In vitro assessment of the effect of magnetic fields on efficacy of biosynthesized selenium nanoparticles by Alborzia kermanshahica

Cyanobacteria represent a rich resource of a wide array of unique bioactive compounds that are proving to be potent sources of anticancer drugs. Selenium nanoparticles (SeNPs) have shown an increasing potential as major therapeutic platforms and led to the production of higher levels of ROS that can present desirable anticancer properties. Chitosan–SeNPs have also presented antitumor properties against hepatic cancer cell lines, especially the Cht-NP (Chitosan–NPs), promoting ROS generation and mitochondria dysfunction. It is proposed that magnetic fields can add new dimensions to nanoparticle applications. Hence, in this study, the biosynthesis of SeNPs using Alborzia kermanshahica and chitosan (CS) as stabilizers has been developed. The SeNPs synthesis was performed at different cyanobacterial cultivation conditions, including control (without magnetic field) and magnetic fields of 30 mT and 60 mT. The SeNPs were characterized by uv-visible spectroscopy, Fourier-transform infrared spectroscopy (FT-IR), Dynamic light scattering (DLS), zeta potential, and TEM. In addition, the antibacterial activity, inhibition of bacterial growth, minimum inhibitory concentration (MIC), and minimum bactericidal concentration (MBC), as well as the antifungal activity and cytotoxicity of SeNPs, were performed. The results of uv-visible spectrometry, DLS, and zeta potential showed that 60 mT had the highest value regarding the adsorption, size, and stabilization in compared to the control. FTIR spectroscopy results showed consistent spectra, but the increased intensity of peaks indicates an increase in bond number after exposure to 30 mT and 60 mT. The results of the antibacterial activity and the inhibition zone diameter of synthesized nanoparticles showed that Staphylococcus aureus was more sensitive to nanoparticles produced under 60 mT. Se-NPs produced by Alborzia kermanshahica cultured under a 60 mT magnetic field exhibit potent antimicrobial and anticancer properties, making them a promising natural agent for use in the pharmaceutical and biomedical industries.

Highly elastic, fatigue-resistant, antibacterial, conductive, and nanocellulose-enhanced hydrogels with selenium nanoparticles loading as strain sensors

The fabrication of highly elastic, fatigue-resistant and conductive hydrogels with antibacterial properties is highly desirable in the field of wearable devices. However, it remains challenging to simultaneously realize the above properties within one hydrogel without compromising excellent sensing ability. Herein, we fabricated a highly elastic, fatigue-resistant, conductive, antibacterial and cellulose nanocrystal (CNC) enhanced hydrogel as a sensitive strain sensor by the synergistic effect of biosynthesized selenium nanoparticles (BioSeNPs), MXene and nanocellulose. The structure and potential mechanism to generate biologically synthesized SeNPs (BioSeNPs) were systematically investigated, and the role of protease A (PrA) in enhancing the adsorption between proteins and SeNPs was demonstrated. Additionally, owing to the incorporation of BioSeNPs, CNC and MXene, the synthesized hydrogels showed high elasticity, excellent fatigue resistance and antibacterial properties. More importantly, the sensitivity of hydrogels determined by the gauge factor was as high as 6.24 when a high strain was applied (400–700 %). This study provides a new horizon to synthesize high-performance antibacterial and conductive hydrogels for soft electronics applications.

Biogenic selenium nanoparticles and selenium/chitosan-Nanoconjugate biosynthesized by Streptomyces parvulus MAR4 with antimicrobial and anticancer potential

BackgroundAs antibiotics and chemotherapeutics are no longer as efficient as they once were, multidrug resistant (MDR) pathogens and cancer are presently considered as two of the most dangerous threats to human life. In this study, Selenium nanoparticles (SeNPs) biosynthesized by Streptomyces parvulus MAR4, nano-chitosan (NCh), and their nanoconjugate (Se/Ch-nanoconjugate) were suggested to be efficacious antimicrobial and anticancer agents.ResultsSeNPs biosynthesized by Streptomyces parvulus MAR4 and NCh were successfully achieved and conjugated. The biosynthesized SeNPs were spherical with a mean diameter of 94.2 nm and high stability. Yet, Se/Ch-nanoconjugate was semispherical with a 74.9 nm mean diameter and much higher stability. The SeNPs, NCh, and Se/Ch-nanoconjugate showed significant antimicrobial activity against various microbial pathogens with strong inhibitory effect on their tested metabolic key enzymes [phosphoglucose isomerase (PGI), pyruvate dehydrogenase (PDH), glucose-6-phosphate dehydrogenase (G6PDH) and nitrate reductase (NR)]; Se/Ch-nanoconjugate was the most powerful agent. Furthermore, SeNPs revealed strong cytotoxicity against HepG2 (IC50 = 13.04 μg/ml) and moderate toxicity against Caki-1 (HTB-46) tumor cell lines (IC50 = 21.35 μg/ml) but low cytotoxicity against WI-38 normal cell line (IC50 = 85.69 μg/ml). Nevertheless, Se/Ch-nanoconjugate displayed substantial cytotoxicity against HepG2 and Caki-1 (HTB-46) with IC50 values of 11.82 and 7.83 μg/ml, respectively. Consequently, Se/Ch-nanoconjugate may be more easily absorbed by both tumor cell lines. However, it exhibited very low cytotoxicity on WI-38 with IC50 of 153.3 μg/ml. Therefore, Se/Ch-nanoconjugate presented the most anticancer activity.ConclusionThe biosynthesized SeNPs and Se/Ch-nanoconjugate are convincingly recommended to be used in biomedical applications as versatile and potent antimicrobial and anticancer agents ensuring notable levels of biosafety, environmental compatibility, and efficacy.

Antibacterial and antioxidant activities of a novel biosynthesized selenium nanoparticles using Rosa roxburghii extract and chitosan: Preparation, characterization, properties, and mechanisms

Developing efficient and safe antibacterial agents to inhibit pathogens including Physalospora piricola and Staphylococcus aureus is of great importance. Herein, a novel compound composed of Rosa roxburghii procyanidin, chitosan and selenium nanoparticle (RC-SeNP) was bio-synthesized, with the average diameter and zeta potential being 84.56 nm and −25.60 mV, respectively. The inhibition diameter of the RC-SeNP against P. piricola and S. aureus reached 18.67 mm and 13.13 mm, and the maximum scavenging activity against DPPH and ABTS reached 96.02% and 98.92%, respectively. Moreover, the RC-SeNP completely inhibited the propagation P. piricola and S. aureus on actual apples, suggesting excellent in vivo antimicrobial capacity. The transcriptome analysis and electron microscope observation indicated that the antibacterial activity would be attributed to adhering to and crack the cell walls as well as damage the cytomembrane and nucleus. Moreover, the RC-SeNP effectively maintained the vitamin C, total acid, and water contents of red bayberry, demonstrating potential application for fruit preservation. At last, the RC-SeNP showed no cell toxicity and trace selenium residual dose (0.03 mg/kg on apple, 0.12 mg/kg on red bayberry). This study would enlighten future development on novel nano-bioantibacterial agents for sustainable agriculture.

Supplementing broiler diets with bacterial selenium nanoparticles enhancing performance, carcass traits, blood indices, antioxidant status, and caecal microbiota of Eimeria tenella-infected broiler chickens

Nanomedicine is a critical therapeutic approach for treating most poultry illnesses, particularly parasitic infections. Coccidiosis is a severe protozoan infection affecting poultry; the emergence of drug-resistant Eimeria strains demands the development of new, safe therapies. Consequently, the objective of this work was to investigate the efficacy of the biosynthesized selenium nanoparticles (SeNPs) byPaenibacillus polymyxa (P. polymyxa) against Eimeria tenella (E. tenella) experimental infection in broiler chickens. The prepared SeNPs absorbed the UV at 270 nm were spherical with a size of 26 nm, and had a surface negative charge of -25 mV. One hundred and fifty, 1-day-old male broiler chicks were randomly allocated into 5 groups (30 birds/group with triplicates each) as follows: T1: negative control (noninfected and nontreated with SeNPs);T2: delivered SeNPs (500 µg/kg diet) for 35 successive days, T3: E. tenella-infected (positive control birds), T4: E. tenella-infected and treated with SeNPs (500 µg/kg diet) and T5: E. tenella-infected chicks and treated with anticoccidial agent (sulfadimidine, 16% solution 8 mL/L of drinking water) for 5 successive days. At 14 d of age, each bird in infected groups was orally treated with 3×103 sporulated oocyst of E. tenella. SeNPs considerably decreased the number of oocysts in broiler feces compared to positive control and anticoccidial drug, followed by a substantial reduction of parasite phase count in the cecum (15, 10, and 8 for meronts, gamonts, and developing oocysts) when compared with positive control birds. The Eimeria experimental infection lowered the activity of antioxidant enzymes, superoxide dismutase (SOD), glutathione peroxidase (GPx), and reduced glutathione (GSH) while increasing the stress parameters nitric oxide (NO) and malonaldehyde (MDA). Moreover, the production of proinflammatory (TNF-α and IL-6) and apoptotic genes (BcL2 and Cas-3) were significantly elevated. Administrating SeNPs to chicks significantly decreased oxidative stress, inflammation, and apoptotic markers in the cecum tissue. Therefore, growth performance, carcass weights, antioxidant enzymes, and blood properties of infected chicks were enhanced. The findings compared the protecting role of Se-nanoparticles against cecum damages in E. tenella-infected broilers.

New investigation of anti-inflammatory activity of Polycladia crinita and biosynthesized selenium nanoparticles: isolation and characterization

BackgroundMarine macroalgae have gained interest recently, mostly due to their bioactive components. Polycladia crinita is an example of marine macroalgae from the Phaeophyceae class, also known as brown algae. They are characterized by a variety of bioactive compounds with valuable medical applications. The prevalence of such naturally active marine resources has made macroalgae-mediated manufacturing of nanoparticles an appealing strategy. In the present study, we aimed to evaluate the antioxidant and anti-inflammatory features of an aqueous extract of Polycladia crinita and biosynthesized P. crinita selenium nanoparticles (PCSeNPs) via a carrageenan-induced rat paw edema model. The synthesized PCSeNPs were fully characterized by UV–visible spectroscopy, FTIR, XRD, and EDX analyses.ResultsFTIR analysis of Polycladia crinita extract showed several sharp absorption peaks at 3435.2, 1423.5, and 876.4 cm−1 which represent O–H, C=O and C=C groups. Moreover, the most frequent functional groups identified in P. crinita aqueous extract that are responsible for producing SeNPs are the –NH2–, –C=O–, and –SH– groups. The EDX spectrum analysis revealed that the high percentages of Se and O, 1.09 ± 0.13 and 36.62 ± 0.60%, respectively, confirmed the formation of SeNPs. The percentages of inhibition of the edema in pretreated groups with doses of 25 and 50 mg/kg, i.p., of PCSeNPs were 62.78% and 77.24%, respectively. Furthermore, the pretreated groups with 25, 50 mg/kg of P. crinita extract displayed a substantial decrease in the MDA levels (P < 0.00, 26.9%, and 51.68% decrease, respectively), indicating potent antioxidant effect. Additionally, the pretreated groups with PCSeNPs significantly suppressed the MDA levels (P < 0.00, 54.77%, and 65.08% decreases, respectively). The results of immune-histochemical staining revealed moderate COX-2 and Il-1β expressions with scores 2 and 1 in rats pre-treated with 25 and 50 mg/kg of free extract, respectively. Additionally, the rats pre-treated with different doses of PCSeNPs demonstrated weak COX-2 and Il-1β expressions with score 1 (25 mg/kg) and negative expression with score 0 (50 mg/kg). Both antioxidant and anti-inflammatory effects were dose-dependent.ConclusionsThese distinguishing features imply that this unique alga is a promising anti-inflammatory agent. Further studies are required to investigate its main active ingredients and possible side effects.

Selenium-Fortified Kombucha-Pollen Beverage by In Situ Biosynthesized Selenium Nanoparticles with High Biocompatibility and Antioxidant Activity.

Biogenic selenium nanoparticles (SeNPs) have been shown to exhibit increased bioavailability. Fermentation of pollen by a symbiotic culture of bacteria and yeasts (SCOBY/Kombucha) leads to the release of pollen content and enhances the prebiotic and probiotic effects of Kombucha. The aim of this study was to fortify Kombucha beverage with SeNPs formed in situ by Kombucha fermentation with pollen. Response Surface Methodology (RSM) was used to optimize the biosynthesis of SeNPs and the pollen-fermented Kombucha beverage. SeNPs were characterized by Transmission electron microscopy energy-dispersive X-ray spectroscopy (TEM-EDX), Fourier-transform infrared spectroscopy (FTIR), Dynamic light scattering (DLS), and Zeta potential. The pollen-fermented Kombucha beverage enriched with SeNPs was characterized by measuring the total phenolic content, antioxidant activity, soluble silicon, saccharides, lactic acid, and the total content of Se0. The polyphenols were identified by liquid chromatography-mass spectrometry (LC-MS). The pollen and the bacterial (nano)cellulose were characterized by scanning electron microscopy-energy dispersive X-ray spectroscopy (SEM-EDX), FTIR, and X-Ray diffraction (XRD). We also assessed the in vitro biocompatibility in terms of gingival fibroblast viability and proliferation, as well as the antioxidant activity of SeNPs and the pollen-fermented Kombucha beverage enriched with SeNPs. The results highlight their increased biological performance in this regard.

Biosynthesized Selenium Nanoparticles Using Epigallocatechin Gallate Protect against Pentylenetetrazole-Induced Acute Epileptic Seizures in Mice via Antioxidative, Anti-Inflammatory, and Anti-Apoptotic Activities.

Several negative outcomes are associated with current anti-epileptic medications. Epigallocatechin gallate (EGCG) is a plant-derived compound called catechin and has many medicinal activities, such as anti-inflammatory and antioxidant activities. Biosynthesized selenium nanoparticles are also showing their neuroprotective effect. The anti-epileptic effect of EGCG, alone or with SeNPs, is still debated. Here, we aimed to investigate the potential anti-seizure effect of biosynthesized SeNPs using EGCG (EGCG-SeNPs) against epileptic seizures and hippocampal damage, which is enhanced by pentylenetetrazole (PTZ) injection in mice. Mice were grouped as follows: control; PTZ-exposed group (epileptic model); EGCG + PTZ-treated group; sodium selenite (Na2SeO3) + PTZ-treated group; EGCG-SeNPs + PTZ-treated group; and valproic acid (VPA) + PTZ-treated group. EGCG-SeNPs administration showed anti-epileptic activity by increasing the latency time and reducing the seizure duration following the PTZ injection. Additionally, EGCG-SeNPs counteracted the PTZ-induced changes in oxidants and antioxidants. Moreover, EGCG-SeNPs inhibited the inflammatory response by suppressing the release of pro-inflammatory cytokines and decreasing the immunoreactivity of the glial fibrillary acidic protein and mRNA expression of glutamate receptor subunit zeta-1 (NMDAR; Grin1), showing their inhibitory effect on epilepsy-associated inflammation. Moreover, EGCG-SeNPs reduced PTZ-induced neuronal apoptosis, as indicated by a reduction in the levels of pro-apoptotic proteins and an elevation of the anti-apoptotic protein. Moreover, EGCG-SeNPs administration significantly modulated the PTZ-induced changes in monoamine levels and acetylcholinesterase activity in the hippocampal tissue. The obtained findings suggest the anti-seizure activity of EGCG-SeNPs via their antioxidant, anti-inflammatory, and anti-apoptotic effects, along with their neuromodulatory effect.

Effect of Biosynthesized Nanoselenium on Controlling Tomato Root-Knot Nematode Meloidogyne incognita

Tomato is a globally important fruit crop, which is easily susceptible to infection by plant-parasitic nematodes during growth. In this study, selenium nanoparticles were biosynthesized through the selenite reduction pathway in a wild-type Pseudomonas stutzeri BB19 and were characterized by uniform nanostructured needle-like forms with an average size of 95.2 nm. The nematicidal effect of biosynthesized selenium nanoparticles (BioSeNPs) at different concentrations (2, 6, and 10 ppm) during the pre- or post-infection of tomato root-knot nematode (RKN), Meloidogyne incognita, was assessed under greenhouse conditions. The BioSeNPs were applied as a foliar spray or a soil drench, compared to ethoprophos nematicide (100 mL/pot). The results showed that the ethoprophos nematicide significantly reduced the number of galls, egg mass, and eggs/egg mass of M. incognita by 94.2, 92.8, and 49.8%, respectively. BioSeNPs, as foliar sprays, significantly reduced the number of galls in post-treated infected tomato plants at 10 ppm and in pre-treated infected tomato plants at 2 ppm by 91.9 and 91.4%, respectively. Concerning the egg mass, BioSeNPs significantly reduced it in pre-treated infected tomato plants at 2 ppm as a foliar spray by 90.9%. Moreover, BioSeNPs significantly reduced the eggs/egg mass in pre-treated infected tomato plants at 2 ppm as a soil drench by 43.3%. On the other hand, the BioSeNPs considerably improved tomato growth, chlorophyll a and b, carotenoid content, and enzymes (i.e., catalase and peroxidase) activity compared to untreated infected tomato plants (negative control). Hence, the BioSeNPs show a significant application potential as a cost-effective and environmentally friendly biocontrol agent for RKN management in tomato plants.

Pharmacological studies of Selenium Nanoparticles synthesized from Syzygium aromaticum

Selenium is an essential element and if taken in trace amounts, then it plays an important role in plant and animal development and reproduction. When compared to other methods of selenium nanoparticles (SeNPs) production, green synthesis using plant extracts has a better probability of eliminating its harmful character and producing beneficial SeNPs. The current work used Syzygium aromaticum to perform biogenic synthesis of SeNPs. The biosynthesized SeNPs were characterized by UV-Vis spectroscopy, X-Ray Diffraction, FTIR, AAS, SEM-EDX and FESEM. The biosynthesized SeNPs showed maximum peak at 276 nm and FESEM image exhibited greater numbers of the agglomerated spherical particles within the diameter range 30-55 nm. The antioxidant activity was measured in vitro using the radical scavenging (DPPH) test, total antioxidant (FRAP) assay and intracellular reactive oxygen species (ROS) assay against the A375 cell line. The formation of free radicals of selenium elements as antioxidant qualities prevented the growth of A375 skin cancer cells. In vitro tests revealed that SeNPs had the highest percentage of inhibition. As a consequence, the current study's findings showed that the lowest concentrations of SeNPs had better antioxidant effects. MTT experiment was used to assess the cytotoxicity of SeNPs against A375 cell lines exhibiting an IC50 value of 27±1.2 g/ mL. Synthesized SeNPs showed less toxicity towards the investigated cell line which remained more than 50% viability at 48 hours of exposure. In vitro anthelmintic activity data demonstrate that the worms experienced paralysis and death time at 25.03± 0.12 and 60.12±1.90 minutes respectively. These SeNPs with diverse objectives may exhibit increased absorption by functionalization and simultaneous drug loading as well as agnostic characteristics in the future.

Anticancer activity of lactoferrin-coated biosynthesized selenium nanoparticles for combating different human cancer cells via mediating apoptotic effects

The present study aims to develop a novel nanocombination with high selectivity against several invasive cancer cells, sparing normal cells and tissues. Bovine lactoferrin (bLF) has recently captured the interest of numerous medical fields owing to its biological activities and well-known immunomodulatory effects. BLF is an ideal protein to be encapsulated or adsorbed into selenium nanocomposites (Se NPs) in order to produce stable nanocombinations with potent anticancer effects and improved immunological functions. The biosynthesis of the functionalized Se NPs was achieved using Rhodotorula sp. strain MZ312359 via a simultaneous bio-reduction approach to selenium sodium salts. The physicochemical properties of Se NPs using SEM, TEM, FTIR, UV Vis, XRD, and EDX confirmed the formation of uniform agglomerated spheres with a size of 18–40 nm. Se NPs were successfully embedded in apo-LF (ALF), forming a novel nanocombination of ALF-Se NPs with a spherical shape and an average nanosize of less than 200 nm. The developed ALF-Se NPs significantly displayed an effective anti-proliferation efficiency against many cancer cells, including MCF-7, HepG-2, and Caco-2 cell lines, as compared to Se NPs and ALF in free forms. ALF-Se NPs showed a significant selectivity impact (> 64) against all treated cancer cells at IC50 63.10 ≤ μg/mL, as well as the strongest upregulation of p53 and suppression of Bcl-2, MMP-9, and VEGF genes. Besides, ALF-Se NPs were able to show the maximum activation of transcrition of key redox mediator (Nrf2) with suppression in reactive oxygen species (ROS) levels inside all treated cancer cells. This study demonstrates that this novel nanocombination of ALF-Se NPs has superior selectivity and apoptosis-mediating anticancer activity over free ALF or individual form of Se NPs.

Exploring the antimicrobial, antioxidant, anticancer, biocompatibility, and larvicidal activities of selenium nanoparticles fabricated by endophytic fungal strain Penicillium verhagenii

Herein, four endophytic fungal strains living in healthy roots of garlic were used to produce selenium nanoparticles (Se-NPs) via green synthesis. Penicillium verhagenii was found to be the most efficient Se-NPs producer with a ruby red color that showed maximum surface plasmon resonance at 270 nm. The as-formed Se-NPs were crystalline, spherical, and well-arranged without aggregation, and ranged from 25 to 75 nm in size with a zeta potential value of −32 mV, indicating high stability. Concentration-dependent biomedical activities of the P. verhagenii-based Se-NPs were observed, including promising antimicrobial activity against different pathogens (Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, Staphylococcus aureus, Candida albicans, C. glabrata, C. tropicalis, and C. parapsilosis) with minimum inhibitory concentration (MIC) of 12.5–100 µg mL–1. The biosynthesized Se-NPs showed high antioxidant activity with DPPH-scavenging percentages of 86.8 ± 0.6% at a concentration of 1000 µg mL–1 and decreased to 19.3 ± 4.5% at 1.95 µg mL–1. Interestingly, the Se-NPs also showed anticancer activity against PC3 and MCF7 cell lines with IC50 of 225.7 ± 3.6 and 283.8 ± 7.5 µg mL–1, respectively while it is remaining biocompatible with normal WI38 and Vero cell lines. Additionally, the green synthesized Se-NPs were effective against instar larvae of a medical insect, Aedes albopictus with maximum mortality of 85.1 ± 3.1, 67.2 ± 1.2, 62.10 ± 1.4, and 51.0 ± 1.0% at a concentration of 50 µg mL–1 for I, II, III, and IV-instar larva, respectively. These data highlight the efficacy of endophytic fungal strains for cost-effective and eco-friendly Se-NPs synthesis with different applications.

In-depth in-vitro and in-vivo anti-diabetic evaluations of Fagonia cretica mediated biosynthesized selenium nanoparticles

Therapeutic moieties derived from medicinal plants as well as plants-based ecofriendly processes for producing selenium nanoparticles have shown great promise in the management of type 2 diabetes mellitus (T2DM). The current study was aimed to assess the anti-diabetic potentials of Fagonia cretica mediated biogenic selenium nanoparticles (FcSeNPs) using in-vitro and in-vivo approaches. The bio-synthesized FcSeNPs were characterized using various techniques including UV–VIS spectrophotometry and FTIR analysis. The in-vitro efficacy of FcSeNPs were assessed against α-glucosidase, α-amylase enzymes as well as the anti-radical studies were performed using DPPH and ABTS free radicals scavenging assays. For in-vivo studies, 20 Male Balb/C albino-mice were randomly divided into 4 groups (n = 5) including normal group, disease group (Diabetic group with no treatment), control group and treatment group (Diabetic group treated with FcSeNPs). Further, biochemistry markers including pancreas, liver, kidney and lipid profile were assessed for all treatment groups. The FcSeNPs exhibited a dose-dependent inhibition against α-amylase and α-glucosidase at 62–1000 µg mL−1 concentration with IC50 values of 92 and 100 µg mL−1 respectively. In antioxidant experiments, the FcSeNPs demonstrated significant radicals scavenging effect against DPPH and ABTS radicals. In STZ-induced diabetic mice, a considerable decline in blood glucose level was observed after treatment with FcSeNPs. Anti-hyperglycemic effect of FcSeNPs treated animals were high (105 ± 3.22**) as compared to standard drug (128.6 ± 2.73** mg dL−1). Biochemical investigations revealed that all biochemical parameters for pancreas, liver function, renal function panel and lipid profile were significantly lowered in FcSeNPs treated animals. Our findings indicate a preliminary multi-target efficacy for FcSeNPs against type-2 diabetes and thus warrant further detailed studies.

Recent clinical and experimental study findings have established oxidative stress as one of the main pathogeneses of PD. Selenium, a trace metals with antioxidant effects, might reverse the neurobehavioral impairments and oxidative stress in rats. Thus, the goal of this study was to ascertain if Selenium Nano Particles (SeNPs) are also effective to protect brain cells from oxidative stress or not. SeNPs were synthesized utilizing Ascorbic acid and chitosan as a reducing and stabilizing agent. Next, eight groups (N: 6) of male Wistar rats were randomly assigned and injected by different dosage (0.1, 0,2, and 0.3mg/kg) of Se and SeNP. Finally, to ascertain the protective benefits of SeNP on PD rats, behavioral evaluation, clinical symptoms, antioxidant activity, and oxidant levels were examined. According to the findings, PD rats' motor functions had developed by SeNP injection. Higher MDA levels and inhibited antioxidant activities (SOD, CAT, and GPX) in lesion group are highlighting the significant role of oxidative stress in dopaminergic neuron death and neurobehavioral abnormalities. SeNP also protect against oxidative stress as compared to the lesion group. The levels of MDA had greatly reduced while the activities of enzymes, TAC, and SeNP both had significantly increased. By enhancing antioxidant activity, administration of SeNP can reduce the hazardousconsequences of oxidative stress.

Antimicrobial and Photocatalytic Activities of Selenium Nanoparticles Synthesized from Elaeagnus indica Leaf Extract

Selenium nanoparticles (Se NPs) have recently received much interest due to their low toxicity, high bioavailability, and wide applications. This study synthesized Se NPs using selenious acid as a starting material and leaf extract from Elaeagnus indica as a reducing agent. Spectroscopic and electron microscopy investigations have demonstrated the production of aggregated amorphous Se NPs with phytochemicals. Furthermore, the reduction of selenious acid into Se NPs by phytochemicals present in the leaf extract of E. indica was confirmed in a prominent band at 269 nm in the UV-visible spectrum. The biosynthesized selenium nanoparticles have a 10–15 nm particle size distribution. The agar well diffusion assay exhibited remarkable dose-dependent, wide-spectrum antimicrobial efficacy of the Se NPs against all the tested microorganisms. Moreover, the lowest minimum inhibitory concentration (10 µg/mL) was noted against Salmonella Typhimurium and Fusarium oxysporum. The prepared Se NPs degraded methylene blue dye by about 89% after 6 h of exposure to sunlight. In conclusion, the synthesis of Se NPs using E. indica leaf extract shows promise as a method for producing Se NPs with significant antimicrobial activity and potential for methylene blue photodegradation. These properties make them potentially valuable in various fields, including water treatment and biomedical applications, in the future.

Unravelling the antibacterial potential of biosynthesized selenium nanoparticles against Salmonella Typhimurium food pathogen: in vitro and in vivo investigation.

It is highly required to find novel alternatives to the antibiotics currently used due to the increasing dissemination of antibiotic resistance among bacteria, especially enteric bacteria. The current study aimed to produce selenium nanoparticles (SeNPs) by Euphorbia milii Des Moul leaves extract (EME). The produced SeNPs were characterized using different techniques. After that, in vitro and in vivo antibacterial activity against Salmonella typhimurium was elucidated. Moreover, phytochemical identification and quantification of the chemical compositions of EME were performed using HPLC. The broth microdilution method determined the minimum inhibitory concentrations (MICs). The MIC values of SeNPs ranged from 128 to 512 µg/mL. Additionally, the impact of SeNPs on membrane integrity and permeability was investigated. A marked decline in the membrane integrity and inner and outer membrane permeability was noticed in 50%, 46.15%, and 50% of the tested bacteria, respectively. Subsequently, a gastrointestinal tract infection model was used to study the in vivo antibacterial potential of SeNPs. Remarkably, treatment with SeNPs resulted in average-sized intestinal villi and colonic mucosa in the small intestine and caecum, respectively. In addition, it was revealed there was no inflammation or dysplasia in the studied tissues. SeNPs also enhanced the survival rate and significantly decreased the number of colony-forming units per gram tissues in the small intestine and caecum. Concerning the inflammatory markers, SeNPs significantly (p < 0.05) decreased interleukins (6 and 1β). The biosynthesized SeNPs revealed antibacterial potential in vivo and in vitro; however, this finding should be elucidated clinically in the future.

High antimicrobial, cytotoxicity, and catalytic activities of biosynthesized selenium nanoparticles using Crocus caspius extract

The green method for synthesizing various nanoparticles is defined as one of the environmentally friendly, promising, and safer technologies. In our study, the selenium nanoparticles (C@SeNPs) were synthesized by Crocus caspius aqueous extract. The existence of functional groups involved in the synthesis of SeNPs that were connected with bioactive compounds was confirmed by the FT-IR spectrum. The TGA curve confirmed about 60 % weight loss between 260 and 500 °C, implying biomolecules surround the metallic core. XRD analysis displayed the trigonal nature of SeNPs. The SEM and TEM images of C@SeNPs demonstrated semi-spherical in shape. EDX analysis identified the intense bond of selenium. Biosynthesized C@SeNPs were discovered to have considerable antioxidant and antibacterial activities on several strains of bacteria. An IC50 value of 67.63 ± 2.5 μg/ml was obtained for their iron-chelating activity. In addition, the fabricated SeNPs have a strong growth inhibitory effect on MCF-7 and AGS cancer cells. C@SeNPs exhibited effective antifungal activity against tested fungi strains and antileishmanial activity against promastigotes. Besides, our NPs were able efficiently to degrade methylene blue (MB) dye in the presence of NaBH4. Thus, the current findings suggest the benefits of using green technology to synthesize SeNPs with potential activity.

Biosynthesized selenium nanoparticles to rescue coccidiosis-mediated oxidative stress, apoptosis and inflammation in the jejunum of mice.

One of the most crucial approaches for treating human diseases, particularly parasite infections, is nanomedicine. One of the most significant protozoan diseases that impact farm and domestic animals is coccidiosis. While, amprolium is one of the traditional anticoccidial medication, the advent of drug-resistant strains of Eimeria necessitates the development of novel treatments. The goal of the current investigation was to determine whether biosynthesized selenium nanoparticles (Bio-SeNPs) using Azadirachta indica leaves extract might treat mice with Eimeria papillata infection in the jejunal tissue. Five groups of seven mice each were used, as follows: Group 1: Non-infected-non-treated (negative control). Group 2: Non-infected treated group with Bio-SeNPs (0.5 mg/kg of body weight). Groups 3-5 were orally inoculated with 1×103 sporulated oocysts of E. papillata. Group 3: Infected-non-treated (positive control). Group 4: Infected and treated group with Bio-SeNPs (0.5 mg/kg). Group 5: Infected and treated group with the Amprolium. Groups 4 and 5 daily received oral administration (for 5 days) of Bio-SeNPs and anticoccidial medication, respectively, after infection. Bio-SeNPs caused a considerable reduction in oocyst output in mice feces (97.21%). This was also accompanied by a significant reduction in the number of developmental parasitic stages in the jejunal tissues. Glutathione reduced (GSH), glutathione peroxidase (GPx), and superoxide dismutase (SOD) levels were dramatically reduced by the Eimeria parasite, whereas, nitric oxide (NO) and malonaldehyde (MDA) levels were markedly elevated. The amount of goblet cells and MUC2 gene expression were used as apoptotic indicators, and both were considerably downregulated by infection. However, infection markedly increased the expression of inflammatory cytokines (IL-6 and TNF-α) and the apoptotic genes (Caspase-3 and BCL2). Bio-SeNPs were administrated to mice to drastically lower body weight, oxidative stress, and inflammatory and apoptotic indicators in the jejunal tissue. Our research thus showed the involvement of Bio-SeNPs in protecting mice with E. papillata infections against jejunal damage.

Green synthesis, Characterization and Antimicrobial potential of Selenium Nanoparticles from Ocimum gratissimum

Ocimum gratissimum L. is a perennial herbaceous plant used in the treatment of fungal and bacterial infections. Green synthesis has provided cost effective, environment friendly procedure and raising safe strategies for the synthesis of nanoparticles. This study was aimed at investigating the potential of O. gratissimum for the synthesis of selenium nanoparticles (SeNPs) and their antimicrobial activities. Phytochemical screening on aqueous extract was carried out using standard procedures. Selenium nanoparticles was biosynthesized by O. gratissimum and characterized using Visual detection, UV-Visible spectroscopy, Scanning Electron Microscope, Transmission Electron Microscope, Energy dispersive X-ray, Fourier Transform Infra-red spectroscopy and X-ray diffraction spectroscopy. Antimicrobial activity of the biosynthesized selenium nanoparticles by O. gratissimum was done using agar well diffusion method. Saponins, tannins, cardiac glycosides, terpenoids and phenols were present. The biosynthesized SeNPs had a strong plasmon resonance band at 300 nm, changes in colour from dark brown to ruby red. The SeNPs were spherical and aggregated with varying shapes and size ranged from 20 – 50 nm. Strong signal of selenium element was observed. Hydroxyl, esters, aldehyde, alkane and amine are present and responsible for the efficient stabilization and bioreduction of Selenium nanoparticle. Furthermore, biosynthesized SeNPs by O. gratissimum (OGSeNPs) exhibited higher antimicrobial activity against both Gram ositive and Gram negative bacteria. Green synthesis of nanoparticles is a promising method in the biomedical field, due to its high bioactive components.

Oxidative stress generated due to photocatalytic activity of biosynthesized selenium nanoparticles triggers cytoplasmic leakage leading to bacterial cell death.

The present work investigates the role of oxidative stress generated at biosynthesized selenium nanoparticles (SeNPs) interface in defining the antimicrobial and anti-biofilm activity. To this end, SeNPs with average size of 119 nm were synthesized rapidly during the growth of Staphylococcus aureus using the principle of green chemistry. The synthesis of SeNPs was confirmed by using different biophysical techniques like UV-vis spectroscopy, X-ray diffraction (XRD), field-emission scanning electron microscope (FE-SEM), EDX and zeta potential analysis. The obtained data from antimicrobial study revealed strong antimicrobial activity against both Gram-positive bacteria like Bacillus subtilis (MTCC 441) and Gram-negative bacteria like Escherichia coli (MTCC 443) and anti-biofilm activity against biofilm forming bacteria. The mechanism behind antimicrobial activity of biosynthesized SeNPs was explored by evaluating the amount of reactive oxygen species (ROS) generated at SeNPs interface due to photocatalytic activity. The experimental data obtained altogether concluded that, the ROS generated at SeNPs interface put stress on bacterial cell membrane causing leakage of cytoplasmic contents, leading to bacterial cell death.
