Whey protein nanoparticles: Enhancing solubility, environmental resistance, and bio-accessibility of pterostilbene

Pterostilbene (Pte) is a dietary phenolic compound derived from plants that offers numerous physiological advantages. Nonetheless, its low water solubility and limited oral bioavailability restrict its bioactivity. To address these limitations, nanoparticles were prepared using anti-solvent precipitation method. Optimal nanoparticles were achieved at a 10:1 mass ratio of whey protein isolate (WPI) to Pte, resulting in a particle size of 104.54 nm, a polydispersity index of 0.42, and an impressive encapsulation efficiency of Pte at 85.02%. The water solubility of Pte in the nanoparticles was found to be 53.14 times higher than that of free Pte and remained stable under various environmental conditions. Furthermore, the DPPH and ABTS+ free radical scavenging activities of the nanoparticles were maximized at 55.58% and 92.97%, respectively. Pte release from WPI-Pte nanoparticles was well-fitted to Korsmeyer-Peppas model and followed a non-Fickian mechanism controlled by both diffusion and erosion. This environmentally friendly, straightforward, and efficient approach shows promise for large-scale production of Pte products with amplified health benefits.

DEVELOPMENT, OPTIMIZATION AND IN VITRO CHARACTERIZATION OF HALOPERIDOL NANOCRYSTALS USING 32 FACTORIAL DESIGN

Objective: The main aim of the present study was to improve the dissolution rate of Haloperidol nanocrystals and thereby increase their bioavailability. Haloperidol is a typical antipsychotic drug and it is used to treat schizophrenia as well as acute mania and mixed states associated with bipolar disorder. Haloperidol falls into the Biopharmaceutics Classification System (BCS-II) class of drugs (poorly soluble aqueous and highly permeable) and has poor bioavailability. Methods: The present study involves the preparation and optimization of Haloperidol nanocrystals by the anti-solvent precipitation method using Polaxomer407 and polyvinyl pyrrolidone K30 (PVP K30). The prepared nanocrystals were evaluated for various parameters like particle size, zeta potential, % drug content, % yield, surface morphology, drug-excipient compatibility studies (Fourier-transform infrared spectroscopy (FT-IR) and Differential Scanning Calorimetry (DSC)), and in vitro dissolution studies. Results: Nine preparations were done and the best preparation amongst them was selected for further studies. F7 preparation containingpolaxomer407 and PVP K30 was selected as optimized preparation based on their evaluation parameters. 32 factorial design was used in the preparation. The particle size of the F7 nanocrystals was 300.2±2.7 nm and the zeta potential-36.3±3.2 mV. The % yield was in the range of 63.62±0.3%-98.21±0.8 %. The drug content of various preparations was found to be in the range of 58.46±0.8%-93.54±0.5 %. In vitro dissolution studies showed the highest % drug release for F7(91.54±0.03%) in 10 h. Conclusion: F7 preparation was found to be having acceptable characteristics and thus selected as optimized preparation.

Preparation of silymarin-loaded zein polysaccharide core–shell nanostructures and evaluation of their biological potentials

Abstract Silymarin-loaded zein polysaccharide core–shell nanoparticles (SZPCS-NPs) were synthesized where sodium alginate and pectin offer stability and controlled release qualities to zein, a maize protein, having excellent biocompatibility. The present study is an attempt to develop zein–silymarin polysaccharide core–shell nanostructures to enhance water solubility, thereby improving bioavailability and producing enhanced biological responses in living systems. SZPCS-NPs were prepared using pH-induced antisolvent precipitation method. Five different types of SZPCS-NPs were synthesized using different combinations of sodium alginate and pectin, namely P100–A00 (non-uniform size ranging from 20 to 100 nm), P70–A30 (spherical and uniform size measuring approximately 80 nm in diameter), P50–A50, P30–A70, and P00–A100 exhibited irregular shapes with the presence of some triangular and oval structures and non-uniform size ranging from 20 to 100 nm. The SZPCS-NPs P70–A30 possessed the best results in terms of shape, size, and other characterization studies. Furthermore, the SZPCS-NPs possessed a percent drug loading of 72.5% and entrapment efficiency of 51.7%, respectively. The resulting SZPCS-NPs exhibited an excellent relative bioavailability percentage of 97.4% in comparison to commercial silymarin, having 58.1%, and crude silymarin, having 46.97% bioavailability percentage, correspondingly. In addition, SZPCS-NPs possessed an almost two folds’ increase in antioxidant activity in comparison to crude and commercially available silymarin. Similarly, SZPCS-NPs also showed better stabilization in hepatic biomarker enzymes and possessed better hepatoprotective activity for a period of 6 weeks, in contrast to commercial and crude silymarin formulations.

Modification of physicochemical properties of chitosan to improve its pharmaceutical and agrochemical potential applications

Chitosan has received much more attention as a functional biopolymer with applications in pharmaceuticals, agricultural, drug delivery systems and cosmetics.The objectives of present investigation were to carry out modification of chitosan for enhancement of aqueous solubility, which will impart increased solubility and dissolution rate of poorly soluble drug itraconazole (ITZ) and also evaluate the modified chitosan for soyabean seed germination studies.The modification of chitosan was accomplished through the antisolvent precipitation method; employing five carboxylic acids. The resulting products were assessed for changes in molecular weight, degree of deacetylation, solubility and solid state characterization. Subsequently, the modified chitosan was complexed with itraconazole using the co-grinding technique. The prepared formulations were evaluated for solubility, FTIR (Fourier-transform infrared spectroscopy), PXRD (Powder X-ray diffraction), in-vitro dissolution studies. Furthermore the effect of modified chitosan has been evaluated on soybean seed germination.Results demonstrated that, modified chitosan improves self and solubility of itraconazole by six folds. As there was increased degree of deacetylation of chitosan leads to improvement in solubility. The results of FTIR showed the slight shifting of peaks in co-grind formulations of itraconazole. Formulations showed reduction in crystallinity of drug which leads to enhancement in dissolution rate as compared to pure itraconazole. Retention of property of seed germination was observed with modified chitosan at optimum concentration of 3 % w/v, with benefit of enhanced aqueous solubility of chitosan.This positive result paves the way for the advancement of pharmaceutical and agrochemical products employing derivatives of chitosan.

Synthesis, physiochemical characterization, molecular docking study, and anti-breast cancer activity of silymarin loaded zein nanoparticles

Breast cancer is a major cause of death in women worldwide leading to requirement of new therapeutic strategies. Silymarin demonstrated the anti-cancer activity however, due to low bioavailability its use is restricted. This study aimed to improve the solubility of silymarin by developing a silymarin loaded zein nanoparticles (SLNPs) which was stabilized by beta cyclodextrin. Comprehensive physiochemical characterization studies based on DLS, FTIR, UV–Vis Spectroscopy, FE-SEM, TEM, XRD, DSC, NMR and TGA confirmed the successful synthesis of SLNPs via an anti-solvent precipitation method. FE-SEM and TEM images demonstrated the uniform size and spherical shape of nanoparticles with encapsulation and loading efficiencies of 84.32 ± 1.9 % and 15.25 ± 2.4 % respectively. The zein protein interaction with silymarin, and β-cyclodextrin was shown to be beneficial via the use of molecular simulations and binding energy calculations. Cellular studies demonstrated dose and time dependent cytotoxicity of SLNPs on MCF-7 breast cancer cell. FACS, qRT-PCR and Western blotting showed Bax (pro-apoptotic) upregulation while Bcl-2 (anti-apoptotic) downregulation. Our findings suggest that these loaded nanoparticles are more efficient than pure drug, enhancing its bioavailability and paving the path for developing it as a promising nutraceutical to treat breast cancer.

Pickering Emulsion Stabilized by Hordein-Whey Protein Isolate Complex: Delivery System of Quercetin.

As a lipophilic flavonol, quercetin has low bioavailability, which limits its application in foods. This work aimed to prepare a hordein-based system to deliver quercetin. We constructed hordein-whey isolate protein fibril (WPIF) complexes (H-Ws) by anti-solvent precipitation method at pH 2.5. The TEM results of the complexes showed that spherical-like hordein particles were wrapped in WPIF clusters to form an interconnected network structure. FTIR spectra revealed that hydrogen bonds and hydrophobic interactions were the main driving forces for the complex formation. H-W1 (the mass ratio of hordein to WPIF was 1:1) with a three-phase contact angle of 70.2° was chosen to stabilize Pickering emulsions with oil volume fractions (φ) of 40-70%. CLSM images confirmed that the oil droplets were gradually embedded in the three-dimensional network structure of H-W1 with the increase in oil volume fraction. The emulsion with φ = 70% showed a tight gel structure. Furthermore, this emulsion exhibited high encapsulation efficiency (97.8%) and a loading capacity of 0.2%, demonstrating the potential to deliver hydrophobic bioactive substances. Compared with free quercetin, the bioaccessibility of the encapsulated quercetin (35%) was significantly improved. This study effectively promoted the application of hordein-based delivery systems in the food industry.

Multi-spectroscopies and molecular docking insights into the interaction mechanism and antioxidant activity of isoquercetin and zein nanoparticles

The purpose of this study was to compare and characterize the theoretical properties and interaction mechanisms of zein and isoquercetin (ISO) from experimental and theoretical perspectives. Zein nanoparticles with different ISO concentrations (ZINPs) were prepared by the antisolvent precipitation method. The experimental results indicated all particles appeared spherical. When the mass ratio of zein to ISO was 10:1, the encapsulation efficiency of ZINPs reached 88.19 % with an average diameter of 126.67 nm. The multispectral method and molecular docking results confirmed that hydrogen bonding and van der Waals force played a dominant role for the binding of ISO to zein, and the primary fluorescence quenching mechanism for zein by ISO was static quenching. Furthermore, ZINPs had greater solubility and antioxidant activity, as well as inhibited the release of ISO during simulated gastrointestinal digestion processes. This research contributes to the understanding of the non-covalent binding mechanism between zein and ISO, providing a theoretical basis for the construction of ISO active carriers.

The Use of Data Mining for Obtaining Deeper Insights into the Fabrication of Prednisolone-Loaded Chitosan Nanoparticles.

The present work explores a data mining approach to study the fabrication of prednisolone-loaded chitosan nanoparticles and their properties. Eight PLC formulations were prepared using an automated adaptation of the antisolvent precipitation method. The PLCs were characterized using dynamic light scattering, infrared spectroscopy, and drug release studies. Results showed that that the effective diameter, loading capacity, encapsulation efficiency, zeta potential, and polydispersity of the PLCs were influenced by the concentration and molecular weight of chitosan. The drug release studies showed that PLCs exhibited significant dissolution enhancement compared to pure prednisolone crystals. Principal components analysis and partial least squares regression were applied to the infrared spectra and the DLS data to extract higher-order interactions and correlations between the critical quality attributes and the diameter of the PLCs. Principal components revealed that the spectra clustered according to the type of material, with PLCs forming a separate cluster from the raw materials and the physical mix. PLS was successful in predicting the ED of the PLCs from the FTIR spectra with R2 = 0.98 and RMSE = 27.18. The present work demonstrates that data mining techniques can be useful tools for obtaining deeper insights into the fabrication and properties of PLCs, and for optimizing their quality and performance. It also suggests that FTIR spectroscopy can be a rapid and non-destructive method for predicting the ED of PLCs.

Formulation of Mentha piperita-Based Nanobiopesticides and Assessment of the Pesticidal and Antimicrobial Potential.

The excessive use of synthetic pesticides has detrimental impacts on humans, non-target organisms, and the environment. Insect pest management strategies are shifting toward biopesticides, which can provide a feasible and environmentally friendly green solution to the pest problem. The key objective of the present research work was the preparation of Mentha piperita-based nanobiopesticides with enhanced stability, solubility, and pesticidal potential. Nanobiopesticides based on the Mentha piperita extract were prepared using the antisolvent precipitation method. The central composite design of response surface methodology (RSM) was utilized to optimize different process parameters, e.g., the amounts of the stabilizer and plant extract. The nanosuspension of Mentha piperita prepared with the stabilizer SLS showed a particle size of 259 nm and a polydispersity index of 0.61. The formulated biopesticides in the form of nanosuspensions showed good antibacterial activities as compared to the Mentha piperita extract against two phytopathogenic bacterial strains, Clavibacter michiganensis and Pseudomonas syringae. The M. piperita nanosuspension had higher antifungal efficacy against A. niger and F. oxysporum than the Mentha piperita extract. The M. piperita extract and its nanosuspensions were tested for pesticidal activity against the stored-grain insects Tribolium castaneum and Sitophilus oryzae. Mentha piperita-based nanobiopesticides demonstrated significantly high (p < 0.05) average mortality of 84.4% and 77.7% against Tribolium castaneum and Sitophilus oryzae, respectively. Mentha piperita-based nanobiopesticides showed enhanced pesticidal potential and could be used as a good alternative to synthetic chemical pesticides.

Fabrication of Nanocrystals for Enhanced Distribution of a Fatty Acid Synthase Inhibitor (Orlistat) as a Promising Method to Relieve Solid Ehrlich Carcinoma-Induced Hepatic Damage in Mice.

Orlistat (ORL) is an effective irreversible inhibitor of the lipase enzyme, and it possesses anticancer effects and limited aqueous solubility. This study was designed to improve the aqueous solubility, oral absorption, and tissue distribution of ORL via the formulation of nanocrystals (NCs). ORL-NC was prepared using the liquid antisolvent precipitation method (bottom-up technology), and it demonstrated significantly improved solubility compared with that of the blank crystals (ORL-BCs) and untreated ORL powder. The biodistribution and relative bioavailability of ORL-NC were investigated via the radiolabeling technique using Technetium-99m (99mTc). Female Swiss albino mice were used to examine the antitumor activity of ORL-NC against solid Ehrlich carcinoma (SEC)-induced hepatic damage in mice. The prepared NCs improved ORL's solubility, bioavailability, and tissue distribution, with evidence of 258.70% relative bioavailability. In the in vivo study, the ORL-NC treatment caused a reduction in all tested liver functions (total and direct bilirubin, AST, ALT, and ALP) and improved modifications in liver sections that were marked using hematoxylin and eosin staining (H&E) and immunohistochemical staining (Ki-67 and ER-α) compared with untreated SEC mice. The developed ORL-NC could be considered a promising formulation approach to enhance the oral absorption tissue distribution of ORL and suppress the liver damage caused by SEC.

Fabrication of zein-tamarind seed polysaccharide-curcumin nanocomplexes: their characterization and impact on alleviating colitis and gut microbiota dysbiosis in mice.

In this work, a zein-tamarind seed polysaccharide (TSP) co-delivery system was fabricated using an anti-solvent precipitation method. The formation mechanism, characterization, and effect on alleviating colitis and gut microbiota dysbiosis in mice of zein-TSP-curcumin (Z/T-Cur) nanocomplexes were investigated. Hydrogen bonding and the hydrophobic effect played a key role in the formation of Z/T-Cur nanocomplexes, and the interactions were spontaneous and driven by enthalpy. The encapsulation efficiency, loading capacity, and bioavailability increased from 60.8% (Zein-Cur) to 91.7% (Z/T-Cur1:1), from 6.1% (Zein-Cur) to 18.3% (Z/T-Cur1:1), and from 4.7% (Zein-Cur) to 20.0% (Z/T-Cur1:1), respectively. The Z/T-Cur significantly alleviated colitis symptoms in DSS-treated mice. Additionally, the prepared nanocomplexes rebalanced the gut microbiota composition of colitis mice by increasing the abundance of Akkermansia. Odoribacter and Monoglobus were rich in the Z-T-Cur treatment group, and Turicibacter and Bifidobacterium were rich in the zein-TSP treatment group. This study demonstrated that the TSP could be helpful in the targeted drug delivery system.

Development of antimicrobial and antioxidant film by incorporation of modified protein self-assembled nanoparticles into Pickering emulsion

In this study, Pickering emulsion (PE) containing zein/Tannic acid (TA) nanoparticles (ZTNPs) and oregano oil (OEO) was incorporated into konjac glucomannan (KGM) film for developing active film with antioxidant and antimicrobial properties. TA anchored the amino group of zein to form covalent bond, and self-assembled to form nanoparticles via the antisolvent precipitation method. The interactions between TA and zein were characterized through FT-IR and molecular docking. The physicochemical properties and morphological characterization of ZTNPs under different zein/TA mass ratios were investigated, significant changes were observed in the Zeta-potential and particle size. ZTNP-0.2 exhibited a uniform and smooth surface (particle size≈126 nm), demonstrating exceptional antioxidant activity and highlighting the enhanced bioavailability of TA. High-speed shearing formed PE loading ZTNP-0.2 and OEO, which was applied to the preparation of active films. The oil phase was evenly distributed in the film matrix, the presence of interfaces between nanoparticles prevented flocculation and aggregation between OEO droplets, and maintained the mechanical and barrier properties of the KGM-P film. The KGM-P film demonstrated remarkable inhibitory effects against S. aureus, E. coli, and A. niger, with inhibition area diameters measuring 39.7 ± 0.46 mm, 19.92 ± 0.45 mm, and 18.70 ± 0.8 mm respectively. Furthermore, the composite KGM-P film exhibited substantial scavenging rates of 87.5% for DPPH and 89.8% for ABTS, underscoring its exceptional antibacterial and antioxidant activities. In conclusion, the active KGM film holds promise for application in food packaging, effectively extending the shelf life of products by retarding oxidation and mitigating microbial intrusion.

Fabrication and Characterization of Zein/PDADMAC Nanoparticles to Encapsulate Tebuconazole by Antisolvent Precipitation Method**

AbstractIn order to improve the sustained release effect and antifungal activity of tebuconazole (TEB), zein /poly dimethyl diallyl ammonium chloride (PDADMAC) nanoparticles (NPs) were prepared in this study. The hollow zein NPs were made by anti‐solvent precipitation method in which sodium carbonate was chosen as sacrificial template, and PDADMAC was combined with zein NPs via electrostatic self‐assembly after loading TEB. The NPs were characterized through particle size, Zeta potential, scanning electron microscopy (SEM), sustained release effect, contact angle, encapsulation efficiency (EE), loading efficiency (LE) and antifungal activity. In this study, TEB/zein/PDADMAC NPs were stable for at least 28 days at room temperature. The release of TEB showed lower release in NPs compared with technical concentrate (TEB TC, 98 %). In addition, NPs also significantly influenced performance such as wetting performance and antifungal activity. The developed functional TEB/zein/PDADMAC NPs with excellent properties can be promising for antifungal pesticide applications.

Zein-yeast carboxymethyl glucan particles formed by anti-solvent precipitation for encapsulating resveratrol

In the work, zein-yeast carboxymethyl glucan (ZY) particles were fabricated by a novel ultrasonic assisted anti-solvent precipitation (ASP) method, which was a good delivery system for resveratrol. The particle size and zeta-potential of ZY samples were detected by Zetasizer Pro analyzer, they gradually increased as the mass ratio of zein and yeast carboxymethyl glucan (YCG) changed from 10:1 to 10:5. The intermolecular interactions were investigated by zeta-potentiometric analyzer, Fourier transform infrared spectroscopy and fluorescence spectroscopy. Electrostatic interaction, hydrogen bonding and hydrophobic effects between zein and YCG molecules were identified as the main driving forces in the formation of ZY particles. The optimized ZY (10:3) binary particles were used as delivery system for encapsulating and protecting resveratrol. They had high encapsulation efficiency (85.4 %) and loading capacity (6.1 %), and increased the retention rate of resveratrol by 2.10 and 1.21 folds after exposure to light and heat conditions, effectively protect resveratrol against light and thermal degradation. These particles also delayed the release of resveratrol in simulated gastrointestinal digestion, which might improve its oral bioavailability. In conclusion, ZY binary particles could be regarded as a useful and promising delivery vehicle, which might contribute to the application of hydrophobic bioactive ingredients in functional foods.

Interactions and characterization of Zein-BSA nanoparticles: Multi-spectral analysis and molecular simulations

Zein-bovine serum albumin (BSA) nanoparticles (NPs) with varying mass ratios were prepared via anti-solvent precipitation method at pH 4.0. Zein-BSA NPs at high Zein mass ratios exhibited superior particle characteristics and surface morphology. Ultraviolet-visible, fluorescence, and X-ray diffraction analyses demonstrated the formation and conformational transition during the binding of Zein and BSA, while ANS hydrophobic binding, fourier transform infrared spectra, and circular dichroism confirmed the hydrophobic exposure and secondary structure assignment. Molecular dynamic simulations revealed that Zein spontaneously bound to the IIB domain of BSA with a binding free energy of −63.52 ± 6.37 kcal/mol, which was dominated by hydrophobic and electrostatic interactions, supplemented by hydrogen bonds. Dissociation experiments supported the key role of hydrophobic and electrostatic interactions in maintaining the stability of the NPs. These findings provided insights into the formation and stabilization mechanisms of Zein-BSA NPs, and their potential applications as encapsulation carriers for bioactive substances or as novel nanoscale food substrates.

Preparation and characterization of particle-filled microgels by chemical cross-linking based on zein and carboxymethyl starch for delivering the quercetin

This work aimed to develop novel particle-filled microgels based on zein and carboxymethyl starch for delivering quercetin (Que). The anti-solvent precipitation and chemical cross-linking methods were combined to produce the zein-carboxymethyl starch particle-filled microgels (SM-Z). The critical finding of the study was that adding zein nanoparticles significantly improved the strength, water holding capacity, and thermal stability of carboxymethyl starch microgel (SM). Besides, SM-Z had good biodegradability, and the particle size was about 44–61 μm. SM-Z successfully encapsulated Que with a high encapsulation efficiency of 86.7 %. Que-loaded SM-Z (Q/SM-Z) significantly enhanced 30 d storage and UV light stability (up to 89.4 % retention rate) of Que than the Que-loaded SM (Q/SM). Q/SM-Z exhibited pH-responsive swelling behavior, and the swelling was greatest in the simulated intestinal fluid (pH = 7). Besides, the Q/SM-Z showed good stability in simulated gastric fluids and sustained release of Que in simulated intestinal fluids, 72.5 % Que was released at 8 h. During Que transport in Caco-2 cell monolayers, Q/SM (5.8 %) and Q/SM-Z (9.7 %) were significantly higher than free Que (1.93 %). Therefore, as an oral delivery system for hydrophobic active substances, SM-Z possesses good biodegradability and pH-responsive intestinal-targeted delivery capability, providing a new strategy for designing starch-based encapsulation materials.

Curcumin loaded Zein-alginate nanogels with “core-shell” structure: formation, characterization and simulated digestion

Zein nanoparticles tend to aggregate in water and are readily digested by enzymes in the gastrointestinal tract. In current study, the Zein-alginate nanogels loaded with curcumin (Cur@ZA) were fabricated with the “core-shell” structure. The Zein “core” was prepared via antisolvent precipitation method, and the alginate gel “shell” was formed by calcium-induced gelation method. The physicochemical properties, microstructure, encapsulation efficiency, stability and simulated digestion characteristics of nanogels were investigated. The results showed that Cur@ZA formed uniform gel spheres with small particle size (415.10 nm), while possessing a dense gel shell on the surface. The Zein “core” and alginate gel “shell” of Cur@ZA are tightly bound to each other by electrostatic adsorption, hydrophobic interaction and hydrogen bonding. Curcumin was able to be loaded in the Cur@ZA nanogels with a higher encapsulation rate (>92 %). Compared with the system which was not induced by calcium ion, the addition of calcium ions improved the photostability and thermal stability of curcumin, and facilitated slow and sustained release of curcumin in the simulated digestion. Therefore, this novel nanogel delivery system has the ideal physicochemical properties, stability and control-release ability, which has the potential to be used in the food industry.

Photocatalytic performance of buoyant TiO2-immobilized poly(ethylene terephthalate) beads for the removal of the anticonvulsant drug pregabalin from water and leachate

The application of TiO2 nanoparticles as photocatalyst for the removal of pharmaceutical compounds from water is extensively applied as an alternative to the conventional treatment processes. However, application of TiO2 as a powder requires energy costing separation stages to remove the catalyst at the end of the process. Thus, immobilization of the catalyst can be an attractive solution. Aiming to find a good support material for TiO2, in this study, the synthesis and characterization of three different PET/TiO2 photocatalytic beads are described along with their application in the photocatalytic degradation of the anticonvulsant drug pregabalin (PGB). The PET/TiO2 beads with different contents of TiO2 filler (10 wt%, 20 wt% and 30 wt%) were synthesized by using an anti-solvent precipitation method and were characterized by different techniques, such as Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), X-ray powder diffraction (XRD), thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC). Among the different composites, PET/TiO2 30 wt% beads proved to be the most efficient, achieving almost complete degradation of PGB within 6 h of treatment. The stability and reuse of the synthesized catalysts were also evaluated. Furthermore, the effect of matrix was examined, revealing slower kinetics in high loaded matrices like a leachate. Identification of the arising transformation products (TPs) was conducted using LC-HRMS analysis. Twenty-two TPs were identified, out of which fifteen were detected for the first time herein, stemming from hydroxylation, amidation, oxidation and oxidative deamination of PGB. Finally, the in silico prediction of their toxicity was conducted based on ECOSAR software revealing mainly the formation of non-toxic compounds. However, amidation can cause the formation of toxic/harmful by-products but they are susceptible to photocatalysis thus being eliminated until the end of treatment.

Preparation and characterization of novel composite nanoparticles using zein and hyaluronic acid for efficient delivery of naringenin.

Hyaluronic acid (HA), a polymer mainly found in animal tissues, plays an important role in food research. In this study, it was used for delivery improvement of naringenin (NAR) by loading it into zein nanoparticles using an anti-solvent precipitation method. The optimal Nar/zein-HA nanoparticles were uniformly spherical with particle sizes of 209.2±1.9nm, polydispersity indexes of 0.146±0.032 and zeta-potentials of -19.0±0.7mV. Moreover, the microstructure of Nar/zein-HA nanoparticles was maintained primarily by hydrophobic, electrostatic, and hydrogen-bonding interactions. Furthermore, Nar/zein-HA nanoparticles showed favorable physical stability and enhanced encapsulation efficiency. Additionally, the antioxidant capacity and release in simulated gastrointestinal digestion of Nar were significantly improved. Overall, these findings indicate that the delivery efficiency of Nar was improved by formulation of ternary nanoparticles.

Impact of solvent polarity on the morphology, physicochemical properties, and digestibility of A-type resistant starch particles.

Resistant starch particles (RSP) formed by antisolvent precipitation method has attracted much attention as a functional food ingredient having beneficial effects on obesity and diabetes. However, the effect of solvent polarity on the physicochemical properties and digestibility of RSP remains unclear. Here, n-propanol, isopropanol, acetone, and ethanol were employed as antisolvents to prepare RSP. The width and length of the resulting RSP decreased from 0.87μm to 0.59μm and from 2.56μm to 1.31μm, respectively, upon increasing the solvent polarity, while dramatically decreasing their crystallinity and the gelatinization enthalpy from 80.5% to 62.3% and from 67.9±14.4J/g to 41.5±8.3J/g, respectively, suggesting that solvent polarity is critical factor determining morphology, crystallinity, and thermostability of RSP. Furthermore, the level of resistant starch in RSP was found to be inversely proportional to the degree of solvent polarity, which would provide a useful means for modulating the digestibility of RSP.
