Edukasi Kualitas Air Bersih Bebas Kontaminasi Bakteri Coliform pada Masyarakat Jungsereh Garut

Indonesian people still use dug wells as a source of clean water. The dug well itself has bacteriological qualities that are easily contaminated. Coliform bacteria such as Escherichia coli can cause diseases, one of which is diarrhea. Jungsereh Garut Village is an area that uses a lot of water from dug wells for daily needs such as drinking water. The aim of this outreach is to determine the quality of clean water free of coliform bacteria contamination in the Jungsereh Garut community. The target of this counseling is the community of Jungsereh Village RT.02 Kec.Mekarsari Garut, totaling 30 people. This outreach activity is a form of increasing public awareness of the quality of clean water for drinking water that is free of coliform bacteria contamination. The method of implementing this extension activity is in the form of material presentation, discussion and question and answer and filling out a questionnaire regarding the level of understanding whether there are differences before and after the intervention.

FOOD SAFETY STUDY IN TERMS OF MICROBIOLOGY IN PORK SATAY IN SANGEH TOURISM VILLAGE

The application of tourism health, such as food safety, is an important matter to pay attention to. Based on the annual report (BPOM, 2019), there are 373 cases of food poisoning in Bali, so Bali ranks fourth with the highest cases of food poisoning in Indonesia. The type of research used is observational. This research was conducted by testing samples to determine microbial contamination on pork satay as well as observing and recording food safety scores where pork satay is sold. The design used was cross-sectional because all research subjects were observed at one time. Food safety scores were tested, and TPC, MPN, and E. coli were tested so that 9 samples (56.25%) were categorized as vulnerable to safe consumption. 16 samples of pork satay met the minimum requirements for microbial contamination, ranging from 20/g to 60,000/g. 1 sample of pork satay (6.25%) with the amount of coliform contamination exceeding the standard, namely 438/gr. There were 6 samples of pork satay (37.5%) positive for E. coli. Based on the results of the Kruskal-Wallis test for total microbial contamination based on the food safety score, the results showed that there was a difference in the results of the microbial contamination test and E. coli with a food safety score. Coliform bacteria contamination based on the food safety score showed that there was no difference from the results of the MPN test with the food safety score based on the Asymp.sig value 0.05. Even so, there are still many traders who have not implemented personal hygiene, so it is necessary to carry out food safety training, monitoring, and developing food quality on a regular basis.

ANALISIS CEMARAN MIKROBIOLOGI BEBERAPA ASPEK PRODUKSI PADA PEDAGANG PENYEDIA MAKANAN DI SEKITAR KAMPUS UNIVERSITAS BUMIGORA MATARAM

Most health problems by food are caused by contamination of microorganisms. This contamination can due because cleanliness or improper implementation of sanitation and hygiene. The aim of this research is to identify microorganism contamination in the production aspects of food traders around the Campus of Bumigora University Mataram. Total microbial testing was carried out using the Total Plate Count (TPC) method for air, table, floor, equipment and water samples. Another method for testing coliforms in water use the principle of the Most Probably Number (MPN) method. The observation results show that the environmental quality as seen from the total microbes in the air is below the threshold, namely <700 col/m3, so it is said to be still safe. Meanwhile, table, floor and equipment samples showed that the total number of microbes was high enough to exceed the required threshold, especially those related to eating utensils such as plates, glasses and spoons. The total microbial test in the water sources used by the three traders showed that the two water sources used contained microbial colonies which were tested via TPC with colony numbers ranging from 87.7 – 99 x 10-5 CFU/ml. The results of the coliform estimation test using the MPN method principle showed that there was no suspicion of any coliform bacteria contamination in the two water samples, which indicated that both water sources also met the clean water requirements according to Minister of Health Regulation No. 492 of 2010.

Analysis of Personal Hygiene, Premises Sanitation and Equipment Sanitation on Coliform Bacteria Content in Bubble Drinks

Abstract. Ice bubble (bubble drink) is a popular ready-to-eat drink and is in great demand by the public because of the variety of flavors with a mixture of bubble topping (boba) in it. The personal hygiene of drink handlers, the cleanliness of sales locations, and the equipment used can all cause microbiological damage to bubble ice, which can lead to coliform bacteria contamination of bubble ice drinks. Coliform bacteria can cause various diseases, one of which is diarrhea. This study aims to determine the relationship between personal hygiene, place sanitation, and equipment sanitation and the content of coliform bacteria in bubble ice drinks in Pontianak City in 2022. According to the findings, coliform bacteria were present in 27 out of the 30 samples that had undergone coliform bacteria testing. This study obtained results of poor personal hygiene for as many as 26 people (86.7%), poor sanitation of selling places for as many as 27 people (90%), and poor sanitation of equipment for as many as 27 people (90%). This study also shows that there is a relationship between personal hygiene and coliform bacteria content with a value of p = 0.039, a relationship between place sanitation and coliform bacteria content with a p value = 0.02, and a relationship between equipment sanitation and coliform bacteria content in ice bubble drinks. with a p value of 0.039. Therefore, it is necessary to pay attention to the cleanliness of tools, the environment, and individual traders so that bubble ice is safe for consumption

Analysis of coliform bacteria contamination in drinking water refill in Ambon City

In recent years, refillable drinking water businesses have grown rapidly in several cities in Indonesia, including Ambon City. On the one hand, Refillable Drinking Water Depots (DAMIU) support efforts to realize a healthy society by expanding the reach of clean water consumption, On the other hand, DAMIU tend to be problematic when faced with business interests. Feces contain coliform bacteria which is an indicator of water contamination. This study aims to determine the presence of coliform bacteria in Ambon City. The samples used in the study came from three sub-districts of refill drinking water depot sources, namely the Teluk Dalam sub-district, Nusaniwe sub-district, and Baguala sub-district. Microbiological testing of refill drinking water using MPN (most probable number) method. The MPN test results showed that three refill drinking water samples were not contaminated with coliform bacteria because there was no turbidity or gas formation in the Durham tube for gas-forming bacteria. This indicates that the three refill drinking water samples tested microbiologically have good quality and are safe for consumption, because in 100 ml of water no coliform bacteria were found in accordance with Permenkes No. 492/Menkes/Per/IX/2010 regulations on the requirements and supervision of water quality, which states that the microbiological requirements for drinking water are MPN Coliform /100 ml sample = 0. There are no coliform bacteria in the refillable drinking water depot in Ambon City so the water is suitable for consumption

The Relationship between Sanitary Hygiene and Coliform Bacteria Contamination at Refill Drinking Water Depots

Objective: This study aims to determine the relationship between sanitation hygiene and Coliform bacteria contamination in refilled drinking water in Waled District in 2023. Methods: This study uses an analytic observational design with a cross-sectional approach where the researcher will simultaneously observe or measure the independent and dependent variables (period). The sampling technique is total sampling with a sample of 30 refilled drinking water depots with 30 workers—data collection through observation and interviews. Data analysis was used, namely data analysis using Fisher's exact test. Results: Based on laboratory tests, it was found that more than half of the refilled drinking water depots were contaminated with coliform bacteria as much as 60%. After analysis, it was found that factors related to Coliform bacteria contamination in filled drinking water were access to sanitation facilities (p = 0.001), and worker behavior (p = 0.001), while factors that were not related were drinking water treatment facilities (p = 0.063), raw water (p = 1.000) and consumer service hygiene (p = 0.418). Conclusion: Of the five sanitation hygiene factors, those that have a significant relationship with coliform bacteria contaminants are sanitation facilities and behavior, while drinking water treatment facilities, raw water, and consumer service hygiene are factors that are not significant in determining bacterial contamination.

Microbiological Quality of Drinking Water and Food in a Rural Community

Safe water and food are essential for good human health. The contamination of water with infected fecal bacteria is common in areas with poor standards of hygiene and sanitation. This study investigated total coliform bacteria, fecal coliform bacteria, and E.coli in drinking water and food stalls using water from a rural community, where the rural living conditions increase the risk of water-related diseases. In total, 40 samples of bottled drinking water, 50 water samples from vending machines, 30 samples of household drinking water, 30 food samples were collected from household food, and 60 food samples were collected from food stalls, and coliform bacteria contamination was determined using the most probable number (MPN) technique. The results revealed that all the water and food samples contained coliform bacteria contamination, while all the water samples and 50% of the food samples contained fecal coliform bacteria. The numbers of household food samples contaminated with coliform bacteria and E.coli were 29 (97%) and 12 (40%), respectively. Entrepreneurs should be regularly maintaining clean drinking water vending machines. The government sector should be urgently solving the problem of drinking water standards to ensure suitable consumer safety.

Coliform Contamination of Raw Beef at the Slaughterhouse and Butchery Levels in Herat City, Afghanistan

Background: Contaminated food products are responsible for causing various food borne diseases all over the world. Microbial contaminants are present in various food products, particularly raw meat. Among the most common microbial contaminants in meat and meat products are coliform bacteria. This study aimed to determine the coliform quality of raw beef with special emphasis on Escherichia coli in Herat city, Afghanistan and to compare the coliform contamination levels in the slaughterhouse and butchery levels. Materials and Methods: In this study, 150 samples of raw beef were randomly selected from butcheries and slaughterhouses of Herat city. Coliform identification was performed by standard bacteriological methods. Findings: The findings of this study revealed that coliform bacteria were detected in 70% and 75 % of beef samples from slaughterhouses and butcheries respectively. A total of 58 % of positive samples were upon the standard accepted ratio of coliforms contaminant per gram. The E. coli contamination ratio of samples in slaughterhouses and butcheries were 52% and 72% respectively. Our study demonstrates a significant difference in coliform bacteria contamination between slaughterhouse samples and butcheries. Conclusion: It was concluded that more than half of the coliform contamination in raw beef from the slaughterhouse and butcheries was exceeded the maximum limit which highlights the need to focus on effective monitoring of the slaughterhouse and butcheries in Herat city for prevention of meat borne intoxication and infection.

ANALISIS UJI MIKROBA PADA MINUMAN COCOGHURT

Cocoghurt is a fermented drink made from coconut milk sediment. This study aims to analyze the microbial test on cocogurt drink. In this study the tests carried out were salmonella, coliform, escherichia coli, and bacillus tests using the Most Likely Number (APM) method in accordance with SNI 01-2897-1992, in addition to this test using a presumptive test approach or estimation and confirmation or confirmation with using liquid media in test tubes and carried out based on the number of positive tubes. Samples that were confirmed positive could be seen by the appearance of gas in the Durham tube and the bacillus test was carried out by observing using a binocular microscope with two temperature treatments, namely 400 C and 40 C. The Coliform and E. coli tests that had been carried out showed that the cocoghurt contained coliform bacteria contamination. of <3 APM/g and Escherichia coli of <3 APM/g. The amount of contaminants from the microbial test results is within the safe limit of the Indonesian National Standard for fermented beverages.

Analysis of Coliform and Colifecal Contamination on Sanjai Chip Using MPN Method

Sanjai chip is a type of traditional food made of cassava processing and it is typical food of West Sumatera. Sanjai chips sold at Pasar Bawah in Pekanbaru City sold in an open state and the location is close to the trash heap, so it is feared to be contaminated by bacteria. The purpose of this research was to find out if there was or there was no contamination of coliform and colifecal bacteria on Sanjai chips. This bacteria contamination was analyzed on seven sanjai chip from different sellers using Most Probable Number (MPN) method with type 5-1-1. The result on seven samples obtained shows that all the samples did not contain coliform marked with there was no formation of gas bubble in the durham tube in the preliminary test. Therefore, affirmation test in this research was not continued. Thereby, Sanjai chips sold at Pasar Bawah Pekanbaru City fulfilled the requirement according to Indonesian National Standard (SNI) Number 7388-2009 about maximum limit of microba contamination in food, where the bacteria number allowed is <3/gram, so it is safe for consumption.

Most Probable Number of Coliform on Beef from Krian Abattoir, Sidoarjo

The problem on food health is the high amount of bacterial contamination of food served by food industries. The quality of beef can be affected by the level of bacterial contamination in meat. Coliform is a group of bacteria used as a sanitary hygiene indicator. The purpose of this study is to measure the value of MPN Coliform in beef at Krian’s abattoir of district Sidoarjo. Thirty samples were taken from the abattoir of Krian, Sidoarjo. Most Probable Number was used to determine the level of Coliform's contaminate beef. The results showed that all beef samples at abattoir were contaminated with Coliform. The average contaminant levels was 706,76 MPN/g. Twenty six samples exceed the maximum limit of Coliform bacteria contamination in accordance with Indonesian National Standard (SNI). The highest Coliform MPN value is ≥ 1600 MPN / g, whereas the lowest MPN Coliform value is 8 MPN/g.

Kualitas Bakteriologis Sumber Mata Air Mumbul Sari Kabupaten Lombok Utara

Water is the main and most important need for living things. The water consumed must meet the physical, chemical, bacteriological requirements, and be free from Coliform bacteria contamination. This research is a qualitative descriptive study that aims to determine the bacteriological quality of the spring water in the Mumbul Sari Village community, Bayan District, North Lombok Regency. The data collection technique used a filter membrane with Colony Forming Units (CFU/100ml). The research data were analyzed descriptively. The results showed that the Mumbul Sari spring in North Lombok Regency was contaminated with Coliform and Escherichia coli bacteria with values exceeding the quality standards set by the Ministry of Health of the Republic of Indonesia. The MPN Coliform value of springs used by women is higher than that of springs used by men (300 CFU/100ml > 250 CFU/100 ml). Similarly, the number of Escherichia coli bacteria (50 CFU/100ml > 12 CFU/100ml).

The Correlation between Personal Hygiene of Greengrocer and Coliform Bacteria Contamination in Lettuce in Jember Traditional Markets

Lettuce is a vegetable that is usually consumed in raw condition or unprocessed by people. Unprocessed vegetables can cause food-borne diseases. Lettuce that is not processed and being consumed directly can cause contamination by Coliform bacteria. The purpose of this study is to know the correlation between Greengrocer’s personal hygiene and Coliform bacteria contamination in lettuce in Jembertraditional markets. This research used a descriptive observational design with the cross-sectional approach. It took place in eight traditional markets in Jember Regency and Microbiology Laboratory, Faculty of Medicine, University of Jember. The samples used were 41 lettuces purchased from 41 traditional market sellers. The microbiological examination was performed in the laboratory using the most probable number method. The results showed that lettuce sellers had bad personal hygiene (92,7%) and the data from the most probable number test found 97,6% contaminated by Coliform bacteria. The bivariate analysis was used to know the relationship between personal hygiene vegetable seller and contamination Coliform bacteria in lettuce. Thep-value was 0,072, which means there was no relationship between personal hygiene vegetable seller and contamination Coliform bacteria in lettuce.

ANALISIS CEMARAN BAKTERI Coliform DAN IDENTIFIKASI Escherichia coli PADA ES BATU BALOK DI KOTA KARAWANG

Ice cubes are one of the food products that are well known by the public and are considered safe for consumption. The Aim of this research is to see the contamination of Coliform bacteria and the presence of Escherichia coli in ice cubes. Test methods used include: Coliform test and identification of Escherichia coli bacteria. The results showed that 2 of the 6 positive samples contained Coliform bacteria and Escherichia coli bacteria with MPN values ​​above the safe limit. The conclusion of this study is that the ice cubes used by the perpetrators around the city of Karawang have microbiological quality that is not suitable for consumption by the public and does not comply with the microbiological quality requirements of the Indonesian National Standard 01-3839-1996 regarding ice cubes, namely the Total Coliform Bacteria exceeds 0/ 100mL.

Contamination of Escherichia coli, Salmonella sp. and Vibrio sp. on ice cubes at food stalls in Karangasem Regency, Bali Province in 2021

Background: Waterborne diseases spread through drinking water and food products made from drinking water, including cooking utensils that are washed using contaminated water. Ice cube is widely used by the people in Indonesia. There was no study regarding bacteria contamination on an ice cube in Karangasem Regency. This study aimed to determine the contamination of Escherichia coli, Salmonella sp., and Vibrio sp. on ice cubes at food stalls in Karangasem Regency in 2021. Methods: This study used a cross-sectional research design and simple purposive sampling as the sampling technique. Ice cubes were taken from 42 food stalls from eight sub-districts. Samples were cultured on Lactose Broth (LB) media for the Most Probable Number (MPN) test procedures and Eosin Methylene Blue Agar (EMBA) to identify the presence of E. coli, Salmonella Shigella Agar (SSA) for Salmonella sp., and Thiosulfate Citrate Bile Sucrose (TCBS) for Vibrio sp. and followed by gram staining. Data processing was carried out descriptively to determine the percentage of samples with positive contamination. Data were analyzed using Microsoft Excel version 2010 for Windows. Results: Based on the study results, it was found that all samples contained coliform bacteria through the MPN test (100.0%). In addition, 5 samples (11.9%) were found positive for E. coli. While the identification of Salmonella sp. and Vibrio sp was found no positive results. Conclusions: There was coliform bacteria contamination in the tested ice cube samples. In addition, E. coli was found in the sample; however, there were no Salmonella sp. and Vibrio sp. on ice cube samples.

SAFETY ASSESSMENT OF STREET FOODS IN THE BOLGATANGA MUNICIPALITY OF THE UPPER EAST REGION, GHANA

Street foods provide a convenient diet for many people in developing countries with the consumption supporting the livelihood of several individuals. However, street food safety is a major global concern due to poor food handling and insanitary conditions that often lead to coliform bacteria contamination resulting in the outbreak of foodborne diseases. This paper analyses the microbial content of food served by food vendors in the Bolgatanga Municipality of Ghana, which has recorded high levels of foodborne diseases. Using a mixed research approach, and a cross-sectional design, a total of 150 food vendors were sampled from 10 communities within the Municipality. Food samples from the vendors were taken and subjected to microbial load analysis. Out of the 66 food samples tested, 42.4 percent recorded the presence of Staphylococcus aureus (S. aureus) with only six recording the presence of S. typhi (S. typhi). All the samples that were isolated for Escherichia coli (E. coli) had microbial loading of 1 and above, with the lowest recorded to be 1.6 ± 0.6 log CFU/g. The highest contamination by E. coli was in the samples of tomato sauce, followed by samples of rice balls and then by groundnut soup. The high microbial quality of the street foods sampled suggests that the safety of food vendors’ food handling practices was largely compromised and so most of the foods were contaminated above acceptable levels for consumption. The study recommends that food vendors should be educated on proper personal hygiene and to avoid making direct skin contact with food.

Depot Sanitation, Hygiene, and Analysis of Total Microba Control in Refilled Drinking Water in The Barus District

Water is the most important requirement for life. People would prefer to refill their drinking water to satisfy their needs because it is easier and more efficient. It is important to implement hygiene and sanitation measures at drinking water depots if the drinking water produced is to be healthy and safe. Contamination of water can occur if drinking water depots are not kept clean and sanitary. The research used is a descriptive study conducted in the Barus District, with the goal of providing a description of the depot's hygiene and sanitation, as well as laboratory analysis to identify the total contamination of coliform bacteria in the Barus District's Refill Drinking Water Depot. According to the Minister of Health of the Republic of Indonesia no 43 in 2014. None of the drinking water depots in Barus district met the physical qualifying requirements. The exam resulted in a score of 70 or higher. and the results of laboratory tests on the bacteriological quality of drinking water replenished with all coliform-contaminated drinking water samples that exceeded the quality satandard. As a result, the depot owner should turn on the disinfection equipment when starting work and always pay attention to building sanitation, employee hygiene and maintaining drinking water treatment equipment to prevent contamination of drinking water.

Analisis Cemaran Bakteri Coliform Pada Minuman Jajanan Dengan Metode MPN (Most Probable Number)

Coliform bacteria contamination of food and beverages may result from various causes, one of which is faecal contamination, leading to serious health issues. Drinking water contamination of coliform bacteria 50/100 mL has been found to be responsible for diarrhea episode. Local regulation has determined the cut-off point of its contamination within commercial beverages not to exceed 3/mL. No such study has ever been conducted in Ternate. To determine the exact value of coliform bacteria within street beverages sold in various city parks in Ternate. This descriptive was carried out through laboratory testing of street beverages obtained from various city parks in Ternate. The most probable number test for coliform bacteria was carried out at the Technical Implementation Unit, Basic and Integrated Laboratory, North Maluku from December 2021-January 2022. The presumption test and the affirmation test were conducted accordingly. From 22 instant powdered ice drinks samples obtained from Toboko, Falajawa 1, Nukila, Landmark, and Film Benteng parks, 90,9% contained coliform bacteria exceeding the cut-off value. Coliform bacteria contamination in the city parks within Ternate has exceeded the quality standard threshold based on Indonesian National Standard 7388 of 2009. Further research is needed to confirm this.

Analisis Kandungan Cemaran Bakteri Coliform Pada Air Rendaman Tahu di Pabrik Tahu Wilayah Kalideres Jakarta Barat

Tofu is a food source containing high vegetable protein. Tofu is in great demand by most of the Indonesian people because it contains high nutrition and the price is relatively cheap and affordable. To produce tofu requires good hygiene and sanitation, such as providing clean raw materials and water sources that are free from contamination. To determine whether the water source has been contaminated with microbes or not, the Coliform bacteria indicator in the water sample is used. In testing Coliform bacterial contamination in water or raw materials, the Most Probable Number (MPN) method is used to calculate the amount or level of microbial contamination in determining the quality or quality of water and raw materials by referring to the MPN table. This study aims to determine the level of Coliform bacteria contamination in tofu soaking water in a tofu factory in the Kalideres area of ??West Jakarta. The design of this research is a descriptive laboratory type which was carried out in March-June 2020 at the Indonesian Social Solidarity STIKes Laboratory. Based on the results of the research on the presumptive test, it was found that 1 positive tube was contaminated with Coliform in the KR tofu factory and 3 positive tubes at the DN factory. The presumptive test was continued to confirmatory test and found 5 positive tubes polluted at the DD factory, 2 positive tubes at the KR factory and 3 positive tubes at the DN factory. The positive tube from the confirmation test was calculated for the contamination index value (MPN) and obtained 21000 mpn/ml for the DD factory, 4000 mpn/ml for the KR factory and 9000 mpn/ml for the DN factory. It can be concluded that there is Coliform bacterial contamination in all tofu soaking water samples in 3 tofu factories with the MPN index value having passed the contamination threshold.

GAMBARAN KUALITAS BAKTERI KOLIFORM AIR BERSIH PADA SUMUR GALI DI DESA TONGUTE TERNATE KECAMATAN IBU KABUPATEN HALMAHERA BARAT PROVINSI MALUKU UTARA

The World Health Organization (WHO) in the Director General of Disease Eradication and Environmental Health states that the average volume of clean water needs for the world's population is different. In developed countries, the water required is approximately 500 liters/or/day. Meanwhile, big cities in Indonesia need 200-400 liters of water/or/day. One of the bacteria that can contaminate water quality is coliform bacteria, which is a group of intestinal bacteria that live in the human digestive tract. And the purpose of this research is to find out the description of Coliform bacteria in clean water of dug wells in Tongute Village, Ternate District, Ibu Subdistrict, West Halmahera Regency, Maluku Province. This research was conducted at the Laboratory of Microbiology, Faculty of Medicine, Hasanuddin University Makassar. The method used in this research is an experimental design with descriptive observation totaling 5 samples of dug wells, carried out with the Most Probable Number (MPN) in the Estimating Test and Confirmation Test for Dug Well Water Samples and Testing on Complementary Tests Using Eosyn Methylene Blue Agar (EMBA) Media Growth Media ). The results showed that in the examination of the estimator test using lactose Broth (LB) media, the 5 samples in the estimator test were marked by a change in the solution to yellow color and the appearance of gas bubbles in the Durham tube so that it was suspected that it contained coliform bacteria contamination. In the confirmation test examination using Brilliant Green Lactosa Broth (BGLB) media, a positive test result in the estimator test is continued to the confirmation test to confirm coliform In the confirmation test using the EMBA medium, the positive test result is confirmed by a confirmation test to determine the type of coliform bacteria his. The result is called fecal coliform if the colonies that grow in the medium are metallic green, if the colonies that grow are brick red, it is called non-fecal coliform. It was concluded that there were coliform bacteria in the five dug wells by showing a metallic green color indicator.
