Quantitative Estimation of Aloin in Aloe Vera and its Commercial Formulation by using UV Spectroscopy

In Indian Ayurvedic system of medicine the leaf of Ghritkumari has been extensively used as a cathartic agent to treat severe stomach constipation. The aloe vera plant is used as food supplement and also used in cosmetic products to treat different skin problems. In order to develop reliable quality control marker to check its potential toxicity and carcinogenic activity and to check the commercial samples of Aloe vera for authenticity and purity, UV Spectroscopy analysis has been done. It has been found that aloin is present in a few commercial samples. HPLC, Infra-Red and thin layer chromatography methods have been reported for aloin determination in commercial samples. Therefore, UV Spectrophotometric method was developed for determination of aloin in commercial formulations.

Determination of Aloin A and Aloin B in Aloe vera Raw Materials and Finished Products by High-Performance Liquid Chromatography: Single-Laboratory Validation.

A single-laboratory validation (SLV) was conducted on an HPLC method for the detection and quantification of aloin A and aloin B in Aloe vera raw materials and finished products. An extraction procedure using sonication with an acidified solvent was used for solid test materials while liquid test materials only required dilution, if necessary, prior to filtration and analysis. Separation was achieved using a fused core C18 column in 18 min under isocratic elution conditions allowing for a single analyte (aloin A) calibration curve to quantify both aloins. Adequate chromatographic resolution (Rs ≥ 1) was achieved for aloin A and aloin B. The calibration curves for aloin A exhibited coefficients of determination (r(2)) of ≥ 99.9% over the linear range of 0.3-50 μg/mL. The LOD values were 0.092 and 0.087 μg/mL, and LOQ 0.23 and 0.21 μg/mL for aloin A and aloin B, respectively. Repeatability studies were performed on nine test materials on each of 3 separate days, with five of the test materials determined to be above the LOQ having repeatability RSD (RSDr) values ranging from 0.61 to 6.30%. Method accuracy was determined through a spike recovery study on both liquid and solid matrixes at three different levels: low, medium, and high. For both aloins, the recovery in the liquid matrix ranged from 92.7 to 106.3% with an RSDr of 0.15 to 4.30%, while for the solid matrix, the recovery ranged from 84.4 to 108.9% with an RSDr of 0.23 to 3.84%. Based on the results of the SLV study, it is recommended that this method be evaluated for reproducibility through a collaborative study.

Determination of aloin of compound aloe ointment by HPLC

Objective To establish a method for the determination of aloin of compound aloe ointment. Methods HPLC method was used with Agilent Zorbax-C18（250 mm×4.6 mm,5μm）column and the mobile phase consisted of acetonitrile-0.08%Sodium 1-heptanesulfonate （22∶78）. The flow rate was 1.0 ml/min,the column temperature was 30 ℃, and the UV detector was set at 355 nm. Results The linear ranges of aloin was 0.037~0.74 mg/ml（r=0.999 8）. The average recovery of aloin was 98.7%. Conclusion The method is simple, repeatable and accurate, it can be applied in quantitative determination of aloin in compound aloe ointment. Comparison of High-Performance Liquid Chromatographic and Thin-Layer Chromatographic Methods for Determination of Aloin in Herbal Products Containing Aloe vera

Aloe vera is a medicinal plant used worldwide to treat a variety of conditions and, as such, has important commercial value. Aloin is a principal component of aloe vera leaves and is used for quality control of products containing it. A semiquantitative thin-layer chromatographic (TLC) method for determining the concentration of aloin in aloe-based products was validated. The results were similar to those of a validated high-performance liquid chromatographic method; therefore, TLC, which is a simple, sensitive, specific, rapid, and cheap method, may be ideal for use in any laboratory for routine analysis of commercial products containing aloe vera.

High-performance liquid chromatographic profiles of aloe constituents and determination of aloin in beverages, with reference to the EEC regulation for flavouring substances

Characteristic HPLC profiles of fresh and aged aloe solutions, detected at 360 and 220 nm, are presented and compared. Several aloe constituents (aloesin, aloeresin A, hydroxyaloin, aloin A and B and aloinoside A and B) were simultaneously separated and identified. The determination of aloin is described (detection limit 0.15 ppm) and discussed. In aloe-based alcoholic beverages, the aloins could not be detected, owing to their instability and degradation in solution; this is discussed in relation to the EEC Council Directive 88/388, which fixed the values of maximum allowable concentrations for aloin in food and beverages. Instead of aloin, other aloe compounds (e.g., aloeresin A or aloesin) should perhaps be used as an index of the presence of aloe in alcoholic beverages.
