GC-MS analysis, preliminary phytochemical screening, physicochemical analysis and anti-diabetic activity of ethanol extract of <i>Jasminum cuspidatum</i> leaves

The purpose of the present study was investigating the GC-MS analysis, preliminary phytochemical screening, physicochemical analysis and anti-diabetic activity of ethanol extract of the leaves of Jasminum cuspidatum. The anti-diabetic activity was investigated in streptozotocin-induced diabetes rats with treatment of ethanol extract at the dose level of 200 and 400 mg/kg, which was compared with glibenclamide at a dose level of 4 mg/kg and the parameter measured being the blood glucose, total cholesterol, HDL, LDL, VLDL, triglycerides, and total protein. An oral glucose tolerance test (OGTT) was also investigated in experimental rats. The GC-MS analysis revealed that the ethanol extract contained seven phytoconstituents. Preliminary phytochemical screening showed the presence of triterpenoids, flavonoids, glycosides and steroids. The result of in vivo anti-diabetic activity revealed that the ethanol extract of J. cuspidatum showed significant anti-diabetic activity.

Hypoglycemic potential of alcoholic root extract of Cassia occidentalis Linn. in streptozotocin induced diabetes in albino mice

ObjectiveCassia occidentalis (CO) (family: Caesalpiniaceae) is a common weed which is widely used to treat inflammation, hepatotoxicity, antimalarial activities, sore eyes, hematuria, rheumatism, typhoid, asthma, leprosy and diabetes in folklore medicine in India. The present study was carried out to investigate the antidiabetic activity of ethanolic extract of C.occidentalis roots. MethodsRoot extract of C.occidentalis (RCO) was administered orally at two doses (250 and 500mg/kg) to normal and streptozotocin (STZ) induced NIDDM. Fasting blood glucose (FBG) level, biochemical parameters like blood glucose, serum cholesterol, high density lipoprotein (HDL) cholesterol, triglycerides (TG), total protein, urea, creatinine, serum glutamate oxaloacetate transaminase (SGOT), serum glutamate pyruvate transaminase (SGPT) levels and physical parameters like change in body weight, food intake, water intake and levels in liver were performed for the evaluation of hypoglycemic effects. ResultsBoth the doses of RCO caused a marked decrease in FBG levels in STZ induced type 2 diabetic mice. RCO decreased the blood glucose, food intake, water intake, organ weight, serum cholesterol, TG, creatinine, SGOT and SGPT levels with significant value and increased the levels of HDL cholesterol and total protein with a significant value (P<0.05–0.01). The decrease in body weight induced by STZ was restored with a significant value (P<0.01) at both doses. ConclusionThe results suggest that ethanolic roots extract of C.occidentalis Linn. possesses hypoglycemic potential for the NIDDM and support the traditional use of the roots of plant as hypoglycemic agent.

Scientific Validation of Antidiabetic Activity of Ethanol Extract of Ficus glomerata Barks and Roots in Streptozotocin-Induced Diabetic Rats

The tribal people of Chhattisgarh state of India use Ficus glomerata for the treatment of diabetes therefore we planned to investigate antidiabetic effect of bark and root extract of bark and root in Streptozotocin (STZ)-induced diabetic rats. Oral administration of ethanol extract of bark and root of Ficus glomerata at the doses of 200 and 400 mg/kg body weight was studied in STZ-induced diabetic rats. After administration of extract the fasting blood glucose levels were significantly decreased. The oral glucose tolerance was also improved. The effect of extracts on serum lipid profile like total cholesterol, triglycerides, low density lipoprotein (LDL) and high density lipoprotein (HDL) were also measured in the diabetic and non diabetic rats. There was significant reduction in total cholesterol, LDL and improvement in HDL in diabetic rats were observed. The activity of the plant extracts was comparable with glibenclamide, a well-known antidiabetic drug. These results indicated that Ficus glomerata possesses significant antidiabetic and antihyperlipidemic effect.

Amelioration of oxidative stress by Tabernamontana divaricata on alloxan-induced diabetic rats.

Objective:The purpose of this study was to evaluate the anti-diabetic activity of ethanol extract of Tabernamontana divaricata (L.) and its ameliorative effect on oxidative stress in alloxan-induced diabetic rats.Materials and Methods:Diabetes was induced by single intraperitoneal injection of alloxan monohydrate (140 mg/kg body weight). Methanol extract of T. divaricata was administered at the doses of 100 and 200 mg/kg body weight in diabetic induced rats including glibenclamide (3 mg/kg) as a reference drug. In the continuous 21 days treatment, fasting blood glucose level was determined on 0, 7, 14 and 21 days. On day 21, serum lipid profiles and glycosylated hemoglobin, liver antioxidant enzymes levels were estimated.Results:Experimental findings showed a significant anti-diabetic potential of the extract in terms of reduction in blood glucose levels and a correct effect on the altered biochemical parameters. Observed data were found statistically significant in correction of antioxidant enzyme level accompanied with diabetes, particularly at the dose of 200 mg/kg body weight.Conclusion:Based on the results, it can be concluded that the T. divaricata is found to be effective in type 2 diabetes in rats and to have an ameliorative effect on the associated oxidative stress.

Anti-hyperglycemic activity of &lt;i&gt;Trichosanthes tricuspidata&lt;/i&gt; root extract

To evaluate the anti-diabetic activity of ethanolic extract of Trichosanthes tricuspidata root in alloxan induced diabetic rats and to perform the phytochemical screening. The extraction, preliminary phytochemical screening, anti-diabetic activity in alloxan induced diabetic rats by oral administration of extract (200 and 400 mg/kg b.w.), the blood glucose level and biochemical parameters like cholesterol, triglyceride, serum protein, SGPT, SGOT, and ALP were estimated. Phytochemical studies shows the presence of carbohy-drates, proteins, glycosides and terpenoids and the ethanolic extract of T. tricuspidata root significantly lowered the blood sugar level. The above findings justified the traditional claim of anti-diabetic activity in this plant.

Antidiabetic Activity of Ethanolic Extract of Hydnocarpus Wightiana Blume Using Stz Induced Diabetes in Sd Rats

Hydnocarpus Wightiana Blume is an Indian traditional plant used in treating diabetes. Chaulmoogra Oil is obtained from the seeds and has been used in treating leprosy from last twenty years. Hydnocarpus Wightiana Blume posses strong antioxidant, α-glucosidase inhibitory activity. And the ethanolic extract of the seed hull also have the chemical constituents like luteolin, hydnocarpin which are responsible for having free radical scavenging nature. The Phytochemical screening of the ethanolic extract of seed hull of Hydnocarpus Wightiana Blume showed the positive results for Flavonoids, Glycosides, Carbohydrates and Amino Acids. From The Phytochemical study we found that the extract has the antioxidant properties. By this Result we extended our work by administering in diabetes induced SD Rats to check whether it has any antidiabetic activity or not. So, after treating the diabetic rats for 28days with our extract we found that the blood glucose levels got decreased when we compared it with the first day of glucose levels. Hence, we confirmed that the ethanolic extract of the seed hull of Hydnocarpus Wightiana Blume has the antidiabetic activity.

Evaluation of antidiabetic activity of ethanolic extract of Cedrus deodara (Pinaceae) stem bark in streptozotocin induced diabetes in mice

Objective: To evaluate the antidiabetic activity of ethanolic extract of bark of Cedrus deodara. Materials and Methods: Ethanolic extract of the Cedrus deodara (EECD) at dose levels of 250 mg/kg and 500 mg/kg body weight was subjected to the streptozotocin induced diabetes mice. The biochemical parameters, serum glutamic oxaloacetic transaminase (SGOT), serum glutamic pyruvic transaminase (SGPT), cholesterol and triglycerides were also assessed in the experimental animals. Results: The EECD exhibited significant antihyperglycemic activity and also lowers the biochemical parameters like SGPT, SGOT, cholesterol and triglycerides. The ethanolic extract at dose level of 500 mg/kg was found to be more potent than 250 mg/kg in lowering the blood glucose level, almost near to the effect of the standard drug. Conclusion: This study supports the traditional claim and the ethanolic extract of this plant could be added in traditional preparations for the ailment of various diabetes-associated complications.

Hypoglycemic and anti-diabetic activity of ethanolic extract of Catharanthus pusillus (murray) g.don

Many plant drugs are studied for effecting in the treatment of Diabetes mellitus. In the present study ethanolic extract of herb of Catharanthus pusillus was evaluated for hypoglycemic effect in Streptozotocin induced diabetic rats using both 18 hour fasted rat model and oral glucose tolerance test. A comparison was made between the actions of ethanolic extract Catharanthus Pusillus and known anti diabetic drug Glibenclamide (5 mg/kg, p.o). The ethanolic extract of Catharanthus Pusillus was administered at different doses to normal and diabetic rats. The ethanolic extract at 500 mg/kg body weight dose level exhibited significant hypoglycemic activity (P < 0.01 and P < 0.001). Phytochemical screening of aqueous, methanolic, ethanolic and chloroform extract revealed the presence of alkaloids, saponins and carbohydrates. It is concluded that ethanolic extract of herb of Catharanthus Pusillus posses anti diabetic activity.

Antidiabetic activity of ethanolic extract of tubers of Dioscorea alata in alloxan induced diabetic rats.

Objective:To evaluate the antidiabetic activity of ethanolic extract of Dioscorea alata in glucose loaded and alloxan induced diabetic rats.Materials and Methods:The authenticated tubers of D. alata (DA) (JSSCPDP/2008/157) were collected from Dharmapuri, Tamil Nadu. The ethanol extract was tested for hypoglycemic activity in normal rats. In oral glucose tolerance test, glucose (3 g/kg, p.o.) was administered to non diabetic control, metformin (250 mg/kg, p.o.) and DA extract (100 and 200 mg/kg, p.o.) to treat treated rats. Diabetes mellitus was induced by alloxan monohydrate (120 mg/kg, i.p.) in physiological saline after overnight fasting for 18 hours. DA extract (100 and 200 mg/kg, p.o.) and standard drug metformin (250 mg/kg, p.o.) were administered to diabetic rats for 21 days. Fasting blood glucose level and changes in body weight were measured on days 0, 7, 14, and 21. At the end of 21st day, serum lipid profile, total protein, albumin, and creatinine were assessed.Results:In glucose loaded normal rats, the treatment with the extract of DA had shown a highly significant reduction (P < 0.001) in blood glucose levels at the doses of 100 and 200 mg/kg, respectively. The extract did not produce hypoglycemic activity at both the dose levels in normal, fasted rats. In alloxan induced diabetic rats, the body weight of the DA extract treated animals had shown a significant increase (P < 0.001) after 21 days treatment. The blood glucose level was reduced significantly by 47.48% and 52.09% after 21 days treatment at dose levels 100 and 200 mg/kg, respectively. Serum lipid levels, total protein, albumin, and creatinine were reversed toward near normal in treated rats as compared to diabetic control.Conclusion:The results indicate that ethanol extract of DA tubers possesses significant antidiabetic activity.

Effect of ethanolic extract of Cassia occidentalis Linn. for the management of alloxan-induced diabetic rats

Aim:As per traditional claims, root, bark, leaf and flower of the plant Cassia occidentalis Linn. (Caesalpiniaceae) have been reported to possess antidiabetic activity. Based on this traditional indication, the aim of this study was to evaluate the antidiabetic activity of ethanolic extract of C. occidentalis in normal and alloxan induced diabetic rats.Materials and Methods:Ethanolic extract of the whole plant of C. occidentalis was orally tested at doses of 100 and 200 mg/kg for evaluating the hypoglycemic effect in normal and alloxan-induced diabetic rats. In addition, changes in body weight, serum cholesterol, triglyceride and total protein levels, assessed in the ethanol extract treated diabetic rats were compared with diabetic control and normal animals. Histopathologic observations during 21 days of treatment were also evaluated.Results:Ethanolic extract of C. occidentalis produced a significant reduction in fasting blood glucose levels in the normal and alloxan-induced diabetic rats at doses of 100 and 200 mg/kg body weight. Treatment with ethanolic extract of C. occidentalis in normal and alloxan-induced diabetic rats led to a dose-dependent fall in blood sugar levels. Significant differences were observed in serum lipid profiles (cholesterol and triglyceride), serum protein and changes in body weight in ethanolic extract treated diabetic animals, when compared with the diabetic control and normal animals. Concurrent histopathologic studies of the pancreas of these animals showed comparable regeneration by ethanolic extract, which were earlier necrosed by alloxan.Conclusion:Ethanolic extract of C. occidentalis exhibited significant antidiabetic activity in normal and alloxan-induced diabetic rats. The rats also showed improvement in parameters like body weight and lipid profiles and also, histopathologic studies showed regeneration of β-cells of pancreas and so it might be of value in the treatment of diabetes.

Antidiabetic Activity of Ethanolic Extract of Zaleya decandra in Alloxan-Induced Diabetic Rats

Diabetes mellitus is a complex disorder that disturbs the metabolism of carbohydrates, fats, and proteins. Medicinal plants play an important role in the management of diabetes mellitus. The present study was aimed to evaluate the antidiabetic potential of Zaleya decandra roots on alloxan-induced diabetes in rats. Oral administration of ethanolic extract of the root (200 mg/kg body weight/day) for 15 days restored the levels of glucose, cholesterol, triglycerides, total proteins, urea, creatinine, lipid peroxidation level, and antioxidant enzymes significantly in diabetic rats. Histopathological studies showed significant changes like necrosis and degeneration in the liver and pancreas of alloxan-induced diabetic rats. Also these histopathological abnormalities were found to be normalized after treatment with Z. decandra extract. The efficacy of the root extract was found to be equivalent when compared to the standard hypoglycemic drug glibenclamide (1.25 mg/kg body weight/day, orally) in diabetic rats.
