Abstract

The two transfer factors from bean rust [ Uromyces phaseoli (Pers.) Wint.] uredospores were resolved, and the aminoacyl-tRNA binding enzyme (Transferase I) was purified for further study. Transferase I catalyzes the polyuridylic acid-directed and GTP-dependent binding of Phe-tRNA to ribosomes, and is required with Transferase II for the synthesis of peptides from aminoacyl-tRNA. Purified Transferase I is free of detectable Transferase II, and has a molecular weight estimated to be 145 000 g per mole by a combination of gel filtration and density gradient techniques.
