Effects of the Syzygium aromaticum L. extract on antioxidation and inhibition of matrix metalloproteinase in human dermal fibroblast

Objective: To investigate cosmetic potential of Syzygium aromaticum L. (S. aromaticum L.) and to determine its antioxidant and anti-wrinkling effects. Methods: Using high-performance liquid chromatography, eugenol component was quantitated. The antioxidant activity of S. aromaticum L. was analyzed by 2,2-diphenyl-1-picrylhydrazyl radical scavenging and superoxide dismutase like activities. To determine cell viability, elastase and matrix metalloproteinase-1 (MMP-1) activity, human dermal fibroblasts (HS68) were treated with S. aromaticum L. The inhibitory effect of S. aromaticum L. on tumor necrosis factor alpha induced MMPs expression in HS68 was analyzed by realtime-PCR. Results: The eugenol content was confirmed in S. aromaticum L. S. aromaticum L. was observed to have high 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity and superoxide dismutase like activity. S. aromaticum L. had no cytotoxicity against the HS68 and dose-dependently increased elastase inhibition. Moreover, S. aromaticum L. significantly decreased MMP-1 content and inhibited gene levels of MMP-1, MMP-2, MMP-3 and MMP-9. Conclusions: The findings suggest that S. aromaticum L. has great potential as a cosmeceutical ingredient with antioxidant and anti-wrinkling effects.

Acetyl hexapeptide 8 (AH8) is a synthetic peptide for anti-wrinkle cosmetics ingredient. It was developed as a mimetic of botox, patternd after N –terminal end of the protein synatosomal-associated protein 25 (SNAP25), a substrate of botulinum toxin. While AH8 has good efficacy and safety profiles, the permeation through the skin is poor. Therefore, we tried to enhance the transdermal delivery of AH8 by using of hyaluonic acid (HA), a linear polysaccharide of N-acetyl glucosamine and glucuronic acid. To investigate the effect of HA on AH8 penetration, we analyzed paraffin sections of MicropigⓇ skin. Fluorescence labeled AH8 was applied to micropig skin with or without HA. The absorption of AH8 was limited to the stratum corneum (SC) without HA. On the other hand, AH8 penetrated to the dermis with HA. Especially, low molecular weight HA (5 kDa) was most efficient compared to 500 kDa HA and 2000 kDa HA. Experiments using fourier-transform infrared (FTIR) spectroscopy revealed that lower molecular weight HA had a tendency to increase the fluidity of the SC lipids more, which means enhancing the skin penetration. Therefore, HA could be expected to enhance the anti-wrinkle effect of AH8.

Cosmeceutical role of Medicinal plants / Herbs: A Review on commercially available Cosmetic ingredients

Cosmeceuticals are cosmetic products intended to improve physical appearance, health, beauty and skin ailments. Drugs used to control acne, anti-wrinkle effects, protect from sun burn, improves complexation, delayed effect of ageing and improve skin textures, etc. are comes under category of cosmeceuticals. Cosmeceuticals may be synthetic, natural or hybrid depending upon their compositions. Herbal cosmeceuticals are gaining great attention now a day’s due to their wide range of availability and fewer side effects. There are enormous plants and natural ingredients available commercially as cosmeceuticals. Herbal cosmetics originated naturally from plants, sea, rock salt and soil, etc. Generally herbal cosmetics are free from harmful synthetic chemicals and considered safe to use. Many medicinal plants and herbs offers effect like cosmeceuticals and used extensively for same purpose globally. Natural herbs such as; Neem, Kapoor, Henna, Hirda, Behada, Amalaki, Bringaraj, Rosary Pea and Mandor, etc. are used in cosmetic industry for various purpose. This article presented a descriptive review on Herbal cosmeceuticals.

Inhibition of HYBID (KIAA1199)-mediated hyaluronan degradation and anti-wrinkle effect of Geranium thunbergii extract.

Hyaluronan (HA) is an essential constituent of extracellular matrix in the skin. HA reduction in the dermis and overexpression of HYBID (KIAA1199), a key molecule for HA degradation in skin fibroblasts, are implicated in facial skin wrinkling. We aimed to obtain anti-wrinkle agent(s) by screening for inhibition of HYBID-mediated HA degradation. Various plant extracts were screened for inhibition of HA degradation in HYBID-stable transfectants in HEK293 (HYBID/HEK293). Inhibition of HA-degrading activity and HYBID mRNA and protein expression by Geranium thunbergii extract was studied in skin fibroblasts and HYBID/HEK293 cells. Size distribution of newly produced HA was evaluated by preparing metabolically radiolabeled HA in skin fibroblasts. A double-blind, randomized, and placebo-controlled study was performed in healthy Japanese women (n=21) by topically treating each side of the face with a lotion formulated with G.thunbergii extract or placebo for 8weeks. Among the plant extracts tested, only G.thunbergii extract abolished HA depolymerization in skin fibroblasts and HYBID/HEK293 cells by down-regulating HYBID mRNA and protein expression and by inhibiting HYBID-mediated HA-degrading activity. Although untreated skin fibroblasts produced polydispersed HA, G.thunbergii extract-treated cells produced high-molecular-weight HA. Treatment with G.thunbergii extract-formulated lotion significantly improved skin elasticity and reduced skin wrinkling scores at the outer eye corner compared with the placebo formulation. Geranium thunbergii extract inhibited HYBID-mediated HA degradation in vitro and showed anti-wrinkle activity in vivo accompanying the improvement in skin elasticity. Our study provides a possible strategy for anti-wrinkle care through inhibition of HYBID-mediated HA degradation.

Conditioned media from human umbilical cord blood-derived mesenchymal stem cells stimulate rejuvenation function in human skin

Developing treatments that inhibit skin aging is an important research project. Rejuvenation, which focuses on prevention of skin aging, is one of the major issues. Recent studies suggested that mesenchymal stem cells (MSCs) secrete many cytokines, which are important in wound healing. In this study, we investigated the effect of human umbilical cord blood-derived mesenchymal stem cells conditioned media (USC-CM) in cutaneous wound healing and collagen synthesis. We found that USC-CM has many useful growth factors associated with skin rejuvenation, such as Epithelial Growth Factor (EGF), basic Fibroblast Growth Factor (bFGF), Platelet Derived Growth Factor (PDGF), Hepatocyte Growth Factor (HGF), Collagen type 1, and especially, one of the rejuvenation factors, the growth differentiation factor-11 (GDF-11). Our in vitro results showed that USC-CM stimulate growth and extracellular matrix (ECM) production of Human Dermal Fibroblasts (HDFs) compared to those of other MSCs conditioned media (CM) from different origins. Moreover, we evaluated the roles of GDF-11. The results showed that GDF-11 accelerates growth, migration and ECM production of HDFs. Our In vivo results showed that topical treatment of USC-CM showed anti-wrinkle effect and significantly increased dermal density in women. In conclusion, USC-CM has various useful growth factors including GDF-11 that can stimulate skin rejuvenation by increasing growth and ECM production of HDFs.

Hydrogel from Acellular Porcine Adipose Tissue Accelerates Wound Healing by Inducing Intradermal Adipocyte Regeneration

As an important component of the skin, intradermal adipocytes are closely associated with skin homeostasis and wound healing. Although studies have focused on the role of fibroblasts, keratinocytes, and inflammatory cells in wound healing, the role of adipocytes has not been fully investigated. Here, we verified whether the induction of adipocyte regeneration in a wound bed can effectively promote wound healing, finding that the hydrogel from acellular porcine adipose tissue in combination with adipose-derived stem cells can induce in situ adipogenesis in the wound microenvironment. The newly regenerated adipocytes enhanced fibroblast migration, accelerated wound closing, and enhanced wound epithelialization. More importantly, newly formed intact skin structure was observed after treating the wound with adipose-derived stem cell-loaded hydrogel from acellular porcine adipose tissue. These results show that hydrogel from acellular porcine adipose tissue might substantially improve re-epithelialization, angiogenesis, and skin-appendage regeneration, making it a promising therapeutic biomaterial for skin wound healing.

Caffeic Acid-layered Double Hydroxide Hybrid: A New Raw Material for Cosmetic Applications

Bioactive ingredients from natural sources possess well-known positive effects in cosmetic applications. Among them, phenolic acids have emerged with very interesting potential. Caffeic acid (CAF) is one of the most promising active compounds because it possess antioxidant, anti-inflammatory, antitumoral and anti-wrinkle effects. In order to increase its local bioavailability in topical applications, the vehiculation of caffeic acid can lead to a new raw material of cosmetic interest. For this purpose, clay minerals possess excellent properties, such as low or null toxicity and good biocompatibility. Clays are able to host a wide range of active ingredients in the interlayer region, using a green process known as intercalation reaction. The hosting of cosmetic actives into the layered structure of anionic clays allows the preparation of new materials with enhanced stability towards oxidation and photodegradation, better local bioavailability, and easier workability. In this paper, the successful vehiculation of caffeic acid into anionic clay is presented. The obtained hybrid is very promising for the cosmetic market because of its higher bioavailability and prolonged antioxidant activity.

Effects on skin of Stichopus japonicus viscera extracts detected with saponin including Holothurin A: Down-regulation of melanin synthesis and up-regulation of neocollagenesis mediated by ERK signaling pathway

Ethnopharmacological relevanceStichopus japonicus (sea cucumber), edible traditional food in Asia, and its extracts are renowned for their wound healing, pain relieving, and cosmetic effects in traditional medicine. Holothurins, toxins isolated from sea cucumber, are thought to be active components for their beneficial effects. However, researchers have yet to outline specific mechanisms thereof. Aim of the studyThe present study was designed to evaluate the anti-melanogenic and anti-wrinkle properties of S. japonicus viscera extracts (VF) on the skin via in vitro and ex vivo experiments and to assess the anti-aging effects of S. japonicus viscera extracts in relation to known wound healing and cosmetic processes. Materials and methodsThe viscera of live S. japonicus specimens were freeze dried and ground into a powder. Aqueous extracts were subsequently prepared from the concentrated powder using a water extraction method. To investigate the inhibitory effects of VF on melanogenesis, mushroom tyrosinase activity assay and melanin assay were performed on Melan-A cells. To further delineate the anti-melanogenic properties of VF, western blot analysis for tyrosinase, TRP-1, TRP-2, MITF, and ERK was conducted. Changes in collagen synthesis in human dermal fibroblast (HDF) were evaluated via CCK-8 assay and immunocytochemistry to determine the anti-wrinkle effects of VF. Finally, anti-aging properties were examined in a human skin equivalent ex vivo model. ResultsIn Melan-A cells, VF treatment reduced melanin contents in a concentration-dependent manner. The anti-melanogenic effects of VF appeared to be due to enzymatic inhibition of tyrosinase. In CCK-8 assay, VF also significantly increased the viability of HDFs in a concentration-dependent manner. Immunoblot analysis revealed phosphorylation of ERK in HDFs treated with VF. In a human skin equivalent ex vivo model (Neoderm®-ED), VF treatment at a concentration of 50 μg/ml enhanced collagen type IV and Ki-67 expression and downregulated MMP-9 expression. ConclusionThis study demonstrated that aqueous extracts from S. japonicus viscera are effective whitening and anti-aging agents that stimulate ERK signaling to inhibit melanin synthesis and promote collagen synthesis.

Phytoconstituents, antioxidant, sun protection and skin anti-wrinkle effects using four solvents fractions of the root bark of the traditional plant Alkanna tinctoria (L.)

IntroductionProtection of skin from wrinkles and pigmentation changes are among the most common challenges for modern science and cosmetics. These are caused by unusual disruption of connective tissue, the formation of free radicals and ultraviolet radiation.The current research is the first study to compare the phytoconstituents, antioxidant, sun protection factor (SPF) and skin wrinkles prevention effects using four different solvents fractions for the traditional plant Alkanna tinctoria (L.) root bark. MethodsQualitative and quantitative phytoconstituents, antioxidant activity, sun protection factor and anti-elastase (skin wrinkles prevention) effects were assessed using standard pharmaceutical and cosmeceuticals assays. ResultsThe results of antioxidant, anti-elastase and sun protection factor activities were the highest for the acetone fraction with IC50 8.51 ± 1.94 μg/ml, 10.02 ± 0.3 μg/ml and SPF value of 6.38, respectively. However, these results may occur due to its high contents of flavonoids and phenols which were 26.55 ± 1.6 mg of QUE/g and 59.48 ± 0.56 mg of GAE/g plant acetone fraction, respectively. ConclusionThe acetone fraction of A. tinctoria root bark could be a promising candidate for cosmetic and pharmaceutical preparations due to the potential antioxidant, anti-elastase enzyme and sun protection activities.

The Anti-Wrinkle Mechanism of Melatonin in UVB Treated HaCaT Keratinocytes and Hairless Mice via Inhibition of ROS and Sonic Hedgehog Mediated Inflammatory Proteins.

Though melatonin is known to improve ultraviolet B (UVB)-induced oxidative damage and inflammatory conditions via the blockade of the nuclear factor (NF)-κB, interleukin (IL)-6, there is no report on the anti-wrinkle effect of melatonin to date. Hence in the present study, the anti-wrinkle mechanism of melatonin was elucidated in UVB treated HaCaT keratinocytes and hairless mice. Herein melatonin protected against a radical initiator tert-Butyl hydroperoxide (t-BOOH) induced reactive oxygen species (ROS) production, matrix metalloprotease 1 (MMP-1), pro-collagen and cytotoxicity in HaCaT keratinocytes. Additionally, melatonin suppressed the expression of sonic hedgehog (SHH) and GLI1 for hedgehog signaling and p-NF-κB, cyclooxygenase (COX-2), phospho-extracellular signal-regulated kinase-1 (p-ERK) for inflammatory responses in UVB treated HaCaT keratinocytes. Furthermore, melatonin protected skin from wrinkle formation, transdermal water loss in hairless mice irradiated by UVB for 8 weeks. Notably, melatonin prevented against epidermal thickness and dermal collagen degradation in UVB irradiated hairless mice by Hematoxylin and Eosin and Masson’s trichrome staining. Taken together, these findings suggest that melatonin reduces wrinkle formation via inhibition of ROS/SHH and inflammatory proteins such as NF-κB/COX-2/ERK/MMP1.

Human skin aging is caused by several factors, such as UV irradiation, stress, hormone, and pollution. Wrinkle formation and skin pigmentation are representative features of skin aging. Although EGF and arbutin are used as anti-wrinkle and skin whitening agents, respectively, they have adverse effects on skin. When more cosmeceutical ingredients are added to cosmetic product, adverse effects are also accumulated. For these reasons, multifunctional and safe cosmetic ingredients are in demand. The aim of the present study is to investigate the novel anti-aging agents, AIMP1-derived peptide (AdP, INCI name: sh-oligopeptide-5/sh-oligopeptide SP) for cosmetic products. To assess the anti-wrinkle effect of AdP, collagen type I synthesis and fibroblast proliferation were determined on human fibroblasts. The anti-wrinkle effect of AdP was examined by ELISA and cell titer glo assay. To assess the whitening, melanin content and tyrosinase activity were determined on melanocytes. The whitening effect of AdP was examined by melanin measurement and enzyme activity assay. The safety of AdP was determined by cytotoxicity and immunogenicity, CCK-8 and TNF-α ELISA assay, respectively. AdP treatment induced the collagen type I synthesis and fibroblast proliferation. Also, AdP treatment inhibited melanin synthesis by regulating tyrosinase activity. The anti-aging effect of AdP is more potent than EGF and albutin. AdP did not show adverse effects. These results show that AdP can be dual functional and safe cosmeceutical agent to prevent skin aging.

Preparation and characterization of nanoparticles from quaternized cyclodextrin-grafted chitosan associated with hyaluronic acid for cosmetics

Hyaluronic acid (HA, 20–50 kDa) is a hydrophilic macromolecule Peer review under responsibility of Shenyang Pharmaceutical University. with anti-wrinkle effects and moisturizing properties. However, its high molecular weight prevents it from penetrating into the deeper layers of the skin and, thus, limits its benefits to topical effects. Thus, the objective of this study is to prepare nanoparticles of quaternized cyclodextrin-grafted chitosan (QCD-g-CS) associated with HA in different molar ratios of QCD-g-CS and HA. The conjugation of the carboxylic moieties of HA and the amides of QCD-g-CS was confirmed by Fourier-transform infrared spectroscopy. Thus, the system was optimized to create nanoparticles with a small size (235.63 ± 21.89 nm), narrow polydispersity index (0.13 ± 0.02), and zeta potential of 16.07 ± 0.65 mV. The association efficiency and loading efficiency were determined by ultra-performance liquid chromatography as 86.77 ± 0.69% and 10.85 ± 0.09%, respectively. The spherical morphology of the obtained nanoparticles was confirmed by transmission electron microscopy. Moreover, the in-vitro hydrating ability was significantly higher (P < 0.001) than that of bulk HA (3.29 ± 0.41 and 1.71 ± 0.05 g water/g sample, respectively). The safety of these nanoparticles at concentrations in the range of 0.01–0.10 mg/ml was confirmed via tests on human skin fibroblasts. Together, these results demonstrate that the developed nanoparticles are promising for future applications in cosmetics.

Background: To obtain useful cosmetic resources, this study aimed to determine the non-saponin fatty acid and inhibitory activities of collagenase and elastase by treatment of Saccharomyces cerevisiae in supercritical fluid extracted oil of the adventitious root culture of wild mountain ginseng.BR Methods and Results: We performed supercritical fluid extraction at various conditions such as pressure, temperature, time, and use of co-solvents, unlike the n-hexane extraction for the adventitious roots culture of wild mountain ginseng. The non-saponin-fatty acid obtained from the oil of the adventitious roots culture was incresed by treatment with S. cerevisiae. The supercritical fluid extraction was conducted using gas chromatography. Non-saponin-fatty acid content, in the oil of adventitious roots culture of wild mountain ginseng treated with S. cerevisiae for 2 days were three times higher than that in the control. In addition, the oil of the adventitious roots culture treated with S. cerevisiae was investigated for the anti-wrinkle effect by using collagenase and elastase. The oil of adventitious roots culture treated with S. cerevisiae exhibited higher collagenase and elastase inhibitory activities than those in the control.BR Conclusions: Supercritical fluid extracted oil of the adventitious roots culture of wild mountain ginseng treated with S. cerevisiae was found to have decreased ratio of saturated fatty acids and incresed ratio and content of unsaturated fatty acids increased. Furthermore, it showed anti-wrinkle effects in vitro.

1456 Tensor effect of anigozanthos flavidus flower extract: Modulation of tenascin-X to regenerate the skin

Wrinkles are characterized by deterioration of the dermal fiber network due to a decreased expression of extracellular matrix proteins and their disorganization. Tenascin-X, a matrix glycoprotein, is known to regulate the structure and stability of elastic fibers and the organization of collagen fibrils in the extra-cellular matrix (ECM), influencing cellular rigidity and elasticity. Therefore, the impact of the modulating tenascin-X expression in the skin is of great interest. To counteract these processes, we have developed a natural Australian plant extract of Anigozanthos Flavidus flower (AF), enabling the regeneration in vitro and in vivo the expression, organization and the functionality of the ECM proteins implicated in the tensor effect on skin aging. In vitro studies revealed the capacity of the AF extract to increase type I collagen, tenascin- X and elastin by 136, 39 and 43%, respectively. Moreover the Glassboxplus® model with equivalent dermis indicated that the extract increased significantly contractile forces of wrinkles fibroblasts by 10 and 19% after 6h and 24h of treatment, respectively. These data were confirmed in vivo: the study was conducted during 28 days with a twice daily application of AF cream at 2% on 42 women volunteers. One hour after the first application we observed a decrease by 5% of wrinkle depth as well as an improvement of the neck sagging by 24%. These effects were sustained for28 days as we improved the total anti-wrinkle effect by 10%. In conclusion, we have demonstrated that AF extract regenerates ECM by increasing matrix essential protein expression, organization and functionality. Anigozanthos Flavidus flower extract could be the new innovative skin care ingredient able to promote fast and long lasting tensor effect to counteract skin aging.

Anti-wrinkle effect of berberine by inhibition of MMP-2 and MMP-9 activity in fibroblasts

우리는 황련에서 분리된 베르베린의 화장품소재로의 활성을 평가하기 위해 항산화와 항주름 효능을 분석하였다. 시료의 독성을 평가하기 위해 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2Htetrazolium bromide (MTT) assay를 수행하였으며 항산화와 항주름 활성평가를 위해 ROS 생성능, hyaluronic acid 생성능, MMP-2, -9, TIMP-1,-2, TNF-α의 발현을 각각 측정하였다. 베르베린에 대한 자외선의 세포 독성은 CCD-986sk 섬유아세포를 이용하여 MTT 분석으로 측정되었고, 세포 독성이 25 μg/mL의 농도 미만에서 관찰되지 않았다. 또한 베르베린이 ROS 생산을 농도 의존적으로 감소시키고 히알루론산의 합성을 촉진한다는 것을 발견했다. 베르베린은 주름 형성과 관련된 MMP-2 및 MMP-9의 protein 단계 및 mRNA 발현을 감소시키고 TIMP-1 및 TIMP-2의 발현을 증가시켰다. 프로 pro-inflammatory cytokine 으로 알려진 TNF-α에 대한 베르베린의 억제효능을 실험한 결과, 농도의존적으로 TNF-α 유전자를 억제하는 효과를 나타내었다. 이러한 결과를 통해 베르베린은 활성산소종의 억제와 TIMP-1과 -2의 발현 조절에 의한 MMP-2,-9의 감소로 항산화 및 항주름에 효과가 있는 화장품 소재로서의 가치를 가진다고 사료된다.

이팝나무 꽃에서 분리한 페놀 화합물의 생리활성 효소 억제효과

This study was designed to determine the biological activities of Chionanthus retusus flower extracts. Water and 90% ethanol extracts of C. retusus flower were prepared. The inhibitory activities of water and ethanol extracts with a phenolic content of 200 μg/mL against xanthine oxidase were 25.60% and 15.92%, respectively. Further, the water extract did not show any inhibitory activity against α-glucosidase whereas the ethanol extract showed 100.00% inhibition of α-glucosidase. The inhibitory activities of the extracts against tyrosinase were 17.27% (water extract) and 36.13% (ethanol extract), which suggest that the extracts may have a whitening effect. The water extract did not inhibit elastase activity but showed a collagenase-inhibitory activity of 20.21%. On the contrary, the ethanol extract showed 96.26% and 35.93% inhibition of collagenase and elastase, respectively. These findings suggest that the extracts may have an anti-wrinkle effect. Lastly, the extracts showed a hyaluronidase inhibitory activity of 36.96% (water extract) and 88.70% (ethanol extract), suggesting that they may have an anti-inflammatory effect. The results indicate that C. retusus flower extracts containing phenolic compounds can be used as functional resources because they have anti-gout, carbohydrate degradation-inhibitory, whitening, anti-wrinkle, and anti-inflammatory effects.
