Anthoxanthum odoratum (Poaceae) as a bioindicator of arbuscular mycorrhizal fungi abundance prior to native revegetation on agricultural soils

ABSTRACT Mycorrhizal fungi, which form symbiotic associations with plant roots, can be critical to the successful establishment of plants. Up to 90% of land plant species form associations with arbuscular mycorrhizal fungi (AMF), which develop fungal structures inside host roots. Common site preparation practices prior to ecological restoration, such as soil disturbance and herbicide application, may lessen the availability of AMF, potentially jeopardising plant establishment. However, the state of the soil microbiome can be difficult to assess directly. This study examines the potential of the widespread pasture grass Anthoxanthum odoratum (Sweet Vernal, Poaceae) as a bioassay of AMF abundance in soils. We present a rapid clearing and staining protocol for visualising AMF structures in the roots of A. odoratum. We then compare % colonisation on roots of naturally established A. odoratum plants collected from undisturbed pastures vs. sprayed and tilled pastures. Roots of A. odoratum collected from sprayed and tilled pastures showed a significantly lower average AMF colonisation at 43.8% compared with 57.6% on roots from undisturbed pastures. However, all plants sourced from the sprayed and tilled pastures still showed some AMF structures. Our results strongly suggest that A. odoratum can be a useful bioindicator of AMF presence and abundance in soils, offering a promising tool for seed-based restoration on retired agricultural land.

Mitigating nitrogen loss in paddy field microcosms through indigenous arbuscular mycorrhizal fungi assemblage

AbstractWhether farmers should consider the role of arbuscular mycorrhizal fungi (AMF) in agriculture is a hotly debated topic. We aimed to investigate the role of indigenous AMF in reducing nitrogen (N) loss from paddy fields via runoff, leaching, NH3 volatilization, and N2O emission. We conducted a pot experiment employing a mycorrhiza‐defective rice mutant (non‐mycorrhizal) as the control, grown in soil containing indigenous AMF. The corresponding AMF treatment used the progenitor of this mutant with the same soil. The plants were fertilized with nitrogen, phosphorus and potassium 6 weeks after sowing. The root colonization was 23% in mycorrhizal rice, and no typical AMF structures were observed in the roots of non‐mycorrhizal rice. Our findings indicated that the mycorrhizal system exhibited lower N concentrations of runoff and leachate further compounded by reduced fluxes of N2O and NH3. This led to 14% decrease (mycorrhizal rice 111 kg N ha−1; the non‐mycorrhizal rice: 129 kg N ha−1) in cumulative N loss within 3 days post‐fertilization. While this AMF effect was consistent across the four tested N loss pathways, differences were observed between NH4+‐N and NO3−‐N in the runoff pathway. Notably, our results revealed no evidence of trade‐offs in AMF effect on N loss among the tested pathways. Additionally, mycorrhizal rice had larger shoots and roots than their non‐mycorrhizal counterparts. Our study underscores the potential benefits of indigenous AMF in paddy fields for mitigating water pollution and reducing greenhouse gas emission.

Micro PIXE mapping proves a differential distribution and concentration of trace elements in fungal structures of Rhizophagus intraradices

Arbuscular mycorrhizal (AM) fungi can sequester different potentially toxic elements, such as trace elements (TEs), within their structures to alleviate the toxicity for its host plant and themselves. To elucidate the role of AM fungi in TEs immobilization in the rhizosphere of host plants, it is important to know the TEs distribution in AM fungal structures. In the present study, we investigated the distribution and concentration of TEs within extraradical spores and mycelium of the AM fungus Rhizophagus intraradices, collected from the rhizosphere of Senecio bonariensis plants grown in a soil polluted with multiple TEs, by using Particle-Induced X-ray Emission with a micro-focused beam (micro PIXE). This technique enabled the simultaneous micrometric mapping of elements in a sample. The calculated values were compared with those in the polluted substrate, measured by the Wavelength Dispersive X-ray Fluorescence technique. The highest concentrations of Fe, P, Ti, Mn, Cr, Cu and Zn were found in AM fungal spores, where they were accumulated, while extraradical mycelium was enriched in Cu. Finally, we demonstrated that AM fungi can simultaneously accumulate high amounts of different TEs in their structures, thus reducing the toxicity of these elements to its host plant.

Unveiling the influence of seawater intrusion and vegetation type on coastal arbuscular mycorrhizal fungal communities in China

AbstractContinuously rising sea levels pose severe threats to the diversity and stability of coastal ecosystems. However, the responses of endospheric and rhizospheric arbuscular mycorrhizal (AM) fungal communities to accelerated seawater intrusion and changing vegetation types unfortunately remains unknown. Such insights will assist in revealing the structure of AM fungi across different tidal zones, enabling the development of effective measures to conserve biodiversity. Here, we investigated changes in endospheric and rhizospheric AM fungal communities associated with four types of coastal vegetation in Dafeng City, Jiangsu Province, China, in response to sea level variations by analyzing their characteristics and connections to selected environmental factors. It was found that the AM fungal communities of coastal vegetation differed significantly in their compositions, diversity, and stability (the resilience or resistance of a community). Despite the responses of endospheric and rhizospheric AM fungi to soil environmental factors, the soil electrical conductivity, cation exchange content, and inorganic carbon were identified as core elements that influenced the characteristics of AM fungal communities between different coastal vegetation species. Lastly, the major endospheric and rhizospheric species of AM fungi were important predictors that explained much of the variation in the soil environment. Consequently, the dynamics of AM fungal communities were correlated with the plant–soil system under different conditions. Apart from the coastal vegetation species, the characteristics of AM fungal communities can also be driven by high salinity and the inorganic carbon content induced by the intrusion of seawater.

Enhanced mycorrhization in four species of columnar cacti under the Sonoran Desert tree <em>Olneya tesota</em>

Background: Columnar cacti survival in deserts occurs mainly under nurse plants, due to better conditions for germination and establishment, as greater humidity, and nutrient concentration. However, interactions like the association between arbuscular mycorrhizal fungi (AMF) and roots remain relatively unexplored. We evaluated the mycorrhizal status and soil rhizosphere properties of four columnar cacti species growing both under and outside their nurse plant. Hypotheses: Soil properties improvement under nurse plants has been correlated with the abundance of AMF. Therefore, a greater number of AMF structures and roots mycorrhization would be in facilitated plants, compared to the same species growing outside the nurse. Studied species: Nurse plant: Olneya tesota, facilitated species: Carnegiea gigantea, Pachycereus pringlei, Pachycereus schottii, and Stenocereus thurberi. Study site and dates: Central Gulf Coast subdivision of the Sonoran Desert, Sonora, Mexico; October 2020. Methods: Cacti roots and soil rhizosphere were collected under and outside the nurse plant. Roots were stained, and mycorrhizal colonization (MC), hairy roots (HR), visual density (VD), and number of spores were determined. Physicochemical soil properties were analyzed. Results: Values of MC, VD, spore number, organic matter (OM), nitrate, and total and assimilable phosphorous were higher under the nurse compared to bare areas. Conclusions: The hypothesis that mycorrhization parameters are greater in roots of cacti growing under the nurse was confirmed, besides previous knowledge about soil quality under nurse plants, suggesting that AMF play a significant role in the cacti facilitation process under nurses plants, and that nurse plant plays a significant role in AMF formation.

Impacts of Managed Vegetation Restoration on Arbuscular Mycorrhizal Fungi and Diazotrophs in Karst Ecosystems.

The crucial functional arbuscular mycorrhizal fungi (AMF) and diazotrophs play pivotal roles in nutrient cycling during vegetation restoration. However, the impact of managed vegetation restoration strategies on AMF and diazotroph communities remains unclear. In this study, we investigated the community structure and diversity of AMF and diazotrophs in a karst region undergoing managed vegetation restoration from cropland. Soil samples were collected from soils under three vegetation restoration strategies, plantation forest (PF), forage grass (FG), and a mixture of plantation forest and forage grass (FF), along with a control for cropland rotation (CR). The diversity of both AMF and diazotrophs was impacted by managed vegetation restoration. Specifically, the AMF Shannon index was higher in CR and PF compared to FF. Conversely, diazotroph richness was lower in CR, PF, and FG than in FF. Furthermore, both AMF and diazotroph community compositions differed between CR and FF. The relative abundance of AMF taxa, such as Glomus, was lower in FF compared to the other three land-use types, while Racocetra showed the opposite trend. Among diazotroph taxa, the relative abundance of Anabaena, Nostoc, and Rhizobium was higher in FF than in CR. Soil properties such as total potassium, available potassium, pH, and total nitrogen were identified as the main factors influencing AMF and diazotroph diversity. These findings suggest that AMF and diazotroph communities were more sensitive to FF rather than PF and FG after managed vegetation restoration from cropland, despite similar levels of soil nutrients among PF, FG, and FF. Consequently, the integration of diverse economic tree species and forage grasses in mixed plantations notably altered the diversity and species composition of AMF and diazotrophs, primarily through the promotion of biocrust formation and root establishment.

Distinct response patterns of soil micro-eukaryotic communities to early-season and late-season precipitation in a semiarid grassland

Semiarid ecosystems are susceptible to changes in precipitation regimes. However, how soil micro-eukaryote communities in semi-arid ecosystems respond to variations in the seasonal distribution of precipitation remains elusive. A 5-year field experiment was performed to investigate the effect of changed precipitation distributions throughout the growing season on soil micro-eukaryotic communities in the temperate steppe. Early-season precipitation did not significantly affect the biomass of arbuscular mycorrhizal fungi (AMF) nor the composition and structure of AMF and protist communities. Late-season precipitation significantly increased the biomass of AMF while significantly altering the relative abundance of dominant genera in AMF and protist communities. There was a clear difference in the community structure of AMF and protists between the late-season precipitation and natural precipitation. In addition, late-season precipitation decreased the amount of consumer protists but enhanced the abundance of parasitic protists. These results indicate that the impact of precipitation limitations in the middle growing season can be weakened by increased precipitation in the early growing seasons, while an increase in precipitation later in the growing season does not improve the impact of a decrease in precipitation in the middle growing season. Our findings highlight the potential impact of intra-annual changes in precipitation in soil micro-eukaryotic communities in arid and semiarid regions.

Improving apple orchard health: The role of arbuscular mycorrhizal fungi in alleviating replant disease and strengthening soil microbial communities

The occurrence of apple replant disease (ARD) is inevitable during the renewal and transformation of aging orchards. This disease seriously threatens the healthy and sustainable development of the apple industry. Arbuscular mycorrhizal fungi (AMF) have shown great potential in promoting plant growth, resisting soil-pathogen infection and improving soil quality. However, there is relatively little information available about the application of AMF in the prevention and control of ARD. In this study, we conducted a pot and field experiment were conducted with inoculation of AMF (Paraglomus sp. SW1) on apple rootstock (Malus hupehensis Rehd. in the pot experiment and M9T337 in the field experiment) to explore the biocontrol effect of AMF on ARD by comparing the changes in fungal and AMF communities after AMF inoculation on apples under replanting conditions. AMF-inoculated apple was used as the treatment, while non-inoculated apple was used as the control. The results showed that AMF-inoculated plants not only promoted the growth of M. hupehensis Rehd., but also stimulated the branch elongation growth of young apple trees, compared to the control. After inoculation with AMF, the community structure of soil fungi and arbuscular mycorrhizal fungi changed significantly, with an increase in the abundance of fungi and AMF in soil, and a significant reduction in the abundance of Fusarium spp., especially Fusarium moniliforme, Fusarium solani, and Fusarium proliferatum. Additionally, the soil enzyme activity increased significantly. Furthermore, the levels of ammonium nitrogen (NH4+), nitrate nitrogen (NO3−), available phosphorus (AP), available potassium (AK), organic matter (SOM), and pH in the rhizosphere soil were significantly related to the changes in the community structure of fungi and AMF. The AP, NH4+ and NO3− was found to drive the fungal communities, while NH4+ and NO3− drove the AM fungal communities in replanting soil. In conclusion, apple-arbuscular mycorrhizal symbionts can effectively alleviate ARD by promoting plant growth and optimizing the structure of soil microbial communities. Future research, should test other gardening plants following AMF inoculation to prepare for a wider application in controlling replant disease.

The influence of a soil amendment on the abundance and interaction of arbuscular mycorrhizal fungi with arable soils and host winter wheat.

Arbuscular mycorrhizal (AM) fungi have been shown to be associated with an estimated 70 % of vascular terrestrial plants. Such relationships have been shown to be sensitive to soil disturbance, for example, tillage in the preparation of a seed bed. From the application of arable soil management, AM fungal populations have been shown to be negatively impacted in abundance and diversity, reducing plant growth and development. The present study aims to utilise two sources (multipurpose compost and a commercial inocula) of mycorrhizal fungi for the amendment of arable soils supporting Zulu winter wheat under controlled conditions and quantify plant growth responses. A total of nine fields across three participating farms were sampled, each farm practicing either conventional, reduced, or zero tillage soil management exclusively. Soil textures were assessed for each sampled soil. Via the employment of AM fungal symbiosis quantification methods, AM fungi were compared between soil amendments and their effects on crop growth and development. The present study was able to quantify a mean 6 cm increase to crop height (P<0.001), 10 cm reduction to root length corresponding with a 2.45-fold increase in AM fungal arbuscular structures (P<0.001), a 1.15-fold increase in soil glomalin concentration corresponding to a 1.26-fold increase in soil carbon, and a 1.32-fold increase in the relative abundance of molecular identified AM fungal sequences for compost amended soils compared to control samples. Mycorrhizal inocula, however, saw no change to crop height or root length, AM fungal arbuscules were reduced by 1.43-fold, soil glomalin was additionally reduced by 1.55-fold corresponding to a reduction in soil carbon by 1.31-fold, and a reduction to relative AM fungal species abundance by 1.26-fold. The present study can conclude the addition of compost as an arable soil amendment is more beneficial for the restoration of AM fungi beneficial to wheat production and soil carbon compared to the addition of a commercial mycorrhizal inocula.

Bruguiera gymnorhiza forms mycorrhizal associations but Rhizophora stylosa does not: A pot experiment using mangrove soil and Rhizophoraceae seedlings

Arbuscular mycorrhizal (AM) fungi often colonize the roots of mangrove plants, forming symbiotic associations with them, but colonization rates differ greatly among mangrove species. To examine differences in the colonization patterns of AM fungi, we focused on two species of mangroves (Rhizophora stylosa and Bruguiera gymnorhiza; Rhizophoraceae) and conducted a pot experiment using seedlings grown in mangrove soil watered with fresh water or brackish water (200 mM NaCl). We observed AM fungal structures such as hyphae, arbuscles, and vesicles in the roots of all B. gymnorhiza seedlings in the freshwater treatment, but rarely in the brackish water treatment. By contrast, we found no AM fungal structures in roots of any R. stylosa seedlings in either the freshwater or brackish water treatment. These results imply that B. gymnorhiza are facultatively mycorrhizal plants that have maintained the ability to form associations with AM fungi, whereas R. stylosa, which dominate habitats more seaward than those of B. gymnorhiza, may have lost the ability to form mycorrhizal associations through adaptation to extreme seaside conditions.

Different Responses of Arbuscular Mycorrhizal Fungal Community Compositions in the Soil and Roots to Nitrogen Deposition in a Subtropical Cunninghamia lanceolata Plantation in China

Elevated nitrogen (N) deposition may stimulate a plant’s dependency on arbuscular mycorrhizal (AM) fungi in phosphorus (P)-deficient subtropical forests. However, the ecological assembly processes and the responses of AM fungal diversity and community structure to N deposition in both the roots and rhizosphere are still unclear. We collected root and soil samples from a Cunninghamia lanceolata plantation forest after four years of N addition and examined the community structure and assembly of AM fungi. Elevated N deposition decreased the AM fungal community diversity in both rhizosphere soil and roots. Glomeraceae was the dominant family of the AM fungal community in both soil and roots across all N addition treatments, followed by Gigasporaceae and Ambisporaceae. However, N addition induced differential variation in the community composition of AM fungi between soil and roots. For soil AM fungi, N addition decreased the Glomeraceae abundance and increased the Gigasporaceae and Ambisporaceae abundance. In contrast, the root AM fungal community was dominated by Glomeraceae under N addition treatments. Furthermore, N addition increased the deterministic community assembly that acted as an environmental filter for soil AM fungi. In contrast, N addition decreased the importance of determinism, implying that the selection of plants on root AM fungi decreased with increasing N addition. Altogether, our findings suggest that the community structure of AM fungi responds differently to N deposition in the soil and roots in subtropical forests and highlight the important role of soil AM fungi in helping host plants respond to N deposition.

Quantification of Arbuscular Mycorrhizal Fungi Colonization Rate in the Study of Invasive Alien Plants.

Arbuscular mycorrhizal fungi (AMF) are widely distributed soil fungi in ecosystems and can form symbiotic associations (mycorrhizae) with the roots of most terrestrial plants. Plants provide carbon sources to AMF through mycorrhizal associations, while AMF hyphae can expand the range of nutrient absorption by roots and promote plant nutrient uptake. There are many different species of AMF, and the symbiotic relationships between different species of AMF and different plants vary. Invasive plants can enrich AMF species with better symbiotic capabilities through root exudates, promoting their growth and thereby increasing their colonization in invasive plant roots. At the same time, invasive plants can also disrupt the symbiotic relationship between AMF and native plants, affecting the local plant community, which is one of the mechanisms for successful plant invasion. The colonization rate of AMF in the roots of invasive and native plants indirectly reflects the role of AMF in the process of invasive plant invasion. In this method, collected plant roots can be processed directly or saved in a fixative for later batch processing. Through decolorization, acidification, staining, and destaining treatment of roots, the hyphae, spores, and arbuscular structures of AMF in the root system can be clearly observed. This method can be completed in a basic laboratory to observe and calculate the colonization rate of AMF in the root systems of invasive plants.

Quantification of arbuscular mycorrhizal fungi root colonization in wheat, tomato, and leek using absolute qPCR

Arbuscular mycorrhizal fungi (AMF) form symbioses with most terrestrial plants and are known to have a positive effect on plant growth and health. Different methodologies have been developed to assess the AMF-plant symbiosis. The most applied method, which involves staining of roots and microscopic observation of the AMF structures, is tedious and time-consuming and the results are highly dependent on the observer. Using quantitative polymerase chain reaction (qPCR) to quantify AMF root colonization represents a reliable, high-throughput technique that allows the assessment of numerous samples. Quantification with qPCR can be performed through two methods: relative quantification and absolute quantification. In relative quantification, the target gene is normalized with a reference gene. On the other hand, absolute quantification involves the use of a standard curve, for which template DNA is serially diluted. In a previous paper, we validated the primer pair AMG1F and AM1 for a relative quantification approach to assess AMF root colonization in Petunia. Here, we tested the same primers with an absolute quantification approach and compared the results with the traditional microscopy method. We evaluated the qPCR method with three different crops, namely, wheat (cv. Colmetta and Wiwa), tomato, and leek. We observed a strong correlation between microscopy and qPCR for Colmetta (r = 0.90, p < 0.001), Wiwa (r = 0.94, p < 0.001), and tomato (r = 0.93, p < 0.001), but no correlation for leek (r = 0.27, p = 0.268). This highlights the importance of testing the primer pair for each specific crop.

Diversity of Endomycorrhizal Fungi in Argan Forest Stands: Implications for the Success of Reforestation Programs

Over the last few decades, argan trees (Argania spinosa L.) skeels have faced harsh ecological conditions and anthropogenic pressure, leading to a dramatic decline in surface and density of cultivation. Nowadays, most techniques used to regenerate argan trees have failed. Arbuscular mycorrhizal fungi (AMF) are root symbionts that increase plant resistance to biotic and abiotic stresses during transplantation. The exploration of these symbiotic fungi from different soils of argan stands is the starting point for the selection and production of high-performance organisms adapted to the reforestation sites. The objective of this study is to investigate the composition of the AMF community associated with the argan tree rhizosphere. Forty adult argan trees were sampled in eight forest sites representative of the distribution and genetic diversity of argan forest stands. Five sub-samples of rhizospheric soil were taken around each tree. Our results revealed the presence of different AMF structures (i.e., hyphae, vesicles/and arbuscules) in root samples. Based on morphological characterization, six genera of AMF spores were identified with a dominance of the genera Septoglomus (34%). In addition, soil organic matter and phosphorus concentrations showed a highly significant correlation with AMF spore density. The chi-square test showed a highly significant dependence of the distribution of genera on the site conditions of forest stands. These AMF could be tested and used during the inoculation of argan seedlings in forest nurseries for the success of restoration and reforestation programs, as well as for the development and sustainable improvement of this agroforestry system.

Reduced Arbuscular Mycorrhizal Fungi (AMF) Diversity in Light and Moderate Fire Sites in Taiga Forests, Northeast China.

Forest fires are an important disturbance factor in forest ecosystems, and obviously change the soil environment. Arbuscular mycorrhizal fungi, as a medium and bridge between vegetation and soil, play a crucial role in mediating plant nutrient uptake and regulating the productivity, stability, and succession of vegetation-soil systems. To investigate the effects of forest fires on the community structure and diversity of arbuscular mycorrhizal fungi in cold-temperate Larix gmelinii forests, we collected soils from light, moderate, and heavy fire disturbance forests and a natural forest as a control forest in Greater Khingan Larix gmelinii forests, in the northeast of China. The community structure and diversity of arbuscular mycorrhizal fungi was sequenced using Illumina MiSeq technology and we analyzed the correlation with the soil physicochemical characteristics. The results showed that the contents of microbial biomass content (MBC), moisture content (MC), total nitrogen (TN), and available phosphors (AP) increased significantly (p < 0.05) with increasing fire intensity (from Light to heavy fire), but available potassium (AK) decreased significantly (p < 0.05). These changes were not significant. A total of 14,554 valid sequences from all sequences were classified into 66 ASVs that belonged into one phylum, one order, four families, and four genera. The genera included Glomus, Ambispora, Paraglomus, and Acaulospora, and Glomus was the dominant genus (the genera with the five most relative abundances) in the control and heavy-fire forests. Non-metric multidimensional scaling (NMDS) analysis showed that forest fires significantly affected the community structure of arbuscular mycorrhizal fungi (p < 0.01). Redundancy analysis (RDA) showed that MBC, SOC, and AP contents significantly affected the composition structure and diversity of arbuscular mycorrhizal fungi communities. This study indicated that forest fires affected the composition and diversity of soil arbuscular mycorrhizal fungi communities through changing the soil physicochemical parameters (MBC, SOC, and AP) in cold-temperate Larix gmelinii forests. The study of soil physicochemical properties and arbuscular mycorrhizal fungi diversity in cold-temperate Larix gmelinii forests in the Greater Khingan Mountains after forest fires provides a reference basis for the revegetation and reconstruction of fire sites.

Effects of Variations in Soil Moisture and Phosphorus Concentrations on the Diversity of the Arbuscular Mycorrhizal Fungi Community in an Agricultural Ecosystem

In farmland ecosystems, phosphorus and water have crucial roles. To elucidate the effects of phosphorus concentration and water management on arbuscular mycorrhizal fungi (AMF), field experiments were conducted in a farmland ecosystem (China). We examined the effects of different treatments, including drought and normal phosphorus, normal water and normal phosphorus, drought and low phosphorus, and normal water and low phosphorus, on the AM fungal biomass, diversity, and community. Results showed great differences in the AMF under different water and phosphorus concentrations. When under a suitable drought treatment, the AMF became more abundant and more conducive to plant growth. The abundance of AMF varied with different phosphorus treatments, and the abundance of AMF in low-phosphorus treatments was higher, which is more suitable for plant growth. In conclusion, as water and phosphorus concentrations change, the community structure of AMF constantly changes. Only under the appropriate water and phosphorus concentration processing can AMF play its role well. Understanding the influence of different phosphorus concentrations and the moisture contents of AMF can play a role in the agricultural production of AMF, and it can also provide improved theoretical support.

Effect of Arbuscular Mycorrhizal Fungi on Growth and Development of Zea Mays L.

In the present study, effect of Arbuscular Mycorrhizal Fungi (AMF) on seed germination, nutrient uptake, plant growth, root colonization and chlorophyll content of the Zea mays L. was evaluated. The soil samples collected from maize field (Morgaon, Pune, Maharashtra) were used for the isolation of AMF spores by using wet sieving and decanting method. The International Culture Collection of (Vesicular) Arbuscular Mycorrhizal Fungi (INVAM) worksheet was used for diagnosing the spores. The completely randomized design with three replicates was used for pot culture experiment. The chlorophyll (a, b and total chlorophyll) content from the leaves of the maize plant was successfully determined by Arnon method. Three AMF spores (Gigaspora sp., Acoulospora sp. and Glomus mosseae) were identified and selected for further study. The application of AMF inoculum significantly increases the overall seed germination percent in saline soil (50% and 25% in nonsterile and sterile saline soil) and root and shoot length (13.68 cm and 10.02 cm increase in nonsterile normal soil while 8.93 cm and 11cm increase in nonsterile saline soil) when compared with control. Highest increase in pigment was found in sterile soil inoculated with AMF [chlorophyll a (47.80%), b (47.76) and total chlorophyll (45.56%)]. In root colonization study, different typical AMF structures (such as vesicles, spores, intra and extra radical hyphae) inside the roots of the maize plant were evaluated by microscopic observations. Finally, the nutrient profiling of the soils from all pots showed increased N, P, K, Ca, Fe, Mn, Cu and Zn when compared with control.

Water availability alters the community structure of arbuscular mycorrhizal fungi and determines plant mycorrhizal benefit

Societal Impact StatementThe world faces major changes in rainfall patterns and water availability, posing a significant threat to plant productions systems and food security. The arbuscular mycorrhizal (AM) fungi associate with most major crops and can support plant nutrient and water uptake. Here, AM fungi were shown to mitigate the negative effects of low water availability on sorghum growth and phosphorus uptake, an effect that was associated with shifts in the fungal community structure. To realise the potential of AM fungi in sustainable agriculture requires more examination of their interactions with edaphic stresses in crop systems.

The impact of ice storms on mycorrhizal fungi varies by season and mycorrhizal type in a hardwood forest

AbstractExtreme weather events, such as ice storms, are increasing and have potentially large impacts on forests, including belowground structures such as fine roots and mycorrhizal fungi. Many forest trees rely on the mutualistic relationship between mycorrhizal fungi and plants; a relationship that, when disrupted, can negatively impact tree net primary productivity. We took advantage of a large‐scale ice storm manipulation in the northeastern United States to test the hypothesis that increasing ice storm intensity and frequency would reduce ectomycorrhizal fungal root tips per unit root length and arbuscular mycorrhizal fungal structures per unit root length, hereafter colonization. We found that ice storm intensity reduced spring ectomycorrhizal fungal and arbuscular mycorrhizal fungal colonization. However, these patterns changed in the fall, where ice storm intensity still reduced ectomycorrhizal fungal root tips, but arbuscular mycorrhizal fungal colonization was higher in ice storm treatments than controls. The amount of ectomycorrhizal fungal root tips and arbuscular mycorrhizal fungal colonization differed seasonally: ectomycorrhizal fungal root tips were 1.7× higher in the spring than in the fall, while arbuscular mycorrhizal fungal colonization was 3× higher in the fall than in the spring. Our results indicate that mycorrhizal fungal colonization responses to ice storm severity vary temporally and by mycorrhizal fungal type. Further, arbuscular mycorrhizal fungi may recover from ice storms relatively quickly, potentially aiding forests in their recovery, whereas ice storms may have a long lasting impact on ectomycorrhizal fungi.
