Cold Exposure Affects Lipid Metabolism, Fatty Acids Composition and Transcription in Pig Skeletal Muscle.

Cold exposure promotes glucose oxidation and modulates the lipid metabolism in adipose tissue, but it is still not fully clear whether cold exposure could affect meat quality and fatty acid metabolism in skeletal muscle of pig in vivo. Here, we kept finishing pigs under cold or room temperature overnight and determined the effects of cold exposure on meat quality, fatty acids composition and transcriptional changes in skeletal muscle of pigs. We found that cold exposure significantly reduced the meat colour24 h and pH24 h, without affecting carcass characteristics and other meat quality traits. Considerable changes were found in the proportions of individual fatty acids and the total content of saturated fatty acid, polyunsaturated fatty acids, monounsaturated fatty acid and n3-fatty acids. RNA-seq results showed upregulated fatty acid biosynthesis genes and downregulated mitochondrial beta-oxidation genes. The lipid metabolism in cold-treated longissimus dorsi muscle might be regulated by functions of the lipoprotein particle, the extracellular matrix, and the PPAR signaling pathways. Our study revealed the potential of cold exposure to regulate the lipid metabolism and fatty acid composition in skeletal muscle of farmed animals.

Maternal butyrate supplementation affects the lipid metabolism and fatty acid composition in the skeletal muscle of offspring piglets.

Maternal sodium butyrate (SB) intake has important effects on offspring growth and development. This study aimed to investigate the impacts of maternal SB supplementation during gestation and lactation on fatty acid composition and lipid metabolism in the offspring skeletal muscle of pigs. Twenty sows (Yorkshire, parity 2 to 3) were assigned to the control group (diets without SB, n = 10) and SB group (diets with 0.1% SB, n = 10). The results showed maternal SB supplementation throughout gestation and lactation increased (P < 0.05) body weight of offspring piglets at weaning. The concentrations of triglyceride in plasma and milk were enhanced (P < 0.05). Maternal SB induced (P < 0.05) lipid accumulation with increased expression of peroxisome proliferator activated receptor γ (PPARγ) by enrichment of the acetylation of H3 acetylation K27 (H3K27) in offspring skeletal muscle. Meanwhile, the concentrations of C18:2n-6, C18:3n-3, total polyunsaturated fatty acid (PUFA), n-6 PUFA and n-3 PUFA decreased (P < 0.05) in skeletal muscle of weaning piglets derived from SB sows. Together, these results showed that maternal SB supplementation could influence offspring growth performance, lipid metabolism and fatty acid composition of the skeletal muscle.

PGC-1α-mediated changes in phospholipid profiles of exercise-trained skeletal muscle

Exercise training influences phospholipid fatty acid composition in skeletal muscle and these changes are associated with physiological phenotypes; however, the molecular mechanism of this influence on compositional changes is poorly understood. Peroxisome proliferator-activated receptor γ coactivator 1α (PGC-1α), a nuclear receptor coactivator, promotes mitochondrial biogenesis, the fiber-type switch to oxidative fibers, and angiogenesis in skeletal muscle. Because exercise training induces these adaptations, together with increased PGC-1α, PGC-1α may contribute to the exercise-mediated change in phospholipid fatty acid composition. To determine the role of PGC-1α, we performed lipidomic analyses of skeletal muscle from genetically modified mice that overexpress PGC-1α in skeletal muscle or that carry KO alleles of PGC-1α. We found that PGC-1α affected lipid profiles in skeletal muscle and increased several phospholipid species in glycolytic muscle, namely phosphatidylcholine (PC) (18:0/22:6) and phosphatidylethanolamine (PE) (18:0/22:6). We also found that exercise training increased PC (18:0/22:6) and PE (18:0/22:6) in glycolytic muscle and that PGC-1α was required for these alterations. Because phospholipid fatty acid composition influences cell permeability and receptor stability at the cell membrane, these phospholipids may contribute to exercise training-mediated functional changes in the skeletal muscle.

Heat stress impairs mitochondria functions and induces oxidative injury in broiler chickens.

The objective of this study was to explore the linkage of oxidative stress occurring in mitochondria, skeletal muscles, and plasma in heat stress-challenged broilers. At d 35, 24 broilers were randomly assigned to 2 treatments: rearing at high temperature (32 ± 1°C; heat stress group) or normal temperature (21 ± 1.2°C; control) for 7 d. The oxidative damage of lipid, DNA, and protein and the activities of antioxidative enzymes were measured, respectively, in plasma, skeletal muscles (breast and thigh muscles), and skeletal muscle mitochondria. The result showed that heat exposure increased (P < 0.01) plasma concentrations of thiobarbituric acid reacting substances (TBARS) and 8-hydroxydeoxyguanosine (8-OHdG) whereas it deceased total antioxidant capacity (P < 0.05) and ability to inhibit hydroxyl radicals (AIHR; P< 0.001). Protein carbonyl and TBARS levels were increased (P < 0.001) by heat stress in breast and thigh muscles. In skeletal muscle mitochondria, heat stress increased (P < 0.05) 8-OHdG and suppressed AIHR. Plasma activity of superoxide dismutase (SOD) was increased (P< 0.001) whereas glutathione peroxidase (GSH-Px) was suppressed by heat stress (P < 0.001). Heat exposure increased SOD and catalase activities in breast muscle (P < 0.01) but the reverse was true in thigh muscle (P < 0.05). Glutathione peroxidase was increased in thigh muscle (P < 0.001) but was not changed in breast muscle (P > 0.05). Heat stress increased SOD (P < 0.05) and decreased GSH-Px activities (P < 0.05) of mitochondria regardless of muscle types. Plasma allantoin level increased (P < 0.01) correspondingly with urate (P < 0.001) in heat-stressed broilers, indicating that urate could serve as an antioxidant to enhance the antioxidative capacity during stress in a concentration-dependent manner. The activities of respiratory chain complexes I and III were estimated in skeletal muscle mitochondria. Mitochondrial complex I activity was suppressed (P < 0.01) by heat exposure in breast and thigh muscles but complex III activity was elevated only in breast muscle (P < 0.01) of heat-stressed broiler. The fatty acid composition in skeletal muscle was not influenced by heat stress. In conclusion, suppressed mitochondrial complex I activity is associated with oxidative stress induced by heat exposure, which, in turn, is linked with the oxidative damages in muscle tissues and plasma.

Genome-wide association and prediction of direct genomic breeding values for composition of fatty acids in Angus beef cattle.

BackgroundAs consumers continue to request food products that have health advantages, it will be important for the livestock industry to supply a product that meet these demands. One such nutrient is fatty acids, which have been implicated as playing a role in cardiovascular disease. Therefore, the objective of this study was to determine the extent to which molecular markers could account for variation in fatty acid composition of skeletal muscle and identify genomic regions that harbor genetic variation.ResultsSubsets of markers on the Illumina 54K bovine SNPchip were able to account for up to 57% of the variance observed in fatty acid composition. In addition, these markers could be used to calculate a direct genomic breeding values (DGV) for a given fatty acids with an accuracy (measured as simple correlations between DGV and phenotype) ranging from -0.06 to 0.57. Furthermore, 57 1-Mb regions were identified that were associated with at least one fatty acid with a posterior probability of inclusion greater than 0.90. 1-Mb regions on BTA19, BTA26 and BTA29, which harbored fatty acid synthase, Sterol-CoA desaturase and thyroid hormone responsive candidate genes, respectively, explained a high percentage of genetic variance in more than one fatty acid. It was also observed that the correlation between DGV for different fatty acids at a given 1-Mb window ranged from almost 1 to -1.ConclusionsFurther investigations are needed to identify the causal variants harbored within the identified 1-Mb windows. For the first time, Angus breeders have a tool whereby they could select for altered fatty acid composition. Furthermore, these reported results could improve our understanding of the biology of fatty acid metabolism and deposition.

Dietary CLA alters intramuscular fat and fatty acid composition of pig skeletal muscle and subcutaneous adipose tissue

The present study was conducted to determinate the responsiveness of different levels of dietary conjugated linoleic acid (CLA) on intramuscular fat (IMF) and fatty acid composition of skeletal muscle and fat in pigs fed from 59.5 to 133.5kg. Forty female Large Whitex(Large WhitexLandrace) pigs were used. Four levels (0%, 0.5%, 1% and 2%) of a commercial enriched CLA oil supplementation (60% of CLA isomers, 30% cis-9, trans-11 and 30% trans-10, cis-12) were fed to pigs. Carcass, ham, foreleg and loin weights were recorded. Dietary CLA enrichment increased the loin weight (P<0.01) and the combined weights of hams+forelegs+loins (P<0.02). IMF content in Longissimus dorsi was also increased by dietary CLA treatment (P<0.001) and a linear response was observed. Dietary CLA increased saturated fatty acids (SFA) and decreased monounsaturated fatty acids (MUFA) in muscle and adipose tissue (P<0.001). Feeding 1% CLA to finishing swine increases IMF in heavy pigs slaughtered at an average weight of 133.5kg.

Fatty acid composition of skeletal muscle and adipose tissue in Spanish infants and children

There is a relationship between the fatty acid profile in skeletal muscle phospholipids and peripheral resistance to insulin in adults, but similar data have not been reported in infancy and childhood. The objective of this study was to investigate the fatty acid composition of skeletal muscle and adipose tissue across the paediatric age range. The fatty acid profile of skeletal muscle phospholipids and adipose tissue triacylglycerols was analysed in ninety-three healthy Spanish infants and children distributed into four groups: group 1 (0 to <2 years, n 10); group 2 (2 to <5 years, n 41); group 3 (5 to <10 years, n 24); group 4 (10 to 15 years, n 18). In skeletal muscle phospholipids, oleic acid (18: 1n-9cis) content decreased significantly whereas that of linoleic (18: 2n-6) acid increased significantly with age (P for trend <0.01). In adipose tissue, the contents of triacylglycerol and linoleic acid increased significantly across the paediatric age range (P for trend <0.01), whereas dihomo-gamma-linolenic (20: 3n-6) and arachidonic (20: 4n-6) showed significant differences between groups. The variations in fatty acid composition observed with age indicated an imbalance in dietary n-3/n-6 long-chain PUFA.

Fatty acid composition of skeletal muscle reflects dietary fat composition in humans

It is still unknown whether the fatty acid composition of human skeletal muscle lipids is directly influenced by the fat composition of the diet. We investigated whether the fatty acid composition of the diet is reflected in the fatty acid profile of skeletal muscle phospholipids and triacylglycerols. Thirty-two healthy adults (25 men and 7 women) included in a larger controlled, multicenter dietary study were randomly assigned to diets containing a high proportion of either saturated fatty acids (SFAs) [total fat, 36% of energy; SFAs, 18% of energy; monounsaturated fatty acids (MUFAs), 10% of energy] or MUFAs (total fat, 35% of energy; SFAs, 9% of energy; MUFAs, 19% of energy) for 3 mo. Within each diet group, there was a second random assignment to supplementation with fish oil capsules [containing 3.6 g n-3 fatty acids/d; 2.4 g eicosapentaenoic acid (20:5n-3) and docosahexaenoic acid (22:6n-3)] or placebo. A muscle biopsy sample was taken from the vastus lateralis muscle after the diet period. Parallel analyses of diet and supplementation effects were performed. The proportions of myristic (14:0), pentadecanoic (15:0), heptadecanoic (17:0), and palmitoleic (16:1n-7) acids in the skeletal muscle phospholipids were higher and the proportion of oleic acid (18:1n-9) was lower in the SFA group than in the MUFA group. The proportion of total n-3 fatty acids in the muscle phospholipids was approximately 2.5 times higher, with a 5 times higher proportion of eicosapentaenoic acid (20:5n-3), in subjects supplemented with n-3 fatty acids than in those given placebo. Similar differences were observed in the skeletal muscle triacylglycerols. The fatty acid composition of skeletal muscle lipids reflects the fatty acid composition of the diet in healthy men and women.

Effects of physical exercise on phospholipid fatty acid composition in skeletal muscle.

The effects of low-intensity exercise on the fatty acid composition in skeletal muscle and in serum were studied in 19 sedentary, middle-aged Swedish men. During a 10-wk period, all subjects were given a standardized diet with an identical fat composition. After 4 wk on this diet, they were randomly allocated to a daily exercise program (55% peak oxygen uptake) or to continue to live a sedentary life for the remaining 6 wk. Aerobic capacity (submaximal bicycle test) and peripheral insulin sensitivity (hyperinsulinemic euglycemic clamp) improved with training, whereas the body weight as well as the body composition (underwater weighing and bioimpedance) were unchanged. The proportions of palmitic acid (16:0) and linoleic acid [18:2(n-6)] and the sum of n-6 fatty acids [18:2(n-6), 20:3(n-6), 20:4(n-6)] were decreased in skeletal muscle phospholipids, whereas the proportion of oleic acid [18:1(n-9)] was increased, by training. The fatty acid profile in skeletal muscle triglycerides remained unchanged. We conclude that regular low-intensity exercise influences the fatty acid composition of the phospholipids in skeletal muscle, which hypothetically may contribute to changes of the skeletal muscle membrane fluidity and influence the peripheral insulin sensitivity.

Tissue specific differences in the fatty acid composition of the marmoset monkey ( Callithrix jacchus)

1. 1. The possible relationship between the fatty acid composition of the adipose tissue of the marmoset monkey ( Callithrix jacchus) and its dietary lipid intake was examined after a long-term feeding trial. 2. 2. Only the proportions of stearic and linoleic acid in the adipose tissue were similar to those in the diet. The proportions of all other saturated and unsaturated fatty acid components were significantly different thus greatly reducing the potential value of this relationship in the marmoset. 3. 3. The phospholipid fatty acid profiles of cardiac and skeletal muscle, aortae, kidney, liver, lung and brain were also compared to those of the plasma, platelets or red blood cells. 4. 4. With the exception of the liver where the fatty acid profile is similar to that of the plasma, no clear relationships were found between the fatty acid profiles of these tissues and the components of the blood. 5. 5. The fatty acid composition of skeletal muscle was very similar to that of the cardiac muscle of the marmoset, suggesting that muscle biopsy might be useful as an index of cardiac muscle composition.

Amino acid composition of skeletal muscle of domestic buffalo (Bos bubalus L.)—II. Fractionation in three protein fractions and studies of their amino acid pattern

1.1. The skeletal muscle of Bos bubalus L. has been fractionated into three different protein fractions: sarcoplasmic, myofibrillar and insoluble.2.2. The amino acid composition of these fractions has been determined. The content of essential amino acids in the various fractions has been found to be very high in decreasing order from the sarcoplasmic to the myofibrillar and insoluble fraction.3.3. The “chemical score” followed by the indication of the limiting amino acid has been calculated to be 74 (Ileu), 71 (Val) and 65 (Phe), respectively, for the sarcoplasmic, myofibrillar and insoluble fraction.4.4. The high nutritional value of meat fractions from the domestic buffalo from the Mediterranean area is supported by experimental data.

Amino acid composition of skeletal muscle of domestic buffalo (Bos bubalus L.)—I. Comparative studies and nutritional value of proteins

1.1. In a search for new sources of protein, meat from the domestic buffalo—widely bred in southern Italy—has been investigated in order to ascertain the nutritional value of the skeletal muscle proteins. Comparative investigations have been performed on the basis of amino acid composition and essential amino acid content.2.2. It has been shown that the total amino acid content is an inverse function of wilderness conditions, when proteins from domestic buffalo skeletal muscle are compared with those from the domestic ox and African buffalo. The amino acid pattern is, however, very similar in the three species when the relative percentages are compared. Also, the total content of essential amino acids is very similar in the three species.3.3. The lysine content is much higher in the domestic buffalo as compared wlth whoie egg proteins (+ 44%), and also with the domestic ox. The results sugget that domestic buffalo skeletal muscle proteins have an amino acid composition which indicates a high nutritional value for the meat of this animal: its use as a food source is indicated on these data.
