Optimization of Pressurized Liquid Extraction (PLE) Parameters for Extraction of Bioactive Compounds from Moringa oleifera Leaves and Bioactivity Assessment.

Moringa oleifera leaves are rich sources of bioactive compounds with potential health benefits, including antioxidants and anti-inflammatory agents. Pressurized liquid extraction (PLE) stands out as a promising technique for effectively extracting valuable compounds from natural sources. In this study, we aimed to optimize PLE parameters, such as temperature, extraction duration, and pressure, to maximize bioactive compound (polyphenols, flavonoids, and ascorbic acid) yield from M. oleifera leaves and evaluate their antioxidant and anti-inflammatory activities. According to the outcomes of this research, the maximum achieved total polyphenol content was 24.10 mg gallic acid equivalents (GAE)/g of dry weight (dw), and the total flavonoid content was increased up to 19.89 mg rutin equivalents (RtE)/g dw. Moreover, after HPLC-DAD analysis, neochlorogenic and chlorogenic acids, catechin and epicatechin, rutin, and narirutin were identified and quantified. As far as the optimum ascorbic acid content is concerned, it was found to be 4.77 mg/g dw. The antioxidant activity was evaluated by three different methods: ferric reducing antioxidant power (FRAP), the DPPH method, and the anti-hydrogen peroxide activity (AHPA) method, resulting in 124.29 μmol ascorbic acid equivalent (AAE)/g dw, 131.28 μmol AAE/g dw, and 229.38 μmol AAE/g dw values, respectively. Lastly, the albumin denaturation inhibition was found to be 37.54%. These findings underscore the potential of PLE as an efficient extraction method for preparing extracts from M. oleifera leaves with the maximum content of bioactive compounds.

Utilization of diamondback puffer (Lagocephalus guentheri) biomass for the production of bioactive oligopeptides and their inflammation suppressing effects invitro

Development of new anti-inflammatory drugs has always been of prime interest due to various side effects exerted by common non-steroidal anti-inflammatory drugs (NSAIDs). Hence, due to the biochemical diversity and efficient anti-inflammatory activities of bioactive peptides from oceanic sources, their demand is increasing. In this study, the common diamondback puffer fish (Lagocephalus guentheri) muscle (PM) and visceral (PV) mass waste was used to produce enzymatic hydrolysates using trypsin, alcalase and papain digestion which were further used to obtain inflammation suppressing peptides. High pressure liquid chromatography (HPLC) analysis showed abundant essential amino acids like arginine, alanine, tyrosine, and glutamic acid in muscle as well as visceral part. The data obtained by the anti-inflammatory assays such as albumin denaturation inhibition and membrane stabilization assay of hydrolysates showed that alcalase 12th hr and 6th hr showed highest activity for PM and PV respectively. Further, out of the fractions obtained through ultrafiltration, 10-3 kDa showed higher anti-inflammatory activity which was subsequently purified with gel filtration chromatography. Moreover, the purified active peaks were analysed using LC-MS/MS to acquire a peptide sequence of MEPLGQG (731 Da) and LLHA (453 Da) for PMpep and PVpep respectively. In vitro studies revealed non-cytotoxic effects of peptide and pro-inflammatory cytokine suppression post LPS-stimulation in RAW264.7 cells. Thus, the puffer fish and other marine waste can be exploited for the isolation of potential nutraceutical and therapeutic compounds.

Pigments, phenolics, and bioactivities of various extracts from flowers of Camellia quephongensis

Camellia quephongensis Hakoda et Ninh, a plant species in the Theaceae family, is native to Vietnam. In this study, the effects of various solvents used for extracting Camellia quephongensis flowers on phenolic content, antioxidative, and in vitro anti-inflammatory activities were assessed. The highest total phenolic and flavonoid contents and antioxidant properties were observed for the ethyl acetate extract. Catechin and epicatechin in the ethyl acetate extract showed the highest concentrations (17.23 ? 0.57 mg/g and 3.86 ? 0.42 mg/g, respectively) whereas epigallocatechin gallate had the highest concentration in methanol extract (10.66 ? 0.33 mg/g). Most of the other phenolics were also found at greater concentrations in the ethyl acetate extract in comparison to the other extracts. The results also demonstrated that ethyl acetate displayed the strongest antioxidant potential assessed by ABTS and DPPH radical scavenging assays. The albumin denaturation inhibition assay showed that the ethyl acetate extract had the greatest efficacy, followed by the ethanol and methanol extracts. Chlorophyll and carotenoids were found in smaller amounts compared to those in other Camellia species' leaves. These findings provide deeper insights into obtaining extracts with high levels of bioactive compounds, antioxidative potential, and anti-inflammatory effects from C. quephongensis.

Fatty acids characterisation by GC-MS, antiglycation effect at multiple stages and protection of erythrocytes cells from oxidative damage induced by glycation of albumin of Opuntia ficus-indica (L.) Mill seed oil cultivated in Eastern Morocco: Experimental and computational approaches

Ethnopharmacological relevanceOpuntia ficus-indica (L.) Mill is frequently observed in the Moroccan traditional medicinal system, where these approaches are employed to mitigate the onset of diabetes and the subsequent complications it may entail. Aim of the studyThe aim of this research was to examine the effectiveness of Opuntia ficus-indica seed oil in preventing diabetic complications. Specifically, the study assessed its ability to counteract glycation at various stages, protected red blood cells from the harmful effects of glycated albumin, and inhibited pancreatic lipase digestive enzymes to understand its potential antihyperglycemic properties. Additionally, the study aimed to identify the chemical components responsible for these effects, evaluate antioxidant and anti-inflammatory properties, and conduct computational investigations such as molecular docking. Materials and methodsThe assessement of Opuntia ficus-indica seed oil antiglycation properties involved co-incubating the extract oil with a bovine serum albumin-glucose glycation model. The study investigated various stages of glycation, incorporating fructosamine (inceptive stage), protein carbonyls (intermediate stage), and AGEs (late stage). Additionally, measurement of β-amyloid aggregation of albumin was performed using Congo red, which is specific to amyloid structures. Additionally, the evaluation of oil's safeguarding effect on erythrocytes against toxicity induced by glycated albumin included the measurement of erythrocyte hemolysis, lipid peroxidation, reduced glutathione. The fatty acid of Opuntia ficus-indica seed oil were analyzed using Gas Chromatography-Mass Spectrometry (GC–MS). The in vitro evaluation of antihyperglycemic activity involved the use of pancreatic lipase enzyme, while the assessement of antioxidant capability was carried out through the utilization of the ABTS and FRAP methods. The in vitro assessement of the denaturation of albumin activity was also conducted. In conjunction with the experimental outcomes, computational investigations were undertaken, specifically employing ADMET (absorption, distribution, metabolism, excretion, and toxicity) analysis. Furthermore, molecular docking was utilized to predict antioxidant and antiglycation mechanisms based on protein targets. ResultsIn vitro glycation assays, Opuntia ficus-indica seed oil displayed targeted inhibitory effects at multiple distinct stages. Within erythrocytes, in addition to mitigating hemolysis and lipid peroxidation induced by glycated albumin. GC-MS investigation revealed a richness of fatty acids and the most abundant compounds are Linoleic acid (36.59%), Palmitic acid (20.84%) and Oleic acid (19.33%) respectively. The findings of antioxidant ability showed a remarkable activity on FRAP and ABTS radicals. This oil showed a pronounced inhibitory impact (p < 0.001) on pancreatic lipase enzyme. It also exerted a notibale inhibition of albumin denaturation, in vitro. ConclusionThe identified results were supported by the abundant compounds of fatty acids unveiled through GC-MS analysis, along with the computational investigation and molecular docking.

Immunomodulatory and anti-inflammatory activities of hydro-ethanolic extract of Securidaca longipedunculata Fresen leaves

Introduction: The use of Securidaca longipedunculata for solving health problems related to immunological disorders is long-standing. The immunomodulatory activity associated with the anti-inflammatory effect of S. longipedunculata has not yet been elucidated by scientific research. The aim of our study is to show that the leaves of S. longipedunculata can solve problems related to immunodeficiency and inflammation. Methods: The phytochemical compounds of the plant were carried out by solubility tests. The anti-inflammatory activity of the plant was evaluated by the chicken egg albumin denaturation inhibition, membrane stabilization, and C-reactive protein (CRP) tests. The 2,2-diphenyl1-picrylhydrazyl, total antioxidant capacity, and inhibition of lipoperoxidation tests were performed to evaluate the antioxidant activity of the plant. Immunosuppression was induced in rats by cyclophosphamide. The immunomodulatory activity of S. longipedunculata was studied by blood count. The lactate dehydrogenase (LDH) titration showed the effect of S. longipedunculata on the energy balance. Results: The extract contained polyphenols, flavonoids, and tannins. The tests of albumin denaturation inhibition, membrane stabilisation, CRP, total antioxidant capacity, and 2,2-diphenyl-1-picrylhydrazyl revealed that the extract had anti-inflammatory and antioxidant effects. The blood count results revealed that the extract non-significantly increased the number of leukocytes except in the case of neutrophils and monocytes at a dose of 400 mg/kg (P&lt;0.05). The extract also restored the energy balance in the rats according to the LDH results. Conclusion: This study reveals that the hydro-ethanolic extract of S. longipedunculata leaves has potential immunostimulant and anti-inflammatory effects.

Comparative Analysis of Pigments, Phenolics, and Bioactivities of Three Camellia Species Growing in Vietnam

Objective/Background: Camellia L. , belonging to the Theaceae family, is a diverse group of plant species native to various regions of Asia. Methods: Chlorophyll and carotenoid contents in leaves of three Camellia species, namely Camellia quephongensis, Camellia yokdonensis, and Camellia longii, were determined using a spectrophotometric method. Phenolic compounds of the leaf extracts were quantified using high-performance liquid chromatography. Additionally, antioxidant activity, anti-α-amylase and in vitro antiinflammatory properties of the extracts were evaluated. Results: The total carotenoid content in C. longii was the highest, whereas the total chlorophyll content in this species was the lowest. Most of the phenolic compounds in C. quephongensis were found at the greatest concentrations compared to the other species. The extract from C. quephongensis exhibited the strongest antioxidant properties estimated by ABTS and DPPH free radical scavenging assays. This activity was shown to be comparable with that of ascorbic acid. All the Camellia extracts may possess a much less potent anti-α-amylase activity in comparison with acarbose. They could also be weaker than diclofenac with respect to in vitro anti-inflammatory activity evaluated by albumin denaturation inhibition assay. Conclusions: The findings of this investigation provide the initial insights into pigments and phenolics present in the various Camellia species and pave the way for the development of functional foods that could potentially offer a range of health benefits.

Anti-inflammatory, anti-diabetic, and biocompatibility properties of aqueous extract of Tamarindus indica L. fruit coat analyses by in-vitro and in-vivo approaches

The anti-inflammatory, anti-diabetic, and biocompatibility nature of Tamarindus indica L. fruit coat aqueous extract were investigated in this research through in-vitro and in-vivo studies. The anti-inflammatory property was determined through albumin denaturation inhibition and antiprotease activities as up to 39.5% and 41.2% respectively at 30 mg mL−1 concentration. Furthermore, the antidiabetic activity was determined through α-amylase and α-glucosidase inhibition as up to 62.15% and 67.35% respectively at 30 mg mL−1 dosage. The albino mice based acute toxicity study was performed by different treatment groups (group I–V) with different dosages of aqueous extract to detect the biocompatibility of sample. Surprisingly, findings revealed that the T. indica L. fruit coat aqueous extract had no harmful impacts on any of the groups. Urine, as well as serum parameter analysis, confirmed this. Moreover, the findings of SOD (Superoxide Dismutase), GST (Glutathione-S-transferase), & CAT (Catalase) as well as glutathione peroxidase as well as reduced glutathione antioxidant enzymes studies stated that the aqueous extract possess high antioxidant ability via a dose-dependent way. These findings indicate that T. indica fruit coat aqueous extract contains medicinally important phytochemicals with anti-inflammatory and anti-diabetic properties, as well as being biocompatible in nature.

Evaluation of the Anti-Inflammatory and Antioxidant, Activities of Parinari Curatellifolia Planch. Ex Benth (Chrysobalanaceae) Methanol Stem Bark Extract

It is used as human food and beverage, and in traditional medicine it is used to treat pneumonia and fever. The aim of this was to investigate its anti-inflammatory and antioxidant activities of Parinari curatellifolia methanol stem bark extract. A 2kg of the dried and powdered plant material was extracted with 80% methanol through cold maceration to obtain the crude methanol extract. The crude methanol extract was sequentially fractionated using n-hexane, ethyl acetate and methanol to yield n-hexane, ethyl acetate and methanol fractions respectively. Acute toxicity study was conducted following the OECD guidelines. The anti-inflammatory activity of the fractions was evaluated was evaluated using inhibition of albumin denaturation, inhibition of egg albumin-induced paw edema in rats and phospholipase A2 (PLA2) inhibition assay models. In vitro antioxidant activity was evaluated using DPPH de-colourizing, Nitric oxide scavenging activity and Ferric Reducing Antioxidant Power (FRAP) assays. Oral acute administration of the extract revealed an LD50 dose &gt; 5000 mg/kg in rats. The extract caused a dose- and time-dependent inhibition of rat paw edema, and a concentration-dependent inhibition of Phospholipase A2 activity. IC50 of the extract for Albumin Denaturation inhibition was 37.22 ug/ml. In the antioxidant assays the IC50 of the extract against DPPH was 25.91 ug/ml, while the Nitric oxide Equivalence (mM)/mg was 42.38.

A facile green synthesis of Ag@MnO2 nanoparticles using Martynia annua plant extract and their biological activity and catalytic reduction of dye

The Ag@MnO2 NPs were characterized by UV, FTIR, HRTEM, SAED, and EDX analysis. UV analysis revealed the characteristic wavelength absorption maxima at 425 nm, which confirmed the synthesis of Ag@MnO2 NPs. Martynia annua plant extract presented phytochemical compounds that act as reducing and capping agents in the synthesis of Ag@MnO2 NPs, as confirmed by FT-IR analysis. HRTEM analysis to determine the Ag@MnO2 NPs morphology images showed the spherical shape of NPs with sizes from 1 to 8 nm. The crystalline nature of Ag@MnO2 NPs was identified using SAED and EDX analysis revealed the composition of these synthesized NPs. In-vitro antioxidant activities of Ag@MnO2 NPs were evaluated using a DPPH assay for the significant radical scavenging ability of these NPs. Our findings revealed that M.annua plant-mediated Ag@MnO2 NPs have been highly active against both bacterial and fungal strains. The BSA and EA methods for inhibition of albumin denaturation were investigated for their anti-inflammatory activity. The Ag@MnO2 NPs showed superior activity as anti-inflammatory agents, reduced the viability of the human lung cancer cell line A549 up to 9.92 % at a concentration of 500 µg/ml, and exhibited a good cytotoxic effect. In the presence of NaBH4, the Ag@MnO2 NPs showed excellent catalytic degradation activity against Fuchsin basic dye.

Evaluation of Gossypium herbaceum Leaf Powder's Nutritional Composition and Nutraceutical Properties

The objective of this study is to identify the proximate composition, phytochemical profile, and anti-diabetic, anti-inflammatory and antioxidant properties of Gossypium herbaceum leaf powder (GLP). The fresh leaves of the G. herbaceum were collected, cleansed with fresh water, drained and allowed to dry in the shade, ground to GLP and analysed. The crude fibre (42.93%) and nitrogen-free extract (36.46 %) have a relatively high proportion in GLP; while ash (2.47%) has the lowest proportion. The GLP has relatively high phenol (219.20 mg/g) when compared to flavonoids (81.03 mg/g), tannins (69.56 mg/g), saponins (66.67 mg/g) and alkaloids (55.80 mg/g). The α-amylase inhibition and α-glucosidase inhibition of GLP were 48.45% and 30.68%, respectively. The percentage of albumin denaturation inhibition and anti-proteinase activity of GLP was 22.88% and 43.87%, respectively. The lipid peroxidation inhibition, vitamin C, Fe chelation and 2,2-diphenyl-1-picrylhydrazyl were 35.43%, 23.87%, 11.76% and 88.16%, respectively. GLP exhibits anti-inflammatory, anti-diabetic and antioxidant properties.

In vitro Anti-Inflammatory and Antioxidant Activity of Seed Ethanolic Extract of Pongamia pinnata

Introduction: Pongamia pinnata Merr., a member of the Fabaceae family, is extensively spread throughout tropical Asia. Historically, several parts of P. pinnata have been utilised in the indigenous medicinal systems of several cultures. Anti-inflammatory and antioxidant properties are basic requisites in treating any oral mucosal lesions. This study aims to investigate the antioxidant and anti-inflammatory properties of these leaf extracts. Materials and methods: pinnata seeds were collected in Chennai, Tamil Nadu, India. For the extraction, shade-dried P. pinnata plant seeds were utilized. P. pinnata seed ethanolic extract was analysed qualitatively for the content of phenols, tannins, saponins, proteins, and acids, among other phytochemicals. Using the DPPH test and suppression of albumin denaturation, the anti-oxidant and anti-inflammatory activity of P. pinnata seed extract was determined. Results: The phytochemical analysis of P. pinnata seed extract revealed the presence of phenols, tannins, saponins, proteins, and acids. The anti-oxidant and anti-inflammatory activity of P. pinnata seed extract was considerably higher at 500 g, as shown by the DPPH test and albumin denaturation inhibition. Conclusion: The results of this study show that P. pinnata seed extracts have both anti-inflammatory and antioxidant properties. Considering the results, P.pinnata shows high potential for management of oral mucosa lesions. Further clinical research needs to be done to analyse the effect of these properties.

Evaluation of phytochemical, antibacterial, thrombolytic, anti-inflammatory, and cytotoxicity profile of Achyranthes aspera aerial part extracts

This investigation was designed and performed to compare the phytochemical profiling, activities of antibacterial, thrombolytic, anti-inflammatory, and cytotoxicity of methanol extract (ME-E) and aqueous extract (AQ-E) of aerial parts of Achyranthes aspera through in-vitro approach. Also characterize the functional groups of bioactive compounds in the ME-E through Fourier-transform infrared (FTIR) spectroscopy analysis. Interestingly, qualitative phytochemical screening proved that the ME-E contain more number of vital phytochemicals such as phenolics. saponins, tannins, alkaloids, flavonoids, cardiac glycosides, steroids, and phlobatannins than AQ-E. Similarly, the ME-E showed notable antibacterial activity as dose dependent manner against Bacillus subtilis, Escherichia coli, Staphylococcus aureus, Klebsiella pneumoniae, and Pseudomonas aeruginosa at 1000 μg mL−1 concentration. ME-E also showed 75.2 ± 2% of clot lysis (thrombolytic activity) at 1000 μg mL−1 dosage and it followed by AQ-E 51.24 ± 3%. The ME-E showed moderate and AQ-E demonstrate poor anti-inflammatory activity evidenced by albumin denaturation inhibition and anti-lipoxygenase assays. Furthermore, the ME-E demonstrated a dose dependent cytotoxicity was noted against brine shrimp larvae. In support of this ME-E considerable activities, the Fourier transform infrared (FTIR) analysis confirmed that this extract contain more number peaks attributed to the stretch of various essential functional groups belongs to different bioactive compounds. Hence this ME-E of A. aspera can be considered for further in depth scientific investigations to validate their maximum biomedical potential.

Anti-proliferative and anti-inflammatory activity of triterpene extracts from plant species belonging to Lamiaceae family

Triterpenes are very important secondary metabolites with wide structural diversity and significant role in pharmacy and medicine.In the present research, a comparative study of pharamacological activities of the triterpene fractions obtained from several plant species belonging to Lamiaceae family, was carried out. In-vitro anti-proliferative activity was performed using a standardproliferation assay based on tetrazolium salts. In vitro anti-inflammatory activity of triterpene fractions was determined by an assay of inhibition of albumin denaturation. In general, the triterpene fractions obtained from plant species belonging to Lamiaceae family showed a strong anti-proliferative activity and anti-inflammatory activity.The triterpene fraction of Rosmarini folium showed the strongest anti-proliferative activity (GI50range from 4 to 37 μg/ml) and the strongest anti-inflammatory activity in the range from 57.27% to 80.69%. This comparative study provides scientific evidence to support the traditional use of Lamiacae plant species for medical purposes as anti-inflammatory and anti-proliferative medicines.

Assessment of multi-biomedical efficiency of Andrographis paniculata shoot extracts through in-vitro analysis and major compound identification

The present investigation looked into the various biomedical potentials of Andrographis paniculata shoot extracts. The results showed that the methanol extract (Met-E) of A. paniculata contains more phytochemicals than the acetone and petroleum ether extracts, including alkaloids, saponins, tannins, phenolics, flavonoids, glycosides, terpenoids, phytosterol, steroids, and protein. Accordingly, the Met-E alone showed considerable bactericidal activity (through agar well diffusion method) against the bacterial pathogens namely Shigella dysenteriae, Bacillus cereus, Salmonella typhi, Enterococcus faecalis, Pseudomonas aeruginosa, Klebsiella pneumoniae, Staphlococcus aureus, E. coli, and B. subtilis. This bactericidal activity was found as dose dependent manner, since at 1000 μg ml concentration, the Met-E showed better antibacterial activity. Similarly, at increased concentration (1000 μg ml) it showed notable antidiabetic (α-amylase inhibition: 74.31% and α-glucosidase inhibition: 72.34%), antioxidant (DPPH: 78.24%), and anti-inflammatory (albumin denaturation inhibition: 79.84% and lipoxigenase inhibition: 69.4%) activities. The phytochemical profiling of Met-E was characterized by UV–visible spectrophotometer (UV–vis), Gas Chromatography–Mass Spectrometry (GC/MS), Fourier transform infrared (FTIR), and High Performance Liquid Chromatography (HPLC) analyses. The results showed the Met-E contain bioactive compounds such as gallic acid, epicatechin, catechin, naringin, vitexin-2-rhamnoside, taxifolin, kaempferol, hesperidin, myricetin, rutin, quercetin, phloretin, and ursolic acid compounds. While most of these substances have been recognised for their pharmacological application perspective, the biological properties of particular substances must be studied in the future using in-vivo strategies.

Assessment of antidiabetic, anti-inflammatory, antioxidant and anticancer activity competence of methonolic extracts of Trianthema ortulacastrum and Andrographis paniculata

An in-vitro investigation was performed to evaluate and compare the phytochemical, antioxidant, antidiabetic, anti-inflammatory, and anti-lung cancer activities of methanol extracts of aerial parts of Andrographis paniculata and Trianthema portulacastrum. Furthermore studied major functional groups of phytochemicals present in the methanol extracts of these plants through Fourier transform infrared (FTIR) analysis. The results showed that the methanol extract of A. paniculata contain more number of pharmaceutically valuable phytochemicals such as alkaloids, flavonoids, terpenoids, saponin, glycoside, phytosterol, and tannin than T. portulacastrum. Similar way the methanol extract of A. paniculata showed considerable dose dependent antioxidant (DPPH: 63%), antidiabetic (α-amylase: 82.31% and α-glucosidase inhibitions: 72.34%), and anti-inflammatory (albumin-denaturation inhibition: 76.3% and anti-lipoxygenase: 61.2%) activities (at 900 μg mL−1 concentration) than T. portulacastrum. However, the anti-lung cancer activities of these test plants against A549 cells were not considerable. According to FTIR analysis, the A. paniculata methanol extract has a larger number of characteristic peaks attributed to the active functional groups of pharmaceutically valuable bioactive components that belong to different types of phytochemicals. These findings imply that A. paniculata methanol extracts can be used for additional research, such as bioactive compound screening and purification, as well as assessing their potential biomedical uses in various in-vitro and in-research settings.

IN-VITRO ANTI-INFLAMMATORY EVALUATION OF METHANOL AND BUTANOL EXTRACTS OF AVOACADO PEAR (PERSEA AMERICANA MILL) FRUIT PEEL

Inflammatory diseases such as arthritis, rheumatoid or non-rheumatoid joint inflammations are dominant among people all around the world. Plant-derived anti-inflammatory drugs are widely used against these disease conditions. This study investigated the in-vitro anti-inflammatory activity of the aqueous methanol and butanol extracts of Persea americana fruit peel. Three different concentrations of test samples: 10, 20 and 40 mg/ml from the concentrated methanol extract (ME) and butanol extracts (BE) were evaluated in vitro by albumin denaturation, heat induced hemolysis, anti-proteinase action, anti-lipooxygenase spectrophotometric assay methods using aspirin as a reference drug. Results showed that at sample concentrations of 10, 20 and 40 mg/ml, the inhibition of albumin denaturation by the ME and BE were in the range 26.826-51.471 % and 40.329- 54.797 % respectively. The inhibition of heat-induced haemolysis by ME at the same concentrations was in the range 30.252-44.727% and 34.52-49.897% for BE. Anti-proteinase inhibitory activity for ME was within the range of 17.073-52.1 20% while 31 .781-37.01 4% was for BE. In all four assays, the BE elicited a relatively higher inhibitory activity than the ME. These suggest that the fruit extract possesses anti-inflammatory properties which may be attributed to the occurrence of bioactive compounds in the fruit peel.

Phytochemical screening, Antiradical and Anti-inflammatory activity of aqueous bark extract acquired from Acacia nilotica

Herbal medicaments are applied in the healing for numerous human illnesses from the sprouting of human civilization. The chemicals existed naturally within the plants displays antiradical features which are the principal agent for the neutralization of reactive oxygen species created within the living entities. Acacia nilotica from the Fabaceae family is a tree with magnificent therapeutic properties. Various sections of this tree are implemented in the traditional healing system. Specifically, the bark are applied to treat numerous health complaints. Here, owned to the medicinal features of bark of A.nilotica in the traditional practices, we prepared the aqueous bark extract from this tree and evaluated the antioxidant (antiradical) and anti-inflammatory activities. Hydroxyl radical scavenging protocol presented dose-based upsurge in the radical inhibition activity of the bark extract with maximum activity at 60 µg/ml and the IC50 value appeared as 43.35µg/ml. ABTS radical scavenging assay executed also displayed dose-lineated upsurge in the radical scavenging property of the tested bark extract with IC50 value as 41.07µg/ml. In both the radical scavenging assays, vitamin C was implemented for reference purpose. Even, the anti-inflammatory nature of the aqueous bark extract was performed applying egg albumin denaturation inhibition protocol. In a dose-lined way, the bark extract presented anti-inflammatory activity with an IC50 calculated value as 73.62µg/ml with diclofenac sodium as reference medicament. Additionally, the phytochemical qualitative scrutiny presented the existence of steroids, saponins, terpenoids, alkaloids, tannins, anthraquinones, carbohydrates, phenols and flavonoids which were chiefly responsible for the pharmacological activities of bark aqueous extract.

Membrane Stabilization, PhospholipaseA2, Albumin Denaturation, Protease Inhibition, as Viable Mechanisms for the Anti-Inflammatory Effects of Methanol Extract of Rauvolfia vomitoria Afzel Leaves

Background: The body uses inflammation as a defence mechanism to eliminate harmful stimuli like germs, damaged cells, or irritants and to initiate the healing process. However, the ongoing discovery of several medicinal plants and the testing of their bioactivity to produce information that will assist doctors and patients in making informed decisions prior to employing them have established.&#x0D; Aims: The study valuate the phosphatelipase A2, membrane stabilization, albumin denaturation, protease inhibition, and platelete aggregation activities as viable mechanisms for the anti-inflammatory effects of the methanol extract of Rauvolfia vomitoria Afzel leaf (MERVL).&#x0D; Methodology: The anti-inflammatory effect of (MERVL) was ascertained using the phosphate lipaseA2, membrane stabilization model, albumin denaturation, protease inhibitor, assay.&#x0D; Place and Duration of the Study: Department of Pharmacology Lab, Enugu State of University of Science and Technology Agbani Nigeria, between March 2021 and August 2021.&#x0D; Results: MERVL inhibited hypotonicity-induced haemolysis by 27.14, 41.10 and 65.70%, at the concentration of 0.4, 0.8 and, 1.0 mg/mL respectively.The highest percentage of inhibition (67.70%) was noticed at the highest concentration of the MERVL. These results were almost analogous to the standard drug (indometacin) used as it exhibited concentration dependent inhibition of albumin denaturation. Protease activity was significantly (P &lt; 0.05) increased at all concentrations which follow the similar tendency as standard drug used. The results showed that MERVL has anti-inflammatory activities.

Determination of Phenolics and Biological Activity of Viola dalatensis Gagnep. by High-Performance Liquid Chromatography (HPLC) and Multivariate Analysis

Viola dalatensis, belonging to the Violaceae family, is an endemic species to Vietnam. The present study aimed to determine phenolic compounds in extracts from aerial parts of the plant species using high performance liquid chromatography with diode-array detection (HPLC-DAD). In addition, free radical scavenging and bovine albumin denaturation inhibition assays were performed to evaluate the antioxidant and in vitro anti-inflammatory activities of the extracts. Most of the phenolic compounds in the methanolic extract were detected at the greatest concentrations compared to the other extracts, which was also supported by the highest total phenolic content (26.70 ± 0.1059 mg GAE/g). In general, the antioxidant activities of the methanolic and ethanolic extracts were found to be stronger than those of the other extracts. The aqueous extract exhibited the most potent inhibitory effect on albumin denaturation (IC50 = 90.39 ± 8.415 µg/mL), comparable with that of diclofenac. Through principal component analysis, 88.60% of the total variance in the phenolic dataset was explained. The correlation analysis showed that chlorogenic acid, ferulic acid, epigallocatechin gallate, rutin, and kaempferol may play important roles in antioxidant activity while gallic acid could contribute to in vitro anti-inflammatory activity of the extracts. The findings of this study provide the first evidence of the bioactive compounds present in V. dalatensis and shed light on the potential health-promoting properties linked to its aerial parts.

Qualitative Phytochemical Screening, Evaluation of Antioxidant and Anti-Inflammatory Potentials of Vitex pinnata L. Leaf Extract

The purpose of the study was to screen phytochemicals and assess the antioxidant as well as anti-inflammatory properties of Vitex pinnata leaf extracts. Alkaloids, glycosides, tannins, terpenoids, saponins, phlobatannins, steroids, quinine, phenol, coumarins, and flavonoids were found during the phytochemical investigation. V. pinnata leaf extract exhibited an IC50 value of 154.33 µg/ml in the DPPH free radical scavenging assay (standard: IC50 = 25.48 µg/ml) IC50 value of 218.8 µg/ml in the albumin denaturation inhibition assay (standard: IC50 = 33.74 µg/ml). According to the findings, the leaves of V. pinnata contain a variety of pharmaceutically important secondary metabolites capable of scavenging DPPH and inhibiting albumin denaturation, demonstrating their antioxidant and anti-inflammatory properties.
