Optimized Lipidomics Extraction of Sphingosine and Sphinganine from Optic Nerve for Signaling Studies.

Interconvertible sphingolipid metabolites represent germane constituents of eukaryotic membranes and are vital in the regulation of cellular homeostasis, proliferation, survival, and induction of autophagy. This protocol describes a step-by-step method for extractions of sphingosine and sphinganine from mammalian tissue samples, particularly from the murine optic nerve. These lipids are partitioned into a binary mixture of chloroform and methanol in a modified Bligh and Dyer method. This is followed with reverse phase ultrahigh-performance liquid chromatography fractionation with a C18+ column and subsequent tandem mass spectrometry (UHPLC-MS-MS) analysis of the biological abundance. These free sphingoid bases dissociate to form structurally distinctive carbocation product ions that can be confirmed with annotations of lipidomic databases or in-house fragmentation software.

Preferential solvation of p-nitroaniline in a binary mixture of chloroform and hydrogen bond acceptor solvents: the role of specific solute-solvent hydrogen bonding.

Preferential solvation of solvatochromic probe p-nitroaniline (PNA) has been studied in binary mixtures of chloroform with different hydrogen bond acceptor (HBA) solvents using the spectroscopic transition energy (ET). Analyses of the solvatochromic shifts of the absorption spectra of PNA in different neat solvents as a function of the solvent polarity parameters reveal the major contribution from the solvent dipolarity/polarizability and HBA basicity in the solvation of PNA. The event of preferential solvation in the chloroform-HBA binary mixtures and the preference of one solvent above the other in the solvation shell have been attributed to the hydrogen bond donor and acceptor ability of the solvent mixtures. HBA solvents form a strong hydrogen bond with the amino group while chloroform forms a hydrogen bond with the nitro group of PNA. This specific functional group recognition increases the local concentration at specific sites resulting in location specific preferential solvation and synergistic preferential solvation. Solvents with comparable polarity have been found to show significant synergistic behavior as a result of the formation of stronger solvent-solvent hydrogen bonded S1-S2 species. These propositions were found to be supported by theoretical solvation models, calculated HOMO-LUMO energy gaps, the effect of deuterated solvent on the extent of PS, and 1H NMR spectral analyses.

Ordered cylindrical micropatterned Petri dishes used as scaffolds for cell growth

Three-dimensional (3D) culture dish patterned with a microwell structure demonstrates a great application potential in biotechnology. This study reports on the easy fabrication of an ordered customizable honeycomb microwell array on the surface of polymer substrates including the commercial Petri dish to create a biological platform for cell culture. The fabrication method is based on a very simple solvent dip-coating technique and the methanol accumulation-induced phase separation in which a binary mixture of chloroform and methanol is used to induce a ternary solution and to guarantee the formation of the ordered pore array on the substrate. The surface topology of the honeycomb substrate is manipulated through varying the experimental conditions; notably, the obtained honeycomb structure is part of the substrate, which reveals an increase in the structure’s stability for the practical applications. Honeycomb-structured Petri dish fabricated using this method is applied as a scaffold for cell growth to demonstrate its potential in biomedical applications.

Solvent effects on the photophysical properties of poly[1,4-dihydroxyanthraquinoneimine-1,3-bis(phenylene-ester-methylene)tetramethyldisiloxane

Absorption and fluorescence spectra of a polyquinoneimine, PQI, built on 1,4-dihydroxyanthraquinone and a siloxane diamine, 1,3-bis(amino-phenylene-ester-methylene)tetramethyldisiloxane, have been investigated in solvents of different polarities. The effect of solvents on the spectral properties was investigated using Lippert–Mataga and Bakhshiev polarity functions and Catalán’s multiple linear regression approach. Absorption and fluorescence spectra in studied solvents exhibit hypsochromic and bathochromic shifts, respectively. The polarity of the solvent was the main parameter which changes the spectral properties of PQI. Also, the binary mixtures of chloroform with methanol and dimethyl sulfoxide were used to analyze the intermolecular interactions and preferential solvation. The preferential solvation parameters (local mole fraction (X2L), excess function (δs2) and preferential solvation constant (KPS)) were calculated from spectral data and discussed as a function of cosolvent content. The values of quantum yield, decreased linearly with increasing solvent polarity (for non-polar and polar solvents).

Non-Aqueous Spray Solvents and Solubility Effects in Desorption Electrospray Ionization

The use of non-aqueous solvents in desorption electrospray ionization mass spectrometry (DESI-MS) is explored by analyzing a set of 43 compounds using binary mixtures of chloroform, tetrahydrofuran, and acetonitrile as the spray solvent. Comparisons of data obtained from chloroform/tetrahydrofuran (1:1) and chloroform/acetonitrile (1:1) spray solvents with the standard aqueous-based spray solvent (methanol/water, 1:1) shows that the non-aqueous systems have practical value for DESI, especially in the analysis of hydrophobic compounds. Non-aqueous spray solvents were used to ionize thermometer molecules (benzyl pyridinium salts) and showed lower internal energies (softer DESI ionization compared with methanol/water, 1:1), a result that has parallels in known solvent effects in electrospray ionization and is explained by solvent effects on surface tension. Consideration of octanol/water partition coefficients (K(ow)) of the 43 analytes in the light of their DESI results reveals the importance of the solubility of analyte in the spray solvent in producing high quality mass spectra. This finding provides additional support for the droplet pick-up description of the DESI mechanism, which is based on analyte dissolution in the spray solvent, followed by splashing of subsequently arriving droplets in the liquid film to form microdroplets of dissolved analyte. DESI solvent optimization can be improved by the use of K(ow) of the analyte as an indication of the polarity of the most appropriate solvent system.

Two new polymorphs of diphenyl(4-pyridyl)methyl methacrylate

The title compound (D4PyMA), C(22)H(19)NO(2), exhibits polymorphism after crystallization by slow evaporation from a binary mixture of chloroform and hexane. Long needle-like crystals have an orthorhombic structure (space group Fdd2), with one molecule in the asymmetric unit, while small tablet-like crystals exhibit a monoclinic crystal structure (space group P2(1)/n), in which two independent but chemically identical molecules comprise the asymmetric unit. The bond lengths and angles are normal, while the torsion angles around the -C-O- bond linking the diphenyl(4-pyridyl)methyl and methacrylate groups show the flexibility of the molecule by way of packing effects. The two polymorphs both contain weak C-H...pi and C-H...O/N contacts but have different conformations.

The Properties of Polymer-Containing BeO Dispersions in an Acetone–Chloroform Binary Medium

The properties of Newtonian and concentrated (non-Newtonian) BeO dispersions in solutions of acidic poly(vinyl acetate-co-vinyl chloride) in binary mixtures of chloroform (a Lewis acid) and acetone (a Lewis base) of different compositions were studied. In contrast to systems characterized by strong acid–base interactions at the particle–medium interface, the role of the steric stabilization factor was found to be extremely low in the studied dispersions. Polymer-containing BeO dispersions in acetone had the highest kinetic stability due to the specific interactions of this solvent with BeO particles and polymer macromolecules contributing to the redispersion of particles and the formation of protective solvation shells. The obtained data on the effect of the dispersion medium composition on the rheological properties of concentrated BeO dispersions can be used for the selection of optimal regimes for preparation of pastes for high-temperature ceramics.

ULTRASONIC DESTRUCTION OF CHLOROFORM AND CARBON TETRACHLORIDE IN AQUEOUS SOLUTION

The sonochemical destruction of binary mixtures of chloroform and carbon tetrachloride in aqueous solution at 30 kHz has been studied. The influences of initial concentration, bicarbonate/chloride, and acoustical intensity on the destruction rates have been observed. It was found that the destruction of the two compounds appeared to follow pseudo-first-order reaction kinetics with the current experimental concentration range. The rate constant of chloroform for initial concentration as high as 100.4 mg/L decreased compared to initial concentration 36.13 μg/L, when the rate constants of carbon tetrachloride hardly changed. However, their destruction rates increased with increasing acoustical intensity and were independent of addition of bicarbonate or chloride. The resulting yield of ionic product (Cl−) for chloroform was determined.

Molecular interactions in chloroform‐diluent mixtures

AbstractPlausible stoichiometries for solvation and association complexes in binary mixtures of chloroform and one of six organic diluents (n‐hexane, p‐dioxane, acetone, diethyl ether, tri‐n‐butyl phosphate, or triethylamine) are examined parametrically. A range of stoichiometric assumptions based on hydrogen bonding are examined, but only a few such combinations yield statistically significant fits to the experimental vapor–liquid equilibria and NMR chemical shift data. Hydrogen bonding considerations and the “true” chemical species are much more important than nonspecific physical factors such as molecular sizes, shape differences, and regular solution terms. Thermodynamic equilibrium constants are presented for the most significant chloroform solvation complexes. Nonspecific dilution shift is important and more helpful in fitting NMR data than the assumption of a chloroform dimer. Physical effects and an orientation factor are used to predict excess enthalpy in chloroform/n‐hexane mixtures where no solvation or association reactions occur. Model parameters are used to predict excess molar volumes and enthalpies. Commonly reported stability constants are concentration dependent.

Thermodynamics of complex formation in chloroform-oxygenated solvent mixtures

Complex formation equilibria in binary mixtures of chloroform with dipropyl ether (PE), diisopropyl ether (IPE), methyl tert-butyl ether (MBE), tetrahydrofuran (THF). 1,4-dioxane (DOX), acetone (AC), and methyl acetate (MA) have been analyzed in detail using several association models. Vapor-liquid equilibria, excess enthalpy and excess heat capacity data for these mixtures have been correlated using a multiproperty global fitting procedure. The thermodynamic properties for chloroform +PE, +IPE, +MBE, +AC, and +MA are best correlated using the ideal association model while for chloroform +THF and +DOX the best model is an athermal solvation model where the Flory-Huggins expression for the species activity coefficients is considered. The model parameters, i.e., the equilibrium constant, enthalpies and heat capacities of complexation, were found to be reliable, well representing the chloroform-oxygenated solvent H-bonded complexes. A detailed discussion is given on the test proposed by McGlashan and Rastogi to decide whether the solution contains only 1∶1 complexes or 2∶1 complexes as well. The complex formation equilibria in chloroform mixtures is compared to those previously examined for halothane (2-bromo-2-chloro-1,1,1-trifluoroethane) mixed with the same oxygenated solvents. It was found that the H-bonds formed by halothane are stronger than those formed by chloroform.

Examination of the inert solvent assumption in the ideal associated solution model. Enthalpy of chloroform–triethylamine complex dissociation

We have made new measurements of the enthalpies of adding cyclohexane to mixtures of chloroform + triethylamine + cyclohexane at 25 °C. The results of these measurements have been analysed in terms of the ideal associated solution model to obtain ΔHθ for the dissociation of the chloroform-triethylamine complex. Our value of ΔHθ is in excellent agreement with a published value that was obtained from results of measurements on binary mixtures of chloroform + triethylamine. This agreement indicates that it is usefully accurate to treat mixtures of chloroform + triethylamine + cyclohexane as ideal mixtures of chloroform, triethylamine, cyclohexane, and chloroform-triethylamine complex. This result has important implications for studies of associated systems dissolved in "inert" solvents and helps to provide a connection between complex formation in dilute solutions and in binary systems (A + B, no solvent). Thermochemical investigations of nearly idela binary solvents. II. Standard heats of solution in systems of nonspecific interactions

Standard heats of solution at 25°C are reported for squalane in mixtures ofn-heptane + iso-octane and in binary mixtures of chloroform, carbon tetrachloride, and cyclohexane; for cyclohexane in chloroform + carbon tetrachloride; and for chloroform in carbon tetrachloride + cyclohexane. General equations based on simple mixing models are developed for the excess thermodynamic properties of a solute at infinite dilution in a binary solvent. For the systems studied, mixing equations based on volume fractions give better agreement than those based on mole fractions, and the agreement is further improved by the use of weighting factors based on the properties of the solute + solvent binary systems.
