Bioavailability of rumen-protected histidine, lysine, and methionine assessed using different in vivo methods.

The objective of this experiment was to estimate the bioavailability (BA) of rumen-protected (RP) His, RPLys, and 2 RPMet products using 3 in vivo methods: area under the curve (AUC), plasma dose-response (PDR), and fecal free AA (FFAA) methods. We used 8 rumen-cannulated cows in a replicated 4 × 4 Latin square experiment with 16-d periods. Treatments were (1) abomasal infusion of water (control), (2) abomasal infusion of free His, Lys, and Met (FAA), (3) administration of RPHis + RPLys + RPMet1 (rumen-protected methionine protected with ethyl cellulose; RPAA1), and (4) administration of RPHis + RPLys + RPMet2 (rumen-protected methionine protected with a pH-sensitive polymer; RPAA2). On d 7 of each experimental period, a pulse-dose of water (control) or FAA were infused into the abomasum of the cows, or RPAA were placed directly in the rumen, and blood samples were taken from the jugular vein through a catheter 11 times over a 24-h period for the AUC method. Following the AA pulse-dose, infusion lines were installed into the abomasum for continuous infusion of FAA for the PDR method, or RPAA were fed from d 12 to d 16 and cows were fitted with urinary catheters for total collection of feces for the FFAA method. Fecal collection and blood sampling were conducted from d 14 to 16. Due to technical issues likely leading to unrealistic BA estimates, data for the PDR method are reported in the supplemental material. Relative BA based on the AUC method (computed as AUC of RPAA treatment plasma AA concentration divided by AUC of FAA treatment plasma AA concentration) was lower for RPMet1 compared with RPMet2 (43% vs. 61%) and was 45% (SEM = 3.35) and 72% (SEM = 5.99), for RPHis and RPLys, respectively. Rumen escape fractions of RPAA, estimated in a previous study using an in situ method, and digestibility data from the current study were used for calculations of BA for the FFAA method. Bioavailability based on the FFAA method was lower for RPMet1 (67%) compared with RPMet2 (91%) and was 87% (SEM = 0.71) and 75% (SEM = 2.75) for RPHis and RPLys, respectively. The relative differences in estimated BA based on both the AUC and FFAA methods between the RPMet products were as expected, based on literature, and data for all 4 RPAA products corresponded well with previously estimated BA using the FFAA method. The unrealistic data for the PDR method were likely caused by technical deviations from the original method (e.g., once-daily dosing of RPAA and inability to capture representative plasma concentration data with the sampling time points). Therefore, comparison of the PDR method with the AUC and FFAA methods were not possible in this study. Further comparisons are needed without deviations from the original PDR method. Variability in BA data and differences in estimated BA between the in vivo methods highlight the current challenges for accurate measurements of relative in vivo BA of RPAA products. Different protection technologies may call for different methodology to be used for BA estimations. Further research and standardization of in vivo BA methods are warranted.

Predicting Human Subcutaneous Bioavailability of Therapeutic Monoclonal Antibodies from Systemic Clearance and Volume of Distribution.

Subcutaneous delivery of monoclonal antibody therapeutics is often preferred to intravenous delivery due to better patient compliance and overall lower cost to the healthcare system. However, the systemic absorption of biologics dosed subcutaneously is often incomplete. The aim of this work was to describe a human bioavailability prediction method for monoclonal antibodies delivered subcutaneously that utilizes intravenous pharmacokinetic parameters as input. A two-compartment pharmacokinetic model featuring a parallel-competitive absorption pathway and a presystemic metabolism pathway was employed. A training data set comprised 19 monoclonal antibodies (geometric mean bioavailability of 68%), with previously reported human pharmacokinetic parameters, while a validation set included data compiled from 5 commercial drug products (geometric mean bioavailability of 69%). A single fitted absorption rate constant, paired with compound-specific estimates of presystemic metabolism rate proportional to compound-specific systemic clearance parameters, resulted in calculations of human subcutaneous bioavailability closely mimicking clinical data in the training data set with a root-mean-square error of 5.5%. Application of the same approach to the validation data set resulted in predictions characterized by 12.6% root-mean-square error. Factors that may have impacted the prediction accuracy include a limited number of validation data set compounds and an uncertainty in the absorption rate, which were subsequently discussed. The predictive method described herein provides an initial estimate of the subcutaneous bioavailability based exclusively on pharmacokinetic parameters available from intravenous dosing.

Investigation on the Potential of Some N-phenylalkanehydrazides as Superoxide Anion Radical Scavengers and Glutathione Peroxidase Binders with ADME Prediction: Comparison with Vitamin C

Objective: An investigation on the antioxidant activity and ADME properties of some N-phenylalkanehydrazides (NPhs) in comparison with ascorbic acid was carried out. Materials and Methods: The power of the compounds to scavenge the superoxide anion radical was examined using cyclic voltammetry. Furthermore, molecular docking was used to examine how NPhs bind to the antioxidant enzyme glutathione peroxidase (GPx). Finally, ADME properties were predicted using the SwissADME webserver. Results: The results showed that the studied compounds had significantly higher EC50 values than Vitamin C and displayed spontaneous binding with GPx with binding energy similar to ascorbic acid. Also, substantial values emerged from the predictive analysis of drug-likeness and bioavailability characteristics. Discussion: First, the EC50 of the compounds under study was determined by assessing the decrease in the anodic current peak intensity; this demonstrated significant radical scavenging activity. Next, the electrostatic way of binding with ROS radical and with antioxidant enzyme was disclosed by the binding free energies; however, the oxygen and nitrogen atoms in the compounds' structures might make many conventional hydrogen bonds with amino acids. Finally, NPh (a) followed all the rules for drug-likeness and bioavailability estimates, making it a promising moiety for synthesising new antioxidant chemicals. Conclusion: The current study is a hybrid of experimental and computational methods that complement each other perfectly.

The critical role of organic matter for cadmium-lead interactions in soil: Mechanisms and risks

Understanding the interaction mechanisms between complex heavy metals and soil components is a prerequisite for effectively forecasting the mobility and availability of contaminants in soils. Soil organic matter (SOM), with its diverse functional groups, has long been a focal point of research interest. In this study, four soils with manipulated levels of SOM, cadmium (Cd) and lead (Pb) were subjected to a 90-day incubation experiment. The competitive interactions between Cd and Pb in soils were investigated using Fourier transform infrared spectrometer (FTIR), X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), and X-ray adsorption near-edge structure (XANES) analysis. Our results indicate that Pb competed with Cd for adsorption sites on the surface of SOM, particularly on carboxyl and hydroxyl functional groups. Approximately 22.6 % of Cd adsorption sites on humus were occupied by Pb. The use of sequentially extracted exchangeable heavy metals as indicators for environment risk assessments, considering variations in soil physico-chemical properties and synergistic or antagonistic effects between contaminants, provides a better estimation of metal bioavailability and its potential impacts. Integrating comprehensive contamination characterization of heavy metal interactions with the soil organic phase is an important advancement to assess the environmental risks of heavy metal dynamics in soil compared to individual contamination assessments.

An in-silico modeling approach to separate exogenous and endogenous plasma insulin appearance, with application to inhaled insulin

The aim of this study was to develop a dynamic model-based approach to separately quantify the exogenous and endogenous contributions to total plasma insulin concentration and to apply it to assess the effects of inhaled-insulin administration on endogenous insulin secretion during a meal test. A three-step dynamic in-silico modeling approach was developed to estimate the two insulin contributions of total plasma insulin in a group of 21 healthy subjects who underwent two equivalent standardized meal tests on separate days, one of which preceded by inhalation of a Technosphere® Insulin dose (22U or 20U). In the 30–120 min test interval, the calculated endogenous insulin component showed a divergence in the time course between the test with and without inhaled insulin. Moreover, the supra-basal area-under-the-curve of endogenous insulin in the test with inhaled insulin was significantly lower than that in the test without (2.1 ± 1.7 × 104 pmol·min/L vs 4.2 ± 1.8 × 104 pmol·min/L, p < 0.01). The percentage of exogenous insulin reaching the plasma, relative to the inhaled dose, was 42 ± 21%. The proposed in-silico approach separates exogenous and endogenous insulin contributions to total plasma insulin, provides individual bioavailability estimates, and can be used to assess the effect of inhaled insulin on endogenous insulin secretion during a meal.

Exploring various measures of the area under the curve for the assessment of dose-proportionality and estimation of bioavailability.

In pharmacokinetics, the area under the concentration versus time curve (AUC) extrapolated to infinity (AUC0-∞) is the preferred metric but it is not always possible to have a reliable estimate of the terminal phase half-life. Here we sought to explore the accuracy of three different area measures to accurately identify dose proportionality and bioavailability. One to three compartment model simulations with different doses for dose-proportionality or different rates and/or extents of bioavailability. Area measures evaluated were AUC0-∞, to the last quantifiable concentration (AUCtlast), and to a common time value (AUCt'). Under linear pharmacokinetics, AUCt' provided the most accurate measure of dose proportionality. Except for the one compartment model where AUC0-∞ provided the best predictor of the true measure, there was no clear advantage to the use of either of the three measures of AUC. With uncertainty about the terminal phase half-life, the use of AUCt' can be a very useful and even the preferred measure of exposure for use in assessing proportionality in exposure between doses. The choice of AUC measure in bioavailability is less clear and may depend on compartmental nature of the drug, and study parameters including assay sensitivity and sampling protocols.

Performance of pharmacokinetic models developed using in vitro-to-in vivo extrapolation (IVIVE) methods may be improved by refining assumptions regarding fraction absorbed (Fabs) through the intestine, a component of oral bioavailability (Fbio). Although in vivo measures of Fabs are often unavailable for non-pharmaceuticals, in vitro measures of apparent permeability (Papp) using the Caco-2 cell line have been highly correlated with Fabs. We measured bidirectional Papp for over 400 non-pharmaceutical chemicals using the Caco-2 assay. A random forest quantitative structure-property relationship (QSPR) model was developed using these and peer-reviewed pharmaceutical data. Both Caco-2 data (R²=0.37) and the QSPR model (R²=0.29) were better at predicting human bioavailability compared to in vivo rat data (R²=0.23). After incorporation into a high throughput toxicokinetics (HTTK) framework for IVIVE, the Caco-2 data were used to estimate in vivo administered equivalent dose (AED) for bioactivity assessed in vitro, The HTTK-predicted plasma steady state concentrations (Css) for IVIVE were revised, with modest changes predicted for poorly absorbed chemicals. Experimental data were evaluated for sources of measurement uncertainty, which were then accounted for using the Monte Carlo method. Revised AEDs were subsequently compared with exposure estimates to evaluate effects on bioactivity:exposure ratios, a surrogate for risk. Only minor changes in the margin between chemical exposure and predicted bioactive doses were observed due to the preponderance of highly absorbed chemicals.

A Review of Mechanistic Models for Predicting Adverse Effects in Sediment Toxicity Testing.

Since recognizing the importance of bioavailability for understanding the toxicity of chemicals in sediments, mechanistic modeling has advanced over the last 40 years by building better tools for estimating exposure and making predictions of probable adverse effects. Our review provides an up-to-date survey of the status of mechanistic modeling in contaminated sediment toxicity assessments. Relative to exposure, advances have been most substantial for non-ionic organic contaminants (NOCs) and divalent cationic metals, with several equilibrium partitioning-based (Eq-P) models having been developed. This has included the use of Abraham equations to estimate partition coefficients for environmental media. As a result of the complexity of their partitioning behavior, progress has been less substantial for ionic/polar organic contaminants. When the EqP-based estimates of exposure and bioavailability are combined with water-only effects measurements, predictions of sediment toxicity can be successfully made for NOCs and selected metals. Both species sensitivity distributions and toxicokinetic and toxicodynamic models are increasingly being applied to better predict contaminated sediment toxicity. Furthermore, for some classes of contaminants, such as polycyclic aromatic hydrocarbons, adverse effects can be modeled as mixtures, making the models useful in real-world applications, where contaminants seldomly occur individually. Despite the impressive advances in the development and application of mechanistic models to predict sediment toxicity, several critical research needs remain to be addressed. These needs and others represent the next frontier in the continuing development and application of mechanistic models for informing environmental scientists, managers, and decisions makers of the risks associated with contaminated sediments. Pharmacokinetics of Two Nanoemulsion Formulations of Δ8-Tetrahydrocannabinol in Rats.

The use of Δ8-tetrahydrocannabinol (Δ8-THC) has increased in recent years. Given that the oral absorption of cannabinoids in oil formulations is typically slow and variable, nanoemulsions may be an improved delivery vehicle. Therefore, we characterized the pharmacokinetics (PK) in Sprague-Dawley rats following the administration of three different oral formulations containing 10 mg/kg Δ8-THC: a translucent liquid nanoemulsion, a reconstituted powder nanoemulsion, and a medium chain triglyceride (MCT) oil solution for comparison. Δ8-THC was also administered intravenously at 0.6 mg/kg. Plasma samples were quantified for Δ8-THC and two metabolites, 11-hydroxy-Δ8-THC (11-OH-Δ8-THC) and 11-carboxy-Δ8-THC (COOH-Δ8-THC). Non-compartmental PK parameters were calculated, and a PK model was developed based on pooled data. Despite a smaller median droplet size of the translucent liquid nanoemulsion (26.9 nm) compared to the reconstituted powder nanoemulsion (168 nm), the PK was similar for both. The median Tmax values of Δ8-THC for the nanoemulsions (0.667 and 1 h) were significantly shorter than the median Tmax of Δ8-THC in MCT oil (6 h). This resulted in an approximately 4-fold higher Δ8-THC exposure over the first 4 h for the nanoemulsions relative to the MCT oil solution. The active 11-OH-Δ8-THC metabolite followed a similar pattern to Δ8-THC. The non-compartmental bioavailability estimates of Δ8-THC for the nanoemulsions (11-16.5%) were lower than for the MCT oil solution (>21.5%). However, a model-based analysis indicated similar bioavailability for all three oral formulations. These results demonstrate favorable absorption properties of both nanoemulsions, despite the difference in droplet sizes, compared to an MCT oil formulation.

Enhancing the Bioavailability of Silymarin Phytopharmaceutical by using Piperine and Fulvic Acid and Simultaneously Quantifying Three Biomarkers in Plasma

Background: Silymarin (SM) is a mixture of seven flavonolignans (FL) and one flavonoid extracted from the seeds of the milk thistle plant. The major FLs include silibinin, silychristin, silydianin, and isosilibinin. Silibinin, a mixture of two diastereomers, i.e., silybin A and silybin B, has been found to be the most active biomarker molecule in the extract of SM. Some studies have also demonstrated the usefulness of other biomarkers. However, most studies have characterized SM extract with respect to silibinin only. In our previous study, we isolated five different biomarkers (silychristin, silibinin, and isosilibinin (mixture of isosilybin A and B)) and denoted them as SM phytopharmaceuticals (PP). In other research, we have combined the SM PP with two bioavailability enhancers, i.e., Piperine (PIP) and fulvic acid (FA). The formulation containing 140 mg SM PP, 10% PIP and 20% FA was optimized using dissolution testing, and its enhanced permeability was demonstrated by ex-vivo gut sac study and Confocal laser scanning microscopy (CLSM). Methods: In this research, we have compared the bioavailability of SM suspension with SM PP and the optimized formulation in C57BL/6J mice. Furthermore, the bioavailability estimation was done for three biomarkers simultaneously, i.e., silychristin, silibinin, and isosilibinin. Results: Comparing the results for three different groups, it was found that the order of Cmax was F1&gt; SM PP&gt; SM suspension for all three biomarkers. For all the three biomarkers, the value of Cmax was at least two times more for F1 as compared to SM suspension. However, the Tmax remained constant i.e., 2 h for all the three groups and for all the three biomarkers. Furthermore, the clearance values were found to be in the order SM suspension &gt; SM PP &gt; F1. Conclusion: The PIP and FA present in this combination increased the clearance value and Cmax, respectively, of the formulation. Hence, this combination of SM PP with PIP and FA has shown a better pharmacokinetic profile than SM suspension, thus, it can serve as a better alternative for the treatment of liver disease.

Design-of-Experiment-Assisted Fabrication of Biodegradable Polymeric Nanoparticles: In Vitro Characterization, Biological Activity, and In Vivo Assessment.

Berberine (BER) is an alkaloid obtained from berberis plant having broad biological activities including anticancer. BER-encapsulated alginate (ALG)/chitosan (CHS) nanoparticles (BER-ALG/CHS-NPs) were developed for long-acting improved treatment in breast cancer. The surface of the NPs was activated by a conjugation reaction, and thereafter, the BER-ALG/CHS-NP surface was grafted with folic acid (BER-ALG/CHS-NPs-F) for specific targeting in breast cancer. BER-ALG/CHS-NPs-F was optimized by applying the Box-Behnken design using Expert design software. Moreover, formulations are extensively evaluated in vitro for biopharmaceutical performances and tested for cell viability, cellular uptake, and antioxidant activity. The comparative pharmacokinetic study of formulation and free BER was carried out in animals for estimation of bioavailability. The particle size recorded for the diluted sample using a Malvern Zetasizer was 240 ± 5.6 nm. The ζ-potential and the predicted % entrapment efficiency versus (vs) observed were +18 mV and 83.25 ± 2.3% vs 85 ± 3.5%. The high % drug release from the NPs was recorded. The analytical studies executed using infrared spectroscopy, differential scanning calorimetry, and X-ray diffraction expressed safe combinations of the components in the formulation and physical state of the drug revealed to be amorphous in the formulation. Cytotoxicity testing demonstrated that the formulation effectively lowered the cell viability and IC50 of the tested cell line in comparison to a raw drug. The cellular uptake of BER-ALG/CHS-NPs-F was 5.5-fold higher than that of BER-suspension. The antioxidant capacities of BER-ALG/CHS-NPs-F vs BER-suspension by the DPPH assay were measured to be 62.3 ± 2.5% vs 30 ± 6%, indicating good radical scavenging power of folate-conjugated NPs. The developed formulation showed a 4.4-fold improved oral bioavailability compared to BER-suspension. The hemolytic assay intimated <2% destruction of erythrocytes by the developed formulation. The observed experimental characterization results such as cytotoxicity, cellular uptake, antioxidant activity, and improved absorption suggested the effectiveness of BER-ALG/CHS-NPs-F toward breast cancer.

A multi-residue method for the analysis of organic pollutants released from atmospheric PM2.5 in simulated biological fluids: Inhalation bioaccessibility and bioavailability estimation

BackgroundIn recent decades, there has been a growing interest within the scientific community regarding the study of the fraction that could be released in simulated biological fluids to estimate in vitro bioaccessibility and bioavailability of compounds. Concerning particulate matter (PM), studies were essentially focused on metal (oid)s probably due to more complex methodologies needed for organic compounds, requiring extraction and pre-concentration steps from simulated fluids, followed by chromatographic analysis. Thus, the development of a simple and sensitive methodology for the analysis of multi-class organic compounds released in different inhalation simulated fluids would represent a great contribution to the field. ResultsIn this work, a methodology for the analysis of 49 organic pollutants, including 18 polycyclic aromatic hydrocarbons (PAHs), 12 phthalate esters (PAEs), 11 organophosphorus flame retardants (OPFRs), 6 synthetic musk compounds (SMCs) and 2 bisphenols released in simulated fluids from PM2.5 samples was developed. After a physiologically based extraction test (PBET) by using artificial lysosomal fluid (ALF) and a simulated body fluid (SBF, filling a dialysis membrane) to obtain in vitro inhalation bioaccessible and bioavailable fractions, respectively; compounds were determined by a vortex-assisted liquid-liquid extraction (VALLE) and a subsequent analysis by programmed temperature vaporization-gas chromatography-tandem mass spectrometry (PTV–GC–MS/MS). Experimental conditions concerning VALLE extraction (extraction time and amount of NaCl (g)) were optimized by using a central composite design (CCD), best MS/MS transitions were selected and matrix-matched calibration combined with use of labelled subrogate standards provided high sensitivity, minimization of matrix effects and recovering losses compensation. SignificanceThe successful validation results obtained for most of the compounds demonstrated the effectiveness of the proposed methodology for the analysis of multi-class organic pollutants released in ALF and SBF for inhalation bioaccessibility and bioavailability assessment, respectively. Furthermore, applicability of the method was proved by analysing 20 p.m.2.5 samples, being the proposed in vitro PBET dialyzability approach for assessing organic pollutant's inhalation bioavailability applied to PM2.5 samples for the first time.

Anticoronavirus Evaluation of Antimicrobial Diterpenoids: Application of New Ferruginol Analogues.

The abietane diterpene (+)-ferruginol (1), like other natural and semisynthetic abietanes, is distinguished for its interesting pharmacological properties such as antimicrobial activity, including antiviral. In this study, selected C18-functionalized semisynthetic abietanes prepared from the commercially available (+)-dehydroabietylamine or methyl dehydroabietate were tested in vitro against human coronavirus 229E (HCoV-229E). As a result, a new ferruginol analogue caused a relevant reduction in virus titer as well as the inhibition of a cytopathic effect. A toxicity prediction based on in silico analysis was also performed as well as an estimation of bioavailability. This work demonstrates the antimicrobial and specifically antiviral activity of two tested compounds, making these molecules interesting for the development of new antivirals.

Preclinical Bioavailability Assessment of a Poorly Water-Soluble Drug, HGR4113, Using a Stable Isotope Tracer.

Drug solubility limits intravenous dosing for poorly water-soluble medicines, which misrepresents their bioavailability estimation. The current study explored a method using a stable isotope tracer to assess the bioavailability of drugs that are poorly water-soluble. HGR4113 and its deuterated analog, HGR4113-d7, were tested as model drugs. To determine the level of HGR4113 and HGR4113-d7 in rat plasma, a bioanalytical method using LC-MS/MS was developed. The HGR4113-d7 was intravenously administered to rats that were orally pre-administered HGR4113 at different doses; subsequently, the plasma samples were collected. HGR4113 and HGR4113-d7 were simultaneously determined in the plasma samples, and bioavailability was calculated using plasma drug concentration values. The bioavailability of HGR4113 was 53.3% ± 19.5%, 56.9% ± 14.0%, and 67.8% ± 16.7% after oral dosages of 40, 80, and 160 mg/kg, respectively. By eliminating the differences in clearance between intravenous and oral dosages at different levels, acquired data showed that the current method reduced measurement errors in bioavailability when compared to the conventional approach. The present study suggests a prominent method for evaluating the bioavailability of drugs with poor aqueous solubility in preclinical studies.

Model Based Estimation of Posaconazole Tablet and Suspension Bioavailability in Hospitalized Children Using Real-World Therapeutic Drug Monitoring Data in Patients Receiving Intravenous and Oral Dosing.

Invasive fungal infections are a major cause of morbidity and mortality for immunocompromised patients. Posaconazole is approved for treatment and prophylaxis of invasive fungal infection in adult patients, with intravenous, oral suspension, and gastroresistant/delayed-released tablet formulations available. In Europe, until very recently, posaconazole was used off-label in children, although a new delayed-release suspension approved for pediatric use is expected to become available soon. A population pharmacokinetic model was developed which uses posaconazole therapeutic drug monitoring data following intravenous and oral dosing in hospitalized children, thus enabling estimation of pediatric suspension and tablet oral bioavailability. In total, 297 therapeutic drug monitoring plasma levels from 104 children were included in this analysis. The final model was a one-compartment model with first-order absorption and nonlinear elimination. Allometric scaling on clearance and volume of distribution was included a priori. Tablet bioavailability was estimated to be 66%. Suspension bioavailability was estimated to decrease with increasing doses, ranging from 3.8% to 32.2% in this study population. Additionally, concomitant use of proton pump-inhibitors was detected as a significant covariate, reducing suspension bioavailability by 41.0%. This is the first population pharmacokinetic study to model posaconazole data from hospitalized children following intravenous, tablet, and suspension dosing simultaneously. The incorporation of saturable posaconazole clearance into the model has been key to the credible joint estimation of tablet and suspension bioavailability. To aid rational posaconazole dosing in children, this model was used alongside published pharmacodynamic targets to predict the probability of target attainment using typical pediatric dosing regimen.

Determination of polyoxymethylene-water partition coefficients for diverse organophosphate esters (OPEs) and prediction of the free-dissolved OPEs in OPE-contaminated soil

Increasing attention on the estimation of bioavailability of organophosphate esters (OPEs) in soil or sediment has urged the development of techniques to measure soil−/sediment-associated porewater concentrations of OPEs. In this study, we investigated the sorption dynamics of 8 OPEs to polyoxymethylene (POM) spanning one order of magnitude of aqueous OPE concentrations and proposed POM-water partitioning coefficients (Kpom/w) for OPEs. The results showed that the Kpom/w values were mainly affected by the hydrophobicity of OPEs. OPEs with high solubility preferentially partitioned into the aqueous phase indicated by the low log Kpom/w values; while lipophilic OPEs were observed to be taken up by POM. The concentration of lipophilic OPEs in the aqueous phase had a strong impact on their sorption dynamics on POM, with higher aqueous concentrations accelerating the sorption dynamics and shortening the time for equilibration. We proposed that the required time to reach equilibration for targeted OPEs should be 42 d. The proposed equilibration time and Kpom/w values were further validated by applying POM to soil artificially contaminated with OPEs to measure OPEs soil-water partitioning coefficients (Ks). The variations of Ks among soil types implied the need to elucidate the effects of soil properties and chemical properties of OPEs on their distribution between soil and water in the future.

591. Adintrevimab (ADI) Population Pharmacokinetics (PPK) in Phase 1 and Phase 2/3 COVID-19 Prevention and Treatment Study Participants

Abstract Background ADI is a fully human IgG1 monoclonal antibody engineered to have an extended half-life with high potency and broad neutralization against SARS-CoV-2 and other SARS-like coronaviruses with pandemic potential. Our objective was to develop a PPK model that describes the serum ADI concentration-time profile following intravenous (IV) and intramuscular (IM) administration. Methods The ADI PPK model was developed on PK data from a Phase 1 single-ascending dose study (24 adults, IV and IM) and from Phase 2/3 COVID-19 prevention (EVADE; 659 adults, IM) and treatment (STAMP; 189 adults, IM) studies. 1,486 PK samples were included in the analysis. The impact of covariates (e.g. body weight, age, and baseline viral load) on ADI serum disposition were evaluated. Prediction-corrected visual predictive check (PC-VPC) plots were used to qualify the PPK model. Participant-specific ADI exposure estimates were generated using individual post hoc PK parameters. Results The PPK model is comprised of 2 systemic compartments, zero-order infusion for IV administration and first-order absorption for IM administration and provided a robust fit to the data based on the PC-VPC plots and goodness-of-fit plots (data not shown). Body weight influenced clearance, intercompartmental clearance, and central and peripheral volume compartments. The relationship between body weight and clearance was not suggestive of the need for dose adjustment over the population weight range studied (38.6 to 178.7 kg). There was no apparent impact of baseline viral load or age on ADI clearance. The median [range] half-lives in days by study; Phase 1 (α1.71 [1.18-2.46]; β 125 [117-149]), Phase 2/3 prevention (α 1.86 [0.640-3.13]; β 136 [105-209]), and Phase 2/3 treatment (α 1.89 [0.631-3.01]; β 136 [108-206]). The population mean IM bioavailability estimate was 90.5%. The figure shows the PPK model median (90% confidence interval) concentration-time profile following a single 300 mg IM ADI dose by study. Conclusion The PPK model provided a precise and unbiased fit to the observed ADI concentration-time data and will be useful for future PK-pharmacodynamic analyses. Moreover, ADI demonstrated high IM bioavailability and a median terminal elimination half-life of 125 to 136 days. Disclosures Christopher M.
