Taxol-induced microtubule asters in mitotic extracts of Xenopus eggs: requirement for phosphorylated factors and cytoplasmic dynein.

Taxol, a microtubule stabilizing drug, induces the formation of numerous microtubule asters in the cytoplasm of mitotic cells (De Brabander, M., G. Geuens, R. Nuydens, R. Willebrords, J. DeMey. 1981. Proc. Natl. Acad. Sci. USA. 78:5608-5612). The center of these asters share with spindle poles some characteristics such as the presence of centrosomal material and calmodulin. We have recently reproduced the assembly of taxol asters in a cell-free system (Buendia, B., C. Antony, F. Verde, M. Bornens, and E. Karsenti. 1990. J. Cell Sci. 97:259-271) using extracts of Xenopus eggs. In this paper, we show that taxol aster assembly requires phosphorylation, and that they do not grow from preformed centers, but rather by a reorganization of microtubules first crosslinked into bundles. This process seems to involve sliding of microtubules along each other and we show that cytoplasmic dynein is required for taxol aster assembly. This result provides a possible functional basis to the recent findings, that dynein is present in the spindle and enriched near spindle poles (Pfarr, C. M., M. Cove, P. M. Grissom, T. S. Hays, M. E. Ultrastructural localization of alpha-actinin and filamin in cultured cells with the immunogold staining (IGS) method.

Monospecific antibodies to chicken gizzard actin, alpha-actinin, and filamin have been used to localize these proteins at the ultrastructural level: secondary cultures of 14-d-old chicken embryo lung epithelial cells and chicken heart fibroblasts were briefly lysed with either a 0.5% Triton X-100/0.25% glutaraldehyde mixture, or 0.1% Triton X-100, fixed with 0.5% glutaraldehyde, and further permeabilized with 0.5% Triton X-100, to allow penetration of the gold-conjugated antibodies. After immunogold staining (De Mey, J., M. Moeremans, G. Geuens, R. Nuydens, and M. De Brabander, 1981, Cell Biol. Int. Rep. 5:889-899), the cells were postfixed in glutaraldehyde-tannic acid and further processed for embedding and thin sectioning. This approach enabled us to document the distribution of alpha-actinin and filamin either on the delicate cortical networks of the cell periphery or in the densely bundled stress fibers and polygonal nets. By using antiactin immunogold staining as a control, we were able to demonstrate the applicability of the method to the microfilament system: the label was distributed homogeneously over all areas containing recognizable microfilaments, except within very thick stress fibers, where the marker did not penetrate completely. Although alpha-actinin specific staining was homogeneously localized along loosely-organized microfilaments, it was concentrated in the dense bodies of stress fibers. The antifilamin-specific staining showed a typically spotty or patchy pattern associated with the fine cortical networks and stress fibers. This pattern occurred along all actin filaments, including the dense bodies also marked by anti-alpha-actinin antibodies. The results confirm and extend the data from light microscopic investigations and provide more information on the structural basis of the microfilament system.

The biochemistry of microtubules. A review.

The whole of the complex field of microtubule structure and function is far too broad to cover in this brief review. The intention has been to give a brief overview of the field and to place emphasis on a few areas of recent significant progress. In particular, the related topics of microtubule “treadmilling,” polarity of assembly, and role of nucleotide hydrolysis have been addressed at some length, as has the topic of tubulin heterogeneity. For a more detailed discussion, several recent books can be recommended. Microtubules (Roberts and Hyams, eds., 1979) contains excellent reviews on the major areas of microtubule structure, chemistry, mechanisms of assembly, and functions. Microtubules and Microtubule Inhibitors (De Brabander and De Mey, eds., 1980) contains briefer articles on many topics of current interest. In Microtubules (1978), P. Dustin has authored a remarkably comprehensive review which is especially strong in providing historical perspective, to which this present review is heavily indebted. Also to be recommended is the more recent book Biological Functions of Microtubules and Related Structures (Sakai et al.,1982).

Modification of Vimentin Polypeptides during Mitosis

A number of cyclic coordinated events take place in mitosis which lead to a major reorganization of the microfilaments (Sanger 1975; Fujiwara and Pollard 1976; Sanger and Sanger 1976; Schroeder 1976; Fujiwara et al. 1978; Herman and Pollard 1979) and microtubules (Brinkley et al. 1976; De Brabander et al. 1979). Contrary to the other two filamentous systems, the intermediate-size filaments remain intact throughout mitosis in most cells studied (Blose and Chacko 1976; Hynes and Destree 1978; Blose 1979; Aubin et al. 1980), although in BHK-21 cells, they disaggregate in prometaphase and reappear as nuclear caps during spreading of the daughter cells (Starger et al. 1978). The studies of Aubin et al. (1980) in PtK2 cells further showed that the vimentin and cytokeratin filaments distribute differently during mitosis and suggested a role for the vimentin filaments in orienting the spindle and/or the chromosomes during mitosis.
