Efficacy of silver-doped Carbon dots in Chemical Castration: a rat model study

This study evaluates silver-doped carbon dots (AgCDs) as a novel agent for chemical castration using a rat model. Six groups of rats (five males and ten females each, except for the surgical group which had only males) were utilized to compare the effects of different concentrations of AgCDs. The groups included control, sham, and three experimental groups injected with 1.25, 50, and 200 µg/mL AgCDs, respectively, along with a surgical castration group. Testosterone levels, sperm parameters, fertility index, oxidative damage, histopathological parameters, and gene expression of P53, Bax, Bcl-2, caspase-3, AKT, and PI3K were analyzed. Results demonstrated that the high-dose AgCDs group significantly reduced testosterone levels, sperm concentration, and motility, resulting in a decreased fertility index. MDA and NO significantly increased, while CAT, SOD, GPx, and TAC significantly reduced in the chemically castrated groups. Histological and genes expression analysis also revealed apoptosis and testicular damage in the AgCDs groups, indicated by significant increases in P53, Bax, and Caspase-3 levels, and significant reductions in AKT, PI3K, and Bcl-2. Based on these findings, AgCDs could be considered a potent and efficient agent for chemical castration, offering a less invasive, cost-effective solution with potential applications for population control.

The investigation of endodontic irrigants and polyhexanide-based solution action on smear layer

AIM. The effect of irrigants on the smear layer on the intracanalicular dentin surface is one of the important issues in endodontics. The objective of this article is to determine the effect of different concentrations of polyhexanide-based solutions and standard irrigants on the smear layer using scanning electron microscopy (SEM).MATERIALS AND METHODS. The present study was conducted on 42 extracted teeth (third molars, first and second premolars) with formed apices that had been extracted for orthodontic indications. The teeth were decoronated and samples were prepared. In order to create a smear layer, the lumen of the canals was instrumented with H-file #40 throughout. A simulation of root canal irrigation was conducted by immersing the specimens in a 5-ml container filled with the proper solution for 1 hour: group 1 – no treatment (n = 6); group 2 – saline (n = 6); group 3 – 2% chlorhexidine (n = 6); group 4 – 3% sodium hypochlorite (n = 6); group 5 – 17% EDTA; group 6 – polyhexanide 0.1% (n = 6); group 7 – polyhexanide 0.2% (n = 6). A ~30 nm thick platinum film was sputtered onto the surface of each sample using a magnetron deposition technique. SEM microphotos were acquired on a Vega3 TESCAN scanning electron microscope (SEM) with an SE secondary electron detector, which enables imaging with high topographic contrast. The initial detector (incamera) was employed for image acquisition, with an electron energy of 30 keV. Images were acquired at magnifications of x20 (overview image), x150, x250, x500, x1000, x2500, x7500 for each sample.RESULTS. The samples from groups 1 and 5 demonstrated the absence of a smear layer on the dentin surface and the absence of smear plugs in the dentinal tubules. Groups 2 and 3 are distinguished by the deposition of a pronounced amorphous smear layer and the obturation of all dentinal tubules. Groups 4, 6, and 7 are distinguished by the deposition of a smaller volume of smear layer on the dentin surface, as well as the presence of semi-open or completely open entrances to the dentinal tubules.CONCLUSION. The data obtained indicates that antiseptic compositions based on polyhexanide in concentrations of 0.1 and 0.2% do not have a pronounced effect on the smear layer. However, in visual evaluation of SEM micrographs, their effect is comparable to that of 3% sodium hypochlorite. In instances where enhanced dissolution of the smear layer and smear plugs from the tubules is necessary to facilitate decontamination of the wall dentin, the combination of antiseptics with chelate compounds, such as 17% EDTA, may be employed.

Trichoderma Rhizosphere Soil Improvement: Regulation of Nitrogen Fertilizer in Saline–Alkali Soil in Semi-Arid Region and Its Effect on the Microbial Community Structure of Maize Roots

In order to reduce the actual impact of a saline–alkali environment on maize production in semi-arid areas, it is particularly important to use the combined fertilization strategy of Trichoderma microbial fertilizer and nitrogen fertilizer. The purpose of this study was to investigate the effects of different concentrations of nitrogen fertilizer combined with Trichoderma on improving the structural characteristics and ecological functions of maize rhizosphere microbial community in semi-arid saline–alkali soil. Through the microbiome analysis of maize rhizosphere soil samples with 60 kg N·ha−1 (N1) and 300 kg N·ha−1 (N2) nitrogen fertilizer combined with Trichoderma (T1) and without Trichoderma (T0), we found that the combination of Trichoderma and different concentrations of nitrogen fertilizer significantly affected the structure of bacterial and fungal communities. The results of this study showed that the combination of Trichoderma and low-concentration nitrogen fertilizer (N1T1) could improve soil nutritional status and enhance its productivity potential, revealing the relationship between beneficial and harmful fungal genera, microbial diversity and abundance, and crop biomass, which is of great significance for improving agricultural production efficiency and sustainable development.

Improving fruit set and quality of Daisy mandarin through foliar application of nutrients

Daisy mandarin is a widely cultivated citrus species valued for its nutritional qualities and organoleptic properties, favored for both fresh consumption and processing. Optimal growth and fruit production is depend upon the availability of essential nutrients. However, soils of the central zones of Punjab are deficient in potassium which hinders the quality and production and their deficiency impairs physiological and biochemical processes, reducing fruit set, yield and quality. Potassium and calcium are vital for metabolic functions and cell wall integrity. A deficiency in any of these nutrients can results in smaller fruits, lower fruit retention and diminished organoleptic and nutritional qualities, such as reduced antioxidants content. So that, a research was conducted to evaluate the effect of different concentrations of potassium nitrate, potassium sulfate, calcium nitrate and boric acid along with paddy straw mulching on Daisy mandarin during the years 2021–22. The pooled data of this research showed that potassium nitrate (2%), is the best treatment with maximum fruit set (14.37%) and retention (72.35%) in Daisy mandarin. Additionally, the average fruit weight (207.50 g), number of fruits (162), fruit yield (33.79 kg/tree), fruit length (6.25 cm) and breadth (6.73 cm) also enhanced. Furthermore, potassium nitrate (2%) application increased the antioxidant activity (39.43%), phenolic content (230.00 mg kg−1 FW) and flavonoid content (3.69 mg RE g−1 FW). The findings suggest that application of potassium nitrate presents a promising strategy for enhancing fruit set, yield and quality of Daisy mandarin, providing valuable insights for improved cultivation practices.

Biodegradable soy protein isolate carvacrol coating controls soft rot on nectarines by enhancing defense response

Nectarine fruits are rich in nutrients, which are susceptible to fungal infection during postharvest, resulting in quality deterioration and decay. This study aimed to assess the effect of different concentrations of carvacrol and soy protein isolate coatings on the fungal infection and quality characteristics of nectarines during postharvest storage. The results showed that soy protein isolate carvacrol (SPI-C) coating inhibited the growth of Rhizopus stolonifer in vitro. The SPI-C coating significantly reduced the incidence of postharvest soft rot, maintained the postharvest quality of nectarines during storage. SPI-C coating induces the defense response to soft rot by enhancing the accumulation of phenolic, flavonoids and lignin, and up-regulating the expression of defense-related genes in nectarines. The SPI-C coating activated the transcription of antioxidant enzymes (CAT, SOD and POD) and eliminated ROS damage. The disease resistance response promotes the production of defense-related substances by upregulating the gene expression of PpPPO and PpPAL. In summary, SPI-C coating significantly reduces the incidence of postharvest soft rot, maintains the postharvest quality and induces defense response to soft rot by enhancing the expression of genes involved in phenylpropanoid metabolism in nectarines during storage.

Effect of different concentrations of soil and foliar applied zinc, boron and iron fertilizers on seedling growth, chlorophyll content and productivity of chickpea seedlings under semi-arid environment

The effectiveness of zinc (Zn), boron (B), and iron (Fe) is reduced in semi-arid regions, which can lead to a deficiency of these nutrients and inhibit chickpea productivity. In this work, three field experiments were executed over two years where soil and foliar applications of Zn, B, and Fe were carried out, including controls (Zn0, B0, and F0), soil application at 4.125 kg/ha (Zn-1, B1, and F1) and 8.25 kg/ha (Zn-2, B2, and F2), and foliar spay at 0.3% at flowering initiating (Zn-3, B3, and F3) and one week after flowering initiation (Zn-4, B4, and F4, respectively). The results indicate that the deficiency of these nutrients inhibited chickpea growth and yield, leading to a reduction in the pigment contents. Nonetheless, soil and foliar application of Zn, B, and Fe significantly improved growth, chlorophyll contents, and yield, showing a dose-dependent effect. The best results were recorded for Zn-3, B2, and F2 treatments which significantly (P<0.05) increased shoot length (20.96-85.19%), root length (42.85-93.65%), shoot fresh (23-76%) and root fresh weight (45-90.32%), compared with the control treatment. Chlorophyll parameters, including chlorophyll a and b contents, showed similar trends. Zn-3, B2, and F2 treatments significantly increased biological and grain yields, which were associated with higher values of the number of pods per plant and the number of seeds per pod. In a nutshell, we suggest that Zn foliar application at 0.3% at flowering initiation and soil application of B and Fe at 25 kg/ha are beneficial for improving the growth, pigment content, and overall productivity of chickpea.

Mitigating Salt Stress with Biochar: Effects on Yield and Quality of Dwarf Tomato Irrigated with Brackish Water.

The increase in the frequency and magnitude of environmental stresses poses a significant risk to the stability of food supplies. In coastal areas of the Mediterranean, brackish water has long been considered a limitation on horticultural production. In this scenario, the use of biochar in agriculture could be considered a valuable tool to cope with the deleterious effects of salt stress. This work aimed to investigate, in a protected environment, the effects of different concentrations of biochar (0, 1, and 2% v/v) obtained from poplar (Populus L.) biomass on the yield and quality of dwarf San Marzano ecotype tomatoes irrigated with saline water at different concentrations of NaCl (0, 40 and 80 mM). The increase in salt concentration from 0 to 80 mM NaCl reduced the total yield (-63%) and the number of fruits (-25%), but improved the main quality parameters such as dry matter (+75%), total soluble solids (+56%), and polyphenol content (+43%). Compared to control conditions, biochar supplementation improved the total yield (+23%) and number of fruits (+26%) without altering the functional and organoleptic characteristics of the fruits. The promising results underscore the potential of biochar as a sustainable solution to amend soils in order to improve tomato production under unfavorable conditions such as high salinity. However, there is a need to clarify which adaptation mechanisms triggered by biochar amending improve production responses even and especially under suboptimal growing conditions.

Effects of biochar application on soil properties and the growth of Melissa officinalis L. under salt stress

Soil salinization is one of the world's most seriously ecological issues. The application of biochar may enhance the properties of the soil and lessen the harm that salt stress causes to plants. In this study, we used the cuttings of Melissa officinalis as experimental materials. The method of pot experiment was used to explore to explore the effects of different concentrations of biochar (0, 10 %, and 20 % w/w) on soil properties and plant physiological characteristics under salt stress (0, 0.20 %, and 0.60 % NaCl+Na2SO4). The results indicate that the physicochemical properties of soil and the plant growth were decreased impacted with increasing salinity level, and these negative impacts were decreased traits were improved with the application of biochar. It was discovered that the application of biochar could increase the soil water holding capacity, total porosity, available P and K content, and soil enzyme activity while also decreasing the soil bulk density under salt stress. Biochar addition promoted the accumulation of plant biomass and the acquisition of nutrients, and reduced Na content in plants. With the addition of biochar, malondialdehyde (MDA) content and electrolyte leakage displayed a significant decrease under salt stress. A reduction in the osmotic substances and secondary metabolite accumulation in the leaves was also evident. The presented results reveal that biochar can contribute to protect M. officinalis against salt stress by alleviating the oxidative stress. Among the test samples, the 20 % biochar application had the best performance, suggesting that this is an advantageous method for improving soil properties and lessening the harm caused by salt stress on M. officinalis.

Effect of Different Concentrations of Tomato Leaf Extract on Aphid Control in Rape Production in Zimbabwe

Rape (Brassica napus) is an important vegetable crop grown by smallholder farmers in Zimbabwe to achieve food and nutritional security. However, productivity and quality of rape are greatly dampened by aphids which reduce its palatability and quality. Chemical pesticides, while effective, pose environmental risks and can lead to pest resistance and resurgence, besides residual effects and bioaccumulation along the food chain. Tomato leaves are rich in phenolic and flavonoid compounds, that have shown promise as a botanical pesticide against aphids in other crops. This research aimed to establish the aphicidal components in tomato leaf extract using TLC and UV-Vis Spectroscopy; establish toxicity of the extract using laboratory bioassay, investigate the potential of different concentrations of tomato leaf extracts as a natural, environmentally friendly and sustainable approach to pest management against aphids in rape production at Kushinga Phikelela Agricultural College in Zimbabwe. A CRD with four treatments (0%, 20%, 40% and 60% of tomato leaf extract) and distilled water (negative control) replicated five times each was used. Aphid infestation (scored on a scale of 0-9), yield, leaf size, and plant height were measured fortnightly. Data was analysed for statistical significance using ANOVA while separation of significant means was done by Fisher’s Protected Least Significant Difference Test (LSD (5%). The results from combined analysis using TLC and UV- Vis Spectroscopy indicated that tomato leaf extract used in this research contained compounds (Solanine, Tomatine, Chaconine) which are known to be toxic to pests. There was a highly significant (p<0.001) difference among treatments on aphid mortality. The highest aphid mortality (95.57%) while the lowest (4%) was observed in 60% and 0% leaf extract respectively. Aphid infestation, leaf size and plant height were highly significant (P<0.001) during weeks 4, 6, 8 and 10 after transplanting. Aphid infestation in week 10 mean score was 8.8, 6.2, 3.8 and 0.8 for 0%, 20%, 40% and 60% tomato leaf extract respectively. Other parameters such as yield and leaf size were highest in the plots that received 60% tomato leaf extract sprays. Highly significant difference (P<0.001) in yield was observed in weeks 8 and 10, while significant difference (P<0.05) in yield was noted in weeks 4 and 6. However, in week two there were no significant differences among the treatments in yield and aphid infestation. Tomato leaf extracts contain secondary metabolites toxic to aphids and this can be used by resource constrained farmers. We recommend the use 60% tomato leaf extracts to control aphids in rape to sustainably achieve high yields.

Evaluation of Anti-Viral Photodynamic Therapy Effects of Different Concentrations of 5-ALA Using Light Irradiation on HSV-1

BackgroundHerpes simplex virus (HSV) infections are prevalent worldwide, posing challenges due to asymptomatic carriage and recurrent outbreaks. Conventional treatments, including antiviral medications, are limited by drug resistance. Photodynamic therapy (PDT) offers a promising alternative, leveraging photosensitizers to induce viral inactivation via reactive oxygen species (ROS) generation upon light activation. MethodsThis study investigates the efficacy of 5-aminolevulinic acid (5-ALA) as a photosensitizer in antiviral PDT against HSV-1. HSV-1 was treated with varying concentrations of 5-ALA and subjected to light irradiation at a 630 nm wavelength. Virus titers were assessed using the 50% Tissue Culture Infectious dose (TCID50) assay. Six experimental groups were used: [1] Control (no 5-ALA or light), [2] HSV-1 with 0.05 g/ml 5-ALA and 630 nm light, [3] HSV-1 with 0.05 g/ml 5-ALA (no light), [4] HSV-1 with 0.1 g/ml 5-ALA and light, [5] HSV-1 with 0.1 g/ml 5-ALA (no light), and [6] HSV-1 with light (no 5-ALA). ResultsVirus treatment with 0.1 g/ml 5-ALA combined with light irradiation significantly reduced HSV-1 titer compared to control groups (p<0.05). Specifically, the virus titer decreased from 10 6.3 TCID50/ml in the control group to 10 4.5 TCID50/ml in the treated group. However, lower concentrations of 5-ALA or light irradiation alone did not yield significant reductions in HSV-1 titer (p>0.05). Group 4, receiving 0.1g/ml 5-ALA with light irradiation, exhibited a significantly greater reduction in virus titer than group 2, receiving 0.05 g/ml 5-ALA with light irradiation (p<0.05). Conclusions5-ALA-mediated PDT demonstrates selective antiviral efficacy against HSV-1, particularly at higher concentrations coupled with light irradiation. These findings underscore the potential of 5-ALA-based PDT as a promising approach for HSV-1 treatment, especially in cases of drug-resistant strains and immunocompromised individuals. Optimization of dosage and treatment protocols is essential for maximizing effectiveness in clinical applications, highlighting the need for further research in this area.

Negative effects of cigarette butt leachate on freshwater phytoplankton communities.

We experimentally tested the effects of different concentrations of cigarette butt leachate on freshwater phytoplankton chlorophyll-a, species richness, cell density, and community composition. For this, we sampled the phytoplankton from a eutrophic lake and acclimated it for 24 h in microcosms. We then conducted the experiment in microcosms maintained for 96 h. The experiment consisted of four treatments: control and leachate from 1 butt L-1 (T1), 5 butts L-1 (T5), and 10 butts L-1 (T10), which were prepared by diluting a stock solution of leachate from 50 butts L-1. We found that algal chlorophyll-a content was not affected by different leachate concentrations. In contrast, phytoplankton cell density decreased in a dose-dependent manner as concentrations of the leachate increased. Similarly, the number of species was highest in the control group relative to all other treatments, with T1 and T5 showing higher species richness than T10. Additionally, the exposition to different concentrations of the leachate impacted community composition across all treatments in comparison to the control group. Our results suggest that cigarette butt leachate alters the number of cells and species, as well as the distribution of abundance, without necessarily reducing chlorophyll-a concentrations. Our findings indicate that to gain a comprehensive understanding of the effects of cigarette butt leachate on freshwater ecosystems, it is essential to evaluate more realistic scenarios that incorporate aquatic communities, rather than isolated species.

Antioxidant and Immunological Properties of Dracocephalum moldavica Extract in Penaeus vannamei

Objectives: The effects of different concentrations of Dracocephalum moldavica extract (DME) supplement on growth, survival, antioxidants, some biochemical parameters and innate immunity of White leg shrimp (Penaeus vannamei) juveniles investigated. Methods: For 56 days, shrimp (4.5-5 g) received diets that were enhanced with 0 (control), 50, 100, 200 mg DME/kg feed to attain apparent satiety. Results: The results revealed that feeding shrimp with DME significantly improved Feed efficiency and Feed conversion ratio with an optimum level of 200 mg/kg diet. Dietary DME supplementation had no significant effects on their carcass composition. Total albumin and protein higher (P &gt; 0.05) in DME-fed particularly at 200 mg/kg treatment On the other hand, shrimp fed 200 mg DME/kg feed experienced significant decreases in aspartate and alanine aminotransferase levels. As dietary extract levels increased, there was a significant (P &lt; 0.05) increase in SOD and CAT activities, as well as LYZ and PO levels, so that treatments with 100 and 200 mg DME/kg feed had the highest values, and Significant (P &lt; 0.05) reductions in MDA and NO levels were observed at high dietary extract levels of 100 and 200 mg DME/kg. Conclusion: P. vannamei's growth, antioxidant capacity and innate immunity were boosted by diets that contained 200 mg DME/kg feed.

Optimization of pH-responsive microgel for the co-delivery of Weizmannia coagulans and procyanidins to enhance survival rate and tolerance

The purpose of this study was to prepare a pH-responsive microgel for co-delivering Weizmannia coagulans 99 (BC99) and procyanidins (PCs) to enhance the survival rate and tolerance of probiotics in complex micro-environment. The effects of different concentrations of PCs on the properties of microgels were optimized, and found that the spherical microgels had higher encapsulation efficiency (90.27 ± 2.51 %) and smaller size when the concentration of PCs was 20 μg/mL. The interaction among PCs, pectin and protein could effectively improve the survival rate of BC99 under different pH, bile salt, digestive enzyme and temperature conditions, maintain their stability in acidic gastric fluid, and realize the release of probiotics in neutral intestinal fluid. Moreover, the microgel was able to protect BC99 against H2O2 and antibiotics. This work provides a pH-responsive co-loaded microgels for BC99 and PCs, and has the potential in the loading and delivery of other probiotics and polyphenols.

Optimization of methylene blue dye degradation using heterogeneous Fenton-like reaction with Fe3O4 nanoparticles/PVDF macrospheres: A response surface methodology approach

Microplastics (MPs) are increasingly recognised for their significant impact on the environment and human health. Understanding MPs is crucial to grasp their widespread presence in various environmental areas. The unique properties of MPs, such as their small size, durability, and potential to adsorb and transport environmental pollutants, underscore the necessity of studying their characteristics. This study aims to investigate the physical and chemical characteristics of polypropylene microplastics (PPMPs) and address the dispersion stability issues associated with them. The PPMPs were characterised using scanning electron microscopy (SEM), revealing a surface structure marked by cracks, fractures, and a rough texture. The PPMPs were observed as irregularly shaped, white particles. Their size distribution spans from 14 to 96 µm, with a mean size of 50.00 µm. Fourier Transform Infrared Spectroscopy (FTIR) confirmed the presence of polypropylene functional groups, specifically identifying characteristic peaks at 2952-2846 cm-1 and 1456 -1376 cm-1, indicating C-H stretching and bending vibrations, respectively, with additional peaks suggesting degradation. The effect of different concentrations of sodium lauryl sulfate (SLS) on PPMPs dispersion indicated that 5% SLS led to superior dispersion of PPMPs, thereby addressing the stability issue. These findings provide comprehensive insights into the physical and chemical attributes of PPMPs and their dispersion stability, offering a foundation for informed environmental assessments and the development of effective mitigation strategies.

Formulation and Characteristics of Film Forming Hydrogel (FFH) of Yellow Root Extract (Arcangelisia Flava (L.) Merr.) As Wound Healing for Burns

The aim of this study was to determine the effect of different concentrations of yellow root extract (Arcangelisia Flava (L.) Merr.) and film-forming hydrogel (FFH) polymer on the physicochemical characteristics and stability of the preparation. The method for making FFH is carried out using varying concentrations (%) of 0,5 extract, 1, and 1,5, as well as variations in concentration (%) of PVP and PVA (2,8:15,2; 2,8:17,2; 2,8:19,2). F1 – F9 were evaluated for physicochemical characteristics, including organoleptic, pH, spreadability, adhesive power, drying time, viscosity, stickiness, and mechanical properties. Next, freeze-thaw stability and data analysis were carried out using the LSD test at a confidence level of 95%. This research indicates that the FFH formulation of yellow root extract has good characteristics in the F1 formula with an extract concentration of 0.5% and a concentration (%) of PVP and PVP polymer (2.8:15.2), which meets all physicochemical characteristic tests. The results of data analysis using SPSS showed that the extract concentration and polymer concentration had a significant effect (0.05&lt;0.05) on the characteristics and stability of the Yellow Root Extract Film Forming Hydrogel (FFH) preparation.

Effects of copper sulphate stress on the morphological and biochemical characteristics of Spinacia oleracea and Avena sativa

Plants are subjected to various biotic and abiotic stresses that significantly impact their growth and productivity. To achieve balanced crop growth and yield, including for leafy vegetables, the continuous application of micronutrient is crucial. This study investigates the effects of different concentrations of copper sulphate (0, 75, 125, and 175 ppm) on the morphological and biochemical features of Spinacia oleracea and Avena sativa. Morphological parameters such as plant height, leaf area, root length, and fresh and dry weights were optimized at a concentration of 75 ppm copper sulfate. At this concentration, chlorophyll a & b levels increased significantly in Spinacia oleracea (462.9 and 249.8 \U0001d707\U0001d454/\U0001d454), and Avena sativa (404.7 and 437.63\U0001d707\U0001d454/\U0001d454). However, carotenoid content and sugar levels in Spinacia oleracea were negatively affected, while sugar content in Avena sativa increased at 125 ppm (941.6 µg/ml). Protein content increased in Spinacia oleracea (75 ppm, 180.3 µg/ml) but decreased in Avena sativa. Phenol content peaked in both plants at 75 ppm (362.2 and 244.5 µg/ml). Higher concentrations (175 ppm) of copper sulfate reduced plant productivity and health. Plants exposed to control and optimal concentrations (75 and 125 ppm) of copper sulpate exhibited the best health and growth compared to those subjected to higher concentrations. Maximum plant height, leaf area, root length, fresh and dry weights were observed at lower concentrations (75 and 125 ppm) of copper sulfate, while higher concentrations caused toxicity. Optimal copper sulfate levels enhanced chlorophyll a, chlorophyll b, total chlorophyll, protein, and phenol contents but inhibited sugar and carotenoid contents in both Spinacia oleracea and Avena sativa. Overall, increased copper sulfate treatment adversely affected the growth parameters and biochemical profiles of these plants.

Salvia Miltiorrhiza Injection Inhibited the Proliferation of AML Cells by Inducing Apoptosis through the p38MAPK Pathway.

The purpose of this study was to explore the antitumor effect and mechanism of Salvia miltiorrhiza injection (SMI) on acute myeloid leukemia (AML) cells in vitro and in vivo. Bioinformatics was used to detect c-Myc mRNA expression in AML patients in the Oncomine database. qRT‒PCR and western blotting were used to detect the mRNA and protein expression of c-Myc and HOXA5 in clinical samples. Different concentrations (6.25, 12.5, 25, 50 and 100 μg/mL) of SMI were added to KG1a and HL60 cells for 24, 48 and 72 h to determine the IC50 value of SMI. A CCK-8 assay was used to detect the effects of different concentrations of SMI and different treatment times on the proliferation of KG1a and HL60 cells. The indicated concentrations of SMI and SB203580 were used to treat KG1a and HL60 cells. The cell cycle distribution was determined by flow cytometry. The percentage of apoptotic cells was detected by Hoechst 33258 staining and flow cytometry. qRT‒PCR was performed to detect the mRNA expression of p38, c-Myc and HOXA5 in KG1a and HL60 cells. Western blotting was used to detect the protein expression of p38, p-p38, c-Myc, HOXA5, cCaspase 3 and cPARP in KG1a and HL60 cells. AutoDock software was used to analyze the molecular docking of the three main active components of SMI with c-Myc. AutoDock analysis revealed that the binding effect of molecular leisure was evaluated by binding energy, and a binding energy <-5 kcal/mol was considered good. SMI decreased the mRNA and protein expression of c-Myc and HOXA5. SMI significantly inhibited the proliferative activity of KG1a and HL60 cells and induced their apoptosis. However, SMI had no significant effect on the cell cycle distribution of KG1a and HL60 cells. With increasing SMI concentrations, the p-p38/p38 ratio increased, while the protein expression of c-Myc and HOXA5 decreased, and the protein expression of cCaspase and cPARP increased. However, SB203580 intervention in addition to SMI reversed these changes. Tanshinone IIA, cryptanshinone and salvianolic acid B can bind to multiple sites of c-Myc. In summary, SMI could be used for the treatment of acute leukemia, and its mechanism may be related to activation of the p38MAPK signaling pathway.

Improvement of fire safety for viscose fabrics based on phytic acid modified tea polyphenols complexed iron ions

In this work, a multifunctional finishing agent named as PATFe was prepared from phytic acid (PA), tea polyphenols (TP), and Fe3+. The optimum weight ratio of PA to TP was determined by exploring the effect on flame retardant and tensile properties of viscose fabrics. Then, the effects of different concentrations of iron ions on the flame retardant and tensile properties of viscose fabrics were further investigated, and finally, multifunctional viscose fabrics, PATFe-9, were prepared. The system was investigated to confer the multifunctional effects on the flame retardant, bacteriostatic, and UV-resistant properties of viscose fabrics under the condition of lower weight gains (about 6.0 wt%). The limiting oxygen index of PATFe-9 reached 33.7 % with a weight gain of 6.1 wt%, and PATFe-9 had an inhibition effect against Staphylococcus aureus, and the ultraviolet protection factor value reached 67. It is worth noting that the breaking force retention rate of this system reached 100 %, which greatly improves the scope of use and added value of viscose fabrics.

Enhancing Cryopreserved Sperm Quality in Chinese Rare Minnow Gobiocypris rarus: The Impact of Antifreeze Proteins.

The Chinese rare minnow (Gobiocypris rarus), an important model fish in China, faces endangerment in the wild. Sperm cryopreservation facilitates the development of new strains and germplasm conservation, but the quality of its cryopreserved sperm remains low. This study evaluates the protective effects of different concentrations of antifreeze proteins (AFP I and AFP III) on the cryopreservation of Chinese rare minnow sperm. Cryopreserved sperm showed significant declines in progressive motility, curvilinear velocity (VCL), average path velocity (VAP), and lifespan compared to fresh sperm, except for straight-line velocity (VSL). The cryomedium containing 10 μg/mL AFP I improved these parameters to their highest levels. However, no significant difference was found in progressive motility and kinetic parameters between cryopreserved sperm with and without AFPs. Cryopreserved sperm with 10 μg/mL AFP I showed the highest plasma membrane integrity, mitochondrial activity, and DNA integrity, significantly better than without AFPs; importantly, the fertilization rate of cryopreserved sperm with 10 μg/mL AFP I was not significantly different from that of fresh sperm. These results indicate that the addition of 10 μg/mL AFP I to the cryomedium for Chinese rare minnow sperm does not improve kinetic parameters but significantly enhances sperm quality, aiding in its new strain development and germplasm conservation.
