Isolation and Evaluation of Cytotoxic, Anti-Inflammatory, Anti-Ulcer Activity of Methanolic Extract of Ceriops decandra leaves

The current study was undertaken to provide scientific validation for the traditional medicinal applications of Ceriops decandra leaves in treating gastrointestinal disorders and inflammation. Additionally, the study aimed to isolate a pure component from the extracted leaves for further analysis. Lupeol was extracted from the crude methanolic extract of Ceriops decandra leaves by column chromatography as part of a phytochemical inquiry. Its structure was determined using 1H and 13C NMR spectroscopy. In order to assess the cytotoxicity, the unrefined methanolic extract was divided into two fractions: a petroleum fraction and an aqueous fraction, employing the modified Kupchan method. The brine shrimp lethality test revealed that both the aqueous and petroleum ether fractions had significant cytotoxic activity, with LC50 values of 1.93 µg/l and 2.04 µg/l, respectively. These values were compared to the LC50 value of the standard Vincristine Sulphate, which was found to be 0.02 µg/l. The results of the anti-inflammatory trial demonstrated that the administration of the extract at doses of 250mg/kg and 500mg/kg resulted in protection rates of 62.5% and 87.5%, respectively, as compared to the carrageenan control group after 3 hours post-injection. It is worth noting that Ibuprofen exhibited a higher level of protection, with a rate of 91.7%. In the context of ethanol-induced stomach ulcer, the administration of extracts at doses of 250 mg/kg and 500 mg/kg resulted in 45.5% and 59.1% protection against gastric ulcer, respectively. These findings were compared to the protective effect of Omeprazole, which demonstrated 63.6% protection and served as the standard reference. The findings suggest that the methanolic leaf extract of Ceriops decandra possesses robust cytotoxic and potent anti-inflammatory and anti-ulcer properties. These results provide support for the traditional application of this extract in the management of gastrointestinal diseases, inflammation, and cancer.

Gastroprotective Activity of Aerial Leaves of Allium cepa

The present study was performed to evaluate the Antiulcer activity of Methanolic extract of Allium cepa leaves (MEAC) in Albino rats. Antiulcer activity of MEAC was investigated by using Ethanol induced gastric mucosal damage and Pylorous ligated gastric ulcers. Evaluation was done on both models comparing with reference standard Omeprazole (20mg/kg). The parameters taken to assess anti-ulcer for pylorus ligation method were volume gastric secretion, pH, free acidity, total acidity and ulcer index. In vivo studies were carried on the plant extract and results were found that protection of the Methanolic extract of Allium cepa leaves against characteristic lesions may be due to both reduction in gastric acid secretion and gastric cycloprotein and this may be due to the presence of Flavanoids. In this study we observed that, Methanolic extract of leaves of Allium cepa (MEAC) provides significant antiulcer activity against gastric ulcers in rats. The result of MEAC showed significant reduction in the ulcer index, when compared to control. The antiulcer activity extract may be attributed to their flavanoids content and antioxidant property. These findings indicate that MEAC display potential antiulcerogenic activity.

Evaluation of antiulcer activity of herbal drugs on experimental animal: Myrica nagi (Myricaceae)

The anti-ulcer activity of Methanolic extract of Myrica nagi (Myricaceae) fruit MEMN was investigated in pylorus ligation and methanol-induced ulcer models in Wistar rats. In both models, the common parameter determined was ulcer index MEMN at doses of 200,400 mg/kg p.o produced significant inhibition of the gastric lesions induced by Aspirin-induced ulcers & pylorus ligation-induced ulcers. The extract (200, 400 mg/kg) showed a significant (P < 0.001) reduction in gastric volume, free acidity and ulcer index as compared to the control. This present study indicates that Myrica nagi fruit extract has potential antiulcer activity in both models. These results may further suggest that methanolic extract was found to possess antiulcer genic as well as ulcer healing properties.

Evaluation of the Antiulcer Activity of Methanolic Extract and Solvent Fractions of the Leaves of Calpurnia aurea (Ait.) Benth. (Fabaceae) in Rats.

Introduction In Ethiopia, traditionally, the leaves of Calpurnia aurea have been utilized to treat peptic ulcer disease. Therefore, the objective of the present study was to examine the antiulcer activity of Calpurnia aurea hydromethanolic leaf extract and solvent fractions in rats. Methods The ulcer-healing potential of the crude test extract was assessed in rats by adopting pyloric ligation-, acidified ethanol-, and acetic acid-induced ulcer methods; while, in solvent fractions, the acidified ethanol-induced ulcer model was used. In all models, three serial test doses (100, 200, and 400 mg/kg) were given and the antiulcer activity was investigated. Standard drugs like sucralfate (100 mg/kg), omeprazole (20 mg/kg), and cimetidine (100 mg/kg) have been used as a positive control; whereas distilled water (10 mL/kg) was used as the negative control. Parameters like ulcer index, total acidity, pH, gastric volume, and gastric mucin level were all measured. Results In an acute toxicity study, the test extract at the limit test dose (2 g/kg) was safe following a single dose administration. In pyloric ligation-induced ulcers, the plant extract at 200 and 400 mg/kg significantly reduced the ulcer index, the volume of stomach secretion, and total acidity while raising gastric pH and mucus content (P < 0.05). Likewise, in the acidified ethanol- and acetic acid-induced ulcer models, the extract at both test doses (200 and 400 mg/kg) also displayed a substantial reduction (P < 0.05) in ulcer index. Among the fractions, the ethyl acetate fraction revealed remarkable cytoprotective activity at all test doses and the aqueous fraction at 400 mg/kg (P < 0.05). In contrast, the effect of chloroform fraction was found to be negligible. The peak ulcer inhibition was noted at 400 mg/kg of ethyl acetate fraction (52.4%). Conclusion The study showed that the crude extract and solvent fractions possess remarkable antiulcer activity.

EVALUATION OF GASTROPROTECTIVE EFFECTS OF METHANOL EXTRACT OF TEPHROSIA VILLOSA AGAINST EXPERIMENTALLY INDUCED ULCERS IN WISTAR RATS

Objective: The present research work was designed to investigate the gastroprotective potentials of methanol extract of Tephrosia villosa. Methods: The aerial parts of Tephrosia villosa were dried under shade, powdered and deffated with petroleum ether and then marc leftover was subjected to methanol extraction using soxhlet apparatus using soxh-let apparatus. Antiulcer activity of methanol extract was determined against stress-induced and aspirin-induced ulcers in experimental animal models. The total number of ulcers formed, ulcer index, percentage inhibition, ulcerated area, protected area, pH and Total acidity were parameters in the study. Results: Methanol extract of Tephrosia villosa at medium (200 mg/kg) and high (200 mg/kg) could significantly (P&lt;0.01) reduced the total number of ulcers formed, ulcer index, ulcerated area and total acidity in therapeutic groups compared to vehicle control and thereby significantly (P&lt;0.01) increased percentage inhibition of ulcers and protected area which was evident by the significant rise in pH of gastric content. The effect of extracts was dose-dependent and results were comparable to that of standard drug omeprazole. Conclusion: The results obtained from the present work suggest that the methanol extract of Tephrosia villosa possess significant anti-ulcer potentials against experimentally induced ulcers in albino rats.

EVALUATION OF ANTI-ULCER ACTIVITY OF METHANOLIC EXTRACT OF LAGENARIA SICERARIA

Introduction: Ulcer is a common gastrointestinal disorder and can be characterized by inflamed lesions of the mucosa and tissue that protect the gastrointestinal tract. These ulcers can develop when the imbalance occurs between the gastroprotective (mucus, bicarbonate, and prostaglandins) and aggressive (acid, pepsin, bile salts, and Helicobacter pylori bacteria). According to research and as other species from genus Lagenaria shows analgesic, anti-inflammatory, immunomodulatory, antihyperlipidemic, diuretic, and antihelmintic activities on the fruit of Lagenaria siceraria.&#x0D; Methods: The anti-ulcer activity of methanolic extract of Lagenaria siceraria fruits was investigated in pylorus ligation, ethanol-induced, Asprin induced and Cold-restraint stress-induced ulcer models in Wistar rats. In all models, the common parameter determined was the ulcer index.&#x0D; Results: The extract (100 mg/kg &amp; 200 mg/kg) showed a significant (P&lt;0.005) reduction in gastric volume, free acidity, and ulcer index as compared to control. This present study indicates that Lagenaria siceraria fruit extract has potential anti-ulcer activity in these models.&#x0D; Discussion: MELS caused a significant decrease in the gastric volume, free acidity, and total acidity compared to the control group, indicating an antisecretory mechanism. Several scientific studies revealed that phytoconstituents like flavonoids, tannins, terpenoids, and saponin were responsible for gastroprotective agents. Further studies are needed to find out their exact mechanism of action on gastric acid secretion and gastric cytoprotection.

Pretreatment of Blumea lacera leaves ameliorate acute ulcer and oxidative stress in ethanol-induced Long-Evan rat: A combined experimental and chemico-biological interaction

Blumea lacera (Burm.f.) DC. is described as a valuable medicinal plant in various popular systems of medicine. The aim of the experiment reports the in vivo antiulcer activity of methanol extract of Blumea lacera (MEBLL) and in silico studies of bioactive constituents of MEBLL. In this study, fasted Long-Evans rat treated with 80 % ethanol (0.5 mL) to induce gastric ulcer, were pretreated orally with MEBLL at different doses (250 and 500 mg/kg, p.o., b.w) and omeprazole (20 mg/kg, p.o.) and distilled water were used as a reference drug and normal control respectively. In silico activity against gastric H+-K+ATPase enzyme was also studied. The findings demonstrated that the treatment with MEBLL attenuated markedly ulcer and protected the integrity of the gastric mucosa by preventing the mucosal ulceration altered biochemical parameters of gastric juice such total carbohydrate, total protein and pepsin activity. Additionally, the experimental groups significantly (p < 0.001) inhibited gastric lesions and malondealdehyde (MDA) levels and upregulated antioxidant enzymes level. Furthermore, nine compounds were documented as bioactive, displayed good binding affinities to against gastric H+-K+ATPase enzyme while these compounds illustrated inhibitory effect. From these studies, it is established MEBLL has ulcer healing property as unveiled by in vivo and in silico studies.

Anti-ulcer activity of methanol extract of the leaves of Hannoa klaineana in rats

Ulcer is a common gastrointestinal disease affecting 5% of the world’s population. Hannoa klaineana is used locally in the management of many gastrointestinal disorders and fevers. The study was conducted to evaluate toxicity profile and anti-ulcer effect of methanol extract of the leaves of Hannoa klaineana (Simaroubaceae). Acute toxicity test was conducted according to OECD guideline 423 using the limit test dose (5000 mg/kg) for 14 days. Sub-chronic toxicity study was carryout according to OECD guideline 407 by daily oral administration of the extract (500 and 1000 mg/kg) for 28 days. Anti-ulcer effect of the extract (100, 200 and 400 mg/kg b.wt) was evaluated using ethanol and indomethacin induced gastric ulcer models. In acute toxicity test, 5000 mg/kg dose of the extract does not caused mortality nor any sign of toxicity observed in the rats, thus, the LD50 value of the extract was above 5000 mg/kg. While sub-chronic toxicity test, the extract demonstrated significant (p&lt;0.01) increase in body weight and weight of the liver, spleen and kidneys of the rats. The result also showed significant (p&lt;0.05) dose-dependent decrease in serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), urea, uric acid and creatinine. Ethanol-induced ulcer model, the extract demonstrated significant (p&lt;0.05) dose-dependent decrease in mean ulcer index, with percentage inhibition (62.84%) of gastric damage at 400 mg/kg. While indomethacin model, the extract exhibited significant (p&lt;0.001) dosedependent decrease in mean ulcer index with percentage gastroprotection (99.20%) at 400 mg/kg. This study suggested that methanol extract of the leaves of Hannoa klaineana is safe for oral administration and exhibited strong anti-ulcer effect, thus validating the traditional use of the plant leaves in ulcer treatment.

Anti-ulcer activity of methanol extract of Tectona grandis Linn

PUD is the most common disease that affects a majority of the population in the world and young people being the most affected out of all the ulcer patients. This disease involves a large number of causes like NSAID, malnutrition and deficiency, unhealthy lifestyles like chronic smoking and stress. In recent years, a rapid progressive understanding had been made concerning the mechanism of ulcers and its treatment protocols. As per the literature, survey says Tectona grandis ethanol extract has been proved to show the anti-ulcer activity and considering the solubility and extractive capabilities of methanol over ethanol; the current research was performed to determine the anti-ulcer potency of methanol extract of Tectona roots. The results showed that Tectona methanol extract is way more efficient in treating ulcers in comparison to the standard drug omeprazole.

Phytochemical Screening, Anti-Oxidant Potential and Anti- Ulcer Activity of Leucas diffusa Plant Extract

Leucas diffusa (lamiaceae) a branched herb collected from the Guntur region, A.P. India. Phytochemical studies of Leucas diffusa, methanolic fraction subjected to column chromatography and estimation of total flavanoid content and to evaluate antioxidant potential and anti-ulcer activity of methanolic extract of Leucas diffusa. The plant powder was subjected to continuous hot extraction in Soxhlet Apparatus &amp; extracted successively with n-hexane, ethyl acetate and methanol solvents. The extracts prepared were tested for the type of chemical constituents present by known qualitative tests. Total flavanoid content of methanolic and ethylacetate extract of Leucas diffusa were done. In vitro Antioxidant activity of methanolic extract was evaluated using hydroxyl radical, reducing power &amp; hydrogen peroxide scavenging abilities. Anti-ulcer activitiy of methanolic extract was evaluated through pylorus ligation method and ethanol induced ulcer models. Statistical analysis was performed by one-way analysis of variance (ANOVA) followed by the tukey’s test for multiple comparisons. Our findings suggest that methanolic extract of whole plant Leucas diffusa have got profound antioxidant effect and showed significant anti-ulcer activity at higher dose.&#x0D; Keywords: Leucas diffusa; Antioxidant; Flavanoid; anti-ulcer activity.

Comparative evaluation of antiulcer activity of combination of extracts of Embilica officinalis Gaertn and Musa sapientum Linn in rats

The present study is to evaluate the anti-ulcer activity of methanolic extract of unripe fruits of Musa Sapientum Linn. and the fresh juice of the fruits of Emblica Officinalis Gaertn in Wistar rats. The methanolic extract unripened fruits of Musa Sapientum Linn. and the fresh juice of the fruits of Emblica Officinalis Gaertn were combined and evaluated for the gastroprotective activity by ethanol-induced model and indomethacin-induced model in rats at a dose of 25mg/kg (p.o) and 50mg/kg (p.o) respectively. Biochemical parameters like ulcer index, the volume of gastric juice, pH of gastric juice, free acidity and total acidity were studied. The unripe fruits of Musa and the fresh juice of Embilica will contain flavonoids, flavones, abscisic acid which shows a significant reduction in gastric volume, pH, free acidity, total acidity and ulcer index at the doses of 25mg/kg (p.o) and 50mg/kg (p.o). The results show that the unripe fruits 0f Musa Sapientum Linn. and fresh juice from the fruits of Emblica Officinalis Gaertn will combinedly possess greater antiulcer activity than that of the individual drugs.

ANTIULCER ACTIVITY OF METHANOLIC EXTRACT OF ROOTS OF BETA VULGARIS, CHENOPODIACEAE

Beta vulgaris (chenopodiacea) is a medicinal plant reported for its variety of ethnic medicinal uses. Beta vulgaris showed antioxidant, anticancer, hepatoprotective, nephroprotective, wound healing, and anti-inflammatory activities. Hence we have planned to screen antiulcer activity of root of the plant with the methanolic extract. Root powders successively extracted with methyl alcohol and were subjected for phytochemical screening to identify different phytoconstituents. The methanolic extracts of roots of Beta vulgaris were investigated for ulcer protective activity against pyloric-ligation, ethanol induced gastric lesion and cold restraint stress induced ulcers. Preliminary phytochemical screening revealed the presence of flavonoids, saponins, sterols, and alkaloids. The extract was tested for their lethal effect up to the dose level of 2000 mg/kg. None of them have produced abnormal behavior or mortality in rats. Further methanolic extract of 200 and 400 mg/kg/p.o significantly (p˂0.01) reduced the ulcer score, ulcer number, ulcer index, free acidity and total acidity in pyloric-ligation, ethanol induced gastric lesion and cold restraint stress induced ulcer models in rats. The present study revealed that the root extract of Beta vulgaris has antiulcer activity.

Evaluation of anti-inflammatory and gastric anti-ulcer activity of Phyllanthus niruri L. (Euphorbiaceae) leaves in experimental rats

BackgroundThe medicinal plants signify a massive basin of potential phytoconstituents that could be valuable as a substitute to allopathic drugs or considered as an analogue in drug development. Phyllanthus niruri L. (Euphorbiaceae) is generally used in traditional medicine to treat ulcer and inflammation. In this project we investigated the methanolic extract of leaves of Phyllanthus niruri for anti-inflammatory and anti-ulcer activity.MethodsThe anti-inflammatory activity of methanol extract of Phyllanthus niruri leaves was evaluated at the doses of 100, 200 and 400 mg/kg, p.o. while using ibuprofen (20 mg/kg, p.o) as the standard drug. The animals used were Swiss albino rats. Inflammation was induced by injecting 0.1 ml carrageenan (1% w/v) into the left hind paw. Paw tissues from the different groups were examined for inflammatory cell infiltration. On the other hand, antiulcer activity of methanolic extract of P. niruri leaves at the doses of 100, 200 and 400 mg/kg, p.o. were examined against ethanol-acid induced gastric mucosal injury in the Swiss albino rats - keeping omeprazole (20 mg/kg, p.o.) as reference. The rats were dissected and the stomachs were macroscopically examined to identify hemorrhagic lesions in the glandular mucosa.ResultsP. niruri significantly (p < 0.01) decreased carrageenan-induced paw edema; it exhibited a reduction of 46.80%, 55.32% and 69.14% at doses of 100, 200 and 400 mg/kg, respectively. These findings were further supported by the histological study. The methanolic extract also disclosed good protective effect against ethanol-acid induced gastric mucosal injury in the rats. Administration of the extract’s doses (100, 200 and 400 mg/kg) demonstrated a significant (p < 0.01) reduction in the ethanol- acid induced gastric erosion in all the experimental groups when compared to the control. The methanolic extract at the higher dose (400 mg/kg) resulted in better inhibition of ethanol-acid induced gastric ulcer as compare to omeprazole (20 mg/kg). Histological studies of the gastric wall revealed that toxic control rats revealed mucosal degeneration, ulceration and migration of numerous inflammatory cells throughout the section. On the other hand, MEPN treatment groups showed significant regeneration of mucosal layer and significantly prevented the formation of hemorrhage and edema.ConclusionsThe investigation suggests that methanolic extract of P. niruri leaf possess anti-inflammatory activity and promotes ulcer protection as ascertained by regeneration of mucosal layer and substantial prevention of the formation of hemorrhage and edema.

Protective effects of edible Rhus tripartita (Ucria) stem extract against ethanol-induced gastric ulcer in rats

Rhus tripartita is a plant which is traditionally used for the treatment of ulcer and diarrhea in Tunisia. The aim of the study is to evaluate the antiulcer activity of methanol extract of Rhus tripartita stem (RSE).The phytochemical analysis of RSE was performed. Fasted rats treated with 80% ethanol (0.5mL) to induce gastric ulcer, were pretreated orally with RSE at different doses (200, 400 and 800mg/kg body weight (BW)) and ranitidine (100mg/kg) and distilled water were used as a positive and negative control respectively.The results showed that the pre-treatment with RSE decreased significantly the ethanol gastric ulcer index value and preserved the integrity of the gastric mucosa by preventing the mucosal ulceration caused by ethanol. The RSE prevented alcohol-induced decrease in stomach antioxidant enzyme activities such as catalase, glutathione peroxidase and superoxide dismutase. Total reduced glutathione and malondialdehyde tissue contents were also reversed in RSE-pretreated rats when compared with the negative control group.

Protective Effect of Hydrolea zeylanica Vahl. Leaf extract in Ethanol and Cold Restraint Stress Induced Ulcers in Rats

The present study was designed to investigate the Antiulcer activity of methanolic extract of the leaves of Hydrolea zeylanica (L) Vahl using two different models namely Ethanol and cold restraint stress induced ulcers models in rats. The extract was administered at a dose of 250 and 500 mg/kg orally 30 min prior to ulcer induction. Ranitidine (50 mg/kg) was used as a reference standard. The antiulcer activity was accessed by determining and comparing the ulcer index in the test group with that of the standard group. Gastric volume, total acid and free acid were estimated in rats. Hydrolea zeylanica leaf methanolic extract (HZLME) (500 mg/kg) showed maximum inhibition of gastric acid, free acid and total acid respectively. The ulcer index in the Hydrolea zeylanica leaf methanolic extract treated animals was found to be significantly less in all the models compared to standard drug treated group. However the antiulcer activity of Hydrolea zeylanica leaf methanolic extract was, found to be lesser than that of ranitidine. The results suggest that Hydrolea zeylanica possesses significant antiulcer property which could be due to cytoprotective action of the drug or strengthening of gastric and duodenal mucosa with an enhancement of mucosal defence mechanism.

Evaluation of Antiulcer Activity of Peltophorum Pterocarpum.

Peltophorum pterocarpum is a rich source for phenols and the wood, leaves, and flowers of the plant are used as medicinal agents in traditional medicine. Peptic ulcer is a major disease of gastrointestinal tract, affecting nearly 40 lac people each year worldwide and affects 10% of world population with different aetiologies. Peltophorum pterocarpum consists of phenols and flavonoids, tannins which have potential antioxidant and cytoprotective activity. To evaluate the antiulcer activity of methanolic extract of the leaves of Peltophorum pterocarpum (MEPP) on albino rats. The rats were divided into four groups as control, standard, test 1 (extract: 100 mg/kg) and test 2 (extract: 200 mg/kg) with six rats in each group. Gastric lesions were induced by oral administration of indomethacin (20 mg/kg) followed by pylorus ligation. Standard group of animals were treated with misoprostol and test group of animals were treated with MEPP at doses of 100 and 200 mg/kg. To determine the antiulcer activity of extract, mean ulcer index, free acidity and total acidity were evaluated. Ulcer index was significantly decreased at p<0.01 in MEPP treated groups as compared to control group. Total and free acidity was significantly decreased at p<0.01 in MEPP treated groups as compared to control group. Histological analysis also supported the gastro protective effect of MEPP treated groups when compared with control groups. The study revealed that MEPP exhibited potential antiulcer activity and showed dose dependent antiulcer effect.

Ulcer Healing and Gastroprotective Activity of Methanolic Extracts of Hyoscyamus Albus and Umbilicus Rupestris Leaves against Gastric Injury Caused by Ethanol in Rats

The present study was performed to evaluate the anti-ulcer activity of methanolic extract of Hyoscyamus albus and Umbilicus rupestris against ethanol-induced gastric ulcer. Experimental groups were orally pre-treated with and 100, and 200 mg/kg b.w of methanolic extracts HAMeOH and URMeOH. Ulcer control group were pre-treated just with absolute ethanol and reference group was orally pretreated with omeprazole (20 mg/kg b.w). After one hour of pretreatment, all rats were received the absolute ethanol to generate gastric mucosal injury. And after one hour, our rats were anesthetized and sacrificed and the ulcer areas of the gastric walls were determined. Grossing evaluation has revealed that the negative control rats exhibited severe mucosal injury, whereas, pre-treatment with U. rupestris leaf extract and H. albus has resulted significantly anti gastric mucosal injury and flattening of the mucosal folds. Histological studies of the gastric wall that the pre-treated with URMeOH and HAMeOH leaf extract was pronounced as gastric protection along followed by reduction or inhibition of leucocytes in the infiltration of the submucosa.
