Abstract

A method for the preparation of tRNA from Escherichia coli is described. A crude tRNA is prepared by phenol and water treatment of cells without the need of centrifugation for the separation of an aqueous phase from a phenolic phase containing cellular debris. Dimethyl sulfoxide and NaCl are used for the purification of the tRNA in good yield without a requirement for column chromatography. Amino acid acceptance assays are used to quantitate the purification and to characterize the preparations.
