Abstract

DNA was extracted from single soil-borne seeds ofOrobanche ramosaL.,O. crenataForsk,O. cumanaLoeffl. andO. aegyptiaca. Species identification was then possible by RAPD-PCR, based on specific DNA markers, and could be verified by Southern hybridization with specific probes. Seed identification was also possible for dead (tetrazolium-negative) seeds. This method is a powerful diagnostic tool forOrobancheclassification in agricultural fields.
