Impact of different severity hyperglycemia on erythrocyte rheological properties1.

The triad "insulin resistance, prediabetes, diabetes" is three independent neologies with characteristic features and development. In addition, each are characterized by progression and the possibility of transition from one form to other. Due to the fact that diabetes is one of the common diseases associated with high rates of disability, it is necessary to improve diagnostic methods and educational regimens for successful prevention and treatment of the disease. We investigated Band 3 protein (B3p) level, osmotic resistance of erythrocytes, the total antioxidant activity (TAA) of blood serum, level of HbA1 in group patients with insulin resistance (IR), prediabetes, and Type 2 Diabetes Mellitus (T2DM) and comparative with health control group. We used original, accurate research methods that measure the essence of the studied quantities. Disruptions of glucose and insulin homeostasis ay lead to the initiation of oxidative stress (in our study demonstrated by a decrease of TAA of blood serum) increased redox-sensitive PTP activity and aberrant band 3 phosphorylation, potentially leading to reduced erythrocyte deformability. At the same time glycation of Hb during T2DM may affect its cross-link with membrane proteins, in particular with B3p, and although appears to limit its cross-linking and decrease its clusterization ability, induces alterations in the cytoskeletal matrix, and thereby decrease erythrocytes' osmotic resistance making them more susceptible to hemolysis. The osmotic resistance of the erythrocytes can be used as a sensitive marker for the detection of the early stages of hyperglycemia (prediabetes). This set of clinical trials will make it possible to identify diseases that make up the triad at an early stage. Early detection of disorders and continued research in this direction will help in the development of a diagnostic scheme for the prevention of such patients. Based on our data, research into anti-oxidation drugs is very important. With the help of the array of studies described in the article and antioxidant treatment, the likelihood of successful treatment will increase.

ВЗАИМОСВЯЗЬ ПОКАЗАТЕЛЕЙ НАПРЯЖЕННОСТИ СИМПАТОАДРЕНАЛОВОЙ СИСТЕМЫ У ПАЦИЕНТОВ С ФИБРИЛЛЯЦИЕЙ ПРЕДСЕРДИЙ ДО И ПОСЛЕ ХИРУРГИЧЕСКОГО ЛЕЧЕНИЯ

Introduction. It is known that cardiovascular diseases, including cardiac arrhythmias, are accompanied by increased activation of the sympathetic-adrenal system (SAS). Hyperactivation of the SAS causes an increase in the level of circulating catecholamines - adrenaline and norepinephrine, resulting in desensitization of β-adrenergic receptors (β-AR), aggravation of myocardial contractile dysfunction and the development of arrhythmia. Indicators of SAS tension are β-adrenergic reactivity of erythrocyte membranes (β-ARM) and the level of circulating catecholamines. One of the most common arrhythmias is atrial fibrillation (AF), which leads to a decrease in the quality of life, disability and high mortality. Therefore, among patients with AF, it is important to study the functional viability of β-AR in different periods of the disease. Purpose: to evaluate β-ARM and the level of adrenaline, norepinephrine in patients with AF before and after surgical treatment. Materials and methods. The sample included 47 patients - 31 men and 16 women, the age in the sample was 51 (26; 77) years. All patients were diagnosed with AF based on ECG and 24-hour monitoring. Among the entire sample, 59.6% of patients had paroxysmal, 25.5% persistent, 14.9% long-term persistent forms of AF. All patients underwent surgical treatment of AF using radiofrequency or cryoablation. β-ARM was determined by the method of assessing changes in erythrocyte osmotic resistance as a result of blockade of β-AR in vitro with a selective β-adrenergic blocker (Β-ARM, AGAT, Russia). Plasma epinephrine and norepinephrine concentrations were determined by enzyme-linked immunosorbent assay using Adrenalin ELISA and Noradrenalin ELISA kits (Tecan IBL International, Germany). The parameters were studied in all patients before treatment, in 44 patients after 3 days, in 18 patients after 3 months and in 9 patients after 12 months after ablation. The significance of changes in β-ARM and catecholamine levels before and after ablation was assessed using the Wilcoxon test. The strength of the linear relationship between quantitative indicators was assessed using the Spearman rank correlation coefficient. Results. In the study sample, there were no statistically significant changes in β-ARM and adrenaline levels before and at different times after ablation. Thus, the level of β-ARM before surgery was 18.2 (12.6; 25.4), after 3 days - 22.3 (14.4; 26.6), after 3 months - 20.3 (9.3 ; 29.3), and 12 months after the operation - 27.7 (16.8; 36.7). The level of adrenaline before surgery, 3 days, 3 months and 12 months after surgery was 4.4 (3.7; 6.9), 4.7 (3.0; 6.4), 4.6 (2 .5; 6.6), 5.6 (9.1; 8.8). However, an increase in the level of norepinephrine was found 12 months after ablation compared with the values before treatment and 3 days after treatment by 1.7 and 1.8 times, respectively (p = 0.028). The level of norepinephrine before surgery, 3 days, 3 months and 12 months after surgery was 40.9 (18.7; 56.6), 38.6 (14.8; 49.6), 46.7 (25 .6; 57.3), 69.3 (65.1; 80.1). Before surgery, β-ARM and norepinephrine levels were linearly correlated (r = 0.335, p = 0.037), but after treatment, the linear correlation disappeared. At 3 and 12 months after ablation, there was a strong linear correlation between adrenaline and norepinephrine levels (r = 0.611, p = 0.012; r = 0.786, p = 0.036, respectively). Conclusion. In the sample of patients with AF, β-ARM and norepinephrine levels were linearly correlated before treatment, but after ablation, the correlation disappeared. At the same time, the values of β-ARM do not change statistically significantly, and the level of norepinephrine increases during a 12-month follow-up. Thus, 12 months after treatment in patients with AF, the activity of the CAS increases, but the functional viability of beta-AR does not change significantly.

Effect of Non-Modified as Well as Surface-Modified SiO2 Nanoparticles on Red Blood Cells, Biological and Model Membranes.

Nanoparticles are extremely promising components that are used in diagnostics and medical therapies. Among them, silica nanoparticles are ultrafine materials that, due to their unique physicochemical properties, have already been used in biomedicine, for instance, in cancer therapy. The aim of this study was to investigate the cytotoxicity of three types of nanoparticles (SiO2, SiO2-SH, and SiO2-COOH) in relation to red blood cells, as well as the impact of silicon dioxide nanoparticles on biological membranes and liposome models of membranes. The results obtained prove that hemolytic toxicity depends on the concentration of nanoparticles and the incubation period. Silica nanoparticles have a marginal impact on the changes in the osmotic resistance of erythrocytes, except for SiO2-COOH, which, similarly to SiO2 and SiO2-SH, changes the shape of erythrocytes from discocytes mainly towards echinocytes. What is more, nanosilica has an impact on the change in fluidity of biological and model membranes. The research gives a new view of the practical possibilities for the use of large-grain nanoparticles in biomedicine.

Effect of Dietary Phosphate Deprivation on Red Blood Cell Parameters of Periparturient Dairy Cows.

Postparturient hemoglobinuria is a sporadic disease characterized by intravascular hemolysis and hemoglobinuria in early lactating dairy cows. The condition has empirically been associated with phosphorus (P) deficiency or hypophosphatemia; however, the exact etiology remains obscure. This paper summarizes two controlled studies investigating the effect of P deprivation during the transition period. In Study I, 36 late pregnant dairy cows were randomly assigned to either a diet with low, or adequate, P content from four weeks before calving to four weeks after calving. In Study II, 30 late pregnant dairy cows were again assigned to either a diet with low, or adequate, P for the last four weeks before calving only. Pronounced hypophosphatemia developed during periods of restricted P supply. In early lactation, a subtle decline of the red blood cell count occurred independently of the dietary P supply. In Study I, anemia developed in 11 cows on deficient P supply, which was associated with hemoglobinuria in five cases. Neither erythrocyte total P content nor osmotic resistance of erythrocytes were altered by dietary P deprivation. Restricted dietary P supply, particularly in early lactation, may lead to postparturient hemoglobinuria, but more frequently causes clinically inapparent hemolysis and anemia in cows.

The aim of the work is to study the osmotic stability and morpho-functional features of peripheral blood erythrocytes of patients with chronic fatigue syndrome depending on the level of tolerance to physical load (TPL) and associated risk factors. groups of factors that may be the cause, so the development of measures to eliminate them is an urgent problem today, which requires the development of effective ways to correct chronic fatigue syndrome. It is known that the peripheral part of erythron actively responds to changes that occur in the whole body after the action of various factors, including those factors that lead to chronic fatigue syndrome (CFS). Material and Methods of Research The examination was performed on the basis of the Department of Vascular Neurology of the Central Clinical Hospital of Ivano-Frankivsk. The study involved 30 patients aged 55-65 years (mean age 60.6 + 1.2 years), who were divided into 3 groups: 1 gr. included 10 patients with chronic fatigue syndrome and a high level of exercise tolerance. The 2nd group included 10 patients with chronic fatigue syndrome and the average level of exercise tolerance, the composition of 3 gr. included 10 patients with chronic fatigue syndrome associated cardiovascular pathology (angina pectoris, hypertension) and low exercise tolerance (3 gr.). Results It is established that chronic fatigue syndrome proceeds in 3 phases and has natural stages of development which are characterized by three groups of etiological factors and the corresponding levels of teletransitivity to physical activity, each of which corresponds to a certain erythrocyte profile and level of osmotic stability of erythrocytes which are offered to use as prognostic and diagnostic characteristics. chronic fatigue syndrome. Conclusions The study of the quantitative composition of peripheral blood erythrocytes and hemoglobin, as well as their ratio (color index) in patients with chronic fatigue syndrome on the background of low levels of exercise tolerance revealed a decrease in erythrocytes and hemoglobin by 42% and 25%, respectively, indicating the presence of anemic hypoxia. 2. In conditions of chronic fatigue there is a decrease in osmotic resistance of erythrocytes, as evidenced by a decrease in the number of osmotically stable erythrocytes with a gradual decrease in the concentration of NaCl solution (3.0%; 0.5%; 0.46%; 0.3%). 3. In chronic fatigue there is a deterioration of membranes in peripheral blood erythrocytes, as evidenced by a decrease in their osmotic stability and an increase in the number of irreversibly altered forms of РВЕ, as well as a decrease in peripheral blood erythrocytes, which is closely correlated with decreased tolerance to exercise.

The purpose of this paper was to study erythrocyte osmotic resistance (EOR) and its relationship with the quantitative and morphofunctional parameters of red blood cells in young women with limited physical activity depending on the I/D polymorphism of the ACE gene. Materials and methods. The study involved 200 healthy women aged 18–22 years leading a sedentary lifestyle. Their blood samples were taken for genotyping and to determine the haemoglobin (HGB) level and red blood cell count (RBC), their mean corpuscular volume (MCV), mean corpuscular haemoglobin (MCH) and mean corpuscular haemoglobin concentration (MCHC) as well as haematocrit (HCT). EOR was studied by measuring the optical density of haemoglobin solutions after erythrocyte destruction in a series of hypotonic NaCl solutions with the concentration decreasing from 0.8 % to 0.1 %. Based on the results of osmotic haemolysis, erythrograms were produced and the NaCl concentrations were determined, at which 10, 50 and 90 % of the red blood cells were destroyed. Results. Carriers of the DD genotype were characterized by the lowest resistance to haemolysis in all three erythrocyte populations. According to the analysis of factor structures, in girls with the DD genotype only relationships between the total and individual parameters of erythrocytes were established. In carriers of the I allele (ID and II genotypes) the dominant factor combined the total red blood cell parameters (RBC, HGB, HCT) and the EOR of old, mature, and young erythrocytes. Thus, carriers of the I allele (ID and II genotypes) of the ACE gene are characterized by a higher resistance of mature and old cell populations than those of the DD genotype, while EOR variations in the former are interconnected with changes in the quantitative parameters of red blood cells. At the same time, an increase in EOR is combined with an increase in erythrocyte functional activity. DD genotype carriers showed no correlations between EOR and the quantitative and morphofunctional parameters of red blood cells.

Protection of Erythrocytes and Microvascular Endothelial Cells against Oxidative Damage by Fragaria vesca L. and Rubus idaeus L. Leaves Extracts-The Mechanism of Action.

The aim of this work is to determine the biological activity of ellagitannins rich extracts from leaves of raspberry (Rubus idaeus L.) and wild strawberry (Fragaria vesca L.) in relation to cells and cell membranes. Detailed qualitative and quantitative analysis of phenolic compounds of the extract was made using chromatographic methods. Cytotoxic and antioxidant activities of tested extracts in relation to erythrocytes and human vascular endothelial cells (HMEC-1) were determined by using fluorimetric and spectrophotometric methods. In order to establish the influence of the extracts on the physical properties of the membrane, such as osmotic resistance and erythrocytes shapes, mobility and/or hydration of polar heads and fluidity of hydrocarbon chains of membrane lipids, microscopic and spectroscopic methods were used. The results showed that the extracts are non-toxic for erythrocytes and HMEC-1 cells (up to concentration of 50 µg/mL), but they effectively protect cells and their membranes against oxidative damage. The increase in osmotic resistance of erythrocytes, formation of echinocytes and changes only in the polar part of the membrane caused by the extracts demonstrate their location mainly in the hydrophilic part of the membrane. The results indicate that tested extracts have high biological activities and may be potentially used in delaying the ageing process of organisms and prevention of many diseases, especially those associated with oxidative stress.

Osmotic Resistance of Rat’s Erythrocytes After Local Cold Injury

The effect of local cold injury in rats on the osmotic resistance of erythrocytes in the early and late reactive terms was investigated. Cryoinfluence was performed by pressing the applicator to the skin on the lateral surface of the rat thigh for 30, 60 and 120 s. Afterwards, the blood was collected from the aorta of animals after 1 and 4 hours and after 24 and 48 hours (early and late reactive periods, respectively). The effect of hypotonic solutions on cells was used as a test for osmotic resistance of erythrocytes. It is shown that the osmotic resistance of erythrocytes in the early reactive term increases (compared with control cells). In the late reactive term, the indices of osmotic resistance of erythrocytes after 24 hours approached the control values and after 48 hours they reached the norm. Osmotic resistance of erythrocytes in both reactive terms after local cold injury does not depend on the duration of cryoinfluence (30, 60, 120 s). The findings are considered from the standpoint of adaptive reactions that have an allostatic nature.

Effect of the water-soluble cholesterol-methyl-β-cyclodextrin complex on the development of pulmonary edema with an analysis of the osmotic resistance of erythrocytes in rats

Introduction. Methyl-β-cyclodextrin is a potent acceptor of cellular membrane cholesterol and, at the same time, used as a solubilizer, which makes it a potential target for targeted delivery of hydrophobic compounds.Aim. To assess the effect of administration of methyl-β-cyclodextrin containing cholesterol on the degree of lung hydration and osmotic resistance of erythrocytes against the background of pulmonary edema in adult rats caused by the administration of mezaton.Materials and methods. We investigated the degree of hydration of the lungs by the value of the pulmonary coefficient and their dry residue. Osmotic resistance of erythrocytes were assessed by the intensity of their hemolysis in a series of hypotonic sodium chloride solutions in adult male Wistar rats (220±40 g) of the control group (intact, n=10) and two experimental groups with pulmonary edema caused by the administration of mezaton (n=10) and with the subsequent introduction of a water-soluble complex cholesterol-methyl-β-cyclodextrin against the background of the development of pulmonary edema (n=10). We used additional criteria for assessing the osmotic resistance of erythrocytes: minimum resistance – the concentration of sodium chloride solution, at which the first “weak” erythrocytes begin to hemolyze; maximum resistance is the concentration of sodium chloride solution at which all or almost all erythrocytes hemolyzed.Results. The introduction of mezaton led to the development of pronounced hemolysis and hydration of the lungs with the development of their edema and a narrowing of the range of minimum and maximum osmotic resistance of erythrocytes. The introduction of a complex of cholesterol with methyl-β-cyclodextrin after edemogenic exposure led to a decrease in the severity of pulmonary edema, determined by a decrease in the pulmonary coefficient and an increase in the dry residue, and to an increase in the osmotic resistance of erythrocytes with an expansion of the range of their minimum and maximum osmotic resistance.Conclusion. The introduction of a water-soluble complex of cholesterol with methyl-β-cyclodextrin against the background of the development of hemodynamic pulmonary edema significantly reduced the degree of hydration of the lungs, which combined with an increase in the osmotic resistance of erythrocytes.

Effects of biologically active substances from brown algae on the resistance of erythrocyte membranes

A study of different modes of administration of bioactive substances (BAS) of algae F. vesiculosus and S. latissima to laboratory animals allowed the authors to establish the doses and duration of administration, at which the complexes under study show moderate cytoprotective activity (15 and 30 mg/kg for S. latissima, 60 and 120 mg/kg for F. vesiculosus; three-day administration). It was found that the complex of BAS F. vesiculosus is characterized by a higher activity with respect to erythrocyte osmotic resistance compared to the complex of BAS S. latissima. At the same time, the complex of BAS laminaria is characterized by a higher activity with respect to erythrocyte peroxide resistance. The above features emphasize the feasibility of combined use of these extracts. At three-day combined administration of complexes of BAS F. vesiculosus and S. latissima at doses of 60 and 15 mg/kg, respectively, their high membrane-protective and antioxidant activity was determined. However, a higher dose or longer administration of the BAS complexes negatively affects the resistance of erythrocyte membranes.

Beneficial Effect of Quercetin on Erythrocyte Properties in Type 2 Diabetic Rats.

Diabetes mellitus is characterized by tissue oxidative damage and impaired microcirculation, as well as worsened erythrocyte properties. Measurements of erythrocyte deformability together with determination of nitric oxide (NO) production and osmotic resistance were used for the characterization of erythrocyte functionality in lean (control) and obese Zucker diabetic fatty (ZDF) rats of two age categories. Obese ZDF rats correspond to prediabetic (younger) and diabetic (older) animals. As antioxidants were suggested to protect erythrocytes, we also investigated the potential effect of quercetin (20 mg/kg/day for 6 weeks). Erythrocyte deformability was determined by the filtration method and NO production using DAF-2DA fluorescence. For erythrocyte osmotic resistance, we used hemolytic assay. Erythrocyte deformability and NO production deteriorated during aging—both were lower in older ZDF rats than in younger ones. Three-way ANOVA indicates improved erythrocyte deformability after quercetin treatment in older obese ZDF rats only, as it was not modified or deteriorated in both (lean and obese) younger and older lean animals. NO production by erythrocytes increased post treatment in all experimental groups. Our study indicates the potential benefit of quercetin treatment on erythrocyte properties in condition of diabetes mellitus. In addition, our results suggest potential age-dependency of quercetin effects in diabetes that deserve additional research.

Influence of cardiopulmonary bypass on the erythrocyte membranes and the method of its protection

The damage to erythrocytes during cardiopulmonary bypass (CPB) remains a recent problem. The aim of this research was to study the effect of fructose-1,6-diphosphate on the state of the erythrocyte membrane during CPB and the level of phosphorus in blood as a marker of the energy potential in the cell. Patients were divided into two groups. The control group 1 (Gr 1) consisted of 75 individuals. The group 2 (Gr 2) included patients to whom fructose-1,6-diphosphate (FDP) was administrated according to the developed scheme as follows 10 g of the drug was diluted in 50 ml of a solvent, 5 g of the drug was injected intravenously with the use of perfusor immediately before initiation of CPB at a rate of 10 ml/min and 5 g at the 30th minute of CPB (before the stage of warming) the same way. When comparing two groups the best results in hemolysis (p<0.01), mechanical (p<0.01). osmotic resistance of erythrocytes (p<0.01), the time of acid hemolysis (p<0.01) and the permeability of the erythrocyte membrane in postperfusion period were in Gr 2. Вefore cardiac surgery hypophosphatemia was detected in 18% out of 150 and in 32% out of 150 patients – a lower limit of normal phosphorus content in the blood. After CPB in Gr 1 phosphorus content in blood was 0.85±0.32 mmol/l and hypophosphatemia was in 53% out of 75 patients. This indicates a pronounced energy deficit in this group. In Gr 2 phosphorus level was 1.7±0.31 mmol/l and there was no hypophosphatemia. As a result, FDP as an endogenous high-energy intermediate metabolite of the glycolytic pathway leads to resistance to hemolysis, protects the erythrocyte membrane from damage and increases the energy potential of the cell during CPB.

Comparison of Osmotic Resistance, Shape and Transmembrane Potential of Erythrocytes Collected from Healthy and Fed with High Fat-Carbohydrates Diet (HF-CD) Pigs-Protective Effect of Cistus incanus L. Extracts.

Long-term high fat-carbohydrates diet (HF-CD) contributes to the formation of irreversible changes in the organism that lead to the emergence of civilization diseases. In this study, the impact of three-month high-fat diet on the physical properties of erythrocytes (RBCs) was studied. Furthermore, the biological activity of Cistus incanus L. extracts, plant known with high pro-health potential, in relation to normal and HF-CD RBCs, was determined. Obtained results have shown that, applied HF-CD modified shape, membrane potential and osmotic resistance of erythrocytes causing changes in membrane lipid composition and the distribution of lipids. The impact of HF-CD on physical properties of RBCs along with atherosclerotic lesions of the artery was visible, despite the lack of statistically significant changes in blood morphology and plasma lipid profile. This suggests that erythrocytes may be good markers of obesity-related diseases. The studies of biological activity of Cistus incanus L. extracts have demonstrated that they may ameliorate the effect of HF-CD on erythrocytes through the membrane-modifying and antioxidant activity.

Use of modulation interference microscopy in applied immunology

Objective: to determine the osmotic resistance of red blood cells using modulation interference microscopy technologies in the light microscopy mode of biological objects to identify the dynamics and determine the possibilities for continuing apitherapy in patients with autoimmune diseases. Methods. Methodological approaches to the use of modulation interference microscopy and computed tomography for the tasks of diagnostic medicine and applied immunology are described. The technology of vital computer dynamic phase metering, special methods for sample preparation of cytoobjects, as well as a computer-aided automated analysis of cytological images were used; recognition algorithms, measurement and identification of micro-objects; methods of statistical data processing. Results. Using the domestic innovative laser microscope MIM340, the osmotic resistance of erythrocytes was estimated using the method of modulation interference microscopy to identify the dynamics and determine the possibilities for continuing apitherapy in patients with rheumatoid arthritis and multiple sclerosis. Using computer methods of cytodiagnostics, new aspects of the functional morphology of living cells were revealed, clinical and morphological parallels were established. It was possible to evaluate the diagnostic and prognostic value of vital cell morphometry in various pathological processes and assess the effectiveness of therapeutic measures. A data bank of graphic images of red blood cells and blood lymphocytes of patients with diseases of the immune system has been created. Findings. The study of the structural features and functional usefulness of circulating blood cells is of great importance in addressing the pathogenesis, diagnosis, assessment of the severity of various pathological conditions and the effectiveness of the therapy. We believe that the study of living cytoobjects using the new method of coherent phase microscopy will provide the most objective data and increase the information content of the analysis, which is undoubtedly an urgent and promising task. The immediate plans include the refinement of mathematical, algorithmic, and software to support decision-making in computer-aided analysis of images of the epidermis and the surface of the dermis in neoplastic processes - malignant skin diseases. It is also necessary to create algorithmic and software tools for computerizing studies of cell models for quantitative and qualitative assessment of the selective accumulation of xenobiotics by laser microscopy.

MEMBRANE-RECEPTOR COMPLEX DYSFUNCTION AS A NEW TARGET FOR THE NEUROTROPHIC THERAPY OF ISCHEMIC STROKE

The aim of the study was the β-adrenergic activity of peripheral blood erythrocyte membranes in the acute period of ischemic stroke (IS). Peripheral blood erythrocytes are the most informative biological tissue for studying many pathogenetic mechanisms, and the work of their membrane-receptor complex (MRC) can very well reflect the functional state of the whole organism, one of the most important criteria for assessing is the adrenergic activity of cytoplasmic membranes in the pathogenesis of IS. This study solves the problem of the finding of the features of changes in the adrenoceptors system in the acute period of IS in the dynamics of the treatment of humans cryopreserved cord blood serum (CCBS). The main scientific results: the obtained results indicate a reduced ability of adrenoceptors (ARs) on erythrocyte membranes to bind blockers due to the desensitisation effect, which is observed from the 1st day of the disease. In the onset of IS, there is a significant increase in the values of β-adrenergic activity of membranes (β-ARM) of erythrocytes in 2.4 times compared with the control. Maximum levels of the indicator (42.43±3.64 CU) are observed in patients with initially severe disease, which indicates significant stress of the sympathoadrenal system (SAS) in these patients. On the 10th day of the disease, there was a decrease in β-ARM relative to the values obtained during hospitalisation. Still, comparing the value of the indicator on the 10th day of IS in both groups, there was an apparent, more significant decrease in β-ARM in the 2nd group of patients, which additionally received CCBS. The use of CCBS contributes to a more pronounced degree of recovery in neurological functions (6.9%) and faster stabilisation of the structure of MRCs in erythrocytes in the form of reducing the values of β-ARM to 16.61±2.86 CU (p<0.05). The area of practical use of the research results: the results of the study can be used to diagnose and treat of the IS. An innovative technological product: A comprehensive approach to assess the β-ARM of peripheral blood erythrocytes in patients with IS by determining the degree of changes in osmotic resistance of erythrocytes (ORE) under the action of adrenergic drugs (AD) is informative and can be used to diagnose of IS. The use of neurotrophic factors is a potentially new method of treating this pathology. The osmotic resistance of erythrocytes and the content of hemoglobin in the blood of carp (Cyprinus carpio L.) and pike (Esox lucius L.) under the action of Fe3+ ions at concentrations of 0.2 and 0.5 mg/dm3, were under study which corresponded to 2 and 5 maximum permissible concentration (MPC). The research demonstrated that increased concentrations of Fe3+ ions lead to changes in the osmotic resistance of erythrocytes and the content of hemoglobin in the blood of carp and pike. The findings suggest that high concentrations of Iron (5 MPC) in carp lead to a decrease in the resistance of erythrocyte membranes, and in pike it led to increased osmotic resistance of erythrocytes, which is apparently due to different mechanisms of adaptation of fish erythrocyte membranes to adverse aquatic environment. Species-specificity is also characterized by changes in the content of hemoglobin in the blood of fish species which were researched. An increased amount of pigment in the blood of carp and a decreased amount of hemoglobin in pike under the action of 5 MPC of metal ions was established. The obtained results can be used to assess the physiological condition of fish and water quality under conditions of contamination of watercourses with metals.

BIOPHYSICAL MARKERS OF ISCHEMIC STROKE

An important and influential factor that directly affects the severity of ischemic stroke (IS) and determines its outcome is the functional state of the membrane-receptor complex (MRC) of cells. One of the most important criteria for assessing this parameter is the β‑adrenergic activity of cytoplasmic membranes (β‑ARM), which plays a leading role in the pathogenesis of IS. The article presents the results of a comprehensive study of the peculiarities of changes in the structural and functional characteristics of brain tissue and β‑adrenoceptors in the acute period of IS to optimize diagnosis. Measurement of changes in the osmotic resistance of erythrocytes (ORE) under the action of β‑blockers was determined by photoelectron colorimetry. The work was based on the materials of a comprehensive examination of 350 patients with the new-onset IS on the 1st, 10th and 21st day of the disease. The severity of the condition and the degree of neurological deficit were objectified using the stroke scale of the National Institutes of Health (NIHSS) with a score in the first hours of the disease, in the dynamics of treatment on the 10th and 21st day. All patients were divided into 2 clinical groups: 1st (n = 183) – patients in moderate severity condition (mean score on the NIHSS scale 11.74 ± 0.33); 2nd (n = 167) – patients in severe condition (mean score on the NIHSS scale 24.06 ± 0.29). As a result of the study, the indicators of β‑ARM of the control group were within normal limits (15.3 ± 4.4 SU), which corresponds to normal β‑ARM. In patients with moderate IS, the indicator exceeded the control values by 1.97 times, which is typical for the average degree of β‑ARM (21–40 SU), and in severe – 2.8 times and was characterized by a low degree of β‑ARM (> 41 SU). An unfavorable sign for the prognosis of the acute period of IS was represented by a further increase in the levels of β‑ARM in the dynamics, which was observed in clinically severe patients.
