Abstract

The recent advances in cell and tissue biology and pathology have been enormous. The combination of genomics and proteomics with imaging techniques has contributed greatly because expression patterns are analyzed in intact cells and tissues in their intact microenvironment. This genomic and proteomic mapping of the physiological and pathological dynamics in cells and tissues does not pinpoint specific regulators that are potentially diagnostic tools or therapeutic targets as the activity of enzymes can be. Activity of enzymes is tightly regulated at all levels between transcription and posttranslational modifications. Its localization in an intact cellular and tissue microenvironment, also called metabolic mapping, provides functional insights into dynamic processes in cells and tissues that can be useful for diagnostics and the development of drugs. Here, protocols are described for valid metabolic mapping of a range of enzymes.
