Associations of exposure to disinfection by-products with blood coagulation parameters among women: Results from the Tongji reproductive and environmental (TREE) study

BackgroundExperimental studies have shown that disinfection byproducts (DBPs) induce coagulotoxicity, but human evidence is scarce. ObjectiveThis study aimed to explore the relationships of DBP exposures with blood coagulation parameters. MethodsAmong 858 women from the Tongji Reproductive and Environmental (TREE) study, urinary dichloroacetic acid (DCAA) and trichloroacetic acid (TCAA) were detected as internal biomarkers of DBP exposures. We measured activated partial thromboplastin time (APTT), fibrinogen (Fbg), international normalized ratio (INR), prothrombin time (PT), and thrombin time (TT) as blood coagulation parameters. Multivariable linear regression models were utilized to estimate the relationships between urinary DCAA and TCAA and blood coagulation parameters. The effect modifications by demographic and lifestyle characteristics were further explored. ResultsElevated tertiles of urinary DCAA concentrations were associated with increased PT and INR (11.29%, 95% CI: 1.66%, 20.92% and 0.99%, 95% CI: 0.08%, 1.90% for the third vs. first tertile, respectively; both P for trends < 0.05). Stratification analysis showed that the positive associations were only observed among younger (< 30 years), leaner (body mass index < 24.0 kg/m2), and non-passive smoking women. Moreover, elevated tertiles of urinary TCAA concentrations in positive associations with PT and INR were observed among younger women (17.89%, 95% CI: 2.50%, 33.29% and 1.82%, 95% CI: 0.34%, 3.30% for the third vs. first tertile, respectively; both P for trends < 0.05) but not among older women (both P for interactions < 0.05). ConclusionHigher levels of urinary DCAA and TCAA are associated with prolonged clotting time among women.

Alkaline chlorination of drinking water: A trade-off between genotoxicity control and trihalomethane formation

The pH of chlorination is an important factor affecting the formation of disinfection byproducts (DBPs). In this study, we discovered that the genotoxicity induced by chlorination can be effectively reduced under alkaline conditions. As the pH of chlorination increased from 6.5 to 8.5, the genotoxicity of investigated waters reduced by ∼30–90 %. By assessing the genotoxicity of the mixture of measured DBPs, it was found that the contribution of measured DBPs to the overall genotoxicity was lower than 5 %, and the significant reduction of genotoxicity was largely associated with unknown DBPs. The result of Pearson's correlation analysis indicated that humified organics and soluble microbial byproducts were likely responsible for the genotoxicity, and their derived genotoxic compounds (i.e., unknown DBPs) tended to decompose during alkaline chlorination. However, the control of genotoxicity by alkaline chlorination was achieved at the expense of promoting trihalomethane (THM) formation. The highest genotoxicity reduction (93 %) was observed for chlorinated granular activated carbon-treated waters, but the formation of THMs was promoted to a level approaching that in untreated waters. The inconsistent trend of overall genotoxicity and THM concentration during alkaline chlorination suggested the inadequacy of THMs as metric for DBP exposure, and considerations should also be given to the toxicity of bulk water in addition to regulated DBPs.

Effects of disinfection by-products in swimming pool environments on the immunological mechanisms of respiratory diseases.

Swimming in pools is a popular and healthy recreational activity. However, potential adverse health effects from disinfection byproduct (DBP) exposure in pool water are concerning. This study evaluated how such DBP exposure affects the respiratory system. DBP exposure was simulated with an animal-specific pool environment model. Experimental animals were exposed to DBPs for a specified duration and frequency over 4 weeks. The wet and dry weights of murine lungs were measured, with no significant differences observed. There were no significant differences in interkeukin (IL)-2/4/10, and interferon-γ levels. However, IL-6 expression decreased in the experimental group. To investigate the effects of DBP exposure on immune cell response, various samples, such as bronchoalveolar lavage fluid, lymph nodes, spleen, and thymus, were collected for T-cell isolation and fluorescence-activated cell sorting. Asthma-related blood cell distribution was analyzed using a complete blood count test; no significant differences were found. Thus, DBP exposure through this model did not induce substantial lung tissue damage, major alterations in cytokine expression (besides IL-6), significant immune cell responses, or changes in asthma-associated blood cell distribution. However, considering earlier results, future studies should focus on specific types, intensity, and duration of exercise that could affect DBP exposure-related immune-inflammatory responses.

Associations between urinary biomarkers of exposure to disinfection byproducts and semen parameters: A repeated measures analysis

Toxicological studies have demonstrated that disinfection byproducts (DBPs), particularly haloacetic acids, cause testicular toxicity. However, evidence from human studies is sparse and inconclusive. This study included 1230 reproductive-aged men from the Tongji Reproductive and Environmental (TREE) cohort to investigate the associations between repeated measures of DBP exposures and semen parameters. Urinary dichloroacetic acid (DCAA) and trichloroacetic acid (TCAA) as biomarkers of DBP exposures and semen parameters in up to three samples from each man were assessed. The linear mixed effect models were applied to explore the associations between urinary biomarkers of DBP exposures and semen parameters. We found inverse associations of urinary DCAA with sperm count, progressive motility, and total motility (e.g., −14.86%; 95% CI: −19.33%, −10.15% in sperm total motility for the highest vs. lowest quartiles; all P for trends < 0.05). Moreover, urinary TCAA modeled as a continuous variable was negatively associated with sperm progressive motility and total motility, while the inverse associations across increasing urinary TCAA quartiles were seen among leaner men (BMI < 25 kg/m2). Exposure to DBPs reflected by urinary DCAA and TCAA was inversely associated with sperm motility and such effects were more evident among leaner men.

Risks of obstructive genitourinary birth defects in relation to trihalomethane and haloacetic acid exposures: expanding disinfection byproduct mixtures analyses using relative potency factors.

Some disinfection byproducts (DBPs) are teratogens based on toxicological evidence. Conventional use of predominant DBPs as proxies for complex mixtures may result in decreased ability to detect associations in epidemiological studies. We assessed risks of obstructive genitourinary birth defects (OGDs) in relation to 12 DBP mixtures and 13 individual component DBPs. We designed a nested registry-based case-control study (210 OGD cases; 2100 controls) in Massachusetts towns with complete quarterly 1999-2004 data on four trihalomethanes (THMs) and five haloacetic acids (HAAs). We estimated temporally-weighted average DBP exposures for the first trimester of pregnancy. We estimated adjusted odds ratios (aORs) and 95% confidence intervals (CIs) for OGD in relation to individual DBPs, unweighted mixtures, and weighted mixtures based on THM/HAA relative potency factors (RPF) from animal toxicology data for full-litter resorption, eye defects, and neural tube defects. We detected elevated aORs for OGDs for the highest of bromodichloromethane (aOR = 1.75; 95% CI: 1.15-2.65), dibromochloromethane (aOR = 1.71; 95% CI: 1.15-2.54), bromodichloroacetic acid (aOR = 1.56; 95%CI: 0.97-2.51), chlorodibromoacetic acid (aOR = 1.97, 95% CI: 1.23-3.15), and tribromoacetic acid (aOR = 1.90; 95%CI: 1.20-3.03). Across unweighted mixture sums, the highest aORs were for the sum of three brominated THMs (aOR = 1.74; 95% CI: 1.15-2.64), the sum of six brominated HAAs (aOR = 1.43; 95% CI: 0.89-2.31), and the sum of nine brominated DBPs (aOR = 1.80; 95% CI: 1.05-3.10). Comparing eight RPF-weighted to unweighted mixtures, the largest aOR differences were for two HAA metrics, which both were higher with RPF weighting; other metrics had reduced or minimally changed ORs in RPF-weighted models.

Enhanced disinfection byproducts formation by fine iron particles intercepted in household point-of-use facilities.

To cope with the demand for good-quality potable water, household point-of-use (POU) facilities such as polypropylene cotton filters (PCFs) are widely used. However, the behaviors of new and used PCFs under discoloration are unclear. In this study, we found that new PCF did not effectively intercept particles under discoloration within the initial 5 d of inflow. In addition, the particles, especially the fine ones, accumulated in the long-used PCF exacerbated the risks of disinfection byproducts (DBPs) and microbes. The concentrations of trihalomethanes (THMs) and haloacetonitriles (HANs) in the effluent run through the PCF all increased over time; interestingly, all sharply increased after 5 d in accordance with the decrease in effluent iron particles. During this stage, maximum increases rate of 117.89% in THMs and 75.12% in HANs were observed. For haloacetic acids (HAAs), it served as the dominant contaminants, with concentrations approximately 10-fold greater than those of THMs and HANs. The increase showed that used PCFscould exacerbate the risks in DBPs exposure. Adenosine triphosphate (ATP) also showed a similar trend, with a maximum increase from 0.0033 to 0.0055 nmol/mL. Thus, PCFs can act only as pretreatment units and should be replaced after yellow water events. This study offers important guidance for PCF usage in drinking water purification, especially under discoloration.

Associations between Urinary Concentrations of Disinfection Byproducts and in Vitro Fertilization Outcomes: A Prospective Cohort Study in China.

Experimental studies show that disinfection byproducts (DBPs) can inhibit oocyte maturation, decrease fertilization capacity, and impair embryo development, but human evidence is lacking. We aimed to evaluate the associations between exposure to drinking water DBPs and in vitro fertilization (IVF) outcomes. The study included 1,048 women undergoing assisted reproductive technology (ART) treatment between December 2018 and January 2020 from a prospective cohort study, the Tongji Reproductive and Environmental study in Wuhan, China. Exposure to DBPs was assessed by dichloroacetic acid (DCAA) and trichloroacetic acid (TCAA) in up to four urine samples, which were collected on the day of both enrollment and oocyte retrieval. Multivariable generalized linear mixed models, accounting for multiple IVF cycles per woman, were applied to evaluate the associations between urinary biomarkers of DBP exposures and IVF outcomes. Stratified analyses were used to explore the potential effect modifiers. The included 1,048 women underwent 1,136 IVF cycles, with 960 (91.6%), 84 (8.0%), and 4 (0.4%) women contributing one cycle, two cycles, and three cycles, respectively. We found that elevated quartiles of urinary DCAA and TCAA concentrations were associated with reduced numbers of total oocytes and metaphase II oocytes and that urinary DCAA concentrations with a lower proportion of best-quality embryos (all p for ). Moreover, elevated quartiles of urinary DCAA concentrations were associated with decreased proportions of successful implantation, clinical pregnancy, and live birth (14%, 15%, and 15% decreases in adjusted means comparing the extreme quartiles, respectively; all p for ). Stratification analyses showed that the inverse associations of urinary TCAA concentrations with multiple IVF outcomes were stronger among women y of age (p for ). Exposure to drinking water DBPs was inversely associated with some IVF outcomes among women undergoing ART treatment. Degradation and dechlorination of trichloroacetic acid induced by an in situ 222 nm KrCl* excimer radiation

Since the coronavirus disease 2019 (COVID-19) pandemic epidemic, the excessive usage of chlorinated disinfectants raised the substantial risks of disinfection by-products (DBPs) exposure. While several technologies may remove the typical carcinogenic DBPs, trichloroacetic acid (TCAA), their application for continuous treatment is limited due to their complexity and expensive or hazardous inputs. In this study, degradation and dechlorination of TCAA induced by an in situ 222 nm KrCl* excimer radiation as well as role of oxygen in the reaction pathway were investigated. Quantum chemical calculation methods were used to help predict the reaction mechanism. Experimental results showed that UV irradiance increased with increasing input power and decreased when the input power exceeded 60 W. Decomposition and dechlorination were simultaneously achieved, where around 78% of TCAA (0.62 mM) can be eliminated and 78% dechlorination within 200 min. Dissolved oxygen showed little effect on the TCAA degradation but greatly boosted the dechlorination as it can additionally generate hydroxyl radical (•OH) in the reaction process. Computational results showed that under 222 nm irradiation, TCAA was excited from S0 to S1 state and then decayed by internal crossing process to T1 state, and a reaction without potential energy barrier followed, resulting in the breaking of C–Cl bond and finally returning to S0 state. Subsequent C–Cl bond cleavage occurred by a barrierless •OH insertion and HCl elimination (27.9 kcal/mol). Finally, the •OH attacked (14.6 kcal/mol) the intermediate byproducts, leading to complete dechlorination and decomposition. The KrCl* excimer radiation has obvious advantages in terms of energy efficiency compared to other competitive methods. These results provide insight into the mechanisms of TCAA dechlorination and decomposition under KrCl* excimer radiation, as well as important information for guiding research toward direct and indirect photolysis of halogenated DBPs.

Challenges of point-of-use devices in purifying tap water: The growth of biofilm on filters and the formation of disinfection byproducts

Currently, polypropylene (PP) and granular activated carbon (GAC) are widely used as the filters in point-of-use devices (POUs). However, the effect of biofilms formation in aged filters and subsequent formation/removal of disinfection byproducts (DBPs) during the use of POUs on purifying tap water remains unclear. In this study, the influence of filter replace event and filter usage time on the removal of DBPs by pristine or aged filters, such as trihalomethanes (THMs) and haloacetic acids (HAAs), was investigated. Additionally, the effect of biofilms-attached PP filters and GAC filters on DBPs concentration in treated water was explored. The results showed that POUs effectively removed over 60% of THMs and HAAs from tap water, while the removal efficiency decreased with the operation time. The increase in usage time of PP filters led to elevated levels of DBPs. Moreover, the pristine pre-GAC filters removed 12.31% of THMs and 6.62% of HAAs, respectively, while aged GAC filters enhanced the removal of HAAs. SEM and high-throughput sequencing analysis confirmed the attached growth of biofilms on aged PP and GAC filters. The biofilms growth on PP filters led to the formation of THMs and HAAs, and the formed DBPs increased with increasing influent chlorine concentration, lengthened filters usage time and stagnation time. Nevertheless, the DBPs in effluent of biofilms-attached GAC filters increased with increasing influent chlorine concentration and filter usage time while decreased with stagnation time. These results indicated that growth of biofilms commonly observed in filters of POUs leads to the formation of DBPs and increases the risk of DBPs exposure to users of POU devices.

Variations in Disinfection By-Product Precursors Bromide and Total Organic Carbon Among U.S. Watersheds

Watersheds with relatively higher concentrations of disinfection by-product (DBP) precursors are of concern for public water systems (PWSs) seeking to control DBP exposure risk. In this study, the occurrence of bromide and total organic carbon (TOC), which are important DBP precursors, was evaluated in PWS source waters on a watershed basis at the hydrologic unit code 2 (HUC2) level. Variations were examined across source water type (e.g., surface water or groundwater) and watershed, and temporally based on seasonality as well as differences between the late 1990s and the years 2018–2020. The median bromide concentrations were higher in groundwater systems compared with surface water (by 25 μg/L), whereas the median TOC concentrations were higher in surface water systems compared with groundwater (by 1.7 mg/L). Surface water sources in multiple midwestern and western watersheds had a combination of elevated bromide and TOC (relative to median values of 21 μg/L and 2.5 mg/L, respectively), which was especially pronounced in the Texas-Gulf and Rio Grande watersheds. None of the HUC2 watersheds had elevated levels of both bromide and TOC in groundwater sources (median values of 50 μg/L and 1.0 mg/L, respectively). Source water bromide median watershed concentrations for very large systems were ∼20% lower in 2018–2020 than in 1998, however, more than 40% higher at the 90th percentile, whereas source water TOC median watershed concentrations for very large systems were ∼25% higher in 2018–2020 than in 1998 but less than 10% higher at the 90th percentile. These temporal differences may be related to the influence of climate change and other anthropogenic effects. In addition, many individual PWSs showed substantial change in bromide and TOC concentrations between those time periods. For example, about 7% of the PWSs had at least double the bromide concentrations, and 5% at least double the TOC concentrations, in 2018–2020 than in 1998.

REGULATORY ISSUES ON WATER DISINFECTION AND THE IMPACT OF DBP GENERATION ON DRINKING WATER QUALITY. Drinking water disinfection has been viewed as one of the major advances in public health protection in history. However, while it has been very successful in assuring microbiological safety, disinfection also brought relevant concerns about chemical safety and the potential unintended health hazards associated with disinfection byproduct (DBP) exposure. DBPs represent one of the most important groups of chemical compounds to which humans are exposed through drinking water, as epidemiological studies have consistently associated long-term exposure with multiple adverse health effects. It is established that disinfection is an essential component of providing microbiologically safe drinking water and shall never be compromised. Yet, chemical safety regarding DBP exposure remains a largely uncovered and highly discussed topic. Several DBPs have been reported in the past few decades, yet research on formation, toxicity and occurrence remains scarce for most compounds. Also, for each identified DBP, many remain undiscovered. In view of an ever-growing list of DBPs and the existing gaps in available research, identifying overall driving agents of toxicity is still a major challenge for public health protection. This review attempts to provide an insight on contemporary issues regarding the overall safety of drinking water and the trade-off between microbiological and chemical risks, considering current challenges in water and sanitation access and existing regulations.

Emerging Disinfection Byproduct 2,6-Dichlorobenzoquinone-Induced Cardiovascular Developmental Toxicity of Embryonic Zebrafish and Larvae: Imaging and Transcriptome Analysis.

Epidemiological studies have observed the potential association of water disinfection byproduct (DBP) exposure with cardiac defects. Aromatic DBPs represent a significant portion of total DBPs, but their effects on cardiovascular development are unclear. In this study, we examined the effects of an aromatic DBP, 2,6-dichlorobenzoquinone (DCBQ), on the cardiovascular development of zebrafish embryos. After exposure to 2, 4, and 8 μM DCBQ, morphological images of growing zebrafish embryos clearly showed cardiovascular malformation. Fluorescent images of transgenic zebrafish strains with fluorescently labeled heart and blood vessels show that DCBQ exposure resulted in deformed atrium-ventricle looping, degenerated abdomen and trunk vessels, pericardial edema, and decreased blood flow. Furthermore, the expression of the marker gene myl7 (essential for the differentiation and motility of cardiomyocytes) was inhibited in a dose-dependent manner by DCBQ exposure. Finally, transcriptome analysis found that in the 4 μM DCBQ exposure group, the numbers of differentially expressed genes (DEGs) were 113 (50 upregulated and 63 downregulated) at 24 hpf, 2123 (762 upregulated and 1361 downregulated) at 48 hpf, and 61 (11 upregulated and 50 downregulated) at 120 hpf; in the 8 μM DCBQ exposure group, the number of DEGs was 1407 (647 upregulated and 760 downregulated) at 120 hpf. The FoxO signaling pathway was significantly altered. The in vivo results demonstrate the effects of 2,6-DCBQ (0-8 μM) on cardiovascular development, contributing to the understanding of the developmental toxicity of aromatic DBP halobenzoquinones (HBQs).

Multi-omics signatures of exposure to disinfection by-products in swimming

Background and aims: Untargeted HRMS metabolomics provides an agnostic global profiling of small molecules in a biological system capturing an overview of exposure to exogenous compounds, and their effects on metabolic pathways, rendering an essential tool for characterising the exposome. Disinfection by-products (DBPs) are a complex mixture of halogenated organic compounds present in drinking and swimming pool water. Although several epidemiological studies have pointed their potential to induce adverse health outcomes such as bladder cancer, their mixture effect and the downstream molecules perturbed by their exposure have been poorly explored. In this work, we aim to identify molecular signatures of exposures to mixtures of DBPs in individuals with measured exposures. Methods: In PISCINA-II study, metabolomics, transcriptomics, proteomics in serum, and levels of DBPs in exhaled breaths, were measured in 60 subjects before and after swimming. Pre-processing of HRMS data from PISCINA-II study was customised to include more halogenated compounds. Multivariate normal (MVN) and partial least square (PLS) models were used to identify metabolic features associated with DBP exposures. Conditional correlation networks were constructed to understand the holistic relationship amongst the metabolic features and their relationship with proteomics and transcriptomics. Results: 98 metabolic features were identified to be associated with exposure to DBP mixture, of which 39 were not identified in the previous analysis without specific reprocessing and inclusion of halogenated compounds. In conditional network analysis, the 98 features showed cluster structures, suggesting they could contribute to different downstream biological pathways. Multi-omic networks showed that transcriptomic and proteomic signals were differentially connected to the different clusters of the metabolic signatures. Conclusions: New data processing approach allowed identification of novel metabolic signatures of DBP mixture exposure. Conditional correlation network implied the functional proximity of these signatures with other measured omic signals. Dietary intakes of trihalomethanes and haloacetic acids from ready-to-eat vegetables in taiwan.

Chlorine-based disinfectants are often used to sanitize fruit and vegetables to produce a product called ready-to-eat (RTE) vegetables. During the disinfection process, disinfection byproducts (DBPs), such as trihalomethanes (THMs) and haloacetic acids (HAAs), might be formed via chlorination. To determine the amounts of DBPs that occur in RTE vegetables in Taiwan, an analytical method which can detect THMs and HAAs simultaneously was developed for this study. For HAAs, dimethyl sulfate (DMS) was first added into the sample as derivatization reagent and tetrabutylammonium hydrogen sulfate (TBA-H2SO4) was used as the ion-pairing agent to improve the derivatization process. Afterwards, the solid-phase microextraction (SPME) procedure coupled with gas chromatography with tandem mass spectrometers (GC/MS/MS) was performed to measure the HAAs derivatives and THMs in the sample. A total of 92 single RTE ingredients were analyzed in this study. Among various THMs and HAAs, the results showed that dibromochloromethane (21%) and dichloroacetic acid (12%) had the highest detection rates, respectively. Compared with fruits, vegetables were more easily to contain DBPs. For adults in Taiwan, the maximum daily exposure of THMs and HAAs estimated via the consumption of RTE vegetables were 28.53 and 77.83 μg, respectively. The findings from this study suggest that the exposure of DBPs from RTE vegetables is an important food safety issue in Taiwan.

Insights to estimate exposure to regulated and non-regulated disinfection by-products in drinking water

BackgroundKnowledge about human exposure and health effects associated with non-routinely monitored disinfection by-products (DBPs) in drinking water is sparse.ObjectiveTo provide insights to estimate exposure to regulated and non-regulated DBPs in drinking water.MethodsWe collected tap water from homes (N = 42), bottled water (N = 10), filtered tap water with domestic activated carbon jars (N = 6) and reverse osmosis (N = 5), and urine (N = 39) samples of participants from Barcelona, Spain. We analyzed 11 haloacetic acids (HAAs), 4 trihalomethanes (THMs), 4 haloacetonitriles (HANs), 2 haloketones, chlorate, chlorite, and trichloronitromethane in water and HAAs in urine samples. Personal information on water intake and socio-demographics was ascertained in the study population (N = 39) through questionnaires. Statistical models were developed based on THMs as explanatory variables using multivariate linear regression and machine learning techniques to predict non-regulated DBPs.ResultsChlorate, THMs, HAAs, and HANs were quantified in 98–100% tap water samples with median concentration of 214, 42, 18, and 3.2 μg/L, respectively. Multivariate linear regression models had similar or higher goodness of fit (R2) compared to machine learning models. Multivariate linear models for dichloro-, trichloro-, and bromodichloroacetic acid, dichloroacetonitrile, bromochloroacetonitrile, dibromoacetonitrile, trichloropropnanone, and chlorite showed good predictive ability (R2 = 0.8–0.9) as 80–90% of total variance could be explained by THM concentrations. Activated carbon filters reduced DBP concentrations to a variable extent (27–80%), and reverse osmosis reduced DBP concentrations ≥98%. Only chlorate was detected in bottled water samples (N = 3), with median = 13.0 µg/L. Creatinine-adjusted trichloroacetic acid was the most frequently detected HAA in urine samples (69.2%), and moderately correlated with estimated drinking water intake (r = 0.48).SignificanceFindings provide valuable insights for DBP exposure assessment in epidemiological studies. Validation of predictive models in a larger number of samples and replication in different settings is warranted.Impact statementOur study focused on assessing and describing the occurrence of several classes of DBPs in drinking water and developing exposure models of good predictive ability for non-regulated DBPs.

Feel the Burn: Disinfection Byproduct Formation and Cytotoxicity during Chlorine Burn Events

Nitrification and biofilm growth within distribution systems remain major issues for drinking water treatment plants utilizing chloramine disinfection. Many chloraminated plants periodically switch to chlorine disinfection for several weeks to mitigate these issues, known as "chlorine burns". The evaluation of disinfection byproduct (DBP) formation during chlorine burns beyond regulated DBPs is scarce. Here, we quantified an extensive suite of 80 regulated and emerging, unregulated DBPs from 10 DBP classes in drinking water from two U.S. drinking water plants during chlorine burn and chloramination treatments. Total organic halogen (TOX), including total organic chlorine, total organic bromine, and total organic iodine, was also quantified, and mammalian cell cytotoxicity of whole water mixtures was assessed in chlorine burn waters for the first time. TOX and most DBPs increased in concentration during chlorine burns, and one emerging DBP, trichloroacetaldehyde, reached 99 μg/L. THMs and HAAs reached concentrations of 249 and 271 μg/L, respectively. Two highly cytotoxic nitrogenous DBP classes, haloacetamides and haloacetonitriles, increased during chlorine burns, reaching up to 14.2 and 19.3 μg/L, respectively. Cytotoxicity did not always increase from chloramine treatment to chlorine burn, but a 100% increase in cytotoxicity was observed for one plant. These data highlight that consumer DBP exposure during chlorine burns can be substantial.

Disinfection byproducts in indoor swimming pool water: Detection and human lifetime health risk assessment

Quantification of regulated and emerging disinfection byproducts (DBPs) in swimming pool water, as well as the assessment of their lifetime health risk are limited in China. In this study, the occurrence of regulated DBPs (e.g., trihalomethanes, haloacetic acids) and emerging DBPs (e.g., haloacetonitriles, haloacetaldehydes) in indoor swimming pool water and the corresponding source water at a city in Eastern China were determined. The concentrations of DBPs in swimming pool water were 1-2 orders of magnitude higher than that in source water. Lifetime cancer and non-cancer risks of DBPs stemming from swimming pool water were also estimated. Inhalation and dermal exposure were the most significant exposure routes related to swimming pool DBP cancer and non-cancer risks. For the first time, buccal and aural exposure were considered, and were proven to be important routes of DBP exposure (accounting for 17.9%-38.9% of total risk). The cancer risks of DBPs for all swimmers were higher than 10−6 of lifetime exposure risk recommended by United States Environmental Protection Agency, and the competitive adult swimmers experienced the highest cancer risk (7.82 × 10−5). These findings provide important information and perspectives for future efforts to lower the health risks associated with exposure to DBPs in swimming pool water.

Occurrence of Disinfection By-Products in Swimming Pools in the Area of Thessaloniki, Northern Greece. Assessment of Multi-Pathway Exposure and Risk.

This study investigated the occurrence of disinfection by-products (DBPs) (trihalomethanes (THMs), haloacetic acids (HAAs), halonitriles (HANs), halonitromethane (TCNM) and haloketones (HKs)) in different type of swimming pools in the area of Thessaloniki, northern Greece by employing the EPA methods 551.1 and 552.3. Moreover, general water quality parameters (pH, residual chlorine, dissolved organic carbon, UV254 absorption, total nitrogen, alkalinity and conductivity) were also measured. The concentrations of DBPs showed great variability among swimming pools as well as within the same pool between sampling campaigns. HAAs exhibited the highest concentrations followed by THMs, HANs, TCNM and HKs. Exposure doses for four age groups (3–<6 y, 6–<11 y, 11–<16 y and adults) were calculated. Route-specific exposures varied among DBPs groups. Inhalation was the dominant exposure route to THMs and TCNM (up to 92–95%). Ingestion and dermal absorption were the main exposure routes to HAAs (40–82% and 18–59%, respectively), depending on the age of swimmers. HANs contributed up to 75% to the calculated cytotoxicity of pool water. Hazard indices for different exposure routes were <1, suggesting non-carcinogenic risk. Inhalation posed the higher carcinogenic risk for THMs, whereas risk via oral and dermal routes was low. Ingestion and dermal contact posed the higher risk for HAAs. Risk management strategies that minimise DBPs exposure without compromising disinfection efficiency in swimming pools are necessary.

Prenatal Exposure to Disinfection Byproducts and Intrauterine Growth in a Chinese Cohort.

Disinfection byproduct (DBP) exposure has been associated with birth size, pregnancy oxidative stress, and other adverse perinatal outcomes. However, little is known about the potential effect of prenatal DBP exposure on intrauterine growth. The present study included 1516 pregnant women from the Xiaogan Disinfection By-Products (XGDBP) birth cohort who were measured for four blood trihalomethanes [i.e., chloroform (TCM), bromodichloromethane (BDCM), dibromochloromethane (DBCM), and bromoform (TBM)] and two urinary haloacetic acids [i.e., dichloroacetic acid (DCAA) and trichloroacetic acid (TCAA)] across pregnancy trimesters. Second- and third-trimester fetal ultrasound measures of the abdominal circumference (AC), head circumference, biparietal diameter, femur length, and estimated fetal weight and birth weight were converted into z-scores. After adjusting for potential confounders, linear mixed models showed a decreasing AC z-score across tertiles of blood brominated THM (Br-THMs, the sum of BDCM, DBCM, and TBM) and total THM (THM4, the sum of Br-THMs and TCM) concentrations (both p for trend <0.01). We also observed a decreasing AC z-score across categories of blood TBM during pregnancy trimesters (p for trend = 0.03). Urinary haloacetic acids were unrelated to fetal growth parameters. In summary, prenatal exposure to THMs, particularly during the first trimester, was associated with reduced fetal abdominal circumference.

Chlorine taste can increase simulated exposure to both fecal contamination and disinfection byproducts in water supplies

Expanding drinking water chlorination could substantially reduce the burden of disease in low- and middle-income countries, but the taste of chlorinated water often impedes adoption. We developed a Monte Carlo simulation to estimate the effect of people's choice to accept or reject drinking water based on chlorine taste and their subsequent exposure to E. coli and trihalomethanes, a class of disinfection byproduct (DBP). The simulation used empirical data from Dhaka, Bangladesh, a megacity with endemic waterborne disease. We drew on published taste acceptability thresholds from Dhaka residents, measured residual chlorine and thermotolerant E. coli inactivation following the addition of six chlorine doses (0.25–3.0 mg/L as Cl2) to untreated piped water samples from 100 locations, and analyzed trihalomethane formation in 54 samples. A dose of 0.5 mg/L, 75% lower than the 2 mg/L dose typically recommended for household chlorination of low-turbidity waters, minimized overall exposure to E. coli. Doses of 1–2 mg/L maximized overall exposure to trihalomethanes. Accounting for chlorine taste aversion indicates that microbiological exposure increases and DBP exposure decreases above certain doses as a higher proportion of people reject chlorinated water in favor of untreated water. Taken together with findings from other modeling analyses, empirical studies, and field trials, our results suggest that taste acceptability should be a critical consideration in establishing chlorination dosing guidelines. Particularly when chlorination is first implemented in water supplies with low chlorine demand, lower doses than those generally recommended for household water treatment can help avoid taste-related objections while still meaningfully reducing contaminant exposure.
