Abstract
This chapter focuses on the results of primed LD typing (PLT). Antigens of the HLA-D complex in man are described by use of the mixed leukocyte culture (MLC) test. A new test called PLT has been developed to measure HLA-D antigens. To prepare a PLT cell, lymphocytes of two haplo-identical related individuals are cultured for approximately 10 days. Discriminatory PLT cells have the ability to pick out cells from some individuals that restimulate strongly in the same range as the cells of the individual initially used for sensitization. HLA-A, -B, and -C loci antigens do not cause strong restimulation. The antigen PL 3 may be associated with a new DW specificity, RE. PLT may be an extremely useful procedure for studying the immunogenetic complexity of the HLA-D region in man.
