Effects of idebenone and coenzyme Q10 on NLRP3/caspase-1/IL-1β pathway regulation on ethanol-induced hepatotoxicity in rats

Chronic and excessive alcohol consumption leads to liver toxicity. There is a need to investigate effective therapeutic strategies to alleviate alcohol-induced liver injury, which remains the leading cause of liver-related morbidity and mortality worldwide. Therefore here, we looked into and evaluated how ethanol-induced hepatotoxicity was affected by coenzyme Q10 (CoQ10) and its analog, idebenone (IDE), on the NLRP3/caspase-1/IL-1 pathway. Hepatotoxicity induced in rats through the oral administration of gradually increasing dosages of ethanol (from 2 to 6 g/kg/day) over 30 days and the effect of CoQ10 (10 or 20 mg/kg) and IDE (50 or 100 mg/kg) were evaluated. Serum hepatotoxicity markers (ALT, AST, GGT, ALP, and TBIL), tissue oxidative stress markers and the mRNA expressions of IL-1β, IL-18, TGF-β, NF-κB, NLRP3, and caspase-1 were evaluated. Masson’s trichrome staining was also used to visualize fibrosis in the liver tissue. The results indicated that ethanol exposure led to hepatotoxicity as well as considerable NLRP3/caspase-1/IL-1β pathway activation. Moreover, CoQ10 or IDE treatment reduced measured parameters in a dosage-dependent manner. Thus, by inhibiting the NLRP3/caspase-1/IL-1 pathway, CoQ10 and IDE can prevent the hepatotoxicity caused by ethanol, although CoQ10 is more effective than IDE. This study will provide insight into new therapeutic avenues that take advantage of the anti-inflammatory and antioxidant properties of CoQ10 and IDE in ethanol-induced liver diseases.

Fruits of Emblica officinalis Linn. and its Phytochemicals in the Prevention of Ethanol-induced Hepatotoxicity: A Mini-Review

Abstract:: Excessive alcohol consumption over time is one of the primary causes of liver disease and death globally. Because there is no effective treatment, the situation has become more complicated, and the emphasis has shifted to the use of alternative and complementary medicine to treat liver problems. Herbs with dietary use have played an important part in the treatment of liver diseases since antiquity. Amla or Indian gooseberry, scientifically known as Phyllanthus emblica or Emblica officinalis, is a fruiting plant of the Phyllanthaceae family that is used in a variety of traditional and folk medical systems. The fruits have both dietary and medicinal value and are used to prevent chemical-induced hepatocarcinogenesis and reduce experimentally induced hyperlipidemia and metabolic syndrome in laboratory animals. They also have hepatoprotective effects against a variety of hepatotoxins such as heavy metals, iron overload, ochratoxins, hexachlorocyclohexane, carbon tetrachloride, therapeutic drugs such as paracetamol, and antitubercular. Amla fruits and phytochemicals such as quercetin, gallic acid, ellagic acid, and kaempferol are known to reduce ethanol-induced liver damage, and mechanistic studies have shown that the beneficial effects of these phytochemicals are mediated in part by free radical scavenging, antioxidant, and anti-inflammatory effects. This review examines the positive effects of amla fruit and its phytochemicals for the first time, as well as the mechanisms underlying the protective effects.

Bulbils of Aerial Yam Attenuate Ethanol-Induced Hepatotoxicity in HepG2 Cells through Inhibition of Oxidative Stress by Activation of the Nuclear Factor Erythroid-2-Related Factor 2 Signaling Pathway.

Bulbil of yam (BY) extract contains various active compounds possessing many pharmacological properties. However, little is known about the effect and underlying mechanism of BY extract on ethanol-induced liver damage. The present study explored the beneficial potential of BY extract on ethanol-induced hepatotoxicity. To evaluate its effectiveness, ethanol-induced HepG2 liver cells were pretreated with BY extract. BY extract effectively rescued cells from ethanol treatment through inhibition of apoptotic cell death as well as inhibiting expression of mitogen-activated protein kinase (MAPK) proteins as stress inducers. BY extract increased the expression of typical antioxidants. Furthermore, BY extract significantly inhibited mitochondrial dysfunction and endoplasmic reticulum (ER) stress, which are major ROS-inducing factors. Finally, as an underlying mechanism of the protective effects of BY extract on ethanol-induced liver damage, it activated Nrf2 protein through translocation from the cytosol to the nucleus, which in turn activated its target oxidative stress suppressor genes. Collectively, our findings demonstrate that BY extract has potential antioxidative effects in ethanol-induced liver cells and contributes to the establishment of a treatment strategy for alcohol-derived liver injuries.

INVESTIGATION OF THE HEPATOPROTECTIVE POTENTIAL OF FRACTIONS AND PHYTOSOMAL COMPLEXES OF TELFAIRIA OCCIDENTALIS HOOK F LEAF EXTRACTS ON ALCOHOL-INDUCED HEPATOTOXICITY IN RAT MODELS

The aim of this work was to evaluate the hepatoprotective potential of fractions and phytosomal complexes of Telfairia occidentalis leaf extracts on ethanol-induced hepatotoxicity in rat models. Phytosomal complexes of ethanol extract fractions were prepared by the thin film hydration technique. Hepatoprotective activities were evaluated by biochemical assays and histopathological examination of liver sections. Intoxication of animals with pure ethanol significantly (p < 0.05) elevated their liver enzyme titers, whereas co-administration with various extracts and phytosomal complexes reversed the increases. Histopathological findings revealed liver tissue protective effects of the extracts with the phytosomal complexes exhibiting greater effects (p < 0.05). Significant differences at p ≤ 0.05 were observed in the liver indices of animals treated with co-administered ethanol and extract fractions or phytosomes complexes versus the positive control group. T. occidentalis leaf extract formulated as phytosomes, therefore, has good potential of enhancing the hepatoprotective activities of the extract.

Establishment and elicitation of liquid adventitious root cultures of Inula crithmoides L. for increased caffeoylquinic acids production and hepatoprotective properties

This study established a liquid adventitious root culture for Inula crithmoides L. focusing on producing extracts rich in hepatoprotective caffeoylquinic acids through elicitation. Adventitious roots were induced from in vitro-grown plant leaves cultured on MS medium supplemented with 1 mg/L IBA. Growth and secondary metabolites accumulation (phenolics, flavonoids, hydroxycinnamic acids) were monitored weekly to determine the optimal harvesting time. Then, the adventitious roots were elicited for 4 weeks with varying concentrations of yeast extract–YE (50, 100 and 200 mg/L) and extracted with ethanol. The extracts were evaluated for their hepatoprotective effects against ethanol-induced hepatotoxicity and analyzed through high-performance liquid chromatography coupled with electrospray ionization mass spectrometry (HPLC–ESI–MS/MS). Roots of greenhouse-cultivated plants were used to compare the results obtained from in vitro -grown adventitious roots. IBA supplementation induced 100% adventitious root formation, with maximum biomass and phenolic accumulation after 4 weeks. Hydroxycinnamic acid levels increased significantly with YE treatment at all the concentrations, particularly in case of chlorogenic acid and di-O-caffeoylquinic acid isomers. The root extracts displayed significant in vitro hepatoprotective effects, particularly at 50 mg/L YE elicitation. Overall, the elicitation of I. crithmoides liquid root cultures with yeast extract offers a promising, efficient, and cost-effective approach for optimizing the production of valuable caffeoylquinic acids having potential pharmaceutical use.

Comparison of the protective effects of CS/TPP and CS/HPMCP nanoparticles containing berberine in ethanol-induced hepatotoxicity in rat

BackgroundAlcoholic liver disease (ALD) is a globally critical condition with no available efficient treatments.MethodsHerein, we generated chitosan (CS) nanoparticles cross-linked with two different agents, hydroxypropyl methylcellulose phthalate (HPMCP; termed as CS/HPMCP) and tripolyphosphate (TPP; termed as CS/TPP), and loaded them with berberine (BBr; referred to as CS/HPMCP/BBr and CS/TPP/BBr, respectively). Alongside the encapsulation efficiency (EE) and loading capacity (LC), the releasing activity of the nanoparticles was also measured in stimulated gastric fluid (SGF) and stimulated intestinal fluid (SIF) conditions. The effects of the prepared nanoparticles on the viability of mesenchymal stem cells (MSCs) were also evaluated. Ultimately, the protective effects of the nanoparticles were investigated in ALD mouse models.ResultsSEM images demonstrated that CS/HPMCP and CS/TPP nanoparticles had an average size of 235.5 ± 42 and 172 ± 21 nm, respectively. The LC and EE for CS/HPMCP/BBr were calculated as 79.78% and 75.79%, respectively; while the LC and EE for CS/TPP/BBr were 84.26% and 80.05%, respectively. pH was a determining factor for releasing BBr from CS/HPMCP nanoparticles as a higher cargo-releasing rate was observed in a less acidic environment. Both the BBr-loaded nanoparticles increased the viability of MSCs in comparison with their BBr-free counterparts. In vivo results demonstrated CS/HPMCP/BBr and CS/TPP/BBr nanoparticles protected enzymatic liver functionality against ethanol-induced damage. They also prevented histopathological ethanol-induced damage.ConclusionsCrosslinking CS nanoparticles with HPMCP can mediate controlled drug release in the intestine improving the bioavailability of BBr.

Hydrogen gas alleviates acute ethanol-induced hepatotoxicity in mice via modulating TLR4/9 innate immune signaling and pyroptosis

Alcoholic liver disease (ALD), which is induced by chronic heavy alcohol consumption, accompanies complicated pathological mechanisms, including oxidative stress, inflammation, cell death, epigenetic changes and acetaldehyde-mediated toxicity. Hydrogen (H2) is the lightest gas with multiple biological effects such as high selective anti-oxidation, anti-inflammation and anti-apoptosis. However, the dose effects and innate immune mechanisms of intraperitoneal injection of H2 on ALD are limited. Here, we used acute ethanol-induced hepatotoxicity mice models to estimate the actions of intraperitoneal injection of H2 on ALD. The effects of H2 on acute ethanol-induced liver damage were examined by hepatic oil red O staining, quantitative PCR (qPCR) for lipid metabolic genes, hepatic triglyceride (TG) and serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels. Hepatic mitochondrial superoxide (MitoSOX), 3-nitrotyrosine (3-NT), malondialdehyde (MDA), and glutathione (GSH) levels were examined to evaluate oxidative stress. Immunoblot, and immunofluorescence staining were used to further confirm the innate immune molecular targets of H2. Our results showed that intraperitoneal injection of H2 improved acute ethanol-induced liver injury in mice in a dose dependent manner, as indicated by decreasing serum ALT and AST levels, hepatic TG levels, and increasing lipid export genes (Mttp and Apob) mRNA levels and reducing fatty acid uptake gene (CD36) mRNA levels. Mechanistically, H2 inhibited hepatic oxidative stress as indicated by reducing reactive oxygen species (ROS), 3-NT, and MDA levels in the liver, while increasing hepatic GSH levels; inhibited the overactived TLR4/9-NF-κB-TNF-α/IL-1β/IL-18 innate immune signaling; suppressed the canonical Caspase-1-GSDMD pyroptosis signaling, and the non-canonical pyroptosis signaling, such as Caspase-11-GSDMD, Caspase-8-GSDMD and Caspase-3-GSDME signaling. Therefore, our study highlights that intraperitoneal injection of H2 may represent a novel therapeutic and safe strategy for ALD via modulating oxidative stress, innate immunity and pyroptosis.

Fermented Protaetia brevitarsis Larvae Ameliorates Chronic Ethanol-Induced Hepatotoxicity in Mice via AMPK and TLR-4/TGF-β1 Pathways.

This study evaluated the hepatoprotective effect of fermented Protaetia brevitarsis larvae (FPB) in ethanol-induced liver injury mice. As a result of amino acids in FPB, 18 types of amino acids including essential amino acids were identified. In the results of in vitro tests, FPB increased alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) activities. In addition, FPB treatment increased cell viability on ethanol- and H2O2-induced HepG2 cells. FPB ameliorated serum biomarkers related to hepatoxicity including glutamic oxaloacetic transaminase, glutamine pyruvic transaminase, total bilirubin, and lactate dehydrogenase and lipid metabolism including triglyceride, total cholesterol, high-density lipoprotein cholesterol, and low-density lipoprotein cholesterol. Also, FPB controlled ethanol metabolism enzymes by regulating the protein expression levels of ADH, ALDH, and cytochrome P450 2E1 in liver tissue. FPB protected hepatic oxidative stress by improving malondialdehyde content, reduced glutathione, and superoxide dismutase levels. In addition, FPB reversed mitochondrial dysfunction by regulating reactive oxygen species production, mitochondrial membrane potential, and ATP levels. FPB protected ethanol-induced apoptosis, fatty liver, and hepatic inflammation through p-AMP-activated protein kinase and TLR-4/NF-κB signaling pathways. Furthermore, FPB prevented hepatic fibrosis by decreasing TGF-β1/Smad pathway. In summary, these results suggest that FPB might be a potential prophylactic agent for the treatment of alcoholic liver disease via preventing liver injury such as fatty liver, hepatic inflammation due to chronic ethanol-induced oxidative stress.

Effect of Lauric acid against ethanol-induced hepatotoxicity by modulating oxidative stress/apoptosis signalling and HNF4α in Wistar albino rats

Ethanol (EtOH) is most widely used in alcoholic beverages to prepare alcohol. As EtOH is mainly metabolised in the liver, the excessive consumption of EtOH forms a primary toxic metabolic product called acetaldehyde, as the gradual increase in acetaldehyde leads to liver injury, as reported. Lauric acid (LA) is rich in antioxidant, antifungal, antibacterial, anticancer, and antiviral properties. LA is an edible component highly present in coconut oil. However, no report on LA protective effects against the EtOH-instigated hepatotoxicity exists. Therefore, the experiment is carried out to investigate the potency effects of LA on EtOH-instigated hepatotoxicity in thirty male albino rats. Rats were divided into five groups (n-6): control DMSO alone, EtOH -intoxicated, EtOH + LA 180 mg/kg, EtOH + LA 360 mg/kg, and LA alone were administered orally using oral gavage. The study measured body weight every weekend in all rat groups. The rats were sacrificed and assessed for serum markers (alkaline phosphatase, alanine aminotransferase, aspartate aminotransferase), antioxidant activity (superoxide dismutase, reduced glutathione, glutathione peroxidase), lipid peroxidation (malondialdehyde), histopathological, cytokine levels (TNF-α, IL-1β and IL-6), protein expression (caspase 3 and caspase 8 and Bcl-2 and HNF4α) were evaluated after the 56-days study period. The impact of EtOH intoxication reduces the rat's body weight by 90 g, upregulates the liver enzyme markers, depletes the antioxidant levels, produces malondialdehyde, changes the histoarchitecture (periportal inflammation and hepatocyte damage), downregulates the Bcl-2 expressions and HNF4α, and elevates the expression of cytokines and apoptotic markers. LA alleviated EtOH-induced liver toxicity by significant (p < 0.05) modulation of biochemical levels, caspase-8/3 signalling, reducing pro-inflammatory cytokines, and restoring the normal histoarchitecture, upregulating the Bcl-2 and HNF4α Expressions. In conclusion, LA treatment can protect the liver against EtOH-induced hepatotoxicity, evidenced by alleviating Oxidative stress, lipid peroxidation, inflammation, apoptosis, and upregulation of HNF4α.

Synergistic Protective Effect of Fermented Schizandrae Fructus Pomace and Hoveniae Semen cum Fructus Extracts Mixture in the Ethanol-Induced Hepatotoxicity.

Schizandrae Fructus (SF), fruits of Schisandra chinensis (Turcz.) Baill. and Hoveniae Semen cum Fructus (HSCF), the dried peduncle of Hovenia dulcis Thunb., have long been used for alcohol detoxification in the traditional medicine of Korea and China. In the current study, we aimed to evaluate the potential synergistic hepatoprotective effect of a combination mixture (MSH) comprising fermented SF pomace (fSFP) and HSCF hot water extracts at a 1:1 (w:w) ratio against ethanol-induced liver toxicity. Subacute ethanol-mediated hepatotoxicity was induced by the oral administration of ethanol (5 g/kg) in C57BL/6J mice once daily for 14 consecutive days. One hour after each ethanol administration, MSH (50, 100, and 200 mg/kg) was also orally administered daily. MSH administration significantly reduced the serum activities of alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, and γ-glutamyl transpeptidase. Histological observation indicated that MSH administration synergistically and significantly decreased the fatty changed region of hepatic parenchyma and the formation of lipid droplet in hepatocytes. Moreover, MSH significantly attenuated the hepatic triglyceride accumulation through reducing lipogenesis genes expression and increasing fatty acid oxidation genes expression. In addition, MSH significantly inhibited protein nitrosylation and lipid peroxidation by lowering cytochrome P450 2E1 enzyme activity and restoring the glutathione level, superoxide dismutase and catalase activity in liver. Furthermore, MSH synergistically decreased the mRNA level of tumor necrosis factor-α in the hepatic tissue. These findings indicate that MSH has potential for preventing alcoholic liver disease through inhibiting hepatic steatosis, oxidative stress, and inflammation.

Synthesis, characterization and hepatoprotective effect of silymarin phytosome nanoparticles on ethanol-induced hepatotoxicity in rats.

Silymarin proved to be a beneficial herbal medicine against many hepatic disorders such as alcoholic liver disease (ALD). However, its application is restricted due to its low bioavailability and consequently decreased efficacy. We herein used a nano-based approach known as "phytosome", to improve silymarin bioavailability and increase its efficacy. Phytosome nanoparticles (NPs) were synthesized using thin film hydration method. NPs size, electrical charge, morphology, stability, molecular interaction, entrapment efficiency (EE %) and loading capacity (LC %) were determined. Moreover, in vitro toxicity of NPs was investigated on mesenchymal stem cells (MSCs) viability using MTT assay. In vivo experiments were performed using 24 adult rats that were divided into four groups including control, ethanol (EtOH) treatment, silymarin/EtOH treatment and silymarin phytosome/EtOH, with 6 mice in each group. Experimental groups were given 40% EtOH, silymarin (50 mg/kg) and silymarin phytosome (200 mg/kg) through the gastric gavage once a day for 3 weeks. Biochemical parameters, containing ALP, ALT, AST, GGT, GPx and MDA were measured before and after experiment to investigate the protective effect of silymarin and its phytosomal form. And histopathological examination was done to evaluate pathological changes. Silymarin phytosome NPs with the mean size of 100 nm were produced and were well tolerated in cell culture. These NPs showed a considerable protective effect against ALD through inverting the biochemical parameters (ALP, ALT, AST, GGT, GPx) and histopathological alterations. Silymarin phytosomal NPs can be used as an efficient treatment for ALD.

In vitro, in vivo, and in silico analysis of synbiotics as preventive interventions for lipid metabolism in ethanol-induced adipose tissue injury

The risk of alcoholic liver disease (ALD) is increased by excessive ethanol drinking. For the prevention of ALD, the effects of ethanol on the liver, adipose tissue, and gut are crucial. Interestingly, garlic and a few probiotic strains can protect against ethanol-induced hepatotoxicity. However, the relationship between adipose tissue inflammation, Kyolic aged garlic extract (AGE), and Lactobacillus rhamnosus MTCC1423 in developing ALD is unknown. Therefore, the present study explored the effect of synbiotics (a combination of prebiotics and probiotics) on adipose tissue to prevent ALD. To investigate the efficacy of synbiotics administration on adipose tissue in preventing ALD, in vitro (3T3-L1 cells, N = 3) groups: control, control + LPS (lipopolysaccharide), ethanol, ethanol + LPS, ethanol + synbiotics, ethanol + synbiotics + LPS; in vivo (Wistar male rats, N = 6) groups: control, ethanol, pairfed, ethanol + synbiotics and in silico experiments were conducted. Lactobacillus multiplies in accordance with the growth curve when exposed to AGE. Additionally, Oil red O staining and scanning electron microscopy (SEM) demonstrated that synbiotics therapy maintained the morphology of adipocytes in the alcoholic model. In support of the morphological changes, quantitative real-time PCR demonstrated overexpression of adiponectin and downregulation of leptin, resistin, PPARγ, CYP2E1, iNOS, IL-6, and TNF-α after administration of synbiotics compared to the ethanol group. In addition, MDA estimation by high-performance liquid chromatography (HPLC) indicated that the synbiotics treatment reduced oxidative stress in rat adipose tissue. Consequently, the in-silico analysis revealed that AGE inhibited the C-D-T networks as PPARγ acting as the main target protein. Nigella sativa L. oil: Study of its toxicity, antiradical activity, and effect on circulating xanthine oxidoreductase

Introduction: Nigella sativa L. is a widely used medicinal plant throughout the world. The low toxic effects and low price of this plant make it an excellent treatment choice for many diseases. The present study aims to investigate the hepatoprotective effect of N. sativa L. total oil (TO) and its neutral lipid fraction (NLF) via the estimation of circulating xanthine oxidoreductase (XOR) level and anti-XOR antibodies titer. Methods: Antiradical activities of TO and NLF, in vitro, were carried out using three reactive oxygen species (ROS), superoxide anion, hydroxyl radical, and hydrogen peroxide. In vivo study was conducted to determine the possible protective effects of TO and NLF against ethanol-induced hepatotoxicity in rats feeding Lieber-DeCarli liquid diet in both sera and liver homogenate. Before conducting the hepatoprotective effects, we assessed the toxicity of our extracts using the same animal model. The administrated doses of 400 mg/kg for TO and 300 mg/kg for NLF showed no toxic effects. Results: ELISA assay indicated a significant increase (P&lt;0.001) in the level of XOR and the titer of anti-XOR antibodies in rats treated with ethanol compared to the control group. After treatment with TO and NLF, the titer of anti-XOR antibodies and the level of XOR decreased significantly compared to the control group. Conclusion: XOR plays an important role in alcohol liver pathologies as a major source of free radicals. In addition, TO and NLF have significant potential as liver protective agents and might be utilized as new antioxidant therapeutics.

Thymol ameliorates ethanol-induced hepatotoxicity via regulating metabolism and autophagy

Alcoholic liver disease represents a serious threat to human health. In terms of safety and acceptability, thymol is widely used in or on foodstuffs to generate odour and taste. The present study aimed to investigate the therapeutic effect and mechanism of thymol against ethanol-induced injury in liver cells. Here we found that thymol is an effective agent for reducing ethanol-induced reactive oxygen species production in mouse liver cells. Thymol improves ethanol-induced lipid accumulation, and this corresponded to altered DGAT2 mRNA expression levels. Metabolomics data analysis showed that thymol alleviated ethanol-induced changes in the levels of thirty-four metabolites including nicotinic acid and l-arginine. By utilizing pathway enrichment analysis, altered metabolites in cells treated with ethanol and ethanol plus thymol were enriched in fourteen pathways including metabolic pathways and arginine and proline metabolism. We further confirmed the alleviation of overdose nitric oxide production in cells treated with ethanol plus thymol compared with that in ethanol-treated cells. It was interesting that up-regulated LC3-II/LC3-I ratio together with higher SQSTM1 protein abundance in ethanol-treated cells were attenuated by treatment with ethanol plus thymol. Thymol ameliorated ethanol-induced reduction of HSPA8 protein abundance. In addition, chloroquine-treated cells exhibited lower HSPA8 protein abundance compared with cells simulated with ethanol plus thymol. These data reveal that improving effect of thymol on ethanol-induced metabolic alteration is related to autophagic flux restoration. Our findings indicate that thymol is an attractive option for treating ethanol-induced liver damage.

AMPK-Regulated Autophagy Contributes to Ursolic Acid Supplementation-Alleviated Hepatic Steatosis and Liver Injury in Chronic Alcohol-Fed Mice.

Alcoholic liver disease (ALD) is a chronic liver disease caused by long-term heavy consumption of alcohol. The pathogenesis of ALD is complex, and there is no effective clinical treatment at present. Ursolic acid (UA), a general triterpenoid with multiple biological roles, is widely distributed in plants. This study aims to explore the therapeutic effect and potential mechanisms of UA that protect against liver injury and hepatic steatosis in an ALD mouse model. In this study, we analyzed the lipid accumulation and the effect of UA treatment in a mouse model of ALD; AML12 and HepG2 cells were used to study the biological effect and potential mechanisms of UA on ethanol-induced hepatotoxicity. The morphologic and histological detections showed that UA significantly reduced alcohol-induced liver injury and hepatic steatosis. In addition, UA dramatically ameliorated alcohol-induced metabolic disorders, oxidative stress, and inflammation. Furthermore, UA treatment activated autophagy via the AMPK-ACC pathway to protect hepatocytes from lipotoxicity. Thus, these findings demonstrate that UA treatment alleviates alcoholic-induced liver injury by activating autophagy through the AMPK-ACC pathway. Therefore, UA may represent a promising candidate for the treatment of ALD.

Evaluation of Pharmacological Profiling of Albizia Odoratissima Bark Extracts on Ethanol-Induced Hepatotoxicity in Albino Rats

Alcohol consumption is a common cause of acute and chronic hepatic toxicity in patients. Discovery of novel therapeutic agents from medicinal plants is most recommended because of the presence of diverse phytochemicals. The aim of the study is to evaluate the therapeutic effect of methanolic extract of bark of Albizia odoratissima on ethanol-induced hepatotoxicity in rats. The following objectives A. odoratissima bark extract was screened for phytochemical analysis, and tests on acute toxicity and hepatoprotection using an Albino rat model were conducted. Twenty-four male albino rats were selected and divided into four groups (n=6). Animals received treatments for a period of 21 days. Normal control (Group-I) received vehicles; toxic control (Group-II) received ethanol (2 ml/kg/p.o.). Treatment groups (Groups-III and IV) received Silymarin (50 mg/kg/p.o.) and plant extract (250 mg/kg/p.o.). Biochemical parameters such as hepatic enzymes, biomarkers and histopathology were estimated to assess the severity of hepatic injury and or disease. Phytochemical analysis of extract reported the presence of secondary metabolites. Ethanol significantly increased SGPT and SGOT, ALP, TB, urea, and creatinine and significantly decreased total protein and albumin levels, indicating hepatic damage. Silymarin ameliorates all hepatic enzymes and biomarkers as compared to hepatotoxic control. Meanwhile, methanolic extract significantly decreased only SGPT, ALP, and creatinine, compared to hepatotoxic control. Histopathological examinations of silymarin and A. odoratissima revealed reduced liver inflammation. This study concludes that a 250mg/kg dose of A. odoratissima showed partial protective effects on ethanol-induced hepatotoxicity in rats as compared with silymarin.Â

Evaluation of Polyherbal preparation of Livina against Ethanol Induced Liver Dysfunction

The present study was performed to evaluate the hepatoprotective activity of ployherbal preparation of livina against ethanol-induced hepatotoxicity in rats. Four groups of adult albino wistar rats were taken for the study. Rats were divided into four groups each containing 6 animals. Group first is normal group received the normal saline 10ml/kg for 21th days. Group second negative control this group received ethanol 12ml/kg bw per day is standard dose of this group is Silymarin (100mg/kg) daily for 21th days and in 21th day administered ethanol. Group third is standard dose of this group is Silymarin (100mg/kg) daily for 21th days and in 21th day administered ethanol. Silymarin and ethanol is administered orally. Fourth group is treated with ethanol + livina (Herbal capsule). The level of Serum Glutamic Pyruvic Transaminase Test (SGPT), Serum Glutamic-Oxaloacetic Transaminase Test (SGOT), alkaline phosphatase (ALP) and total bilirubin were determined to assay hepatotoxicity. Ethanol administration caused severe hepatic damage in rats as evidenced by elevated serum SGPT, SGOT, ALP and total bilirubin levels. The polyherbal preparation of livina and silymarin administration prevented the toxic effect of ethanol. The present study concludes polyherbal preparation of livina that has significant hepatoprotective activity against ethanol induced hepatotoxicity, which may be associated with its bioactive component.

The potential effects of HECTD4 variants on fasting glucose and triglyceride levels in relation to prevalence of type 2 diabetes based on alcohol intake

Excessive alcohol intake is an important cause of major public health problem in East Asian countries. Growing evidence suggests that genetic factors are associated with alcohol consumption and the risk for alcohol-associated disease, and these factors contribute to the risk of developing chronic diseases, including diabetes. This study aims to investigate the association of type 2 diabetes with genetic polymorphisms within HECTD4 based on alcohol exposure. We performed a genome-wide association study involving the cohorts of the KoGES-HEXA study (n = 50,028) and Ansan and Ansung study (n = 7,980), both of which are prospective cohort studies in Korea. The top three single-nucleotide polymorphisms (SNPs) of the HECTD4 gene, specifically rs77768175, rs2074356 and rs11066280, were found to be significantly associated with alcohol consumption. We found that individuals carrying the variant allele in these SNPs had lower fasting blood glucose, triglyceride, and GGT levels than those with the wild-type allele. Multiple logistic regression showed that statistically significant associations of HECTD4 gene polymorphisms with an increased risk of type 2 diabetes were found in drinkers. Namely, these SNPs were associated with decreased odds of diabetes in the presence of alcohol consumption. As a result of examining the effect of alcohol on the expression of the HECTD4 gene, ethanol increased the expression of HECTD4 in cells, but the level was decreased by NAC treatment. Similar results were obtained from liver samples of mice treated with alcohol. Moreover, a loss of HECTD4 resulted in reduced levels of CYP2E1 and lipogenic gene expression in ethanol-treated cells, while the level of ALDH2 expression increased, indicating a reduction in ethanol-induced hepatotoxicity.

Metabolic Signature of Ethanol-Induced Hepatotoxicity in HepaRG Cells by Liquid Chromatography-Mass Spectrometry-Based Untargeted Metabolomics.

Alcoholic liver disease is highly prevalent but poorly identified and characterized, leading to knowledge gaps, which impairs early diagnosis. Excessive alcohol consumption is known to alter lipid metabolism, followed by progressive intracellular lipid accumulation, resulting in alcoholic fatty liver disease. In this study, HepaRG cells were exposed to ethanol at IC10 and 1/10 IC10 for 24 and 48 h. Metabolic alterations were investigated intra- and extracellularly with liquid chromatography-high-resolution mass spectrometry. Ion mobility was added as an extra separation dimension for untargeted lipidomics to improve annotation confidence. Distinctive patterns between exposed and control cells were consistently observed, with intracellular upregulation of di- and triglycerides, downregulation of phosphatidylcholines and phosphatidylethanolamines, sphingomyelins, and S-adenosylmethionine, among others. Several intracellular metabolic patterns could be related to changes in the extracellular environment, such as increased intracellular hydrolysis of sphingomyelins, leading to increased phosphorylcholine secretion. Carnitines showed alterations depending on the size of their carbon chain, which highlights the interplay between β-oxidation in mitochondria and peroxisomes. Potential new biomarkers of ethanol-induced hepatotoxicity have been observed, such as ceramides with a sphingadienine backbone, octanoylcarnitine, creatine, acetylcholine, and ethoxylated phosphorylcholine. The combination of the metabolic fingerprint and footprint enabled a comprehensive investigation of the pathophysiology behind ethanol-induced hepatotoxicity.

Evaluation of Endophytic Fungal Fractions of Phyllanthus amarus schum andthonn Leaves in Paracetamol and Ethanol Induced Hepatotoxicity

Background Endophytes are the microbes residing inside the plant tissues forming relations such as mutualistic symbiotic and trophobiotic. Paracetamol is a widely used analgesic and antipyretic drug higher doses of which leads to serious liver disorders. Alcohol a psychoactive drug causes deleterious effects on the liver with high consumption. In both cases pathological conditions vary from basic intoxication to lifethreatening pathological states.nbspAim of the study An attempt was made to study the effects of fractions of endophytic fungi Nigrospora sp. CMH213 PALF-2 isolated in our previous report from leaves of Phyllanthus amarus in paracetamol and ethanol-induced hepatotoxicity.Materials and Methods In this study the effects of earlier isolated endophytic fungal fractions namely P2EA ethyl acetate and P2nB n butanol were studied on paracetamol and ethanol-induced hepatotoxicity at doses of 50 mgkg amp 100 mgkg. Various biochemical parameters and histopathology of the livers were also studied in both models.Results In both the models the elevated biochemical parameters such as aspartate aminotransferase AST alanine aminotransferase ALT alkaline phosphatase ALP total bilirubin direct bilirubin and triglycerides TG were reversed by P2EA and P2nB 50 mgkg amp 100 mgkg as compared to induced groups plt0.001. P2EA and P2nB 50 mgkg amp 100 mgkg also restored the lipid peroxidation LPO superoxide SOD and catalase CAT levels.Conclusion P2EA and P2nB exhibited hepatoprotective activity significantly in paracetamol and ethanolinduced models from endophyte Nigrospora sp.
