Photosynthetic pigments and color of wild Undaria pinnatifida for wakame production (Chubut, Patagonia Argentina)

The invasive brown seaweed Undaria pinnatifida was first recorded in the Southwest Atlantic in 1992 in Golfo Nuevo, northern Patagonia, Argentina. Since then, it has spread over most of the Argentine coast developing dense seasonal kelp beds. This seasonal biomass could be used for wakame production, for which the color is an important quality variable. The aim of this study was to determine the temporal and ontogenetic variations of pigments content and color of Undaria from Golfo Nuevo, and its relation to water temperature, irradiance, and nutrient concentration. Sporophytes present in five 1m2 quadrats were collected from a 2 m-depth hard bottom in five occasions between September and November 2018. Morphometric variables were measured in all the sporophytes collected (N = 200). Color (CIE Lab) and pigments (Chlorophyll a and c, and fucoxanthin) were measured on each juvenile and adult sporophyte. Color was also measured after boiling. Water temperature and irradiance were measured 70 cm above the bottom with a 10-minute frequency. Nitrate+nitrite, ammonium, and phosphate concentrations (μM) were measured from surface seawater samples taken on each sampling date. Higher pigment content and better color parameters (lower L* and b*) were observed in adult sporophytes with thicker blades. In juvenile sporophytes with thin blades, L*and b* color parameters were strongly affected by pigment content. Temporal fluctuations of color parameters and pigment content were smooth and not significant. GLMs showed that variations in color parameters and pigments were mostly related to development and blade thickness, and at a lower extent, by nutrients and irradiance. We conclude that adult sporophytes have better color than juveniles for wakame production during the austral spring in the study area. However, further research would be necessary to analyze other quality parameters (e.g. hairs, wrinkles, and hardness) and their geographical variations.

Monthly fluctuations in the content and monosaccharide composition of fucoidan from Undaria pinnatifida sporophylls from northern Patagonia

The invasive brown seaweed Undaria pinnatifida was first recorded in 1992 in Golfo Nuevo, northern Patagonia, Argentina (hereafter Undaria). Like other brown seaweeds, Undaria synthesizes fucoidans, a unique class of sulfated polysaccharides, which display an array of biological activities and have important commercial value. In this work we have measured the content, sulfate, and monosaccharide composition of fucoidans in Undaria sporophylls from Golfo Nuevo. These results were analyzed in relation to harvest month and development stage of algal thalli. Samples were collected between November 2015 and March 2016 and classified according to morphological traits into previously defined development stages. Acid extraction (0.01 M HCl, pH 2) was carried out at room temperature and at 70 °C. Average fucoidan content was 18.1% dry wt. and slightly increased with month progression and in senescent individuals. Average sulfate content in fucoidan extracts was 20.3% dry wt. decreasing with month and development. Predominant sugars were fucose and galactose averaging a total of 91 mol% of neutral sugars. This study confirms that sporophyll fucoidans from Patagonian Undaria are sulfated galactofucans. Fucoidan content significantly increases from 14.5 to 19.2% dry wt. with sporophyte development. Slight increments with month progression are not statistically significant. Molar proportion of neutral sugars is constant between maturity stages and varies slightly with month progression. Sulfate content of fucoidan decreased significantly with development and month progression. Pooled with previous reports about abundance and seasonality of Undaria in Patagonia, our results suggest that sporophylls could be harvested for fucoidan production at least during 5 months between November and March.

Study of the potential use of the invasive marine algae Undaria pinnatifida in the preliminary development of a functional textile

Seaweeds are considered to be an important source of relevant chemical substances or bioactive compounds of great usefulness in the medical, cosmetic, pharmaceutical or food industry. Recently, some seaweed had been used to produce or incorporate in fibers in order to manufacture technical textiles in which functional ingredient of the seaweed are exploited in the health and body care area. The brown seaweed Undaria pinnatifida (Undaria) has an important nutritive value being consumed as food in several Asian countries. As well Undaria is required in cosmetic or dermatology medicine given the presence of other bioactive chemical substances known as phycocolloids. The objective of this study was to explore the potential use of Undaria in a preliminary development of a functional textile. A soft bilaminate textile with a high crease recovery and an efficient release of mucilage, was achieved using a fine dust (particle of <0.26 mm) of Undaria between two layers of nonwoven. The best combination of the two layers of nonwoven was laminate and fine spundbond. The extract of Undaria used in our experimentation had minerals and vitamins with positive effect to the human skin, besides it is no irritant when applied to healthy or abraded skin. Further research focused in the incorporation of this alga in nonwoven fibers is required. We expect to obtain a textile with a dermatologic use in the near future; so as to turn Undaria into a resource susceptible to exploitation given the specie is considered a worldwide plague.

Characterization and hypoglycemic effects of sulfated polysaccharides derived from brown seaweed Undaria pinnatifida

The brown seaweed Undaria pinnatifida polysaccharides show various biological activities, but their hypoglycemic activity and the underlying mechanism remain unclear. Here, three fractions of sulfated polysaccharides Up-3, Up-4, and Up-5 were prepared by microwave-assisted extraction from U. pinnatifida. In vitro assays demonstrated that Up-3 and Up-4 had strong α-glucosidase inhibitory activity, and Up-3, Up-4, and Up-5 could improve the glucose uptake in insulin-resistant HepG2 cells without affecting their viability. In vivo studies indicated Up-3 and Up-4 markedly reduced postprandial blood glucose levels. Up-U (a mixture of Up-3, Up-4, and Up-5), reduced fasting blood glucose levels, increased glucose tolerance and alleviated insulin resistance in HFD/STZ-induced hyperglycemic mice. Histopathological observation and hepatic glycogen measurement showed that Up-U alleviated the damage of the pancreas islet cell, reduced hepatic steatosis, and promoted hepatic glycogen synthesis. These findings suggest that Up-U could alleviate postprandial and HFD/STZ-induced hyperglycemia and was a potential agent for diabetes treatment.

Physiological and ultrastructural responses of the brown seaweed Undaria pinnatifida to triphenyltin chloride (TPTCL) stress

Triphenyltin chloride (TPTCL) is a well-known marine pollutant that may constitute major environmental threats to seaweed mariculture. In the present study, the toxic effects of TPTCL on physiology and ultrastructure of cultivated sporophytes of Undaria pinnatifida were investigated under different TPTCL concentrations ranging from 0 to 100 μg L−1. Significant negative effects of increased TPTCL concentration were detected in the relative growth rates, survival percentages and chlorophyll a contents of young and adult sporophytes. Low TPTCL concentrations could significantly stimulate the activities of enzymes related to nitrogen metabolism. The chloroplast, mitochondria and nucleus inside cells were greatly damaged by TPTCL. Meanwhile, significant increases of electron dense deposits and physodes were found. Additionally, young sporophytes exhibited greater tolerance to TPTCL stress than adult sporophytes. The results of this study indicate that coastal TPTCL pollution could reduce the productivity and quality of cultivated U. pinnatifida.

Oral Administration of Edible Seaweed Undaria Pinnatifida (Wakame) Modifies Glucose and Lipid Metabolism in Rats: A DNA Microarray Analysis.

Wakame is an edible seaweed that is a common constituent in the Japanese diet. Previous studies showed that wakame consumption is associated with the prevention of metabolic syndrome, but the molecular mechanisms underlying the protective effects are poorly understood. To determine if the expression of hepatic genes is affected by ingestion of the brown seaweed Undaria pinnatifida (wakame), rats were fed a diet containing 0, 0.1, or 1.0 g per 100 g dried wakame powder for 28 days. Administration of 1% wakame significantly decreased serum total cholesterol levels. Hepatic gene expression was investigated using DNA microarray analysis, and the results showed that wakame suppresses the lipogenic pathway by downregulating SREBF-1. Moreover, bile acid biosynthesis and gluconeogenesis were promoted by upregulation of the PPAR signaling pathway, which leads to a reduction in the accumulation of cholesterol and promotion of β-oxidation. These results suggest that wakame ingestion affects glucose and lipid metabolism by altering the expression of SREBF-1 and PPAR signal-related genes.

Undaria marching on; late arrival in the Republic of Ireland

The Asian invasive brown seaweed Undaria pinnatifida was found for the first time in the Republic of Ireland in Kilmore Quay in Co. Wexford in July 2016. As this brown kelp is of considerable economic importance and is cultivated in Asia as well as in Europe, it opens up the discussion if this invasive species is socially acceptable to be cultivated in the Republic of Ireland for food and other purposes. This paper briefly examines the global economic importance, cultivation aspects compared to the European native equivalents such as Alaria esculenta and Saccharina latissima, cultivation yield, economic considerations and the ecological impact of the spread of Undaria into non-native areas. Based on the information and facts presented, it is concluded that Undaria from a physical, social and economic point of view can be cultivated in Ireland.

Development of an eco-protocol for seaweed chlorophylls extraction and possible applications in dye sensitized solar cells

Seaweeds are a reserve of natural dyes (chlorophylls a, b and c), characterized by low cost and easy supply, without potential environmental load in terms of land subtraction, and also complying with the requirements of an efficient waste management policy. In particular, the brown seaweed Undaria pinnatifida is a species largely present in the Venice Lagoon area, and for it a removal strategy is actually mandatory. In this paper, we set-up an eco-protocol for the best extraction and preparation procedures of the pigment, with the aim of finding an easy and affordable method for chlorophyll c extraction, exploring at the same time the possibility of using these algae within local sustainable management integrated strategies, among which the possible use of chlorophylls as a dye source in dye sensitized solar cells (DSSCs) is investigated. Experimental results suggest that the developed protocols are useful to optimize the chlorophyll c extraction, as shown by optical absorption spectroscopy measurements. The DSSCs built with the chlorophyll extracted by the proposed eco-protocol exhibit solar energy conversion efficiencies are similar to those obtained following extraction protocols with larger environmental impacts.

Anti-inflammatory Activity and Chemical Composition of Essential Oil Extracted with Supercritical CO2 from the Brown Seaweed Undaria pinnatifida

Essential oil extracted with supercritical CO2 fluid from the brown seaweed Undaria pinnatifida showed high anti-inflammatory activity. Optimal extraction with supercritical CO2 was observed at 45°C and 20 MPa. The essential oil was active against mouse ear inflammation induced by phorbol myristate acetate, with median inhibition concentration IC50 values of 87, 134, and 158 µg per ear for edema, erythema, and blood flow, respectively. The major component compounds were palmitic acid (26 %), 2-(9,12- octadecadienyloxy)-,(z,z)-ethanol (16 %), eicosa-5,8,11,14,17-pentaenoate (EPA; 13 %), and 5,7,11- eicosatrienoic acid (mead acid; 12 %). These findings are consistent with the claims that the seaweed U. pinnatifida can be used in remedies for inflammation-related symptoms.

Mekabu fucoidan: Structural complexity and defensive effects against avian influenza A viruses

Fucoidan from the sporophyll (Mekabu) of brown seaweed Undaria pinnatifida (wakame) is interesting due to its various biological activities. Mekabu fucoidan (Mw ∼9kDa) of this study (MF) was previously isolated and characterized by chemical and separation methods including GPC and methylation analysis (Lee, Hayashi, Hashimoto, Nakano, & Hayashi, 2004). It was found that this fucoidan composed of partially sulphated (DS ∼0.72) fucose and galactose at approximately equal amounts. Methylation analyses revealed complex structure of MF. However, it has been still unclear about the linkages between units and substitution patterns. To solve these structural tasks, spectroscopic methods (FTIR, FT Raman and NMR) were used in the analysis of native MF and its deesterified derivatives. According to obtained results, this polysaccharide was defined as O-acetylated sulphated fucogalactan. The defensive effects of MF were evaluated on mice infected with avian influenza A viruses (H5N3 and H7N2 subtypes); its efficacy was determined in reducing viral replication and increasing antibody production. Oral administration of MF resulted in suppressing virus yields. In addition, the production of neutralizing antibodies and mucosal IgA in the animals inoculated with the avian influenza A viruses was significantly increased. These results suggested that MF could be used for the prevention of viral infection.

Zoospore-derived monoecious gametophytes in Undaria pinnatifida (Phaeophyceae)

The annual life cycle of the brown seaweed Undaria pinnatifida (Harvey) Suringer comprises a macroscopic diploid sporophyte stage and a microscopic haploid gametophyte stage. In 2011, an unusual zoospore-derived monoecious gametophyte isolate (designated as line 10-5-3) of U. pinnatifida was observed. To understand this phenomenon, a comprehensive screening of eighty-two previously identified male gametophyte cultures, isolated from three randomly selected cultivars (lines 10, 7, and 5) was performed. Thirty-six of the isolates developed both antheridia and oogonia on the same filamentous fragment in a standard gametogenesis test (SGT: 18A degrees C, 60 mu mol photons/(m(2)center dot s)). Selfing of the monoecious gametophyte or crossing it with a normal male gametophyte both gave rise to morphologically normal sporophytic offspring. However, crossing resulted in a much higher fertilization rate (89.7%). The hybrid and selfed sporophytic offspring were grown to maturity in flow tanks at an ambient temperature of 10-18A degrees C over a period of 69 days. Active zoospores were released from both types of mature sporophylls. The majority of these developed into male gametophytes, while 15%-20% developed into the observed monoecious structures on the same filament. Using PCR amplification it was found that all the monoecious gametophyte isolates and the sporophytic offspring resulting from the selfing and crossing lacked the femalelinked microsatellite sequence (a part of the locus Up-AC-2A8, GenBank accession No. AY738602.1), indicating their male nature. U. pinnatifida is an invasive species in some regions and the implications of the above findings for this species in nature are briefly discussed.

Enzyme-assisted extraction of fucoxanthin and lipids containing polyunsaturated fatty acids from Undaria pinnatifida using dimethyl ether and ethanol

A novel method for the efficient extraction of fucoxanthin and lipids containing polyunsaturated fatty acids (PUFAs) from the brown seaweed Undaria pinnatifida was developed and demonstrated at a laboratory scale. U. pinnatifida, also known as Wakame, contains a number of biologically active lipophilic compounds, particularly fucoxanthin, which has anti-oxidant, anti-cancer, anti-obesity and anti-inflammatory properties. The yield of fucoxanthin and lipids containing PUFAs was determined by extraction from wet and freeze-dried seaweed using dimethyl ether (DME) and ethanol and from enzyme-pretreated seaweed using the same solvents. The highest yields of fucoxanthin (94%) and lipids (94%) rich in PUFAs were obtained from fresh (wet) U. pinnatifida by enzyme pre-processing, followed by extraction using DME with ethanol as a co-solvent. In comparison, ethanol extraction resulted in lower extraction yields for both fucoxanthin (86%) and lipids (73%) under the conditions described. Enzyme pre-processing using alginate lyase resulted in the hydrolysis of cell wall polysaccharides, resulting in high extraction yields. The hydrolysis time, pH and temperature were found to be the most important parameters for the enzyme pre-processing step and for minimizing fucoxanthin losses due to oxidative degradation. The removal of water-soluble compounds (polysaccharides) following the enzyme pre-treatment prior to DME extractions doubled the throughput and maximized the yield. The residual biomass was colorless or a pale-brown color after the DME extraction, which indicated the highly effective extraction of fucoxanthin. The PUFA content and fucoxanthin levels were not affected by the enzyme or extraction using the described enzyme-assisted DME+ethanol co-solvent process.

Fucoidan derived from Undaria pinnatifida induces apoptosis in human hepatocellular carcinoma SMMC-7721 cells via the ROS-mediated mitochondrial pathway.

Fucoidans, fucose-enriched sulfated polysaccharides isolated from brown algae and marine invertebrates, have been shown to exert anticancer activity in several types of human cancer, including leukemia and breast cancer and in lung adenocarcinoma cells. In the present study, the anticancer activity of the fucoidan extracted from the brown seaweed Undaria pinnatifida was investigated in human hepatocellular carcinoma SMMC-7721 cells, and the underlying mechanisms of action were investigated. SMMC-7721 cells exposed to fucoidan displayed growth inhibition and several typical features of apoptotic cells, such as chromatin condensation and marginalization, a decrease in the number of mitochondria, and in mitochondrial swelling and vacuolation. Fucoidan-induced cell death was associated with depletion of reduced glutathione (GSH), accumulation of high intracellular levels of reactive oxygen species (ROS), and accompanied by damage to the mitochondrial ultrastructure, depolarization of the mitochondrial membrane potential (MMP, Δψm) and caspase activation. Moreover, fucoidan led to altered expression of factors related to apoptosis, including downregulating Livin and XIAP mRNA, which are members of the inhibitor of apoptotic protein (IAP) family, and increased the Bax-to-Bcl-2 ratio. These findings suggest that fucoidan isolated from U. pinnatifida induced apoptosis in SMMC-7721 cells via the ROS-mediated mitochondrial pathway.

Novel means for variety breeding and sporeling production in the brown seaweed Undaria pinnatifida (Phaeophyceae): Crossing female gametophytes from parthenosporophytes with male gametophyte clones

SummaryThe unialgal haploid gametophyte clones are frequently used for variety breeding and sporeling production in the brown seaweed Undaria pinnatifida because a single crossing of a pair of the selected male and female gametophyte clones can generate sporophytic offspring with identical genotype and phenotype. As the seeds to be sprayed on the collectors, the detachment rate of the filamentous gametophytes is high in comparison to the seeded spores. In this investigation, we report the use of parthenogenesis to achieve the same goal in variety selection and sporeling production but with higher efficiency. The selected female unialgal gametophyte clone (Code: 06‐8‐1F) was induced to produce parthenosporophytes. These sporophytes were grown up in a controlled system and used to release zoospores. All zoospores generated into female gametophytes. These female gametophytes were allowed to go through parthenogenesis for the second year to confirm the applicability of this means. In the third year, the zoospores released from the parthenosporophytes were seeded on collectors over summer in female gametophyte form. In the early autumn, a selected male unialgal gametophyte clone (code: 5#F1‐2‐5M) was used to cross the seeded female gametophytes on the collectors. When the sporelings reached a mean length of 780 μm, they were transplanted to open sea on longlines for growing up. At harvest, the average length, width and wet weight of the adult sporophytes were 211 cm, 48.8 cm and 373 g, respectively. The sporophytic blades were uniformly smooth without wrinkles on both sides of the midrib, indicating top quality of the products. Amplified fragment length polymorphism analyses confirmed the identical genotypes of sporophytic offspring. These results suggested that this novel variety breeding and sporeling production method could serve as an efficient alternative to the traditional breeding technique for U. pinnatifida and possibly other commercial kelps that have identical life cycles.

Fucoidan extract derived from Undaria pinnatifida inhibits angiogenesis by human umbilical vein endothelial cells

In recent years, anti-angiogenic therapy has become an effective strategy for inhibiting tumor growth. Fucoidan is a class of fucose-enriched sulfated polysaccharides found in brown algae, and it is known to have strong anti-tumor property. Using a human umbilical vein endothelial cells (HUVEC)-based cell culture model, the present study investigated the anti-angiogenic activity of fucoidan extracted from the brown seaweed Undaria pinnatifida. Treatment of HUVECs with various concentrations of fucoidan resulted in significant inhibition of cell proliferation, cell migration, tube formation and vascular network formation. However, significant inhibition of cell proliferation only occurred with longer treatment time (48h instead of 24h or less). About 40% of cell proliferation and cell migration and 61% of tube formation by HUVECs were inhibited by 400μg/ml fucoidan, the maximum concentration tested. These results appeared to suggest that modulation of angiogenesis by fucoidan might not occur through growth inhibition and apoptosis. Ex vivo angiogenesis assay demonstrated that at 100μg/ml, fucoidan caused significant reduction in microvessel outgrowth. Western blot and RT-PCR analyses indicated that at 400μg/ml, fucoidan significantly reduced the expression of the angiogenesis factor VEGF-A in the suppression of angiogenesis activity. Our results showed that fucoidan isolated from U. pinnatifida may have a new therapeutic potential in the prevention angiogenesis-related diseases.

In vitro cytotoxic effect of ethanol extract prepared from sporophyll of Undaria pinnatifida on human colorectal cancer cells.

Brown seaweed Undaria pinnatifida (Harvey) Suringar is popular as a foodstuff, and used for medical care in East Asian countries. The major components of this seaweed are shown to benefit hypertension and hyperlipidemia, and considered to reduce the risks of infarction and ischemic diseases. Furthermore, the intake of dietary fiber of seaweeds is considered to prevent the production and proliferation of cancer in the gastrointestinal tract. The direct effect of an ethanol extract prepared from Undaria pinnatifida sporophyll (mekabu) on HCT116 human colorectal cancer cells was examined, and the mekabu extract was shown to induce the non-oxidative apoptotic damage to the cells, thus resulting in the reduction of their viabilities in a concentration-dependent manner. Moreover, the cytotoxic effects of carcinostatic drugs, such as 5-fluorouracil (5-FU) and irinotecan (CPT-11), were observed only in the medium containing sera, while the mekabu extract could effectively reduce the cell viabilities even in the serum-free medium. These findings suggest that the mekabu extract may contain a potential active substance inducing the non-oxidative apoptotic cell death probably through a mechanism different from those of 5-FU and CPT-11, and hence mekabu is possibly useful as an auxiliary drug to the chemotherapy of colorectal cancer.

Anti-Edema Effects of Brown Seaweed (Undaria pinnatifida) Extract on Phorbol 12-Myristate 13-Acetate-Induced Mouse Ear Inflammation

The brown seaweed Undaria pinnatifida (Harvey) Suringar is used in traditional medicine to treat fever, urination problems, lumps and swelling, and as a dietary supplement for post-childbirth women. We examined the anti-inflammatory activities of the seaweed. The methanol extract of the seaweed was active against mouse ear edema induced by phorbol myristate acetate (PMA), with an IC(50) of 10.3 mg/ml. The extract reduced the edema to a half-maximal level when applied at the concentration of 40 mg/ml within 3 hours before or 2 hours after application of PMA. Extract taken from the blade section of the seaweed demonstrated the highest activity. The Northern form of U. pinnatifida was more active than the Southern form. In the analgesic test, the methanol extract suppressed the acetic acid-induced writhing response, with an IC(50) of 0.48 g/kg body weight. The extract also demonstrated antipyretic activity in yeast-induced hyperthermic mice. Activity-related constituents were arachidonic, eicosapentaenoic, and stearidonic acids.

Antioxidant activity of enzymatic extracts from the brown seaweed Undaria pinnatifida by electron spin resonance spectroscopy

Enzyme assisted extraction of Undaria pinnatifida was performed using five proteases (Alcalase, Flavourzyme, Neutrase, Trypsin and Protamex) and six carbohydrases (AMG, Dextrozyme, Maltogenase, Promozyme, Viscozyme and Celluclast) in order to acquire extracts rich in antioxidants. Antioxidant activity of the extracts was investigated using electron spin resonance (ESR) spectroscopy on 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl and superoxide radicals. Extracts exhibited strong radical scavenging activity on DPPH and hydroxyl radical, and activity increased with increasing extract concentration. In contrast to DPPH and hydroxyl radical scavenging activity, extracts showed weak scavenging activity on superoxide radical. Extracts exhibited weak radical scavenging activity compared to vitamin C as a reference compound. However, they are still a good source of antioxidant materials that are safe for consumption and show water-soluble properties.
