Corticosteroids regulate brain hippocampal 5-HT1A receptor mRNA expression

Using in situ hybridization techniques, the expression of 5-HT1A receptor mRNA was measured within the hippocampal formation after bilateral adrenalectomy (ADX). After 24 hr ADX, 5-HT1A receptor mRNA expression was significantly increased in all hippocampal subfields in ADX animals relative to sham-operated controls (SHAM). The magnitude of the increase was most pronounced within CA2 (127%) and CA3/4 (94%)-subfields of dorsal hippocampus, intermediate in the dentate gyrus (73%), and least within CA1 (60%). Administration of exogenous corticosterone (CORT) at the time of ADX maintained the level of 5-HT1A receptor mRNA expression within the range of SHAM animals. In vitro receptor autoradiographic analysis of 5-HT1A receptors in adjacent sections from the same animals indicated a simultaneous increase in 5-HT1A binding throughout the hippocampus in response to ADX. 5-HT1A binding increased to a similar extent (approximately 30%) in CA subfields and dentate gyrus but remained within SHAM levels in CORT-replaced animals. 5-HT1A receptor mRNA levels were also increased in hippocampal subregions of 1 week ADX animals relative to SHAM animals. Within both CA1 and CA2 subfields, the increments were approximately double those observed after 1 d ADX. 5-HT1A receptor binding was increased in every hippocampal subfield to a similar extent as that observed after 1 d ADX. Increases in both 5-HT1A receptor mRNA expression and 5-HT1A receptor binding were preventable by administration of exogenous CORT at the time of ADX. Hippocampal 5-HT1C receptor mRNA and D1 receptor mRNA expression were not significantly altered by either acute or chronic ADX treatment. These data indicate that adrenal steroids may selectively regulate hippocampal 5-HT1A receptors at the level of 5-HT1A receptor mRNA expression.

Stress-induced modulation of the primary cellular immune response to herpes simplex virus infection is mediated by both adrenal-dependent and independent mechanisms

A murine model of herpes simplex virus (HSV) infection was used to examine the role of the adrenal gland in restraint stress-induced suppression of viral immunity. Adrenal-dependent mechanisms were important for suppressing the generation of HSV-specific cytotoxic T lymphocytes (CTL) but not the associated diminished lymphadenopathy in response to local HSV infection. While exogenous corticosterone administration alone was unable to suppress lymphadenopathy and CTL generation in adrenalectomized mice, an adrenal-independent mechanism induced by restraint stress functioned in synergy with corticosterone to suppress lymphadenopathy and CTL development. These results suggest that both adrenal-dependent and independent mechanisms contribute to stress-induced modulation of HSV immunity.

Changes in brain dopamine and acetylcholine release during and following stress are independent of the pituitary-adrenocortical axis

Microdialysis was employed to assess extracellular dopamine from medial prefrontal cortex, nucleus accumbens, nucleus caudatus, and acetylcholine from the hippocampus of conscious rats during and after 120 min restraint stress. Restraint stress rapidly stimulated the release and the metabolism of dopamine in the medial prefrontal cortex and in the nucleus accumbens, and acetylcholine release in the hippocampus. Fifty-sixty min later, although rats were still restrained, dopamine and acetylcholine release gradually returned to basal levels. When the animals were freed a considerable increase in the release of both neurotransmitters was observed. No changes in the striatum were observed throughout the experiments. The time-course of plasma corticosterone did not parallel that of dopamine and acetylcholine release, increasing during the whole stress procedure, and decreasing when the animals were released. Adrenalectomized rats responded to stress and liberation in much the same way as intact rats. The administration of exogenous corticosterone (0.5-1.5 mg/kg s.c.) did not change the release of dopamine from the prefrontal cortex and nucleus accumbens, and of acetylcholine from the hippocampus, while the dose of 3.0 mg/kg which stimulated them, raised plasma corticosterone to very high concentrations which had never been attained during stress. Moreover, RU 38486, an antagonist of brain glucocorticoid receptors, did not antagonize the stress-induced increase of neurotransmitter release.(ABSTRACT TRUNCATED AT 250 WORDS)

Glucocorticoid receptor regulation: The effects of adrenalectomy, exogenous glucocorticoid, and stress on hepatic receptor number in male and female mice

Although glucocorticoids are known to regulate their own receptor number, the physiologic significance of this process is not known. In order to assess this process in intact animals the effects of adrenalectomy, stress, and exogenous glucocorticoid on the number of hepatic glucocorticoid receptors in Swiss-Webster mice were evaluated. In males 24 hr after adrenalectomy there was a clear 2- to 2.5-fold increase (upregulation) in glucocorticoid receptor number. Conversely, 24 hr after the ip administration of exogenous corticosterone there was a clear downregulation of receptor number. In each case (upregulation and downregulation) female mice were much less responsive than males. Three stressors were used to evaluate the effect of the endogenous secretion of glucocorticoids on downregulation. Male mice were exposed to ether, vibration, and confinement either once or daily for periods up to 3 days. Animals were sacrificed 24 hr after the last stress and hepatic receptor number was compared to an unstressed control. Cytosolic receptor number was not influenced by any of these stimuli. It is concluded that although glucocorticoids clearly regulate glucocorticoid receptor number, as demonstrated by adrenalectomy and the administration of steroids to adrenalectomized animals, the physiologic significance of this process is uncertain as receptor number does not appear to be changed by stimuli of adrenal glucocorticoids in the intact animal.

Adrenal Glucocorticoids Regulate Adipsin Gene Expression in Genetically Obese Mice

Adipsin expression at the protein and mRNA levels is greatly reduced in several distinct syndromes of obesity in the mouse: genetic obesity due to the db/db and ob/ob genes, and a chemically induced model secondary to neonatal exposure to monosodium glutamate. We considered first the possibility that the adipsin gene might be identical to the db or ob locus and the lowered expression of this protein might result from a mutation in this gene. We show here that the adipsin structural gene is located on chromosome 10 and hence is physically distinct from any obesity genes so far identified in the mouse. A major role for the adrenal gland and adrenal glucocorticoids in the aberrant regulation of adipsin in these models of obesity is indicated by several experiments. Adrenalectomy of the ob/ob mouse raises the circulating levels of adipsin protein and the amount of this mRNA in epididymal fat pads (5-fold), although neither is increased to the levels seen in lean controls. Exogenous administration of corticosterone completely blocks the effects of adrenalectomy on adipsin, suggesting that the effect of this endocrine ablation is through reduction of adrenal glucocorticoids. Corticosterone administration also causes suppression in the levels of adipsin mRNA and protein in lean mice, although this decrease is never as severe as that seen in obese mice. The effect of exogenous corticosterone in lean mice occurs within 2 days and hence is not secondary to the obesity which these hormones eventually elicit. These results indicate that glucocorticoids can regulate adipsin expression in vivo and strongly suggest that the hyperglucocorticoid state seen in certain obese models plays a significant role in lowering adipsin mRNA and protein levels. Quantitative analysis of these experiments suggests that other as yet unknown neuroendocrine factors also function to suppress adipsin in obesity.

Circulating corticosterone in the infant rat: the mechanism of age and thyroxine effects.

Infant rats exhibit a marked rise in serum concentrations of corticosterone between postnatal days 12 and 24. As this rise appears to be cued by L-thyroxine, the aim of the current study was to determine the physiological bases of the effects of both age and thyroid status on serum corticosterone. The in vivo response to either adrenocorticotropic hormone (60 mU/g body weight) or dibutyryl 3',5'-cyclic adenylate (0.3 mg/g body weight) increased between 10 and 16 days and was advanced and delayed by hyper- and hypothyroidism, respectively. In contrast, in vitro studies with adrenals from rats aged 10 and 16 days showed no effect of age on either basal or adrenocorticotropic hormone-stimulated production of corticosterone. Chronic administration of exogenous corticosterone to hyperthyroid animals resulted in significantly higher concentrations of serum corticosterone than did an identical administration to hypothyroid animals. As hyperthyroidism was associated with marked elevations in the concentration of corticosteroid-binding globulin and in the extent of binding of corticosterone, these results suggest that the effects of both age and thyroid status on serum concentrations of corticosterone may reflect changes in the metabolic clearance of the hormone.

DEVELOPMENT OF SYMPATHETIC NEURONES IN VIVO: AN INVESTIGATION OF THE POSSIBLE ROLE OF GLUCOCORTICO‐STEROIDS IN REGULATING TRANSMITTER TYPE

In vivo the majority of sympathetic neurones are adrenergic, whereas in vitro they can become cholinergic. This atypical cholinergic development can be suppressed by addition of a glucocorticoid to the culture medium (McLennan, Hill and Hendry, 1980). In this paper we have investigated the possibility that glucocorticoids influence the choice of sympathetic transmitter type in vivo. Reduction in circulatory glucocorticoids by adrenalectomy of 2-day-old rat pups did not result in increased cholinergic development in their superior cervical ganglia. Furthermore, administration of exogenous corticosterone to increase circulatory glucocorticoids in young rats did not prevent the developmental switch in transmitter type of the sympathetic innervation of the sweat gland from adrenergic to cholinergic. It is concluded that the developmental regulation of sympathetic transmitter type in vitro may not be analogous to that in vivo.

Stress down-regulates corticosterone receptors in a site-specific manner in the brain.

We have examined whether corticosterone receptor number within the brain can be regulated by its own ligand and whether such autoregulation reduces receptor number after the sustained secretion of corticosterone during repeated stress. Glucocorticoid receptors were measured in cytosolic preparations from acutely adrenalectomized rats using [1,2,6,7-3H] dexamethasone; maximal binding and receptor affinity parameters were determined by Scatchard analysis. Sustained elevations of circulating corticosterone, whether by repeated stress or exogenous corticosterone administration, did not change receptor affinity for [3H]dexamethasone, but significantly reduced cytosolic corticosterone receptor number. This reduction in total receptor number could not be attributed to residual tissue contamination with endogenous corticosterone after adrenalectomy or to translocation of cytosolic receptors to cell nuclei. The receptor reductions were anatomically specific, occurring in the hippocampus and amygdala, but not in the hypothalamus or pituitary, and were limited, in that exogenous corticosterone plus stress reduced receptor number no more than did stress alone. Further, 3 weeks of daily administration of 5 mg corticosterone caused reductions similar to those seen after only 4 days of treatment. Finally, the declines in glucocorticoid receptor number were reversible; receptor concentrations returned to normal levels within 1 week after the cessation of treatment with exogenous corticosterone. Thus, this study presents evidence that glucocorticoid down-regulation may constitute a physiological phenomenon.

Effect of corticosterone on myofibrillar protein turnover in diabetic rats as assessed by Ntau-methylhistidine excretion.

The effect of corticosterone on myofibrillar protein breakdown in diabetic rats was investigated in order to assess the possible counteracting effects of the secondary rise in plasma insulin concentrations which normally accompanies such treatment. N(tau)-Methylhistidine excretion, an index of myofibrillar protein breakdown, was compared before and after corticosterone treatment (4.0 mg/100 g body wt. per day) of normal control, adrenalectomized, 10-day-streptozotocin-diabetic and adrenalectomized diabetic rats. Diabetic rats received 1.5 units of insulin/100 g body wt. per day throughout the experiment and showed marked hyperglycaemia and glucosuria during corticosterone treatment, whereas non-diabetic rats had only mild hyperglycaemia but elevated insulin concentrations. Corticosterone treatment increased the average rate of myofibrillar protein breakdown by 68% and 95% respectively in non-diabetic and diabetic rats. Net loss of muscle non-collagen protein for the same 7-day period was greater in diabetic than in non-diabetic animals (4.15 versus 2.84% per day), and the calculated average synthesis rates were lowest in diabetic rats. Adrenalectomy had little effect except to decrease slightly the rate of muscle protein breakdown. These results show that the rise in plasma insulin concentrations that accompanies exogenous corticosterone administration to non-diabetic rats diminishes the catabolic effect of this glucocorticoid on muscle. Insulin appears to antagonize the effects of the glucocorticoid by attenuating the increased rates of myofibrillar protein breakdown and, to a lesser extent, by limiting the decrease in synthesis rates.

Adrenal involvement in conditioned immunosuppression

Abstract A taste aversion was induced in rats by pairing cyclophosphamide, an immunosuppressive drug, with the consumption of saccharin, a novel drinking solution. Three days after conditioning, animals were injected with sheep erythrocytes; hemagglutinating antibody titers were determined 6 days later. Relative to control groups, conditioned animals provided with saccharin at the time of antigen injection showed an attenuated antibody response, confirming the results of previous studies. The injection of LiCl to elicit an adrenocortical response or the exogenous administration of corticosterone in place of the conditioned stimulus (saccharin) at the time of antigen injection did not lower antibody titer significantly. The results provide no support for the hypothesis of an adrenocortical mediation of conditioned immunosuppressive effects.

The influence of adrenalectomy and of corticosterone administration on the ether-induced increase in plasma prolactin in ovariectomized estrogen-treated rats.

SummaryOvariectomized, polyestradiol-phosphate (PEP) injected, catheterized rats subjected to continuous ether anesthesia showed an increase in plasma prolactin level at 10 min followed by lower levels at 30, 60, and 120 min. Plasma corticosterone levels were increased significantly under continuous ether anesthesia. In contrast to intact animal, bilaterally adrenalectomized animals under ether anesthesia showed a sustained elevation of plasma prolactin at all time periods. In adrenalectomized animals subjected to ether anesthesia the exogenous administration of corticosterone (either 500 μg/rat or 25 μg/rat) prevented the increase in plasma prolactin at 10 min period when plasma level of corticosterone was high. In subsequent periods as the plasma corticosterone decreased, the plasma prolactin increased. It is suggested that corticosterone has an inhibitory feedback effect on the ether-induced prolactin release.

Schedule-induced polydipsia in rats: adrenocortical and hippocampal modulation.

Onset of schedule-induced polydipsia (SIP) is related to adrenal gland weight. Adrenalectomy, but not demedullation, hastened the emergence of SIP, and exogenous corticosterone administration tended to reverse this effect. Hippocampal lesions were followed by a rapid and uniform release of SIP. None of the above manipulations influenced normal (home-cage) drinking. A synthesis of present findings with the literature suggests that the hippocampus and the adrenal cortex interact and that the equilibrium established within this system is reflected, for any particular rat, in its adjunctive behavior.
