Abstract

tRNA and aminoacyl-tRNA synthetases were obtained from undifferentiated virginal and differentiated (lactating and non-lactating) bovine and rabbit mammary glands. Comparative study indicates that the quantitative ratio of some individual tRNA's and possibly aminoacyl-tRNA synthetases in total preparations changes during differentiation in the mammary gland. The ability of IO − 4 to inactivate uncharged tRNA was used to examine charging levels of some tRNA's in vivo. The pool of aminoacyl-tRNA's in the lactating mammary gland proved to correspond to the amino acid composition of the main secretory proteins. The results of the study on the heterogeneity of leucyl-tRNA show that lactating cells synthesize leucine tRNA's of which one species is either absent in the non-lactating gland or present in undetectable concentration. Considerable difference was found in the ability of enzymes of virginal, lactating and non-lactating mammary glands to cross-react with heterologous tRNA.
