Publisher Summary This chapter reviews the reports on the recognition of the mixed lymphocyte cultures (MLC) determinants by typing cells. It is unsettled whether the MLC determinants are simple monovalent structures or are the sum of a number of subunits. There is a strong linkage disequilibrium between MLC (D) and four (B) locus determinants. Responder cells pre-sensitized against a certain MLC determinant can recognize that determinant also in unrelated individuals.Lymphoblastoid cellines made from typing cells can be used for MLC typing. In this way, the need for repeated bleedings of the typing cell donors is evaded. The chapter further reviews some reports on the recognition of the MLC determinants and other closely linked loci by serology. It describes a study of sera with antibodies with presumed LD specificity and also presents a summary of the results obtained from a workshop on the typing of human MLC determinants.
