The antioxidant and antimicrobial activity of ethanolic extract in roots, stems, and leaves of three commercial Cymbopogon species

BackgroundCymbopogon is a member of the family Poaceae and has been explored for its phytochemicals and bioactivities. Although the antimicrobial activities of Cymbopogon spp. extracts have been extensively studied, comprehensive analyses are required to identify promising compounds for the treatment of antimicrobial resistance. Therefore, this study investigated the antioxidant and antimicrobial properties of Cymbopogon spp. ethanolic extracts in every single organ.MethodsEthanolic extracts were obtained from three Indonesian commercial species of Cymbopogon spp., namely Cymbopogon citratus (L.) Rendle, Cymbopogon nardus (DC.) Spatf., and Cymbopogon winterianus Jowitt. The leaf, stem, and root extracts were evaluated via metabolite profiling using gas chromatography-mass spectrometry (GC–MS). In silico and in vitro analyses were used to evaluate the antioxidant and antimicrobial properties of the Cymbopogon spp. ethanolic extracts. In addition, bioactivity was measured using cytotoxicity assays. Antioxidant assays were performed using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2-azino-bis [3-ethylbenzothiazoline-6-sulfonic acid (ABTS) to determine toxicity to Huh7it-1 cells using a tetrazolium bromide (MTT) assay. Finally, the antimicrobial activity of these extracts was evaluated against Candida albicans, Bacillus subtilis, Staphylococcus aureus, and Escherichia coli using a well diffusion assay.ResultsGC–MS analysis revealed 53 metabolites. Of these, 2,5-bis(1,1-dimethylethyl)- phenol (27.87%), alpha-cadinol (26.76%), and 1,2-dimethoxy-4-(1-propenyl)-benzene (20.56%) were the predominant compounds. C. winterianus and C. nardus leaves exhibited the highest antioxidant activity against DPPH and ABTS, respectively. Contrastingly, the MTT assay showed low cytotoxicity. C. nardus leaf extract exhibited the highest antimicrobial activity against E. coli and S. aureus, whereas C. winterianus stem extract showed the highest activity against B. substilis. Furthermore, computational pathway analysis predicted that antimicrobial activity mechanisms were related to antioxidant activity.ConclusionsThese findings demonstrate that the leaves had strong antioxidant activity, whereas both the leaves and stems showed great antimicrobial activity. Furthermore, all Cymbopogon spp. ethanolic extracts showed low toxicity. These findings provide a foundation for future studies that assess the clinical safety of Cymbopogon spp. as novel drug candidates.

Phytochemicals and antimicrobial properties of Thai edible plant extracts and their prebiotic-like effects

In this study, the phytochemicals and antimicrobial activities of ethanolic extracts of Thai edible plants, green tea, red cotton tree flower, fingerroot and ginger were evaluated. The plant extracts were taken for evaluation of antimicrobial activities against Cutibacterium acnes DMST 14916, Staphylococcus epidermidis TISTR 518, and Staphylococcus aureus TISTR 746. The minimum inhibitory concentrations (MICs) of green tea, fingerroot, and ginger extracts against C. acnes DMST 14916 were 3.92, 0.49, and 7.85 mg cm-3, respectively and the minimum bacteriostatic concentrations (MBCs) were 3.92, 0.49, and 7.85 mg cm-3, respectively. The MICs and MBCs of fingerroot extract against S. epidermidis TISTR 518 were 0.12 and 0.49 mg cm-3, respectively, while those against S. aureus TISTR 746 were 0.12 and 0.98 mg cm-3, respectively. Red cotton tree flower extract showed no antimicrobial activity against the acne-causing bacteria. By scanning electron microscopy (SEM) evaluation, the bacterial cells treated with the plant extracts revealed visible shrinkages compared to the smooth cell surfaces of the controls. The phytochemicals in the plant extracts were analysed by liquid chromatography with tandem mass spectrometry (LC-MS/MS). Well-known antimicrobial compounds like azelaic acid, embelin and kaempferol 3-rutinoside-4′-glucoside were identified in all extracts. The cytotoxic effects of the plant extracts on human cell lines were further investigated. The green tea extract was slightly toxic to HaCaT cells found at the initial concentration of 62.5 mg cm-3, but not toxic to MRC-5 cells. The fingerroot and ginger extracts had no cytotoxicity on HaCaT cells, but promoted the MRC-5 cell proliferation. The combination effects of the plant extracts were prebiotic-like and indifferent effects. Regarding all results, the ethanolic extracts of green tea, fingerroot, and ginger could be used individually as natural anti-acne ingredients capable of further product development to improve human skin health.

Antimicrobial activity of amazon medicinal plants

In recent decades, an increase in resistance of bacteria and fungi to antimicrobials has been observed. Therefore, have each other investigated the antimicrobial potential of plants with medicinal properties. In this context, the Amazon has a high variability of plants with potentially active compounds. Thus, the present study aimed to evaluate the antimicrobial activity of ethanol extracts from Amazonian medicinal plants. Leaves and stems of 19 plant species were dried, crushed and extracted by cold maceration with ethanol. The extracts were dried and solubilized at a concentration of 100 mg mL-1 to done antimicrobial activity assays using the agar diffusion method against gram-positive and gram-negative bacteria, and fungi of the genus Candida. Of the total of 38 extracts analyzed, 14 (36.8%) had antimicrobial activity against at least one of the microorganisms tested. The Brazilnut tree stem extract was active against the highest number of microorganisms, three bacteria (S. aureus, E. coli and K. pneumoniae) and three fungi (C. tropicalis, C. parapsilosis and C. krusei). The mulateiro leaf extract was active against three microorganisms, the saracura-mira stem extract against two, and the other extracts only against one microorganism. K. pneumoniae was the most sensitive microorganism, and the most resistant was C. albicans. The lowest values for minimum inhibitory concentration (MIC) for antibacterial activity were for stone-breaker stem extract against K. pneumoniae (0.048 mg mL-1), and for antifungal activity for Brasilnut tree stem against C. krusei (3.125 mg mL-1), stone-breaker leaf against C. guillermondii (3.125 mg mL-1), and mulateiro leaf against C. tropicalis (3.125 mg mL-1). The lowest values of minimum microbicidal concentration (MMC) were for the extracts of stone-breaker stem against K. pneumoniae (0.192 mg mL-1) and mulateiro leaf against C. tropicalis (3.125 mg mL-1). The tested Amazonian medicinal plants had antimicrobial activity, especially Brazilnut tree, this being the first report for this plant species.

Evaluation of Antimicrobial Activity of Alocasia Cordfolia Rhizomes Extract

Objective: Discovering new antibacterial drugs with high efficacy and low toxicity from medicinal plants has received increasing attention worldwide. The aim of this study was to investigate the antimicrobial activity of ethanolic extract of Alocasia cordfolia rhizomes (ACRE). Methods: S.aureus, E.coli and C.albicans were selected as the tested microorganisms. The diameter of the inhibition zone was determined by the disc diffusion method, and MIC and MBC were measured using the broth microdilution method. Results: The diameter of the inhibition zone, MIC and MBC values of ACRE against S. aureus were 7.67±0.58 mm, 160 μg/μL and 320 μg/μL, and that of ACRE against C.albicans were 8.33±1.15 mm, 80 μg/μL and 160 μg/μL, respectively. However, ACRE did not show antimicrobial activity against E. coli. Conclusion: ACRE may be a potentially promising natural antimicrobial product, and further studies are needed to identify the specific active compounds present in this extract and to elucidate their antimicrobial mechanisms.

Utilization of Piper betle L. Extract for Inactivating Foodborne Bacterial Biofilms on Pitted and Smooth Stainless Steel Surfaces.

Biofilms are a significant concern in the food industry. The utilization of plant-derived compounds to inactivate biofilms on food contact surfaces has not been widely reported. Also, the increasing negative perception of consumers against synthetic sanitizers has encouraged the hunt for natural compounds as alternatives. Therefore, in this study we evaluated the antimicrobial activities of ethanol extracts, acetone extracts, and essential oils (EOs) of seven culinary herbs against Salmonella enterica serotype Typhimurium and Listeria innocua using the broth microdilution assay. Among all tested extracts and EOs, the ethanol extract of Piper betle L. exhibited the most efficient antimicrobial activities. To evaluate the biofilm inactivation effect, S. Typhimurium and L. innocua biofilms on pitted and smooth stainless steel (SS) coupons were exposed to P. betle ethanol extract (12.5 mg/ml), sodium hypochlorite (NaClO; 200 ppm), hydrogen peroxide (HP; 1100 ppm), and benzalkonium chloride (BKC; 400 ppm) for 15 min. Results showed that, for the untreated controls, higher sessile cell counts were observed on pitted SS versus smooth SS coupons. Overall, biofilm inactivation efficacies of the tested sanitizers followed the trend of P. betle extract ≥ BKC > NaClO > HP. The surface condition of SS did not affect the biofilm inactivation effect of each tested sanitizer. The contact angle results revealed P. betle ethanol extract could increase the surface wettability of SS coupons. This research suggests P. betle extract might be utilized as an alternative sanitizer in food processing facilities.

Tamarindus indica Extract as a Promising Antimicrobial and Antivirulence Therapy

The worldwide crises from multi-drug-resistant (MDR) bacterial infections are pushing us to search for new alternative therapies. The renewed interest in medicinal plants has gained the attention of our research group. Tamarindus indica L. (T. indica) is one of the traditional medicines used for a wide range of diseases. Therefore, we evaluated the antimicrobial activities of ethanolic extract of T. indica. The inhibitions zones, minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), and fractional inhibitor concentration indices (FICI) against Gram+ve and -ve pathogens were detected. The bioactive compounds from T. indica extract were identified by mass spectroscopy, thin-layer chromatography, and bio-autographic assay. We performed scanning electron microscopy (SEM) and molecular docking studies to confirm possible mechanisms of actions and antivirulence activities, respectively. We found more promising antimicrobial activities against MDR pathogens with MIC and MBC values for Staphylococcus aureus (S. aureus) and Pseudomonas aeruginosa (P. aeruginosa), i.e., (0.78, 3.12 mg/mL) and (1.56, 3.12 mg/mL), respectively. The antimicrobial activities of this extract were attributed to its capability to impair cell membrane permeability, inducing bacterial cell lysis, which was confirmed by the morphological changes observed under SEM. The synergistic interactions between this extract and commonly used antibiotics were confirmed (FICI values < 0.5). The bioactive compounds of this extract were bis (2-ethylhexyl)phthalate, phenol, 2,4-bis(1,1-dimethylethyl), 1,2-benzenedicarboxylic acid, and bis(8-methylnonyl) ester. Additionally, this extract showed antivirulence activities, especially against the S. aureus protease and P. aeruginosa elastase. In conclusion, we hope that pharmaceutical companies can utilize our findings to produce a new formulation of T. indica ethanolic extract with other antibiotics.

Biological activity of Mesua ferrea (Nageswar) seed extracts: An in vitro and in silico study

Overuse of antimicrobial drugs has led to selective resistance to existing antibiotics, necessitating the development of different and improved alternatives. Natural substances, particularly those derived from plants, are well-known for their therapeutic characteristics, including antibacterial and antifungal properties. Over the last decade, several pharmacological investigations on medicinal plants have been carried out based on their traditional uses in complementary therapies. In our current study, we have evaluated the pharmacological properties of Mesua ferrea seed ethanolic extract. To test their antioxidant properties, a 1,1-diphenyl-2-picryl-hydrazyl (DPPH)-radical scavenging activity test was performed. Using the agar well diffusion method, the antimicrobial activity of ethanolic extracts of M. ferra seed was investigated against four bacteria such as Salmonella typhi, Bacillus cereus, Staphylococcus aureus and Escherichia coli. The ethanol extract of M. ferra seed was shown to be efficient against gram positive and gram-negative bacteria in this investigation, although Salmonella typhi, Escherichia coli. Demonstrated resistance to this extract. The results revealed that gram positive bacteria were more sensitive than gram negative bacteria to the ethanolic extract of M. ferra seed at all concentrations utilized (from 20 μL to 100 μL). The lowest inhibition zone was produced by a dose of 20 μL with an inhibition zone equal to 11 mm and 14.33 mm, while the maximum inhibition zone was produced by a high concentration of 100 μL with an inhibition zone 19.33 mm and 18 mm against Bacillus cereus and Staphylococcus aureus respectively. Gas Chromatography-Mass Spectroscopy (GC-MS) was performed to identify the compounds in the extract to determine their pharmacological effects and then we performed in-silico experiments to determine the biological activity of the compounds. We have concluded from the study that M. ferrea seed can be a potential source for lead compound development.

Phytochemical and antimicrobial investigations on the aerial plant parts of pergularia daemia forsk. (asclepiadaceae): a perennial twinning herb

Scientific investigations of medicinal plants have been initiated in many parts of our country because of their contribution to health care. The medicinal properties of plants are due to the presence of phytochemicals. The phytochemicals have anti-oxidant and anti-microbial properties. The resistance to antibiotics by bacteria is increasingly becoming a concern to public health. Currently used antibiotic agents fail to bring an end to many bacterial infections due to super-resistant strains. Medicinal plants represent a rich source of antimicrobial agents from which new drugs can be obtained. The phytochemical constituents of plants are desirable, not only for the discovery of therapeutic agents, but also because such information may be of value in disclosing new sources. The present study has been carried out on the phytochemical, anti- oxidant and anti-bacterial character of aerial plant parts of Pergularia daemia. The phytochemical result shows that the flavonoids, phenols and tannins were present abundantly in the Ethanolic extracts of aerial plant parts. Both total phenol and tannins are high in ethanolic extract. The antioxidant activity of the plant was done by using DPPH radical scavenging assay. The antimicrobial activity of ethanolic extract of Pergularia daemia aerial plant parts was tested against Klebsiella pneumoniae and Citrobacter freundii.

Evaluation of Anticholinergic, Antidiabetic and Antioxidant Activity of Astragalus dumanii, an Endemic Plant

The research was conducted to separately evaluate and detect the possible in vitro antioxidant, antimicrobial activity of ethanol extracts prepared from aerial parts and roots of Astragalus dumanii and anti-cholinesterase and α-glucosidase inhibitory activity from only aerial parts of its The antioxidant capacity was tested by scavenging of DPPH and ABTS free radicals. Compared with the standard antioxidant compound gallic acid; Root and aerial part extract showed lower DPPH radical scavenging activity, however aerial part extract demonstrated higher ABTS radical scavenging activity. The phenolic contents were detected as 5.31 ± 0.03 and 13.23 ± 0.05 mg gallic acid equivalent g-1 extract, flavonoid contents were found as 8.26 ± 0.004 and 7.93 ± 0.005 mg Qercetin equivalent g-1 extract. In addition, the effects of the extracts obtained from aerial parts of the plant on acetylcholinesterase, butyrylcholinesterase and a-glycosidase enzymes were investigated in vitro and IC50 values were obtained as 1.47, 0.83 and 0.48 µg mL-1, respectively. When these values were compared with standard substances, it was seen that Astragalus dumanii could be a good enzyme inhibitory agent. Biological activities and determination of the mode of action of Tunisian Globularia alypum and Cistus monspeliensis ethanolic extracts

ABSTRACT This study was designed to evaluate the antioxidant and antimicrobial activity of ethanolic extracts (EEs) of Cistus monspeliensis and Globularia alypum. C. monspeliensis showed the highest values of phenolic compounds. Further, it was shown that EE of C. monspeliensis displayed the highest DPPH (IC50 = 8.3 ± 1.08 mg/mL). The chemical profiles demonstrated a total of 12 and 13 phenolic compounds for C. monspeliensis and G. alypum, respectively. EEs of both plants possessed broad-spectrum antimicrobial activity when tested against Escherichia coli and Staphylococcus aureus. Treatment of studied strains with these extracts at their MICs reduced considerably the bacterial viability. The extracts did not induce total bacterial lysis, as determined by the measurement of optical density at 620 nm. Strains treated with EEs at Minimum Inhibitory Concentrations showed significant loss of tolerance to NaCl. Our results contribute to understanding the antibacterial mechanism of ethanolic extracts of the studied medicinal plants.

Salvia microstegia Merikarplarının Antimikrobiyal ve Antibiyofilm Aktiviteleri Üzerine Bir Araştırma

Abstract Biofilm-associated infections cause important health problems all over the world. Because of the resistance to antimicrobials and their possible side effects, researchers' attention has been focused on plant extracts and their components. The aim of this study is to evaluate the antimicrobial and antibiofilm potentials of Salvia microstegia Boiss. et Bal. mericarps. The antimicrobial activity of ethanolic extract obtained from the mericarps was evaluated using the microdilution method against seven standard bacteria and three standard yeast strains. In addition, the effect of the extract on inhibiting biofilm formation and preformed biofilm of Pseudomonas aeruginosa was determined using the crystal violet method. The used extract showed moderate to low antimicrobial activity against tested bacteria and yeasts with minimum inhibitory concentration (MIC) values ranging from 62.5 to 250 µg/mL. Moreover, the extract's potential to inhibit preformed biofilms was found to be greater compared to its potential to inhibit biofilm formation. In conclusion, the tested extract obtained from Salvia microstegia mericarps showed antibiofilm potential, however, additional studies involving different strains are needed to reveal its antimicrobial potential in a more holistic perspective.

In Vitro Antioxidant, Antibacterial and Mechanisms of Action of Ethanolic Extracts of Five Tunisian Plants against Bacteria

Pistacia lentiscus, Rosmarinus officinalis, Erica multiflora, Calicotome villosa, and Phillyrea latifolia were considered important medicinal herbs and were used to treat various ailments. The present study was designed to evaluate the antioxidant and antimicrobial activities of ethanolic extracts (EEs). P. lentiscus and R. officinalis were the richest species in phenolic compounds. Similarly, both species showed the highest values of flavonoids. While the EEs of P. lentiscus, E. multiflora, and C. villosa had higher amounts of tannins. These phenolic compounds were evaluated by two different tests, namely diphenyl picrylhydrazyl (DPPH) and ferric iron-reducing power (FRAP). The IC50 values were found to be significant (p &lt; 0.05) for P. lentiscus and E. multiflora. Similarly, both plants showed the highest ferric-reducing antioxidant power (FRAP). This study has been conducted to evaluate the antibacterial potential of EEs against selected bacteria—Gram-positive bacteria (Staphylococcus aureus ATCC 29213, Listeria monocytogenes ATCC 7644) and Gram-negative bacteria (Escherichia coli ATCC 8739, Salmonella typhimurium NCTC 6017)—and determine their modes of action. The ethanolic extracts inhibited bacterial growth by producing concentration-dependent zones of inhibition. Treatment with these extracts at their minimum inhibitory concentrations (MICs) showed a significant reduction (p &lt; 0.05) in the viability of bacteria. The extracts did not induce total lysis. Bacteria organisms treated with EEs at MICs showed a significant (p &lt; 0.05) loss of tolerance to NaCl (5%). Our results highlighted the use of plant extracts as natural antibacterials that can be safely used in health care and led to the understanding of the antibacterial mechanism of plant extracts.

Determination of antioxidant and antimicrobial activities of ethanolic extract from Suillus granulatus

In this study, Suillus spp. after determining the dry matter and protein amounts of the mushroom, the antioxidant and antimicrobial activities of the ethanolic extracts were investigated. Free radical scavenging activity was determined by DPPH method and phenolic content was determined by HPLC. Antimicrobial activity of Suillus spp. was determined against microorganisms that Staphylococcus aureus, Escherichia coli, Salmonella Typhimurium, Listeria monocytogenes and Pseudomonas florescens microorganisms. Suillus mushroom had a antioxidant activity and It was determined that the phenolic components with the highest amount of phenolic acid were ellagic acid, caffeic acid and gentisic acid, respectively. It was stated that Suillus extract inhibited the growth of S. aureus and E. coli, while suppressing the growth of S. Typhimurium, L. monocytogenes and P. florescens.

Antimicrobial Activity of Vernonia amygdalina, Ocimum gratissimum and Gongronema latifolium Synergy on Common Foodborne Pathogenic Bacteria

The search for antimicrobial agents from natural sources such as diverse plant species against foodborne pathogens has less side effect than chemically synthesized compounds. In this study, antimicrobial activity of ethanolic extract, hot and cold aqueous extracts of Vernonia amygdalina, Ocimum gratissimum and Gongronema latifolium was tested against Escherichia coli, Staphylococcus aureus and Listeria monocytogenes. The leaf extracts of Vernonia amygdalina did not show antimicrobial activity against the test isolates. The diameter of zone of inhibition shown by ethanolic leaf extract of Gongronema latifolium and Ocimum gratissimum against S. aureus is 12.0±1 mm and 10±2 mm, respectively. With regards to ethanolic leaf extract of Gongronema latifolium and Ocimum gratissimum, the diameter of zone of inhibition demonstrated against L. monocytogenes and E. coli is 18.0±2 mm and 6.0±1 mm, respectively. A synergistic ethanolic or aqueous extracts of the leaves did not show antimicrobial activity against the test isolates. However, ethanolic extracts of Gongronema latifolium and Ocimum gratissimum used separately is an effective antimicrobial agent the test isolates.

Antimicrobial Properties of Bacterial Cellulose Films Enriched with Bioactive Herbal Extracts Obtained by Microwave-Assisted Extraction.

The use of bacterial cellulose (BC) as scaffold for active biofilms is one of the most interesting applications, especially for the biomedical and food industries. However, there are currently few studies evaluating the potential of incorporating herbal extracts into various biomaterials, including BC. Thus, the aim of this study is to report a screening of the total phenolic content and antioxidant and antimicrobial activity of ethanolic extracts of oregano, rosemary, parsley, and lovage. At the same time, the bioactive potential of BC enriched with the four ethanolic extracts is described. Microwave-assisted extraction was used to extract bioactive compounds from the four selected herbs. The physical, mechanical, structural, and chemical properties of BC were also assessed. Next, BC was enriched with the extracts, and their effect against Escherichia coli, Staphylococcus aureus, and Candida albicans was evaluated. The results showed that the bioactivity of the herbs varied significantly, with rosemary extract being the most bioactive. The BC films possessed good mechanical properties, and a three-dimensional network fibrillar structure appropriate for ethanolic-extract incorporation. The BC samples enriched with rosemary extracts had the highest antibacterial activity against S. aureus, while E. coli. and C. albicans seemed to be resistant to all extracts, regardless of herbs.
