Abstract

This chapter focuses on online monitoring of glucose in batch fermentations of E. coli with a commercially available biosensor analyzer-fermentation control system. During the cultivation of E. coli, the consumption of glucose is monitored in the range of 20 gl−1 to practically 0 gl−1 over 8 h. In a study described in the chapter, a monitoring was performed with a new analytic system provided by B. Braun Biotech International, Melsungen, and FRG. Glucose was detected by a specially modified version of the biochemical analyzer Yellow Springs Instruments (YSI) 2700 coupled to a host computer. The biosensor electrode in this apparatus is one of the most widespread ones in the world. Incorporated in analyzers as model YSI 27, it is often used as reference method in cultivation technology. The correlations to off-line measurements including Boehringer enzyme assay as reference were good.
