Characterization of polyphenols and carbohydrates exuded by Phaeodactylum tricornutum diatom grown under Cu stress

This study is focused on analysing polyphenols and carbohydrates released by Phaeodactylum tricornutum (P. tricornutum) diatoms cultured in natural seawater enriched with sublethal and lethal Cu doses. Cu concentrations of 0.31, 0.79 and 1.57 µM reduced cell densities by 37, 82 and 91%, respectively, compared to the control. The total sum of all identified polyphenols and total carbohydrates released by cells grown under lethal Cu levels increased up to 18.8 and 107.4 times, respectively, compared to data from a control experiment. Four different in vitro assays were used to estimate the antioxidant activities of the extracellular compounds: 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical inhibition, cupric ion reducing antioxidant capacity (CUPRAC), ferric reducing antioxidant power and Cu complexing ability (CCA). The highest antioxidant activities were observed in the Cu lethal treatments, where the CCA assay exhibited a greater increase (up to 32.2 times higher than that found in the control experiment) to reduce the concentration of free Cu in the medium and its toxicity. The presence of Cu stimulated the release of polyphenols and carbohydrates to the medium as a detoxification mechanism to survive under lethal levels of Cu regulating its speciation.

Characterization and antioxidant activity of peel extracts from three varieties of citrus sinensis

High volume of postharvest materials including peels from citrus fruits is periodically generated, which contributes to environmental pollution. Investigating the chemical composition cum antioxidant property of these ‘wastes’ would be instructive in achieving value addition in the food and pharmaceutical value chain. On this premise, this study carried out phytochemical screening and antioxidant activity of three (3) commonly cultivated citrus varieties namely Citrus sinensis ‘valencia’, Citrus sinensis ‘washinton’ and Citrus sinensis ‘thompson navel’. The peels were extracted using ethanol and hexane in a Soxhlet extractor and thereafter subjected to phytochemical and Gas Chromatography/Mass Spectrometry (GC/MS) analyses, ferric ion reducing antioxidant power (FRAP), hydrogen peroxide scavenging and cupric ion reducing antioxidant capacity (CUPRAC) assays to evaluate their antioxidant potentials. Results show that Citrus sinensis peel extracts contain alkaloids, flavonoids, phenols, phytosterols, diterpenes, tannins and glycosides. GC/MS analysis identified about 48 compounds in each extract; with the predominant bioactive compounds being limonene (16.5%), ascorbic acid (17.7%), stearic acid (26.3%), linalool (4.7%), linoleic acid (16.18%), palmitic acid (15.23%), pentadecyclic acid (1.1%). Ethanol and hexane extracts of Valencia exhibited higher FRAP (9.09 ± 0.13) and CUPRAC (2.04 ± 0.06) values while the ethanol extract of Ibadan sweet demonstrated greater hydrogen peroxide scavenging activity (1.39 ± 0.00). Citrus peels are rich in bioactive compounds with excellent antioxidant activity and may serve as potential sources of natural antioxidants for food products or pharmaceutical formulations.

CYTOTOXICITY SCREENING AND ANTIOXIDANT CAPACITY ASSESSMENT OF THE INNER PERIANTH SEGMENTS OF 14 RUMEX SPECIES GROWN IN TÜRKİYE

Objective: Breast cancer is one of the most prevalent cancer types worldwide. Antioxidant sources may prevent the occurrence of cancer. Natural sources rich in phenolics, thus, may provide alternate agents in the management of breast cancer. Rumex species are widely distributed in Turkish flora. Emerging evidence has pointed out the antitumoral property of Rumex species on a variety of cancer cells. In the present study, we propose to test the ethanolic extracts of the inner perianth segments of 14 Rumex species on four breast cancer cells with different origins. We also demonstrated their toxicity on healthy cells. Material and Method: We performed the resazurin reduction assay to examine the cytotoxicity and toxicity. Furthermore, we determined the phenolic contents of the extracts as an indicator of their antioxidant profile and ascertained their antioxidant activities by DPPH radical, ABTS radical cation scavenging activity and cupric ion-reducing antioxidant capacity assays. Result and Discussion: The ethanolic extracts of the inner perianth segments of Rumex species exhibited remarkable cytotoxicity profiles neither on breast cancer cells nor on healthy H9c2 rat myoblastoma cells. However, they usually displayed strong antioxidant activities due to possessing high phenolic content.

Synthesis, antioxidant, DNA interaction, electrochemical, and spectroscopic properties of chromene-based Schiff bases: Experimental and theoretical approach

Two chromene-based Schiff base ligands were synthesized by the condensation of 5-hydroxy-2,2-dimethyl-2H-chromene-6-carbaldehyde with 2-aminodiphenylamine and 1,8-diaminonaphthalene: (E)-2,2-dimethyl-6-(((2-(phenylamino)phenyl)imino)methyl)-2H-chromen-5-ol (2) and (E)-6-(((8-aminonaphthalen-1-yl)imino)methyl)-2,2-dimethyl-2H-chromen-5-ol (3). The compounds were characterized by Fourier transform infrared (FT-IR), absorption (UV-Vis) and nuclear magnetic resonance (NMR) spectroscopies, in addition to mass spectrometry (MS) analyses; the results of which were rationalized using density functional theoretical (DFT) calculations. Antioxidant activity of the Schiff bases was determined using the DPPH free radical scavenging and CUPric Ion Reducing Antioxidant Capacity (CUPRAC) assays. Compound 3 (IC50 = 14 μM) showed greater free radical scavenging ability than compound 2 (35 μM), and was also more effective than ascorbic acid and Trolox. DFT studies indicate that the HAT mechanism is favored over SET for DPPH scavenging. Both compounds exhibited stronger Cu(II) ion reducing capacity than Trolox and ascorbic acid with Trolox equivalent activity coefficient (TEAC) values of 2.0 and 1.6 for compounds 2 and 3, respectively. Furthermore, DNA cleavage and binding properties were determined using gel electrophoresis, UV-Vis spectroscopy, and cyclic voltammetry. The compounds showed copper-mediated cleavage of plasmid DNA and strong binding to DNA, with binding constants on the order of 104 M−1. The strength of DNA binding was verified by molecular docking analysis using two DNA models: a B-DNA dodecamer (-8.4 to -9.3 kcal/mol) and a hexamer (-7.8 to -7.9 kcal/mol). Overall, the chromene Schiff bases show promise as antioxidant and DNA binding agents.

Phytochemical analyses by LC-HRMS, FTIR spectral analysis, antioxidant, antidiabetic and antityrosinase activity of Crataegus orientalis Pall. ex M. Bieb fruit extracted with various solvents.

Crataegus orientalis Pall. ex M. Bieb fruit (COPMB) is extensively used as a source of various products in the medicinal-aromatic field and holds the potential for erosion control, ornamental purposes, food source, and economic benefits for forest villagers from its fruits. This study aims to determine the chemical components and biological activities of extracts prepared from COPMB using different solvents. The present work was designed to define the antioxidant activity [phosphomolybdenum (total antioxidant capacity), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), ferric reducing antioxidant power (FRAP), cupric ion-reducing antioxidant capacity (CUPRAC) and metal chelating activity (MCA)], phytochemical screening analysis, enzyme inhibitor (α-amylase, α-glucosidase and tyrosinase) potential, and liquid chromatography-high resolution mass spectrometry (LC-HRMS) secondary metabolite profiling in different extracts of COPMB. The results of LC-HRMS revealed that fumaric acid was the main phenolic compound in all extracts. Among the extracts, ethyl acetate extract has the highest phytochemical and antioxidant properties [total phenolic content (TPC): 32.5 mg GAE/g, total flavonoid content (TFC): 12.2 mg QE/g, ABTS: 213.0 mg TE/g; CUPRAC: 126.0 mg TE/g, MCA: 145.0 mg EDTA/g; FRAP: 122.8 mg TE/g; TAC: 2.8 mmol TE/g]. Ethyl acetate and methanol extracts are more effective in α-amylase (0.27 ± 0.01 mg/mL; 0.12 ± 0.00 mg/mL), α-glucosidase (0.63 ± 0.02 mg/mL; 0.77 ± 0.02 mg/mL) and tyrosinase (0.03 ± 0.00 mg/mL; 0.03 ± 0.00 mg/mL) enzyme inhibition potentials compared to standard acarbose (0.75 ± 0.02 mg/mL for α-amylase; 1.11 ± 0.03 mg/mL for α-glucosidase) and kojic acid (0.04 ± 0.00 mg/mL). The findings from this study suggest that COPMB could serve as a valuable source of natural agents for the food and pharmaceutical industry. © 2024 Society of Chemical Industry.

In vitro research to evaluate the antioxidant effects, inhibiting enzymes, methicillin-resistant Staphylococcus aureus strains of Terminalia catappa extracts

AimContribute to the well-being of the Ivorian population through scientific proof of the use of Terminalia cattapa leaves extracts in traditional medicine. MethodologyThe antioxidative effect of aqueous, water-alcoholic, ethyl acetate and hexane derivatives of Terminalia catappa leaves was measured by six tests: DPPH (2-diphenyl-1-picrylhydrazyl), ABTS (2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)), FRAP (ferric ion reducing antioxidant power), PPBD (phosphomolybdenum), CUPRAC (cupric ion reducing antioxidant capacity) and MCA (metal chelating activity). The inhibition of the activity of five enzymes (acetylcholinesterase, butyrylcholinesterase, tyrosinase, amylase and glucosidase) was also determined. In addition, the broadcast method was used to evaluate impact of the samples on methicillin-resistant Staphylococcus aureus (MRSA) strains. ResultsHighest antioxidant capacity was obtained in the ethanolic sample for DPPH, ABTS, FRAP and PPBD tests. Ethyl acetate sample produced the best CUPRAC and MCA tests. Among inhibitory effect of samples on the five enzymes, aqueous sample showed the strongest anti-acetylCholynesterase and anti-butyrylCholynesterase activity. Best anti-tyrosinase and anti-amylase activity were obtained with the ethanolic sample. Hexane sample obtained the best anti-glucosidase activity while ethanolic and aqueous samples obtained none. All extracts showed no activity on the MRSA strains tested. ConclusionThe chemical compounds present in Terminalia cattapa leaves extracts would not be responsible for the use made in traditional medicine against infections.

Antioxidant Activities of Stem, Leaves and Fruits Extracts of Pepino (Solanum muricatum Aiton).

<b>Background and Objective:</b> Pepino (<i>Solanum muricatum</i> Aiton), rich with vitamin C and flavonoids, constitutes an abundant source of potent antioxidants. This research was conducted to determine antioxidant activity from three different parts of pepino based on equivalence with ascorbic acid, to analyze the relationship between total phenolic content (TPC) and total flavonoid content (TFC) on antioxidant activities and to determine flavonoid compounds. <b>Materials and Methods:</b> Antioxidant activities were determined using 2,2-Diphenyl-1-Picrylhydrazyl (DPPH) and Cupric Ion Reducing Antioxidant Capacity (CUPRAC) methods. The TPC and TFC were determined by UV-visible spectrophotometry. The correlation between TPC, TFC and antioxidant activity was analyzed using Pearson's method. Flavonoid compound content was performed by HPLC. <b>Results:</b> The ethyl acetate pepino fruit extract expressed the highest antioxidant activity by DPPH and CUPRAC assays. The highest TPC was obtained from the ethyl acetate extract of pepino stem (18.493 g GAE/(100 g)), while the highest TFC was obtained from the hexane extract of pepino leaves (9.541 g QE/(100 g)). <b>Conclusion:</b> The DPPH and CUPRAC assays demonstrated that pepino exhibits potential as a source of natural antioxidants, especially in its fruit part.

Cytotoxicity of Sambucus nigra L. on Cancer Cell Line and In Vitro Antioxidant Properties

Objective: Essential oils, free fatty acids, flavonoids, glycosides, phenolic acids, carotenoids, vitamins, and minerals are found in significant quantities in the characteristic chemical composition of Sambucus nigra L. This study aimed to evaluate the antioxidant potential of Sambucus nigra L. Fructus and evaluate the cytotoxicity on the cancer cell line.
 Methods: The Sambucus nigra L. fruits were collected from Yalova Atatürk Horticultural Central Research Institute in September 2021. The ethanol extract was prepared. Antioxidant property of Sambucus nigra L. fruit extracts was evaluated with 2,2-diphenyl-1-picryl-hydrazyl- hydrate (DPPH) radical scavenging activity, Cupric Ion Reducing Antioxidant Capacity (CUPRAC). Also, total phenolic content, total flavonoid content, and total anthocyanin content were calculated. Liver hepatocellular carcinoma cell line (HepG2) was used for cytotoxicity assay and an 2,5-diphenyl-2H-tetrazolium bromide (MTT) assay was applied.
 Results: The total phenolic, total flavonoid contents and total monomeric anthocyanins were 9.75±0.92 mg GAE/ mg fruit extract, 0.07437±0.004 mg quercetin/ mg fruit extract, respectively. 0.45±0.0014 mg catechin/gram of fruit extract and 2.08±0.025 mg Cyanidin-3 glucosides/g fruit extract. CUPRAC and DPPH results showed that Sambucus nigra L. extract has strong antioxidant activity. The results of the cytotoxicity assay indicated that while concentrations of the extract increased, the viability of HepG2 decreased.
 Conclusion: Our findings suggest that the Sambucus fruit extract is particularly rich in antioxidant components that are possibly modulating their beneficial use for hepatocellular malignancies, significantly reducing the number of viable cancer cells and inducing cell death.

Liposomes as a carrier for 4–hydroxycoumarin: characterisation, stability and antioxidant potential

<p>The aims of the present research were to develop and characterize 4-hydroxycoumarin-loaded liposomes <em>via </em>the determination of encapsulation efficiency, antioxidant capacity, vesicle size, polydispersity index (PDI), zeta potential, conductivity, mobility, density, surface tension, viscosity, and 60-day storage stability. The encapsulation efficiency of 4-hydroxycoumarin in liposomal particles was 96.7 ± 1.2%. The ABTS and DPPH radical scavenging capacity of the prepared liposomes was 89.76 ± 0.56% and 93.18 ± 0.23%, respectively, whereas cupric ion-reducing antioxidant capacity amounted to 0.367 ± 0.003 mmol Trolox equivalent (TE)/L. The density of liposomes with 4-hydroxycoumarin was 1.007 ± 0.002 g/cm<sup>3</sup>, surface tension was 22.7 ± 0.2 mN/m, and viscosity was 14.3 ± 0.2 mPa·s. Vesicle size and PDI of 4-hydroxycoumarin-loaded liposomes were changed from 1286.3 ± 73.8 nm to 2077.3 ± 63.2 nm and from 0.409 ± 0.050 to 0.676 ± 0.064, respectively, during the 60-day stability study. The zeta potential of the obtained liposomes was changed from -16.73 ± 0.47 mV to -10.31 ± 0.42 mV, while mobility varied from -1.311 ± 0.036 µmcm/Vs to -0.806 ± 0.031 µmcm/Vs. The conductivity did not change during 60 days and amounted to approximately 0.020 mS/cm. Overall, due to the high encapsulation efficiency and antioxidant capacity, the obtained results qualify liposomes to be used as 4-hydroxycoumarin carriers for future examination of its biological activities and potential application in medicine and pharmaceutical products. However, future experiments should include the optimization of the liposomal composition with the aim of improving 4-hydroxycoumarin-loaded liposome stability.</p>

Genistein Co-Amorphous Systems with Amino Acids: An Investigation into Enhanced Solubility and Biological Activity.

Genistein, an isoflavone known for its antioxidant and antidiabetic effects, suffers from the drawback of low solubility. To overcome this limitation, co-amorphous systems were synthesized by incorporating amino acids that were chosen through computational methods. The confirmation of the amorphous state of lysine and arginine-containing systems was ascertained by X-ray powder diffraction. Subsequently, the characterization of these systems was extended by employing thermo-gravimetry, differential scanning calorimetry, Fourier-transform infrared spectroscopy, and scanning electron microscopy. The investigation also included an assessment of the physical stability of the samples during storage. The apparent solubility of the systems was studied in an aqueous medium. To evaluate the in vitro permeability through the gastrointestinal tract, the parallel artificial membrane permeability assay was employed. The biological properties of the systems were assessed with regard to their antioxidant activity using 2,2-diphenyl-1-picrylhydrazyl and cupric ion-reducing antioxidant capacity assays, as well as their ability to inhibit α-glucosidase. The systems' glass transition temperatures were determined, and their homogeneity confirmed via differential scanning calorimetry analysis, while Fourier-transform infrared spectroscopy analysis provided data on molecular interactions. Stability was maintained for the entire 6-month storage duration. The co-amorphous system containing lysine displayed the most pronounced apparent solubility improvement, as well as a significant enhancement in antioxidant activity. Notably, both systems demonstrated superior α-glucosidase inhibition relative to acarbose, a standard drug for managing type 2 diabetes. The results indicate that co-amorphous systems with lysine and arginine have the potential to significantly enhance the solubility and biological activity of genistein.

Catechin Composition, Phenolic Content, and Antioxidant Properties of Commercially-Available Bagged, Gunpowder, and Matcha Green Teas

The leaves of the Camellia sinensis plant are used to produce many different types of teas, and green tea is particularly noted for its health promoting properties which are attributed to high concentrations of phenolic compounds known as catechins. Green tea is available in forms such as bagged, gunpowder (rolled leaf), and matcha, a fine powder. This study evaluated the phenolic content, catechin composition, caffeine concentration, and antioxidant properties of fifteen commercially-available green teas with an emphasis on identifying differences between ceremonial and culinary matcha. Gunpowder and bagged green teas had higher total phenolic contents and greater CUPRAC (cupric ion reducing antioxidant capacity) and ORAC (oxygen radical absorbance capacity) antioxidant capacities than matcha teas. Among matcha samples, less expensive culinary teas had greater total phenolic contents and higher antioxidant capacities than ceremonial products. Hierarchical cluster analysis was used to determine similarities and differences in catechin composition and revealed that bagged and gunpowder green teas had the highest levels of the stereoisomers epi/catechin gallate, which correlated with their strong antioxidant properties. Bagged and gunpowder teas were the lowest cost products in this study, indicating that they are an economical choice for consumers who want a green tea beverage with high catechin content, although they generally had lower caffeine levels than matcha.

Comparative Study on the Chemical Profile, Antioxidant Activity, and Enzyme Inhibition Capacity of Red and White Hibiscus sabdariffa Variety Calyces.

Hibiscus sabdariffa L. (Family: Malvaceae) is believed to be domesticated by the people of western Sudan sometime before 4000 BC for their nutritional and medicinal properties. This study aimed to investigate the chemical profile, antioxidant activity, and enzyme inhibition property of extracts from red roselle (RR) and white roselle (WR) varieties grown in Sudan. Three aqueous extracts obtained by maceration, infusion, and decoction, in addition to the methanolic one, were prepared from the two roselle varieties. Results showed that the highest total phenolic and flavonoid contents of RR were obtained from the extracts prepared by infusion (28.40 mg GAE/g) and decoction (7.94 mg RE/g) respectively, while those from the WR were recorded from the methanolic extract (49.59 mg GAE/g and 5.81 mg RE/g respectively). Extracts of RR were mainly characterized by high accumulation of chlorogenic acid (6502.34-9634.96 mg kg-1), neochlorogenic acid (937.57-8949.61 mg kg-1), and gallic acid (190-4573.55 mg kg-1). On the other hand, neochlorogenic acid (1777.05-6946.39 mg kg-1) and rutin (439.29-2806.01 mg kg-1) were the dominant compounds in WR. All extracts from RR had significant (p < 0.05) higher antioxidant activity than their respective WR except in their metal chelating power, where the methanolic extract of the latter showed the highest activity (3.87 mg EDTAE/g). RR extracts prepared by infusion recorded the highest antioxidant values (35.09, 52.17, 65.62, and 44.92 mg TE/g) in the DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid), CUPRAC (cupric ion reducing antioxidant capacity), and FRAP (ferric reducing antioxidant power) assays, respectively. All aqueous extracts from the WR exerted significant (p < 0.05) acetylcholinesterase (AChE) inhibitory activity (3.42-4.77 mg GALAE/g; GALAE = galantamine equivalents), while only one extract, obtained by maceration, from RR exerted AChE inhibitory activity (4.79 mg GALAE/g). All extracts of the RR showed relatively higher BChE (butyrylcholinesterase) inhibitory activity (3.71-4.23 mg GALAE/g) than the WR ones. Methanolic extracts of the two roselle varieties displayed the highest Tyr (tyrosinase) inhibitory activity (RR = 48.25 mg KAE/g; WR = 42.71 mg KAE/g). The methanolic extract of RR exhibited the highest amylase (0.59 mmol ACAE/g) and glucosidase (1.46 mmol ACAE/g) inhibitory activity. Molecular docking analysis showed that delphinidin 3,5-diglucoside, rutin, isoquercitrin, hyperoside, and chlorogenic acid exerted the most promising enzyme inhibitory effect. In conclusion, these findings indicated that the chemical profiles and biological activity of roselle varied according to the variety, extraction solvent, and technique used. These two roselle varieties can serve as a valuable source for the development of multiple formulations in food, pharmaceutical, and cosmetic industries.

Determination of the antioxidant and enzyme-inhibiting activities and evaluation of selected terpenes' ADMET properties: in vitro and in silico approaches

The in vitro antioxidant capacity of eugenol, eucalyptol, linalool, and carvacrol was performed using six test systems: phosphomolybdenum, DPPH (2,2′-diphenyl-1-picrylhydrazyl radical), ABTS (2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonate), CUPRAC (cupric ion reducing antioxidant capacity), FRAP (Ferric reducing ability of plasma), and metal chelating effect. Their inhibitory activities on cholinesterases, α-amylase, α-glucosidase, and tyrosinase were assessed as well. Molecular docking helped to explain the obtained outcomes while in silico ADMET (absorption, distribution, metabolism, excretion, and toxicity) prediction served for assessing the physicochemical and pharmacokinetic profiles. Our findings showed that the examined terpenes have promising antioxidative activity with eugenol and carvacrol displaying the highest efficiency. Linalool presented the highest anti-AChE and anti-tyrosinase activities (7.931 ± 0.034 mg GALAE/g and 58.707 ± 5.120 mg KAE/g, respectively). Eucalyptol exhibited the strongest anti-BChE activity (2.704 ± 0.025 mg GALAE/g), while carvacrol exhibited the highest rates for α-glucosidase and α-amylase inhibition (1.414 ± 0.042 and 1.023 ± 0.054 mM ACAE/g, respectively). Molecular docking revealed that all compounds interacted in varied ways with the five enzymes’ active sites. Excellent binding profiles and greatest inhibitory activity to AChE (−7.4 kcal/mol), α-glucosidase (−6.5 kcal/mol), and α-amylase (−6.5 kcal/mol) was attributed to carvacrol. As regards to tyrosinase, eugenol (−6.2 kcal/mol) was the potent one whereas carvacrol and eugenol (−6.5 kcal/mol for both) strongly interacted with BChE. Furthermore, in silico ADMET-related physicochemical and pharmacokinetic properties revealed good profiles. To sum up, here we demonstrate the powerful antioxidant potency and enzymatic inhibitory effects of the four terpenes which may enable the discovery of lead compounds that are extremely potent antioxidants and enzyme inhibitors.

Neuroprotective and Antioxidant Activities of Algerian Punica granatum L. Peel Extracts

Background: Oxidative stress is implicated in many neurodegenerative disorders, such as Alzheimer’s disease. As an alternative, Punica granatum peels are widely used in the treatment of several ailments. To date, this is the first study on the anti-acetyl/butyrylcholinesterase effects of diethyl ether extract from Algerian pomegranate peels. Objective: This research aimed to evaluate the content of total phenolics, flavonoids, flavonols, and tannins in Punica granatum peel crude extract and its fraction diethyl ether, as well as to assess its antioxidant and neuroprotective activities. Methods: The total phenolic, flavonoid, flavonol, and tannin contents in the extracts were evaluated using spectrophotometric methods. The inhibitory effects on acetyl/butyrylcholinesterase were assessed using the in vitro models. In addition, several methods were used to investigate the antioxidant capacity of extracts, including 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline- 6-sulfonate) (ABTS) free radicals scavenging, phenanthroline, reducing power and cupric ion reducing antioxidant capacity (CUPRAC) assays. Results: Phytochemical analysis revealed that the diethyl ether extract recorded the highest content of total phenolics, flavonoids, flavonols, and tannins with values of 643.3 ± 9.96 μgGAE/mg, 78.97 ± 9.97 μgQE/mg, 31.32 ± 1.79 μgQE/mg and 96.6 ± 13.65 μgTAE/mg, respectively. Moreover, the diethyl ether extract exhibited the strongest activity in the DPPH scavenging assay (IC50=3.36 ± 0.11 μg/mL), ABTS scavenging assay (IC50=1.46 ± 0.10 μg/mL), CUPRAC assay (CA0.5=3.62 ± 0.36 μg/mL), phenanthroline assay (CA0.5=1.04 ± 0.10 μg/mL), reducing power assay (CA0.5=4.14 ± 0.32 μg/mL), antiacetylcholinesterase assay (IC50=114.86 ± 13.66μg/mL), and anti-butyrylcholinesterase assay (IC50=147.00 ± 10.41 μg/mL). Conclusion: The present study has shown the extracts from Punica granatum peel to be a rich source of bioactive compounds, potent antioxidants, and cholinesterase inhibitors. They could be considered as natural agents that can be used in anti-neurodegenerative pharmaceutical formulations.

Optimization of pressurized ethanol extraction for efficient recovery of hyperoside and other valuable polar antioxidant-rich extracts from Betula pendula Roth leaves

Silver birch (Betula pendula Roth) leaves, known for their long-standing uses in traditional medicine, are bioactive compound-rich by-products of the birch logging industry, abundantly available throughout the northern hemisphere. Considering the current demands for the sustainable valorization of various plant materials and their processing by-products, this study aimed to optimize pressurized ethanol extraction (PLE-EtOH) parameters in order to produce hyperoside and other valuable polar antioxidant-rich extracts from silver birch leaves. A central composite design along with response surface methodology (CCD-RSM) was used to optimize the temperature and time of PLE-EtOH for yield, total phenolic (TPC) and flavonoid (TFC) content, Trolox equivalent antioxidant capacity (TEAC) measured in oxygen radical absorbance capacity (ORAC), cupric ion reducing antioxidant capacity (CUPRAC), and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) assays, and hyperoside content measured by ultraperformance liquid chromatography-electrospray tandem mass spectrometry (UPLC-ESI/MS2) analysis. The optimal PLE-EtOH conditions were 86 °C and 39 min, which produced an extract with 32.2% yield, high TPC (97.3 mg gallic acid equivalents/g), TFC (37.3 mg quercetin equivalents/g) and proanthocyanidin (301.2 mg catechin hydrate equivalents/g) content, also strong in vitro antioxidant capacity in ORAC, CUPRAC and ABTS assays (999.3, 297.0 and 392.3 mg Trolox equivalents/g, respectively). Hyperoside (54.1 mg/g), betuloside (18.3 mg/g) and chlorogenic acid (11.5 mg/g) were the prime phenolic compounds in the PLE-EtOH extract. Additionally, a high sun protection factor (37.4 at 1 mg/mL) was determined by in vitro method for this extract. Such products from B. pendula leaves have potential cosmetical, nutraceutical, and pharmaceutical applications.

The performance of hypochlorous acid modified Ag nanoparticle-based assay in the determination of total antioxidant capacity of Boswellia Serrata and Aronia

In this study, the SNPAC method was developed by using hypochlorous (ClO−) at pH 9.3. Polyvinylpyrrolidone (PVP) was used as a stabilizer. The existence of HOCl enables to formation stronger and more stable yellow emission which gives sensitivity. As a result, lower LOD and LOQ values were achieved by the AgNP–HOCl method. The obtaining lower values was attributed to the reoxidation of silver atoms with ClO− ions in the presence of PVP. The newly modified method was applied to the total antioxidant capacity (TAC) measurement of Boswellia Serrata, Aronia Melanocarpa tea, and juice. The results were compared with the results obtained by application of CUPRAC (Cupric ion Reducing Antioxidant Capacity) and FRAP (Ferric Reducing Antioxidant Power) assays. The TAC values of Boswellia Serrata, Aronia Melanocarpa tea, and juice were found as 6.18, 137.98, and 12.08 mmol TEAC/kg, respectively.

In-vitro anti-diabetic, anti-Alzheimer, anti-tyrosinase, antioxidant activities of selected coumarin and dihydroisocoumarin derivatives

Benzo-α-pyrone structured coumarin derivatives are secondary metabolites first obtained from Coumarouna odorata in 1822. Coumarin and its structural isomer dihydroisocoumarin derivatives are found in many different sources in nature. Several different bioactivities of these compounds have been reported. In this study, preliminary activity screening and comparison of four purchased coumarin derivatives (esculetin, esculin monohydrate, umbelliferon, scoparone) and four previously isolated 3-phenyl-3,4-dihydroisocoumarin derivatives (thunberginol C, scorzocreticoside I, scorzocreticoside II, scorzopygmaecoside) from a medicinal plant were carried out by in-vitro methods. α-Glucosidase, acetylcholinesterase, butyrylcholinesterase, tyrosinase inhibitor activities and antioxidant potentials of the compounds were evaluated. Consequently, thunberginol C (free – not glycosylated form of 3,4-dihydroisocoumarin structure) showed better potential in all enzyme inhibitory activities compared to coumarin structure. Particularly, α-glucosidase inhibitory activity of this compound with a very low IC50 value (94.76±2.98 µM) compared to standard acarbose (1036.2±2.70 µM) should be noted. Glycosylation and/or methoxy substitution of 3,4-dihydroisocoumarin structure resulted a significant decrease in all tested enzyme inhibitory activities. The structures of esculin MH, umbelliferone, scoparone, scorzocreticoside I, and scorzopygmaeceoside might be considered in further synthetic studies as selective acetylcholinesterase inhibitors. Thunberginol C has a promising potential in tyrosinase inhibitory activity. Esculetin and thunberginol C showed the best results with high potentials in antioxidant activity via 2,2-diphenyl-1-picryl-hydrazyl-hydrate free radical scavenging, 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid cation radical decolorization, and cupric ion reducing antioxidant capacity assays compared to the standards.

Molecular features, antioxidant potential, and immunological expression assessment of thioredoxin-like protein 1 (TXNL1) in yellowtail clownfish (Amphiprion clarkii)

Thioredoxin-like protein 1 (TXNL1) is a redox-active protein belonging to the thioredoxin family, which mainly controls the redox status of cells. The TXNL1 gene from Amphiprion clarkii (AcTXNL1) was obtained from a pre-established transcriptome database. The AcTXNL1 is encoded with 289 amino acids and is predominantly localized in the cytoplasm and nucleus. The TXN domain of AcTXNL1 comprises a34CGPC37 motif with redox-reactive thiol (SH-) groups. The spatial distribution pattern of AcTXNL1 mRNA was examined in different tissues, and the muscle was identified as the highest expressed tissue. AcTXNL1 mRNA levels in the blood and gills were significantly increased in response to different immunostimulants. In vitro antioxidant capacity of the recombinant AcTXNL1 protein (rACTXNL1) was evaluated using the ABTS free radical-scavenging activity assay, cupric ion reducing antioxidant capacity assay, turbidimetric disulfide reduction assay, and DNA nicking protection assay. The potent antioxidant activity of rAcTXNL1 exhibited a concentration-dependent manner in all assays. Furthermore, in the cellular environment, overexpression of AcTXNL1 increased cell viability under H2O2 stress and reduced nitric oxide (NO) production induced by lipopolysaccharides (LPS). Collectively, the experimental results revealed that AcTXNL1 is an antioxidant and immunologically important gene in A. clarkii.

Modeling of Sensory Properties of Poppy Sherbet by Turkish Consumers and Changes in Quality Properties during Storage Process.

Poppy is an important edible plant containing bioactive components. This study aimed to produce good-tasting poppy sherbet by determining the content using a response surface methodology (RSM). At the same time, bioactive components, phenolic compounds, and color properties were investigated in optimum poppy sherbet during storage; 0.26 g of dried corn poppy flowers, 0.15 g of citric acid, and 4.29 g of sucrose values were the most promising, achieving high scores for color, smell, taste, and general acceptance from sensory properties (sensory score of 8.55 for color; 7.19 for smell; 8.38 for taste; 7.98 for general acceptability). A total of nine polyphenols were detected in the optimum poppy sherbet sample; gallic acid was the most common. There was no statistically significant difference between the samples stored on the 0th and 30th days regarding gallic acid content (23.886 ± 0.164 μg/mL, 23.403 ± 0.343 μg/mL) and protocatechuic acid (1.146 ± 0.048 μg/mL, 1.047 ± 0.038 μg/mL). Total flavonoid contents (TFC), total phenolic contents (TPC), CUPRAC (cupric ion reducing antioxidant capacity), DPPH (e free radical diphenylpicrylhydrazyl), total monomeric anthocyanin (TAC), and color values were found to decrease as the storage period increased. It was considered that a highly palatable and rich bioactive component product could be obtained.

Chemical Composition and In Vitro Antimicrobial, Antioxidant, and Tyrosinase-Inhibiting Activity of Essential Oils Extracted From the Leaves and Twigs of Calocedrus macrolepis var. formosana

In this study, we focused on evaluating the antimicrobial, antioxidant, and tyrosinase-inhibiting properties of leaf and twig essential oils from Calocedrus macrolepis var. formosana. Additionally, we determined the major components associated with these effects. The findings revealed that α-pinene (36.4%) and limonene (23.6%) were the dominant components in the leaf essential oil, while α-pinene (36.2%), ferruginol (22.5%), and limonene (13.1%) were the major constituents in the twig essential oil. However, when comparing the biological activities of the 2 essential oils, the twig essential oil demonstrated excellent inhibitory effects against skin pathogens, as well as strong antioxidant and tyrosinase-inhibiting activities, in contrast to the leaf essential oil. The twig essential oil also exhibited excellent antimicrobial activity against 5 skin pathogenic strains, with the strongest inhibitory effect being that against Staphylococcus aureus, S epidermidis, and Candida albicans, which was accompanied by an inhibition zone of 46 to 48 mm, a minimum inhibitory concentration (MIC) of 125 μg/mL, and a minimum bactericidal concentration (MBC) of 250 μg/mL. The inhibitory activity of the twig essential oil against lipid peroxidation was found to be potent, as evidenced by an IC50 value of 216 ± 1.9 μg/mL. It also exhibited strong scavenging activity against free radicals, including 2,2-diphenyl-1-picrylhydrazyl (DPPH) (IC50 = 121.6 ± 0.5 μg/mL) and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) (IC50 = 136.8 ± 1.6 μg/mL) cation radicals. In addition, it exhibited a high cupric ion reducing–antioxidant capacity (CUPRAC), with the A0.50 value determined to be 186.28 ± 0.5 μg/mL. It also exhibited adequate activity in terms of inhibiting in vitro tyrosinase activity, as evidenced by an IC50 value of 168.5 ± 0.6 μg/mL. Further analysis revealed that ferruginol was the active component responsible for the antimicrobial, antioxidative, and tyrosinase-inhibiting activities observed in twig oil. These findings suggest that the twig oil is a promising candidate for further research because of its favorable antimicrobial, antioxidative, and tyrosinase-inhibiting activity.
