Abstract

Rapid and complete lysis of Escherichia coli cells was obtained by treating the cells with ethanol prior to the treatments with EDTA, lysozyme and detergent. The lysis was observed in the cells in stationary phase as well as in log phase. The use of ethanol is expected to interrupt cellular metabolism immediately yielding non-degraded DNA in a sucrose gradient solution containing sodium dodecylsulfate.
