Next-generation 3D printed multipurpose prevention intravaginal ring for prevention of HIV, HSV-2, and unintended pregnancy

Globally, nearly half of all pregnancies are unintended, ∼1.3 million new human immunodeficiency virus (HIV) infections are reported every year, and more than 500 million people are estimated to have a genital herpes simplex virus (HSV-2) infection. Here we report the first 3D printed multipurpose prevention technology (MPT) intravaginal ring (IVR) for prevention of HIV, HSV-2, and unintended pregnancy. The IVRs were fabricated using state-of-the-art Continuous Liquid Interface Production (CLIP™) 3D printing technology using a biocompatible silicone-urethane based resin. Anti-HIV drug (Dapivirine, DPV), anti-herpes drug (Pritelivir, PTV) and a contraceptive drug (Levonorgestrel, LNG) were loaded in a macaque size IVR (25 mm outer diameter, OD; 6.0 mm cross-section, CS) allometrically scaled from the human size (54 mm OD; 7.6 mm CS) IVR analogue. All three active pharmaceutical ingredients (APIs) were loaded in the IVR using a single-step drug loading process driven by absorption. DPV, PTV, and LNG elicited zero-order release kinetics in vitro in simulated vaginal fluid (SVF) at pH 4 and pH 8 relevant to human and macaque vaginal pH respectively. CLIP 3D printed MPT IVRs remained stable after 6 months of storage at 4 °C with no change in physical, dimensional, or mechanical properties and no change in drug concentration and absence of drug degradation byproducts. The MPT IVRs elicited sustained release of all three APIs in macaques for 28 days with median plasma concentrations of 138 pg/mL (DPV), 18,700 pg/mL (PTV), and 335 pg/mL (LNG). Safety studies demonstrated that the MPT IVRs were safe and well tolerated in the macaques with no observed change or abnormalities in vaginal pH and no significant changes in any of the 22 mucosal cytokines and chemokines tested including pro-inflammatory (IL-1β, IL-6, IL-8, IFN-γ, TNF-α, IL-17, IL-18) and anti-inflammatory (IL-10, IL-12) cytokines while the MPT IVR was in place or after its removal. Additionally, MPT IVRs elicited no observed alterations in systemic CD4+ and CD8+ T cells during the entire study. Collectively, the proposed MPT IVR has potential to expand preventative choices for young women and girls against unintended pregnancy and two highly prevalent sexually transmitted infections (STIs).

Evaluation of the Safe Water Optimization Tool to Provide Evidence-Based Chlorination Targets in Surface Waters: Lessons from a Refugee Setting in Uganda.

The Safe Water Optimization Tool (SWOT) generates evidence-based point-of-distribution free residual chlorine (FRC) targets to adjust chlorine dosing by operators and ensure water quality at point-of-consumption. To investigate SWOT effectiveness in surface waters, we conducted two before-and-after mixed-method evaluations in a Uganda refugee settlement served by piped and trucked surface water systems. We surveyed 888 users on water knowledge, attitudes, and practices; collected 2768 water samples to evaluate FRC,Escherichia coli, and disinfection by-products (DBPs) concentrations; and conducted nine key-informant interviews with system operators about SWOT implementation. After baseline data collection, SWOT chlorination targets were generated, increasing point-of-distribution FRC targets from 0.2 to 0.7-0.8 mg/L and from 0.3 to 0.9 mg/L for piped and trucked systems, respectively. At endline, household point-of-consumption FRC ≥ 0.2 mg/L increased from 23 to 35% and from 8 to 42% in the two systems. With these increases, we did not observe increased chlorinated water rejection or DBPs concentrations exceeding international guidelines. Informants reported that SWOT implementation increased knowledge and capacity and improved operations. Overall, SWOT-generated chlorination targets increased chlorine dosage, which improved household water quality in surface waters although less than previously documented with groundwater sources. Additional operator support on prechlorination water treatment processes is needed to ensure maximally effective SWOT implementation for surface water sources.

Novel catalyst-free activation of chlorine by visible light for micropollutant abatement

This study systematically investigated the direct activation of chlorine by visible light emitting diode (Vis-LED). Vis-LED could effectively activate chlorine to degrade micropollutants with degradation efficiency and pseudo-first-order degradation rate constant range of 64.3–100 % and 0.0340–0.195 min−1, respectively. Quenching experiments and modeling results suggested that reactive chlorine species (RCS, including ClO•, Cl2•-, and Cl•) and hydroxyl radical (•OH) were involved in the degradation of atenolol (ATL). The contribution ratio of ClO•, free available chlorine, Cl•, Cl2•-, and •OH to ATL degradation were 58.7 %, 17.4 %, 15.6 %, 1.8 %, and 5.9 %, respectively, in Vis-LED448/chlorine process. Moreover, the innate quantum yields of HClO and ClO- decreased from 0.229 and 0.0206 to 0.0489 and 0.0109 mol·Einstein−1, respectively, as the wavelength increased from 448 to 513 nm, leading to a decrease in ATL degradation, which was consistent with the model results. Experimental and modeling results have confirmed that ATL degradation decreased when pH increased from 4.0 to 9.0. Cl- had little effect on the degradation of ATL, while HA and HCO3- affected ATL degradation by scavenging reactive species and/or shielding effect. The concentration of disinfection by-products decreased with the increase of wavelength and pH. In summary, Vis-LED/chlorine is an efficient water treatment process even without a catalyst.

Cotton Gin By-Products Utilization: Past, Present, and Future

The regional concentration of cotton gin by-products (CGB) has increased significantly over the past three decades because of the consolidation of smaller gins into larger gins. Although several studies have investigated the potential of various waste treatment/valorization technologies to improve the management of CGB, most of the technologies' economic feasibility remains a challenge. Therefore, there is a need to review the existing and emerging technologies vis-à-vis process economics and changes in the cotton ginning industry. This study reviews the published status of these existing technologies (in terms of the challenges and potentials), analyzes the prospects of some emerging waste treatment technologies for CGB, and discusses future economic and environmental sustainability directions. Where appropriate, new information from studies conducted in the U.S. and Australia is provided to support existing published data. The results show that the main limiting factors include production costs (e.g., energy and feedstock/additives costs) and logistics/supply. Additionally, the results support a solid potential for the sustainable use of CGB for various applications. However, the potential can be achieved by optimizing specific treatment processes, combining multiple processes through hybrid or integrated systems, co-treating CGB with other affordable and readily available materials (such as by-products from other agricultural operations), and improving return on investment for products developed.

Revealing the impact of sample enrichment method on concentration and cytotoxicity of volatile disinfection byproducts in drinking water: A quantitative study for liquid-liquid extraction

Liquid-liquid extraction (LLE) combined with the N2 blow-down method is a promising tool for bioanalysis of drinking water. However, detailed information on which disinfection byproduct (DBP) classes are retained in LLE extracts is currently unavailable. In this study, the recovery of seven classes of volatile DBPs and total adsorbable organic halogens (TOX) during the LLE method, combined with three common N2 blow-down methods, for bioanalysis in real tap water was analyzed at a 2-L scale, along with their corresponding cytotoxicity. The total concentration of seven classes of volatile DBPs in drinking water in Suzhou ranged from 64.6 to 83.0 µg/L, with the majority contributed by trihalomethanes (THMs: 59.9 µg/L), haloaldehydes (HALs: 5.4 µg/L), haloacetamides (HAMs: 3.4 µg/L), and haloacetonitriles (HANs: 3.2 µg/L). During the LLE - N2 blow-down process for bioanalysis, about 69–85 % of targeted volatile DBPs and 64–75 % of TOX were lost, respectively. Seven classes of volatile DBPs accounted for 52.8–64.3 % and 23.8–61.3 % of TOX in tap water and LLE - N2 blow-down samples, respectively, suggesting that targeted aliphatic DBPs are the key contributors to TOX. Furthermore, although LLE - solvent exchange had a better recovery performance than other N2 blow-down methods, the recoveries of volatile DBPs using this method were still not ideal. For example, HALs and HAMs had a slightly better recovery (>50 %), while most volatile DBPs had a poor recovery, including iodo-trihalomethanes (I-THMs, 0 %), haloketones (28 %), THMs (26 %), halonitromethanes (33 %), and HANs (38 %). During LLE - solvent exchange, 31 % and 36 % of targeted DBPs and TOX, respectively, in real tap water can be retained, which shows better performance than non-ionic macroporous copolymers (XAD). More importantly, the water volume required in this method for cytotoxicity analysis is 2 L, which greatly reduces the burden of water sample collection, transport, and pre-treatment compared to XAD (which typically requires 5 or 10 L). In general, this paper reveals the fate of volatile DBPs during LLE - N2 blow-down and indicates that LLE - solvent exchange is a good substitute for the XAD method in bioanalysis.

Removal of livestock odor gas ammonia, hydrogen sulfide, methanethiol by electron beam in a continuous flow system

Odor is becoming one of the serious environmental issues as the industry develops. Electron beam technology has recently attracted attention as an AOP (Advanced Oxidation Process) for air purification, water treatment, and soil remediation. This work studied the removal of major livestock odor such as ammonia (NH3), hydrogen sulfide (H2S), and methanethiol (CH3SH) using a continuous gas flow electron beam treatment process. The effects of initial odor gas concentration, type of background gas, absorbed dose of the electron beam, voltage of the electron accelerator, and odor gas flow rate on the removal of odor gas were all studied in this continuous gas flow treatment system. As the absorbed dose of electron beam was increased, the removal efficiencies of odor gases NH3, H2S, and CH3SH all increased gradually. As the initial concentration of odor gas was decreased and as the flow rate of odor gas was increased, the removal efficiencies also increased. Regarding background gas, the odor gas removal efficiency was found to be in descending order of O2 > air > N2. The lower the voltage (MeV) of the electron beam, the higher the odor gas removal efficiency. This was attributed to the fact that, to set the same absorbed dose (kGy), as the voltage decreases, the current value (mA) increases. For the byproducts, as the absorbed dose of electron beam was increased, the concentration of byproducts increased. Regarding the gas-phase continuous electron beam treatment reactions, the optimal operating conditions must be derived by considering various operational variables, including the initial concentration, electron beam dose, background gas type, gas flow rate, and electron beam voltage. The results of this study show that, among several possibly relevant variables in removing odor gases in the continuous flow electron beam irradiation process, the current value can be considered one of the most important variables, along with the initial concentration and background gas.

A model of the spatiotemporal distribution of ozone-squalene reaction and ozonolysis by-products from human body

Emissions of ozone and its by-products from ozonolysis on human surfaces lead to indoor air pollution. However, the spatiotemporal distribution of such emissions in indoor environments remains unclear, which may introduce bias when assessing human exposure to ozone and ozonolysis byproducts. This study developed a computational fluid dynamics model to describe the physical and chemical processes involved in the emission of ozone-dependent volatile organic compounds from the human body. The results showed that the reaction probability of ozone in the human body depends on the ozone concentration in the bulk air. For ozone concentrations ranging from 28 ppb to 200 ppb, the reaction probabilities ranged from 5.9 × 10−5 to 1.5 × 10−4. The concentrations of ozone and ozonolysis byproducts obtained from the experimental measurements were used for model validation. The ozonolysis by-products were found to be uniformly distributed in the chamber, whereas the ozone distribution showed less uniformity. The ozone concentration near the human surface was approximately 30 %∼50 % of that in the ambient air. Overall, a model was developed to understand the effect of ozone-surface interactions on indoor air quality. This model can be applied to analyze human exposure to ozone and ozonolysis byproducts and for health risk assessment in built environments.

Disinfection Byproducts in Drinking Water from the Tap: Variability in Household Calculated Additive Toxicity (CAT).

Recent studies have implemented a calculated additive toxicity (CAT) approach that sums measured disinfection byproduct (DBP) concentrations weighted by their respective in vitro bioassay potencies to estimate their associated risk in disinfected waters. In this study, the CAT approach was used to systematically investigate 21 regulated and unregulated DBPs measured in drinking water at the household level. Water samples from the tap were collected from over 120 randomly selected participants supplied by eight public water systems using four distinct source water types, two types of disinfection processes, and across two seasons. The purpose of this study was to compare CAT using multiple biological end points, examine household variability, identify DBPs driving toxicity, and assess if current regulated DBPs are adequate predictors of unregulated DBPs. Our results support the significance of unregulated DBPs, particularly haloacetonitriles and iodoacetic acid, as drivers of toxicity. Simple linear models between regulated versus unregulated concentrations and CAT were overall weak with 67% considered poor (r 2 < 0.3). These results reveal that current regulatory monitoring approaches may not be adequately capturing true household exposure due to higher contribution of unregulated DBPs to CAT and poor predictability between regulated and unregulated DBP-mediated CAT.

Availability, distribution and quality of agro-industrial byproducts and compound feeds in Ethiopia

Livestock production is a major contributor to the national economy in Ethiopia, of which ruminants, particularly cattle, are the dominant and most important livestock species. Limited supply of quality feeds is a major constraint to the improvement of livestock production in Ethiopia. The expansion of agro-industries in Ethiopia, over the last three decades, has created an opportunity for an increased supply of agro-industrial byproducts as a source of feed for livestock. This study was conducted to assess the availability, distribution, and quality of agro-industrial byproducts and compound feeds in Ethiopia. About 310 flour mill factories, 194 oil factories, 13 brewery factories, 7 sugar factories, 4 malt factories, 2 meat and bone meal processing plants, 8 limestone factories, and 112 feed processing plants were surveyed to assess the availability and distributions of agro-industrial byproducts and compound feeds. A total of 757 feed samples were collected for evaluating nutritional values. The annual production of agro-industrial byproducts ranges from 18,065 tons DM/year (abattoir byproducts) to 3,092,035 tons DM/year (malt byproducts) and the total annual production of the different agro-industrial by-products amounts to 5,245,854 tons DM. The annual production of compound feeds was reported to be 5,812,608 tons DM. All agro-industries are processing under their capacity, ranging from 11.7% in sugar factories to 93% in breweries. Shortage and seasonal fluctuation of supply of raw materials (34.8%), high price and price fluctuations of raw materials (17.5%) and electric power interruptions (17.5%) were the major challenges faced by different agro-industries. The crude protein (CP) concentrations of agro-industrial byproducts ranges from 6.2% in maize grain screening to 15.9% in wheat bran for flour mill byproducts; 28% in cotton seed cake to 49.7% in groundnut cake for oilseed cakes and 14.8% in areqe atela to 24.8% in brewery spent grains for brewery and local distillery byproducts. High protein or energy contents of agro-industrial byproducts indicated their potential to be utilized in intensive livestock rations. Thus, it is necessary to create enabling conditions to allow the existing agro-industries to operate at full capacity and to attract new ones into the business to boost the production and availability of agro-industrial byproducts needed as main inputs to manufacture compound feeds.

Evaluation of distribution system water quality during a free chlorine conversion.

Chloraminated drinking water systems commonly use free chlorine conversions (FCCs) to prevent or control nitrification, but unintended water quality changes may occur, including increased disinfection by-product and metal concentrations. This study evaluated water quality in a chloraminated drinking water system and residential locations before, during, and after their annual, planned FCC. Water quality alternated between relatively consistent and variable periods when switching disinfectants. During the FCC, regulated four trihalomethane and five haloacetic acid concentrations increased by four and seven times, respectively, and exceeded corresponding maximum contaminant levels. Implications of disinfection by-product sampling during an FCC were assessed, and an approach to account for increased FCC disinfection by-product concentrations was proposed. For metals, the FCC had minor impacts on distribution system concentrations and did not appear to impact residential concentrations. Overall, observed variable water quality appeared primarily associated with switching disinfectants and depended on distribution system hydraulics.

Optimizing dilute acid pretreatment for enhanced recovery and co-fermentation of hexose and pentose sugars for ethanol and butanol production

A systematic study of dilute acid pretreatment of sugarcane bagasse was performed to optimize the recovery of hexose-rich solid and pentose-rich liquid while minimizing fermentation inhibitors. Pretreatment experiments evaluated the effects of pretreatment time (10–90 min) and acid concentration (0.5–3.0 % w/w). The optimized pretreatment (1.4 % w/w sulfuric acid, 56.4 min at 121 °C) led to a solid fraction with 60.4 % cellulose and a liquid stream rich in pentose, in which 92.7 % of sugars was recovered as xylose (17.8 g/L). Besides that, low concentrations of by-products (3 g/L – the sum of acetic and formic acid, furfural, and HMF) were identified in the liquid fraction. Both fractions were integrated into enzymatic processes for further co-fermentation of C5- and C6- sugars. Two fermentative routes were evaluated: simultaneous isomerization of xylose and co-fermentation of glucose and xylulose (SIcF) for 2G ethanol production and acetone-butanol-ethanol (ABE) fermentation of glucose and xylose for biobutanol, utilizing Saccharomyces cerevisiae and Clostridium acetobutylicum, respectively. SIcF yielded 0.48 gethanol/gsugars (24 g/L ethanol) with complete glucose consumption and 58.2 % xylose conversion. ABE fermentation produced 0.36 gABE/gsugars (15.7 g/L ABE and 9.5 g/L butanol), with 96 % glucose consumption and 29.6 % xylose conversion. The proof-of-concept demonstrated that the hydrolysates were satisfactorily fermentable by microorganisms, thus validating the pretreatment optimization for fermentation applications.

Investigation of pollutants accumulation in the submerged zone for pyrite-based bioretention facilities under continuous rainfall events

Submerged zone in bioretention facilities for stormwater treatment has been approved to be an effective structure amendment to improve denitrification capability. However, the role and influence of water quality changes in the submerged zone under natural continuous random rainfall patterns are still not clear, especially when the rainfall is less than the pore water in the submerged zone. In this study, continuous rainfall events with different rainfall volume (light rain-light rain-heavy rain) were designed in a lab-scale woodchip mulched pyrite bioretention facility to test the effects of rainfall pattern. The results exhibited that light rain events significantly affected the pollutant removal performance of bioretention for the next rainfall. Different effects were observed during the long-term operation. In the 5th month, light rain reduced the ammonia removal efficiency of subsequent rainstorm events by 8.70%, while in the 12th month, when nitrate leakage occurred, light rain led to a 40.24% reduction in the next heavy rain event's nitrate removal efficiency. Additionally, light rain would also affect the concentration of by-products in the next rainfall. Following a light rain, the concentration of sulfate in the subsequent light rainfall can increase by 24.4 mg/L, and by 11.92 mg/L in a heavy rain. The water quality in the submerged zone and media characteristics analysis suggested that nitrogen conversion capacity of the substrate and microbes, such as Nitrospira (2.86%) and Thiobacillus (35.71%), as well as the in-situ accumulation of pollutants under light rain played important roles. This study clarifies the relationship between successive rainfall events and provides a more comprehensive understanding of bioretention facilities. This is beneficial for field study of bioretention facilities in the face of complex rainfall events.

Stability testing of iomeprol and iopamidol formulations subjected to X-ray radiation

IntroductionExposure of iodinated contrast media (ICM) to X-rays is not uncommon, as contrast media are often stored in close proximity to radiological equipment. However, the interaction between X-rays and ICM is not widely investigated in literature. The present study aims to investigate the chemical stability of iomeprol and iopamidol, two commercial iodinated ICM commonly used in diagnostic imaging, under X-rays exposure. MethodsDifferent formulations of iopamidol and iomeprol (iodine concentration 9 to 400 mgI/mL, volume 50–500 mL) were exposed to three different conditions of X-ray irradiation: i) 1 month storage in CT room (≈5–15 mGy); (ii) low-dose protocol (≈10 mGy); (ii) stressed protocol (≈100 mGy). Unexposed and exposed solutions were characterized by high-performance liquid chromatography in terms of concentration of active pharmaceutical ingredient (API), iodine species and by products. In addition, appearance and colour of the solutions were inspected and pH measured. ResultsAPI concentrations, appearance, colour and pH of the exposed formulations remained unaffected by X-rays. Measured concentrations of iodine species and by products were observed well within the acceptability criteria, i.e. values turned out to be lower than specifications limits established by the manufacturer, considering both release and shelf-life values. ConclusionsUp to 100 mGy X-ray exposure did not induce any alteration of iomeprol and iopamidol formulation, nor a detectable increase in the concentration of iodine species or by-products. Implications for practiceOur study strengthens the hypothesis that ICM are stable under X-rays exposure up to 100 mGy.

Grain by-products and Saccharomyces cerevisiae application in paper packaging material: impact on physical–mechanical and barrier properties

AbstractRenewable materials and material circularity are key priorities for the coming decades. While paper is the most utilized material in the packaging sector, its applications in food packaging are limited due to low barrier properties. Coating it with petroleum-based polymer barrier coatings renders it unrecyclable both mechanically and biologically. Bio-coated paper packaging, made from agricultural waste or by-products, presents environmentally favorable solutions that can benefit the biological cycle of the circular economy. The main objective of this study is to assess the applicability of grain and beer production by-products in wood pulp food packaging materials. It examines the effects of different grain by-products (industrial wheat grain processing residues and wheat bran) particles and concentrations (15 wt% and 40 wt%), as well as the impact of brewer’s yeast (Saccharomyces cerevisiae) at a concentration of 10 wt%, on the physical–mechanical and barrier properties of Northern bleached softwood kraft (NBSK) and chemithermomechanical (CTMP) pulps. Material composites were fabricated using a standard sheet forming method. Physical–mechanical properties were analyzed through tensile strength, strain at break, tear resistance, and bending stiffness tests. Barrier-related properties were analyzed through roughness, air permeance, and water contact angle tests. Results revealed that the vast majority of physical–mechanical properties decreased with the increase of both types of grain production fillers. Industrial wheat grain processing residues had a lower impact on physical–mechanical properties than wheat bran filler. A higher percentage of property decrease was absorbed in NBSK pulp than in CTMP. Roughness of CTMP can be improved by adding both industrial wheat grain processing residues and wheat bran. Wheat bran particles can increase hydrophobicity. CTMP pulp strength properties can be increased with the addition of yeast; however, the yeast additive’s effect on air permeance demonstrated a negative impact. In the case of NBSK pulp, which has higher primary strength properties, the addition of yeast does not make any significant changes. The conducted experiments demonstrated that, overall, the addition of these fillers resulted in a decline in physical–mechanical properties such as tensile strength, strain at break, tear resistance, and bending stiffness. This decline was more pronounced with higher concentrations of fillers. In the case of CTMP pulp, both types of fillers exhibited similar trends in affecting properties, whereas for NBSK pulp, wheat bran had a more significant impact compared to wheat grain processing residues. The addition of yeast generally led to a reduction in physical–mechanical properties, particularly in tensile strength and tear resistance. However, samples containing yeast displayed increased flexibility compared to controls. The influence on barrier properties varied: while yeast increased air permeability, it also enhanced surface hydrophobicity, thereby reducing the paper’s receptivity to liquids.

Enhancing the Retention and Oxidative Stability of Volatile Flavors: A Novel Approach Utilizing O/W Pickering Emulsions Based on Agri-Food Byproducts and Spray-Drying.

Spray-drying is a commonly used method for producing powdered flavors, but the high temperatures involved often result in the loss of volatile molecules. To address this issue, our study focused on a novel approach: developing O/W Pickering emulsions with agri-food byproducts to encapsulate and protect D-limonene during spray-drying and storage. Emulsions formulated with lupin hull, lupin-byproduct (a water-insoluble protein-fiber byproduct derived from the production of lupin protein isolate), and camelina press-cake were subjected to spray-drying at 160 °C. The results revealed that these emulsions exhibited good stability against creaming. The characteristics of the dry emulsions (powders) were influenced by the concentration of byproducts. Quantitative analysis revealed that Pickering emulsions enhanced the retention of D-limonene during spray-drying, with the highest retention achieved using 3% lupin hull and 1% camelina press-cake. Notably, lupin-stabilized emulsions yielded powders with enhanced oxidative stability compared to those stabilized with camelina press-cake. Our findings highlight the potential of food-grade Pickering emulsions to improve the stability of volatile flavors during both processing and storage.

Spatial and seasonal variation in disinfection byproducts concentrations in a rural public drinking water system: A case study of Martin County, Kentucky, USA.

To increase our understanding of the factors that influence formation of disinfection byproducts (DBPs) in rural drinking systems, we investigated the spatial and seasonal variation in trihalomethane (THM) and haloacetic acid (HAA) concentrations in relation to various chemical and physical variables in a rural public drinking water system in Martin County, Kentucky, USA. We collected drinking water samples from 97 individual homes over the course of one year and analyzed them for temperature, electrical conductivity, pH, free chlorine, total chlorine, THMs (chloroform, bromodichloromethane, dibromochloromethane, dichlorobromomethane, and bromoform) and HAAs (monochloroacetic acid, dichloroacetic acid, trichloroacetic acid, bromoacetic acid, and dibromoacetic acid). Spatial autocorrelation analysis showed only weak overall clustering for HAA concentrations and none for THMs. The relationship between modeled water age and TTHM or HAA5 concentrations varied seasonally. In contrast, there was strong variation for both HAA and THMs, with concentrations of HAA peaking in mid-summer and THMs peaking in early fall. Multiple regression analysis revealed that THM concentrations were strongly correlated with conductivity, while HAA concentrations were more strongly correlated with water temperature. Individual DBP species that only contained chlorine halogen groups were strongly correlated with temperature, while compounds containing bromine were more strongly correlated with conductivity. Further investigation revealed that increased drinking water conductivity associated with low discharge of the Tug Fork River, the source water, is highly correlated with increased concentrations of bromide. Discharge and conductivity of the Tug Fork River changed dramatically through the year contributing to a seasonal peak in bromide concentrations in the late summer and early fall and appeared to be a driver of brominated THM concentrations. Brominated DBPs tend to have higher toxicity than DBPs containing only chlorine, therefore this study provides important insight into the seasonal factors driving risk from exposure to DBPs in rural drinking water systems impacted by bromide.

Removal of Anabaena by ultrasonic pretreatment enhancing-coagulation and water treatment processes

Cyanobacteria bloom brings great challenges to the safety of the drinking water process. Ultrasonic pretreatment can effectively enhance coagulation to remove algae cells from algae-laden water. However, due to the special chain-like structure of Anabaena, the removal characteristics of Anabaena in water treatment process are quite different from those of unicellular cyanobacteria. No study has yet systematic studied on the enhancement effect of ultrasonic enhanced water treatment process on Anabaena. Therefore, this paper studied the characteristics and mechanism of ultrasonic pretreatment to enhance the removal of Anabaena, and carried out pilot experiments. The results showed that ultrasound irradiation at 740 kHz and 1.12 MHz had a great effect on the destruction of the chain-like structure and cell structure of Anabaena. After 15 min, the removal rate of Anabaena cells reached 16.0% and 30.4%, respectively, and the release of intracellular organic matter (IOM) reached 4.3 and 6.0 mg/L, in which phycocyanin released more. The damage of dynamic ultrasound irradiation on algae cells is weak, which can avoid the influence of uneven ultrasound irradiation on algae cells. The enhanced coagulation effect was better after 740 kHz ultrasonic pretreatment for 1 min, and the removal rates of algal cells, turbidity and dissolved organic carbon (DOC) increased by 7%, 9.4% and 12.6%, respectively. Pilot scale experiments demonstrate that ultrasonic pretreatment leads to a notable enhancement in the removal efficiency during the coagulation and sedimentation stages. After 4 h of operation, the turbidity and algal cell removal in the sedimentation stage increased by 6.1% and 14.0%, respectively. In addition, the removal of DOC in the final treated water increased by 6.7%. Furthermore, the disinfection by-products (DBPs) concentration experiences a reduction exceeding 200 μg per liter. Together, these results show a substantial improvement in filtration efficiency and effluent quality. This study provides guidance for the ongoing study of ultrasonic pretreatment to remove chain-like cyanobacteria.

Urinary and household chemical exposures in pet dogs with urothelial cell carcinoma.

Urothelial cell carcinoma (UCC) has been linked to environmental chemical exposures in people, but these risk factors are not well understood in dogs with UCC. We hypothesised that household chemical exposures contribute to the risk of UCC in pet dogs. This prospective cross-sectional case-control study included 37 dogs with UCC and 37 unaffected breed-, sex-, and age-matched controls. Dog owners completed an environmental questionnaire and household samples were collected and analysed for arsenic (in tap water and room dust) and acrolein (in room air). Urine samples from UCC dogs, control dogs, and consenting owners were analysed for inorganic arsenic species, the acrolein metabolite 3-HPMA, and the phenoxy herbicide 2,4-D. Public data on chlorination byproducts (total trihalomethanes) in municipal drinking water were also compared between case and control households. Dogs with UCC were more likely to swim in a pool (15.2%) compared with control dogs (0%) (OR 1.69, 95% CI = 1.69-∞; p = .02). Dogs with UCC also had more than 4-fold higher reported municipal water concentrations of chlorination byproducts (median 28.0 ppb) compared with controls (median 6.9 ppb; p < .0001). Dust arsenic concentrations were unexpectedly lower in case households (median 0.277 ng/cm2) compared with control households (median 0.401 ng/cm2; p = .0002). Other outcomes were not significantly different between groups. These data suggest that dog owners, especially those of breeds known to be at higher risk for UCC, consider limiting access to swimming pools and installing water filtration units that remove total trihalomethanes.

Segmentation and evaluation of pathway module efficiency: Quantitative approach to monitor and overcome evolving bottlenecks in xylose to ethanol pathway

Engineering microbes that can efficiently ferment xylose to ethanol is critical to the development of renewable fuels from lignocellulosic biomass. To accelerate the strain optimization process, a method termed Segmentation and Evaluation of Pathway Module Efficiency (SEPME) was developed to enable rapid and iterative identification and removal of metabolic bottlenecks. Using SEPME, the overall pathway was segmented into two modules: the upstream xylose assimilation pathway and the downstream pentose phosphate pathway, glycolysis, and fermentation. The efficiencies of both modules were then quantified to identify the rate controlling module, followed by analyses of control coefficients, reaction rates, and byproduct concentrations to narrow down targets within the module. SEPME analysis revealed that as the strain was engineered with increasing xylose-to-ethanol yields, the bottlenecks shifted within a module and across the two modules. Guided by SEPME, these bottlenecks were removed one by one, and a strain approaching the theoretical ethanol yield was obtained.

Rapid in situ regeneration of phenol-saturated activated carbon fiber by an electro-permonosulfate-ozone process: Performance, operators and mechanism

In this study, a new method that encompasses electricity, permonosulfate (PMS), and ozone (E-PMS-O3) was proposed and applied in the regeneration of phenol-saturated activated carbon fiber (ACF). Compared with traditional regeneration technology, the E-PMS-O3 process simultaneously regenerated exhausted ACF in situ and mineralized the desorbed pollutants effectively (78.67%) with relatively low energy consumption (4.338 kWh m−3). Furthermore, the E-PMS-O3 regeneration process only required 2 h, which was shorter than other well-documented electro-advanced oxidation processes (EAOPs), such as the E-O3 process lasting for 3 h, the E-persulfate process for 6 h, and the E-Fenton process for 6 h. Possible pathways in the regeneration process include direct electron transfer, ozone oxidation, PMS oxidation, non-radical reactions, and reactive oxygen species (ROS) oxidations. Two main ROS — hydroxyl radical (O•H) and sulfate radical (SO4•−) were probed and their contribution ratio was approximately 1:1. An external electric field visibly enhanced the recovery adsorption capacity of ACF and accelerated the decomposition of PMS. Interestingly, additional experiments revealed that adding two different oxidants (PMS/O3) provided a better protection effect on the properties of ACF against oxidation of oxidants and ROS. It's important to mention that the maximum concentration of byproducts remained below 5 mg g−1, which effectively minimized the potential for subsequent pollution. In the end, multiple regeneration cycles demonstrated that the regeneration efficiency was nearly 60% even after 6 cycles. Hence, the E-PMS-O3 process was promising for further exploration and discussion when applied in regeneration of exhausted AC materials.
