The development of methods for determination the total flavonoids in buds of Populus alba L.

White poplar (Populus alba L., family Saliaceae ) has a similar chemical composition with other species of the genus Populus L. and it can consider as a promising source of raw materials containing phenolic compounds, in particular flavonoids. The pharmacological activity of the buds of pharmacopeial species of genus Populus L., and also of the white poplar is caused by biologically active compounds, mainly of a phenolic nature, including flavonoids (pinostrobin, piriocembrin, quercetin, etc.), phenylpropanoids (caffeic acid, etc.) and simple phenols (salicin).. One of the most well-known biologically active compounds of white poplar is quercetin, for which an antihistamine, anti-inflammatory action has been demonstrated. Differential spectrophotometry carried out in accordance with the OFS was used as a research method.1.2.1.1.0003.15 "Spectrophotometry in the ultraviolet and visible regions". Spectral characteristics of water-alcohol extracts were evaluated on a Specord 40 spectrophotometer (Analytik Jena AG, Germany) in cuvettes with a layer thickness of 10 mm. It was determined that in all electronic spectra of the water-ethanolic extractions from the buds of the white poplar there are two absorption maxima in the area of 290 nm and 370 nm, due to flavanones and flavonols, respectively. It was established that in the electronic spectra of the water-ethanolic extractions from the buds of the white poplar, a significant bathochromic shift of the long-wavelength band in the presence of aluminum chloride is observed, which confirms the presence of flavonoids. Under the conditions of differential spectrophotometry UV spectrum of the water-ethanolic extractions from the buds of the white poplar, an absorption maximum is observed at wavelength of 430 nm, which indicates the expediency of using quercetin in the analysis technique, which has an absorption maximum at a wavelength of 430±2 nm. As a result of the study, the method has been developed for the quantitative determination of the amount of flavonoids in the buds of white poplar using differential spectrophotometry calculated on quercetin at an analytical wavelength of 430 nm. The optimal parameters of extraction of raw materials were determined: extractant – 90% ethyl alcohol, the ratio of "raw material-extractant" – 1:30, extraction time – 60 minutes. The content of the total flavonoids calculated on quercetin in the buds of the white poplar is varied from 0,45±0,02 % to 0.69 ± 0.03%

Efficient two-step excitation energy transfer in artificial light-harvesting antenna based on bacteriochlorophyll aggregates

Chlorosomes of green photosynthetic bacteria are large light-harvesting complexes enabling these organisms to survive at extremely low-light conditions. Bacteriochlorophylls found in chlorosomes self-organize and are ideal candidates for use in biomimetic light-harvesting in artificial photosynthesis and other applications for solar energy utilization. Here we report on the construction and characterization of an artificial antenna consisting of bacteriochlorophyll c co-aggregated with β-carotene, which is used to extend the light-harvesting spectral range, and bacteriochlorophyll a, which acts as a final acceptor for excitation energy. Efficient energy transfer between all three components was observed by means of fluorescence spectroscopy. The efficiency varies with the β-carotene content, which increases the average distance between the donor and acceptor in both energy transfer steps. The efficiency ranges from 89 to 37% for the transfer from β-carotene to bacteriochlorophyll c, and from 93 to 69% for the bacteriochlorophyll c to bacteriochlorophyll a step. A significant part of this study was dedicated to a development of methods for determination of energy transfer efficiency. These methods may be applied also for study of chlorosomes and other pigment complexes.

Monitor Calibrator as an Alternative to Spectrofluorimeter: Determination of Quinine in Beverages and Medicinal Preparations

A possibility of using a monitor calibrator for the determination of luminescent compounds is shown on an example of quinine. The determination is based on the irradiation of a sample with broadband radiation in the visible and near UV spectral regions from a built-in source exciting phosphor molecules and the simultaneous registration of the radiation incident on the detector. Measurement conditions are selected. Quinine can be determined in the range of 60−750 µM, the limit of detection is 20 µM. The determination is not affected by common inorganic ions, as well as sweeteners and acidity regulators present in many beverages. The developed method of determination is applicable to the analysis of carbonated drinks and drugs. Compared to a traditional spectrofluorimeter, monitor calibrator is characterized by compactness, mobility, ability of detecting luminescence in cuvettes of various sizes and shapes, and lower cost.

Zinc Ion-Based Switch-on Fluorescence-Sensing Probes for the Detection of Tetracycline

Tetracycline (TC) is an antibiotic that has been widely used in the animal husbandry. Thus, TC residues may be found in animal products. Developing simple and sensitive methods for rapid screening of TC in complex samples is of great importance. Herein, we demonstrate a fluorescence-sensing method using Zn2+ as sensing probes for the detection of TC. Although TC can emit fluorescence under the excitation of ultraviolet light, its fluorescence is weak because of dynamic intramolecular rotations, leading to the dissipation of excitation energy. With the addition of Zn2+ prepared in tris(hydroxymethyl)amino-methane (Tris), TC can coordinate with Zn2+ in the Zn2+-Tris conjugates to form Tris-Zn2+-TC complexes. Therefore, the intramolecular motions of TC are restricted to reduce nonradiative decay, resulting in the enhancement of TC fluorescence. Aggregation-induced emission effects also play a role in the enhancement of TC fluorescence. Our results show that the linear dynamic range for the detection of TC is 15-300 nM. Moreover, the limit of detection was ~7 nM. The feasibility of using the developed method for determination of the concentration of TC in a complex chicken broth sample is also demonstrated in this work.

Determination of Genotoxic Impurity N-Nitroso-N-methyl-4-aminobutyric Acid in Four Sartan Substances through Using Liquid Chromatography-Tandem Mass Spectrometry.

N-nitroso-N-methyl-4-aminobutyric acid (NMBA) is the third N-nitrosamine impurity found in sartans. Herein, a sensitive and stable LC-MS/MS method with multiple reactions monitoring mode has been developed for the quantitative determination of NMBA in four sartan substances. The effective separation of NMBA and sartan substances was achieved on a C18 column under gradient elution conditions. The mass spectrometry method of the atmospheric pressure chemical ionization source and internal standard method was selected as the quantitative analysis method of NMBA. Then, this proposed LC-MS/MS analysis method was validated in terms of specificity, sensitivity, linearity, accuracy, precision and stability. Good linearity with correlation coefficient over 0.99 was obtained at the NMBA concentration of 3-45 ng/mL, and the limit of quantification was 3 ng/mL. Additionally, the recoveries of NMBA in four sartan substances ranged from 89.9% to 115.7%. The intra-day and inter-day relative standard deviation values were less than 5.0%. In conclusion, this developed determination method for NMBA through liquid chromatography-tandem mass spectrometry showed the characteristics of good sensitivity, high accuracy and precision, which will be of great help for the quantitative analysis of NMBA in sartan products.

Development of Artificial Intelligence Methods for Determination of Methane Solubility in Aqueous Systems

Accurate determinations of water (H2O) content in natural gases especially in the methane (CH4) phase are highly important for chemical engineers dealing with natural gas processes. To this end, development of a high performance model is necessary. Due to importance of the solubility of methane in the aqueous solutions for natural gas industries, two novel models based on the Decision Tree (DT) and Adaptive Neuro-Fuzzy Interference System (ANFIS) have been employed. To this end, a total number of 204 real methane solubility points in aqueous solution containing NaCl under different pressure and temperature conditions have been gathered. The comparisons between predicted solubility values and experimental data points have been conducted in visual and mathematical approaches. The R2 values of 1 for training and testing phases express the great ability of proposed models in calculation of methane solubility in pure water systems.

1-Ethyl-2-pyrrolidone (NEP) is a colorless liquid with ammonia-like odor. It belongs to the organic compounds from the lactam group, i.e., the ethyl derivative of 2-pyrrolidone. 1-Ethyl-2-pyrrolidone, due to similar physicochemical proper-ties, is used in industry as a substitute for 1-methyl-2-pyrrolidone (NMP). It is used as a solvent in polymer, petroche-mical, paint and varnish, and electronic industries. Moreover, it has been used as a cleaning agent for removing paints, varnishes, adhesives, oil or grease. 1-Ethyl-2-pyrrolidone can be absorbed through the skin as well as through inhalation and food. The aim of the this study was to develop and validate a method for determining 1-ethyl-2-pyrrolidone in work-place air. The developed method of NEP determination consists in adsorption of vapors of this substance on coconut shell charcoal, extraction with a dichloromethane and chromatographic analysis of the obtained solution. The study was performed using a gas chromatograph coupled with mass spectrometer (GC-MS), equipped with a polar ZB-WAXplus capillary column (length 60 m, diameter 0.25 mm and the film thickness of the stationary phase 0.5 µm). The developed method is linear in the concentration range of 15.0–320.0 µg/ml, which corresponds to the range of 1.5–32.0 mg/m3 for a 10-L air sample. The analytical method described in this paper makes it possible to determine 1-ethyl-2-pyrrolidone in workplace air in the presence of comorbid substances. The method is precise, accurate and it meets the criteria for procedure for measuring chemical agents listed in Standard No. PN-EN 482. Developed method of determining 1-ethyl- 2-pyrrolidone at workplace air has been recorded as an analytical procedure (see Appendix). EDTA salt modified carbon paste electrode for square wave voltammetric determination of theophylline in pharmaceutical tablet formulation.

In this study, a square wave voltammetric method for determination of theophylline in tablet formulation based on EDTA salt modified carbon paste electrode is presented. CV, FT-IR, and EIS results confirmed modification of the carbon paste with EDTA salt. In contrast to the unmodified carbon paste electrode, the modified carbon paste electrode showed irreversible oxidation of theophylline with considerable current enhancement. Investigation of the effect of scan rate on the Ip and Ep response of the modified electrode for theophylline revealed predominantly diffusion controlled oxidation kinetics. Under the optimized conditions, square wave oxidative peak current of theophylline in pH 7.0 PBS showed linear dependence on concentration in the range 10–200 μM with determination coefficient (R2), limit of detection, and limit of quantification of 0.99782, 0.0257 μM, and 0.0857 μM, respectively. Detection of an amount of theophylline in the analyzed tablet formulation with 1.85% error from its nominal content (120 mg/tablet) confirmed the accuracy of the developed method. Spike and interference recovery results of 98.59%, and 95.7–100%, respectively validated the applicability of the developed method for determination of theophylline content in tablet samples.

Development of continuous flow injection analysis method for determination of oxymetazoline and vancomycin hydrochloride in pharmaceutical preparations

ABSTRACT. Continuous flow injection (CFI)-spectrophotometric method has been developed for oxymetazoline (OXY) and vancomycin hydrochloride (VAN) determination in pure and dosage forms. Sulfadimidine drug was used as a safe chromogenic reagent by diazotization coupling with studied drugs to produce sensitive azo dyes with maximal wavelengths of 498 and 441 nm, respectively, for OXY and VAN drugs. The effect of various chemical and physical conditions on the signal response has been studied. Under optimum conditions, Beer’s law was linear over the concentration range of 5–200 and 6–200 µg/mL with limits of detection of 2.19 and 1.79 µg/mL for OXY and VAN, respectively. The CFI systems gave a sample throughput of more than 120 samples per hour for 150 μL injection volume for both drugs. The proposed methods were used for the estimation of microgram quantities of both drugs in their different dosage forms with great effectiveness. The method's validity and applicability were thoroughly investigated and the recoveries values were compared with those of standard pharmacopeia methods. The specified approaches were used without the excipients interfering in pharmaceutical applications.

Development of method for determination and quantification of Alternaria mycotoxins in food by high resolution mass spectrometry

High-resolution liquid chromatography-mass spectrometry (LC-HRMS) is a highly sensitive, specific and accurate method for the simultaneous detection and quantification of three mycotoxins Alternaria (Alternariol (AOH), Alternariol Monomethyl Ete) (AME), Tenuazonic acid (TeA)). Chromatographic conditions include: column reversed phase C18, mobile phases in gradient mode with two channels of H2O and MeOH containing 0.1 % formic acid and 10 mM ammonium formate. HRMS high resolution mass spectrometer detector with negative electron ionization (ESI-) source is used. Samples were cleaned and enriched using an Oasis HLB solid phase extraction column. The method has a limit of quantitation (LOQ) for AOH and AME of 1.0 µg/kg and TeA of 1.0 µg/kg with recovery from 80.0 to 114.8 %, repeatability from 0.07 - 9.9 %, meeting the requirements of AOAC and European regulations. The method was applied to detect and quantify Alternaria mycotoxins in 80 food samples collected in Hanoi city. AOH was detected in 04/20 vegetable samples (3.3 - 178 µg/kg), 03/20 fruit samples (5.6 - 7.3 µg/kg), 06/20 cereal samples (13.4 - 17.4 µg/kg), 03/20 oil seed samples (13.2 - 25.2 µg/kg); detected AME in 01/20 vegetable samples (7.5 µg/kg), 02/20 fruit samples (< 3 µg/kg), 01/20 cereal samples (9.5 µg/kg), 02/20 oil seed samples (< 3 µg/kg); detected TeA in 02/20 vegetable samples (9.0 µg/kg), 01/20 cereal samples (< 3 µg/kg), 02/20 oil seed samples (< 3 µg/kg), not detected in fruit samples.

Influence of Geometric Parameters of Conical Acrylic Portholes on Their Stress-Strain Behaviour.

Translucent elements in the form of truncated cones, which are made of organic glass, are widely used in the structures of portholes, submersible vessels, space vehicles, pressure chambers, teleboxes and other types of technical equipment. The decisive factor in designing portholes is to ensure the strength of their translucent elements. In order to reduce the weight of portholes and, accordingly, to increase the payload, it is necessary to optimise the geometric parameters of the translucent elements, which include the tapering angle and the ratio of thickness to radius of the smaller base. The paper deals with development of the applied (engineering) method for determining the stress–strain behaviour of the conical translucent elements of portholes made of organic glass under the action of a uniform hydrostatic pressure. Finite-element modelling of the translucent element of the conical porthole is performed, with the calculation of its stress–strain behaviour. External hydrostatic pressure of 10 MPa, absence of loads from the inside and continuous sliding of the translucent element with friction along the conical supporting surface of the porthole metal body are the boundary conditions for the computational model. Full-scale tests of translucent elements of portholes made of organic glass were performed under the action of uniform hydrostatic pressure. Analysis of the influence of geometric characteristics of the portholes on stress–strain behaviour showed that the increase in the tapering angle at the constant relative thickness of the translucent element reduced its axial displacement in all cases. Equivalent stresses acquire minimum values when the tapering angle is in the range from 75° to 105° (when the relative thickness increases, the optimal tapering angle becomes smaller). It is shown that the developed method for determination of the stress–strain behaviour of the conical translucent elements of portholes made of organic glass reflects the real picture of deformation and agrees with the results of full-scale tests. Results of the work allow us to choose the rational parameters of the translucent element for increasing the reliability of portholes through the creation of an effective distribution of stresses and strains in the translucent element, and improving its optical characteristics due to a relatively small deflection in operation.

Development of method for determination and preconcentration of uranium in water samples using XAD-4 resin loaded with Br-PADAP

In the present work, a minicolumn of XAD-4 loaded with 2-(5-bromo-2-pyridylazo)-5-(diethylamino)-phenol (Br-PADAP) is proposed as a preconcentration system for uranium determination in well, tap and mineral water samples by spectrophotometer using arsenazo III as the chromogenic reagent. Initially, a two-level (23) full factorial design was used for the preliminary evaluation of three factors, involving the following variables: sampling flow rate, elution flow rate, and pH. This design has revealed that, for the studied levels, buffer concentration and pH were significant factors. When the experimental conditions established in the optimization step were pH = 8.6, and an elution flow rate of 8.6 mL min−1 using 0.5% m/v ascorbic acid, this system has allowed for the determination of uranium with a detection limit (LOD) (3σ/S) of 0.05 μg L−1 and a quantification limit (LOQ) (10σ/S) of 0.16 μg L−1. The precision expressed as the relative standard deviation (R.S.D.) of 0.8% and 1.9% at 10.0 and 1.0 μg L−1, respectively- and a preconcentration factor of 184.5 for a sample volume of 50.0 mL. Accuracy was confirmed by uranium determination in the standard reference material, NIST SRM 1566b trace element units in Oyster Tissue samples, and spike tests with recuperations ranging from 93.2 to 105%; the procedure were applied for uranium determination in tap water, well water, and drinking water samples collected from Caetité and Cruz das Almas Cities, Bahia, Brazil. Determination of hydroquinone and benzoquinone in pharmaceutical formulations: critical considerations on quantitative analysis of easily oxidized compounds.

Hydroquinone is a skin-lightening agent used as an active ingredient in topical dermatological formulations prescribed for treating cutaneous diseases caused by hyperpigmentation. Despite being widely used, some toxicological aspects have been associated with these products, mainly due to overdosage and long-term use combined with the easy oxidation of hydroquinone. In this work, an investigative study has been done to gather enough data for selecting a quantitative analytical method for quality control purposes that considers the ease of oxidation not only within the product but also during the experimental procedures. After studying the influence of pH, reversibility, sampling, and standard solution preparation on the redox reaction between hydroquinone and benzoquinone by using spectroscopic, electrophoretic, and electroanalytical measurements, a reliable, fast, and selective chronoamperometric method was achieved. The optimized method was used for the analysis of samples, previously diluted in Britton-Robinson (BR) buffer (pH 5.5) and methanol (1 : 9, v/v), by applying a potential fixed at 0.4 V. A glassy-carbon working electrode, lab-made Ag/AgCl(sat) and platinum wire as a reference electrode and auxiliary electrodes, respectively, and BR buffer (pH 5.5) as supporting electrolyte were the additional experimental conditions used. Analytical performance parameters were verified to confirm the applicability of the new method (LOD 4.22 μmol L-1 and LOQ 14.1 μmol L-1; recovery mean value of 100% with 0.22% RSD). A gel topical formulation containing 4% (w/w) hydroquinone was analyzed through the developed method for determination of dosage and oxidation traces, and a content of 3.53 ± 0.095% (w/w) was found with no indications of degradation.

Novel poly(diaquabis(1,10-phenanthroline)copper(II)chloride) modified glassy carbon electrode for square wave voltammetric determination of aspirin in tablet samples

In this study, potentiodynamically fabricated poly (diaquabis(1,10-phenanthroline) copper(II)chloride) modified glassy carbon electrode (poly(A2P2CuC)/GCE) is reported for detection of aspirin in tablet samples. EIS and CV analysis of Fe(CN)6]3-/4- at the modified electrode revealed surface modification by an electroactive polymer material that improved electrode effective surface area and charge transfer resistance. An oxidative peak without a reductive peak in the reverse scan direction for aspirin at poly(A2P2CuC/GCE augmented by peak potential shift with scan rate showed the irreversibility of the electrochemical oxidation of ASA at the electrode. A better R2 (0.9983) for dependence of peak current on square root of scan rate than R2 (0.9612) on scan rate indicated that oxidation of aspirin at poly(A2P2CuC)/GCE was predominantly diffusion mass transport controlled. Under optimized PBS pH (5.5), and square wave parameters, oxidative peak current response of poly(A2P2CuC)/GCE showed linear dependence on the concentration of aspirin in the range 1–200 ​μM with associated %RSD under 3.7% (n ​= ​3), limit of detection 0.039 ​μM, and limit of quantification and 0.13 ​μM. Spike recovery result in the range 96.5–100.5% with %RSD under 1.9% (n ​= ​3), and interference recovery in the range 97.2–103.4% in the presence of 50–200% of uric acid, ascorbic acid, and glucose as potential interferents validated the applicability of the developed method for determination of aspirin content in tablet samples. Detection of aspirin in the studied tablet samples in an amount 99.4–101.5% of their nominal values with %RSD under 2.6% using the present method showed the applicability of the method to control the aspirin content in tablet samples. The present method showed better performance over the previously reported methods for determination of aspirin in tablet samples making it an excellent candidate.

Enantioselective analyses of chloroquine and hydroxychloroquine in rat liver microsomes through chiral liquid chromatography-tandem mass spectrometry.

An efficient, sensitive and selective liquid chromatography-tandem mass spectrometry (LC-MS/MS) chiral analysis method was established for determination of chloroquine and hydroxychloroquine enantiomers in rat liver microsomes. Effects of polysaccharide chiral stationary phases and basic additives on chiral separations of two analytes were discussed in detail. Amylose tris(3, 5-dimethylphenylcarbamate)-coated chiral stationary phase showed the best separation performance for them with acetonitrile-diethylamine-ethanol-diethylamine mixture (90:0.1:10:0.1, v/v/v/v) among four chiral stationary phases. Then, multiple reaction monitoring mode was selected as the data acquisition for determination of two pairs of enantiomers. The proposed LC-MS/MS chiral analysis method was validated in terms of linearity, accuracy, precision, and specificity. Good linearity with correlation coefficient over 0.998 was obtained in the concentration range of 0.05-5 μM. Limits of quantification for chloroquine and hydroxychloroquine enantiomers were 5.0 and 1.0 nM, respectively. The recoveries ranged from 81.14% to 111.09%. The intra-day and inter-day relative standard deviation were less than 6.5%. Moreover, concentrations of chloroquine and hydroxychloroquine enantiomers in rat liver microsomes were determined through the proposed LC-MS/MS analysis method. After incubated with rat liver microsomes for 10 min, the enantiomeric factor of hydroxychloroquine decreased from 0.50 to 0.45 (p < 0.001). In brief, our developed determination method for chloroquine and hydroxychloroquine enantiomers through LC-MS/MS spectrometry showed the characteristics of high-efficiency, fast speed, and very low detection limit, and would be greatly beneficial for screening and quantitation of them in biological matrices.

Simultaneous Detection of Multiple Plant Growth Regulator Residues in Cabbage and Grape Using an Optimal QuEChERS Sample Preparation and UHPLC-MS/MS Method.

At present, plant growth regulators (PGRs) are widely used in agricultural and forestry production. PGRs, like traditional pesticides, have certain toxicities. Naively excessively applying them will cause the acute and chronic poisoning of humans and animals and potentially harm human health. In order to assess, prevent, and control the residues of PGRs in fruits and vegetables, a set of quick, easy, cheap, effective, rugged, and safe (QuEChERS) analytical methods that simultaneously detect multiple PGR residues are urgently needed for quality and safety inspection of agricultural product. In this study, grapes (representative of fruits) and cabbages (representative of vegetables) were used as the detected objects. The 30 commercial product residues of PGRs were detected in both with an ultra-high performance liquid chromatography (UHPLC)-tandem mass spectrometry (MS/MS) method, based on optimized chromatographic, MS, and preparation conditions (extraction solvent and cleanup conditions). Grape and cabbage samples were extracted with acetonitrile containing 5% (v/v) acetic acid, dehydrated using a salt package, purified using the QuEChERS method, ionized using electrospray ionization under positive and negative ion switching mode, detected using multi-reaction monitoring, and quantification using an external standard method of matrix matching standard curve. Methanol was selected as the strong elution phase. A methanol-0.1% formic acid-5 mmol/L ammonium acetate solution was selected as the best mobile phase. The optimal extraction solvent was acetonitrile containing 5% acetic acid. Primary secondary amine cleanup could met the determination requirements of PGR residues. The developed method for determination of 30 commercial products of PGR, such as betaine, showed excellent linearity in 1-500, 10-1000, ∼500, ∼2000, and 100-10 000 μg/kg (R ≥ 0.98). At the 0.001 (0.01), 0.05, 0.20, and 1.00 mg/kg additive concentrations, the average addition standard recovery of 30 commercial products of PGR were 61-132% with the relative standard deviations of 1-14% and the LOQs were confirmed to be 1.0-100 μg/kg through the actual addition values of samples. The set of optimized QuEChERS UHPLC-MS/MS methods simultaneously detect residues of PGRs in fruits and vegetables with one-time sample preparation for high-throughput, rapid quantitative screening, and confirmation. The methods cover a wide range of PGRs with simple and convenient preparation and small amounts of solvent, and can provide technical support for the supervision of PGR residues in fruits and vegetables. The optimizations of extraction solvent screening, different ratios of various purification packages in the QuEChERS method, and UPLC-MS conditions were conducted and the precision, sensitivity, and recovery rates of the methods were investigated in order to establish a QuEChERS UPLC-MS/MS method for simultaneously detecting 30 kinds of PGR residues in fruits and vegetables. The methods allow high-throughput determination of multiple PGR residues in fruits and vegetables and can also provide technical references for related compound residue detection of other matrixes.
