Effect of temperature fluctuation on the physiological stress response of hybrid pearl gentian grouper during waterless keeping alive.

Temperature fluctuations are inevitable and have an important impact on the survival of fish during transportation. Therefore, the effect of temperature fluctuation (15 ± 1 °C, 15 ± 2 °C, 15 ± 3 °C) on the muscle quality, physiological, and immune function of hybrid pearl gentian grouper before waterless keeping alive, during keeping alive (0 h, 3 h, 6 h, 9 h, 12 h), and after revival for 12 h was investigated. The plasma glucose concentration of grouper gradually decreased to 0.645 ± 0.007 mg/mL, 0.657 ± 0.006 mg/mL, and 0.677 ± 0.004 mg/mL after keeping alive for 12 h under different temperature fluctuations of 15 ± 1 °C, 15 ± 2 °C, and 15 ± 3 °C, respectively. The cortisol concentration and lysozyme activity of pearl gentian grouper significantly increased (P < 0.05) during the keeping alive period. The results suggested that fish bodies would produce acute stress response, strengthen immune defense ability, and quickly consume a lot of energy to adapt to the low-temperature anhydrous environment. In all treatment groups, the activities of plasma alanine transaminase (ALT) and aspartate aminotransferase (AST) and the content of creatinine gradually increased with the prolongation of the survival time. The hardness and springiness of muscle decreased from 5965.99 ± 20.15 and 0.90 ± 0.00 to 3490.69 ± 27.59 and 0.42 ± 0.01, respectively. In the meanwhile, the change of glycogen and lactic acid content was opposite, indicating that temperature fluctuation harmed the liver, kidney function, and muscle quality. In the later stage of keeping alive, the superoxide dismutase (SOD) and catalase (CAT) activities decreased, especially in the temperature fluctuation group of ±3 °C (125.99 ± 5.48 U/mgprot, 44.21 ± 0.63 U/mgprot), leading to an imbalance of fish immunity. In summary, higher temperature fluctuation would influence the physiological function and immune defense ability and decrease the quality of pearl gentian grouper.

Ecotoxicological Assessment of Ponds Contaminated by Wastewater of Livestock Enterprises by Fish Biomarkers

The morphometric characteristics of internal organs, the morphological composition of blood, and indicators of metabolic processes in the blood plasma and hepatopancreas of the two-year-old carp (&lt;i&gt;Cyprinus carpio&lt;/i&gt; L.) from the conditionally clean pond and contaminated with sewage from a livestock enterprise pond were examined. It was established that the considered contamination caused increase of the spleen mass and its index respectively 1.63 and 1.65 times, but did not affect live weight, body length and height, body structure index, hepatopancreas mass and its index. The hemoglobin concentration, number of erythrocytes, thrombocytes and monocytes in blood of carps from the contaminated pond was lower than in control, whereas number of the segmented neutrophils and ESR were higher, and the number of leukocytes, eosinophils, lymphocytes and rod-nucleated neutrophils did not differ. Wastewater from the livestock enterprise affected metabolic processes in carp, particularly caused increase of the glucose and total protein concentration, the lactate dehydrogenase, aspartate aminotransferase and creatine kinase activity in blood plasma and hepatopancreas. At the same time, there were no differences between specimens from the conditionally clean and contaminated pond in terms of content of triglycerols, cholesterol, urea, creatinine, calcium, inorganic phosphorus, AlAT and AsAt activity in blood plasma and hepatopancreas and total protein content and LHD activity in hepatopancreas, content of total protein, glucose, creatinine, urea, the glucose content in hepatopancreas. The established changes in internal organs, morphological composition of blood and metabolic parameters, particularly concentration of hemoglobin and the erythrocytes number in blood, glucose and total protein content, and activity of lactate dehydrogenase and creatine kinase in carp enabled to propose to use these parameters as supplementary over assessment of the ecotoxicological state of ponds contaminated by wastewater from livestock enterprises.

Determination of Weight and Biochemical Changes in Layers Experimentally Infected with Escherichia coli

This study determined the effect of experimental exposure of layer chickens to Escherichia coli (E. coli) on weight, egg production and some plasma biochemical parameters. A total of 20 laying chickens (20 weeks old) were acquired and assigned, at random, into infected and control groups, of 10 layers each. Birds in the infected group were challenged with 0.5 ml of bacterial aliquot containing 109 colony forming units (CFU) of the bacteria, administered intra tracheally. Five birds, from each group were selected at random and their live body weights were determined on days 0, 2, 4, 6, 14, 21, 28, 35 and 42. Also, egg production was monitored, and the weekly percentage egg production of birds was calculated. Two (2) ml of blood were collected from each bird in both groups and used for plasma biochemical analyses. Finding from the research showed that E. coli infection caused reduction in feed and water consumption by day 3 post infection (pi); with subsequent reduction in body weight and egg production. The mean plasma aspartate aminotransferase (AST) activity in the infected group progressively increased from day 6 pi to reach a significantly higher (p &lt; 0.05) value (66.4 ± 0.93 IU/L) on day 28 pi while the mean plasma alanine aminotransferase (ALT) activity progressively increased in the infected group from day 6 pi to attain the highest significant (p &lt; 0.05) value (74.4 ± 2.11 IU/L) on day 21 pi. The mean plasma total protein concentration in the infected group decreased from day 4 pi and reached the lowest significant (p &lt; 0.05) value (5.12 ± 0.28 mg/dl) on day 35 pi. In conclusion, the drop in egg production, loss of body weight, increase hepatic transaminases and decrease plasma protein may be attributed to anorexia or possible impairment of intestinal, renal reabsorption of nutrients from the infection.

Biochemical Response and Histopathologic Alterations in Papyrocranus afer from Banegbe River, Delta State, Nigeria

Aim: This study investigated the effects of pollutants on the biochemical and histopathological components of Papyrocranus afer obtained from the Banegbe River at different locations (upstream, middle-stream, and downstream). The effect of the pollutant on the checked were, levels of alkaline phosphatase (ALP), alanine aminotransferase (ALT), and aspartate aminotransferase (AST) in blood samples of the fishes and the histopathologic alterations of the gills and liver of the fishes. &#x0D; Methodology: The gills and liver tissue of the fish were collected and processed using routine histologic techniques for fixing and embedding in paraffin and staining sections with hem eosin. All the indices of the enzyme tests were estimated with the aid of Randox Diagnostic Test Kits following the manufacturer’s methods. &#x0D; Results: Evaluation of the plasma enzymes showed significant increases (p&lt;0.05) in plasma ALT, AST, and ALP activity in all the sampled points of Banegbe River when compared to the control 1 (Orogodo river) with middle-stream having significant (p&lt;0.05) higher concentrations when compared to down-stream and up-stream. The main alterations observed in the liver of the fish at the Banegbe River were Kupffer cell hyperplasia and focal necrosis of the liver. While the gills showed alteration of the structure of the epithelium, hyperplasia of the epithelium of the primary lamella, epithelial lifting, vacuolization, alteration of the structure, and occurrence of aneurysms in the secondary lamella, all these alterations were as a result of pollutant in the river. Control 1 showed normal parenchymal cells of the liver and normal gill structure. &#x0D; Conclusion: The increase in enzyme activities suggests leakage of these enzymes from the liver cytosol into the bloodstream as a result of liver damage by contaminants. From our results, it can be concluded that the Banegbe River is polluted as a result of effluents channeled in the river by industries within the environs.

Hepatic Health and Humoral Immunological Parameters of Common Carp (Cyprinus carpio) Fed Lactic Acid-supplemented Diets

Background: Organic acids and their salts are known as appropriate substitutes in feed for improving the health, growth, and performance of fish. Objectives: The present study was conducted to determine the effects of dietary lactic acid supplementation on immunological factors, hepatic enzyme activity, and plasma proteins in common carp, Cyprinus carpio. The fish were fed the diets mentioned above for 56 days, then their growth performance, humoral immunity, and plasma hepatic enzymes were assessed. Methods: A total of 180 fish (mean weight=25 g) were randomly distributed in twelve tanks (150 L water in each tank) as four treatments, fed diets containing 0, 2.5, 5, and 10 g/kg lactic acid (T0, T1, T2, and T3, respectively). Results: At the end of the feeding trial, T2 showed significantly higher growth performance than T0. Plasma total protein, albumin, and globulin levels of T1-T3 were significantly higher than that of T0. However, plasma protein levels decreased significantly by elevation in dietary lactic acid concentration (10 g/kg). No significant differences were observed in plasma aspartate aminotransferase (AST) and alkaline phosphatase (ALP) activity among the treatments, although alanine aminotransferase (ALT) activity decreased significantly in fish-fed dietary lactic acid supplementation (T1-T3) compared with the control group (T0). All humoral immunity parameters (lysozyme, complement, immunoglobulin, and bactericidal activity) increased significantly in T1-T3 treatments compared to the T0 group. Conclusion: Overall, dietary lactic acid supplementation improves growth performance, humoral immunological parameters, and hepatic health. According to the results, dietary lactic acid (2.5-5 g/kg) is recommended for preparing common carp feed.

Plasma Creatine Kinase and Aspartate Aminotransferase Activities Before, During, and After 5 Days of Intramuscular Penicillin Administration in Sedentary Horses.

Muscle damage can result in leakage of intracellular enzymes such as creatine kinase (CK) and aspartate transaminase (AST) into plasma. There are no controlled documentations of the effects of intramuscular antibiotic drug administration on plasma CK and AST activities in horses. The objective of this experiment was to test the hypothesis that 5 days of intramuscular procaine penicillin G injection in normal horses would result in increased plasma activities of CK and AST. Nine healthy adult horses were sampled for 7 days preceding, 5 days during, and 32 days following procaine penicillin G (22,000 IU/kg) administration intramuscularly twice daily. Heparinized jugular venous blood samples were obtained daily before treatment and were analyzed the same day for plasma activities of CK and AST. Repeated measures ANOVA and post hoc Tukey's Test were used to identify days where CK or AST were elevated compared to control means at a significance level of P < .05. Beginning the day after first injection, plasma CK increased above the reference range, peaking at 2,046 ± 627 U/L after 3 days, and returned to 227 ± 57.3 U/L (within the reference range) 9 days after treatment began. Beginning the day after first injection, plasma AST increased, peaking at 703 ± 135 U/L on the day after the last injection. Plasma AST did not return to the reference range in all individual horses until 29 days after the last injection (mean 247 ± 33 U/L). Compared to the control period, plasma CK and AST elevations lasted for 8 and 28 days, respectively, after the onset of treatment (P < .001 to P=.03) and lasted for 4 and 24 days, respectively, after the last day of treatment (P < .001 to P=.03).

Evaluation of Cottonseed Meal as an Alternative to Fish Meal in Diet for Juvenile Asian Red-Tailed Catfish Hemibagrus wyckioides.

A 10-week trial was performed to investigate the effects of replacing fishmeal with cottonseed meal (CSM) on the growth rate, protein metabolism, and antioxidant response of Asian red-tailed catfish Hemibagrus wyckioides. Five isonitrogenous and isocaloric diets (C0, C8.5, C17.2, C25.7, and C34.4) were prepared to contain 0%, 8.5%, 17.2%, 25.7%, and 34.4% CSM replacing fishmeal, respectively. The weight gain, daily growth coefficient, pepsin, and intestinal amylase activities initially increased and then decreased with the raising dietary CSM levels; the highest values were observed in the C17.2 group (P < 0.05). However, feed cost exhibited the opposite trend. With the increasing dietary CSM levels, the protein efficiency ratio and intestinal trypsin activity decreased but feed conversion rate increased gradually; while no differences were observed among the C0, C8.5, and C17.2 groups (P > 0.05). Dietary CSM inclusion regardless of levels increased the plasma growth hormone level as well as hepatic aspartate aminotransferase (AST) and γ-glutamyl transpeptidase activities but decreased the plasma glutamate dehydrogenase and AST activities (P < 0.05). With the increasing dietary CSM levels, the plasma alkaline phosphatase (AKP) and hepatic superoxide dismutase activities decreased but malondialdehyde content increased gradually, while no differences were observed among the C0, C8.5, and C17.2 groups (P > 0.05). The plasma immunoglobulin M content and hepatic glutathione reductase activity initially increased but then decreased with the raising dietary CSM levels; the highest values were found in the C17.2 group. These results indicated that dietary CSM inclusion level up to 17.2% improved the growth rate, feed cost, digestive enzyme activity, and protein metabolism without compromising antioxidant capacity of H. wyckioide, whereas these parameters were depressed by further inclusion of CSM. CSM is a potentially cost-effective alternative plant protein source in diet of H. wyckioide.

Effects of triterpenoids in fatty products on liver condition of laboratory animals with acute toxic hepatitis

One of the principles of prevention and non-medicamentous treatment of liver diseases, including hepatitis of different etiology, is the normalization of the diet through the consumption of food with physiologically active ingredients, in particular betulin, which helps to eliminate the causes of metabolic and oxidative disorders within liver cells. The aim of the research was to assess in vivo the influence of triterpene alcohol betulin extracted from Betula pendula Roth. birch bark in fat-containing products (for example mayonnaise) on the blood biochemical parameters and liver morphological structure of rats with initiated acute toxic hepatitis. Material and methods. Hepatoprotective and antioxidant activities of betulin as part of mayonnaise samples has been investigated in vivo on the model of toxic hepatitis initiated by carbon tetrachloride in male Wistar rats weighing 210-265 g. The animals were divided into 4 groups of 10 animals each: CG-1 - intact, CG-2 and MG - with carbon tetrachloride initiated toxic hepatitis. rats of the main groups were orally administered mayonnaise once a day at a dosage of 1 ml for 21 days after the formation of the model pathology: OG-1 with the added betulin (1 mg per 1 kg of body weight), OG-2 without betulin. Disorders of metabolic and oxidative processes in liver cells of animals were evaluated by biochemical indicators of blood plasma: the level of glucose, albumin, total cholesterol, triglycerides and urea and the activity of alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase and γ-glutamyltransferase. Oxidative stress in rats was estimated by the activity of catalase and superoxide dismutase in blood hemolysate (at a dilution of 1:200 and 1:10, respectively); the total prooxidant (in blood plasma) and total antioxidant (in blood hemolysate at a dilution of 1:10) activity were determined spectrophotometrically (colored complexes of TWIN-80 oxidation products with thiobarbituric acid). The morphological structure of rats' liver was estimated by microscopy of prepared cuts of hepatic tissue. Results. Based on biochemical parameters of rat blood plasma, it has been established that the administration of mayonnaise with betulin prevents the development of cytolic syndrome and suppresses the process of peroxidation by directly neutralizing free radicals. Aspartate aminotransferase and alkaline phosphatase activity in blood plasma of the experimental animals of the main group MG-1 reduced by 20.7 and 35.2% compared with indicators of the rats of the main group MG-2. Glucose concentration normalized to the level of the control group CG-1. The concentration of bilirubin and triglycerides decreased by 22.9 and by 48.1%, which indicates a significant reduction in the indicators of cholestatic syndrome in the group of animals OG-1 compared to OG-2. The total prooxidant activity and the concentration of thiobarbiturate-reactive products decreased compared to the CG-2 and MG-2 groups, which indicates the suppression of oxidative stress and, as a result, an improvement in liver conditions of animals with toxic hepatitis even when taking a fat-containing product. In liver histopeparates of animals receiving mayonnaise with betulin, necrobotic changes were less pronounced in comparison with the group MG-2. They were estimated at 1 point: small-drip dystrophy spots were found, haemorrhages in the interregional septum with inflammatory infiltration in the course of hemorrhages against the presence of necrosis hepatocytes with pronounced adipose dystrophy in the centres of the lobules, step necrosis with signs of replacing the damaged hepatocytes of the connective tissue, accompanied by centrolobular hemorrhages in MG-2 rats. Conclusion. Introduced into the composition of mayonnaise betulin, reduces the development of cytolic syndrome in toxic hepatitis and suppresses the process of peroxidation, on the basis of which fat-containing foods with betulin can be recommended for clinical examination as specialized products in acute and chronic liver diseases, including complicated cholestasis.

Comparison of the Retention Rates of Synthetic and Natural Astaxanthin in Feeds and Their Effects on Pigmentation, Growth, and Health in Rainbow Trout (Oncorhynchus mykiss).

The coloring efficiency and physiological function of astaxanthin in fish vary with its regions. The aim of this study was to compare the retention rates of dietary astaxanthin from different sources and its effects on growth, pigmentation, and physiological function in Oncorhynchus mykiss. Fish were fed astaxanthin-supplemented diets (LP: 0.1% Lucantin® Pink CWD; CP: 0.1% Carophyll® Pink; EP: 0.1% Essention® Pink; PR: 1% Phaffia rhodozyma; HP: 1% Haematococcus pluvialis), or a diet without astaxanthin supplementation, for 56 days. Dietary astaxanthin enhanced pigmentation as well as the growth of the fish. The intestinal morphology of fish was improved, and the crude protein content of dorsal muscle significantly increased in fish fed with astaxanthin. Moreover, astaxanthin led to a decrease in total cholesterol levels and alanine aminotransferase and aspartate aminotransferase activity in plasma. Fish fed on the CP diet also produced the highest level of umami amino acids (aspartic acid and glutamic acid). Regarding antioxidant capacity, astaxanthin increased Nrf2/HO-1 signaling and antioxidant enzyme activity. Innate immune responses, including lysozyme and complement systems, were also stimulated by astaxanthin. Lucantin® Pink CWD had the highest stability in feed and achieved the best pigmentation, Essention® Pink performed best in growth promotion and Carophyll® Pink resulted in the best flesh quality. H. pluvialis was the astaxanthin source for achieving the best antioxidant properties and immunity of O. mykiss.

Neutrophil Depletion Attenuates Acute Liver Stress after Exhaustive Exercise in Mice.

Exhaustive exercise induces acute liver stress; however, the precise mechanisms remain unclear. We investigated the effects of neutrophil depletion in male C57BL/6J mice. Male C57BL/6J mice were divided into four groups: sedentary with control antibody ( n = 20), sedentary with antineutrophil antibody ( n = 20), exhaustive exercise with control antibody ( n = 20), and exhaustive exercise with antineutrophil antibody ( n = 20). Antineutrophil antibodies (1A8) or control antibodies were administered intraperitoneally before running on a treadmill. Immediately and at 24 h after running to exhaustion on a treadmill at a 7% gradient and a speed of 24 m·min -1 , blood neutrophil counts were measured by flow cytometry. Plasma activities of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were also measured. Hematoxylin-eosin staining was performed to calculate the liver stress score, and hepatic tumor necrosis factor-α was measured using enzyme-linked immunosorbent assay. Exercise increased blood neutrophil and neutrophil infiltration into the liver. Plasma AST and ALT activities were significantly higher immediately after exhaustive exercise than after sedentary control (AST, sedentary with control antibody: 52.2 ± 0.4, exhaustive exercise with control antibody: 210.0 ± 19.8; ALT, sedentary with control antibody: 29.8 ± 2.2, exhaustive exercise with control antibody: 87.2 ± 15.8). However, AST and ALT activities were significantly decreased with the 1A8 antibody (AST, 102.2 ± 12.9; ALT, 39.2 ± 4.0). In addition, the liver stress score increased after exercise but was significantly reduced by prior 1A8 antibody administration. The 1A8 antibody treatment also decreased hepatic tumor necrosis factor-α levels after exhaustive exercise. These results suggested that neutrophils play a critical role in increasing liver stress by regulating inflammation.

Different Types of Non-Starch Polysaccharides Alter the Growth, Intestinal Flora and Serum Metabolite Profile of Grass Carp, Ctenopharyngodon idella.

Dietary non-starch polysaccharides (NSPs) broadly influence fish intestinal flora and physiological metabolism, but limited information is available on grass carp (Ctenopharyngodon idella). This study investigated the effects of different types of NSPs on the growth, nutrient metabolism status, gut microbiota, and serum metabolome of grass carp. Fish were fed with diets containing 4.4% insoluble NSPs (INSP), 9.24% soluble NSPs (SNSP), 13.64% NSPs (4.4% INSP + 9.24% SNSP, NSP) and non NSPs (FM), respectively, for 9 weeks. Results showed that dietary SNSP decreased protein efficiency ratio and serum protein content, but increased feed coefficient ratio, feed intake, plasma blood urea nitrogen content, and plasma aspartate aminotransferase activity (AST); conversely, dietary INSP decreased plasma AST activity. Dietary INSP and SNSP increased serum free cholesterol content. Dietary NSPs altered the abundance of dominant bacteria and serum metabolite profiles. The differential metabolites between groups were significantly enriched in amino acid synthesis and metabolic pathways. In conclusion, dietary INSP exhibited a growth-promoting effect compared to SNSP. Dietary INSP is beneficial for improving nutrient metabolism and intestinal health. Moreover, dietary NSPs may regulate the physiological metabolism and feeding behavior of grass carp by altering amino acid synthesis and metabolism.

Effects of dietary Hyssop, Hyssopus officinalis, extract on physiological and antioxidant responses of rainbow trout, Oncorhynchus mykiss, juveniles to thermal stress.

The present study aimed at assessing the effects of dietary Hyssop, Hyssopus officinalis, extract on rainbow trout, Oncorhynchus mykiss, responses to thermal stress. The juveniles (69.8 ± 0.38 g) were stocked in 12 through-flow tanks at a density of 12 fish per tank. Methanolic extract of Hyssop (HME) was added to diet at 0, 100, 250, and 500 mg/kg and the fish were fed (3% of biomass) over a 70-d period: 62 d at 13.3 ± 0.08°C and 7 d at 21-22°C. At the end of the trial, the plasma alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH), triiodothyronine (T3), thyroxin (T4), cortisol, glucose, lactate, total antioxidant capacity (TAC), ascorbate, and the gill glutathione peroxidase (GPx), glutathione reductase (GR), glutathione-S-transferase (GST), and malondialdehyde (MDA). The results showed that HME had no significant effects on fish growth performance, survival, and feed efficiency. Dietary 250 mg/kg HME significantly decreased plasma ALT activity (P < 0.001), but showed no significant effects on plasma AST) (P = 0.106) activity, T3 (P = 0.992), and T4 (P = 0.070) levels. Thermal stress significantly (P < 0.001) increased plasma ALT and AST activities, but lowered plasma T3 and T4 levels. Dietary HME and thermal stress had interaction effects on plasma cortisol (P < 0.001), glucose (P = 0.007), lactate (P = 0.010), LDH (P = 0.005), TAC (P = 0.038), ascorbate (P < 0.001), and the gill GPx (P = 0.001), GR (P < 0.001), GST (P < 0.001), and MDA (P = 0.001). Thermal stress significantly increased plasma cortisol, glucose, lactate, and LDH, the gill GPX, GR, and GST, but dietary HME supplementation significantly reduced such elevations, particularly at 250 mg/kg level. Dietary HME significantly increased plasma TAC before the thermal stress and mitigated the stress-induced decreased in TAC, particularly at 250 mg/kg level. Dietary HME significantly decreased the gill MDA before and after the thermal stress, and lowest MDA was observed in 250 mg/kg HME level. Based on the present results, 250 mg/kg HME is recommended as suitable dose to improve antioxidative responses and hepatoprotection in rainbow trout under heat stress.

The Preliminary Results for Evaluating Cocoa Butter's Hepatoprotective Effects against Lipid Accumulation and Inflammation in Adult Male Rats Chronically Fed Ethanol.

The purpose of this study was to clarify the role of saturated fats from cocoa butter (plant source) compared with lard (animal source) on alcoholic liver damage in rats. Male Wistar rats were fed either a control diet (C) or an ethanol diet (E), and the dietary fats (corn oil, olive oil, and safflower oil) of these two diets were further replaced by lard (CL, EL) or cocoa butter (CC, EC). After 8-week feeding, plasma aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities, hepatic triglyceride (TG) levels, plasma intercellular adhesion molecular (ICAM)-1 levels, hepatic cytochrome P450 2E1 (CYP2E1) protein expression, and hepatic interleukin (IL)-1β significantly increased in the E group compared to the C group. In addition, hepatic histopathological scores of fatty changes, inflammatory cell infiltration, and degeneration and necrosis in the E group were significantly higher compared to those in the C group. However, fatty changes were significantly inhibited only in the EC group as well as hepatic inflammatory cell infiltration, degeneration, and necrosis being significantly lower in the EL and EC groups. Plasma ICAM-1 and hepatic tumor necrosis factor (TNF)-α, IL-1β, IL-6, and IL-10 levels were significantly lower in the EL and EC groups than those in the E group. Moreover, a correlation analysis showed that hepatic histopathological scores of degeneration and necrosis were significantly positively correlated with erythrocytic oleic acid (C18:1) and were negatively correlated with linoleic acid (C18:2). In conclusion, cocoa butter protected the liver against lipid accumulation and inflammation in rats chronically fed ethanol.

Effect of Dietary Ginger Rhizome Powder Addition on Reproductive Performance of Rabbit Bucks

This study was conducted to evaluate the effect of the addition of ginger rhizome powder (GRP) to the rabbit bucks ration on semen quality and semen plasma traits. A total of 28 male New Zealand rabbits (7- month- old), divided into four groups; the first treatment served as a control (Con; 0% ginger); the second, third, and fourth treatments were supplemented with 0.5%, 1.0%, and 1.5% ginger powder in the diet, respectively. The results revealed no change in ejaculate volume, a significant increase in the mass sperm motility, sperm concentration, total sperm output, total motile sperm, live sperm, and Total functional sperm fraction. Dietary ginger reduced the reaction time, semen pH, and the percentage of abnormal sperm. Bucks that received 1.0% or 1.5% dietary ginger had a higher seminal plasma total proteins, albumin, globulin concentrations, alkaline phosphatase and acid phosphatase activities. Conversely, seminal plasma aspartate aminotransferase and alanine aminotransferase activities were significantly decreased compared to the control group. Practically, it can be concluded that ginger is a suitable supplement for improving semen quality and semen plasma in adult male rabbits.

Safety of the anthelminthic drugs levamisole, fenbendazole, and ivermectin administered in therapeutic baths for the common carp Cyprinus carpio

Due to the safe "off label" use of selected antiparasitic drugs in fish, we assessed the effects of a therapeutic bath with levamisole (50 mg/l), fenbendazole (25 mg/l), and ivermectin (0.031 mg/l) on the haematological and biochemical blood indices, oxidative and antioxidant variables, and gill histology of the common carp (Cyprinus carpio). Levamisole did not affect the haematological profile, but significantly increased (P &amp;lt; 0.01) the plasma glucose, lactate, and ammonia concentrations, alkaline phosphatase, and alanine aminotransferase activities, TBARS (muscle, liver), total superoxide dismutase activity (muscle), and catalase activity (liver) and significantly decreased (P &amp;lt; 0.01) plasma aspartate aminotransferase and glutathione reductase activity (gill, liver, muscle). Ivermectin led to a significantly (P &amp;lt; 0.01) greater muscle total superoxide dismutase activity compared to the controls, whereas the haematological and biochemical indices remained unchanged. On the other hand, fenbendazole did not affect the haematological or biochemical indices, and the oxidative stress parameters and antioxidant indices did not differ from the controls. The bath in FBZ can be recommended for safe antiparasitic treatment in carp.

Effect of in ovo manganese injection on the embryonic development, antioxidation, hatchability, and performances of offspring broilers under normal and high temperatures

Two experiments were carried out to study the effect of in ovo manganese (Mn) injection on the embryonic development, antioxidation, hatchability, and performances of offspring broilers under normal temperature (NT) and high temperature (HT). Experiment 1 was conducted to investigate the effect of in ovo Mn injection on the embryonic hatchability of Arbor Acres broiler breeders. On D 9 of incubation, a total of 684 fertilized eggs were randomly allocated to 6 treatments: the non-injected positive control (niPC) and treatments injected with 0 (the negative control, iNC), 6.25, 12.5, 25.0, or 50.0 μg Mn/egg as Mn sulfate. Experiment 2 was conducted to investigate the effect of in ovo Mn injection on the embryonic development, antioxidation and performance of offspring broilers under NT and HT. A total of 792 fertilized eggs were randomly allocated to 6 treatments in a 1 (niPC) + 1 (iNC) + 2 (injected Mn sources: Mn sulfate and Mn proteinate) × 2 (injected Mn levels: 12.5 and 25.0 μg/egg) factorial arrangement during the embryonic stage and D1 to 28 at NT. Then, 288 birds were allotted to 12 treatments in a 6 (the above embryonic treatments) × 2 (environmental temperatures: NT-22℃ vs HT-34℃) factorial arrangement from D 29 to 42. The results showed that Mn injection affected (P < 0.03) the hatchability and the maximum level of in ovo injected Mn was 25.0 μg Mn/egg. The Mn injection upregulated (P < 0.05) Mn-containing superoxide dismutase mRNA expression in the embryonic heart compared to the iNC. Hyperthermia decreased (P < 0.05) ADG and ADFI, breast muscle percentage, plsma alkaline phosphatase activity, and red color values of breast and thigh muscles, but increased (P < 0.05) F/G, plasma aspartate aminotransferase and lactic dehydrogenase activities, total cholesterol, uric acid and triiodothyronine contents, abdominal fat, light values of breast and thigh muscles of offspring broilers. The results suggest that in ovo Mn injection can enhance antioxidant ability in the chick embryonic heart.

A new single-cell protein from Clostridium autoethanogenum as a functional protein for largemouth bass (Micropterus salmoides).

Clostridium autoethanogenum protein (CAP) is a new single-cell protein source originating from inactivated bacteria. An in vitro digestion experiment and an 8-wk growth experiment were conducted to evaluate the molecular weight distribution of the CAP hydrolysate, and the effects of dietary CAP levels on the growth performance, plasma parameters, hepatic and intestinal health, and the diversity of gut-adherent microbiota of largemouth bass (Micropterus salmoides). The fish (initial body weight of 47.99 ± 0.01 g) were fed diets where CAP gradually replaced 0% (CAP0), 12.5% (CAP12.5), 25% (CAP25), 37.5% (CAP37.5) and 50% (CAP50) of low-temperature steam dried anchovy fish meal (LTFM) in the diet. Results showed that the content of peptides below 1,000 Da in the CAP hydrolysate (0.56 mg/mL) was higher than that of the LTFM hydrolysate (0.48 mg/mL). Dietary CAP inclusion had no negative effect on growth performance, while whole-body lipid content significantly reduced in the CAP25 and CAP50 groups (P < 0.05). The plasma alanine aminotransferase activities and triglyceride concentrations in the CAP inclusion groups were significantly lower than those in the CAP0 group (P < 0.05). The plasma aspartate aminotransferase activity was significantly reduced in the CAP37.5 group (P < 0.05). The richness and diversity of the gut-adhesive microbiota and the proportion of Clostridium sensu stricto 12 in the CAP50 group were significantly higher than those in the CAP0 group (P < 0.05). Dietary CAP inclusion inhibited inflammatory responses by down-regulating the mRNA levels of interleukin 1β (IL1β), IL10 and transforming growth factor β1 (P < 0.05) in the liver. The mRNA levels of acetyl-CoA carboxylase 1 were significantly down-regulated in the CAP12.5, CAP25 and CAP37.5 groups (P < 0.05), while that of fatty acid synthase was significantly down-regulated in the CAP50 group (P < 0.05). These results demonstrate that dietary CAP inclusion could improve the hepatic and intestinal health of largemouth bass, and can be helpful to further develop CAP as a functional feed ingredient.

Glycine alleviated diquat-induced hepatic injury via inhibiting ferroptosis in weaned piglets.

ObjectiveThe beneficial effects of glycine were tested in piglets with diquat-induced hepatic injury.MethodsThirty-two piglets were assigned by a 2×2 factorial experimental design including glycine supplementation and diquat challenge. After 3 weeks of feeding with a basic diet or a 1% glycine supplemented diet, piglets were challenged with diquat or saline. After 1 week later, the piglets were slaughtered and samples were collected.ResultsOur results indicated that glycine alleviated diquat induced morphological hepatic injury, decreased the activities of plasma alanine aminotransferase, aspartate aminotransferase and glutamyl transpeptidase in the piglets under diquat challenge, and increased total antioxidant capacity and antioxidative enzyme activity significantly. Adding glycine enhanced the concentrations of hepatic adenosine triphosphate and adenosine diphosphate. Transmission electron microscope observation showed that diquat induced clear hepatocytes ferroptosis and its effect could be alleviated by glycine to a certain degree. Moreover, glycine significantly affected mRNA and protein expression of ferroptosis-related signals in the liver.ConclusionThese results demonstrated that glycine attenuated liver damage via inhibiting ferroptosis.

Acyl-CoA:diacylglycerol acyltransferase 1 inhibition in the small intestine increases plasma transaminase activity via the activation of protein kinase C pathway.

Acyl-CoAdiacylglycerol acyltransferase 1 (DGAT1) is a key enzyme in the fat absorption step in enterocytes. We previously reported that the pharmacological inhibition of DGAT1 increased plasma alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activity in corn oil-loaded rats without any sign of hepatotoxicity. In this study, we investigated this mechanism. We found that this elevation occurred only during the pharmacologically active period of a DGAT1 inhibitor and the magnitude did not depend on the volume of corn oil. In addition, this elevation was not accompanied by increases in ALT or AST mRNA levels in the small intestine and liver. To clarify a lipid component responsible for this elevation, rats were treated with free fatty acids instead of corn oil and no plasma ALT elevation was observed. Next, rats were pretreated with inhibitors of monoacylglycerol acyltransferase 2 and intestinal microsomal triglyceride transfer protein instead of the DGAT1 inhibitor, but no plasma ALT elevation was observed after corn oil loading. Since the results suggested a possible role of diacylglycerol (DAG), which activates protein kinase C (PKC), we measured PKC activity in the small intestine and found that the activity was increased by treatment with the DGAT1 inhibitor and corn oil. Moreover, rats pretreated with a PKC inhibitor in combination with the DGAT1 inhibitor showed suppression of plasma ALT elevation. Taken together, the present results suggest that DAG accumulation induced by pharmacological DGAT1 inhibition and resultant PKC activation in enterocytes are involved in the increase in plasma ALT and AST activity in rats.

Mulberry (Morus alba L.) ethanol extract attenuates lipid metabolic disturbance and adipokine imbalance in high-fat fed rats.

An imbalanced adipokine profile in obesity increases the susceptibility to obesity-related cardiometabolic alterations, including type 2 diabetes, hypertension, dyslipidemia, and non-alcoholic fatty liver disease. The mulberry plant has been reported to have health benefits, such as hypolipidemic and hepatoprotective effects. This study examined the effects of a mulberry (Morus alba L.) fruit ethanol extract (MBEE) on dyslipidemia, liver steatosis, and adipokine imbalance in response to a high-fat diet. Male Sprague-Dawley rats were assigned to one of 4 groups containing 6 rats each and fed either a control diet (CON), a high-fat diet (HFD), or a high-fat diet with MBEE of 150 mg/kg/day (LMB) or 300 mg/kg/day (HMB). The triglyceride (TG), total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), alanine aminotransferase (ALT), and aspartate aminotransferase (AST) activities were measured spectrophotometrically. The leptin, adiponectin, and plasminogen activator inhibitor-1 (PAI-1) levels were determined by an enzyme-linked immunosorbent assay. The plasma TG levels were similar in the 4 groups. Plasma cholesterol and low-density lipoprotein cholesterol (LDL-C) levels and TC/HDL-C ratio increased in the HFD group compared with the CON group, whereas those values decreased in the LMB group (P < 0.05), indicating that MBEE had a plasma lipid-lowering effect. HDL-C decreased in the HFD group, but MBEE did not affect the HDL-C level. The HFD rats significantly increased hepatic TG and cholesterol levels and plasma ALT and AST activities compared to the CON group. The hepatic TG level and ALT and AST activities were reduced markedly by the MBEE treatment. The HFD group showed a higher PAI-1 level, whereas MBEE treatment, especially in the HMB group, significantly reduced leptin level, and leptin/adiponectin and PAI-1/adiponectin ratios. These findings suggest that MBEE altered the imbalance between the pro- and anti-inflammatory adipokines to a more anti-inflammatory state. MBEE could protect against abnormal lipid metabolism and hepatic steatosis induced by a high-fat diet, lowering plasma cholesterol, LDL-C and TC/HDL-C, and hepatic TG. These findings are associated with the regulating effect of MBEE on the leptin/adiponectin and PAI-1/adiponectin ratios.
