Classification of Red Blood Cells Using Time-Distributed Convolutional Neural Networks from Simulated Videos

The elasticity of red blood cells (RBCs) plays a vital role in their efficient movement through blood vessels, facilitating the transportation of oxygen within the bloodstream. However, various diseases significantly impact RBC elasticity, making it an important parameter for diagnosing and monitoring health conditions. In this study, we propose a novel approach to determine RBC elasticity by analyzing video recordings and using a convolutional neural network (CNN) for classification. Due to the scarcity of available blood flow recordings, computer simulations based on a numerical model are employed to generate a substantial amount of training data. The simulation model incorporates the representation of RBCs as elastic objects within a fluid flow, allowing for a detailed understanding of their behavior. We compare the performance of different CNN architectures, including ResNet and EfficientNet, for video classification of RBC elasticity. Our results demonstrate the potential of using CNNs and simulation-based data for the accurate classification of RBC elasticity.

An automated neural network-based stage-specific malaria detection software using dimension reduction: The malaria microscopy classifier

Due to climate change and the COVID-19 pandemic, the number of malaria cases and deaths, caused by the Plasmodium genus, of which P. falciparum is the most common and lethal to humans, increased between 2019 and 2020. Reversing this trend and eliminating malaria worldwide requires improvements in malaria diagnosis, in which artificial intelligence (AI) has recently been demonstrated to have a great potential. One of the main reasons for the use of neural networks (NNs) is the time saving through automatising the process and the elimination of human error. When classifying with two-dimensional images of red blood cells (RBCs), the number of parameters fitted by the NN for the classification of RBCs is extremely high, which strongly influences the performance of the network, especially for training sets of moderate size. The complicated handling of malaria culturing and sample preparation does not only limit the efficiency of NNs due to small training sets, but also because of the uneven distribution of red blood cell (RBC) categories. To boost the performance of microscopy techniques in malaria diagnosis, our approach aims at resolving these drawbacks by reducing the dimension of the input data and by data augmentation, respectively. We assess the performance of our approach on images recorded by light (LM), atomic force (AFM), and fluorescence microscopy (FM). Our tool, the Malaria Stage Classifier, provides a fast, high-accuracy recognition by (1) identifying individual RBCs in multi-cell microscopy images, (2) extracting characteristic one-dimensional cross-sections from individual RBC images. These cross-sections are selected by a simple algorithm to contain key information about the status of the RBCs and are used to (3) classify the malaria blood stages. A Machine-Learning Derived Red Blood Cell Morphology Tool Enables Differential Diagnosis and Novel Single-Cell Analyses

Microscopic examination of red blood cell (RBC) morphology in a peripheral blood smear is a standard clinical test with broad utility even in resource-limited settings. Yet this analysis remains subjective, semi-quantitative, and low throughput, limiting its potential in clinical practice and translational research. Prior attempts to develop quantitative RBC morphology tools have been hampered by limited scope, reproducibility challenges, and sparse clinical validation. Here, we present a novel, open-source machine-learning approach leveraging durable geometric features to quantify clinically relevant RBC morphologies-elliptocytes, microcytes, macrocytes, schistocytes, sickle cells, spiculated cells, and teardrop cells-and generate an RBC differential ('RBC-diff'), analogous to the widely used white cell differential. Analysis of >300 thousand smear images across two medical centers shows algorithm concordance with expert human morphology estimates at both the individual cell and full-field smear level, and when compared to standard clinical grading of smears. This algorithm illustrates high intra-sample reproducibility while providing greatly improved speed and resolution comparative to current standards. To test the utility of this new tool we examined: (i) diagnostic signatures in the RBC differential, (ii) large-scale cohort prognostics, (iii) single-cell mechanistic studies, and (iv) an animal model of hematologic disease. Diagnostic RBC differential signatures were discovered by applying the RBC-diff to a cohort of patients with thrombotic microangiopathy (TMA). In particular, the RBC-diff facilitated rapid identification and differential diagnosis of patients with thrombotic thrombocytopenic purpura (TTP) or hemolytic uremic syndrome (HUS), providing greater specificity than manual review (72% vs. 41%, p < 0.001), while maintaining high sensitivity (94-100%). Because TTP is a life-threatening disorder requiring immediate therapy, this new rapid diagnostic tool may provide significant benefit, as existing gold-standard diagnostic tests for TTP are slow (hours to days). Large-scale cohort prognostic value was uncovered through an analysis of smears from a general cohort of 58,950 inpatients that found that elevated schistocyte counts are associated with increased all-cause mortality (9.5% six-month mortality for >1% schistocytes vs. 4.7% for schistocytes < 0.5%, p < 0.001), after adjusting for patient demographics, comorbidities, and manual smear review flags. This tool also allowed for analysis of morphologically annotated single-cell volume distributions, providing insights into how dysmorphology can affect mean corpuscular volume (MCV) and the red cell distribution width (RDW); note that current technology provides only whole-population measures for these variables. For example, in iron-deficient patients who underwent intravenous iron transfusions, this technique highlighted that successful hematologic resolution involved volume increases for all cell types, rather than an isolated reduction in microcyte levels. Finally, without any retraining the model was successfully applied to animal-derived blood smears, highlighting expected differences between a wild-type (WT) and sickle cell disease mouse model (SCD) (0% sickle cells for WT, 1.2% for SCD), demonstrating potential value for basic research investigations. Usage of the red cell differential has the potential to greatly benefit hematologic research and clinical practice. Here we present the RBC-diff, an extensively validated method for efficient calculation of this differential from commonly collected blood smear images. As a resource to the community, we provide open-source code for calculating the RBC-diff. We also provide a repository of 5000 individual RBC images, and 50 full-field smears, each manually labelled by a team of expert pathologists, to facilitate further developments of improved computational methods of RBC classification. Reducing data dimension boosts neural network-based stage-specific malaria detection

Although malaria has been known for more than 4 thousand years1, it still imposes a global burden with approx. 240 million annual cases2. Improvement in diagnostic techniques is a prerequisite for its global elimination. Despite its main limitations, being time-consuming and subjective, light microscopy on Giemsa-stained blood smears is still the gold-standard diagnostic method used worldwide. Autonomous computer assisted recognition of malaria infected red blood cells (RBCs) using neural networks (NNs) has the potential to overcome these deficiencies, if a fast, high-accuracy detection can be achieved using low computational power and limited sets of microscopy images for training the NN. Here, we report on a novel NN-based scheme that is capable of the high-speed classification of RBCs into four categories—healthy ones and three classes of infected ones according to the parasite age—with an accuracy as high as 98%. Importantly, we observe that a smart reduction of data dimension, using characteristic one-dimensional cross-sections of the RBC images, not only speeds up the classification but also significantly improves its performance with respect to the usual two-dimensional NN schemes. Via comparative studies on RBC images recorded by two additional techniques, fluorescence and atomic force microscopy, we demonstrate that our method is universally applicable for different types of microscopy images. This robustness against imaging platform-specific features is crucial for diagnostic applications. Our approach for the reduction of data dimension could be straightforwardly generalised for the classification of different parasites, cells and other types of objects.

Red Blood Cell Classification Based on Attention Residual Feature Pyramid Network.

Clinically, red blood cell abnormalities are closely related to tumor diseases, red blood cell diseases, internal medicine, and other diseases. Red blood cell classification is the key to detecting red blood cell abnormalities. Traditional red blood cell classification is done manually by doctors, which requires a lot of manpower produces subjective results. This paper proposes an Attention-based Residual Feature Pyramid Network (ARFPN) to classify 14 types of red blood cells to assist the diagnosis of related diseases. The model performs classification directly on the entire red blood cell image. Meanwhile, a spatial attention mechanism and channel attention mechanism are combined with residual units to improve the expression of category-related features and achieve accurate extraction of features. Besides, the RoI align method is used to reduce the loss of spatial symmetry and improve classification accuracy. Five hundred and eighty eight red blood cell images are used to train and verify the effectiveness of the proposed method. The Channel Attention Residual Feature Pyramid Network (C-ARFPN) model achieves an mAP of 86%; the Channel and Spatial Attention Residual Feature Pyramid Network (CS-ARFPN) model achieves an mAP of 86.9%. The experimental results indicate that our method can classify more red blood cell types and better adapt to the needs of doctors, thus reducing the doctor's time and improving the diagnosis efficiency.

Urine red blood cell classification based on Siamese Network

Urine examination is an important examination commonly used in medical in vitro examination. The morphological classification of red blood cells in urine plays an important role in the diagnosis of hematuria and renal diseases. In order to improve the accuracy and efficiency of urine red blood cell classification, a urinary red blood cell classification algorithm based on Siamese Network of dual-mode contrastive loss function was proposed. In the dual-mode contrastive loss function, the Cosine Similarity measure and a weight factor were added as the constraint of Euclidean distance in the Contrastive Loss function, which improves the similarity judgment ability of the Contrastive Loss function for the two input eigenvectors. The experimental results show that the Cosine Similarity measure has a positive effect on the Contrastive Loss function, and can effectively improve the accuracy of the whole classification model.

Classification of red blood cell aggregation using empirical wavelet transform analysis of ultrasonic radiofrequency echo signals

Grading red blood cell (RBC) aggregation is important for the early diagnosis and prevention of related diseases such as ischemic cardio-cerebrovascular disease, type II diabetes, deep vein thrombosis, and sickle cell disease. In this study, a machine learning technique based on an adaptive analysis of ultrasonic radiofrequency (RF) echo signals in blood is proposed, and its feasibility for classifying RBC aggregation is explored. Using an adaptive empirical wavelet transform (EWT) analysis, the ultrasonic RF signals are decomposed into a series of empirical mode functions (EMFs); then, dominant empirical mode functions (DEMFs) are selected from the series. Six statistical characteristics, including the mean, variance, median, kurtosis, root mean square (RMS), and skewness are calculated for the locally normalized DEMFs, aiming to form primary feature vectors. Random forest (RDF) and support vector machine (SVM) classifiers are trained with the given feature vectors to obtain prediction models for RBC classification. Ultrasonic RF echo signals are acquired from five groups of six types of porcine blood samples with average numbers of aggregated RBCs of 1.04, 1.20, 1.83, 2.31, 2.72, and 4.28, respectively, to test the classification performance of the proposed method. The best subset with regard to the variance, kurtosis, and RMS is determined according to the maximum accuracy based on the RDF and SVM classifiers. The classification accuracies are 84.03 ± 3.13% for the RDF classifier, and 85.88 ± 2.99% for the SVM classifier. The mean classification accuracy of the SVM classifier is 1.85% better than that of the RDF classifier. In conclusion, the machine learning method is useful for the discrimination of varying degrees of RBC aggregation, and has potential for use in characterizing and monitoring the RBC aggregation in vessels.

Red Blood Cell Classification Using Image Processing and CNN

In the medical field, the analysis of the blood sample of the patient is a critical task. Abnormalities in blood cells are accountable for various health issues. Red blood cells (RBCs) are one of the major components of blood. Classifying the RBC can allow us to diagnose different diseases. The traditional, time-consuming technique of visualizing RBC manually under the microscope, is a tedious task and may lead to wrong interpretation because of the human error. The various health conditions can change the shape, texture, and size of normal RBCs. The proposed method has involved the use of image processing to classify the RBCs with the help of convolution neural networks. The algorithm can extract the feature of each segmented cell image and classify it into 9 various types. Images of blood slides were collected from the hospital. The overall accuracy was 98.5%. The system has been developed to provide accurate and fast results that can save patients’ lives.

Red blood cell classification in lensless single random phase encoding using convolutional neural networks.

Rapid cell identification is achieved in a compact and field-portable system employing single random phase encoding to record opto-biological signatures of living biological cells of interest. The lensless, 3D-printed system uses a diffuser to encode the complex amplitude of the sample, then the encoded signal is recorded by a CMOS image sensor for classification. Removal of lenses in this 3D sensing system removes restrictions on the field of view, numerical aperture, and depth of field normally imposed by objective lenses in comparable microscopy systems to enable robust 3D capture of biological volumes. Opto-biological signatures for two classes of animal red blood cells, situated in a microfluidic device, are captured then input into a convolutional neural network for classification, wherein the AlexNet architecture, pretrained on the ImageNet database is used as the deep learning model. Video data was recorded of the opto-biological signatures for multiple samples, then each frame was treated as an input image to the network. The pre-trained network was fine-tuned and evaluated using a dataset of over 36,000 images. The results show improved performance in comparison to a previously studied Random Forest classification model using extracted statistical features from the opto-biological signatures. The system is further compared to and outperforms a similar shearing-based 3D digital holographic microscopy system for cell classification. In addition to improvements in classification performance, the use of convolutional neural networks in this work is further demonstrated to provide improved performance in the presence of noise. Red blood cell identification as presented here, may serve as a key step toward lensless pseudorandom phase encoding applications in rapid disease screening. To the best of our knowledge this is the first report of lensless cell identification in single random phase encoding using convolutional neural networks.

Automated Semantic Segmentation of Red Blood Cells for Sickle Cell Disease.

Red blood cell (RBC) segmentation and classification from microscopic images is a crucial step for the diagnosis of sickle cell disease (SCD). In this work, we adopt a deep learning based semantic segmentation framework to solve the RBC classification task. A major challenge for robust segmentation and classification is the large variations on the size, shape and viewpoint of the cells, combining with the low image quality caused by noise and artifacts. To address these challenges, we apply deformable convolution layers to the classic U-Net structure and implement the deformable U-Net (dU-Net). U-Net architecture has been shown to offer accurate localization for image semantic segmentation. Moreover, deformable convolution enables free-form deformation of the feature learning process, thus making the network more robust to various cell morphologies and image settings. dU-Net is tested on microscopic red blood cell images from patients with sickle cell disease. Results show that dU-Net can achieve highest accuracy for both binary segmentation and multi-class semantic segmentation tasks, comparing with both unsupervised and state-of-the-art deep learning based supervised segmentation methods. Through detailed investigation of the segmentation results, we further conclude that the performance improvement is mainly caused by the deformable convolution layer, which has better ability to separate the touching cells, discriminate the background noise and predict correct cell shapes without any shape priors.

A deep learning AlexNet model for classification of red blood cells in sickle cell anemia

Sickle cell anemia (SCA) is a serious hematological disorder, where affected patients are frequently hospitalized throughout a lifetime and even can cause death. The manual method of detecting and classifying abnormal cells of SCA patient blood film through a microscope is time-consuming, tedious, prone to error, and require a trained hematologist. The affected patient has many cell shapes that show important biomechanical characteristics. Hence, having an effective way of classifying the abnormalities present in the SCA disease will give a better insight into managing the concerned patient's life. This work proposed algorithm in two-phase firstly, automation of red blood cells (RBCs) extraction to identify the RBC region of interest (ROI) from the patient’s blood smear image. Secondly, deep learning AlexNet model is employed to classify and predict the abnormalities presence in SCA patients. The study was performed with (over 9,000 single RBC images) taken from 130 SCA patient each class having 750 cells. To develop a shape factor quantification and general multiscale shape analysis. We reveal that the proposed framework can classify 15 types of RBC shapes including normal in an automated manner with a deep AlexNet transfer learning model. The cell's name classification prediction accuracy, sensitivity, specificity, and precision of 95.92%, 77%, 98.82%, and 90% were achieved, respectively.

Implementation of Convolutional Neural Network in the classification of red blood cells have affected of malaria

Malaria is a disease caused by plasmodium which attacks red blood cells. Diagnosis of malaria can be made by examining the patient's red blood cells using a microscope. Convolutional Neural Network (CNN) is a deep learning method that is growing rapidly. CNN is often used in image classification. The CNN process usually requires considerable resources. This is one of the weaknesses of CNN. In this study, the CNN architecture used in the classification of red blood cell images is LeNet-5 and DRNet. The data used is a segmented image of red blood cells and is secondary data. Before conducting the data training, data pre-processing and data augmentation from the dataset was carried out. The number of layers of the LeNet-5 and DRNet models were 4 and 7. The test accuracy of the LeNet-5 and DrNet models was 95% and 97.3%, respectively. From the test results, it was found that the LeNet-5 model was more suitable in terms of red blood cell classification. By using the LeNet-5 architecture, the resources used to perform classification can be reduced compared to previous studies where the accuracy obtained is also the same because the number of layers is less, which is only 4 layers

Deep Learning Models for Classification of Red Blood Cells in Microscopy Images to Aid in Sickle Cell Anemia Diagnosis

Sickle cell anemia, which is also called sickle cell disease (SCD), is a hematological disorder that causes occlusion in blood vessels, leading to hurtful episodes and even death. The key function of red blood cells (erythrocytes) is to supply all the parts of the human body with oxygen. Red blood cells (RBCs) form a crescent or sickle shape when sickle cell anemia affects them. This abnormal shape makes it difficult for sickle cells to move through the bloodstream, hence decreasing the oxygen flow. The precise classification of RBCs is the first step toward accurate diagnosis, which aids in evaluating the danger level of sickle cell anemia. The manual classification methods of erythrocytes require immense time, and it is possible that errors may be made throughout the classification stage. Traditional computer-aided techniques, which have been employed for erythrocyte classification, are based on handcrafted features techniques, and their performance relies on the selected features. They also are very sensitive to different sizes, colors, and complex shapes. However, microscopy images of erythrocytes are very complex in shape with different sizes. To this end, this research proposes lightweight deep learning models that classify the erythrocytes into three classes: circular (normal), elongated (sickle cells), and other blood content. These models are different in the number of layers and learnable filters. The available datasets of red blood cells with sickle cell disease are very small for training deep learning models. Therefore, addressing the lack of training data is the main aim of this paper. To tackle this issue and optimize the performance, the transfer learning technique is utilized. Transfer learning does not significantly affect performance on medical image tasks when the source domain is completely different from the target domain. In some cases, it can degrade the performance. Hence, we have applied the same domain transfer learning, unlike other methods that used the ImageNet dataset for transfer learning. To minimize the overfitting effect, we have utilized several data augmentation techniques. Our model obtained state-of-the-art performance and outperformed the latest methods by achieving an accuracy of 99.54% with our model and 99.98% with our model plus a multiclass SVM classifier on the erythrocytesIDB dataset and 98.87% on the collected dataset.

Red blood cell classification on thin blood smear images for malaria diagnosis

Parasite detection is important for the diagnosis of many blood-borne diseases including malaria. As part of a program to develop a fast, accurate, and affordable automatic device for diagnosing malaria, a critical step is to automatically classify individual red blood cells in thin blood smear images. To automatically recognize malaria parasites in an image, this paper presents a red blood cell classification study for malaria diagnosis. To diagnose malaria, the threshold-based segmentation is implemented using the Otsu’s method succeeded by the distance transform and statistical classifier. The methods are applied to red blood cell images obtained from Kaggle. These experimental results show that the classification recognizes malaria parasite with 94.60% accuracy, 96.20% specificity, and 93% sensitivity.

The most common abnormal RBCs are considered as anemic. The RBC features are sufficient to identify the type of anemia and the disease that caused it. Therefore, we found that several features extracted from RBCs in the blood smear images are not significant for classification when observed independently but are significant when combined with other features. The number of feature vectors is reduced from 271 to 8 as time resuming in training and accuracy percentage increased to 98%.

Diagnosis of Malaria Using Wavelet Coefficients and Dynamic Time Warping

Malaria remains one of the world’s most deadly infectious disease and arguably, the greatest menace to modern society in terms of morbidity and mortality. Demand of time is to identify the methods, which can diagnose the disease and are economical and accurate. In this work, we have proposed a technique based on scientific computations. The aim is to provide a method which can do classification of Red Blood Cells (RBCs) into the malaria infected and healthy ones. The images of RBCs are captured using low cost digital holographic interferometric microscope (DHIM), these images are pre-processed for noise removal and then used for computing the discrete wavelet coefficients (DWC). The DWC are then used, as feature vectors, in dynamic time warping algorithm to find similarities or dissimilarities, between the RBCs. This allows the classification of RBCs into healthy or malaria infected. The proposed method shows excellent results for classification of RBCs for diagnosis of disease. Such automated detection methods, using low cost DHIM devices and minimal human intervene, will be good aid to medical area and are demand of time.

Non-invasive analysis of stored red blood cells using diffuse resonance Raman spectroscopy.

A method to acquire the Raman spectra of sub-surface components using diffusely focused radiation in a microscope sampling configuration is described. This procedure generates Raman scattering at various sample depths by producing a converging beam at the back aperture of the objective lens. This method requires illumination of the sample with a defocused laser, while simultaneously increasing the number of CCD pixels that are binned along the spatial axis of the detector. We applied this diffuse sampling method to the analysis of stored red blood cells (RBCs). During storage, biochemical changes to RBCs occur (the "storage lesion"). However, there are no existing non-invasive methods to assess this. We evaluated the instrumental parameters needed to maximize the diffusely scattered signal, including pixel binning, slit width, and bandwidth. We demonstrated the effectiveness of this diffuse resonance Raman spectroscopy (DRRS) method by detecting RBCs through a blood bag segment (1 mm wall thickness). We directly compared the DRRS method to the more common stand-off Raman spectroscopy (SORS) method using both 633 nm and 785 nm excitation. Time-dependent DRRS spectra were used in a multivariate model for classification of RBCs in polymer segments by storage age. Young (6-8 day) RBCs were differentiated from old (35-40) RBCs with 100% sensitivity and 98.5% selectivity. These data indicated that DRRS is a promising, non-invasive technique for acquiring the spectra of sub-surface components, and is particularly applicable when the underlying sample can be resonantly enhanced.

A deep convolutional neural network for classification of red blood cells in sickle cell anemia.

Sickle cell disease (SCD) is a hematological disorder leading to blood vessel occlusion accompanied by painful episodes and even death. Red blood cells (RBCs) of SCD patients have diverse shapes that reveal important biomechanical and bio-rheological characteristics, e.g. their density, fragility, adhesive properties, etc. Hence, having an objective and effective way of RBC shape quantification and classification will lead to better insights and eventual better prognosis of the disease. To this end, we have developed an automated, high-throughput, ex-vivo RBC shape classification framework that consists of three stages. First, we present an automatic hierarchical RBC extraction method to detect the RBC region (ROI) from the background, and then separate touching RBCs in the ROI images by applying an improved random walk method based on automatic seed generation. Second, we apply a mask-based RBC patch-size normalization method to normalize the variant size of segmented single RBC patches into uniform size. Third, we employ deep convolutional neural networks (CNNs) to realize RBC classification; the alternating convolution and pooling operations can deal with non-linear and complex patterns. Furthermore, we investigate the specific shape factor quantification for the classified RBC image data in order to develop a general multiscale shape analysis. We perform several experiments on raw microscopy image datasets from 8 SCD patients (over 7,000 single RBC images) through a 5-fold cross validation method both for oxygenated and deoxygenated RBCs. We demonstrate that the proposed framework can successfully classify sickle shape RBCs in an automated manner with high accuracy, and we also provide the corresponding shape factor analysis, which can be used synergistically with the CNN analysis for more robust predictions. Moreover, the trained deep CNN exhibits good performance even for a deoxygenated dataset and distinguishes the subtle differences in texture alteration inside the oxygenated and deoxygenated RBCs.

Extreme learning machine based microscopic red blood cells classification

The digitalization of blood slides introduced pathology to a new era. Despite being the most powerful prognostic tool; automated analysis of microscopic blood smear images is still not used in routine clinical practices as manual pathological image analysis methods are still in use that is tedious, time consuming and subjective to technician dependent variation, furthermore it also needs training and skills. In this work, we present novel method based on extreme machine learning approach for the classification of red blood cells (RBC) images. Segmentation of RBC is initiated with statistical based thresholding to retrieve those pixels which are most relevant to RBC followed by Fuzzy C-means for the image segmentation and boundary detection. Different texture and geometrical features are extracted for the classification of normal and abnormal cells. The classification technique is rigorously evaluated against the dataset to evaluate the accuracy of classifier. We have compared the results with state of the art techniques. So far the proposed technique has produced more promising results as compared to the existing techniques.
