Antibacterial activity on methicillin-resistant Staphylococcus aureus (MRSA) and antioxidant properties of silver nanoparticles synthesized by Hunteria umbellata (K. Schum.) seeds

Silver nanoparticles (AgNPs) have gained significant attention over the years due to their unique physicochemical properties and diverse applications in various areas including medicine, electronics, and so on. Hunteria umbellata (HU), is a glabrous tree native to West Africa that belongs to the Apocynaceae family with various medicinal properties. The medicinal applications; anti-microbial and anti-oxidant properties of synthesized and characterized silver nanoparticles by using HU seeds, were studied. Aqueous and ethanol extracts of HU were obtained for the synthesis process. UV-Vis spectra analysis was used to reveal the distinct absorption patterns for AgNPs synthesized by ethanol and aqueous extracts. FTIR spectra exhibited characteristic transmittance peaks, indicating the presence of functional groups such as hydroxyl groups. XRD analysis confirmed the crystalline nature of AgNPs with identifiable peaks at specific planes while SEM/EDX was used to provide insights into the size distribution and morphology of AgNPs, reinforcing the data from other characterization methods. Medicinal properties were assessed through antibacterial assays using Methicillin-Resistant Staphylococcus aureus (MRSA) and DPPH radical scavenging activity, showcasing the potential biomedical applications of AgNPs-HU complex. Comparative studies with standard compounds like ascorbic acid validated their efficacy as antioxidants. The findings of this study suggest promising antibacterial and antioxidant properties of AgNPs-HU complex synthesized by HU seeds extracts and also contribute to the understanding of nanomedicine and underscore the potential of green synthesis for biomedical applications.

Antibacterial Activity of Silver Nanoparticles Prepared from Camellia Sinensis Extracts in Multi-Drug Resistant Staphylococcus Aureus.

The nano-method has been used as a technique for creating novel, non-traditional antimicrobial agents. This effective method for treating infectious diseases has many advantages over conventional antibiotics, including increased efficacy against species that have developed drug resistance, and the ability to circumvent the development of resistance that disrupts a number of biological pathways. As a result, the objective of this study was to synthesize and characterize silver nanoparticles using phenolic compounds obtained from Camellia sinensis . The nanoparticles were then used as antibacterial agents on the multidrug resistant Staphylococcus aureus, as well as, biofilm formation mechanism were also investigated. Ten isolates of Staphylococcus aureus were acquired from the labs of the University of Baghdad's Genetic Engineering and Biotechnology Institute. The VITEK-2 system was used to confirm the diagnosis. Aqueous and methanolic extracts of Camellia sinensis leaves were used to create silver nanoparticles and obtain CAgNPs, which were then characterized using Atomic Fluorescence Microscopy (AFM), X-ray diffractometer (XRD), and Zeta potential analyzers. The extracts were put through a series of assays, including High-performance liquid chromatography (HPLC), antibacterial activity assessments, and the microtiter plate method to determine the lowest inhibitory concentration (MIC) and antibiofilm formation. Both aqueous and methanolic extracts containing silver nanoparticles included spherical nanoparticles that may be single or combined. The HPLC results showed the presence of two phenolic compounds (gallic acid and caffeine) by comparing their retention durations to those of the reference compounds. The results of the antibacterial activity of (CAgNPs) showed that the methanolic (CAgNPs) extract was more effective than the aqueous (CAgNPs) extract, producing inhibitory zones of 15.67 ± 0.58 and 20.33 ± 0.58 mm, at 375 and 750 ppm respectively, when compared to the aqueous (CAgNPs) extract, which produced inhibitory zones of 12.33 ± 0.58 and 15.67 ± 0.58 mm, respectively. The MIC result also showed that the CAgNPs methanolic extract was more effective than the CAgNPs aqueous extract. The MIC of the CAgNPs methanolic extract on S. aureus isolates were 11.718 and 23.43 µg/ml, while the MIC of the CAgNPs aqueous extract on all S. aureus isolates were 46.87 µg/ml except isolate No. 3 and 6 which was 11.718 and 93.75 µg/ml respectively. Additionally, The anti-biofilm in S. aureus was increased when CAgNPs methanolic extract were used compared with the CAgNPs aqueous extract, the CAgNPs methanolic extract inhibited 80%, 90% and 100% of the biofilm formation of S. aureus in 23.43, 46.87 and 93.75 µg/ml respectively, while the anti-biofilm activity of the CAgNPs aqueous extract on S. aureus isolates was 80% and 100% of the biofilm formation in 46.87 and 93.75 µg/ml respectively. The methanolic and aqueous leaves extracts from Camellia sinensis are a successful method for producing CAgNPs. The synthesized CAgNPs also have significant antibacterial activity against S. aureus, depending on the concentrations, and inhibit S. aureus biofilm formation.

Asymmetric flow field-flow fractionation for comprehensive characterization of hetero-aggregates made of nano-silver and extracellular polymeric substances

The present study explores the capability of asymmetrical flow field-flow fractionation (AF4) coupled online with diode array (DAD), fluorescence detectors (FLD), multi-angle light scattering (MALS) and dynamic light scattering (DLS) to characterize silver nanoparticles (nAg) hetero-aggregates formed with diatoms derived extracellular polymeric substances (EPS). The content of EPS varied from 10.5 to 105 mgC L-1 and nAg were dispersed at 4 mg L-1 in a freshwater medium. Good recoveries (∼ 76.9 ± 8.4 %) of nAg-EPS were obtained from AF4-DAD signals, but an anomalous elution was observed as EPS concentration increased: AF4 retention times decreased despite average gyration radii measured by MALS for nAg-EPS increased (from 16 nm to 24 nm), which suggests a change in the aggregation state, as evaluated by UV–Vis scans obtained from DAD. A regular Brownian relaxation of nAg-EPS was proven for each EPS concentration using these 2 detectors. The comparison of on-line and batch DLS measurements validated in addition, that no (dis)aggregation occurs upon injections. After a thorough comparison with classical AF4 using standards, the frit-inlet-AF4 was used. Slightly higher recovery (79.6 ± 4.6 %) was obtained but similar deviation of nAg-EPS elution occurred, excluding the implication of membrane differential fouling of nAg-EPS /conditioning effects of EPS. The investigation of physico-chemical parameters controlling the Brownian relaxation of nAg-EPS suggests the influence of nAg-EPS structure and EPS loading. This study demonstrates the suitable use of AF4 coupled to multiple detectors to probe-out eco-corona formation and characterize polydisperse systems containing NPs, EPS and their hetero-aggregates in freshwaters, even under a non-ideal size-fractionation scenario.

Antimicrobial, anticancer and photocatalytic dye degradation activities of silver nanoparticles phytosynthesized from Secamone emetica aqueous leaf extract

Secamone emetica (Family: Apocynaceae) is traditionally used for several ailments among indigenous people due to the bioactive components present in it. Nanomedicine has undergone impressive modifications and the development of new drugs with significant healthcare outcomes. The purpose of the present study was to green synthesize and characterize silver nanoparticles (Ag-NPs) using aqueous leaf extract of S. emetica, and to study their in vitro antimicrobial, anticancer and photocatalytic dye degradation activities. Various spectroscopic and microscopic analytical tools were used to characterize the Ag-NPs. Selective G+ve, G−ve bacteria and fungal species were used to test the Ag-NPs antibacterial and antifungal activity. Anticancer efficacy of the Ag-NPs was appraised by MTT test using MDA-MB 231 human breast cancer cells. Degradation of Rhodamine B dye by the Ag-NPs under visible light was assessed. The characterization studies confirmed the formation of Ag-NPs which were crystalline cubical symmetry with an average size of 26.69 nm. Dose dependent antimicrobial activity displayed a maximum zone of inhibition of 22.39 mm against E. coli followed by 21.28 mm against S. aureus at 100 µL/mL concentration of Ag-NPs. The IC50 value of synthesized Ag-NPs on MDA-MB 231 cell lines was 0.5 μg/mL. The Ag-NPs catalyst degraded 96 % of Rhodamine B dye in 150 min under visible light. Phytosynthesized Ag-NPs showed strong antimicrobial and anticancer properties and also revealed excellent dye degradation capacity. It is possible to further utilize the biosynthesized Ag-NPs as a possible source of antimicrobial, anticancer, and dye-degradation activities.

Chitosan Coupled Silver Nanoparticles Electrocatalyst Synthesis and Characterization

This study sought to find a suitable technique for synthesizing and characterizing silver nanoparticles due to its applications in various fields such as nanotechnology and medical. The synthesized silver nanoparticles were characterized using SEM microscopy, FT-IR and UV-Vis spectroscopy. The maximum absorbance from the UV-Vis spectrophotometer showed the formation of silver nanoparticles with λ max of 418 nm. The FTIR analysis envisaged a strong symmetrical stretching at 1400 cm-1 to 1200 cm-1 with the major peaks recorded at 1390 cm-1 and 1380 cm-1, indicating the presence of nitro components in the sample. The signal at 1658 cm−1 corresponded to the amide (C=O) bonds stretching vibrations, at 1089 cm−1 C-O-C bonds and at 564 cm−1 plane with bends NH, plane-out and bends C−O. C-H stretching vibrational signal was noted at 2927 cm−1, and a broader band at 3426 cm−1 with an overlap between the O−H stretching vibration and N−H stretching vibration of the oligosaccharide applied in the capping. The concentration of chitosan on transmittance was done using 0.5%, 1.0%, 1.5% and 2.0% solutions with the results from FT-IR showing elaborate intensity at 2.0 % chitosan capping and the least at 0.5 %. Scanning microscope validated characterized silver nanoparticles morphology at 500nm. The results obtained from SEM depicted a size range of 100 nm with resolutions between 0.5 to 4 nm

New Study on the Effect of Scopoletin and Silver Nanoparticles of Fenugreek Extract as Pancreatic Cancer Inhibition Agents

Pancreatic cancer is a highly aggressive and lethal form of cancer with limited treatment options. Therefore, exploring novel therapeutic strategies is crucial to improve patient outcomes. Researchers are currently investigating the potential therapeutic effects of scopoletin and silver nanoparticles on various diseases, including pancreatic cancer. Specifically, they have synthesized and characterized silver nanoparticles using fenugreek extract and tested different concentrations of these nanoparticles and scopoletin to determine their anti-cancer and anti-angiogenic properties. The aim is to identify the most effective dose that can reduce the levels of cyclooxygenase-2 (COX-2) and lactate dehydrogenase (LDH), which are markers associated with cancer progression. The study was performed in a laboratory setting, dividing the samples into three separate groups. The first group, known as the control group, consisted of 30 samples from healthy individuals of both genders, aged between 23 and 45 years. The second group included 30 male and female patients aged between 45 and 65 years, with stage II pancreatic cancer. Lastly, the third group comprised 30 male and female patients with stage III pancreatic cancer, aged between 52 and 79 years. This study found that, compared to the control group, patients with stage IΙ and stage IΙΙ pancreatic cancer experienced the greatest decrease in enzyme concentration at the concentration of the nanoparticle solution 2 ppm where the Cyclooxygenas-2 enzyme concentrations values were 2.43±0.22ng/ml, 3.04±0.31ng/ml and 1.59±0.06ng/ml, respectively. As for the concentration of the enzyme Lactate dehydrogenase, it was 1.20±0.02ng/ml, 1.14±0.02ng/ml and 0.85±0.18ng/ml respectively. Also, as compared to the control 1.97±0.17ng/ml, 1.72±0.10ng/ml, and 1.22±0.07ng/ml, the concentration of the scopoletin compound solution 8ppm results in the best reduction in COX-2 enzyme concentration As for the concentration of the enzyme LDH, it was 0.93±0.07 ng/ml, 0.97±0.11 ng/ml and 0.71±0.10 ng/ml In individuals with stage IΙ and stage IIΙ PC. The results of this research demonstrate that scopoletin compounds and Ag-NPs derived from Fenugreek extract function as anti-cancer and anti-angiogenesis agents by reducing the levels of cyclooxygenase-2 (COX-2) and lactate dehydrogenase (LDH) enzymes in the blood serum of individuals with stage II and stage III pancreatic carcinoma in Iraq.

Silver nanoparticles biogenically synthesised using Maclurodendron porteri extract and their bioactivities

Silver nanoparticle is widely used in various field including medical, cosmetic, food and industrial purposes due to their unique properties in electrical conductivity, thermal, and biological activities. In the medical field, silver nanoparticles (AgNPs) have been reported to have strong antimicrobial and cytotoxic activities. This study aimed to synthesize and characterize silver nanoparticles (AgNPs) using Maclurodendron porteri (MP) extract and to evaluate the antimicrobial and cytotoxic activities of the synthesised MP-AgNPs. Green method of Ultrasound Assisted Extraction (UAE) was used to extract the leaves of M. porter. Liquid Chromatography -Mass Spectrometry/Quadrupole time-of-flight (LC-MS/QTOF) was used to identify the compounds in the leaf extract of M. porteri. Characterisation of the synthesised nanoparticles involved ultraviolet–visible (UV–Vis), Fourier Transform Infrared (FTIR), scanning electromagnetic microscopy (SEM), Zeta potential Analyzer and Particle Size Analyzer. The cytotoxic assay was conducted on MCF-7 and Caco-2 cell lines by MTT assay. Antimicrobial activity was tested on Gram-negative and Gram-positive bacteria using the disc diffusion method. Based on LC-MS/QTOF analysis, 430 compounds were found. The identified major compounds consist of amino acids, polyphenols, steroids, terpenoids and heterocyclic compounds which possibly act as reducing agents. 1 mM, 5 mM and 10 mM of silver nitrate solution were mixed with the leaf extract to form silver nanoparticles. 1.2 mg/ml of MP-AgNPs were found to have antibacterial activity against B. subtilis, S. aureus, E. coli, and P. aeruginosa with inhibitory zones of 8.0 ± 0.36 mm, 8.5 ± 0.45 mm, 7.5 ± 0.36 mm, and 9.0 ± 0.40 mm respectively. MP-AgNPs showed no cytotoxic activity against Caco-2 and MCF-7 cells. In conclusion, the presence of major amine compounds such as 10,11-dihydro-10,11-dihydroxyprotriptyline and harderoporphyrin in the extract facilitated the synthesis of AgNPs and the nanoparticle showed weak bioactivities in the assay conducted.

Synergistic antibacterial and mosquitocidal effect of Passiflora foetida synthesized silver nanoparticles.

Silver nanoparticles are opted to have various applications in different fields ranging from traditional medicines to culinary items. It is toxic and most effective against bacteria, fungi viruses, parasites, parasite carrying vectors such as mosquitoes and their larvae and other eukaryotic microorganisms at low concentration without any side effects and toxicity to humans. In view of these data, the present research has been investigated by synthesizing silver nanoparticles using 1mM silver nitrate and aqueous extract of Passiflora foetida. The variation of nanoparticles in size and shape concerning the concentration of extract prepared were analysed. The formation of silver nanoparticles was confirmed by colour changing from yellowish green to reddish-brown implicating the surface plasmon resonance. Further, it was concluded by obtaining an absorbance peak at 420 nm using UV-Visible spectrophotometer analysis. FTIR analysis was used to identify the capping ligands, which included alkanes, aromatic groups and nitro compounds. The average grain size of ~12 nm to 14 nm with crystalline phase was revealed by X-ray Diffraction studies. The SEM images depicted the surface morphology with agglomeration; TEM studies showed the shape of nanoparticles as spherical and hexagonal with sizes ranging from 40 nm to 100 nm and EDAX analysis confirmed the presence of elemental silver as the principal constituent. The characterized silver nanoparticles were then tested for synergistic antibacterial effects with tetracycline, and the results show that they are more active against E. coli and S. aureus, but moderately effective against B. cereus and K. pneumoniae . It also had a strong larval and pupal toxic effects on the dengue vector, Aedes aegypti with the highest mortality. As a result, silver nanoparticles could be a viable alternative for a variety of applications.

Synergistic effect of synthesized silver nanoparticles combined with amoxicillin/clavulanic acid against Pseudomonas aeruginosa isolated from clinical samples

Abstract Background: Pseudomonas aeruginosa is one of the most prevalent nosocomial pathogenic microorganisms that affect and cause a life-threatening situation. Objective: The study aimed to investigate the synergistic effect of silver nanoparticles (NPs) with amoxicillin/clavulanic acid antibiotic on P. aeruginosa growth. Materials and Methods: A total of 220 clinical samples were collected. A sample has been subjected to isolation and identification by standard microbiological procedures. The extract of Citrus aurantium was used to synthesize silver NPs. The characterization of silver NPs was achieved at the University of Tehran, by using UV–visible spectrophotometry, X-ray diffraction (XRD), and transmission electron microscope (TEM). The antibacterial activity of AgNPs and combination with amoxicillin/clavulanic acid antibiotic were tested against bacteria using the agar well diffusion method. Results: Out of the 220 samples collected, 33 (15%) isolates were positive for P. aeruginosa. Highest resistance of P. aeruginosa was found to amoxicillin/clavulanic acid in percentage of 100. NPs have been characterized by a UV–visible spectrometer which revealed a broad peak at 426 cm-1. XRD showed the purity of the prepared silver NPs that the particle size was equal to 21.26 nm. TEM measurement showed the presence of sphere-like structures with sizes of 20 nm for regular particles and 40 nm for irregular particles. AgNPs, amoxicillin/clavulanic acid, and mixture (amoxicillin/clavulanic acid + AgNPs) have high inhibition activity of P. aeruginosa in concentration of 100 µg/mL and recorded 21.33 ± 3.06, 13.00 ± 0.00, and 22.00 ± 3.46 mm, respectively. Conclusion: The results supported a green synthesis approach for the synthesis of AgNPs with antimicrobial. P. aeruginosa has synergistic combinations of AgNPs with amoxicillin/clavulanic acid, with a great inhibitory effect on the growth of the bacteria.

Determination of the Cytotoxicity and Antibiofilm Potential Effect of Equisetum arvense Silver Nanoparticles.

This study aimed to synthesize and characterize silver nanoparticles (AgNPs) by green synthesis from Equisetum arvense (Ea) extracts and to investigate their cytotoxicity, antibiofilm activity, and α-glucosidase enzyme inhibition. Diverse characterization techniques were applied to verify the production of nanoparticles. SEM examination confirmed that the size of nanoparticles is in the range of 40-60nm. Also, interactions between silver and natural compounds of plant extract were confirmed through FT-IR and EDX analyses. It was determined that Equisetum arvense silver nanoparticles had antibiofilm activity against three different clinical strains with high biofilm-forming ability. AgNPs reduced the biofilm-forming capacity of clinical A. baumannii isolate with strong biofilm-forming capacity by approximately twofold, while the capacity of clinical K.pneumonaie and E.coli isolates decreased by 1.5 and 1.2 fold, respectively. The α-glucosidase enzyme inhibition potential of the AgNPs, which is determined as 93.50%, was higher than the plant extract with, and the α- 30.37%. MTT was performed to assess whether incubation of nanoparticles with A549 and ARPE-19 cell lines affected their viability, and a dramatic reduction in cell growth inhibition of both A549 and ARPE-19 cells was observed. It has been shown that A549 cells treated with 200 and 150µg/mL nanoparticles had less cell proliferation compared to control cells at 24-h and 48-h incubation time. According to these results, Ea-derived AgNPs appear to have potential anticancer activity against A549 cancer cells. Investigating the effects of green synthesis nanoparticles on microbial biofilm and various tumors may be important for developing new therapies. The outcomes of this study have showed that Ea-AgNPsmay have a high potential both in the treatment of pathogenic strains that form biofilms, as well as in anticancer therapy use.

Facile Synthesis, Characterization, and Antimicrobial Assessment of a Silver/Montmorillonite Nanocomposite as an Effective Antiseptic against Foodborne Pathogens for Promising Food Protection.

Foodborne pathogens can have devastating repercussions and significantly threaten public health. Therefore, it is indeed essential to guarantee the sustainability of our food production. Food preservation and storage using nanocomposites is a promising strategy. Accordingly, the present research's objectives were to identify and isolate a few foodborne pathogens from food products, (ii) synthesize and characterize silver nanoparticles (AgNPs) using wet chemical reduction into the lamellar space layer of montmorillonite (MMT), and (iii) investigate the antibacterial potential of the AgNPs/MMT nanocomposite versus isolated strains of bacteria. Six bacterial species, including Escherichia coli, Salmonella spp., Pseudomonas aeruginosa, Staphylococcus aureus, Listeria monocytogenes, and Bacillus cereus were isolated from some food products (meat, fish, cheese, and vegetables). The Ag/MMT nanocomposite was synthesized and characterized using UV-visible spectroscopy, transmission electron microscopy, particle size analyzer, zeta potential, X-ray diffraction (XRD), and scanning electron microscopy with dispersive energy X-ray (EDX). The antibacterial effectiveness of the AgNPs/MMT nanocomposite further investigated distinct bacterial species using a zone of inhibition assay and microtiter-based methods. Nanoparticles with a narrow dimension range of 12 to 30 nm were identified using TEM analysis. The SEM was employed to view the sizeable flakes of the AgNPs/MMT. At 416 nm, the most excellent UV absorption was measured. Four silver metallic diffraction peaks were found in the XRD pattern during the study, and the EDX spectrum revealed a strong signal attributed to Ag nanocrystals. AgNPs/MMT figured out the powerful antibacterial action. The AgNPs/MMT nanocomposite confirmed outstanding minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) against six isolates of foodborne pathogens, ranging from 15 to 75 µg/mL, respectively. The AgNPs/MMT's antibacterial potential against gram-negative bacteria was noticeably better than gram-positive bacteria. Therefore, the AgNPs/MMT nanocomposite has the potential to be used as a reliable deactivator in food processing and preservation to protect against foodborne pathogenic bacteria. This suggests that the nanocomposite may be effective at inhibiting the growth and proliferation of harmful bacteria in food, which could help to reduce the risk of foodborne illness.

Bio-Fabrication of Silver Nanoparticles Using Citrus aurantifolia Fruit Peel Extract (CAFPE) and the Role of Plant Extract in the Synthesis.

The green synthesis of silver nanoparticles has been proposed as an eco-friendly and cost-effective substitute for chemical and physical methods. The aim of this study was to synthesize and characterize silver nanoparticles using the peel extract of Citrus aurantifolia fruit, and to determine the possible phytochemical constituents' presence in the plant extracts that might be responsible for the synthesis. Citrus aurantifolia fruit peel extraction was followed by phytochemical studies of secondary metabolites, FTIR analysis confirmation of functional groups, and GC-MS analysis. Silver nanoparticles were synthesized through bio-reduction of silver ions (Ag+) to silver nanoparticles using CAFPE and characterized using UV-Vis spectroscopy, HR-TEM, FESEM, EDX, XRD, DLS, and FTIR. The presence of plant secondary metabolites such as alkaloids, flavonoids, tannins, saponins, phenols, terpenoids, and steroids was detected. The FTIR analysis of the extract revealed the presence of functional groups like hydroxyl, carboxyl, carbonyl, amine, and phenyl, whereas the GC-MS analysis indicated presence of chemical compounds such as 1,2,4-Benzenetricarboxylic acid, Fumaric acid, nonyl pentadecyl, and 4-Methyl-2-trimethylsilyloxy-acetophenone, etc., with similar functional groups. The synthesized silver nanoparticle (AgNP) has displayed the characteristics of a surface plasmon resonance (SPR) band peak from 360-405 nm. High resolution transmission electron microscope (HR-TEM) and field emission scan electron microscope (FESEM) confirm polydisperse, spherical shaped, and smooth surface nanoparticles with an average size of 24.023 nm. Energy dispersive X-ray (EDX) analysis further revealed that silver is the most abundant element found in the micrograph of the nanoparticles, and FTIR analysis further confirmed the presence of different functional groups in the surface of the nanoparticle. The XRD analysis also confirmed that the nanoparticles synthesized are crystalline in nature. Based on the findings of this study, it is understood that the variety of natural compounds that are present in plant extracts of Citrus aurantifolia fruit peel can act as both reducing and stabilizing agents for the synthesis of silver nanoparticles. It is, therefore, concluded that Citrus aurantifolia peel extract can be potentially used for the large production of silver nanoparticles for several applications.

In vitro antiplasmodial activity, hemocompatibility and temporal stability of Azadirachta indica silver nanoparticles

Recently green nanotechnology has gained great interest as a promising tool for drug discovery. In the present study, we synthesized and characterized silver nanoparticles (AgNPs) using Azadirachta indica (AI) and evaluated their hemocompatibility and effect against Plasmodium falciparum strains. AI leaves and barks were used for aqueous extracts (AIL and AIB) and AgNPs synthesis. AgNPs were characterized using spectroscopic, diffraction, electron microscopic and electrostatic techniques. Anti-plasmodial and haemolytic activity were assessed following the SYBR Green I fluorescence assay and Miki et al. protocol, respectively. The normalized fluorescence counts were plotted against the log-transformed drug concentration and half-maximal inhibitory concentration (IC50) determined by analyzing the dose–response curves. AgNPs were stored for 120 days at room temperature-RT, +4 °C and −20 °C and subsequently their stability was evaluated by spectroscopy. Both NPs were predominantly spheroidal, crystalline in nature, stable, well dispersed with mean size of 13.01 nm for AIL-NPs and 19.30 nm for AIB-NPs and exhibited good antiplasmodial activity against 3D7 and RKL9 P. falciparum strains with IC50 of 9.27 µg/mL and 11.14 µg/mL for AIL-NPs, 8.10 µg/mL and 7.87 µg/mL for AIB-NPs, respectively. A. indica contain bioactive phyto-compounds indicating great potential for anti-malarial drug development through green nanotechnology. The AgNPs were structurally stable after 120 days but antiplasmodial activity was considerably affected. A significant haemolytic activity (>25%) was observed with AIL- and AIB-AgNPs at concentrations ≥125 µg/mL.

An Evaluation of Antimicrobial, Anticancer, Anti-Inflammatory and Antioxidant Activities of Silver Nanoparticles Synthesized from Leaf Extract of Madhuca longifolia Utilizing Quantitative and Qualitative Methods.

In the current decade, nanoparticles are synthesized using solvents that are environmentally friendly. A number of nanoparticles have been synthesized at room temperature using water as a solvent, such as gold (Au) and silver (Ag) nanoparticles. As part of nanotechnology, nanoparticles are synthesized through biological processes. Biological methods are the preferred method for the synthesis of inorganic nanoparticles (AgNPs) as a result of their simple and non-hazardous nature. Nanoparticles of silver are used in a variety of applications, including catalysts, spectrally selective coatings for solar absorption, optical objectives, pharmaceutical constituents, and chemical and biological sensing. Antimicrobial agents are among the top uses of silver nanoparticles. In the current study, silver nanoparticles were biologically manufactured through Madhuca longifolia, and their antibacterial activity against pathogenic microorganisms, anticancer, anti-inflammatory, and antioxidant activities were assessed. UV-Vis spectroscopy, XRD (X-ray diffraction), transmission electron microscopy, Zeta Potential, and FTIR were used to characterize silver nanoparticles. The current work describes a cheap and environmentally friendly method to synthesize silver nanoparticles from silver nitrate solution by using plant crude extract as a reducing agent.

Green Synthesis and Characterization of Silver Nanoparticles of Psidium guajava Leaf Extract and Evaluation for Its Antidiabetic Activity.

Diabetes mellitus (DM) and its complications are a severe public health concern due to the high incidence, morbidity, and mortality rates. The present study aims to synthesize and characterize silver nanoparticles (AgNPs) using the aqueous leaf extract of Psidium guajava (PGE) for investigating its antidiabetic activity. Psidium guajava silver nanoparticles (PGAg NPs) were prepared and characterized by various parameters. The in vivo study was conducted using PGE and PGAg NPs in Streptozotocin (STZ)-induced diabetic rats to assess their antidiabetic properties. STZ of 55 mg/kg was injected to induce diabetes. The PGE, PGAg NPs at a dose of 200 and 400 mg/kg and standard drug Metformin (100 mg/kg) were administered daily to diabetic rats for 21 days through the oral route. Blood glucose level, body weight changes, lipid profiles, and histopathology of the rats’ liver and pancreas were examined. In the diabetic rats, PGE and PGAg NPs produced a drastic decrease in the blood glucose level, preventing subsequent weight loss and ameliorating lipid profile parameters. The histopathological findings revealed the improvements in pancreas and liver cells due to the repercussion of PGE and PGAg NPs. A compelling effect was observed in all doses of PGE and PGAg NPs; however, PGAg NPs exhibited a more promising result. Thus, from the results, it is concluded that the synthesized PGAg NPs has potent antidiabetic activity due to its enhanced surface area and smaller particle size of nanoparticles.

Characterization of Asafoetida Resin Silver Nanoparticles and Efficacy of Antioxidant, Antimicrobial Inhibition

Background: In recent years, biological precursor-based synthesis, particularly plant-based green synthesis, has piqued interest in the field of nanotechnology. The plant-based approach is a promising technique for the biosynthesis of nanoparticles. 
 Aim of the study: Biosynthesis nanoparticles (AgNPs) using the ethanolic extract of a resin exuded from Ferula asafoetida (resin) and evaluate its anti actinobacterial, and antioxidant properties. 
 Method: Ferula asafoetida resin was collected and processed for ethanolic extraction by soxhlet apparatus. Preliminary phytoconstituents were quantified by GC MS and NMR. Consequently, the biosynthesis of silver nanoparticles was carried out with ethanolic extract. Characterization studies of synthesized Asafoetida AgNPs were done by UV Vis spec, XRD and SEM. Bioactivity of As-AgNPs was evaluated for anti-actinobacterial activity against selected Streptomyces toxytricini D2, Streptomyces tritici D5 and antioxidants properties were proved by Antioxidant assays. 
 Result: Ferula asafoetida resin ethanolic extract can be successfully used for the biosynthesis of Ag NPs. It showed excellent antioxidant activity, including DPPH and hydroxyl radical scavenging abilities comparable to standard ascorbic acid antioxidants. Characterized silver nanoparticles have moderate antimicrobial properties.
 Conclusion: Currently, the biosynthesis of silver nanoparticles has fostered the development of a new generation of nanoproducts as well as increased scientific promise in the nanomaterial field. Asafoetida based AgNPs as novel materials can be utilized by antioxidant, antimicrobial inhibition

Biosynthesis and characterization of silver nanoparticles using marasmius palmivorus MG717877.1 and their antifungal activity

Silver nanoparticles (AgNPs) were produced utilizing Marasmius palmivorus MG717877.1 in the current investigation. The fungal cell filtrate was used to accomplish external production of silver nanoparticles (AgNPs) from silver nitrate (AgNo3) solution. The aqueous silver (Ag) ions in a 1mM AgNo3 aqueous solution were decreased when it was exposed to fungal cell filtrate, resulting in highly stable AgNPs. When challenged with 1mM AgNo3 solution, the fungal biomass acquired AgNPs on its surface, inside the cytoplasmic membrane, and within the cytoplasm in 72 hours. UV was used to characterize the AgNPs that had been produced. For M.palmivorus MG717877.1, the greatest absorbance of AgNPs was recorded at 400nm in the visible spectrum. Furthermore, the FTIR and SEM analyses of silver nanoparticles on these fungus were used to characterize silver nanoparticles. In addition, we tested the antifungal activity of M.palmivorus MG717877.1 at various concentrations, including 25, 50, 100, and 150 mg/ml. The findings revealed that silver nanoparticles produced in a concentration-dependent manner have strong antifungal activity on isolates. At 150 mg of AgNPs, the highest decrease was seen for this isolate.

Green synthesis and characterization of silver nanoparticles using Eugenia roxburghii DC. extract and activity against biofilm-producing bacteria

The green synthesis of silver nanoparticles (AgNPs) and their applications have attracted many researchers as the AgNPs are used effectively in targeting specific tissues and pathogenic microorganisms. The purpose of this study is to synthesize and characterize silver nanoparticles from fully expanded leaves of Eugenia roxburghii DC., as well as to test their effectiveness in inhibiting biofilm production. In this study, at 0.1 mM concentration of silver nitrate (AgNO3), stable AgNPs were synthesized and authenticated by monitoring the color change of the solution from yellow to brown, which was confirmed with spectrophotometric detection of optical density. The crystalline nature of these AgNPs was detected through an X-Ray Diffraction (XRD) pattern. AgNPs were characterized through a high-resolution transmission electron microscope (HR-TEM) to study the morphology and size of the nanoparticles (NPs). A new biological approach was undertaken through the Congo Red Agar (CRA) plate assay by using the synthesized AgNPs against biofilm production. The AgNPs effectively inhibit biofilm formation and the biofilm-producing bacterial colonies. This could be a significant achievement in contending with many dynamic pathogens.

Ajwa-Dates (Phoenix dactylifera)-Mediated Synthesis of Silver Nanoparticles and Their Anti-Bacterial, Anti-Biofilm, and Cytotoxic Potential

Green nanotechnology is the evolution of cost-effective and environmentally friendly processes for the production of metal-based nanoparticles due to medicinal importance and economic value. The aim of the present study was to biosynthesize and characterize silver nanoparticles (AgNPs) using the seed extract of Ajwa dates (Aw). The anti-bacteriostatic activity of biosynthesized Aw–AgNPs against Gram-positive and Gram-negative bacterial strains was evaluated. The anti-biofilm activity was examined by the tissue culture plate method. Lastly, the anti-cancer potential of Aw–AgNPs was investigated against the human breast cancer cell line HCC712. UV–visible absorption spectra exhibited the plasmon resonance peak at 430 nm, with the solution undergoing rapid color changes that verified the existence of biosynthesized silver nanoparticles in the solution. TEM and SEM images illustrated that the Aw–AgNPs were spherical and between 15 and 80 nm in diameter. The reduction and stabilization of Aw–AgNPs was due to the functional groups present in the biomolecules of the Ajwa seeds, as identified by FTIR. The Aw–AgNPs exhibited significant anti-bacterial activity against all the tested bacterial strains. Moreover, the Aw–AgNPs efficiently hampered the biofilm formation of the bacterial strains and exhibited cytotoxicity at various concentrations. Overall, these findings suggest that biosynthesized Aw–AgNPs may be used as a potential therapeutic formulation against bacterial infections and breast cancer.

Silver Nanoparticles as Chlorhexidine and Metronidazole Drug Delivery Platforms: Their Potential Use in Treating Periodontitis.

PurposePeriodontal disease (PD), defined as oral inflammation caused by dental plaque, is an emerging problem. PD may lead to tooth loss, and treatment options are limited. In this study, we designed, synthesized, and characterized silver nanoparticles (AgNPs) conjugated with chlorhexidine (AgNPs-CHL) or metronidazole (AgNPs-PEG-MET) to determine whether they can be used to treat PDs.Materials and MethodsAgNPs were synthesized and characterized by transmission electron microscopy, UV-vis spectrometry, thermogravimetric analyses, and dynamic light scattering. We determined the safety and the antimicrobial and anti-inflammatory properties of synthesized AgNPs in an in vitro model of periodontitis. Antimicrobial properties were determined by measuring the minimum inhibitory concentration (MIC) and minimum biofilm eradication concentration (MBEC) on reference strains of bacteria and fungi. Human gingival fibroblast (HGF-1), murine macrophage (RAW264.7) and human foetal osteoblast (hFOB1.19) cells were used in the study. Lipopolysaccharide (LPS) was used to induce inflammation. Cytokine levels were measured using an enzyme-linked immunosorbent assay; metalloproteinase expression was measured using Western blotting.ResultsThe synthesized AgNPs were spherical and narrow-dispersed with an average diameter of 13.4 nm ± 3.0 nm in the case of AgNPs-CHL and 3.72 nm ± 0.72 nm in the case of AgNPs-PEG-MET. Both types of AgNPs were active against bacteria and fungi. AgNPs-CHL proved to be a more potent antimicrobial agent, although they were more cytotoxic than AgNPs-PEG-MET; however, both demonstrated beneficial properties in nontoxic concentrations. AgNPs-CHL and AgNPs-PEG-MET decreased the production of proinflammatory cytokines IL-1β, IL-6, IL-8 and TNFα. Both agents also decreased the levels of metalloproteinases MMP3 and MMP8, which may indicate that they will inhibit tissue degradation.ConclusionAgNPs-CHL and AgNPs-PEG-MET may be possible therapeutic options for PD, as they have antibacterial and anti-inflammatory properties. However, to fully understand the potential of AgNPs, our in vitro findings must be evaluated in an in vivo model.
