Simultaneous determination of Propranolol hydrochloride and Hydrochlorothiazide in Tablets formulation using spectrophotometric technique (Simultaneous Equation Method)

A new spectrophotometric method was developed for simultaneous determination of compounds with interfering spectra in binary mixtures without previous separation, showing significant advantages over the conventional methods regarding minimal data manipulation and applicability. The proposed method was applied for the determination of Propranolol hydrochloride and Hydrochlorothiazide in Tablets formulation, for determination of sampling wavelength, 10 ?g/ml of each of PRO and HCT were scanned in 200-400 nm range and sampling wavelengths were 289 nm for PRO and 270 nm for HCT are selected for development and validation of simultaneous equation method. For this method linearity observed in the range of 10-50 ?g/ml for PRP and 5-25 ?g/ml for HCT, and in their pharmaceutical formulation with mean percentage recoveries 100.13 0.86 and 100.07 0.58, respectively. The method was validated according to ICH guidelines and can be applied for routine quality control testing.

Development and validation of RP-HPLC, HPTLC and UV-visible spectrophotometric methods for simultaneous estimation of alprazolam and propranolol hydrochloride in their combined dosage form

Three accurate, sensitive and reproducible methods are described for the quantitative determination of alprazolam (ALP) and propranolol hydrochloride (PNL) in their combined dosage form. The first method involves an RP-HPLC separation on the C18 column using acetonitrile-25 mM ammonium acetate buffer and 0.2% triethylamine (pH of buffer adjusted to 4 with glacial acetic acid) in the ratio of 35: 65 (v/v) as mobile phase. Symmetrical peaks with good separation, ALP at 9.3 min and PNL at 3.5 min, were achieved. Quantification was done with photo diode array detection at 255 nm over the concentration ranges of 0.5–50 and 10–250 μg/mL for ALP and PNL, respectively. The second method is based on the separation of drugs by HPTLC using chloroform-methanol-ammonia 7: 0.8: 0.1 (v/v/v) as mobile phase. Quantification was achieved using UV detection at 248 nm over the concentration range of 100–600 ng/spot and 5–30 μg/spot for ALP and PNL, respectively. The third method involves dual wavelength UV-visible spectrophotometric method. It is based on the determination of PNL at 319.4 nm using its absorptivity value and ALP at 258.2 nm after deduction of absorbance due to PNL. Quantification was achieved over the concentration range of 1–40 and 80–200 μg/mL for ALP and PNL, respectively. All methods were validated according to ICH guidelines and successively applied to marketed pharmaceutical formulation, and the results of all three methods were compared statistically as well. No interference from the tablet excipients was found.

Quantitative Spectrophotometric Determination of Propranolol Hydrochloride in Pharmaceuticals Using Cerium(IV) Sulphate as Oxidimetric Reagent

Two simple, selective and economic spectrophotometric methods have been developed and validated for the analysis of the propranolol hydrochloride (PPH) in bulk drug and pharmaceuticals. The methods are based on the oxidation of PPH with cerium(IV) in acid medium and subsequent measurement of the excess cerium(IV) either by its reaction with p-dimethylamino benzaldehyde (p-DMAB) and measuring the resulting color at 460 nm (method A) or by its reaction with sulphanilic acid (SAA) to give a pink colored product peaking at 540 nm (method B). The decrease in the absorption intensity of the colored products, due to the presence of the drug has been correlated with its concentration in the sample solution. Different experimental variables affecting the reaction are carefully studied and optimized. Under the optimum conditions, linear relationships with good correlation coefficients are found in the concentration ranges of 0.4–7.0 μg ml−1 for method A and 10–200 μg ml−1 for method B, respectively. The calculated molar absorptivity values are 3.72 × 104 for method A and 1.43 × 103 for method B. The methods have been validated for accuracy, precision, ruggedness and robustness and the results are found to be satisfactory. The proposed methods are successfully applied to the determination of PPH in pure and pharmaceutical dosage forms without any interference from the common excipients. The results obtained by the proposed methods are comparable with those obtained by the official methods.

Development and validation of HPTLC method for simultaneous estimation of propranolol hydrochloride and flunarizine dihydrochloride in combined tablet dosage form

A simple, precise, and accurate high-performance thin-layer chromatography (HPTLC) method for the simultaneous determination of propranolol hydrochloride (PRP) and flunarizine dihydrochloride (FLU) in their combined tablet dosage form has been developed and validated. Propranolol hydrochloride and flunarizine dihydrochloride from the formulation were separated on silica gel 60 GF254 HPTLC plates with solutions in the proportion of methanol-chloroform-toluene-GAA (3:7.5:7.5:0.3 v/v) as mobile phase. Densitometric quantification was performed at 260 nm. Well-resolved bands were obtained with RF values 0.35 and 0.64 for propranolol hydrochloride and flunarizine dihydrochloride, respectively. The method was validated for precision, accuracy, and specificity. The calibration curve was found to be linear in the concentration range of 3000–15,000 ng band−1 and 750–3700 ng band−1 of PRP and FLU respectively. Enzymatic kinetic method for determination of propranolol hydrochloride in pharmaceuticals based on its inhibitory effect on cholinesterase

Propranolol, a widely used beta-blocker, inhibits the reaction of enzyme cholinesterase hydrolysis. Measurements of the hydrolysis rate differences between and inhibited reactions, allows the development of a kinetic method for its determination. Both systems, enzyme-substrate-chromogen and enzyme-substrate-chromogen-inhibitor, were characterized through biochemical kinetic parameters (KM = 0.326-0.330 mmol/L; Vmax = 40-42.99 ?mol/Lmin), inhibition type was recognized as competitive, and inhibition constant, Ki, was determined to be 22.60 ?mol/L. The detection and quantification limits were calculated as 0.004 and 0.0136 ?mol/L, respectively. Accuracy and precision of proposed methods were tested. Proposed method is characterized with good sensitivity, selectivity, simplicity and rapidity, thus it is convenient for clinical applications.

Development and validation of a dissolution method for novel fixed dose combination of etodolac and propranolol hydrochloride tablets by RP-HPLC

Summary The present study reports the dissolution method for a novel fixed dose combination (FDC) containing etodolac (ET) and propranolol hydrochloride (PH) developed utilizing USP Apparatus 1 (basket) at 100 rpm with 1000 mL of phosphate buffer (pH 6.8; 0.05 M) medium at 37°C. An isocratic reversed-phase liquid chromatographic (RPLC) method was also developed for the simultaneous determination of ET and PH on an octadecylsilica column using phosphate buffer (pH 5.5) and acetonitrile (60:40, υ/υ) as the mobile phase with ultraviolet (UV) detection at 292 nm. Validation data were obtained, which demonstrated that the dissolution methodology is accurate, precise, linear, and rugged for the combination tablets.

Liquid Chromatographic Determination of Propranolol Hydrochloride in Various Dosage Forms

A simple and rapid liquid chromatographic method is described for the determination of propranolol hydrochloride in pharmaceutical preparations. The separation was achieved on a reverse-phase octylsilane (C8) column by using a mobile phase composed of a mixture of 0.5 g dodecyl sodium sulfate in 18 mL (0.15 M) H3PO4 plus 90 mL methanol, 90 mL acetonitrile, and 52 mL water. Detector response was linear for 0.03-3.1 mg/mL of propranolol. Recoveries from synthetic mixtures ranged from 99.6 to 101.7%. The results obtained by the proposed method were similar to those obtained by the USP XXI method.
