Interruption after Short-Term Nitrogen Additions Improves Ecological Stability of Larix olgensis Forest Soil by Affecting Bacterial Communities.

Atmospheric nitrogen deposition can alter soil microbial communities and further impact the structure and function of forest ecosystems. However, most studies are focused on positive or negative effects after nitrogen addition, and few studies pay attention to its interruption. In order to investigate whether interruption after different levels of short-term N additions still benefit soil health, we conducted a 2-year interruption after a 4-year short-term nitrogen addition (10 and 20 kg N·hm-2·yr-1) experiment; then, we compared soil microbial diversity and structure and analyzed soil physicochemical properties and their correlations before and after the interruption in Larix olgensis forest soil in northeast China. The results showed that soil ecological stabilization of Larix olgensis forest further improved after the interruption compared to pre-interruption. The TN, C:P, N:P, and C:N:P ratios increased significantly regardless of the previous nitrogen addition concentration, and soil nutrient cycling was further promoted. The relative abundance of the original beneficial microbial taxa Gemmatimonas, Sphingomonas, and Pseudolabrys increased; new beneficial bacteria Ellin6067, Massilia, Solirubrobacter, and Bradyrhizobium appeared, and the species of beneficial soil microorganisms were further improved. The results of this study elucidated the dynamics of the bacterial community before and after the interruption of short-term nitrogen addition and could provide data support and a reference basis for forest ecosystem restoration strategies and management under the background of global nitrogen deposition.

Biochar as a green solution to drive the soil carbon pump

Biochar is a nature-based green solution to lift soil carbon storage and mitigate carbon release. Here, we propose a novel concept of biochar carbon pump (BCP) that bridges microbial carbon pump (MCP) and mineral carbon pump (MnCP), facilitating effective carbon sequestration. The BCP functions to promote carbon storage by introducing biochar-derived persistent C into soil, enhancing negative priming effects, altering soil microbial communities, and reinforcing organo-organic and organo-mineral interactions. Recognizing the value of BCP in bridging MCP and MnCP to facilitate diverse natural reactions for soil carbon sequestration is particularly significant in addressing climate change.

Adaptive multi-paddock grazing increases mineral associated soil carbon in Northern grasslands

Grassland soils play an important role in sequestering carbon (C) and are primarily used for livestock grazing. Grazing management can increase the amount of C stored in soils and the distribution of C in different soil fractions by altering soil microbial community structure, thereby influencing the persistence of soil C over time. Adaptive multi-paddock grazing (AMP), grazing cattle at high stock densities with long periods between grazing events, is considered to enhance grassland sustainability, productivity, and soil C compared to more conventional grazing practices (continuous to slow rotational). However, whether and how AMP grazing might affect the amount of C stored in different soil fractions has received little attention. Soil samples (0–15 cm depth) were collected from 24 ranches in a paired design, where 12 ranches practiced AMP grazing and 12 neighboring ranches practiced conventional grazing. Soil organic C (SOC) and total nitrogen were measured in different soil particle size [fine (<53 µm), medium (53–250 µm) and coarse (>250 µm)], and density [light (>1.6 g cm−3), and heavy (<1.6 g cm−3)] fractions. Soil bacterial and fungal biomass and abundances were obtained from phospholipid fatty acid analysis. Mean weights of soil microaggregates and heavy fractions were higher in soil exposed to AMP grazing, whereas mean weights of macroaggregates and light fractions were higher in conventionally grazed soils. In warmer and drier climatic conditions, SOC in both bulk soils and within various size and density fractions tended to decrease. Soil organic C (both concentration and stock) were significantly higher in the fine soil fractions under AMP management than conventionally grazed grasslands. Given that SOC in fine fractions, is more mineral associated and recalcitrant, our results indicate that AMP grazing increased the size of the stable SOC pool. The increased carbon pool in the fine fraction was associated with greater fungal to bacterial ratios, suggesting a possible biological mechanism for the increase. Overall, the results indicate that AMP grazing may be beneficial for sequestering more stable carbon that helps mitigate the effects of climate change.

Effect of n-hexadecanoic acid on N2O emissions from vegetable soil and its synergism with Pseudomonas stutzeri NRCB010

Recently, it has been recommended to utilize specific plant root metabolites to decrease nitrous oxide (N2O) emissions from agricultural soil. However, only a limited number of plant root metabolites have been shown to decrease soil N2O emissions, and their combined effects with plant growth-promoting rhizobacteria, as well as the mechanisms involved in mitigating N2O emissions, remain to be explored. This study investigated the impact and microbial mechanisms of three tomato root exudate metabolites, namely, methyl hexadecanoate, methyl stearate, and n-hexadecanoic acid, on N2O emissions. In a pure culture experiment, n-hexadecanoic acid significantly enhanced Pseudomonas stutzeri NRCB010 auxin production, nitrogen removal, N2O reduction capacity, and N2O reductase activity. Methyl stearate significantly promoted NRCB010 N removal and the N2O reduction capacity. In soil microcosm and greenhouse pot experiments, n-hexadecanoic acid decreased N2O cumulative emissions by 34.3 % and 46.6 %, respectively, and further decreases were observed when combined with NRCB010. Additionally, n-hexadecanoic acid, both independently and in combination with NRCB010, significantly promoted tomato growth. N-hexadecanoic acid, alone or with NRCB010, also altered soil microbial communities and nitrogen-cycle functional gene abundance. Structural equation models revealed that n-hexadecanoic acid, either alone or with NRCB010, decreased N2O cumulative emissions by modifying the environmental conditions to be more conducive to N2O mitigation (e.g., higher soil pH and organic matter content), inhibiting N2O production (through a decrease in ammonia-oxidizing bacteria and nirK gene copy numbers), and promoting N2O reduction (by increasing nosZII gene copy numbers). Our results support the potential use of n-hexadecanoic acid in enhancing crop productivity and mitigating N2O emissions in agricultural soils.

Continuous cropping system altered soil microbial communities and nutrient cycles.

Understanding the response of microbial communities and their potential functions is essential for sustainability of agroecosystems under long-term continuous cropping. However, limited research has focused on investigating the interaction between soil physicochemical factors and microbial community dynamics in agroecosystems under long-term continuous cropping. This study probed into the physicochemical properties, metabolites, and microbial diversity of tobacco rhizosphere soils cropped continuously for 0, 5, and 20 years. The relative abundance of bacterial genera associated with nutrient cycling (e.g., Sphingomonas) increased while potential plant pathogenic fungi and beneficial microorganisms showed synergistic increases with the duration of continuous cropping. Variations in soil pH, alkeline nitrogen (AN) content, and soil organic carbon (SOC) content drove the shifts in soil microbial composition. Metabolites such as palmitic acid, 3-hydroxypropionic acid, stearic acid, and hippuric acid may play a key role in soil acidification. Those results enhance our ability to predict shifts in soil microbial community structure associated with anthropogenic continuous cropping, which can have long-term implications for crop production.

Metagenomics reveals the response of desert steppe microbial communities and carbon-nitrogen cycling functional genes to nitrogen deposition.

Nitrogen (N) deposition seriously affects the function of carbon (C) and N cycling in terrestrial ecosystems by altering soil microbial communities, especially in desert steppe ecosystems. However, there is a need for a comprehensive understanding of how microorganisms involved in each C and N cycle process respond to N deposition. In this study, shotgun metagenome sequencing was used to investigate variations in soil C and N cycling-related genes in the desert steppe in northern China after 6 years of the following N deposition: N0 (control); N30 (N addition 30 kg ha-1 year-1): N50 (N addition 50 kg ha-1 year-1). N deposition significantly increased the relative abundance of Actinobacteria (P < 0.05) while significantly decreased the relative abundances of Proteobacteria and Acidobacteria (P < 0.05). This significantly impacted the microbial community composition in desert steppe soils. The annual addition or deposition of 50 kg ha-1 year-1 for up to 6 years did not affect the C cycle gene abundance but changed the C cycle-related microorganism community structure. The process of the N cycle in the desert steppe was affected by N deposition (50 kg ha-1 year-1), which increased the abundance of the pmoA-amoA gene related to nitrification and the nirB gene associated with assimilation nitrite reductase. There may be a niche overlap between microorganisms involved in the same C and N cycling processes. This study provides new insights into the effects of N deposition on soil microbial communities and functions in desert steppe and a better understanding of the ecological consequences of anthropogenic N addition.

Soil C:N impacts on soil biological health and consequences on weed control in soybean and corn systems

Abstract Nitrogen availability has an important influence on agricultural weed growth since many weeds in annual cropping systems are more competitive in high-nitrogen soils. A potential method to control nitrogen availability is through soil carbon amendments, which stimulate soil microbial growth and immobilize nitrogen. Additionally, carbon amendments may alter soil microbial community composition, increase soil biological functioning, and improve soil health. In a two-year field experiment in corn (Zea mays L.) and soybean [Glycine max (L.) Merr.], we implemented five amendment treatments to test their ability to alter weed and crop growth through soil nitrogen availability and soil biological functioning. The treatments included: an untreated control, an unamended weed-free control, rye hay adding 3,600 kg C·ha-1 and 3,400 kg C·ha-1 each year, sawdust adding 5,000 kg C·ha-1 and 4,400 kg C·ha-1 each year, and a rye hay and sawdust combined treatment adding 8,600 kg C·ha-1 and 7,700 kg C·ha-1 each year. Each treatment was replicated 5 times in corn and 6 times in soybean. Each season, we explored correlations between crop and weed biomass and weed community composition and nitrogen immobilization measured through soil respiration and nitrogen availability. We also explored changes to the soil microbial community composition and soil health as a secondary result of the carbon amendment treatments. Nitrogen availability was lowest in plots treated with the highest C:N amendment. Increasing carbon improved soil health metrics, but the microbial community composition was most affected by the rye hay treatment. Amendments with high C:N reduced weed growth in both soybean and corn plots but only selected for specific weed communities in soybeans, leading to improved soybean competitiveness against weeds. In corn, crop growth and weed community composition remained consistent across amendment treatments. Targeted nitrogen immobilization may improve leguminous crop competition in some weed communities as part of an integrated weed management program.

Inoculation with Stutzerimonas stutzeri strains decreases N₂O emissions from vegetable soil by altering microbial community composition and diversity.

Plant growth-promoting rhizobacteria (PGPR) have been applied to mitigate nitrous oxide (N₂O) emissions from agricultural soils, but the microbial ecological mechanisms underlying N₂O mitigation are poorly understood. That is why only limited PGPR strains can mitigate N₂O emissions from agricultural soils. Therefore, it is of substantial significance to reveal soil ecological mechanisms of PGPR strains to achieve efficient and reliable N₂O-mitigating effect after inoculation. Inoculation with Stutzerimonas stutzeri strains decreased N₂O emissions from two soils with contrasting textures probably by altering soil microbial community composition and gene abundance involved in nitrification and denitrification. Our findings provide detailed insight into soil ecological mechanisms of PGPR strains to mitigate N₂O emissions from vegetable agricultural soils.

Afforestation alters soil microbial community composition and reduces microbial network complexity in a karst region of Southwest China

AbstractSoil microbial community plays important roles in altering ecological processes and biogeochemical cycles in ecosystems. However, little is known about how afforestation influences the diversity, composition, and ecological network of soil microbial community in karst regions. In this study, soil samples were collected from one farmland (FL) and three afforestation lands including one bamboo forest (BA), one landscape tree planting forest (LAT), and one orange orchard (ORO) in a karst region of Southwest China. The bacterial and fungal communities were characterized using the high‐throughput sequencing approach, and soil properties including soil organic carbon, pH, soil water content, and soil total and available nutrient contents were measured under different land use treatments. Results showed that conversion from FL to BA and LAT significantly reduced the Shannon diversity of bacterial community. At the phylum level (top 10), genus level (top 30), and operational taxonomic units (OTUs) level, afforestation from FL resulted in significant changes in nine phyla, 24 genera, and 31.32% OTUs of bacterial community, and in three phyla, 13 genera, and 11.62% OTUs of fungal community. The number of nodes, number of negative edges, connectivity, average degree, and relative modularity of the microbial network under afforestation lands decreased by 9.33%–18.66%, 47.98%–72.75%, 0.45%–5.93%, 14.73%–22.29%, and 6.46%–23.50%, respectively, compared with FL. The soil organic carbon, total potassium and total phosphorus were identified as the key soil properties affecting the microbial community. Compared with LAT and ORO, the changes in bacterial and fungal communities under BA were more obvious because of the higher contents in soil organic carbon (13.48%) and total potassium (27.18%). In conclusion, afforestation significantly changed compositions and ecological network complexity of soil microbial community in karst regions, and conversion from FL to BA had stronger influences on the changes of soil microbial community than other afforestation lands in Southwest China. These findings provide the context necessary to evaluate the responses of soil microbial community to land use changes in karst regions.

Chronic enhanced nitrogen deposition and elevated precipitation jointly benefit soil microbial community in a temperate forest

Global change profoundly impacts carbon and nitrogen (N) cycling processes in terrestrial ecosystems by altering soil microbial communities. However, how enhanced N deposition and elevated precipitation jointly affect soil microbes and the underlying mechanisms remain poorly understood, particularly in forest ecosystems. In a long-term field experiment conducted in a temperate forest in China, we added N and/or water above the forest canopy to investigate their effects on soil microbial communities. Our results showed that N addition led to a decrease in bacterial biomass while increasing fungal biomass, resulting in an alteration of soil microbial community structure. Conversely, water addition did not significantly impact soil microbial communities. However, when N and water were added together, both fungal and bacterial biomass notably increased, leading to alterations in the soil microbial community structure. These results suggested that the effect of N addition could be mediated by elevated precipitation. Precipitation increment amplified the positive impact of N deposition on soil fungi but shifted its impact on bacteria from negative to positive. Additionally, soil bacteria were affected by soil N availability and acidification, as well as litter quality, whereas fungi were regulated by litter quantity and soil organic carbon. This study provides crucial evidence that concurrent enhanced N deposition and elevated precipitation favor soil microbial communities in temperate forests. It highlights the potential of elevated precipitation to alleviate the negative impacts of enhanced N deposition on temperate forest ecosystems, thereby sustaining ecological stability under global change scenarios.

Effects of sweet pepper straw biochar on soil microbial communities and growth of continuously cropped cucumber

PurposeThis study evaluates biochar from crop residues as a solution to soil degradation in continuous monoculture within greenhouse agriculture, focusing on its impact on soil microbial communities and cucumber plant growth.MethodsWe analyzed biochar derived from tomato straw (TSB), sweet pepper straw (SPSB), and eggplant straw (ESB), assessing their nutrient content, cation exchange capacity, and adsorption rates. This study examined the effects of three concentrations (2.5%, 5%, and 7.5% w/w) of the more promising SPSB on soil properties and cucumber growth.ResultsSPSB showed significantly higher levels of nitrogen, phosphorus, and potassium, along with superior adsorption capacity compared to TSB and ESB. The 5% w/w SPSB concentration notably improved cucumber growth, increasing plant height by 13.01%, stem thickness by 20.79%, leaf area by 50.26%, and dry weight by 58.56% relative to the control. High-throughput sequencing revealed this concentration significantly altered soil microbial community structure, enhancing bacterial and fungal diversity. It increased beneficial bacterial groups (Firmicutes, Actinobacteria, Bacillus) and modified fungal communities, with a decrease in Ascomycota and Aspergillus and shifts in Penicillium abundance. Functional genomic analysis indicated enrichment in bacterial metabolic pathways and fungal replication and expression genes.ConclusionSPSB, especially at a 5% w/w concentration, emerges as an effective soil amendment in greenhouses affected by continuous monoculture. This approach represents a sustainable method to enhance soil health and crop productivity.

Soil fungal community is more sensitive than bacterial community to modified materials application in saline-alkali land of Hetao Plain.

Soil salinization has become a major challenge that severely threatens crop growth and influences the productivity of agriculture. It is urgent to develop effective management measures to improve saline-alkali soil. Thus, in this study, soil properties, microbial communities, and function under desulfurization gypsum (DE), soil amendment (SA), farm manure (FA), and co-application of desulfurization gypsum, soil amendment, and farm manure (TA) in a field experiment were examined by high-throughput sequencing. The results showed that the application of modified materials is an effective approach in improving saline-alkali soil, especially TA treatment significantly increased the content of available phosphorus (AP), available potassium (AK), soil organic matter (SOM), and alkaline hydrolysis nitrogen (AHN) and decreased pH, bulk density (BD), and electrical conductivity (EC). The application of modified materials resulted in notable enhancement in fungal diversity and altered the composition and structure of the fungal community. Conversely, the effect on the bacterial community was comparatively minor, with changes limited to the structure of the community. Regarding the fungal community composition, Ascomycota, Mortierellomycota, and Basidiomycota emerged as the dominant phyla across all treatments. At each taxonomic level, the community composition exhibited significant variations in response to different modified materials, resulting in divergent soil quality. The TA treatment led to a decrease in Mortierellomycota and an increase in Ascomycota, potentially enhancing the ability to decompose organic matter and facilitate soil nutrient cycling. Additionally, the sensitivity of fungal biomarkers to modified materials surpassed that of the bacterial community. The impact of modified materials on soil microbial communities primarily stemmed from alterations in soil EC, AP, AK, and SOM. FUNGuild analysis indicated that the saprotroph trophic mode group was the dominant component, and the application of modified materials notably increased the symbiotroph group. PICRUSt analysis revealed that metabolism was the most prevalent functional module observed at pathway level 1. Overall, the application of modified materials led to a decrease in soil EC and an increase in nutrient levels, resulting in more significant alterations in the soil fungal community, but it did not dramatically change the soil bacterial community. Our study provides new insights into the application of modified materials in increasing soil nutrients and altering soil microbial communities and functions and provides a better approach for improving saline-alkali soil of Hetao Plain.

Drought legacy interacts with wildfire to alter soil microbial communities in a Mediterranean climate-type forest

Mediterranean forest ecosystems will be increasingly affected by hotter drought and more frequent and severe wildfire events in the future. However, little is known about the longer-term responses of these forests to multiple disturbances and the forests' capacity to maintain ecosystem function. This is particularly so for below-ground organisms, which have received less attention than those above-ground, despite their essential contributions to forest function. We investigated rhizosphere microbial communities in a resprouting Eucalyptus marginata forest, southwestern Australia, that had experienced a severe wildfire four years previously, and a hotter drought eight years previously. Our aim was to understand how microbial communities are affected over longer-term trajectories by hotter drought and wildfire, singularly, and in combination. Fungal and bacterial DNA was extracted from soil samples, amplified, and subjected to high throughput sequencing. Richness, diversity, composition, and putative functional groups were then examined. We found a monotonic decrease in fungal, but not bacterial, richness and diversity with increasing disturbance with the greatest changes resulting from the combination of drought and wildfire. Overall fungal and bacterial community composition reflected a stronger effect of fire than drought, but the combination of both produced the greatest number of indicator taxa for fungi, and a significant negative effect on the abundance of several fungal functional groups. Key mycorrhizal fungi, fungal saprotrophs and fungal pathogens were found at lower proportions in sites affected by drought plus wildfire. Wildfire had a positive effect on bacterial hydrogen and bacterial nitrogen recyclers. Fungal community composition was positively correlated with live tree height. These results suggest that microbial communities, in particular key fungal functional groups, are highly responsive to wildfire following drought. Thus, a legacy of past climate conditions such as hotter drought can be important for mediating the responses of soil microbial communities to subsequent disturbance like wildfire.

Increased precipitation alters the effects of nitrogen deposition on soil bacterial and fungal communities in a temperate forest

Anthropogenic nitrogen (N) deposition and increased precipitation are known to alter soil microbial communities. However, the combined effects of elevated N deposition and increased precipitation on soil microbial community dynamics and co-occurrence networks in temperate forests remain elusive. In this study, we conducted a field manipulation experiment by applying N solution and water to the forest canopy to simulate natural N deposition and increased precipitation in a temperate forest. We collected samples in the litter layer, organic soil layer, and mineral soil layer in 2018–2019 after 6–7 years of N and water treatments, and explored how elevated N deposition and increased precipitation regulate soil microbial diversity, community composition, and co-occurrence networks in different soil layers and at different sampling times. We found that the effects of N deposition and increased precipitation on soil microbial communities varied with soil layers and sampling times. Compared to the ambient environment, single canopy N addition (CN) or single canopy water addition (CW) did not affect bacterial Shannon diversity in the mineral soil layer in 2018, but the combined canopy N and water additions (CNW) decreased it in this layer at this time. CN increased fungal OTU richness in the organic and mineral soil layers in 2018; however, CW and CNW did not have an effect on it in the same layer at the same time. CW and CNW, but not CN, significantly affected bacterial and fungal community compositions in the litter layer in 2018 and in the organic soil layer in 2019. In contrast, CN, but not CW or CNW, significantly affected fungal community composition in the litter layer in 2019. CNW exhibited higher complexities of bacterial and fungal co-occurrence networks than CN and the ambient environment, indicating increased precipitation can strengthen the effect of N deposition on the complexity of bacterial and fungal co-occurrence networks. Our findings suggest that increased precipitation alters the effects of atmospheric N deposition on soil bacterial and fungal communities in this temperate forest, depending on soil layer and sampling time. Moreover, both bacterial and fungal community compositions are sensitive to increased precipitation, but the bacterial community composition is more sensitive to N deposition than the fungal community composition in the organic and mineral soil layers in this forest.

Chemical composition of soil carbon is governed by microbial diversity during understory fern removal in subtropical pine forests

Understory vegetation has an important impact on soil organic carbon (SOC) accumulation. However, little is known about how understory vegetation alters soil microbial community composition and how microbial diversity contributes to SOC chemical composition and persistence during subtropical forest restoration. In this study, removal treatments of an understory fern (Dicranopteris dichotoma) were carried out within pine (Pinus massoniana) plantations restored in different years in subtropical China. Soil microbial community composition and microbial diversity were measured using phospholipid fatty acids (PLFAs) biomarkers and high-throughput sequencing, respectively. The chemical composition of SOC was also measured via solid-state 13C nuclear magnetic resonance (13C NMR). Our results showed that fern removal decreased alkyl C by 4.2 % but increased O-alkyl C by 15.6 % on average, leading to a decline of alkyl C/O-alkyl C ratio, suggesting altered chemical composition of SOC and lowered SOC recalcitrance without fern. Fern removal significantly lowered the fungi-to-bacteria ratio, and it also reduced fungal and bacterial diversity. Partial correlation analysis revealed that soil nitrogen availability was a key factor influencing microbial diversity. Bacterial diversity showed a close relationship with the Alkyl C/O-alkyl C ratio following fern removal. Furthermore, the microbial community structure and bacterial diversity were responsible for 18 % and 55 % of the explained variance in the chemical composition of SOC, respectively. Taken together, these analyses jointly suggest that bacterial diversity exerts a greater role than microbial community structure in supporting SOC persistence during understory fern removal. Our study emphasizes the significance of understory ferns in supporting microbial abundance and diversity as a means of altering SOC persistence during subtropical forest restoration.

Land management of formerly subtropical Atlantic Forest reduces soil carbon stocks and alters microbial community structure and function

Soil carbon (C) sequestration has been proposed as one means to mitigate the effects of climate change and to guarantee food safety. However, forest conversion to managed land uses can deplete soil C stocks and affect soil microbial communities, potentially impacting a wide range of ecosystem services. Our objective was to investigate the effect of forest conversion to intensive land uses on soil C dynamics and soil microbial community structure and function in the subtropical portion of the Atlantic Forest biome. We evaluated four land uses - forest fragment, grassland, and agriculture with five or fifty years of cultivation. We analyzed soil C pools, edaphic variables, and soil bacterial/archaeal (16S) and fungal (ITS) communities. Forest conversion to managed land uses reduced soil C stocks and altered soil microbial community structure, with implications for soil C dynamics. Bacterial/archaeal and fungal community composition were strongly correlated with pH, base saturation, and calcium (Ca) and magnesium (Mg) content. However, soil C pools were not associated with potential extracellular enzyme activity or microbial diversity. Finally, the fungal species Mortierella minutissima ASV_1 and several bacterial groups were positively correlated with soil C stocks, potentially representing indicators of soil C storage. Our study indicates that land use change affects the interplay between soil microbial communities and soil C pools, with consequences to soil C storage. Our findings may support the development of soil C enhancement programs within the Atlantic Forest biome.

Application of Italian ryegrass residue changes the soil biochemical properties and bacterial communities in Southern China

Green manure application is a feasible agricultural practice to effectively improve soil fertility. However, our understanding of the effects and mechanisms of different green manure residue amounts on soil fertility improvement remains insufficient. Regarding a new type of Italian ryegrass (Lolium multiflorum L.)-rice (Oryza sativa L.) rotation (IRR) system in Southern China, the effects of six Italian ryegrass residue amounts (IRR0: 15,700 kg·ha−1, IRR1: 30,700 kg·ha−1, IRR2: 35,400 kg·ha−1, IRR3: 40,100 kg·ha−1, IRR4: 44,800 kg·ha−1, IRR5: 49,500 kg·ha−1) on the soil nutrient contents, soil enzyme activities, and bacterial communities were evaluated. Compared to the CK, appropriate amounts of Italian ryegrass residue application increased total nitrogen (36.45–37.38 %), soil organic carbon (15.34–26.55 %), microbial biomass carbon (14.07–33.93 %) and microbial biomass nitrogen (30.20–47.65 %) (P < 0.05). The activities of soil polyphenol oxidase, urease, β-glucosidase, peroxidase, cellulase, and total enzyme were also improved significantly in comparison with the CK (P < 0.05). Bacterial community structures in response to IRR were different from those in the CK. The application of Italian ryegrass residue significantly enhanced the relative abundance of beneficial taxa, including Acinetobacter, Pseudomonas, and Arenimonas (P < 0.05). The 30,700–35,400 kg·ha−1 Italian ryegrass residue exhibited the most practical amounts for enhancing soil fertility, soil enzyme activities, and altering soil microbial communities, suggesting that the improvement effects of green manure application on soil were dependent on the residue returning amounts. These findings suggest that the returning of Italian ryegrass residue is an effective land management measure to realize efficient and sustainable utilization of paddy fields.

Interactions of long-term grazing and woody encroachment can shift soil biogeochemistry and microbiomes in savanna ecosystems

Semi-arid grasslands and savannas in the southern Great Plains USA are extensively used for livestock grazing. Over the past century, Juniperus (juniper) and Quercus (oak) species abundance have increased due to intensive grazing and reduced fire frequency. We investigated the interactions between livestock grazing history (none, moderate, heavy) and vegetation cover (grassland, juniper, oak) using a ∼ 70-year grazing experiment in west-central Texas. We explored effects on soil organic carbon (SOC), total nitrogen (TN), total phosphorus (TP), microbial community composition, and function. SOC and TN were 50–150 % higher under juniper and oak compared to grasslands, and 10–30 % lower in grazed vs. ungrazed areas. Vegetation × grazing interaction showed greater SOC and TN loss under oak than juniper or grasslands. Ungrazed controls had higher soil TP than grazed areas, with oak and juniper soils having more TP than grasslands. Bacterial and fungal communities differed between grassland and woody vegetation. Grazing affected only bacterial communities. SOC, TN, TP accounted for differences in community structure. Abundances of genes related to methane, nitrogen, sulfur metabolisms, and dominant fungal trophic modes were linked to soil C, N, P ratios. These findings highlight how long-term livestock grazing and woody plant encroachment influence soil C, N, P cycles, altering soil microbial community structure and function. This study provides insights for savanna ecosystem management and integrating land cover effects into biogeochemical models for global change scenarios.

Direct and indirect effects of Basta®, a glufosinate-based herbicide, on banana plantation soil microbial diversity and function

Herbicide applications have doubled over the past 30 years to control weeds, but their use may influence soil microbial communities, which mediate important ecosystem services. Here, we characterised the direct effects of a single application of Basta® (glufosinate), at 1× and 2× the recommended rate (5 and 10 kg product ha−1), as well as the indirect effects of vegetation loss on the diversity and function of soil microbial communities within a banana plantation. A 10-month pre-experimental period using Basta® to remove additional vegetation from half of the experimental area created conditions for assessing the indirect herbicide effects. Direct assessments on soil microbial communities began when the Basta® herbicide treatments were applied to the areas with and without vegetation and continued over multiple time-points, for 56 days. Herbicide treatment had no significant direct impacts on basal, and substrate induced respiration rates, or the potential activities of microbial enzymes as inferred from fluorescein diacetate hydrolysis (FDA) and β-glucosidase assays. Similarly, phylogenetic marker gene sequencing indicated that Basta® application did not significantly influence the diversity of soil bacterial or fungal communities. Indirectly, Basta® had a greater influence on the soil microbial activity and functions by removing understory vegetation cover around banana plants. This suggested that continual use of herbicides to reduce soil vegetation cover under bananas had a greater impact on soil microbial communities than a single application of the herbicide. Furthermore, the presence or absence of vegetation cover, significantly altered the abundance of Fusarium oxysporum, plus an additional eight bacterial and ten fungal taxa. Our results indicated that a single application of glufosinate as Basta® at the recommended or double the recommended rate, did not adversely affect soil microbial communities or their activities in banana plantations directly. However, application of herbicides, such as Basta® in crops like banana, indirectly alters soil microbial communities and their activities through loss of vegetation cover and should only be used as a component of an integrated weed management system.
