A role for a cell wall localized glycine‐rich protein in dehydration and rehydration of the resurrection plant Boea hygrometrica

The acquisition of desiccation tolerance in dicotyledonous angiosperms requires the induction of a co-ordinated programme of genetic and biochemical processes during drying and the adaptive mechanisms are primarily protoplasmic in nature. Recent studies have shown that changes in cell wall structure and composition are also important for recovery after drying, however, the molecular mechanisms that underpin these adaptive responses are largely unknown. Here, the desiccation-tolerant plant Boea hygrometrica was used as a model system to investigate the changes in gene expression and cell wall adaptation that take place during extreme dehydration. A cDNA macroarry analysis of dehydration-inducible genes led to the identification of a gene encoding a glycine-rich protein (BhGRP1). The corresponding transcript was up-regulated during drying in B. hygrometrica leaves. In silico analysis revealed that BhGRP1 is targeted to the cell wall and this was confirmed in planta. Morphological changes in the cell wall architecture were also observed during the process of drying and re-watering. Concomitant with this observation, cell wall profiling by Fourier transform infrared spectroscopy indicated that protein levels increased upon desiccation and remained broadly similar upon re-watering. These findings suggest that the deposition of BhGRP1 may play a role in cell wall maintenance and repair during dehydration and rehydration in B. hygrometrica.

Genomic and proteomic analysis of the Alkali-Tolerance Response (AlTR) in Listeria monocytogenes 10403S.

BackgroundInformation regarding the Alkali-Tolerance Response (AlTR) in Listeria monocytogenes is very limited. Treatment of alkali-adapted cells with the protein synthesis inhibitor chloramphenicol has revealed that the AlTR is at least partially protein-dependent. In order to gain a more comprehensive perspective on the physiology and regulation of the AlTR, we compared differential gene expression and protein content of cells adapted at pH 9.5 and un-adapted cells (pH 7.0) using complementary DNA (cDNA) microarray and two-dimensional (2D) gel electrophoresis, (combined with mass spectrometry) respectively.ResultsIn this study, L. monocytogenes was shown to exhibit a significant AlTR following a 1-h exposure to mild alkali (pH 9.5), which is capable of protecting cells from subsequent lethal alkali stress (pH 12.0). Adaptive intracellular gene expression involved genes that are associated with virulence, the general stress response, cell division, and changes in cell wall structure and included many genes with unknown functions. The observed variability between results of cDNA arrays and 2D gel electrophoresis may be accounted for by posttranslational modifications. Interestingly, several alkali induced genes/proteins can provide a cross protective overlap to other types of stresses.ConclusionAlkali pH provides therefore L. monocytogenes with nonspecific multiple-stress resistance that may be vital for survival in the human gastrointestinal tract as well as within food processing systems where alkali conditions prevail. This study showed strong evidence that the AlTR in L. monocytogenes functions as to minimize excess alkalisation and energy expenditures while mobilizing available carbon sources.

Cell wall structural changes in wheat straw pretreated for bioethanol production

BackgroundPretreatment is an essential step in the enzymatic hydrolysis of biomass and subsequent production of bioethanol. Recent results indicate that only a mild pretreatment is necessary in an industrial, economically feasible system. The Integrated Biomass Utilisation System hydrothermal pretreatment process has previously been shown to be effective in preparing wheat straw for these processes without the application of additional chemicals. In the current work, the effect of the pretreatment on the straw cell-wall matrix and its components are characterised microscopically (atomic force microscopy and scanning electron microscopy) and spectroscopically (attenuated total reflectance Fourier transform infrared spectroscopy) in order to understand this increase in digestibility.ResultsThe hydrothermal pretreatment does not degrade the fibrillar structure of cellulose but causes profound lignin re-localisation. Results from the current work indicate that wax has been removed and hemicellulose has been partially removed. Similar changes were found in wheat straw pretreated by steam explosion.ConclusionResults indicate that hydrothermal pretreatment increases the digestibility by increasing the accessibility of the cellulose through a re-localisation of lignin and a partial removal of hemicellulose, rather than by disruption of the cell wall.

Generation of ESTs in Vitis vinifera wine grape (Cabernet Sauvignon) and table grape (Muscat Hamburg) and discovery of new candidate genes with potential roles in berry development

We report the generation and analysis of a total of 77,583 expressed sequence tags (ESTs) from two grapevine ( Vitis vinifera L.) cultivars, Cabernet Sauvignon (wine grape) and Muscat Hamburg (table grape) with a focus on EST sequence quality and assembly optimization. The majority of the ESTs were derived from normalized cDNA libraries representing berry pericarp and seed developmental series, pooled non-berry tissues including root, flower, and leaf in Cabernet Sauvignon, and pooled tissues of berry, seed, and flower in Muscat Hamburg. EST and unigene sequence quality were determined by computational filtering coupled with small-scale contig reassembly, manual review, and BLAST analyses. EST assembly was optimized to better discriminate among closely related paralogs using two independent grape sequence sets, a previously published set of Vitis spp. gene families and our EST dataset derived from pooled leaf, flower, and root tissues of Cabernet Sauvignon. Sequence assembly within individual libraries indicated that those prepared from pooled tissues contributed the most to gene discovery. Annotations based upon searches against multiple databases including tomato and strawberry sequences helped to identify putative functions of ESTs and unigenes, particularly with respect to fleshy fruit development. Sequence comparison among the three wine grape libraries identified a number of genes preferentially expressed in the pericarp tissue, including transcription factors, receptor-like protein kinases, and hexose transporters. Gene ontology (GO) classification in the biological process aspect showed that GO categories corresponding to ‘transport’ and ‘cell organization and biogenesis’, which are associated with metabolite movement and cell wall structural changes during berry ripening, were higher in pericarp than in other tissues in the wine grape studied. The sequence data were used to characterize potential roles of new genes in berry development and composition.

Intracellular Gene Expression Profile of Listeria monocytogenes

Listeria monocytogenes is a gram-positive, food-borne microorganism responsible for invasive infections with a high overall mortality. L. monocytogenes is among the very few microorganisms that can induce uptake into the host cell and subsequently enter the host cell cytosol by breaching the vacuolar membrane. We infected the murine macrophage cell line P388D1 with L. monocytogenes strain EGD-e and examined the gene expression profile of L. monocytogenes inside the vacuolar and cytosolic environments of the host cell by using whole-genome microarray and mutant analyses. We found that approximately 17% of the total genome was mobilized to enable adaptation for intracellular growth. Intracellularly expressed genes showed responses typical of glucose limitation within bacteria, with a decrease in the amount of mRNA encoding enzymes in the central metabolism and a temporal induction of genes involved in alternative-carbon-source utilization pathways and their regulation. Adaptive intracellular gene expression involved genes that are associated with virulence, the general stress response, cell division, and changes in cell wall structure and included many genes with unknown functions. A total of 41 genes were species specific, being absent from the genome of the nonpathogenic Listeria innocua CLIP 11262 strain. We also detected 25 genes that were strain specific, i.e., absent from the genome of the previously sequenced L. monocytogenes F2365 serotype 4b strain, suggesting heterogeneity in the gene pool required for intracellular survival of L. monocytogenes in host cells. Overall, our study provides crucial insights into the strategy of intracellular survival and measures taken by L. monocytogenes to escape the host cell responses.

Cryo-FE-SEM & TEM immuno-techniques reveal new details for understanding white-rot decay of lignocellulose

High-resolution Cryo-Field Emission Scanning Electron Microscopy (HR-Cryo-FE-SEM) and immuno-cytochemistry were used to reveal novel details on the morphological events and spatial distribution of oxidoreductive enzymes during the degradation of birch wood by the white-rot fungi Phlebia radiata and mutant strain P. radiata Cel 26. Cryo-observations of fractured fibres showed degradation across the cell wall by P. radiata (wild) to progress by delamination and removal of concentric orientated aggregates from the secondary S2 cell wall. Decay by P. radiata Cel 26 progressed by removal of materials (lignin and hemicelluloses) between the aggregates (primarily cellulose) that remained even after advanced decay. With both decay patterns, extracellular slime materials were present uniting lumina hyphae with the attacked fibre wall. The extracellular slime material had two morphological forms: viz a fibrillar (often tripartite) and a ‘gel-form’, the former found in discrete bands progressing across the lumen onto the fibre wall. Using TEM immunocytochemistry, laccase, manganese peroxidase (MnP) and diarylpropane enzymes were localized in the periplasmic space of luminal hyphae, in association with the cell membrane, periplasmic vesicles and fungal cell wall. Extracellularly, the three enzymes were found associated with the slime and tripartite membranes and with the birch cell walls at all stages of attack through to middle lamella corner decay. Enzyme distribution was correlated with morphological changes in cell wall structure. The association of extracellular slime with these enzymes and sites of decay strongly suggests a major role for this matrix in fibre cell wall decomposition. To cite this article: G. Developmental changes in cell wall structure of phloem fibres of the bamboo Dendrocalamus asper.

Bamboo culms have excellent physical and mechanical properties, which mainly depend on their fibre content and anatomical structure. One of the features which is known to contribute to the high tensile strength in bamboo is the multilayered structure of the fibre cell wall. The aim of this study was to characterize the development of the layered structure in fibre cell walls of developing and maturing culms of Dendrocalamus asper. Cell wall development patterns were investigated in phloem fibre caps of vascular bundles in the inner culm wall areas of Dendrocalamus asper of three different age classes (<6 months old, 1 year old, 3 years old). A combination of light microscopy and image analysis techniques were employed to measure cell wall thickness and to determine number of cell wall layers, as well as to describe the layering structure of fibre walls. Two-dimensional maps showing the distribution pattern of fibres according to the number of cell wall layers were produced. The cell walls of fibres in phloem fibre caps located in the inner part of the culm wall of D. asper developed rapidly during the first year of growth. Six different fibre types could be distinguished based upon their cell wall layering and all were already present in the young, 1-year-old culm. In the mature stage (3 years of age) the multilayering was independent of the cell wall thickness and even the thinner-walled fibres could have a large number of wall layers. The multilayered nature of cell wall structure varied considerably between individual cells and was not exclusively related to the cell wall thickness. Nevertheless, fibres at the periphery of the fibre bundles and immediately adjacent to the phloem elements exhibited a consistent and high degree of layering in their cell walls. The multilayered structure of fibre cell walls was formed mainly during the first year of growth by the deposition of new wall layers of variable thickness, resulting in a high degree of heterogeneity in the layering patterns amongst individual fibres. A degree of 'order' in the distribution of multilayered fibres within the caps does exist, however, with multilayered cell walls common in fibres adjacent to phloem elements and around the edge of the fibre cap. These findings confirm the observations, primarily in Phyllostachys viridi-glaucescens. The layering structure was not found to be specifically related to the thickness of the cell wall.

Anatomical changes during culm maturation in Bambusa bambos (L.) Voss and Dendrocalamus strictus Nees

The anatomical changes occurring during the maturation of culms were investigated in two common species of Indian bamboos, namely Bambusa bambos (L.) Voss and Dendrocalamus strictus Nees by a comparative study of culm internodal material of different ages between 2 and 60 months. The culm wall consisted of ground parenchyma tissue enclosing a large number of fibro-vascular bundles, which varied in size, structure and abundance in different parts of the culm, both in the axial and radial direction. The main change that occurred during culm maturation was the thickening of cell walls and lignification. Cell-wall thickening and lignification progressed from the outer to the inner parts of the culm wall, and from the culm base towards the top. Within a fibro-vascular bundle, these changes first occurred in fibres contiguous to the vascular tissues and then progressed to outer parts of the bundle. In the peripheral fibrous strands of the bundles, the outermost fibres matured first followed by the inner ones. Cell-wall thickening of fibres was accomplished by addition of lamellae leading to a polylamellate cell-wall structure. In ground parenchyma, although the wall thickening was evident, lamellation was not distinct. These changes in cell-wall structure led to increase in basic density of the culm material. The increase in density was dramatic during the first two years in both B. bambos and D. strictus. It became more gradual during the third year and stabilized thereafter. The moisture content percentage, which showed an inverse relationship with density, declined rapidly during the first two years and reached a stable value in later years. The study suggested that the maturation process of newly emerged culms was rapid during the first two years, and stabilized after the third year in both species.

Are changes of the cell membrane structure causally involved in the aging process?

Lipid peroxidation is recognized by proliferation, wounding, and aging. The connecting link between these different events is a change in cell wall structure, which activates membrane bound phospholipases. These cleave phospholipids. Thus liberated polyunsaturated fatty acids (PUFAs) are substrates for lipoxygenases, which accept equally well linoleic acid and arachidonic acid and generate lipid hydroperoxides (LOOHs). If the amount of free PUFAs exceeds a certain amount, lipoxygenases commit suicide. The consequence is liberation of free iron ions that react with LOOHs by formation of radicals. These start a chain reaction. LOO* radicals produced in the course of this process attack proteins, nucleic acids, and also double bonds of all unsaturated compounds by epoxidation. Morever LOOHs are decomposed to toxic epoxy acids and alphabetagammadelta-unsaturated aldehydes. Both species react with glutathione. The resulting products seem to induce apoptosis. Since the products generated by wounding or aging are formed by decomposition of LOOHs the investigation of the aging processes can be simplified by studying the physiological action of artificially generated lipid peroxidation products derived from pure PUFAs. Degradation products of LOOHs are generated by thermal decomposition of fat-containing PUFAs. These products are induced into the body by adsorption in the intestine. They are at least partly incorporated in low density lipoproteins (LDLs). Primarily investigations seem to indicate that an overload of a diet rich in PUFAs induces only after two days an increase in oxidized LDL/PUFAs for a factor up to two in young people and for a factor of more than two in old individuals.

Expression of afibrobacter succinogenes 1,3-1,4-β-glucanase in Potato (Solanum tuberosum)

The potential development of potato (Solanum tuberosum) as a low-cost eukaryotic system for the production of a commercially valuable enzyme feed supplement was examined. AFibrobacter succinogenes 1,3-1,4-β-glucanase [1,3-1,4-β-D-glucan 4-glucanohydro-lase] gene under the control of the constitutive cauliflower mosaic virus 35S promoter was transferred into the potato cultivar, Desiree. The presence of the β-glucanase cDNA in the plant genome of independent transgenic potato lines was confirmed by PCR and Southern analysis. Northern analysis identified the presence of the β-glucanase mRNA in the leaf tissue of transgenic plants. Furthermore, western analysis showedF. succinogenes β-glucanase accumulations of 0.1% and 0.05% of total soluble protein in the leaves and tubers, respectively. Specific activities of the enzyme in leaves (1693 units mg-1 β-glucanase) and tubers (2978 units mg-1 β-glucanase) were comparable to that previously reported for the enzyme produced in bacteria. Lyophilization of leaves had no effect on the specific activity of the β-glucanase, and only marginally influenced the specific activity of the enzyme expressed in tubers. Relative to the control line (cv. Desiree), tuber yields were significantly reduced by 28%-72% in all lines expressing theF. succinogenes β-glucanase, and microscopy showed that expression of the β-glucanase caused changes in cell wall structure. Results of this study demonstrate that a 1,3-1,4-β-glucanase can be expressed in potato tissues, and that potato plants have the potential to be used for the commercial production of heterologous enzymes.

Ultrastructural effects of cellulose biosynthesis inhibitor herbicides on developing cotton fibers.

Cotton fibers are often utilized as a model system to investigate cellulose biosynthesis and cell wall elongation. In this study, we grew cotton fibers in vitro, with ovules dissected at day zero post anthesis as the explant source, in the presence of three herbicides that inhibit cellulose biosynthesis. Cultures were sampled for electron microscopy and immunocytochemistry 1-2 days after beginning the treatments. After dichlobenil treatment, the fibers were much shorter than the controls and assumed a variety of abnormal shapes, from shortened versions of the control fiber to nearly spherical. The inner layers of the fiber wall often contained juxtaposed electron-translucent and -transparent areas; this layer reacted strongly with antibodies to callose. Cellulase-gold labeling in these newly developed fibers grown in the presence of dichlobenil was present at only about 3% of the control labeling. After treatment with either isoxaben or flupoxam, the fibers assumed spherical shapes and frequently (more than 60% of fibers) exhibited a new cell plate within the fiber, indicating that cell division had occurred, a process that rarely occurred in the controls. Unlike the dichlobenil-treated fibers, fibers grown in the presence of isoxaben or flupoxam contained an extensive accumulation of chiefly deesterified pectins, replacing the entire wall with an elaborated version of the pectin sheath found in control cotton fibers. These data indicate that all three herbicides are effective disrupters of cellulose biosynthesis and cause radical changes in cell wall structure and composition. Moreover, these data indicate that the composition of the walls may influence indirectly cell cycle kinetics, keeping these fiber cells in a more meristematic mode.

Compression wood-responsive proteins in developing xylem of maritime pine (Pinus pinaster ait.).

When a conifer shoot is displaced from its vertical position, compression wood (CW) is formed on the under side and can eventually return the shoot to its original position. Changes in cell wall structure and chemistry associated with CW are likely to result from differential gene/protein expression. Two-dimensional polyacrylamide gel electrophoresis of differentiating xylem proteins was combined with the physical characterization of wooden samples to identify and characterize CW-responsive proteins. Differentiating xylem was harvested from a 22-year-old crooked maritime pine (Pinus pinaster Ait.) tree. Protein extracted from different samples were revealed by high-resolution silver stained two-dimensional polyacrylamide gel electrophoresis and analyzed with a computer-assisted system for single spot quantification. Growth strain (GS) measurements allowed xylem samples to be classified quantitatively from normal wood to CW. Regression of lignin and cellulose content on GS showed that an increase in the percentage of lignin and a decrease of the percentage of cellulose corresponded to increasing GS values, i.e. CW. Of the 137 studied spots, 19% were significantly associated with GS effect. Up-regulated proteins included 1-aminocyclopropane-1-carboxylate oxidase (an ethylene forming enzyme), a putative transcription factor, two lignification genes (caffeic O-methyltransferase and caffeoyl CoA-O-methyltransferase), members of the S-adenosyl-L-methionine-synthase gene family, and enzymes involved in nitrogen and carbon assimilation (glutamine synthetase and fructokinase). A clustered correlation analysis was performed to study simultaneously protein expression along a gradient of gravistimulated stressed xylem tissue. Proteins were found to form "expression clusters" that could identify: (a) Gene product under similar control mechanisms, (b) partner proteins, or (c) functional groups corresponding to specialized pathways. The possibility of obtaining regulatory correlations and anticorrelations between proteins provide us with a new category of homology (regulatory homology) in tracing functional relationships.

Identification of a peptide synthetase involved in the biosynthesis of glycopeptidolipids of Mycobacterium smegmatis.

Five rough colony mutants of Mycobacterium smegmatis mc2155 were produced by transposon mutagenesis. The mutants were unable to synthesize glycopeptidolipids that are normally abundant in the cell wall of wild-type M. smegmatis. The glycopeptidolipids have a lipopeptide core comprising a fatty acid amide linked to a tetrapeptide that is modified with O-methylated rhamnose and O-acylated 6-deoxy talose. Compositional analysis of lipids extracted from the mutants indicated that the defect in glycopeptidolipid synthesis occurred in the assembly of the lipopeptide core. No other defects or compensatory changes in cell wall structure were detected in the mutants. All five mutants had transposon insertions in a gene encoding an enzyme belonging to the peptide synthetase family. Targeted disruption of the gene in the wild-type strain gave a phenotype identical to that of the five transposon mutants. The M. smegmatis peptide synthetase gene is predicted to encode four modules that each contain domains for cofactor binding and for amino acid recognition and adenylation. Three modules also have amino acid racemase domains. These data suggest that the common lipopeptide core of these important cell wall glycolipids is synthesized by a peptide synthetase.

Gene-specific changes in alpha-tubulin transcript accumulation in developing cotton fibers.

The fibers of cotton (Gossypium hirsutum) are single-cell trichomes that undergo rapid and synchronous elongation. Cortical microtubules provide spatial information necessary for the alignment of cellulose microfibrils that confine and regulate cell elongation. We used gene-specific probes to investigate alpha-tubulin transcript levels in elongating cotton fibers. Two discrete patterns of transcript accumulation were observed. Whereas transcripts of alpha-tubulin genes GhTua2/3 and GhTua4 increased in abundance from 10 to 20 d post anthesis (DPA), GhTua1 and GhTua5 transcripts were abundant only through to 14 DPA, and dropped significantly at 16 DPA with the onset of secondary wall synthesis. This is the first report, to our knowledge, of gene-specific changes in tubulin transcript levels during the development of a terminally differentiated plant cell. The decrease in abundance of GhTua1 and GhTua5 transcripts was correlated with pronounced changes in cell wall structure, suggesting that alpha-tubulin isoforms may be functionally distinct in elongating fiber cells. Although total alpha-tubulin transcript levels were much higher in fiber than several other tissues, including the hypocotyl and pollen, none of the alpha-tubulins was specific to fiber cells.

Synthesis and oxidative insolubilization of cell-wall proteins during osmotic stress.

The cell walls in the new white roots of jack pine (Pinus banksiana Lamb.) were observed to constrict around the shrinking protoplast of osmotically stressed roots, and pressure was maintained via an apparent adjustment of cell-wall size and elasticity. These elastic alterations of the cell wall permitted the root cells to maintain full turgor despite the loss of most of the water in the tissue. The constriction of the root cell wall around the dehydrating protoplasts to maintain turgor may reflect changes in cell wall structure. We found that these shrinking root cells synthesize and secrete into the intercellular fluid a set of proteins. These proteins become tightly associated (i.e. guanidine HCl- and sodium dodecyl sulfate-insoluble) with the cell wall but can be released from the matrix, after briefly boiling in 0.1% sodium dodecyl sulfate, by the combination of guanidine HCl, CaCl2 and dithiothreitol. However, these cell-wall proteins became insoluble with time. The proteins could subsequently be destructively extracted from the wall with acid NaClO2 treatments. After these proteins were incorporated into the cell walls, the roots adopted a new, smaller maximal tissue volume and elastic coefficients returned to normal levels.

An extensin peroxidase is associated with white-light inhibition of lupin (Lupinus albus) hypocotyl growth

The spatial distribution of the major basic (B2; pI 8.8) peroxidase of the intercellular fluid has an inverse relation with extension rate in etiolated hypocotyls of Lupinus albus L., suggesting its possible role in the control of cell expansion. White-light irradiation of etiolated hypocotyls resulted in growth inhibition and the induction of B2 and acidic (A2, pI 4.7–5.2) isoperoxidases (EC 1.1.11.7) to higher physiological activities. However, only the activities of the B2 isoperoxidases underwent quantitative changes in both space and time which suggested their role in growth-retardation. We have purified the B2 and A2 (pI 5.2) peroxidases to apparent electrophoretic homogeneity. To corroborate evidence obtained elsewhere that growth cessation coincides with cell wall structural changes and cell wall rigidification, we have shown that the B2 peroxidase, and not A2 peroxidase, cross-links tomato extensin in vitro. The B2 peroxidase may therefore catalyse the developmentally and light regulated formation of a covalently cross-linked cell wall extensin matrix in lupin hypocotyls. The cell wall would be more rigid or more recalcitrant to wall-loosening and subsequently contribute to the control of cell expansion.

Strategies for drought resistance in leaves of two almond cultivars

Potted plants of two cultivars of almond ( Amygdalus communis L.), Ramillete and Garrigues, growing under field conditions were subjected to drought by withholding irrigation for 28 days. Stressed plants were reirrigated and the recovery was studied for 8 days. Control plants were drip irrigated daily maintaining the soil matric potential at about −30 kPa. Predawn leaf water potential declined progressively due to water stress effect, reaching −0.80 MPa in Garrigues and −0.98 MPa in Ramillete at the end of the stress period. The reduction in midday leaf conductance in plants under water stress could be related to the reduction in midday leaf water potential. The reductions in leaf conductance values were about 62% and 79% for Garrigues and Ramillete, respectively. Garrigues exhibited osmotic adjustment during the greater part of the stress period and this was sufficient to maintain the leaf turgor. The relationship between leaf conductance and turgor pressure at midday appeared to be nearly linear over the range of turgor pressure observed. Ramillete presented lower leaf water potential at turgor loss point than did Garrigues. Both cultivars presented high relative apoplastic water contents. The increase in these parameter values by water stress effect observed in Ramillete could indicate changes in cell wall structure. After rewatering, most of the parameters studied recovered quickly, although leaf conductance was delayed.

Fourier Transform Infrared Microspectroscopy of Plant Tissues

Fourier transform infrared (FT-IR) microspectroscopy was applied to three distinct types of plant tissue. Reflectance microspectroscopy of nutshells highlighted the differences between the chemistries of the inner and outer surfaces and the tissue as a whole. The outer surfaces were suberized, while the inner surfaces contained absorbances indicative of lignin or tannins or both. Transmission microspectroscopy was used to follow the changes in cell wall structure and composition of flax epidermal cells during development and showed that initial development was accompanied by suberin and lignin deposition, which was followed by polysaccharide deposition characteristic of secondary cell wall formation. These results were compared with those obtained from bamboo. Transmission microspectroscopy was also used to study the infection of potato tubers by Erwinia carotovora ssp. carotovora in aerobic and anaerobic conditions. The spectra suggested that both infective conditions produced cell wall degradation, whereas anaerobic infection was accompanied by extensive breakdown of starch and plasma membrane.

Fourier-Transform Infrared Microspectroscopy of Anatomically Different Cells of Flax (Linum usitatissimum) Stems during Development

FT-IR microspectroscopy was used to study the changes in cell wall structure of four anatomically different cells of flax (Linum usitatissimum) stems during growth. The cell types were xylem, fiber, epidermal, and gland, a specialized epidermal cell. FT-IR spectra suggested that xylem growth from 5 to 20 days was accompanied by de-esterification of pectin and an increse in lignification. The principal feature in the spectra of the fiber cell walls was the predominance of cellulose absorbances at 7-20 days. Associated with these were acetyl absorbances which suggest the presence of acetylated glucomannan and/or xylan. The spectra of both the epidermal and gland cell walls contained suberin/cutin and protein absorbances. Epidermal development produced increased suberin/cutin deposition, while gland cell wall development was accompanied by protein accretion.

Response of soybean leaf water relations to tropospheric ozone

Tropospheric O3 and water stress cause significant reductions in crop growth and yield. The effects of chronic O3 exposures on leaf water relations have been less thoroughly studied. Soybeans were grown in two years in open-top field chambers equipped to control O3. The seasonal mean O3 concentrations for the charcoal-filtered controls and supplemental O3 treatments were 24 and 83 nL∙L−1 for the first year (1990) and 20 and 99 nL∙L−1 for the second year (1992). In 1990 leaves were sampled during four intervals of the 106-d growing season and subjected to potential–volume analysis. In 1992, leaves were sampled over a 3-week period, centered on 49 days after planting for potential–volume analysis as well as for midday xylem pressure potentials and leaf conductance. Ontogenetic changes in most of the parameters were large compared with treatment effects. O3 treatment consistently caused decreased symplastic volume, specific leaf mass, and tissue elasticity. In 1992, these effects were accompanied by decreased leaf conductances with no discernable change in xylem pressure potential, although midday turgor increased by 32% and stomatal competency was maintained. Tissue elasticity decreases may be related to O3-induced changes in cell wall structure during leaf expansion and may result in decreased symplastic volume.
