Abstract

Somatic embryogenesis is a widespread phenomenon in alfalfa callus and cell cultures. However, many problems have to be solved before this technique can be routinely applied to breeding and agriculture. In this study we tried to clarify the significance of several factors related to the improvement of somatic embryogenesis in Medicago sativa L. cvs. Rangelander, 74RS, and Furez; M. varia cv. 868 Rambler; and M. falcata cvs. 39, 47, and 49. To simplify the examination, only B5 modified media (B5-II) was used in the course of this study: macro- and microsalts, vitamins [Gamborg et al. (1968) Exp. Cell Res. 50:151–158], casein hydrolisate (500 mg/1), myoinositol (500 mg/1), FeEDTA [Murashige and Skoog (1962) Physiol. Plant 15:473–497], sucrose (30 g/1), L-glutathione (10 mg/1), 2,4-dichlorophenoxyacetic acid (2,4-D) (1 mg/1), kinetin (0, 2 mg/1), adenine (1 mg/1), and bactoagar (8 g/1).
