Determination of Total Antioxidant Capacities as Ascorbic Acid Equivalent of Tea Extract Samples from Different Brands Using Digital Image-Based Colorimetric Detection Method

In this study, the Digital Image-Based Colorimetric Detection Method developed by Bakırdere et al. was used to find the TAC (Total Antioxidant Capacity) value of tea samples from different brands. To determine the total amount of antioxidants in tea samples, the CUPRAC (cupric ion reducing antioxidant capacity) method, which is widely used in antioxidant determination, was combined with a digital image-based colorimetric detection system. To use in our study, a box with opaque wood material measuring 24 cm x 19 cm x 17 cm (width/length/depth) was designed and manufactured. In the analysis, the oxidation reaction between the chromogenic copper(II)-neocuproine (Cu(II)-Nc) reagent and antioxidants was utilized. The color change that occurs as a result of the oxidation was calculated using an application on smartphones. In our study, analyzes were performed on 4 different brand tea extract samples (tea A, tea B, tea C, tea D) to determine the total antioxidant capacity of ascorbic acid equivalent. The TAC values for ascorbic acid equivalent in tea extract samples were found as 380 ± 8 mg/L (tea A), 402 ± 4 mg/L (tea B), 213 ± 3 mg/L (tea C), 232 ± 4 mg/L (tea D) using the digital image-based colorimetric detection systems.

Kırgız Cumhuriyeti Issyk-Kul Bölgesinde Yetişen Yalanci İğde (Hippophae rhamnoides L.) Meyvelerinin Kimyasal Kompozisyonu ve Antioksidan Aktivitesi

The fruit of sea-buckthorn (Hippophae rhamnoides L.) is a rich source of nutrients and phytochemicals. This study determined the chemical composition and antioxidant activity of sea-buckthorn grown in Issyk-Kul region, Kyrgyz Republic, during maturation from August to November. Three different assays [ferric reducing antioxidant power (FRAP), DPPH radical scavenging activity (DPPH), and cupric ion reducing antioxidant capacity (CUPRAC)] were used to determine the antioxidant activity of sea-buckthorn fruits. Sea-buckthorn fruits had a sugar content of 6.57-8.08 g/100g, a dietary fiber content of 0.39-0.83 g/100g, an ash content of 0.49-0.99 g/100g, a lipid content of 6.56-6.98 g/100g and titratable acidity of 1.38-1.92%. The total phenolic content and total flavonoids of sea-buckthorn fruits were 408.52–886.48mg GAE/kg and 260.81-345.37mg CE/kg, respectively. Antioxidant activity values determined by FRAP, DPPH, and CUPRAC were 88.51-1041.22mg TE/kg, 1011.93-2673.22mg TE/kg, and 755.40-1232.55mg TE/kg, respectively. Total carotenoid content was 16.6-18.8 mg/kg. Results showed the relationship between the change in the color of sea-buckthorn berries and the chemical composition by the months of their ripening and the influence of weather conditions. All these results indicated that sea-buckthorn fruits could be beneficial for human nutrition while they might have a great potential to be used in a wide range of products to enrich their functional properties.

Chemical Profile and Skin-Beneficial Activities of the Petal Extracts of Paeonia tenuifolia L. from Serbia.

Without being aware of its chemical makeup, many ancient societies have used Steppe peony in their traditional medicine. Given that modern phytopreparation intended for use on human skin requires, above all, knowledge of its chemical composition, the goal of this study was to make a screening of the composition of aqueous and methanolic extracts of the petals of P. tenuifolia L. and to examine them for various skin-beneficial properties. The extracts were prepared by maceration, ultrasound-assisted, and microwave-assisted extraction procedures. The chemical profiling was conducted by the use of UHPLC-LTQ-OrbiTrap MS and UHPLC/MS, and spectrophotometric methods for the determination of total polyphenol and total flavonoid contents. The biological activities entailed antioxidant ABTS, DPPH, CUPRAC (Cupric Ion Reducing Antioxidant Capacity), and FRAP (Ferric Reducing Antioxidant Power) assays, antimicrobial (antibacterial and antifungal) and antibiofilm activities, cytotoxicity, wound healing potential, as well as the adhesion and invasion of Staphylococcus lugdunensis. The results showed that the petals are rich in phenolic acids and flavonoids, which are commonly associated with numerous biological activities. The aqueous extracts were more efficient in the majority of the bioactivity assays then the methanolic ones, whereas the optimal extraction method varied between the assays. This study is the first step towards the safe use of the aqueous extracts of P. tenuifolia petals for therapeutic skin treatments.

Colorimetric Nanobiosensor Design for Determining Oxidase Enzyme Substrates in Food and Biological Samples.

Biological enzymes have high catalytic activity and unique substrate selectivity; their immobilization may provide cost reduction and reusability. Using magnetic nanoparticles (MNPs) as support materials for enzymes ensures easy separation from reaction media by an external magnetic field. Thus, MNPs were prepared by the coprecipitation method, coated with silanol groups, then -NH2-functionalized, and finally activated with glutaraldehyde. Finally, three different oxidase enzymes, i.e., uricase, glucose oxidase, and choline oxidase, were separately immobilized on their surfaces by covalent attachment. Hence, colorimetric nanobiosensors for the determination of three biologically important substrates, i.e., uric acid (UA), glucose (Glu), and choline (Ch), were developed. Hydrogen peroxide liberated from enzyme-substrate reactions was determined by the cupric ion reducing antioxidant capacity (CUPRAC) reagent, bis-neocuproine copper(II) chelate. In addition, UA-free total antioxidant capacity could also be measured via the developed system. Kinetic investigations were carried out for these nanobiosensors to enable the calculation of their Michaelis constants (K m), revealing no affinity loss for the substrate as a result of immobilization. Enzyme-immobilized MNPs could be reused at least five times. The linear concentration ranges were 5.43-65.22 μM for UA, 11.1-111.1 μM for Glu, and 2.78-44.4 μM for Ch, and the limit of detection (LOD) values with the same order were 0.34, 0.59, and 0.20 μM, respectively. Besides phenolic and thiol-type antioxidants, UA could be determined with an error range of 0.18-4.87%. The method is based on a clear redox reaction with a definite stoichiometry for the enzymatically generated H2O2 (which minimizes chemical deviations from Beer's law of optical absorbances), and it was successfully applied to the determination of Glu and UA in fetal bovine serum and Ch in infant formula as real samples.

Farklı Tekniklerle Ekstrakte Edilen Propolisin Polifenol İçerikleri Ve Antioksidan Aktivitelerinin Değerlendirilmesi

Propolis is classified as an opotherapeutic medicine due to the botanical origin of the resins. The chemical composition of propolis is greatly influenced by the honeybee species, botanical source and extraction techniques. Within this frame, we compared the same propolis’ polyphenol contents and antioxidant activities prepared with different techniques. Four types of extracts were prepared. The first type was prepared classically by ethyl alcohol (POH). The second and third types were extracted by sterile distilled water kept as both sterilised (PS) and non-sterilized (PN). The fourth one was prepared with full vacuumed and dried propolis with honey (PH). The antioxidant activity of extracts was evaluated with DPPH radical scavenging, ABTS radical cation scavenging, Cupric ion reducing antioxidant capacity. Also total phenolic and flavonoid content of extracts were investigated. POH extract showed significantly high content of total phenol and flavonoids which followed by PN, PS and PH. POH showed approximately two times higher activity on DPPH radical (IC50=4,39μg/mL) compared with quercetin as references. The highest activity on DPPH is shown by POH with 4,39 μg/mL of IC50 value which was followed by aqueous extracts 18,08. The lowest activity was shown by PS with 4,39 μg/mL of IC50 value. The highest scavenging activity against ABTS radical cation was shown by POH (73,37 mg TE/g extract) and the lowest activity was shown by PS (34,21 mg TE/g extract). According to the results, the new aqueous extraction technique is promising with relatively high polyphenol contents and antioxidant activities. Also honey with propolis can be an alternative product, although it has relatively lower values of antioxidant activity.

Antioxidant Activity of DPPH, CUPRAC, and FRAP Methods, as well as Activity of Alpha-Glucosidase Inhibiting Enzymes from Tinospora crispa (L.) Stem Ultrasonic Extract

Aims: The goal of this work was to get the stem extract of Tinospora crispa (L.) using ultrasoundassisted extraction (UAE) with duration and amplitude changes to achieve optimal extraction conditions. The antioxidant potential of the extract as measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH), CUPRAC (Cupric Ion Reducing Antioxidant Capacity), FRAP (Ferric Reducing Ability of Plasma), and its antidiabetic potential. Results: The third test of antioxidant methods showed that the measurement of antioxidant activity with, DPPH, CUPRAC, and FRAP gave significantly different results, but CUPRAC and FRAP gave the same response to the ethanol extract of Tinospora crispa stems, it can be concluded that the extract of Tinospora crispa stems in all treatments had strong antioxidant activity using either the CUPRAC or methods FRAP. In addition, it is very active as an antidiabetic by inhibiting alpha glucosidase. The optimum extract selected was extract B (extraction time was 35 minutes and amplitude 65%) with IC50 values for antioxidant activity in the CUPRAC method of 72.53 ± 0.18 mg/L and the FRAP method of 152.29 ± 2.16 mg/L, while the inhibition of alpha glucosidase was 0.30 ± 0.006 mg/L.

Phenolic compounds from Gomphrena globosa L.: phytochemical analysis, antioxidant, antimicrobial, and enzyme inhibitory activities in vitro

ABSTRACT In the present study, phenolic compounds from Gomphrena globosa L. (GGLPCs) including total phenols, flavonoids, flavonols, flavanols, and phenolic acids were measured by spectrophotometric method. The antioxidant activities of GGLPCs were assessed by radical scavengings of 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2′-azinobis-(3-ethylbenzothiazoline -6-sulfonic acid)-diammonium salt (ABTS) cation, oxygen radical absorbance capacity (ORAC), phosphomolybdenum antioxidative power (PAP), ferric reducing antioxidant power (FRAP), and cupric ion reducing antioxidant capacity (CUPRAC) assays. Antimicrobial and enzyme inhibitory activities of GGLPCs in vitro were investigated using micro-plate dilution method and colorimetry, respectively. The results showed that GGLPCs containing abundant phenolic compounds, had obvious antioxidant and antimicrobial activities (especially against Escherichia coli and Listeria monocytogenes), and significant inhibitory effects on lipase, α-amylase, α-glucosidase, and trypsin in vitro. Thus, the results showed that GGLPC may be used as a natural antioxidant and antibacterial agent, as well as drugs for the treatment of hyperlipidemia, diabetes, and inflammation-related diseases.

Enzymatic determination of hypoxanthine in fish samples as a freshness indicator using the CUPRAC colorimetric sensor

Fish consumption is essential for a healthy diet. However, all seafood including fish are susceptible to deterioration unless properly preserved. Controlling the freshness of fresh or packaged fish is a challenging issue for the food industry in terms of human health and shelf life determination. One of the main indicators showing the freshness of fish is undoubtedly the amount of hypoxanthine (Hx). As soon as the organism dies, Hx begins to be released with the cessation of ATP synthesis and shows a gradual increase over time. Therefore, Hx determination is an important indicator in the control of fish freshness. Based on this fact, a colorimetric method for the enzymatic determination of Hx using the CUPRAC (Cupric ion Reducing Antioxidant Capacity) sensor was developed. Uric acid (UA) and H2O2 are enzymatically produced by xanthine oxidase (XOD) from Hx, and both products respond to the CUPRAC reagent to produce the cuprous neocuproine (Cu(I)-Nc) chromophore chelate formed in situ on a Nafion anionic membrane on which the cationic Cu(II)-Nc complex was fixed. Hx was measured at different time intervals in the meat samples taken from sea bass (Dicentrarchus labrax), which was left to stand at room temperature for a time period between 0 and 24 h; the level of spoilage was determined from the coloration of the CUPRAC membrane sensor (via absorbance measurement at 450 nm). It was observed that there was a linear increase in the amount of Hx during the measurement period. The method was optimized for Hx determination, verified with interference analysis and standard additions to real samples, and validated against HPLC. The linear detection range of the developed method for Hx was 2.0–32.0 μM with an LOD of 0.79 μM, and early stages of fish degradation could be detected at several nanomoles of Hx per gram of fish meat. The proposed method was demonstrated to have distinct superiority over many recent colorimetric sensors of fish freshness in regard to its lower LOD for Hx, wider linear range, capability to cope with interferents (including biologically important antioxidants, such as cysteine, reduced glutathione, ascorbic acid, UA and α-tocopherol) and applicability to real samples.

Impact of Aqueous Extracts of Turkish Wild Edible Plants on Acrylamide Formation in Potato Crisps

Background: Antioxidants have the ability to influence acrylamide formation. This study aimed to evaluate the impact of aqueous extracts of six wild edible plants on the acrylamide formation in potato crisps.
 Methods: Sliced potatoes were submerged in the plant extracts at a concentration of 0, 5, and 10 g/L for 1, 5, and 10 min. Before being fried and their acrylamide levels were calculated by Liquid Chromatography-Tandem Mass Spectrometry (LC-MS/MS).
 Results: Aqueous extract of ribwort plantain was found the most effective trial at 10 g/L for 5 min because it reduced acrylamide concentration by 57% compared to control without significantly affecting potato crisps’ sensory and color parameters (p>0.05). The aqueous extract of shepherd’s-needle yielded the highest Total Antioxidant Capacity (TAC) in 2,2′-Azino-bis (3-ethylbenzothiazoline-6-sulfonic acid; ABTS) and Cupric ion Reducing Antioxidant Capacity (CUPRAC) assay, the highest Total Phenolic Content (TPC), and Total Flavonoid Content (TFC). Similarly, no significant correlation was found between TAC, TPC, and TFC of watery plant extracts with acrylamide level of potato crisps produced after immersion of these extracts (at 5 g/L for 5 min).
 Conclusion: Wild edible plants have the potential to be used for acrylamide reduction in potato crisps.

Synthesis, spectroscopic characterization, density functional theory study, antimicrobial and antioxidant activities of curcumin and alanine-curcumin Schiff base

In this work, a novel Schiff-base derived from curcumin and L-Alanine was synthesized under microwave conditions in excellent yield. The structural characterization has been carried out from their elemental analyses, FTIR, UV–Vis and 13C-NMR and 1H-NMR spectral techniques. The Schiff base (Cur-Ala) and curcumin (Cur) have been screened for their antimicrobial activity toward some pathogens clinically important microorganisms: Bacillus subtilis, Escherichia coli and Staphylococcus aureus, Aspergillus niger and Candida albicans. Result found that the Schiff base was more active than the curcumin. The antibacterial and antifungal activities of Cur-Ala can be attributed to its greatest dipole moment, as shown by theoretical calculations. Also, the antioxidant activity of Schiff base and curcumin were studied by DPPH, cupric ion reducing antioxidant capacity and o-phenanthroline techniques. Results indicate that Cur-Ala and Cur show more antioxidant activities than the standard antioxidants (BHT and BHA). Quantum chemical parameter calculations of Cur-Ala and Cur have been investigated by DFT using B3LYP/6-31G (d,p) basis set method to calculate the optimized structure, atomic charges, MESP, global reactivity descriptors and thermomolecular proprieties of both molecules. Antioxidant Capacity and Potential as an Alpha-Glucosidase Inhibitor in Phaleria macrocarpa (Scheff.) Boerl Fruit Peel Ultrasonic Extract

Phaleria macrocarpa (Scheff.) Boerl is native to the tropical region of Papua Island, Indonesia, and has traditionally been used as a herbal drink, either alone or in combination with other medicinal plants, to treat diseases such as cancer, hypertension, and diabetes mellitus. This study aimed to obtain the fruit peel extract of Phaleria macrocarpa (Scheff.) Boerl. through the application of ultrasound-assisted extraction (UAE) with variations in time and amplitude to produce optimal extraction conditions. The potential of the extract as an antioxidant using the cupric ion reducing antioxidant capacity (CUPRAC) and ferric reducing antioxidant power (FRAP) methods, and its potential as an antidiabetic through alpha glucosidase inhibition. The optimum extract selected was extract C (extraction time was 45 minutes and amplitude 60%) with IC50 values for antioxidant activity in the CUPRAC method of 39.63 ± 0.009 mg/L and the FRAP method of 77.37 ± 0.8 mg/L, while the inhibition of alpha glucosidase was 0.45 ± 0.007 mg/L. It can be concluded that the ethanol extract of Phaleria macrocarpa fruit peel has the potential as a source of antioxidants and anti-diabetic.

Effects of Methods and Durations of Extraction on Total Flavonoid and Phenolic Contents and Antioxidant Activity of Java Cardamom (Amomum compactum Soland Ex Maton) Fruit

Free radicals contribute to the pathophysiology of degenerative diseases which increase mortality globally, including mortality in Indonesia. Amomum compactum Soland. Ex Maton fruit from the Zingiberaceae family, also known as Java cardamom, contains secondary metabolites that have high antioxidant activities. The antioxidant activity of the methanol extract of Java cardamom fruit correlates with its flavonoid and phenolic compound contents, which can be affected by different methods and durations of extraction. This study aimed to measure and compare the effects of extraction methods and durations on total flavonoid and phenolic contents (TFCs and TPCs) and subsequent antioxidant activities by the 2,2′-diphenyl-1-picrylhydrazyl (DPPH) radical, ferric reducing antioxidant power (FRAP), 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonate) (ABTS), and cupric ion reducing antioxidant capacity (CUPRAC) assays. Methanol extracts of Java cardamom were produced by continuous shaking (CSE), microwave-assisted (MAE), or ultrasonic-assisted extractions (UAE) for three different durations. CSE for 360 min resulted in the highest TFCs (3.202 mg Quercetin Equivalent/g dry weight), while the highest TPCs (1.263 mg Gallic Acid Equivalent/g dry weight) were obtained by MAE for 3 min. Out of the investigated methods, MAE for 3 min resulted in the highest antioxidant activity results for the extracts. We conclude that the polyphenolic antioxidant yield of Java cardamom depends on two parameters: the method and the duration of extraction.

Lipase-Catalyzed Synthesis, Antioxidant Activity, Antimicrobial Properties and Molecular Docking Studies of Butyl Dihydrocaffeate.

Green chemistry approaches, such as lipase-catalyzed esterification, are promising methods for obtaining valuable chemical compounds. In the case of the use of lipases, unlike in aqueous environments, the processes of the ester bond formations are encountered in organic solvents. The aim of the current research was to carry out the lipase-catalyzed synthesis of an ester of dihydrocaffeic acid. The synthesized compound was then evaluated for antioxidant and antimicrobial activities. However, the vast majority of its antioxidant activity was retained, which was demonstrated by means of DPPH· (2,2-diphenyl-1-picrylhydrazyl) and CUPRAC (cupric ion reducing antioxidant capacity) methods. Regarding its antimicrobial properties, the antifungal activity against Rhizopus oryzae is worth mentioning. The minimum inhibitory and fungicidal concentrations were 1 and 2 mM, respectively. The high antifungal activity prompted the use of molecular docking studies to verify potential protein targets for butyl ester of dihydrocaffeic ester. In the case of one fungal protein, namely 14-α sterol demethylase B, it was observed that the ester had comparable binding energy to the triazole medication, isavuconazole, but the interacted amino acid residues were different.

Unused Parts of Jackfruit (Artocarpus heterophyllus): Prospective In Vitro Antioxidative Activity

BACKGROUND: Free radical is unstable and highly reactive, which may lead to oxidative stress that causes various diseases, that is, diabetes mellitus. Antioxidant can prevent oxidation process by scavenging free radicals. Jackfruit (Artocarpus heterophyllus) is a native tropical fruit that can easily be found in Indonesia. When the flesh is commonly eaten, the unused parts – such as the leaves, fruit peels, and pulps will be considered waste to be thrown away. However, these unused parts of Jackfruit are rich in antioxidant compounds that potentially can work as therapeutic agents. AIM: The aim of the study was to determine the antioxidant properties of leaves, peels, and pulps of A. heterophyllus by calculating their antioxidant activity index (AAI) with 2,2-diphenyl-1-picrylhydrazyl (DPPH) and Cupric Ion-Reducing Antioxidant Capacity (CUPRAC) method; total phenolic content (TPC) and total flavonoid content (TFC); observing the correlation between TPC and TFC with AAI DPPH and CUPRAC; as well as the correlation between AAI DPPH and CUPRAC. MATERIALS AND METHODS: Extraction process was carried out using reflux method using three different polarity solvents. UV-visible spectrophotometer was used to determine the TPC, TFC, AAI DPPH, and AAI CUPRAC. Pearson’s method was used to observe the correlation between TPC and TFC with AAI DPPH and CUPRAC, as well as the correlation between both methods. RESULTS: The AAI in DPPH method were varied from 0.0310 to 36.8852, while CUPRAC from 0.1156 to 1.2503. Ethanol leaves extract gave the highest TPC value (5.53 g GAE/100 g) and n-hexane peels extract exposed the highest TFC value (16.07 g QE/100 g). The correlation between TPC and AAI of leaves, peels, and pulps extracts with DPPH method, as well as between TFC and AAI CUPRAC of peels extracts was positive and significant. Rutin was determined as the marker compound, valuing at 0.0106%. CONCLUSION: Phenols and flavonoids (including rutin) content contributed to DPPH and CUPRAC antioxidant activity. The antioxidant property between both methods was not linear in leaves, peels, and pulps extracts. Unused parts (peels and leaves) of A. heterophyllus might be potential to be developed as natural antioxidant sources.

Stability and Antioxidant Activity of Hydro-Glyceric Extracts Obtained from Different Grape Seed Varieties Incorporated in Cosmetic Creams

Grape seeds are agro-industrial by-products, which if improperly managed, may be responsible for socioeconomic and environmental problems. Nevertheless, it is possible to effectively valorize them by means of extraction of the bioactive compounds, especially the antioxidant phenolic molecules, using a safe, green, and environmentally-friendly extractive medium (i.e., hydro-glyceric solution). In the present study, the extraction was performed using seeds from two Lebanese varieties, Obeidi and Asswad Karech, and three international varieties, Marselan, Syrah, and Cabernet Franc. The type and amount of phenolic compounds were identified by High-Performance Liquid Chromatography (HPLC). Marselan was the extract richer in catechins (132.99 ± 9.81 μg/g of dried matter), and it also contained a higher amount of phenolic compounds (49.08 ± 0.03 mg of gallic acid equivalent/g of dry matter and 10.02 ± 0.24 mg of proanthocyanidin content/g of dry matter). The antioxidant capacity of the extracts was assessed using three different colorimetric assays including 2,2-DiPhenyl-1-PicrylHydrazyl (DPPH), CUPRIC ion Reducing Antioxidant Capacity (CUPRAC), and Ferric Reducing Antioxidant Power (FRAP). As expected, Marselan exhibited the highest antioxidant activity; as well, the total phenolic and proanthocyanidin content were the highest. The stability of the Marselan extract incorporated into a commercial cream, was performed at three different temperatures (4, 25, and 50 °C), and four different concentrations (5, 4, 3, 2%), over a period of 4 months, using different methods such as centrifugation, Heat-Shock Cycles, pH, and viscosity. All Marselan hydro-glyceric extract formulations were proven to be stable over the entire 4 months, where the highest stability was achieved at 4 °C and the least at 50 °C. This study supports the suitability of the incorporation of phenolic extracts into commercial creams to enrich the cosmetic industry with effective, natural, and safe skincare products.

Feline obesity causes hematological and biochemical changes and oxidative stress - a pilot study.

Obesity, an extremely important factor in feline clinical practice, is estimated to affect up to one third of the feline population. Moreover, it can trigger chronic inflammation, which could predispose to oxidative stress by increasing reactive oxygen species, thereby generating potentially irreversible cellular damage. This study analyzed hematological, biochemical and oxidative stress profiles at various degrees of feline obesity. Forty-five cats were selected and divided into three groups: control (n = 17), overweight (n = 13) and obese (n = 15), after clinical and laboratory evaluation and body condition score. Biochemical and oxidative stress analyses were performed using a photocolorimeter and hematological analyses were performed in a veterinary cell counter. Obese cats showed increased mean corpuscular volume (MCV), red cell distribution width (RDW), HDL cholesterol and triglycerides and decreased activity of gamma-glutamyl transferase (GGT) than control cats, although within the reference ranges for the species. As for oxidative stress, obese cats showed higher total antioxidant capacity (TAC), by the inhibition of 2,2'-Azino-Bis-3-Ethylbenzthiazoline-6-Sulfonic Acid (ABTS), inhibition of ABTS associated with horseradish peroxidase (ABTS + HRP), cupric ion reducing antioxidant capacity (CUPRAC) and ferric reducing antioxidant power (FRAP) methods, while overweight cats had a higher TAC-ABTS + HRP and TAC-FRAP than control cats. We conclude that the conditions of natural obesity and overweight in the feline species alter its hematological, biochemical and oxidative stress parameters.

Contribution to Pharmacological Valorisation of Algerian Arctium minus (Hill) Bernh. Subsp. Atlanticum (Pomel) Maire; Antioxidant an d Acetylcholinesterase Inhibitory Activities

Background: The traditional pharmacopoeia is full of potential resources for molecules with therapeutic effects involving the inhibition of enzymes linked to some diseases. Objective: This work aimed to test in vitro neuroprotective activity against Alzheimer's disease (AD) combined with the antioxidant effect of root extracts obtained by water, water/methanol, and ethyl acetate of the endemic Arctium minus. subsp. Atlanticum, a native of Algeria. Methods: The different extracts of the root of the studied plant were obtained by decoction or maceration. The inhibitory effect of acetyl/butyrylcholinesterase (AChE/BChE) was achieved by a colorimetric method. Similarly, the antioxidant activity was measured based on several mechanisms: 1, 1- diphenyl-2-picryl-hydrazyl (DPPH) and galvinoxyl (GOR) radicals free scavenging assays, β-carotene bleaching inhibition activity, and cupric ion reducing antioxidant capacity (CUPRAC). Results: In the various tests carried out, the ethyl acetate extract (EAE) possessed the most powerful antioxidant and anticholinesterase activities compared to the other samples. The IC50 and A0.5 values of DPPH, GOR, β-carotene, CUPRAC, anti-AChE, and anti-BChE assays were 69.45±5.49, 28.87±0.18, 121.58±16.76, 37.41±1.59, 265±21, and 240±6.3 μg / mL, respectively. Likewise, a correlation could be deduced between the type of extract and the potent antioxidant/anticholinesterase potential. Conclusion: This work examines for the first time the anticholinesterase potential combined with the antioxidant effect of Algerian Arctium minus. subsp. atlanticum. This association between the two effects could be effective in the fight against AD, and therefore, the use of this natural resource may be possible in combating this aspect of neurodegeneration.

Characteristics of seaweed salt residue Sargassum polycystum and coffee dregs as raw materials for body scrubs

Body scrub contains coarse grains that act as sandpaper (obrasiver). Seaweed salt residue can be a substitute for microbeads in body scrub. The production body scrub from the residue of Sargassum polycystum seaweed can be added with coffee dregs. This study aims the characteristics of the raw material and the effect of the addition of coffee dregs on the characteristics of a body scrub. The research contained three-stage, namely preparation of seaweed salt residue, coffee dregs, and production of the body scrub. The body scrub preparations were carried out by mixing the water phase and the oil phase. The addition of coffee dregs with a concentration (0; 1; 2; 3%) to the body scrub. The selected product was a body scrub with 7% Sargassum polycystum seaweed salt residue and 3% coffee dregs (S 7:3). The pH value of the selected product was 6.76 and the dispersion was 3.57 cm. The emulsion stability test showed that the cream was in a stable state. The antioxidant activity of the selected product using the 1.1-diphenyl-2-picryl-hydrazyl (DPPH) method was 55.54 ppm and the Cupric Ion Reducing Antioxidant Capacity (CUPRAC) was 78.84 (μM/g). The results showed that body scrubs have good moisture for the skin.

Phytochemical analysis, UPLC-ESI-Orbitrap-MS analysis, biological activity, and toxicity of extracts from Tripleurospermum limosum (Maxim.) Pobed

Tripleurospermum limosum (TL) has been used in folk medicine to treat gastritis. Toward the further development and use of TL, we report the phytochemical profiling, determination of active components, and antioxidant and enzyme inhibitory activities of TL. Nineteen compounds were identified by ultra-performance liquid chromatography-electrospray ionization-orbitrap-mass spectrometry for the first time in this plant. Phytochemical studies indicated that TL contained 11 types of phytochemicals. The active components [total carbohydrate content (TCC), total protein content (TProC), total triterpenoid content (TTC), total phenolic content (TPheC), total flavonoid content (TFC), total phenolic acid content (TPAC), condensed tannin content (CTC), and gallotannin content (GC)] of eight different solvent extracts were determined by ultraviolet–visible spectrophotometry. Aqueous extract had highest TProC, TPheC, and GC values. Methanol extract exhibited highest TCC and TFC values. Ethanol extract showed highest TPAC and CTC values and dichloromethane extract exhibited highest TTC value. Methanol extract showed strongest ability to scavenge 2,2-diphenyl-1-picrylhydrazyl radicals, 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) diammonium salt cation radicals, and hydroxyl radicals, and also exhibited highest antioxidant activity in ferric-reducing antioxidant power and cupric ion-reducing antioxidant capacity assays. Best iron and copper chelating activity and H2O2 scavenging ability were shown by aqueous extract. Ethanol extract showed strongest ability to scavenge superoxide radicals and effectively prevent β-carotene bleaching. Acetone extract had highest inhibitory activity toward α-glucosidase, α-amylase, and xanthine oxidase. Ethyl ether extract had highest inhibitory activity towards urease and angiotensin converting enzyme. Aqueous and ethanol extracts had strongest inhibitory activity toward acetylcholinesterase. Methanol extract showed highest inhibitory activity toward tyrosinase. Methanol extract showed good stability and antioxidant capacity during heating, at different pH values, and after in vitro digestion and had low toxicity. The efficacy of methanol extract in stabilizing olive and sunflower oils was studied, the results suggested that methanol extract had a protective effect on the primary oxidation of the two oils. TL may be useful as a source of active components for application in human nutrition and/or phytomedicine and methanol extract of TL could be used as a natural oil stabiliser.

Impact of Bioactive Compounds of Plant Leaf Powders in White Chocolate Production: Changes in Antioxidant Properties during the Technological Processes.

Bioactive compounds present in the powdered leaves of matcha green tea (Camellia sinensis L.) (MGTP) and moringa (Moringa oleifera) (MOLP) seem to be related to health benefits due to their antioxidant properties. The growing accessibility of these powders has led to their being more widely used in food production. The aim of this study was to evaluate the total phenolic content (TPC) and antioxidant capacity (AC) of white chocolate (WCh) supplemented with MGTP and MOLP. AC was determined by 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), cupric ion-reducing antioxidant capacity (CUPRAC), and ferric-reducing antioxidant power (FRAP) assays, whereas TPC was determined by the Folin–Ciocalteu (FC) method. Both additives were incorporated at four levels (1, 2, 3 and 4%) in two chocolate processing steps (conching and tempering). Additionally, the amounts of phenolic acids, tocopherols, and carotenoids in WCh samples enriched by MGTP and MOLP were determined to explain their influence on AC. The results showed that the chocolates supplemented with MGTP were characterized by higher antioxidant properties than those with MOLP. In turn, MOLP significantly increased the content of lipophilic antioxidants in chocolates, tocopherols and carotenoids, which also exhibit pro-health effects. Furthermore, the incorporation of these additives during the tempering process was more relevant to the improvement of the antioxidant properties of WCh.
