Xyloglucan deficiency leads to a reduction in turgor pressure and changes in cell wall properties, affecting early seedling establishment

Xyloglucan is believed to play a significant role in cell wall mechanics of dicot plants. Surprisingly, Arabidopsis plants defective in xyloglucan biosynthesis exhibit nearly normal growth and development. We investigated a mutant line, cslc-Δ5, lacking activity in all five Arabidopsis cellulose synthase like-C (CSLC) genes responsible for xyloglucan backbone biosynthesis. We observed that this xyloglucan-deficient line exhibited reduced cellulose crystallinity and increased pectin levels, suggesting the existence of feedback mechanisms that regulate wall composition to compensate for the absence of xyloglucan. These alterations in cell wall composition in the xyloglucan-absent plants were further linked to a decrease in cell wall elastic modulus and rupture stress, as observed through atomic force microscopy (AFM) and extensometer-based techniques. This raised questions about how plants with such modified cell wall properties can maintain normal growth. Our investigation revealed two key factors contributing to this phenomenon. First, measurements of turgor pressure, a primary driver of plant growth, revealed that cslc-Δ5 plants have reduced turgor, preventing the compromised walls from bursting while still allowing growth to occur. Second, we discovered the conservation of elastic asymmetry (ratio of axial to transverse wall elasticity) in the mutant, suggesting an additional mechanism contributing to the maintenance of normal growth. This novel feedback mechanism between cell wall composition and mechanical properties, coupled with turgor pressure regulation, plays a central role in the control of plant growth and is critical for seedling establishment in a mechanically challenging environment by affecting shoot emergence and root penetration.

Exploring wood micromechanical structure: Impact of microfibril angle and crystallinity on cell wall strength

Since wood has recently acquired increasing attention from structural engineers due to its green and low-carbon attribute, a growing interest in understanding the intricate relationships between the microstructure of wood and the mechanical properties of building components. This study was aimed at investigating the wood micromechanical structure of plantation Chinese fir (Cunninghamia lanceolata (Lamb.) Hook), focusing on assessing the impact of microfibril angle (MFA), chemical components and crystallinity on cell wall strength. Understanding these microstructural features is crucial, as they directly influence the overall strength of timber. Hence, significant factors contributing to the mechanical properties of the wood, i.e., the S2 layer's thickness and microfibril angle, along with the crystallinity and cellulose content, were comprehensively examined. A model was proposed to quantitatively forecast wood cell wall mechanical properties using information about MFA, chemical components, and crystallinity. This model can be used to predict the mechanical performances of wood components based on these key structural characteristics due to the close strength relationship between the micro- and macro-structures of wood.

Pectin methylesterification state and cell wall mechanical properties contribute to neighbor proximity-induced hypocotyl growth in Arabidopsis.

Plants growing with neighbors compete for light and consequently increase the growth of their vegetative organs to enhance access to sunlight. This response, called shade avoidance syndrome (SAS), involves photoreceptors such as phytochromes as well as phytochrome interacting factors (PIFs), which regulate the expression of growth-mediating genes. Numerous cell wall-related genes belong to the putative targets of PIFs, and the importance of cell wall modifications for enabling growth was extensively shown in developmental models such as dark-grown hypocotyl. However, the contribution of the cell wall in the growth of de-etiolated seedlings regulated by shade cues remains poorly established. Through analyses of mechanical and biochemical properties of the cell wall coupled with transcriptomic analysis of cell wall-related genes from previously published data, we provide evidence suggesting that cell wall modifications are important for neighbor proximity-induced elongation. Further analysis using loss-of-function mutants impaired in the synthesis and remodeling of the main cell wall polymers corroborated this. We focused on the cgr2cgr3 double mutant that is defective in methylesterification of homogalacturonan (HG)-type pectins. By following hypocotyl growth kinetically and spatially and analyzing the mechanical and biochemical properties of cell walls, we found that methylesterification of HG-type pectins was required to enable global cell wall modifications underlying neighbor proximity-induced hypocotyl growth. Collectively, our work suggests that plant competition for light induces changes in the expression of numerous cell wall genes to enable modifications in biochemical and mechanical properties of cell walls that contribute to neighbor proximity-induced growth.

Pectin methylesterase inhibitors GhPMEI53 and AtPMEI19 improve seed germination by modulating cell wall plasticity in cotton and Arabidopsis

The germination process of seeds is influenced by the interplay between two opposing factors: pectin methylesterase (PME) and pectin methylesterase inhibitor (PMEI), which collectively regulate patterns of pectin methylesterification. Despite the recognized importance of pectin methylesterification in seed germination, the specific mechanisms that govern this process remain unclear. In this study, we demonstrated that the overexpression of GhPMEI53 is associated with a decrease in PME activity and an increase in pectin methylesterification. This leads to the softening of the cell wall in seeds, which positively regulates cotton seed germination. AtPMEI19, the homologue in Arabidopsis thaliana, plays a similar role in seed germination to GhPMEI53, indicating a conserved function and mechanism of PMEI in seed germination regulation. Further studies revealed that GhPMEI53 and AtPMEI19 directly contribute to promoting radicle protrusion and seed germination by inducing cell wall softening and reducing mechanical strength. Additionally, the pathways of ABA and GA in the transgenic materials underwent significant changes, suggesting that GhPMEI53/AtPMEI19-mediated pectin methylesterification serves as a regulatory signal for the related phytohormones involved in seed germination. In summary, GhPMEI53 and its homologs alter the mechanical properties of cell walls, influencing the mechanical resistance of the endosperm or testa. Moreover, they impact cellular phytohormone pathways (e.g., ABA, GA) to regulate seed germination. These findings enhance our understanding of pectin methylesterification in cellular morphological dynamics and signaling transduction, and contribute to a more comprehensive understanding of the PME/PMEI super-gene family in plants.

Structural changes in cell wall pectic polymers contribute to freezing tolerance induced by cold acclimation in plants

Subzero temperatures are often lethal to plants. Many temperate herbaceous plants have a cold acclimation mechanism that allows them to sense a drop in temperature and prepare for freezing stress through accumulation of soluble sugars and cryoprotective proteins. As ice formation primarily occurs in the apoplast (the cell wall space), cell wall functional properties are important for plant freezing tolerance. Although previous studies have shown that the amounts of constituent sugars of the cell wall, in particular those of pectic polysaccharides, are altered by cold acclimation, the significance of this change during cold acclimation has not been clarified. We found that β-1,4-galactan, which forms neutral side chains of the acidic pectic rhamnogalacturonan-I, accumulates in the cell walls of Arabidopsis and various freezing-tolerant vegetables during cold acclimation. The gals1 gals2 gals3 triple mutant, which has reduced β-1,4-galactan in the cell wall, exhibited impaired freezing tolerance compared with wild-type Arabidopsis during initial stages of cold acclimation. Expression of genes involved in the galactan biosynthesis pathway, such as galactan synthases and UDP-glucose 4-epimerases, was induced during cold acclimation in Arabidopsis, explaining the galactan accumulation. Cold acclimation resulted in a decrease in extensibility and an increase in rigidity of the cell wall in the wild type, whereas these changes were not observed in the gals1 gals2 gals3 triple mutant. These results indicate that the accumulation of pectic β-1,4-galactan contributes to acquired freezing tolerance by cold acclimation, likely via changes in cell wall mechanical properties.

Abnormal cell wall structure caused by boron nutrient imbalance in orchards could affect psyllid feeding behaviour, resulting in epidemic variation of Asian citrus psyllid.

The control of Huanglongbing (HLB), one of the most destructive pests of citrus, relies heavily on the reduction of Asian citrus psyllid (ACP), Diaphorina citri Kuwayama. An in-depth understanding of ACP feeding behaviours among citrus plants is urgent for comprehensive management of orchards. An investigation was conducted in 37 citrus orchards in HLB epidemic areas, sampling shoots in the area with aggregation feeding of ACP (ACPf) and shoots in a neighbouring area without ACP feeding (CK), to study the interaction between leaf chemical composition and ACP psyllid feeding behaviours. Results of FTIR showed a strong absorption peak intensity, mainly representing functional groups originating from cell wall components in the leaf with ACP feeding. As compared with the control, cell wall components, such as alkali-soluble pectin, water-soluble pectin, total soluble pectin, cellulose, and hemicellulose, of the cell wall of ACPf increased by 134.0%, 14.0%, 18.0%, 12.5%, and 20.35%, respectively. These results suggest that cell wall mechanical properties significantly decreased in the term of decreases in pectin performance and cellulose mechanical properties. In addition, there was a remarkably lower boron (B) content in leaves and cell wall components with ACP feeding. Further analysis indicated that leaf B content significantly affected leaf cell wall components. Taken together, we provide evidence to demonstrate that the regional distribution of nutrient imbalance in orchards could affect psyllid feeding behaviour by weakening the cell wall structure, resulting in epidemic variation in ACP. This could help us to understand the management of psyllid infections in orchards with unbalanced nutrition.

Hydration and mechanical properties of arabinoxylan, (1,3;1,4)-β-glucan, and cellulose multilayer films simulating the cell wall of wheat endosperm

The cell walls of wheat endosperm, which play a pivotal role in seed germination, exhibit a laminated structure primarily composed of polysaccharides. In this study, composite multilayer films were prepared using arabinoxylan (AX), (1,3;1,4)-β-D-glucan (MLG), and cellulose nanofibers (CNFs), and the effect of polymer blend structure on cell wall hydration and mechanical properties was investigated. Atomic force microscopy and X-ray diffraction indicated that the network structure of MLG/CNF exhibits a higher degree of continuity and uniformity compared to that of AX/CNF. Mechanically, the extensive linkages between MLG and CNFs chains enhance the mechanical properties of the films. Moreover, water diffusion experiments and TD-NMR analysis revealed that water molecules diffuse faster in the network structure formed by AX. We propose a structural model of the endosperm cell wall, in which the CNFs polymer blend coated with MLG serves as the framework, and the AX network fills the gaps between them, providing diffusion channels for water molecules.

Mutation of AtPME2, a pH-Dependent Pectin Methylesterase, Affects Cell Wall Structure andHypocotyl Elongation.

Pectin methylesterases (PMEs) modify homogalacturonan's chemistry and play a key role in regulating primary cell wall mechanical properties. Here, we report on Arabidopsis AtPME2, which we found to be highly expressed during lateral root emergence and dark-grown hypocotyl elongation. We showed that dark-grown hypocotyl elongation was reduced in knock-out mutant lines as compared to the control. The latter was related to the decreased total PME activity as well as increased stiffness of the cell wall in the apical part of the hypocotyl. To relate phenotypic analyses to the biochemical specificity of the enzyme, we produced the mature active enzyme using heterologous expression in Pichia pastoris and characterized it through the use of a generic plant PME antiserum. AtPME2 is more active at neutral compared to acidic pH, on pectins with a degree of 55-70% methylesterification. We further showed that the mode of action of AtPME2 can vary according to pH, from high processivity (at pH8) to low processivity (at pH5), and relate these observations to the differences in electrostatic potential of the protein. Our study brings insights into how the pH-dependent regulation by PME activity could affect the pectin structure and associated cell wall mechanical properties.

Cell wall polysaccharides, phenolic extractability and mechanical properties of Aleatico winegrapes dehydrated under sun or in controlled conditions

Aleatico is an Italian red winegrape variety traditionally used partially dehydrated to produce Passito wines. The aim of this research is to assess the effect of two different off-vine withering techniques (natural sun-exposed: SUN, and controlled temperature and humidity conditions: CTR) on grapes physicochemical modifications, focusing on mechanical properties, phenolic profile, and cell-wall polysaccharide structures. The composition and extractability of polyphenols were studied before and after the two withering processes, differentiating the tissues (skin, pulp, seeds). A stronger effect on cell-wall polysaccharides composition pectin was highlighted by chemical and immunochemical approaches in SUN skins at the level of pectin chains and de-esterification, affecting grapes mechanical properties, especially for skin rigidity (<Esk). Sun-dehydrated grapes showed the highest pH (about 0.3 units more than FRESH and CTR), and the lowest acidity (−0.71 g/L vs FRESH) and anthocyanin content in wine-like solution (−41%), whereas these parameters were better preserved in controlled conditions. Instead, seeds tannins were higher in SUN than other samples, with a lower oligomers/polymers ratio. Indeed, after withering the phenolic extractability decreased in skins (total phenolics: from 60% for FRESH to 40% for SUN; anthocyanins: from 72% for FRESH to 46% for SUN) and increased in seeds (total phenolics: from 30% for FRESH to 41% for SUN) compared with fresh samples, in both cases more markedly for SUN than CTR. Skin phenolic composition after 10 days of simulated-maceration was affected by the combination of the loss/concentration balance, together with their modified release in the wine-like solution. As regards seeds, instead, the differences highlighted by the extractable profiles were due to the modified extractability as consequence of the different withering conditions and not to chemical quantitative/qualitative modifications of phenolic compounds caused by dehydration.

The influence of microstructural characteristics and cell wall material properties on the mechanical behaviors of different tissues of sorghum stems

Sorghum stems comprise different tissue components, i.e., rind, pith, and vascular bundles in the rind and pith regions, of different cell morphologies and cell wall characteristics. The overall responses of stems to mechanical loadings depend on the responses of these tissues themselves. Investigating how each tissue deforms to various loading conditions will inform us of the failure mechanisms in sorghum stems when exposed to wind loadings, which can guide the development of lodging-resistant variants. To this end, numerical analyses were implemented to investigate the effects of cell morphologies and cell wall properties on the overall mechanical responses of the above four tissues under tension and compression. Microstructures of different tissues were constructed from microscopic images of the tissues using computer-aided design (CAD), which were then used for finite element (FE) analyses. Shell finite elements were used to model the cell walls, and the classical lamination model was used to determine the overall mechanical responses of cell walls having different fiber composite arrangements. The results from the numerical analyses helped explain how the loading (boundary) conditions, the cell microstructures, the mechanical properties of cell walls of different tissues, the cell wall thickness, the microfibril angle (MFA) of fiber composites of the cell walls, and the turgor pressure affected the overall mechanical responses of the tissues. Tissue stiffening or softening behaviors were attributed to different microstructural deformations, i.e., local or global buckling of cell walls, cell collapse, densifications of cells, or reorientation and rearrangement of cells. The mechanical properties and thickness of cell walls only affected the stiffness and load-bearing ability of the tissues. The turgor pressure affected the compressive responses but its effect on tensile responses was negligible. The MFA had a significant influence on the stiffness and load-bearing ability when the tissues were loaded along their longitudinal axis, but it had an insignificant effect on loading in the transverse direction. Tissues with smaller cell sizes and denser cells were stronger and stiffer than those with larger cell sizes. The numerical simulations also revealed that rind and rind vascular bundles were stiffer and had higher load-bearing ability than pith and pith vascular bundles.

Dual preservative strategy for facilitating bamboo durability using cinnamaldehyde and diethylenetriamine and its reaction characteristics on bamboo cell wall

Cinnamaldehyde exhibits excellent anti-mold properties that can preserve bamboo. However, establishing a stable bonding linkage (CN) requires cinnamaldehyde modification, which can be used to manipulate cell wall characteristics and improve bamboo durability. In this study, cinnamaldehyde's chemical fixation into bamboo through the Mannich reaction was successfully achieved, resulting in enhanced anti-mold capacity and dimensional stability. The modified bamboo exhibited remarkable anti-mold properties, with a 40.0% increase in resistance to swelling caused by moisture fluctuations. Nanoindentation analysis also revealed that the internal bamboo cell wall's mechanical properties improved following modification, including a significant improvement in surface hydrophobicity from 13.9° to 67.3°. Concerning the impact of the modification on bamboo cells, the sclerenchyma cell of bamboo was determined to be the focus. We used guaiacol as a lignin model compound to elucidate the potential mechanism of cinnamaldehyde modification, specifically Schiff base formation, using nuclear magnetic resonance spectra. This study shows a viable dual preservative strategy to extend bamboo lifespan.

Stiffness transitions in new walls post-cell division differ between Marchantia polymorpha gemmae and Arabidopsis thaliana leaves

Plant morphogenesis is governed by the mechanics of the cell wall-a stiff and thin polymeric box that encloses the cells. The cell wall is a highly dynamic composite material. New cell walls are added during cell division. As the cells continue to grow, the properties of cell walls are modulated to undergo significant changes in shape and size without breakage. Spatial and temporal variations in cell wall mechanical properties have been observed. However, how they relate to cell division remains an outstanding question. Here, we combine time-lapse imaging with local mechanical measurements via atomic force microscopy to systematically map the cell wall's age and growth, with their stiffness. We make use of two systems, Marchantia polymorpha gemmae, and Arabidopsis thaliana leaves. We first characterize the growth and cell division of M. polymorpha gemmae. We then demonstrate that cell division in M. polymorpha gemmae results in the generation of a temporary stiffer and slower-growing new wall. In contrast, this transient phenomenon is absent in A. thaliana leaves. We provide evidence that this different temporal behavior has a direct impact on the local cell geometry via changes in the junction angle. These results are expected to pave the way for developing more realistic plant morphogenetic models and to advance the study into the impact of cell division on tissue growth.

Apple metabolism under oxidative stress affects plant cell wall structure and mechanical properties

Several studies have shown beneficial effects of short exposure to oxidative stress on stored fruit, such as better preservation, increased firmness, preservation of polyphenolic compounds, and reduced risk of postharvest disorders such as bitter pit and superficial scald in apples. In this study the effect of short-term oxidative stress conditions on the physiology of apple fruit was investigated. Apple fruit of three cultivars were exposed to hypoxic storage conditions of various lengths to induce anaerobiosis. The response of apple fruit to short-term oxidative stress was evaluated by means of cell wall immunolabeling and atomic force microscopy. In addition, the antioxidant capacity and antioxidative activity of apple peels was assessed. Through various techniques, it was shown that short-term oxidative stress conditions promote specific enzymatic activity that induces changes in the cell wall of apple fruit cells. Exposure to short-term stress resulted in the remodeling of cell wall pectic polysaccharides, observed as an increase in the size and complexity of extracted oxalate pectin. Structural changes in the cell wall were followed by an increase in Young’s modulus (compressive stiffness of a solid material, expressed as the relationship between stress and axial strain) of the cell wall material. The data presented in this paper show in a novel way how storage under short-term oxidative stress modifies the cell wall of apple fruit at the molecular level.

Variability in the mechanical properties of cell walls of Bambusa arundinacea (Retz.) Willd. based on nanoindentation method,

Bambusa arundinacea (Retz.) Willd. is a large and thorny bamboo species used for construction and scaffolding purposes. Bamboo fibers, which are an important part of bamboo, give bamboo its excellent mechanical properties. The goal of this work was to get more information about fiber in order to learn more about bamboo material. It was tested for different ages, heights, and radial positions using the nanoindentation method. The results showed that the mean values of indentation modulus of elasticity and hardness of this species were 20.79 GPa and 497.01 MPa, respectively, and that the mechanical properties of the fiber cell walls were less affected by the three factors and were generally consistent. In regards to the age factor, it had little effect on the indentation modulus of elasticity, while the hardness increased slightly with age. The indentation modulus of elasticity and hardness tended to increase with increasing height. The fiber strength was relatively stable in different parts of the radial direction.

In vivo measurement of the Young’s modulus of the cell wall of single root hairs

Root hairs are cells from the root epidermis that grow as long tubular bulges perpendicular to the root. They can grow in a variety of mechanical or chemical environments. Their mechanical properties are mainly due to their stiff cell wall which also constitutes a physical barrier between the cell and its environment. Thus, it is essential to be able to quantify the cell wall mechanical properties and their adaptation to environmental cues. Here, we present a technique we developed to measure the Young’s (elastic) modulus of the root hair cell wall. In essence, using custom-made glass microplates as cantilevers of calibrated stiffness, we are able to measure the force necessary to bend a single living root hair. From these experiments one can determine the stiffness and Young’s modulus of the root hair cell wall.

The Modification of Cell Wall Properties Is Involved in the Growth Inhibition of Rice Coleoptiles Induced by Lead Stress.

Lead (Pb) is a widespread heavy metal pollutant that interferes with plant growth. In this study, we investigated the effects of Pb on the mechanical and chemical properties of cell walls and on the growth of coleoptiles of rice (Oryza sativa L.) seedlings grown in the air (on moistened filter paper) and underwater (submerged condition). Coleoptile growth of air-grown seedlings was reduced by 40% by the 3 mM Pb treatment, while that of water-grown ones was reduced by 50% by the 0.5 mM Pb. Although the effective concentration of Pb for growth inhibition of air-grown coleoptiles was much higher than that of water-grown ones, Pb treatment significantly decreased the mechanical extensibility of the cell wall in air- and water-grown coleoptiles, when it inhibited their growth. Among the chemical components of coleoptile cell walls, the amounts of cell wall polysaccharides per unit fresh weight and unit length of coleoptile, which represent the thickness of the cell wall, were significantly increased in response to the Pb treatment (3 mM and 0.5 mM Pb for air- and water-grown seedlings, respectively), while the levels of cell wall-bound diferulic acids (DFAs) and ferulic acids (FAs) slightly decreased. These results indicate that Pb treatment increased the thickness of the cell wall but not the phenolic acid-mediated cross-linking structures within the cell wall in air- and water-grown coleoptiles. The Pb-induced cell wall thickening probably causes the mechanical stiffening of the cell wall and thus decreases cell wall extensibility. Such modifications of cell wall properties may be associated with the inhibition of coleoptile growth. The results of this study provide a new finding that Pb-induced cell wall remodeling contributes to the regulation of plant growth under Pb stress conditions via the modification of the mechanical property of the cell wall.

Boron deficiency mediates plant-insect (Diaphorima citri) interaction by disturbing leaf volatile organic compounds and cell wall functions.

In the present study, a hydroponic culture was carried out to evaluate the effects of boron deficiency on psyllid feeding process to decode the molecular/biochemical basis of host-psyllid interaction. Boron deficiency was observed to play a major role in accelerating the release of volatile organic compounds, especially methyl salicylate, affecting the shikimic acid pathway through an elevated synthesis of shikimic acid, l-phenylalanine, 3-phenylpyruvic acid and salicylic acid. These changes made citrus leaf more attractive to psyllid adults. Meanwhile, boron deficiency evidently decreased the boron concentration of leaf cell wall fractions, thereby, weakened the structural stability by affecting pectin and cellulose formations. A significant decrease of cell wall mechanical strength was observed in boron-deficiency leaf, which could be the critical reasons to reduce piercing and to increase phloem ingestion during psyllid feeding. Our study demonstrated that boron deficiency facilitated the feeding behavior of psyllid adults through elevated release of methyl salicylate, coupled with weakened mechanical properties of cell wall.

An Arabidopsis PTH2 Gene Is Responsible for Gravity Resistance Supporting Plant Growth under Different Gravity Conditions.

Terrestrial plants respond to and resist gravitational force. The response is termed “gravity resistance”, and centrifugal hypergravity conditions are efficient for investigating its nature and mechanism. A functional screening of Arabidopsis T-DNA insertion lines for the suppression rate of elongation growth of hypocotyls under hypergravity conditions was performed in this study to identify the genes required for gravity resistance. As a result, we identified PEPTIDYL-tRNA HYDROLASE II (PTH2). In the wild type, elongation growth was suppressed by hypergravity, but this did not happen in the pth2 mutant. Lateral growth, dynamics of cortical microtubules, mechanical properties of cell walls, or cell wall thickness were also not affected by hypergravity in the pth2 mutant. In other words, the pth2 mutant did not show any significant hypergravity responses. However, the gravitropic curvature of hypocotyls of the pth2 mutant was almost equal to that of the wild type, indicating that the PTH2 gene is not required for gravitropism. It is suggested by these results that PTH2 is responsible for the critical processes of gravity resistance in Arabidopsis hypocotyls.

Mechanisms of plant cell wall surveillance in response to pathogens, cell wall-derived ligands and the effect of expansins to infection resistance or susceptibility.

Cell wall integrity is tightly regulated and maintained given that non-physiological modification of cell walls could render plants vulnerable to biotic and/or abiotic stresses. Expansins are plant cell wall-modifying proteins active during many developmental and physiological processes, but they can also be produced by bacteria and fungi during interaction with plant hosts. Cell wall alteration brought about by ectopic expression, overexpression, or exogenous addition of expansins from either eukaryote or prokaryote origin can in some instances provide resistance to pathogens, while in other cases plants become more susceptible to infection. In these circumstances altered cell wall mechanical properties might be directly responsible for pathogen resistance or susceptibility outcomes. Simultaneously, through membrane receptors for enzymatically released cell wall fragments or by sensing modified cell wall barrier properties, plants trigger intracellular signaling cascades inducing defense responses and reinforcement of the cell wall, contributing to various infection phenotypes, in which expansins might also be involved. Bamboo flattening technology ebables efficient and value-added utilization of bamboo in the manufacture of furniture and engineered composites

Bamboo flattening, a novel and efficient bamboo processing and utilization technology, can effectively flatten bamboo culms into crack-free board. The technology enables the efficient and value-added utilization of bamboo in the manufacture of furniture and engineered composites. In this study, we compared the effects of three types of flattening technologies on the macro-mechanical, macro-physical, micro-morphology, and micro-mechanical properties of untreated, softened a flattened bamboo samples. As expected, the hemicellulose and cellulose concentrations of the saturated steam treated sample (18.05% and 38.9%, respectively) were less than those the untreated sample (21.3% and 40.1%, respectively). Moreover, scanning electron microscopy (SEM) analysis showed that the notched flattening technology (NFT) inserted indents and grooves on the bamboo surface. However, the “non-notched flattening technology (NNFT)” only compressed the bamboo tissue. The flattening process increased the equilibrium moisture content and density of bamboo. The enhanced density, lignin content, and cellulose crystallinity index enhanced the bamboo cell wall properties. The non-notched-flattened bamboo board exhibited a higher cell wall modulus of elasticity and hardness (20.1 and 0.89 GPa, respectively) than the untreated bamboo samples (13.7 and 0.49 GPa, respectively). The enhanced bamboo cell wall mechanical properties of the non-notched-flattened bamboo board. Finally, we explored the practical applications, economic benefits, and limitations of the current “Non-notched bamboo flattening technology”.
