Moderate protective effect of Kyotorphin against the late consequences of intracerebroventricular streptozotocin model of Alzheimer's disease.

The established decrease in the level of endogenous kyotorphin (KTP) into the cerebrospinal fluid of patients with an advanced stage of Alzheimer's disease (AD) and the found neuroprotective activity of KTP suggested its participation in the pathophysiology of the disease. We aimed to study the effects of subchronic intracerebroventricular (ICV) treatment (14days) with KTP on the behavioral, biochemical and histological changes in rats with streptozotocin (STZ-ICV)-induced model of sporadic AD (sAD). Three months after the administration of STZ-ICV, rats developed increased locomotor activity, decreased level of anxiety, impaired spatial and working memory. Histological data from the STZ-ICV group demonstrated decreased number of neurons in the CA1 and CA3 subfields of the hippocampus. The STZ-ICV group was characterized with a decrease of total protein content in the hippocampus and the prefrontal cortex as well as increased levels of the carbonylated proteins in the hippocampus. KTP treatment of STZ-ICV rats normalized anxiety level and regained object recognition memory. KTP abolished the protein loss in prefrontal cortex and decrease the neuronal loss in the CA3 subfield of the hippocampus. STZ-ICV rats, treated with KTP, did not show significant changes in the levels of the carbonylated proteins in specific brain structures or in motor activity and spatial memory compared to the saline-treated STZ-ICV group. Our data show a moderate and selective protective effect of a subchronic ICV administration of the dipeptide KTP on the pathological changes induced by an experimental model of sAD in rats.

Peach Fruit Development: A Comparative Proteomic Study Between Endocarp and Mesocarp at Very Early Stages Underpins the Main Differential Biochemical Processes Between These Tissues.

Peach (Prunus persica) is an important economically temperate fruit. The development follows double sigmoid curve with four phases (S1–S4). We centered our work in the early development. In addition to S1, we studied the very early stage (E) characterized by the lag zone of the exponential growing phase S1, and the second stage (S2) when the pit starts hardening. “Dixiland” peach fruit were collected at 9 (E), 29 (S1), and 53 (S2) days after flowering (DAF) and endocarp and mesocarp were separated. There was a pronounced decrease in total protein content along development in both tissues. Quantitative proteomic allowed the identification of changes in protein profiles across development and revealed the main biochemical pathways sustaining tissue differentiation. Protein metabolism was the category most represented among differentially proteins in all tissues and stages. The decrease in protein synthesis machinery observed during development would be responsible of the protein fall, rather than a proteolytic process; and reduced protein synthesis during early development would reroute cell resources to lignin biosynthesis. These changes were accompanied by net decrease in total amino acids in E1–S1 and increase in S1–S2 transitions. Amino acid profiling, showed Asn parallels this trend. Concerted changes in Asn and in enzymes involved in its metabolism reveal that increased synthesis and decreased catabolism of Asn may conduct to an Asn increase during very early development and that the β-Cyano-Alanine synthase/β-Cyano-Alanine hydratase could be the pathway for Asn synthesis in “Dixiland” peach fruit. Additionally, photosynthetic machinery decays during early development in mesocarp and endocarp. Proteins related to photosynthesis are found to a higher extent in mesocarp than in endocarp. We conclude mesocarpic photosynthesis is possible to occur early on the development, first providing both carbon and reductive power and latter only reductive power. Together with proteomic, histological tests and anatomical analysis help to provide information about changes and differences in cells and cell-walls in both tissues. Collectively, this work represents the first approach in building protein databases during peach fruit development focusing on endocarp and mesocarp tissues and provides novel insights into the biology of peach fruit development preceding pit hardening.

Maternal Body Mass Index and Breastmilk Energy, Fat, and Protein Content: A Systematic Review and Regression Analysis of Simulated Data (OR30-06-19)

ObjectivesGiven increased incidence of maternal postpartum obesity and limited understanding of the degree to which maternal body composition is related to energy and macronutrient content of breastmilk, we conducted a systematic review to assess the associations between maternal body mass index (BMI) and breastmilk energy, fat, and/or total protein content. MethodsA protocol was registered prior to the completion of this systematic review (PROSPERO 2018 CRD42018098808). The electronic databases MEDLINE, Embase, CINAHL, and Web of Science were searched. The three outcomes of interest were breastmilk energy (kcal/dL), fat (g/dL), and total protein (g/dL) from mothers of term-born infants. Articles that reported maternal BMI or weight and height, collected breastmilk, and measured energy, fat, or protein concentration in breastmilk one to six months postpartum were eligible. If breastmilk was collected at multiple timepoints, the measures closest to three months postpartum were used. A simulated dataset was generated in Stata 14 based on respective sample sizes and means and variance of maternal BMI and breastmilk energy, fat, and protein content of the included studies, assuming normality of each of these variables based on the literature. Linear regressions were performed to examine associations between maternal BMI (kg/m2) and breastmilk energy and individual macronutrients. ResultsA total of 2,012 articles were identified from the search and reference lists of included studies. After full-text screening by two reviewers, 39 articles were included with breastmilk samples from 2,837 mothers. Based on the simulated data, maternal BMI was not significantly associated with breastmilk energy concentration (β = −0.0044, 95% CI [−0.14, 0.14], P > 0.9). However, BMI was positively associated with breastmilk fat concentration (β = 0.015, 95% CI [0.0022, 0.027] P = 0.02) and was negatively associated with total protein concentration (β = −0.0068, 95% CI [−0.011, −0.0027], P = 0.001). Thus, every 1 kg/m2 increase in BMI is associated with a 0.0022 g/dL increase in fat and a 0.011 g/dL decrease in total protein concentration. ConclusionsStatistically significant associations were found between maternal BMI and breastmilk fat and protein content. Protective Roles of Kolaviron Extract from Garcinia kola Seeds against Isoniazid-induced Kidney Damage in Wistar Rats

Background: Garcinia kola seeds have been observed to be medically important and kolaviron, a bioflavonoid obtained from the seeds was studied for its biological activities. The study investigated the protective effect of kolaviron extract obtained from the seed of Garcinia kola against isoniazid-induced kidney damage.
 Methodology: Kolaviron was extracted from fresh seeds of Garcinia kola (2 kg) using soxhlet extractor and partitioned with chloroform. Nephrotoxicity was induced in wistar rats by oral administration of isoniazid (20 mg/kg bwt) while kolaviron was administered on wistar rats an hour before isoniazid administration and lasted for 30 days. Protective effect of kolaviron was measured in the plasma of wistar rats by estimating the levels of key metabolites used as kidney biomarkers which are total protein, creatinine, urea and uric acid concentration.
 Results: The isoniazid-treated group showed a significant (p < 0.05) decrease in total protein concentration of 3.57 ± 0.12 (mg/dl) while there was a significant (p < 0.05) increase in urea, uric acid and creatinine concentrations with values of 70.30 ± 4.77, 55.71 ± 11.15 and 18.04 ± 5.33 (mg/dl) respectively. However, kolaviron-treated group showed a remarkable increase (6.15 ± 0.96) in total protein concentration while urea, uric acid and creatinine concentrations significantly decreased to 45.25 ± 2.29, 35.60 ± 11.01 and 13.28 ± 4.41 (mg/dl) respectively.
 Conclusion: Kolaviron extract obtained from Garcinia kola seeds exhibited a remarkable protective effect against kidney damage caused by isoniazid by regulating renal biomarkers and preventing toxic affront of isoniazid. Thus, it may be relatively safe when used therapeutically at this dose in the treatment and management of diseases associated with kidney damage.

Biochemical effect of low metabolic calories diets on experimentally induced obesity on rats

This study aimed to evaluate the effect of weight loss diets in experimental obesity. Forty –five male albino rats, 6-8 weeks old, were randomly assigned into four groups: Group 1 (Control normal group) : Comprised 10 rats, provided only with regular diet and plenty of fresh. Group 2 ( Control obese group with 75 calories diet) : Included 10 rats ,was received a high fat diet for 4 weeks period then each rat was fed daily with 75 calories diet for 8weeks .Group 3 (Obese group with 60 calories diet): Included 10 rats ,was received a high fat diet for 4 weeks period then each rat was fed daily with 80% calories of the Control obese diet for 8weeks (60 calories diet) .Group 4 (Obese group with 45 calories diet) : Included 10 rats ,was received a high fat diet for 4 weeks period then each rat was fed daily with 60% calories of the Control obese diet for 8weeks (45 calories diet).Blood samples were collected after 8 and 12weeks of supplementation for biochemical analysis. As compared to the control normal group, control obese group exhibited a significant decrease in serum high density lipoprotein-cholesterol (HDL-c), transferrin and total iron bending capacity(TIBC), along with increase in aspartate aminotransferase (AST) activity, total protein, albumin, urea, creatinine, total cholesterol ,plasma triacylglycerol, very low density lipoprotein-cholesterol (VLDL-c), low density lipoprotein-cholesterol (LDL-c),sodium ,calcium , iron concentration and Plasma ferritin concentration. Meanwhile , the obese group provided with 60 calories diet exhibited a significant decrease in AST activity, total protein, albumin , urea, creatinine, total cholesterol , triacylglycerol ,VLDL-c, LDL-c, sodium ,calcium , iron concentration and ferritin along with increase in HDL-c, transferrin and TIBC when compared with obese group. However, the obese group provided with 45 calories diet, exhibited a significant decrease in total protein concentration, albumin, T. cholesterol, triacylglycerol, VLDL-c, LDL-c, sodium, calcium, iron concentration and ferritin along with increase in AST activity, urea, creatinine, HDL, transferrin and TIBC, when compared with obese group. In conclusion, Low calorie diet reduces calorie intake without incurring malnutrition or a reduction in essential nutrients compared with very low-calorie diet.

Effect of co-administration of Bee honey and some chemotherapeutic drugs on dissemination of hepatocellular carcinoma in rats.

Alternative and complimentary usage of the natural compound has raised hopes of finding curative options for liver hepatocarcinogenesis. In the present study, the curative effect of bee honey against diethylnitrosamine (DEN) (50 mg/kg) and carbon tetrachloride (CCl4) (2 mg/Kg)-induced hepatocellular carcinoma (HCC) in male rats in the presence or absence of some chemotherapeutic drugs, Cisplatin (Cis), Cyclophosphamide (CY) and 5- Fluorouracil (5-FU) were investigated. The obtained results demonstrated that treatment with DEN/CCl4 caused oxidative stress as assigned by the increase in malondialdehyde (MDA) and fall in glutathione (GSH) content. Meantime detraction in the antioxidants, including superoxide dismutase (SOD), catalase (CAT), glutathione-s-transferase (GST) and glutathione peroxidase (GPx) was observed. Also, the results showed induction of inflammation as reflected by an increase in the levels of both α- fetoprotein and α- fucosidase in the liver. This was accompanied by changes in the hepatic function biomarkers which characterized by the increased levels of transaminases (AST, ALT), alkaline phosphatase (ALP) and γ-Glutamyl transferase (γ-GT) and decrease in total protein content in the serum. In conclusion, the combination of the selected drugs and bee honey may be an effective chemo- preventive and therapeutic strategy for treating DEN and CCl4-induced HCC.

Chitinases and thaumatin-like proteins in Sauvignon Blanc and Chardonnay musts during alcoholic fermentation

Protein precipitation, also referred to as protein instability, may lead to haziness in bottled wines and result in significant commercial losses. To avoid problems of this nature, fining finished wines with clay (bentonite) is the most commonly applied methodology. However, bentonite fining reduces yield and may affect wine quality. Protein haze has been primarily linked to grape pathogenesis-related proteins, in particular chitinases and thaumatin-like proteins. To better understand the persistence of these proteins during fermentation, reverse phase chromatography was used to monitor the evolution of total grape proteins as well as of chitinases and thaumatin-like proteins during alcoholic fermentation. The data confirm a previously reported significant decrease in total protein content during fermentation. This reduction in total protein levels was observed throughout fermentation, and was affected by factors such as fermentation temperature, yeast strain or grape cultivar. However, significant changes in the concentration of free chitinases were observed in a yeast strain-dependent manner. The data thus confirm the correlation between the levels of yeast cell wall chitin and changes in chitinase concentration, and suggest that it is primarily the amount of lateral chitin, and not the chitin in bud scars, that is responsible for this activity.

Biochemical defence responses of resistant and susceptible rice genotypes against blast pathogen Magnaporthe oryzae

Rice blast is the leading fungal disease which is caused by Magnaporthe oryzae that contributes for the significant decline in the rice yield throughout the globe. There is a need for the understanding of biochemical changes in rice plant during blast infection for the development of novel disease control strategies. In the present study, we isolated M. oryzae from the local paddy fields and the fungal isolates (VCF and PON) were identified by ITS-PCR using genomic DNA samples. Further, we inoculated resistant (BR2655 and TUNGA) and susceptible (INTAN and HR12) rice cultivars with PON and VCF isolates. PON isolate showed relatively high virulence compared to VCF and standard MTCC fungal strains. Therefore, we evaluated the effect of PON on the total protein content and plant defence-related key enzymes (peroxidase, polyphenol oxidase, phenylalanine ammonia lyase, β-glucosidase, chitinase and lipoxygenase) activities between 24- and 120-hour post-inoculation (hpi). The results demonstrated the decrease in total protein content in all the inoculated cultivars. In addition, we observed the variation in the activity of peroxidase, polyphenol oxidase, β-glucosidase, chitinase and lipoxygenase at different time points in all the tested rice plants compared to respective controls. However, no significant difference was observed in the phenylalanine ammonia lyase activity relative to its control. Taken together, this study emphasizes on the variation in the activities of plant defence enzymes in different plant cultivars against the tested fungal pathogen and also implementation of defence enzymes as biochemical markers for resistant breeding.

Multiple biomarkers based appraisal of deltamethrin induced toxicity in silver carp (Hypophthalmichthys molitrix)

Deltamethrin (DLM) is α-cyano (type II) synthetic pyrethroid. DLM exposure leads to strong neurotoxic effects and a number of complex toxicological syndromes. The current study assessed DLM mediated oxidative stress, behavioral, hematological, histopathological, and biochemical toxic effects on silver carp (Hypophthalmichthys molitrix). Exposure to an acute concentration (2 μg/L) of DLM resulted in different behavioral inconsistencies and a time-dependent significant (P < 0.05) change in the hematology and serum biochemistry of silver carp. A significant (P < 0.05) increase in the activities of reactive oxygen species, lipid peroxidation, and antioxidant enzymes whereas a significant decrease in total protein contents in the liver, gills, brain, and muscle tissues were observed. DLM exposure increased the activities of metabolic enzymes in the gills, muscles, and liver of silver carp. A significant (P < 0.05) increase in DNA damage in peripheral blood erythrocytes was evident. DLM exposure led to a time-dependent significant (P < 0.05) increase in the whole-body cortisol and blood glucose level, while a significant decrease in acetylcholine esterase activity in the brain, liver, and muscle tissues. Different histopathological changes in the liver, gills, brain, and intestine were observed, however, no significant change in the gross anatomy and morphometric parameters of the fish was observed. The current study provides valuable information for devising better strategies regarding environmental management, chemicals' risk assessment, biodiversity conservation, and monitoring of the aquatic organisms. DLM was concluded to be highly toxic to fish. The extensive use of DLM should be prohibited or allowed under strict environmental laws; otherwise, it might lead to the extermination of the susceptible wildlife, such as commercially very valuable but nearly threatened silver carp.

Metabolomics and proteomics reveal drought-stress responses of leaf tissues from spring-wheat

To reveal the integrative biochemical networks of wheat leaves in response to water deficient conditions, proteomics and metabolomics were applied to two spring-wheat cultivars (Bahar, drought-susceptible; Kavir, drought-tolerant). Drought stress induced detrimental effects on Bahar leaf proteome, resulting in a severe decrease of total protein content, with impairments mainly in photosynthetic proteins and in enzymes involved in sugar and nitrogen metabolism, as well as in the capacity of detoxifying harmful molecules. On the contrary, only minor perturbations were observed at the protein level in Kavir stressed leaves. Metabolome analysis indicated amino acids, organic acids, and sugars as the main metabolites changed in abundance upon water deficiency. In particular, Bahar cv showed increased levels in proline, methionine, arginine, lysine, aromatic and branched chain amino acids. Tryptophan accumulation via shikimate pathway seems to sustain auxin production (indoleacrylic acid), whereas glutamate reduction is reasonably linked to polyamine (spermine) synthesis. Kavir metabolome was affected by drought stress to a less extent with only two pathways significantly changed, one of them being purine metabolism. These results comprehensively provide a framework for better understanding the mechanisms that govern plant cell response to drought stress, with insights into molecules that can be used for crop improvement projects.

Heavy Metal Stress and Its Consequences on Exopolysaccharide (EPS)-Producing Pantoea agglomerans.

Currently, the heavy metal pollution is of grave concern, and the part of microorganism for metal bioremediation should take into account as an efficient and economic strategy. On this framework, the heavy metal stress consequences on exopolysaccharide (EPS)-producing agricultural isolate, Pantoea agglomerans, were studied. The EPS production is a protective response to stress to survive and grow in the metal-contaminated environment. P. agglomerans show tolerance and mucoid growth in the presence of heavy metals, i.e., mercury, copper, silver, arsenic, lead, chromium, and cadmium. EDX first confirmed the metal accumulation and further, FTIR determined the functional groups involved in metal binding. The ICP-AES identified the location of cell-bound and intracellular metal accumulation. Metal deposition on cell surface has released more Ca2+. The effect on bacterial morphology investigated with SEM and TEM revealed the sites of metal accumulation, as well as possible structural changes. Each heavy metal caused distinct change and accumulated on cell-bound EPS with some intracellular deposits. The metal stress caused a decrease in total protein content and increased in total carbohydrate with a boost in EPS. Thus, the performance of P. agglomerans under metal stress indicated a potential candidate for metal bioremediation. Graphical Abstract ᅟ.

Utilization of stagnant non-potable pond water for cultivating oleaginous microalga Chlorella minutissima for biodiesel production

Utilization of waste resources to cultivate microalgae for biofuel production has the potential to improve the economics along with sustainable reuse of discarded nitrogen and phosphorous sources. The present investigation aims to cultivate an oleaginous microalga (Chlorella minutissima) in stagnant non-potable pond (SNP) water collected from different seasons (summer, winter and rainy) for biomass and lipid synthesis. The C. minutissima showed enhanced biomass productivity (245 mg/L/d) and lipid content (47 %) when grown in SNP water as compared to basal media, synthetic wastewater and recycled media respectively. The adaption in the SNP media by C. minutissima was attributed to an increase in cell size (∼2 folds) with decrease in total protein content (∼2 folds) and total carbohydrate content (∼1 fold) respectively. The fatty acid profile indicated the presence of C16:0, C16:2, C18:0, C18:1 and C18:2 as major methyl esters in the derived biodiesel. Further, the measured biodiesel properties such as specific density (0.90 g/cm3), kinematic viscosity (3.24 mm2/s), flash point (143 °C), iodine value (40.14 g I2/100 g) and acid value (3.24 mgKOH/g) abided by the ASTM D6751-02 and EN14214 standards. Therefore, such kind of SNP's have dual advantage; microalgal cultivation for biodiesel production and providing economical solution for bioremediation.

The role of mitochondria in the myocardium of senescent Meriones unguiculates

According to the mitochondrial theory of aging, changes in the functional state of mitochondria, which lead to excessive formation of active forms of oxygen, are the main factor in the development of age changes in organs and tissues of the whole organism. The assessment of the mitochondria state of the heart of senescent gerbils (Mongolian Gerbilia, Meriones unguiculates Milne-Edwards, 1867). It is proved that the aging of the heart is preceded by the appearance of dysfunction in mitochondria. The disturbance of metabolic processes in the myocardium of gerbils over the age of two years was established, which was accompanied by activation of oxidative stress by increasing the concentration of TBK-active compounds. An increase in the concentration of cytochrome C in cytosol has been shown due to the destructive effects of oxidized products on the outer membrane of mitochondria and enhancement of its permeability. The violation of bioenergetic processes, increase of the anaerobic respiration and the accumulation of lactate and unoxidative metabolites, which increases oxidative stress and cell damage, are determined. We established that for gerbils the critical age for senescence is 24 months. Major metabolic changes in the heart occur mostly at this age. This is marked by activation of prooxidants formation, proteolytic processes (decrease in total protein concentration) and inhibition of aminotransferase activity in cytosol. The switching of metabolic processes in the mitochondria of the heart with the participation of aminotransferases has been registered: increased activity of the mitochondrial isoenzyme alanine aminotransferase and reciprocal reduction of aspartate aminotransferase. After two years of age in the experimental gerbils the intensification of adaptive processes was established: activation of catalase, γ-glutamyltransferase, relative restoration of the activity of alanine and aspartate aminotransferase, thus maintaining the processes of the vital activity of the whole organism, but at a new metabolic level. With age, irreversible damage to cardiomyocytes occurs, which gradually lose the ability to convert lactate, resulting in its increase, and the processes of its utilization are inhibited. The results confirm the key role of mitochondria in the process of aging of the myocardium. However, when gerbils reach the 36 months of age the metabolic disturbances in the myocardium reach their peak, resulting in large-scale cell damage.

Oleanolic acid from antifilarial triterpene saponins of Dipterocarpus zeylanicus induces oxidative stress and apoptosis in filarial parasite Setaria digitata in vitro

Absence of a drug that kills adult filarial parasites remains the major challenge in eliminating human lymphatic filariasis (LF); the second leading cause of long-term and permanent disability. Thus, the discovery of novel antifilarial natural products with potent adulticidal activity is an urgent need. In the present study, methanol extracts of leaves, bark and winged seeds of Dipterocarpus zeylanicus (Dipterocarpaceae) were investigated for macro and microfilaricidal activity. Two antifilarial triterpene saponins were isolated from winged seed extracts by bioactivity guided chromatographic separation and identified using Nuclear Magnetic Resonance and mass spectroscopic analysis as oleanolic acid 3-O-β-D- glucopyranoside (1) (IC50 = 20.54 μM for adult worms, 19.71 μM for microfilariae ) and oleanolic acid 3-O-α-L-arabinopyranoside (2) (IC50 = 29.02 μM for adult worms, 25.99 μM for microfilariae). Acid hydrolysis of both compounds yielded oleanolic acid (3) which was non or least toxic to human peripheral blood mono nuclear cells (Selectivity index = >10) while retaining similar macrofilaricidal (IC50 = 38.4 μM) and microfilaricidal (IC50 = 35.6 μM) activities. In adult female worms treated with 50 and 100 μM doses of oleanolic acid, condensation of nuclear DNA, apoptotic body formation and tissue damage was observed by using Hoechst 33342 staining, TUNEL assay and Hematoxylin and Eosin staining respectively. A dose dependent increase in caspase 3/CED3 activity and decrease in total protein content were also observed in these parasites. A dose dependant DNA fragmentation was observed in adult parasites and microfilariae. Decreased levels of reduced glutathione (GSH) and elevated levels of glutathione S transferase (GST), superoxide dismutase (SOD) and reactive oxygen species (ROS) were also observed in parasites treated with oleanolic acid indicating an oxidative stress mediated apoptotic event. Compound 3/oleanolic acid was thus identified as a potent and safe antifilarial compound in vitro.

Temperature and Time Effects of Sous Vide on Tenderness in Beef Semitendinous Muscles

ObjectivesTo determine the time and temperature combination required to improve tenderness in undervalued cuts of beef while maintaining quality attributes; and to understand the biochemical effects of sous vide cooking and how it might be advantageous to tough cuts of meat.Materials and MethodsWhole beef semitendinosus muscles (IMPS #171C) were purchased from 2 beef groups: fed beef (< 30 mo. of age; n = 20) and cow carcasses (> 42 mo. of age; n = 20). Beef muscles were portioned into 6 cm roasts using a template. Roasts were randomly assigned to 1 of 2 cook times: 2 or 8 h at 2 different temperature treatments: 55°C and 70°C. Percent cook loss, objective color (L*a*b*, Minolta), and Warner-Bratzler shear force (WBSF, kg) were analyzed. Purge accumulated in the cook-in bag was saved and analyzed to determine soluble and total protein concentrations. Soluble protein concentrations were determined using a ThermoFisher Scientific BCA assay and total protein concentrations were determined using a Bio- Rad RC DC assay. Data were analyzed with a mixed model in JMP. The LSMeans were compared within an age classification using a Student’s t test and considered significant at P ≤ 0.05.ResultsWBSF values decreased as time increased for roasts representing the cow group when cooked at 55°C (P < 0.0144). However, roasts representing the fed group resulted in a decrease in WBSF values when temperature was increased from 55°C to 70°C (P < 0.002). Lightness values (L*) increased in both groups as time and temperature increased (P < 0.0611); whereas, redness values (a*) significantly decreased across all cooking treatments (P < 0.0145). Additionally, percent cook loss significantly increased as cooking temperature and time increased in both groups (P < 0.0322). Total protein concentrations in the purge significantly decreased as temperature and time increased in both groups (P < 0.0287). Cooking loss could be correlated to the decrease in total protein concentrations. As cooking temperature and time increase the amount of water expelled from the muscle (purge) could increase, resulting in a dilution of protein concentration. Soluble protein concentration in both age groups significantly decreased as cooking temperature and time increased (P < 0.0266).ConclusionThrough the application of sous vide cooking we can improve tenderness in tough cuts of beef, especially undervalued beef cuts originating from cows. To capitalize on value in dealing with cow meat cuts, cooking to an internal degree of doneness of 55°C (rare) for 8+ hours may be the most suitable.

Protective effects of fennel oil extract against sodium valproate-induced hepatorenal damage in albino rats.

Foeniculum vulgare (Apiaceae) is commonly known as fennel. This herb is well-known worldwide and traditionally used as curative herbal therapy for the treatment of epileptic disease, seizurescarminative, digestive, lactogogue, diuretic, treating respiratory and gastrointestinal disorders. The aim of present study is to investigate the possible effect of fennel oil against the toxicity of Sodium-Valproic (SVP) in albino rats. In order to assess the protection of fennel oil on SVP induced hepato- and nephro-toxicity, male albino rats were treated with 1 ml/kg b.w fennel oil 3 days/week for 6 weeks. The biochemical analyses of hepatic enzymes were evaluated by estimating blood biomarkers of liver and renal damage along with histological examination. The results obtained from this work showed that treating animals with SVP lead to many histopathological alterations in the liver and kidney tissues. The effect appeared in the liver tissue include leukocyte infiltrations, cytoplasmic vacuolization of the hepatocytes, fatty degeneration and congestion of blood vessels. This commonly used chemical (SVP) caused some unwanted effects on the kidney cortex which histologically observed as degeneration in renal tubules, atrophy of the glomeruli and edema. Biochemical results also revealed an abnormal increase in the enzyme level of AST, SAT, ALP, bilirubin, creatinine and urea-nitrogen, with a noticed decrease in total protein content. However, the results of treated rats with SVP plus fennel oil showed some positive histopathological changes in both the liver and kidney tissues. These results have confirmed that fennel oil has positive effects on the histological structure of the liver and kidney and the biochemical levels of AST, ALT, ALP, bilirubin, total proteins, creatinine and urea. It is concluded that fennel oil has various pharmacological properties including antioxidant, anti-cancer activity, anti-inflammatory. These valuable effects might be due to the presence of aromatic compounds trans-anethole. This useful properties of fennel plant could be due to its antioxidant activity that prevents the toxicity of SVP.

Erratum to: The effects of two preservatives on peroxidase activity and total protein content in Strelitzia reginae during cold storage

This study aimed to assess changes in peroxidase activity and total protein content of cut flowers of Strelitzia reginae that were treated with the chemical preservatives 8-hydroxyquinoline citrate (8-HQC) or chlorine during cold storage. The bases of the cut flowers were immersed in 1.5 L of tap water with 48 h pulsing treatments (a control with no chemical preservatives (0 mg·L-1, 100 mg·L-1 chlorine, 250 mg·L-1 8-HQC or 500 mg·L-1 8-HQC) during cold storage at 10.5°C and 90% relative humidity. The solutions were then replaced with tap water and the cut flowers remained in cold storage for 12 days. For biochemical characterization of peroxidase activity and total protein content, tissue samples (sepals, petals, stems, and bracts) were collected and analyzed at 0, 4, 8, and 12 days after treatment. There was an increase in peroxidase activity in petals, stems, and bracts and a decrease in total protein content, regardless of the treatment used. In this experiment, therefore, differences in peroxidase activity and total protein content due to treatments with 8-HQC or chlorine were not identified.

Virulence of Entomopathogenic Nematodes Steinernema glaseri and Heterorhabditis bacteriophora Poinar (HP88 strain) Against the Black Cutworm, Agrotis ipsilon.

Virulence of entomopathogenic nematodes, Steinernema glaseri and Heterorhabditis bacteriophora Poinar (HP88 strain) were studied against 3rd, 4th, 5th and 6th instar larvae of the black cutworm, Agrotis ipsilon. The observed mortality caused by the both tested nematodes at different time intervals was recorded. The establishment of nematodes in a host depends greatly on its ability to manage host defences so, the host haemocyte interaction with entomopathogenic nematodes were observed by encapsulation, nodule formation and phagocytosis processes. Phagocytosis is known to stimulate production of lysosomal enzymes. Acid phosphatase acts as a lysosome marker so; the activity and the pattern of acid phosphatase during the course of infection were recorded. Also, total protein and protein pattern were screened. Results indicated that, the two nematodes had variable significantly effects. S. glaseri recorded a highly significantly affect than H. bacteriophora. In addition, there are relationship between mortality rate and time exposure of S. glaseri and H. bacteriophora against 3rd, 4th, 5th and 6th larval instars of A. ipsilon i.e. the time exposure increased the susceptibility also increased. At the lower concentration, fourth and/or fifth instars were the most susceptible instars to S. glaseri ,while the 3rd and 4th instar larvae were most vulnerable instars to the infection with H. bacteriophora. Results was recorded that, six haemocyte types were identified in A. ipsilon 6th instar larvae: prohaemocytes, plasmatocytes, granulocytes, spherulocytes, oenocytoids, and adipohaemocytes. S. glaseri in A. ipsilon escape encapsulation and overcome host immunity faster than H. bacteriophora which recognized by host haemocytes. Prohemocytes, plasmatocytes or granulocytes are phagocytic cells in A. ipsilon 6th instar larvae. Phagocytosis was expressed in A. ipsilon haemocytes infected with H. bacteriophora at a higher rate compared with S. glaseriinfection. Data demonstrated both S. glaseri and H. bacteriophora induced a significant decrease in total protein contents after 24hrs from infection. While, there was significant increase in acid phosphatase activity after 6 hrs of infected groups of S. glaseri and H. bacteriophora. This increase may be due to phagocytosis processes which recorded in the present study at this time because phagocytosis is known to stimulate production of lysosomal enzymes. There is an acid phosphatise band detected only in H. bacteriophora and S. glaseri samples with Rf 0.86 after 6 and 12hrs from infection. This band may refer to nematodes exudates which contain high amount of this enzyme that the nematodes can easily penetrate and digest host tissues. There is a characterestic band of S. glaseri protein pattern with Mw 47KDa. It was identified as surface coat protein of S. glaseri which play important roles in defeating the host immune system by its antiphagocytic activity.

Rhizome extracts of Curcuma zedoaria Rosc induce caspase dependant apoptosis via generation of reactive oxygen species in filarial parasite Setaria digitata in vitro

Human lymphatic filariasis (LF) is mainly caused by filarial parasite Wuchereria bancrofti and is the second leading cause of long term and permanent disability in tropical countries. To date, incapability to eliminate long lived adult parasites by current drugs remains the major challenge in the elimination of LF. Hence, in the current study, the efficacy of rhizome extracts of Curcuma zedoaria (a plant traditionally used in Sri Lanka in the management of LF) was evaluated as an effective filaricide in vitro. Sequential solvent extracts of C. zedoaria rhizomes were screened for in vitro antifilarial activity at 0.01–1 mg/mL concentrations by motility inhibition assay and 3-(4, 5 dimethylthiazol-2-yl)-2, 5 diphenyl tetrazolium bromide (MTT) reduction assay using cattle parasite Setaria digitata as a model organism. Exposure of parasites to hexane and chloroform extracts of C. zedoaria caused a dose dependant reduction in motility and viability of microfilariae (IC50 = 72.42 μg/mL for hexane extract, 191.14 μg/mL for chloroform extract) and adult parasites (IC50 = 77.07 μg/mL for hexane extract, 259.87 μg/mL for chloroform extract). Both extracts were less toxic to human peripheral blood mononuclear cells when compared to filariae. A dose dependant increase in caspase 3/CED 3 and a decrease in total protein content, cyclooxygenase (COX) and protein tyrosine phosphatase (PTP) activities were observed in adult parasites treated with hexane or chloroform extract. A significant degree of chromatin condensation and apoptotic body formation were also observed in these worms by Hoechst 33342 and terminal deoxynucleotidyl transferase-mediated dUTP biotin nick end labeling (TUNEL) staining respectively. Dose dependant chromosomal DNA laddering was observed in treated adult worms but not in microfilariae in response to both extracts. Oxidative stress parameters such as reduction in reduced glutathione (GSH) levels and increase in glutathione s transferase (GST), superoxide dismutase (SOD) and catalase activities, increased reactive oxygen levels (ROS) and lipid peroxidation were also observed indicating that an apoptotic event is induced by reactive oxygen species.

Toxic Effects of Quinalphos on Protein Content and Growth Rate in Freshwater Teleost, &lt;I&gt;Cyprinus carpio&lt;/I&gt; (L.)

Organophosphate based pesticides account for a major percentage of pesticides used in domestic, agricultural and industrial applications throughout the world. They are highly popular because they are effective, non persistent and relatively less expensive. Quinalphos is a synthetic organophosphate which has become a matter of concern because of its potentiality and hazardous effect. Hence, the present investigation was designed to assess the impact of quinalphos on total protein content in the selected tissues and growth rate of freshwater fish, Cyprinus carpio. The lethal toxicity of quinalphos to the fish exposed for 96 h was found to be 2.75 ppm. For sublethal toxicity study, the fish were exposed to two concentration viz., 1/5 th of LC 50 (0.550 ppm) and 1/10 th of LC 50 (0.275 ppm) along with control as reference for 4, 8, 16 and 32 days. Decrease in total protein concentration and growth rate were observed in 4 th and 8 th day of exposure, but on 16 th and 32nd day of exposure all the values reached nearer to normal condition. The decline in total protein levels and relative growth rate at initial exposure periods may indicates the high energy demand associated with imposed quinalphos stress. Restoration of protein fractions to normal, implies that after 8 days of exposure there seems to exist an oscillatory phase in protein turnover towards a more synthetic phase leading to the establishment of recuperation and adaptation phenomena.
