscozyme L) to substrate (2 fungal beta-glucanase units (FBG)) at 0.02, reaction at pH 3.7, 50 degrees C for 12 h, and ethanol concentration of 70% were chosen as the most favorable extraction condition. Total phenolic content (TPC), reducing sugar content (RSC), and extraction yield increased by about 3, 1.5, and 2 times, respectively, compared with control. The contents of p-coumaric acid, ferulic acid, and caffeic acid increased to 8, 4, and 32 times, respectively. The enzyme-aided polyphenol extraction process from unripe apples might be applied to food industry for enhancing bioactive compound production.

Optimum extraction Process of polyphenols from the bark of Phyllanthus emblica L. based on the response surface methodology

Phyllanthus emblica L. is an economic plant used in Chinese medicine for the treatment of various diseases. The bark of P. emblica is rich in polyphenols and its extractions have shown strong antioxidative and radical scavenging activity. Response surface methodology (RSM) was used to assess the optimal extraction of polyphenols from P. emblica bark. Various extraction parameters including ethanol concentration, extraction time, temperature, solid-liquid ratio, and extraction times were chosen to identify their effects on polyphenols extraction. Among these parameters, extraction times and solvent concentration were found to have significant effect on polyphenols extraction. RSM was applied to obtain the optimal combination of solvent concentration, extraction time, temperature, and extraction time for maximum rate of extraction. The most suitable condition for the extraction of polyphenols was at ethanol concentration 75%, extraction time 25 min, extraction temperature 45 degrees C, and extraction times 3. At these optimal extraction parameters, the maximum extraction of polyphenols obtained experimentally was found to be very close to its predicted value. The extraction rate of polyphenols was 19.78% at the optimum conditions. The mathematical model developed was found to fit with the experimental data of polyphenols extraction.
