Studying mass spectrometric behaviors of {Au6 Ag2 (C)[PPh2 (4-CH3 -Py)]6 }(BF4 )4 and {Au8 [(PPh3 )2 O]3 (PPh3 )2 }(NO3 )2 by electrospray time-of-flight mass spectrometry and electrospray ion trap mass spectrometry.

Mass spectrometry (MS) has been recognized as a powerful technique to detect accurate chemical information about metal clusters. Maintaining metal clusters intact, which is a great challenge in MS analysis, was achieved in this work by choosing a suitable mass analyzer and carefully optimizing analysis parameters. Electrospray ionization time-of-flight mass spectrometry (ESI-TOF-MS) and electrospray ion trap mass spectrometry (ESI-IT-MS) were applied to characterize the synthesized ligand-protected metal clusters [Au6 Ag2 (C)(L(1) )6 ](BF4 )4 (L(1) =2-diphenylphosphanyl-4-methylpyridine) (1) and [Au8 (L(2) )3 (L(3) )2 ](NO3 )2 (L(2) =bis(2-diphenylphosphinophenyl)ether, L(3) =triphenyl-phosphane) (2). Three kinds of buffer gas (helium, mass: 2; nitrogen, mass: 28; argon, mass: 40) and various radiofrequency (RF) amplitudes (from 70 to 330) were chosen to study the fragmentation rate during the "collision cooling" process in the ion trap analyzer. In the ESI-TOF-MS analysis, metal clusters 1 and 2 were mainly observed as intact clusters, which were Au6 Ag2 (C)(L(1) )6 (BF4 )2 (2+) , Au6 Ag2 (C)(L(1) )6 (BF4 )(3+) , Au6 Ag2 (C)(L(1) )6 (4+) for 1 and Au8 (L(2) )3 (L(3) )2 (2+) for 2. While, in the ESI-IT-MS analysis, only fragments could be found, such as Au6 Ag(C)(L(1) )6 (BF4 )(2+) , Au6 (C)(L(1) )6 (2+) , Au5 Ag(C)(L(1) )4 (2+) , Au6 Ag(C)(L(1) )6 (3+) , Au(L(1) )(+) for 1 and Au8 (L(2) )3 (L(3) )(2+) , Au8 (L(2) )3 (2+) , Au6 (L(2) )3 (2+) for 2. It is obvious that the two kinds of mass analyzers caused different MS behaviors of metal clusters. In the ion trap (IT) mass analyzer, particularly, "collision cooling" was contributing to further dissociation of fragile compounds, in which a higher RF amplitude and a larger mass buffer gas led to more fragmentation. In this work, intact metal clusters were obtained in ESI-TOF-MS, instead of ESI-IT-MS, in which the "collision cooling" process caused more cluster dissociation. It was concluded that the analyzer in ESI-TOF-MS is "softer" than that in ESI-IT-MS for metal clusters. Copyright © 2016 John Wiley & Sons, Ltd.

UV fragmentation and ultrafast dynamics of trinuclear silver/1-methylthymine and silver/1-methyluracil metal-base pairs in an ion trap

We report on gas phase UV action spectroscopy and photodynamics of [Ag3(1MT-H/1MU-H)2]+ comprised of a linear silver string and two deprotonated 1-methyl-thymine/uracil (1MT/1MU) ligands. We applied pump–probe femtosecond laser photofragmentation in an electrospray ion trap mass spectrometer and high-level ab initio calculations at the level of approximate coupled-cluster singles-doubles theory. The experimental UV band at 283/275nm is assigned to a red shifted 1ππ∗ nucleobase located transition. Relaxation of the 1ππ∗ state occurs with time constants of 0.2/1.1ps and 0.2/4.2ps for the 1MT and 1MU complexes, respectively, on a similar ultrafast time scale as non-metalated uracil derivatives.

Ion-pairing liquid chromatography with on-line electrospray ion trap mass spectrometry for the structural analysis of N-unsubstituted heparin/heparan sulfate.

The rare N-unsubstituted glucosamine (GlcNH3(+)) residues in heparan sulfate (HS) have important biological and pathophysiological roles. In this study, a high-resolution method for the separation and analysis of N-unsubstituted disaccharides of heparin/HS is described. Four N-unsubstituted disaccharides, together with eight N-substituted species, can be well-separated by ion-pair reverse-phase ultra-performance liquid chromatography. Each disaccharide can then be detected and its relative abundance quantified using electrospray ionization mass spectrometry in the negative mode. Because of its high sensitivity, without interference from proteins and other sample impurities, this method is particularly useful in the analysis of low content GlcNH3(+) residues in small amounts of biological materials, eg. sera, tissue and cell culture-derived samples. This would lead to a better understanding of the biological origin of GlcNH3(+) residues and their increasingly important function in human health and disease.

Production of Angiotensin-I-Converting-Enzyme-Inhibitory Peptides in Fermented Milks (Lassi) Fermented by Lactobacillus acidophillus with Consideration of Incubation Period and Simmering Treatment

The lassi, fermented milks product containing angiotensin-I-converting-enzyme (ACE)-inhibitory peptides were produced by using selected Lactobacillus acidophilus NCDC-15 and the incubation period and simmering effect was also optimized for production of ACE-inhibitory peptides. The time–temperature combination for the heat treatment was optimized using RSM. The biological activity was measured in the supernatant of the fermented milk after centrifugation. The lowest IC50 values for the inhibition of angiotensin-converting enzyme (ACE) was found 28.9 ± 0.95 μg protein/ml in the supernatant of milk fermented by L. acidophilus and heated at 78 °C for 10 h. The fractions which showed the highest ACE-inhibitory indexes were further purified by different techniques including solid phase extraction, RP-HPLC and FPLC and the related peptides were identified by LC–MS/MS using the Ultimate 3000 nano HPLC system (Dionex) coupled to a 4000 Q TRAP electro-spray ionization mass spectrometry. The high ACE-inhibitory activity containing fractions of the milk fermented by L. acidophilus contained the sequences of b-casein (b-CN) fragment. The fraction-III showed minimum IC50 value i.e. 14.57 ± 0.72 μg/ml compared with fraction-I and fraction-II. Among these peptides 14 peptides have been identified from the fraction-I of the lassi prepared from L. acidophilus i.e. β-CN f47–56, β-CN f47–57, β-CN f199–209, β-CN f176–182, β-CN f176–183, β-CN f176–184, β-CN f1–7, β-CN f57–68, β-CN f166–175, β-CN f195–206, β-CN f195–207, β-CN f195–209, β-CN f94–106 and β-CN f169–176 showed partially or completely homology to that the milk protein bioactive peptides having ACE inhibitory. The two peptides KVLPVPQK (β-CN f169–176) and YQEPVLGPVRGPFPIIV (β-CN f193–209) have the same sequence as ACE inhibitory peptides (Maeno et al. in J Dairy Sci 79(8):1316–1321, 1996; Yamamoto et al. in J Dairy Sci 77:917–922, 1994b).

“Aroeira” (Myracrodruon urundeuva) methanol extract: the relationship between chemical compounds and cellular effects

Context: “Aroeira” [Myracrodruon urundeuva Allemão (Anacardiaceae)] is a tree whose leaves have been studied for therapeutic purposes in medicine and dentistry.Objective: The study chemically identifies the leaf extract of aroeira and determines its effect on human gingival fibroblasts.Materials and methods: An 80% methanol leave extract was obtained by maceration and chemically identified through flow-injection analysis–electrospray ionization–ion trap–tandem mass spectrometry (FIA–ESI–IT–MSn). Cytotoxicity of the aroeira’s methanol extract was evaluated in lineage of fibroblasts. Adherent cells were treated with different concentrations of aroeira’s methanol extract in the medium: 0.1, 1, 10, 100 and 1000 μg/mL. Control cells were cultivated in the medium only. Analyses were done at 24, 48, 72 and 96 h of culture by neutral red assay; and at 24, 48 and 96 h by crystal violet assay.Results: FIA–ESI–IT–MS analysis determined the presence of compounds, for the first time in the species: quercetin-O-glucuronide and quercetin-O-deoxyhexose-O-glucose in the extract. On one hand, neutral red and crystal violet assay showed a reduction (to 50% up until 100%) of cellular viability of groups of 100 and 1000 μg/mL compared with control at 96 h (p < 0.05). On the other hand, lower concentrations (0.1; 1 and 10 μg/mL) of the extract were similar to that of the control at 96 h (p < 0.05), in general.Conclusions: In view of the results, we can conclude that the extract of aroeira presents tannins and flavonoids. Furthermore, the extract is capable of modulating the viability of human gingival fibroblasts according to its concentration.

Identification and characterization of a library of surfactins and fengycins from a marine endophytic Bacillus sp.

An endophytic bacterial strain from a marine green alga, Ulva lactuca, was isolated and identified by 16S rRNA gene sequencing method. The bacterial isolate was found to secrete two major families of cyclic depsilipopeptides, surfactins, and fengycins. Sequencing of the isolated lipopeptides was carried out using the MSn data obtained from an electrospray ionization (ESI) ion trap mass spectrometer coupled to an HPLC system. The assigned sequences were confirmed by a chemical derivatization approach involving esterification followed by mass spectrometric analysis. Distinction of leucine residues from isoleucine was established through a combined electron transfer dissociation-collision-induced dissociation (ETD-CID) method. The fengycins described in this study were found to cause significant delay of growth of two plants, Vigna radiata (mung bean) and Oryza sativa (rice). To the best of our knowledge, this is the first study describing identification and characterization of cyclic peptides from an endophytic Bacillus sp. isolated from marine algae.

High-Performance Liquid Chromatography with Diode Array Detector and Electrospray Ionization Ion Trap Time-of-Flight Tandem Mass Spectrometry to Evaluate Ginseng Roots and Rhizomes from Different Regions.

Ginseng, Panax ginseng C. A. Meyer, is an industrial crop in China and Korea. The functional components in ginseng roots and rhizomes are characteristic ginsenosides. This work developed a new high-performance liquid chromatography coupled with electrospray ionization ion trap time-of-flight multistage mass spectrometry (LC–ESI-IT-TOF-MSn) method to identify the triterpenoids. Sixty compounds (1–60) including 58 triterpenoids were identified from the ginseng cultivated in China. Substances 1, 2, 7, 15–20, 35, 39, 45–47, 49, 55–57, 59, and 60 were identified for the first time. To evaluate the quality of ginseng cultivated in Northeast China, this paper developed a practical liquid chromatography–diode array detection (LC–DAD) method to simultaneously quantify 14 interesting ginsenosides in ginseng collected from 66 different producing areas for the first time. The results showed the quality of ginseng roots and rhizomes from different sources was different due to growing environment, cultivation technology, and so on. The developed LC–ESI-IT-TOF-MSn method can be used to identify many more ginsenosides and the LC–DAD method can be used not only to assess the quality of ginseng, but also to optimize the cultivation conditions for the production of ginsenosides.

Identification and Biological Activities of Long‐Chain Peptaibols Produced by a Marine‐Derived Strain of Trichoderma longibrachiatum

Six long-chain peptaibols, 1-6, were identified from agar cultures of a marine-derived Trichoderma longibrachiatum Rifai strain (MMS151) isolated from blue mussels. The structure elucidation was carried out using electrospray ionization ion trap mass spectrometry (ESI-IT-MS) and GC/EI-MS. The long-chain peptaibols exhibited the general building scheme Ac-Aib-Ala-Aib-Ala-Aib-XXX-Gln-Aib-Vxx-Aib-Gly-XXX-Aib-Pro-Vxx-Aib-XXX-Gln-Gln-Pheol and were similar or identical to recurrent 20-residue peptaibols produced by Trichoderma spp. Three new sequences were identified and were called longibrachins A-0, A-II-a, and A-IV-b. The isolated peptaibols were assayed for cytotoxic, antibacterial, and antifungal activities, and acute toxicity on Dipteran larvae.

Rapid screening of transferrin-binders in the flowers of Bauhinia blakeana Dunn by on-line high-performance liquid chromatography–diode-array detector–electrospray ionization–ion-trap–time-of-flight–mass spectrometry–transferrin–fluorescence detection system

Transferrin (Transferrin, TRF, TF) has drawn increasing attention in cancer therapy due to its potential applications in drug delivery. TF receptor, highly expressed in tumor cells, recognizes and transports Fe3+-TF into cells to release iron into cytoplasm. Thus, discovering TF-binding compounds has become an active research area and is of great importance for target therapy. In this study, an on-line analysis method was established for screening TF-binding compounds from the flowers of Bauhinia blakeana Dunn using a high-performance liquid chromatography–diode-array detector–multi-stage mass spectrometry–transferrin–fluorescence detector (HPLC–DAD–MSn–TF–FLD) method. As a result, 33 of 80 identified or tentatively characterized compounds in the sample were TF-binding active. Twenty-five flavonol glycosides and eight phenolic acids were identified as TF-binders. Twelve of these active compounds together with six standard compounds were used to study the dose-response effects and structure-activity relationships of flavonoids and phenolic acids. The method was validated by vitexin with a good linearity in the range of concentrations used in the study. The limit of detection for vitexin was 0.1596 nmol. Our study indicated that the established method is simple, rapid and sensitive for screening TF-binding active compounds in the extract of Bauhinia blakeana Dunn, and therefore is important for discovering potential anti-cancer ingredients from complex samples for TF related drug delivery.

Multiclass Carcinogenic DNA Adduct Quantification in Formalin-Fixed Paraffin-Embedded Tissues by Ultraperformance Liquid Chromatography-Tandem Mass Spectrometry.

DNA adducts are a measure of internal exposure to genotoxicants and an important biomarker for human risk assessment. However, the employment of DNA adducts as biomarkers in human studies is often restricted because fresh-frozen tissues are not available. In contrast, formalin-fixed paraffin-embedded (FFPE) tissues with clinical diagnosis are readily accessible. Recently, our laboratory reported that DNA adducts of aristolochic acid, a carcinogenic component of Aristolochia herbs used in traditional Chinese medicines worldwide, can be recovered quantitatively from FFPE tissues. In this study, we have evaluated the efficacy of our method for retrieval of DNA adducts from archived tissue by measuring DNA adducts derived from four other classes of human carcinogens: polycyclic aromatic hydrocarbons (PAHs), aromatic amines, heterocyclic aromatic amines (HAAs), and N-nitroso compounds (NOCs). Deoxyguanosine (dG) adducts of the PAH benzo[a]pyrene (B[a]P), 10-(deoxyguanosin-N(2)-yl)-7,8,9-trihydroxy-7,8,9,10-tetrahydrobenzo[a]pyrene (dG-N(2)-B[a]PDE); the aromatic amine 4-aminobiphenyl (4-ABP), N-(deoxyguanosin-8-yl)-4-aminobiphenyl (dG-C8-4-ABP); the HAA 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP), N-(deoxyguanosin-8-yl)-PhIP (dG-C8-PhIP); and the dG adducts of the NOC 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK), O(6)-methyl-dG (O(6)-Me-dG) and O(6)-pyridyloxobutyl-dG (O(6)-POB-dG), formed in liver, lung, bladder, pancreas, or colon were recovered in comparable yields from fresh-frozen and FFPE preserved tissues of rodents treated with the procarcinogens. Quantification was achieved by ultraperformance liquid chromatography coupled with electrospray ionization ion-trap multistage mass spectrometry (UPLC/ESI-IT-MS(3)). These advancements in the technology of DNA adduct retrieval from FFPE tissue clear the way for use of archived pathology samples in molecular epidemiology studies designed to assess the causal role of exposure to hazardous chemicals with cancer risk.

Biochemical characterization of the novel α-1, 3-galactosyltransferase WclR from Escherichia coli O3

Glycosyltransferases (GTs) catalyze the formation of regio- and stereo-specific glycosidic linkages between specific sugar donors and recipients. In this study, the function of the gene wclR from the Escherichia coli O3 O-antigen gene cluster that encodes an α 1, 3-galactosyltransferase (GalT) that acts on the linkage Gal α 1, 3-GlcNAc was biochemically characterized. WclR was expressed in E. coli BL21 (DE3), and the enzymatic product was identified by liquid chromatography-mass spectrometry (LC-MS), collision-induced dissociation electrospray ionization ion trap multiple tandem MS (CID-ESI-IT-MSn) and galactosidase digestion, using UDP-Gal as the donor substrate and the synthetic acceptor substrate GlcNAc-PP-De (decyl diphosphate N-acetylglucosamine). The physiochemical properties and the substrate specificity of WclR were investigated. WclR is the first bacterial GalT characterized that acts on the linkage Gal α 1, 3-GlcNAc. This study enhanced our knowledge of the diversified functions of GTs and provided a novel enzyme source for possible pharmaceutical application.

Automated Modular High Throughput Exopolysaccharide Screening Platform Coupled with Highly Sensitive Carbohydrate Fingerprint Analysis.

Many microorganisms are capable of producing and secreting exopolysaccharides (EPS), which have important implications in medical fields, food applications or in the replacement of petro-based chemicals. We describe an analytical platform to be automated on a liquid handling system that allows the fast and reliable analysis of the type and the amount of EPS produced by microorganisms. It enables the user to identify novel natural microbial exopolysaccharide producers and to analyze the carbohydrate fingerprint of the corresponding polymers within one day in high-throughput (HT). Using this platform, strain collections as well as libraries of strain variants that might be obtained in engineering approaches can be screened. The platform has a modular setup, which allows a separation of the protocol into two major parts. First, there is an automated screening system, which combines different polysaccharide detection modules: a semi-quantitative analysis of viscosity formation via a centrifugation step, an analysis of polymer formation via alcohol precipitation and the determination of the total carbohydrate content via a phenol-sulfuric-acid transformation. Here, it is possible to screen up to 384 strains per run. The second part provides a detailed monosaccharide analysis for all the selected EPS producers identified in the first part by combining two essential modules: the analysis of the complete monomer composition via ultra-high performance liquid chromatography coupled with ultra violet and electrospray ionization ion trap detection (UHPLC-UV-ESI-MS) and the determination of pyruvate as a polymer substituent (presence of pyruvate ketal) via enzymatic oxidation that is coupled to a color formation. All the analytical modules of this screening platform can be combined in different ways and adjusted to individual requirements. Additionally, they can all be handled manually or performed with a liquid handling system. Thereby, the screening platform enables a huge flexibility in order to identify various EPS.

Dehydrogenation of icosahedral carborane anions via gas-phase collisional activation.

The icosahedral carborane anion HCB11 H11-1 is well known for its exceptional stability in solution and the solid state, but the gas-phase properties of this molecule have not previously been explored. Electrospray ionization ion trap mass spectrometry, collisional activation, and isotopic labelling were used to examine the formation and reaction pathways of covalent and noncovalent carborane clusters. Covalent attachment of an amino group to the C-vertex of HCB11 H11-1 dramatically alters its stability in the gas phase. Interestingly, covalently bound carborane dimers are produced during electrospray ionization. Subsequent collisional activation of these dimers leads to facile hydrogen losses (7 H2 molecules). The loss of H2 does not follow a stochastic pattern, but occurs preferentially by loss of 3 H2 , followed by loss of 2 H2 and then another 2 H2 molecules. Screening anaphylactic components of MaiLuoNing injection by using rat basophilic leukemia-2H3 cell membrane chromatography coupled with HPLC-ESI-TOF-MS.

MaiLuoNing injection is a traditional Chinese medicine that used clinically since the 1950s in China. However, anaphylactic reactions, through the potentiation of mast cell degranulation, have been reported. In the present study, a rat basophilic leukemia-2H3 cell membrane chromatography coupled with high-performance liquid chromatography and electrospray ionization-ion trap-time of flight-mass spectrometry method was established for screening, analyzing, and identifying the potential anaphylactic components of MaiLuoNing injection. Harpagoside, a potential degranulator of rat basophilic leukemia-2H3 cells, was retained in rat basophilic leukemia-2H3 cell membrane chromatography. We aimed to evaluate the retained components to determine which of those were capable of inducing degranulation of basophilic leukemia cells. A β-hexosaminidase assay revealed that harpagoside can induce rat basophilic leukemia-2H3 cell degranulation in a dose-dependent manner. BLBA/c mice also exhibit passive cutaneous anaphylaxis in response to harpagoside. These results indicate that rat basophilic leukemia-2H3 cell membrane chromatography coupled with high-performance liquid chromatography and electrospray ionization ion trap time-of-flight mass spectrometry is effective in screening for the anaphylactic components of MaiLuoNing injection.

Characterization of the Anticoagulative Constituents of Angelicae Sinensis Radix and Their Metabolites in Rats by HPLC-DAD-ESI-IT-TOF-MSn.

Angelicae Sinensis Radix is commonly used in traditional Chinese medicine. Pharmacological studies show that Angelicae Sinensis Radix has clear anticoagulant activity. Therefore, in this study, the anticoagulant activity of crude Angelicae Sinensis Radix extracts was investigated by measuring the thrombin times of the extracts. The results revealed that the petroleum ether-soluble fraction of Angelicae Sinensis Radix exhibited significant anticoagulant activity in vitro, and 26 compounds were characterized by high-performance liquid chromatography with diode array detection combined with electrospray ionization ion trap time-of-flight multistage mass spectrometry. In addition, 5 prototype constituents, 24 in vivo metabolites in rat urine and 7 prototype constituents, and 9 in vitro metabolites in the rat hepatic S9 incubation system of the petroleum ether-soluble fraction were tentatively identified. All metabolites were found from Angelicae Sinensis Radix for the first time. Among them, 13 (three ferulic acid-related constituents, six senkyunolide D-related constituents, and four senkyunolide F-related constituents) were identified as new metabolites (new compounds). This study is the first to qualitatively characterize the chemical constituents of the potent anticoagulative extract of Angelicae Sinensis Radix and to explore its metabolism. The result is a notable improvement in the discovery of Angelicae Sinensis Radix metabolites, and it provides the chemical basis for the effective forms and pharmacodynamic substances (prototypes, metabolites, or both) of the anticoagulant activity of Angelicae Sinensis Radix.

Protective Effect of Deer Bone Oil on Cartilage Destruction in Rats with Monosodium Iodoacetate (MIA)-Induced Osteoarthritis.

The anti-osteoarthritic activity of the methanol fraction of deer bone oil extract (DBO-M) was evaluated in interleukin (IL)-1β-inflamed primary rabbit chondrocytes and in rats with monosodium iodoacetate (MIA)-induced osteoarthritis. The active compound in DBO-M was analyzed using a direct infusion liquid chromatography quadrupole (LCQ) ion-trap electrospray ionization (ESI)-mass spectrometer (MS). DBO-M significantly suppressed the IL-1β-induced sulfated-glycosaminoglycan (s-GAG) release from chondrocyte, and lowered mRNA levels of the collagen-degrading enzymes matrix metalloproteinase (MMP)-1 and MMP-3 in a dose-dependent manner. Upon treatment with high doses of DBO-M, the levels of IL-1β, tumor necrosis factor (TNF)-α, and IL-6 decreased by around 40, 70, and 50%, respectively, compared to the control in the serum of rats with MIA-induced osteoarthritis. Bone volume fraction (BV/TV) and trabecular thickness (Tb.Th) increased by over 40% in rats treated with DBO-M compared to the values reported for the MIA-treated control group, while trabecular separation (Tb.Sp) showed a significant decrease (ca. 38%), as confirmed through micro-computed tomography (CT) analysis of MIA-induced destruction of articular bones. Furthermore, direct infusion ESI-MS analysis showed that DBO-M contains gangliosides, which are glycosphingolipids with monosialic acid (GM3), as a major compound. Our results suggest that DBO-M effectively improves MIA-induced osteoarthritis by suppressing inflammatory responses, and that gangliosides could be one of the DBO-derived anti-inflammatory components.

Sensitive analysis of phthalate esters in plastic bottled water via on-line capillary solid-phase microextraction liquid chromatography electrospray ionization-ion trap mass spectrometry

An online capillary solid-phase microextraction coupled liquid chromatography electrospray ionization-ion trap mass spectrometry (cSPME-LC-ESI-IT-MS) has been developed and applied for analyzing six phthalate esters (PAEs) in commercially available plastic bottled water.

Study of the hydrolytic properties of the trivalent Y-ion in chloride medium

Hydrolysis reactions of YCl3 aqueous solutions were studied by potentiometric titrations at 310K in the pH range ca.3–9 and total Y3+ ion concentration range 0.5–10.0mmol/dm3. The experimental data were evaluated with the aid of the Hyperquad2006 suite of programs, which indicated the formation of the following hydrolytic complexes (logconc. overall stability constants βp,q defined as βp,q=Mp(OH)q[H]q/[M]p), Y(OH)2+ (−7.71(2)); Y(OH)2+ (−16.42(2)); Y(OH)3 (−26.01(3)); Y2(OH)24+ (−14.23(4)); Y3(OH)52+ (−34.20(4)); Y4(OH)62+ (−37.10(5)). The hydrolysis of the Y3+ ion was also monitored by electrospray ionization ion-trap mass spectrometry (ESI-MS) in the pH range 2.63–7.10. The influence of the sample cone voltage on the product distribution was significant, indicating the contribution of ion source fragmentation reactions. The optimal cone voltage was set to 70V for cationic and 100V for anionic spectra. The cationic and anionic ESI mass spectra of the Y3+ solutions produced strong evidence for the presence of a variety of monomeric and polymeric hydrolytic complexes. The Cl− ion appeared to be a strong competitor to the OH− ion for the oxo-yttrium core. The hydrolytic precipitation reactions with NaOH or urea as precipitating agents were studied. The obtained precipitates were characterized by the potentiometric technique, elemental analysis, thermal analysis (TG-DSC), infrared spectroscopy, X-ray diffractions and the SEM technique. Hydrothermal synthesis of the precipitate in an autoclave at T=420K and P=10bar with NaOH as the precipitating agent produced a crystalline precipitate in the form of hexagonal needles (P63/m space group). At atmospheric pressure only an amorphous precipitate was produced with both precipitating agents. The concentration solubility product K∗so of Y(OH)3 was evaluated from the potentiometric titrations curves and logK∗so=−22.85(4). The synthesized precipitate with NaOH as a precipitating agent had a composition of Y(OH)3·2H2O, while the amorphous precipitate obtained with urea had a composition of Y2(OH)2(CO3)2·3H2O.

A Fragmentation Study of Six C21 Steroidal Aglycones by Electrospray Ionization Ion-Trap Time-of-Flight Tandem Mass Spectrometry

The fragmentation patterns of six C21 steroidal aglycones, metaplexigenin (1), caudatin (2), qingyangshengenin (3), penupogenin (4), 20-cinnamoylsarcostin (5), and gagamine (6), were analyzed by high-resolution electrospray ionization ion-trap time-of-flight tandem mass spectrometry (HR-ESI-IT-TOF-MS(n)). The [M-H]+ ions of steroids 1-3 that contain a carbonyl functional group at C-20 (Type I) and [M+H]+ ions of steroids 5-6 that possess a hydroxyl group at C-20 (Type II) were readily observed in MS analyses. The fragmentation pathways and diagnostic fragment ions for these six steroidal aglycones were proposed on the basis of their MS(n) analyses. The common fragmentation pathways for type I steroidal aglycones include the neutral loss of the ester group at C-12 and the hydroxyl moieties on the steroid skeleton, as well as the cleavage of ring D. Their diagnostic fragment ions were identified as m/z 361(B), 343 (C), 325 (D), 307 (F), 283 (G), 259 (E), and 243 (H). The fragmentation behavior of penupogenin (4) in type II was similar to those of type I, with m/z 363 (B'), 345 (C'), 327 (D'), 309 (F'), 283 (G), and 243 (H) as its diagnostic fragment ions. The ester group at C-20 was difficult to cleave in the MS(n) analyses of 20-cinnamoylsarcostin (5) and gagamine (6) so that the loss of this ester group was slower than that at C-12 and hydroxyl groups; the key ions at m/z 329 (I), 311 (J), 293 (K), and 275 (L) were characteristic for 5 and 6. The base ion peaks were derived from the loss of the substituent group at either C-12 or C-17 for both type I and type II steroidal aglycones.
