AbstractThe cover picture shows the design and synthesis of two successive generations of analogues of embelin, a natural product with proapoptotic and anticancer properties. This research was performed in the Indo‐French “Joint Laboratory for Sustainable Chemistry at Interfaces&2dquo; between the University of Rennes I/CNRS and the Indian Institute of Chemical Technology/CSIR, Hyderabad. This cooperation is highlighted by the best‐known monuments from both cities (Charminar in Hyderabad and Parliament of Brittany in Rennes). Details of this research are discussed in the article by R. Synthesis of a Focused Chemical Library Based on Derivatives of Embelin, a Natural Product with Proapoptotic and Anticancer Properties

AbstractThe synthesis of new derivatives of embelin, a natural inhibitor of X‐linked inhibitor of apoptosis protein (XIAP) is described. The design of these new molecules involved introduction of aromatic groups directly linked to the benzoquinone core. To allow a large flexibility in the nature and the length of the added chain, the strategy involves first aSuzuki–Miyaura reaction with functionalized aromatics, yielding a first generation of molecules. Then, by appropriate use of the functional groups, a second generation of representative embelin derivatives was prepared.

Synthesis and Evaluation of Analgesic and Anti-inflammatory Activities of Most Active Free Radical Scavenging Derivatives of Embelin-A Structure-Activity Relationship

Antioxidant and related properties of the plant Embelia ribes and embelin are well known. In the present study embelin was condensed with various aromatic substituted primary amines to yield ten new and one reported derivatives along with monomethyl embelin. All these compounds along with embelin were evaluated for in vitro antioxidant activity using 2,2'-azino-bis(3-ethylbenzo-thiazoline-6-sulfonic acid) diammonium salt (ABTS) and 2,2'-diphenyl-1-picryl hydrazyl (DPPH) methods. Two para-substituted embelin derivatives showed potent antioxidant activity. These compounds along with embelin were studied for analgesic and anti-inflammatory activities at 10 and 20 mg/kg doses by standard methods. Potent analgesic activity higher than the standard pentazocine was observed. Embelin and both of its derivatives almost completely abolished the acetic acid induced writhing. p-Sulfonylamine phenylamino derivative showed better anti-inflammatory activity than embelin.

An efficient synthesis of embelin derivatives through domino Knoevenagel hetero Diels–Alder reactions under microwave irradiation

The synthesis of novel pyrano embelin derivatives has been achieved through domino Knoevenagel hetero Diels–Alder reactions of embelin ( 1) with paraformaldehyde and electron rich alkenes. This synthetic approach is highly efficient when microwave irradiation is used.

Design, synthesis, and characterization of new embelin derivatives as potent inhibitors of X-linked inhibitor of apoptosis protein

X-Linked inhibitor of apoptosis protein (XIAP) is a promising molecular target for the design of new anticancer drugs aiming at promoting apoptosis in cancer cells. We have previously identified embelin as an inhibitor of XIAP through computational structure-based database screening. Herein, we report the design, synthesis, and evaluation of new embelin analogues as inhibitors of XIAP. Our efforts led to the identification of new and more potent inhibitors. For example, compound 6g has a K i value of 180 nM binding to XIAP BIR3, in a competitive binding assay and represents a promising lead compound for further optimization.

Embelin Derivatives and Their Anticancer Activity through Microtubule Disassembly

Biological activities of the 1,4-benzoquinone derivatives 5- O-ethylembelin ( 1) and 5- O-methylembelin ( 2) were investigated. Both of them showed antiproliferative activity against a panel of human tumor cell lines upon comparison to normal marsupial kidney cells (PtK2). They arrested HL-60 cells in the G(0)/G(1) phase of the cell cycle in a dose- and time-dependent manner. In HeLa cells, exposure to 100 microM of 1 or 2 for 6 h induced a complete disassembly of the microtubule network and an increased number of cells blocked in mitotic stages. Treatment with 10 microM of 1 and 2 for 24 h induced apoptosis in HL-60 cells. This evidence suggests that both 1 and 2 are promising novel antimitotic and anticancer molecules targeting microtubular proteins.
