Sulfamic Acid Catalyzed One-Pot Synthesis of Polyhydroquinolines via the Hantzsch Four Component Condensation Reaction

The synthesis of polyhydroquinoline derivatives has been achieved using catalytic amounts of green, inexpensive and environmentally benign sulfamic acid. The process is operationally simple and has good to excellent yields.

An efficient one pot synthesis of 1,4-dihydropyridines using alumina sulfuric acid (ASA) catalyst

An efficient synthetic method for 1,4-dihydropyridines has been developed using 3 or 4 component condensation reactions of aldehydes, 1,3-dicarbonyl compounds, and ammonium acetate in the presence of alumina sulfuric acid catalyst in minimum methanol at reflux temperature. This procedure offers several advantages including high yields, an environmentally friendly procedure, short reaction times, and a simple work-up procedure.

Synthesis of Proteasome Inhibitor Omuralide Featuring Stereocontrolled Ugi Reaction and Novel Convertible Isocyanide

Stereocontrolled formal total synthesis of proteasome inhibitor omuralide is described. This is featured by the application of Ugi 4–center 3–component condensation reaction to prepare a highly functionalized pyroglutamic acid. To this end, a novel convertible isocyanide was developed, which is readily available by preparative synthesis from nitrotoluene. The hydrolysis of the sterically hindered C–terminal amide derived from the isocyanide was facilitated by the conversion to N–acylindole. By careful design of the γ–ketoacid precursor, a substrate–controlled diastereoselective Ugi reaction was successfully accomplished. Also, several highly functionalized pyroglutamic acid amides were prepared with high diastereoselectivity.

Nano-sized copper as an efficient catalyst for one pot three component synthesis of thiazolidine-2,4-dione derivatives

Abstract A simple, efficient, stereoselective, one pot three component condensation reaction between thiazolidine-2,4-dione, aldehyde and amine derivatives using monodispersed, recyclable and inexpensive Cu-nanoparticles for the synthesis of thiazolidine-2,4-dione derivatives in excellent yields and high purity has been described here for the first time.

An Efficient One‐Pot Biginelli Condensation of Aliphatic Aldehydes Catalyzed by Zinc Bromide under Solvent‐Free Conditions

Zinc bromide catalyzes the three component condensation reaction of an aldehyde, urea, and β‐ketoester or β‐diketone under solvent‐free conditions to afford the corresponding dihydropyrimidinones (DHPMs) with moderate to high yields in short reaction time. The present method is very effective for the Biginelli condensation of aliphatic aldehydes.

Comparison of Catalytic Effect of Alkali and Alkaline Earth Metals Hydrogen Sulfate: As the Promoter for an Efficient Synthesis of 3,4-Dihydropyrimidin-2(1H)-ones under Solvent-Free Conditions

Alkali and alkaline earth metals hydrogen sulfate catalyzed the one-pot three component condensation reactions of aldehydes, 1,3-dicarbonyl compounds and urea or thiourea under solvent-free conditions leading to 3,4-dihydropyrimidin-2(1H)-ones in high yields at 80 °C. Our results showed that Biginelli reaction not only is pH dependent, but the cation of catalyst plays an important role.

1,4-Dipolar cycloaddition in organic synthesis: A facile route to isoquinoline fused heterocycles

The three component condensation reactions involving isoquinoline, dimethyl acetylenedicarboxylate and carbonyl dipolarophiles such as o- and p-benzoquinones and N- substituted isatins constitute a one-pot synthesis of a variety of (1,3)oxazino isoquinoline derivatives via 1,4-dipolar cycloaddition.

Bismuth nitrate pentahydrate catalyzes the three component condensation reaction of an aromatic aldehyde, urea and a β‐ketoester or a β‐diketone under solvent‐free conditions to afford the corresponding dihydropyrimidinones (DHPMs) in high yields. The present method is also effective for the selective condensation of aryl aldehydes in the presence of aliphatic aldehydes.

Silicagel supported sodium hydrogensulfate as a heterogenous catalyst for high yield synthesis of 3,4-dihydropyrimidin-2 (1H)-ones

Silicagel supported sodium bisulfate catalyzes efficiently the three component condensation reaction of aldehyde, β-ketoester and urea in refluxing acetonitrile to afford the corresponding dihydropyrimidinones. Compared to the classical Biginelli reaction conditions, this new approach consistently has the advantage of excellent yields (84–93%) and short reaction times 1–2.5 h.
